The need to reduce the use of chemical pesticides currently fosters great interest in eco-friendly biological control agents. In addition, the isolation of plant allelopathic substances and the evaluation of their phytotoxic effects can lead to the discovery of new natural herbicides. In this context, our study aimed to assess the herbicidal activity of ten crude extracts obtained from aerial parts of Tunisian spontaneous plants against Trifolium incarnatum, Silybum marianum and Phalaris minor. It confirmed that the Cynara cardunculus methanolic extract best inhibited weed germination and seedling growth, and caused necrosis or chlorosis. Following a bioassay-guided fractionation, five main phenolic compounds, (1) syringic acid, (2) p-coumaric acid, (3) myricitrin, (4) quercetin and (5) naringenin were identified in the most active crude methanolic extract. Then, only 3 of the flavonoids contained in the most active fraction were tested on Trifolium incarnatum. The 3 compounds had a significant phytotoxic effect and therefore could be employed in a new composition of botanical herbicides to control crop weeds. Besides, a novel herbicide composition was designed to improve the post-emergence activity of the methanolic extract. The formulation containing the C. cardunculus crude methanolic extract showed the same herbicidal activity as the standard industrial bioherbicide containing pelargonic acid. These results make C. cardunculus a suitable source of natural compounds potentially usable as natural herbicides.

The effect of drying methods (sun, oven and shade drying) on aniseeds was investigated in terms of their essential oils, phenolics and antioxidant activities. The optimum yield of essential oil was found in shade drying (2.62%). Fourteen volatile compounds were determined in all samples with variation of the main component proportions depending on drying methods. Thus, trans-anethole (84.21%) and estragole (3.82%) proportions significantly increased in shade drying. The highest total phenol and flavonoid contents of aniseeds were recorded in shade drying (42.70 mg of GAE/g and 53.55 mg of QE/g, respectively) while the lowest contents in oven drying at 60 °C (31.15 mg of GAE/g and 46.20 mg of QE/g, respectively). In all drying methods, naringin (41.04–43.76%), chloroginic acid (23.13–27.19%) and rosmarinic acid (12.26–15.95%) were the predominant phenol compounds. Although shade drying increased the antioxidant activity, aniseed extracts exhibited higher radical scavenging (IC50 = 10.15 µg/mL), reducing power (EC50 = 187.24 µg/mL) and chelating (IC50 = 6.85 mg/mL) capacities than essential oils (IC50 = 114.87 µg/mL, EC50 = 548.05 µg/mL and IC50 = 58.65 mg/mL, respectively). In conclusion, Shade drying method was found to enhance essential oils, phenols and antioxidant activities in aniseeds.

The aim of this study was to evaluate the replacement aspects of conventional methods (petroleum-based solvent and Folch assay) by alternative methods (bio-based and biodegradable solvent 2-methyltetrahydrofuran (MeTHF) and supercritical CO2 (SC-CO2)) for seed oil extraction from anise (Pimpinella anisum L.) and fennel (Foeniculum vulgare Mill.). Results showed that the highest oil yield of aniseeds was obtained by using Folch (24.07%) and MeTHF (23.65%) extraction methods whereas fennel seeds had 20.02% and 18.72%, respectively. Fatty acid composition of both seed oils obtained by the two green extraction methods was similar to the conventional ones with the predominance of petroselinic acid (54.22–61.25% in fennel and 42.39–48.97% in anise). Besides, SC-CO2 method allowed to obtain the maximum of sterol content in anise (3.85 mg/g of oil) and fennel (4.64 mg/g of oil) seed oils. Chemical composition and antioxidant activity of four aniseed populations (Egyptian, Serbian, Tunisian and Turkish) were investigated during three developmental stages. The highest oil yield was achieved at full maturity in all aniseed accessions ranged from 11.93% (Serbia) to 13.80% (Tunisia). Fatty acid profile of aniseed oil was characterized by high proportions of palmitic (4.90–57.18%) and petroselinic (10.48–46.60%) acids which had an antagonist evolution during maturation. The essential oil yield reached its maximum at the beginning of ripening process in all aniseed accessions ranged from 1.94% (Serbia) to 3.09% (Tunisia). The main essential oil compound was trans-anethole (66.34–93.05%) during aniseed ripening in all accessions. Phenolic content patronized its maximum at the last stage of aniseed ripening ranged from 17.11 mg GAE/g DW (Serbia) to 25.16 mg GAE/g DW (Tunisia). The main phenolic compound of aniseed was naringin (17.55–32.49%) and its accumulation was followed by the reduction of gallic, rosmarinic, ellargic and syringic acids during aniseed ripening in all accessions. Concerning antioxidant activity, DPPH scavenging activity, chelating ability and reducing power were maximal at full maturity in all aniseed accessions. Our findings indicate that the determination of optimal periods and provenances for antioxidant accumulation can be used to evaluate the quality of aniseeds and could be important for industries.

