Abstract

• Bioengineering of Lactic Acid Producing Bacterial (LAB) vector secreting functionally bioactive chicken IL-17A. • Pre-treatment of primary chicken embryo fibroblasts (CEFs) with recombinant IL-17A induced a strong antiviral host state. • sChIL-17A induced pro-inflammatory responses inhibit and delay onset of Type A influenza virus replication. • The findings underscore the prophylactic potential of LAB vector expressing cytokine against AIV infections in chickens. The recent advances in our understanding of the host factors in orchestrating qualitatively different immune responses against influenza Type A virus (IAV) have changed the perception of conventional approaches for controlling avian influenza virus (AIV) infection in chickens. Given that infection-induced pathogenicity and replication of influenza virus largely rely on regulating host immune responses, immunoregulatory cytokine profiles often determine the disease outcomes. However, in contrast to the function of other inflammatory cytokines, interleukin-17A (IL-17A) has been described as a 'double-edged sword', indicating that in addition to antiviral host responses, IL-17A has a distinct role in promoting viral infection. Therefore, in the present study, we investigated the chicken IL-17A mediated antiviral immune effects on IAVs infection in primary chicken embryo fibroblasts cells (CEFs). To this end, we first bioengineered a food-grade Lactic Acid Producing Bacteria (LAB), Lactococcus lactis (L. lactis), secreting bioactive recombinant chicken IL-17A (sChIL-17A). Next, the functionality of sChIL-17A was confirmed by transcriptional upregulation of several genes associated with antiviral host responses, including granulocyte-monocyte colony-stimulating factor (GM-CSF) (CSF3 in the chickens), interleukin-6 (IL-6), interferon-α (IFN-α), -β and -γ genes in primary CEFs cells. Consistent with our hypothesis that such a pro-inflammatory state may translate to immunoprotection against IAVs infection, we observed that sChIL-17A pre-treatment could significantly limit the viral replication and protect the primary CEFs cells against two heterotypic IAVs such as A/turkey/Wisconsin/1/1966(H9N2) and A/PR/8/1934(H1N1). Together, the data presented in this work suggest that exogenous application of sChIL-17A secreted by modified LAB vector may represent an alternative strategy for improving antiviral immunity against avian influenza virus infection in chickens. Abstract

The grand-scale laboratory investigation on finding influenza type A virus in wild and synanthropic birds on vast territories of Eastern Siberia was conducted for the first time. Alongside with the ornithological investigations, we also conducted collection of over 17,000 samples of laboratory investigations of 165 bird species. In the south of Central Siberia in 2006-2011, the antibodies to antihemagglutination were found in 484 samples, 179 out of which were from synanthropic birds. During the investigation of PCR of RNA avian influenza 167 samples were isolated, 17 (10,2 %) out of which were from synanthropic species. In 2014, antibodies were isolated from 30 species, RNA avian influenza were isolated from 26 species of the following bird orders: Podicipediformes - 3/1 (in hemagglutination-inhibition reaction/polymerase chain reaction) of species, Ciconiiformes - 0/1, Anseriformes - 12/10, Gruiformes - 1/1, Charadriiformes - 7/4, Passeriformes - 6/8. In the following years the participation in epizootic process of new species/ subspecies were increased. The maximum virus carriage (40.2 %) and birds infection takes place during the period from July 15 till August 28, which is characterized mainly by the presence of a large number of multipleaged young-of-the-year water fowls on the ponds and riparian vegetation. In the process of work, there was compiled not only the list of species/subspecies of avian influenza virus carriers and vectors, but also revealed their connection with wintering areas and the main location of the recombinant subtypes of highly pathogenic virus H5N1. The cases of virus spreading and its influence on the birds of various ecologic groups were described.

Abstract

In 2004, canine influenza virus subtype H3N8 emerged in greyhounds in the United States. Subsequent serologic evidence indicated virus circulation in dog breeds other than greyhounds, but the virus had not been isolated from affected animals. In 2005, we conducted virologic investigation of 7 nongreyhound dogs that died from respiratory disease in Florida and isolated influenza subtype H3N8 virus. Antigenic and genetic analysis of A/canine/Jacksonville/2005 (H3N8) and A/canine/Miami/2005 (H3N8) found similarity to earlier isolates from greyhounds, which indicates that canine influenza viruses are not restricted to greyhounds. The hemagglutinin contained 5 conserved amino acid differences that distinguish canine from equine lineages. The antigenic homogeneity of the canine viruses suggests that measurable antigenic drift has not yet occurred. Continued surveillance and antigenic analyses should monitor possible emergence of antigenic variants of canine influenza virus.

