A quick diagnosis of the ecological quality of five sections of the Macacu-Guapiaçu hydrographic complex was performed using visual assessment protocol (VAP), conventional analysis, composition and structure of ciliates and the occurrence of benthic macro invertebrates. Water samples were collected from five sampling stations (A-E) in July of 2010. Protists were sampled with aid of artificial substrates, and macro invertebrates were sampled using the multi-habitat method. A total of 22 taxa of protozoa and 51 taxa of macro invertebrates was registered at the five stations. Reference station (A) showed greater protist diversity and higher values of BMWP-CETEC and IBE-IOC indices relating to macro invertebrates. The impacted stations (B and E) presented lower diversity and higher abundance of protozoa, as well as the lowest values for the rates. The four methods used were convergent to diagnose reference station (A), the impacted stations (B and E) and those in intermediate conditions (C and D). Thus, the methods were complementary, as the conventional parameters and ciliate assembly were more sensitive to detecting the increase in organic pollution and VAP and macro invertebrate assembly detected changes in geomorphology and habitat degradation.

Little is known about the role of protists and bacteria interactions during hydrocarbon biodegradation. This work focused on the effect of oil on protists from three different locations in Guanabara Bay and bacteria from Caulerpa racemosa (BCr), Dictyota menstrualis (BDm) and Laurencia obtusa (BLo) during a 96 h bioassay. Cryptomonadida (site 1, 2 and 3), Scuticociliatida (site 2) and Euplotes sp.1 and Euplotes sp.2 (site 3) appeared after incubation. The highest biomass observed in the controls was as follows: protist site 3 (6.0 µgC.cm–3, 96 h) compared to site 3 with oil (0.7 µgC.cm–3, 96 h); for bacteria, 8.6 µgC.cm–3(BDm, 72 h) and 17.0 µgC.cm–3(BCr with oil, 24 h). After treatment, the highest biomasses were as follows: protists at site 1 and BLo, 6.0 µgC.cm–3 (96 h), compared to site 1 and BLo with oil, 3.31 µgC.cm–3 (96 h); the bacterial biomass was 43.1 µgC.cm–3 at site 2 and BDm (96 h). At site 3 and BLo with oil, the biomass was 18.21 µgC.cm–3 (48 h). The highest biofilm proportions were observed from BCr 1.7 µm (96 h) and BLo with oil 1.8 µm (24 h). BCr, BLo and BDm enhanced biofilm size and reduced the capacity of protists to prey.

Species of Spirostomum Ehrenberg, 1838 are widely used as model organisms in ecological studies of environmental impacts and symbioses between ciliates and human pathogenic bacteria. However, the taxonomy of this genus is confused by the superficiality of the morphological descriptions of its included species, and the use of only a few characters for their differentiation. The present study provides details of total infraciliature, nuclear apparatus, morphometric data and 18S rDNA gene sequences of Spirostomum teres Claparède & Lachmann, 1858 and Spirostomum minus Roux, 1901, isolated from a sewage treatment plant and a freshwater lake in the city of Rio de Janeiro, Brazil, respectively. For the morphological descriptions of S. teres and S. minus, living cells were observed using bright-field and differential interference contrast (DIC) microscopy, the total infraciliature and nuclear apparatus were revealed by staining with protargol, and ciliary patterns were observed also with scanning electron microscopy (SEM). The complete sequences of the 18S rDNA of S. teres and S. minus were obtained using eukaryotic universal primers, and then compared with sequences of other species and populations of Spirostomum deposited in the GenBank database. Living S. minus measured 400-800 µm in length and 55-115 µm in width, with the following characteristics: adoral zone of membranelles approximately 112 µm long; inconspicuous paroral kinety; 30-40 kineties in somatic ciliature; moniliform macronucleus with 9-25 nodes, approximately 12 micronuclei; single and posterior contractile vacuole; and yellow-brown cytoplasm. Living and fully extended S. teres measured approximately 250 µm in length and 65 ìm in width, with the following characteristics: adoral zone of membranelles approximately 92 µm long; approximately 30 somatic kineties; compact macronucleus, approximately five micronuclei; macronuclear groove present; single and posterior contractile vacuole; and colorless cytoplasm. Evidence from 18S rDNA sequences confirms the identification of S. teres and suggests the existence of cryptic species closely related to S. minus. The use of silver impregnation technique (protargol) allowed the observation and description of a greater number of characters in S. minus and S. teres, thus assisting the research that require identification of these species.