In this work, LC-ESI-TOF-MS and GC-EI-MS were used to assess the potential of Artemisia herba alba as a source of health-promoting constituents. Besides, the antioxidant, the antimicrobial and the cytotoxic potentials were evaluated. A total of 86 metabolites, including C-glycosylated and methylated flavones, quinic acid derivatives, coumarins, sesquiterpenes lactones, terpenoids, fatty acids, carbohydrates, organic acids and alkaloids were identified, sixty five of them were reported for the first time in A. herba alba. The main compounds were di-O-caffeoylquinic acids, artemisinic acid, menthol, α-ketoglutaric acid, scopolin, isoschaftoside and sucrose. Furthermore, results showed that A. herba alba possess high total antioxidant activity (Total antioxidant activity=423mg gallic acid equivalent/g dry weight), strong potential anticancer capacity against MCF-7 breast cancer and HeLa human cervical cell lines (IC50 of 15 and 70μg/ml, respectively), and moderate antibacterial activity against S. aureus, B. thurigiensis and A. hydrophila. These results support the use of this plant as alternative bioactive ingredient for functional foods, dietary supplements or nutraceuticals.

In this study, the effects of salinity on growth, fatty acid, essential oil, and phenolic composition of cumin (Cuminum cyminum L.) seeds as well as the antioxidant activities of their extracts were investigated. Plants were treated with different concentrations of NaCl treatment: 0, 50, 75, and 125 mmoL. Plant growth was significantly reduced with the severity of saline treatment. This also caused important reductions in the seed yield and yield components. Besides, NaCl treatments affected fatty acid composition. Petroselinic and linoleic acids proportions diminished consistently with the increase in NaCl concentration, whereas palmitic acid proportion increased. Furthermore, NaCl enhanced essential oil production in C. cyminum seeds and induced marked changes on the essential oil quality. Essential oil chemotype was modified from γ-terpinene/1-phenyl-1,2 ethanediol in control to γ-terpinene/β-pinene in salt stressed plants. Total polyphenol content was higher in treated seeds, and salinity improved the amount of individual phenolic compounds. Moreover, antioxidant activities of the extracts were determined by four different test systems, namely 2,2-diphenyl-1-picrylhydrazyl, β-carotene/linoleic acid chelating, and reducing power assays. The highest antioxidant activities were reveled in severe stressed plants. In this case, cumin seeds produced under saline conditions may function as a potential source of essential oil and antioxidant compounds, which could support the utilization of this plant in a large field of applications such as food industry.

This study was designed to examine the effect of provenance on the phytochemical and antioxidant properties of essential oil and methanolic extract from Tunisian and French fennel seeds (FFS). Analysis of the essential oil showed that although the same main compounds were found in Tunisian and FFS cultivars, some differences were present in their proportions allowing to classify them in two chemotypes. The first class was composed by trans-anethole (63.41%-78.26%) for Tunisian cultivars and the second one by estragole (44.72%-88.92%) for French cultivars. The phenolic composition of all fennel seed extracts was characterized by its richness in quinic acid, 4-O-caffeoylquinic acid, p-coumaric acid, and 4-O-caffeoylquinic acid. All fennel seed extracts showed a better antioxidant potential than their essential oils depending on the origin. Principal component analysis showed a dispersion of the cultivars on three groups depending on the chemotype diversity. PRACTICAL APPLICATIONS: Recently, much attention has been focused on fennel due to the nutritional and health-protective value of their seeds. Several studies have highlighted the importance of fennel seed extracts and essential oils as key ingredients rich in bioactive compounds serving in formulation of new functional food products. This investigation designed to examine the effect of provenance on phytochemical and antioxidant potentials of Tunisian and French fennel seed extracts and essential oils.