Influenza is associated with high morbidity and may result in serious complications such as life threatening viral or bacterial pneumonia. Especially, young children, older adults, patients with chronic diseases such as heart, lung, kidney, and diabetes and immunosuppressed people are at higher risk for complications and death from influenza virus infections. The aim of this study was to determine the incidence of influenza type A and B virus infections and influenza A virus subtypes in hospitalized patients with respiratory tract infections by real-time reverse transcriptase-polymerase chain reaction (RT-PCR, Sacace, Italy), conventional RT-PCR and direct immunofluorescence antibody (DFA, Argene SA, France) tests. Nasopharyngeal swab specimens were collected from a total of 476 patients with respiratory tract symptoms by using flocked swabs (Copan Diagnostics, Italy) between 1 April 2012 and 31 December 2013. Influenza A virus was detected in 20.5% (98/476) and influenza B virus in 3.3% (16/476) of the cases by real-time RT-PCR test. During the study period, 63.3% of 98 influenza virus isolates were found as influenza A(H1N1) pdm09 and 36.7% were influenza A(H3N2) subtypes. Influenza A (H1N1) pdm09 subtype was observed in 12 cases in January 2013 and influenza A(H3N2) subtype was observed in 11 cases in December 2013 as the highest values. When the real-time RT-PCR test was regarded as the reference test, the sensitivities of DFA test for influenza A and B and conventional RT-PCR test with WHO primers (M30F2/08 and M264R3/08) for influenza A were detected as 72.4%, 75%, 96% and the specificities were detected as 99.2%, 99.5% and 100%, respectively. In conclusion, influenza A virus infection was detected rather high with a rate of 20.5% in the study group. The monitoring of influenza virus types and subtypes is required for the evaluation of influenza vaccine strains and circulating influenza viruses and for the identification of subtypes with pandemic potential. Planning for appropriate antiviral therapy using real-time RT-PCR in the early diagnosis of influenza virus infections will significantly contribute to the management of the patient's treatment. Thus, unnecessary drug use will be prevented and controlled with effective treatment of the disease at the time of infection.

İnfluenza virus enfeksiyonları yüksek morbidite ile ilişkilidir ve pnömoni gibi ciddi komplikasyonlara sebep olabilir. Bu çalışmanın amacı solunum yolu infeksiyonu olan ve hastaneye yatırılan hastalarda influenza tip A ve B virus enfeksiyonlarının insidansı ve influenza A virus alttiplerini real-time RT-PCR test, konvansiyonel RT-PCR test ve direk immunfloresan antikor (DFA) testi ile tespit etmektir. Toplam 476 hastadan 1 Nisan 2012 ve 31 Aralık 2013 tarihleri arasında flocked eküvyon (Copan Diagnostics, Italy) ile nazofaringeal sürüntü örnekleri toplanmıştır. Real-time RT-PCR test (Sacace, Italy) ile influenza A virus vakaların %20.5 (98/476) ve influenza B virus %3.3’ünde (16/476) tespit edilmiştir. Çalışma süresi boyunca 98 influenza virus izolatının %63.3’ünün A(H1N1)pdm09 ve %36.7’sinin A(H3N2) olduğu bulunmuştur. İnfluenza A(H1N1)pdm09 alttipinin Ocak 2013 (12 vaka) ve A(H3N2) alttipinin (11 vaka) Aralık 2013’de baskın olduğu gözlenmiştir. Real-time RT-PCR test referans alındığında influenza A ve B için DFA testinin (Argene SA, France) ve influenza A için WHO primerlerinin (M30F2/08 ve M264R3/08) kullanıldığı konvansiyonel RT-PCR testinin sensitivitesi sırasıyla %72.4, %75, %96 ve spesifitesi %99.2, %99.5 ve %100 bulunmuştur. Sonuç olarak, çalışma grubunda influenza A virus enfeksiyonu %20.5 oranla oldukça yüksek tespit edilmiştir. İnfluenza virus tipleri ve alttiplerinin izlenmesi, influenza aşı suşlarının dolaşımdaki influenza virusları ile uyumunun değerlendirilmesi ve pandemik potansiyeli olan alttiplerin tanısı için gereklidir. Etkili korunma ve kontrol önlemlerinin zamanında alınması, gereksiz antibiyotik kullanımının azaltılması ve antiviral tedaviye başlamak için influenza virus enfeksiyonlarının erken tanısında real-time RT-PCR testinin kullanımı önerilir.