During an ecological study of the epibiotic relationship between ciliate protists and Pomacea figulina (Spix, 1827) (Gastropoda, Ampullariidae), originating from an urban stream in southeast Brazil, a high infestation by the peritrich ciliate Carchesium polypinum (Linnaeus, 1758) Ehrenberg, 1830 (Ciliophora, Peritrichia) associated to the shell of one mollusc among 23 was observed. We provided a morphological and morphometric study of C. polypinum using observations of specimens in vivo, after protargol staining, and examined using scanning electron microscopy. The Brazilian-population of C. polypinum is characterized by: size of zooid in vivo 89 µm x 57 µm on average; colony regularly dichotomously branched with usually up to 40 zooids; macronucleus usually J-shaped; single contractile vacuole located in the upper third of body; myoneme not continuous throughout the colony; stalks contract despite the discontinuity of their individual myonemes; polykinety comprises three peniculi, each consisting of three kineties. The high infestation showed here could be related to the preference for eutrophic environments showed by C. polypinum and suggested that ciliate epibionts may be ecologically important in aquatic habitats.

In samples of raw sewage collected from a sewage treatment plant in Rio de Janeiro (ETE-Penha), we found populations of two species, Deviata estevesi Paiva & Silva-Neto, 2005 and Deviata brasiliensis sp. nov. The organisms were studied in vivo under phase contrast microscopy, differential interference contrast (DIC), and after protargol-impregnation. The population of D. estevesi exhibited more extensive variation in cirral pattern than previously described. The interphasic organisms of new species D. brasiliensis sp. nov. are distinguishable from their congeners based on a series of morphometric features: cirral row R3 usually presents 1-3 cirri behind the right frontal cirrus, on average there are four macronuclear nodules, and, during morphogenesis of cell division, primordium V of the proter originates from the anterior region of cirral row R5 instead of row R6, as in the type species D. abbrevescens Eigner, 1995. In D. estevesi, the ventral cirral rows replicate by within-row primordia, which develop independently for the proter and for the opisthe, suggesting that it belongs to or is closely related to Parastrongylidium, hence the combination P. estevesi comb. nov. is established.

ABSTRACT Physiological reorganization is described for the first time for the hypotrich ciliate genus Apoamphisiella Foissner, 1997, based on a population of A. vernalis (Stokes, 1887) Berger, 2006 collected from an eutrophic pond in the state of Minas Gerais, Brazil. Stomatogenesis is epiapokinetal, and the cirral pattern reorganizes from six typical fronto-ventral-transverse primordia, plus at least one short primordium located between IV and V. Primordia I and II originate from disaggregating undulating membranes and buccal cirrus, respectively; primordium III originates from left frontoventral cirrus plus a streak of basal bodies extending from the anterior end of oral primordium; primordium IV and the short extra primordia are formed from basal bodies perhaps associated to the posterior end of III plus the disaggregation of anteriormost cirri of the left ventral row. Primordia V and VI arise within the right ventral row as a single streak, splitting to form the two primordia. Marginal primordia develop initially within the pre-existent marginal rows, continuing to reorganize outside, pushing the old marginal cirri leftwards. Two dorsomarginal kineties develop associated to the right marginal row primordium. Dorsal ciliature also reorganizes from within the pre-existent dorsal kineties. The two macronuclear nodules approach each in middle reorganizers, briefly touching each other. At least one micronucleus undergo division. The process of physiological reorganization in A. vernalis resembles that of the North American Paraurostyla weissei complex representative, and along with features of the interphase morphology, indicate that Apoamphisiella belongs or is related to the Cyrtohymena-Paraurostyla group, within the Dorsomarginalia.