Introduction The consumption of health-promoting products such as oil seeds may improve human health and prevent certain diseases. Carvi seeds have the potential to produce oil with nutritional and functional properties rich in active compounds. Objective To extract bioactive lipids from Carum carvi seeds using green methodologies. Material and methods Supercritical-carbon dioxide (Sc-CO2 ) and ethanol as co-solvent and bio-based solvent 2-methyltetrahydrofuran (MeTHF) were used to extract the oil from Carum carvi. The yield, the chemical composition, as well as antioxidant and anti-inflammatory activities of green extracted oils were investigated and compared to those obtained with conventional methods (hexane and Folch system). Results MeTHF extraction gave higher oil yield than that obtained by hexane. Fatty acids composition of the two obtained green extracted oils was similar to conventional extracted ones where petroselinic (39-43%), linoleic (29-31%) and oleic (19-21%) acids were the major compounds. Furthermore, MeTHF and Sc-CO2 green extracted oils were enriched of bioactive compounds including sterols (5.4 and 7.3 mg/g oil) and total polyphenols (9.3 and 7.6 mg GAE/g oil) which were correlated to enhanced antiradical capacity. Moreover, the green extracted oils exhibited high anti-inflammatory capacity inhibiting nitric oxide (NO) release in lipopolysaccharide (LPS)-induced RAW 264.7 macrophages with IC50 values of 28 and 24 μg/mL. Conclusion Green solvents are a good alternative to petroleum solvents to recover oil from carvi seeds with high amount of nutritionally important fatty acids, along with significant antioxidant and anti-inflammatory potential.

In order to reduce the use of chemical pesticides, great interest has been focused on environment-friendly biological control agents and botanicals that preserve biodiversity. In this context, our study aimed to assess the antifungal and herbicidal activities of Rosmarinus officinalis essential oil (EO) to find an alternative to synthetic pesticides. The chemical composition of R. officinalis essential oil was determined by gaz chromatography-mass spectrometry analysis (GC-MS). Results showed that R. officinallis EO was rich in monoterpenes and the major constituents were 1,8-cineole (54.6%), camphor (12.27%) and α-pinene (7.09%). However, under laboratory conditions, two tests were carried out. The first one consisted on the study of EO antifungal activity using ELISA microplates and the second one consisted on evaluating the effect of EO on seedling growth of weeds. It was confirmed that this EO significantly inhibits spore germination of Fusarium oxysporum, Fusarium culmorum, Penicillium italicum and at 6 mM, the percentage of inhibition reached 100% on Fusarium oxysporum. Indeed, EO slows down seedling growth of Trifolium incarnatum, Silybum marianum, and Phalaris minor. In fact, EO at 5 mM completely inhibits seed germination. On the other hand, another experiment was carried out to evaluate the herbicidal activity by spraying EO on weeds. This showed that a novel herbicide formulation was set up for the first time to improve the activity of R. officinalis EO on post-emergence. Overall, R. officinalis EO can be suggested as a potential eco-friendly pesticide and suitable source of natural compounds potentially usable as natural pesticides.

This study was conducted to investigate potentially protective and curative effects of Curcuma longa root (turmeric) powder on CCl(4)-induced hepatotoxicity in rats. Turmeric was administered before (preventive effect) or after (curative effect) treatment with CCl(4). Total phenolic and flavonoid levels were 26.35 mg GAE/g and 12.35 mg CE/g, respectively. Using HPLC analysis, turmeric powder was rich in curcumin (62.97%), demethoxycurcumin (20.86%) and bisdemethoxycurcumin (16.17%). Curcuma longa powder showed important in vitro antioxidant activities. Results showed that the activities of aspartate aminotransaminase and alanine aminotransaminase, and the levels of bilirubin and serum lipids were increased in CCl(4)-treated animals. However, total protein and albumin levels and antioxidant enzyme activities were decreased. Turmeric administration, before or after CCl(4) treatment, significantly decreased the activities of marker enzymes and lipid levels in serum. Moreover, total protein and albumin contents were restored to nearly normal levels after turmeric administration accompanied with increase of antioxidant enzymes activities.

The present study aimed to assess antioxidant activities of three organs (flower, fruit, and stem) extracts of Tunisian Ferula (F.) communis. Various experimental models were used to characterize the antioxidant activities in vitro as well as on ROS-induced fluorescence using dichlorofluorescein technique from phorbol myristate acetate (PMA)-stimulated human myeloid cell line HL-60. Results showed that the antioxidant activities varied considerably with organs. Thus, flower exhibited higher DPPH-scavenging ability, reducing and chelating power than stem and fruit. Also, antioxidant capacities using ORAC method and a cell-based assay showed that fruit and stem exhibited statistically similar antioxidant activities. Moreover, F. communis contains high amounts of flavonoids with various health benefits attributed to their antioxidant potential. Likewise, to obtain biologically relevant information, the antioxidant activities of the extracts were evaluated on cellular models implicating the antioxidant activities; this test generally showed that F. communis flower extracts have the highest antioxidant capacities correlated to the highest total phenolic content. The identification of phenolic compounds in F. communis extracts using RP-HPLC revealed that resorcinol, ferulic, and syringic acids together with coumarin were the major molecules.

The antioxidant and antibacterial activities of phenolic compounds from cultivated and wild Tunisian Ruta chalepensis L. leaves, stems, and flowers were assessed. The leaves and the flowers exhibited high but similar total polyphenol, flavonoid, and tannin content. Moreover, two organs showed strong, although not significantly different, total antioxidant activity, 2,2-diphenyl-1-picrylhydrazyl scavenging ability, and reducing power. Investigation of the phenolic composition showed that vanillic acid and coumarin were the major compounds in the two organs, with higher percentages in the cultivated organs than in the spontaneous organs. Furthermore, R. chalepensis extracts showed marked antibacterial properties against human pathogen strains, and the activity was organ- and origin-dependent. Spontaneous stems had the strongest activity against Pseudomonas aeruginosa. From these results, it was concluded that domestication of Ruta did not significantly affect its chemical composition and consequently the possibility of using R. chalpensis organs as a potential source of natural antioxidants and as an antimicrobial agent in the food industry.

In the current study, we determined the effects of seven drying methods on total phenolics, flavonoids, individual phenolics, and antioxidant activity of the methanol extract of Salvia officinalis L. As compared with total phenolic content (TPC) of fresh plants, results showed that the highest TPC was recorded in plants dried by microwave (MW) at a power of 800 W/30 g of fresh plant and was 4.2 times higher than that of fresh plants whereas the lowest content was found in the case of plants dried by far-infrared (FIR) at 45 °C. The analysis of the different extracts by RP-HPLC showed a predominance of phenolic acids particularly in fresh plants and those dried by MW (600 W/30 g of fresh plant) whereas flavonoids predominate in the case of plants dried by FIR (65 °C). The assessment of the radical scavenging activity (RSA) against the stable radical 1,1-diphenyl-1-picrylhydrazyl (DPPH) showed an increase in the scavenging effect particularly in MW (800 W/30 g of fresh plant) dried plants with an IC50 = 13.49 μg ml−1 (IC50 is the concentration required to cause 50 % DPPH inhibition). The complementary assessment of the RSA using the β-carotene/linoleic acid system showed an increase of this activity for all extracts and particularly for the extract derived from MW (600 W/30 g of fresh plant) dried plants as compared to fresh ones. Finally, all the plant extracts showed moderate reducing power as assessed by the ferric-reducing antioxidant potential. These results suggested that MW drying could be applied to retain phenolic contents and to enhance antioxidant activity of sage plant materials.