The water retention capacity of porous materials is crucial in various geotechnical and environmental engineering applications such as slope stability analysis, landfill management, and mining operations. Filtered tailings stacks are considered an alternative to traditional tailings dams. Nevertheless, the mechanical behaviour and stability of the material under different water content conditions are of concern because these stacks can reach considerable heights. The water behaviour in these structures is poorly understood, particularly the effects of the water content on the stability and potential for liquefaction of the stacks. This study aims to investigate the water retention and flow characteristics of compacted iron ore tailings in high columns to better understand their hydromechanical behaviour. The research used 5 m high columns filled with iron ore tailings from the Quadrilátero Ferrífero region in Minas Gerais, Brazil. The columns were prepared in layers, compacted, and instrumented with moisture content sensors and suction sensors to monitor the water movement during various stages of saturation, drainage, infiltration, and evaporation. The sensors provided consistent data and revealed that the tailings exhibited high drainage capacity. The moisture content and suction profiles were effectively established over time and revealed the dynamic water retention behaviour. The comparison of the data with the theoretical soil water retention curve (SWRC) demonstrated a good correlation which indicates that there was no hysteresis in the material response. The study concludes that the column setup effectively captures the water retention and flow characteristics of compacted tailings and provides valuable insights for the hydromechanical analysis of filtered tailings stacks. These findings can significantly help improve numerical models, calibrate material parameters, and contribute to the safer and more efficient management of tailings storage facilities.

ABSTRACTTrypanosoma cruzi is the etiologic agent of the most prevalent human parasitic disease in Latin America, Chagas disease. Its genome is rich in multigenic families that code for virulent antigens and are present in the rapidly evolving genomic compartment named Disruptive. DNA replication is a meticulous biological process in which flaws can generate mutations and changes in chromosomal and gene copy numbers. Here, integrating high-throughput and single-molecule analyses, we were able to identify Predominant, Flexible, and Dormant Orc1Cdc6-dependent origins as well as Orc1Cdc6-independent origins. Orc1Cdc6-dependent origins were found in multigenic family loci, while independent origins were found in the Core compartment that contains conserved and hypothetical protein-coding genes, in addition to multigenic families. In addition, we found that Orc1Cdc6 density is related to the firing of origins and that Orc1Cdc6-binding sites within fired origins are depleted of a specific class of nucleosomes that we previously categorized as dynamic. Together, these data suggest that Orc1Cdc6-dependent origins may contribute to the rapid evolution of the Disruptive compartment and, therefore, to the success of T. cruzi infection and that the local epigenome landscape is also involved in this process.IMPORTANCETrypanosoma cruzi, responsible for Chagas disease, affects millions globally, particularly in Latin America. Lack of vaccine or treatment underscores the need for research. Parasite’s genome, with virulent antigen-coding multigenic families, resides in the rapidly evolving Disruptive compartment. Study sheds light on the parasite’s dynamic DNA replication, discussing the evolution of the Disruptive compartment. Therefore, the findings represent a significant stride in comprehending T. cruzi’s biology and the molecular bases that contribute to the success of infection caused by this parasite.

Skeletal muscle regeneration after injury is a complex process involving inflammatory signaling and myoblast activation. Pro-inflammatory cytokines like tumor necrosis factor-alpha (TNF-α) are key mediators, but their effects on gene expression in proliferating myoblasts are unclear. We performed the RNA sequencing of TNF-α treated C2C12 myoblasts to elucidate the signaling pathways and gene networks regulated by TNF-α during myoblast proliferation. The TNF-α (10 ng/mL) treatment of C2C12 cells led to 958 differentially expressed genes compared to the controls. Pathway analysis revealed significant regulation of TNF-α signaling, along with the chemokine and IL-17 pathways. Key upregulated genes included cytokines (e.g., IL-6), chemokines (e.g., CCL7), and matrix metalloproteinases (MMPs). TNF-α increased myogenic factor 5 (Myf5) but decreased MyoD protein levels and stimulated the release of MMP-9, MMP-10, and MMP-13. TNF-α also upregulates versican and myostatin mRNA. Overall, our study demonstrates the TNF-α modulation of distinct gene expression patterns and signaling pathways that likely contribute to enhanced myoblast proliferation while suppressing premature differentiation after muscle injury. Elucidating the mechanisms involved in skeletal muscle regeneration can aid in the development of regeneration-enhancing therapeutics.