This study is designed to examine the yield components, fatty acid, and essential oil compositions and phenolic contents fruit essential oil composition, the total phenolic amounts as well as the antioxidant activities of cumin (Cuminum cyminum L.) seeds under drought. This plant is one of the most common aromatics in the Mediterranean kitchen. Plants were treated with different levels of water deficit: control (C), moderate water deficit (MWD) and severe water deficit (SWD). Our results indicated that MWD improved the number of umbels per plant as well as the number of umbellets per umbel and the seed yield, in comparison to the control, but it decreased under SWD. Fatty acid composition analysis indicated that petroselinic acid was the major fatty acid (55.9%) followed by palmitic (23.82%) and linoleic (12.40%) acids. Water deficit enhanced the palmitic acid percentage and affected the double bound index of the fatty acid pool and thus the oil quality. The essential oil yield was 1.64% based on the dry weight and increased by 1.40 folds under MWD. Nevertheless it decreased by 37.19% under SWD in comparison to the non treated seeds. Drought results on the modification of the essential oil chemotype from γ-terpinene/phenyl-1,2 ethanediol in the control seeds to γ-terpinene/cuminaldehyde in stressed ones. Besides, total phenolic contents were higher in the treated seeds (MWD and SWD). Results suggest that water deficit treatment may regulate the production of bioactive compounds in cumin seeds, influencing their nutritional and industrial values. Besides, antioxidant activities of the extracts were determined by four different test systems, namely DPPH, β-carotene/linoleic acid chelating and reducing power assays and showed that treated seeds (MWD and SWD) exhibited the highest activity.

Chemical composition and antioxidant activity of Salvia officinalis essential oil (EO) were studied under different drying methods of plant material. Results obtained showed that dried plant material yielded more essential oils than did the fresh one. The highest EO yields were obtained by infrared drying at 45 °C (0.39%) followed by air drying (0.30%) and oven drying at 45 °C (0.26%). The analysis of EOs by gas chromatography–mass spectrometry, showed in each of 55 identified compounds belonging mostly to oxygenated monoterpenes. Polyphenol contents and 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) scavenging activity of cumin (Cuminum cyminum L.) seed extracts were compared depending on their geographical origin (Tunisia and India: TCS and ICS, respectively) and the extraction solvent polarity. The β-carotene bleaching assay, the chelating ability and the reducing power of the most promising solvent extracts were also assessed. In addition, TCS and ICS extracts were acid-hydrolyzed and the phenolics identified by reversed-phase high-performance liquid chromatography (HPLC). Seed phenolic contents and antioxidant activity appeared to be accession and solvent dependent. Extraction with 80% acetone led to the highest polyphenol (18.60 and 16.50 mg gallic acid equivalents (GAE)/g dry weight (DW)), flavonoid (5.91 and 4.99 mg catechin equivalents (CE)/g DW) and tannin (83.23 and 80.23 mg CE/g DW) contents, respectively for TCS and ICS. DPPH scavenging activity, β-carotene bleaching assay, chelating ability and reducing power were maximal in 80% acetone for both TCS and ICS. HPLC analysis revealed several phenolic compounds in C. cyminum seeds, with p-coumaric (4.83 and 2.33 mg/g DW), trans-2-dihydrocinnamic (1.09 and 1.20 mg/g DW) and rosmarinic (0.70 and 1.04 mg/g DW) acids as major phenolics in TCS and ICS, respectively. Thus, phenolic composition of cumin seeds is also origin dependent. Taken together, our findings indicate that cumin might constitute a rich and novel source of natural antioxidants as food additives in food industry and that acetone 80% would be the most appropriate solvent for seed extraction.

Giant fennel (Ferula communis L.) is well known in folk medicine for the treatment of various organ disorders. The biological importance of members of genus Ferula prompted us to investigate the leaves of the endangered Tunisian medicinal plant F. communis L. not previously investigated. An estimate of genetic diversity and differentiation between genotypes of breeding germplasm is of key importance for its improvement. Thus, four F. communis populations were RAPD fingerprinted (63 RAPD markers generated by 7 primers) and the composition of their leaf essential oils (EO) (134 EO compounds) was characterized by GC-MS. Cluster analysis based on the leaf volatiles chemical composition of F. communis accessions defined three chemotypes according to main compounds have been distinguished: α-eudesmol/β-eudesmol/γ-terpinene; α-eudesmol/α-pinene/caryophyllene oxide and chamazulene/α-humulene chemotypes. A high genetic diversity within population and high genetic differentiation among them, based on RAPDs, were revealed (H(pop)=0.320 and GST=0.288) caused both by the habitat fragmentation, the low size of most populations and the low level of gene flow among them. The RAPD dendrogram showed separation of three groups. Populations dominated by individuals from the β-eudesmol/γ-terpinene; chemotype showed the lowest gene diversity (H=0.104), while populations with exclusively α-pinene/caryophyllene oxide chemotype showed the highest value (H=0.285). The UPGMA dendrogram and PCA analysis based on volatiles yielded higher separation among populations, indicated specific adaptation of populations to the local environments. Correlation analysis showed a non-significant association between the distance matrices based on the genetic markers (RAPD) and chemical compounds of essential oil (P>0.05) indicating no influence of genetic background on the observed chemical profiles. These results reinforce the use of both volatile compounds and RAPD markers as a starting point for in situ conservation. The analysis of chemical constitution of oil of the populations from a specific region revealed predominance of specific constituents indicating possibility of their collection/selection for specific end uses like phytomedicines. Sufficient molecular and biochemical diversity detected among natural populations of this species will form the basis for the future improvement. The correlation between matrices of RAPD and essential oils was not significant. The conservation strategies of populations should be made according to their level of genetic and chemical diversity in relation to geographic location of populations. Our results give some insights into the characterization of this as yet little investigated plant.

The aim of this work was to characterize Tunisian cactus pear (Opuntia ficus indica L.) flower through its lipids, essential oils, phenolics, and antioxidant activity. The total lipid content of cactus pear flower was about 2% with the neutral lipid amount, especially monoacylglycerols, being the highest. The lipids were rich in linoleic, oleic, and palmitic acids. The essential oil yield was 0.01% with the presence of 29 components in a complex mixture of monoterpenes. The main components of the essential oil were benzenacetatealdehyde, D-3-carene, hexanol, and α-pinene. Solvents with different polarities had significant effect on polyphenol contents and antioxidant activity of cactus pear flower. RP-HPLC analysis allowed characterization of 20 phenolic compounds mainly belonging to phenolic acid and flavonoid classes. Based on its phytochemical composition and antioxidant activity, cactus pear flower could be used in food, industrial, and biomedical applications. Practical applications The interest in naturally occurring antioxidants has recently increased for use in food, cosmetic, and pharmaceutical products to replace synthetic antioxidants, which are being restricted because of their carcinogenicity. The cactus pear flower constitutes one of the by-products in cactus industry and generally discarded by separation from the fruit. This study aimed to valorize cactus pear flower through its lipids, essential oils, phenolics, and antioxidant activity.

The effects of two extraction methods, used at three ripening stages on the total polyphenol contents and the antioxidant activities of Cumin (Cuminum cyminum L.) seed extracts were studied. The ripening stage effect on some physical properties of cumin seed was significant. The increase of dry matter (from 10.3 to 87.5 %) during ripeness was correlated negatively with that of moisture content (from 89.7 to 12.5 %). Besides results showed that the full ripe seeds were richer on polyphenols and condensed tannin than unripe ones, and consequently exhibited higher antioxidant activities. However, the unripe seeds had a higher total flavonoid content compared to those of half ripe and full ripe ones. The comparison of two extraction methods showed that soxhlet extracts contained the greatest amount of polyphenols and flavonoids, while maceration samples exhibited higher antiradical and bleaching power assay. Total phenolic contents and IC50 (concentration required to cause a 50 % DPPH inhibition) values in cumin seed during their maturation allowed to conclude that antioxidant activity does not depend only on the high content of total phenolics but also on the phenolic composition. A total of 19 phenolic compounds were successfully identified by HPLC analysis during the ripening of cumin seeds. Rosmarinic acid was the major phenolic acid for the unripe seeds. Furthermore, half ripe and full ripe seeds were dominated by p-coumaric acid. These compounds might be considered as interesting bioactive natural substances that may be used in several fields, such as nutraceuticals, cosmetics and agro-food industry.

Our results indicated that, in both varieties, moderate water deficit (MWD) improved the number of umbels per plant as well as the number of umbellets per umbel and the seed yield, in comparison to the control, but it decreased under severe water deficit (SWD). Besides, total phenolic contents were higher in the treated seeds and drought increased the level of total and individual polyphenols. This increase was appreciably more important in TCS than in ICS. Moreover, antioxidant activities of the extracts were determined by 4 different test systems, namely 2,2-diphenyl-1-picrylhydrazyl, β-carotene/linoleic acid chelating, and reducing power assays, and showed that treated seeds exhibited the highest activity, for both TCS and ICS.