Citrus orchards in northeastern Brazil are mostly rainfed and comprised basically of ‘Pera CNPMF D-6’ sweet orange budded on ‘Rangpur’ lime, for the drought tolerance and productivity imparted by this rootstock. Therefore, the selection of new varieties is needed to broaden the genetic basis of citrus cultivated in this region. Accordingly, this study compared vegetative, productive, and fruit quality traits of eight sweet orange scions grafted on ‘Rangpur’ lime over eleven years under the tropical rainfed conditions of northeastern Brazil. ‘Kona’ trees excelled in yield performance associated with bulk canopy, precocity, sweet fruit with intermediate acidity, and high vitamin C contents in spite of proneness to alternate yields and low ratio (maturity index). ‘Valencia Montemorelos’ and ‘Rubi’ trees, in turn, had high yield performances coupled with intermediate canopies, sweet fruit, intermediate acidity (‘Rubi’) and vitamin C contents, low propensity for yield fluctuation (‘Valencia Montemorelos’), and high precocity (‘Rubi’), albeit low ratio. Overall, our results emphasize ‘Kona,’ ‘Valencia Montemorelos,’ and ‘Rubi’ as superior sweet orange varieties for diversification of tropical rainfed orchards for their outstanding yield performance and good fruit quality.

An ever-growing collection of commercial biostimulants is becoming available in a wide variety of forms and compositions to improve crop performance. Given the intricate nature of deciphering the underlying mechanisms of commercial products, which typically comprise various biological components, it is crucial for research in this area to have robust tools to demonstrate their effectiveness in field trials. Here, we took a multi-attribute approach to evaluating the impact of biostimulants on crop performance. First, we assessed the impact of a biostimulant on the soil and rhizosphere microbiomes associated to crops in eight reference farms, including corn (3 farms), soybean (2), cotton (2) and sugarcane (1), in different biomes and production contexts in Brazil and Paraguay. Second, we modeled a set of integrated indicators to measure crop responses to biostimulant application, including five analytical themes as follows: i) crop development and production (9 indicators), ii) soil chemistry (9), iii) soil physics (5), iv) soil biology (6) and v) plant health (10). Amplicon 16S rRNA and ITS sequencing revealed that the use of the biostimulant consistently changes the structure of bacterial and fungal communities associated with the production system for all evaluated crops. In the rhizosphere samples, the most responsive bacterial taxa to biostimulant application were Prevotella in cotton; Prauserella and Methylovirgula in corn; and Methylocapsa in sugar cane. The most responsive fungal taxa to biostimulant use were Arachnomyces in soybean and cotton; and Rhizophlyctis in corn. The proposed integrated indicators yielded highly favorable positive impact indices (averaging at 0.80), indicating that biostimulant-treated fields correlate with better plant development and crop performance. Prominent indices were observed for indicators in four themes: soil biology (average index 0.84), crop production (0.81), soil physics (compaction reduction 0.81), and chemical fertility (0.75). The multi-attribute approach employed in this study offers an effective strategy for assessing the efficacy of biostimulant products across a wide range of crops and production systems.

Abstract: The morphology of Blepharisma sinuosum resting cysts and the dynamics of pigmentation at different stages of encystment are presented for the first time. Cyst morphometrics are similar to other Blepharisma species, with three‐wall layers, bacteria surrounding the ectocyst, a conical plug, and wrinkly surface toward the plug in mature stages. The vegetative moniliform macronucleus changes to a horseshoe shape, and at early stages, the cystic cytoplasm is homogeneously pigmented, comprising a contractile vacuole; later, pigments polarize toward the plug, decorate the cortical layer, and become brownish. Objective: To evaluate the applicability of transcutaneous electrical nerve stimulation (TENS) as a complementary treatment method to non-opioid analgesics for acute postoperative pain in patients undergoing surgical treatment due to ankle fractures. Methods: A prospective, randomized, analytical, cross-sectional study performed after ankle fracture surgery. Two groups were randomized as follows: group 1 (Intervention) received TENS and non-opioid analgesic (dipyrone), and group 2 (Control) received non-opioid analgesic (dipyrone). Results: This study demonstrated that TENS in patients undergoing surgical treatment of ankle fractures reduces the use of rescue opioids significantly to control postoperative pain. Conclusion: TENS devices may be another safe option to control postoperative pain and reduce the use of opioids, avoiding adverse effects from this class of analgesic.
