Les membranes plasmàtiques protegeixen la cèl·lula d’invasions exògenes, fet que dificulta l’arribada de certs fàrmacs a les seves dianes terapèutiques intracel·lulars. En els últims anys, s’han desenvolupat diferents pèptids sintètics per internalitzar diferents molècules bioactives. Aquest article tracta de la síntesi en fase sòlida de dos pèptids tetradecamèrics híbrids mitjançant l’estratègia Fmoc/Boc. Aquests pèptids estan constituïts per un g-aminoàcid ciclobutànic i un residu de trans-g-amino-l-prolina funcionalitzat, enllaçats alternadament. Alguns d’aquests pèptids han donat bons resultats d’internalització en cèl·lules HeLa i en el paràsit leishmània, i es preveu realitzar un estudi en profunditat de les seves propietats de penetració., Plasma membranes protect the cell from exogenous invasions and this may impede the arrival of some drugs to their intracellular therapeutic target. In recent years, various synthetic cell-penetrating peptides have been developed which permit the internalization of different bioactive molecules. This paper deals with the solid-phase synthesis of two hybrid tetradecameric peptides using the Fmoc/Boc strategy. These peptides consist of a g-cyclobutane amino acid that is alternately linked with a functionalized residue of trans-g-amino-l-proline. Some of these peptides have provided good results in internalizing HeLa cells and Leishmania parasites, and their internalization properties will continue to be studied.

En aquesta tesi, diferents sistemes que contenen una unitat ciclobutànica substituïda en posició  o han estat sintetitzats i estudiats com a foldamers, agents de penetració cel·lular i agents de contrast. Tot i no tenir estructures directament relaciones entre elles, les mateixes estratègies sintètiques i tècniques han estat utilitzades per a preparar els diferents compostos. La present tesi consisteix en una introducció general que inclou alguns precedents dels nostre grup de recerca seguit dels objectius generals (Capítol 2). Els resultats obtinguts durant la elaboració de la tesi estan distribuïts en tres capítols diferents (Capítol 3-5), que al mateix temps estan dividits en diferents seccions. Les conclusions generals estan recollides al Capítol 6 i la metodologia experimental es pot trobar al Capítol 7. ♣Capítol 3. Sis famílies de pèptids, amb diferent estereoquímica, constituïts a partir d'un aminoàcid que conté un anell ciclobutànic en combinació amb la -amino-L-prolina han estat sintetitzats. L'estudi de la seva habilitat de plegament s'ha realitzat utilitzant tècniques de RMN d'alta resolució per tal de detectar la formació d'enllaços d'hidrogen que poden conduir a estructures secundaries. La informació obtinguda en aquest capítol va ser molt útil per a dissenyar nous pèptids de penetració cel·lular. ♣Capítol 4. Diferents pèptids han estat sintetitzats mitjançant tècniques de fase sòlida i han estat estudiats com a potencials agents de penetració en dos sistemes biològics: cèl·lules HeLa i paràsits de Leishmania. Els millors pèptids de penetració cel·lular han estat conjugats amb un fàrmac per tal d'estudiat el seu efecte en paràsits de Leishmania. ♣Capítol 5. Un nou lligand que conté una diamina basada en l'anell ciclobutànic ha estat sintetitzat i complexat amb diferents ions metàl·lics. L'estabilitat termodinàmica i el número d'hidratació d'aquests complexos ha estat estudiada mitjançant diferents tècniques. El seu potencial com a nou agent de contr, In this thesis, different systems containing a - or -substituted cyclobutane scaffold have been synthesized and studied as foldamers, as cell penetrating peptides (CPP) or as contrast agents. Despite having different structures not directly related between them, the same synthetic strategies and techniques have been used to prepare the different compounds. The thesis has been structured with a general introduction with some precedents from our research group, followed by the general objectives (Chapter 2). The results of the present thesis are presented in three different chapters (Chapters 3-5), which are divided in different sections. General conclusions are reported in Chapter 6 and the experimental methodologies can be found in Chapter 7. ♣ Chapter 3. Six families of peptides with different stereochemistry built with a cyclobutane containing amino acid (CBAA) in combination with -amino-L-proline were synthesized. The study of their folding ability was carried out by using high resolution NMR techniques in order to detect the formation of hydrogen bonds that could induce secondary structures. The information obtained in this chapter was very useful to design new cell penetrating peptides. ♣ Chapter 4. Different new peptides were synthesized by solid phase synthesis techniques and studied as potential penetrating agents in two biological systems: HeLa cells and Leishmania parasites. The best cell penetrating peptides were conjugated with a drug to study their effect in Leishmania parasites. ♣ Chapter 5. A new linear ligand containing a cyclobutane-based diamine was synthesized and complexed with different metal ions. The thermodynamic stability and the hydration number of these complexes were studied by means of different techniques. Their potential as new contrast agents for magnetic resonance imaging (MRI) was studied by different NMR methodologies.

New enantiomerically pure C16-alkyl diamides derived from trihydroxy cyclohexane-1,2-dicarboxylic acid have been synthesized from (−)-shikimic acid. The hydroxyl groups in these compounds are free or, alternatively, they present full or partial protection. Their gelling abilities towards several solvents have been tested and rationalized by means of the combined use of Hansen solubility parameters, scanning electron microscopy (SEM), and circular dichroism (CD), as well as computational calculations. All the results allowed us to account for the capability of each type of organogelator to interact with different solvents and for the main mode of aggregation. Thus, compounds with fully protected hydroxyl groups are good organogelators for methanol and ethanol. In contrast, a related compound bearing three free hydroxyl groups is insoluble in water and polar solvents including alcohols but it is able to gelate some low-polarity solvents. This last behavior can be justified by strong hydrogen bonding between molecules of organogelator, which competes advantageously with polar solvent interactions. As an intermediate case, an organogelator with two free hydroxyl groups presents an ambivalent ability to gelate both apolar and polar solvents by means of two aggregation patterns. These involve hydrogen bonding interactions of the unprotected hydroxyl groups in apolar solvents and intermolecular interactions between amide groups in polar ones

Els resultats d'aquesta tesi estan dividits en tres capítols. 1) Dos gelificadors basats en pèptids estudiats prèviament en el nostre grup de recerca han estat funcionalitzats amb un derivat de la terpiridina. La influencia d'aquesta nova funció en la habilitat gelificant ha estat investigada. Per a aquest estudi s'ha utilitzat el test de la inversió del tub, microscòpia electrònica, dicroisme circular i càlculs teòrics, suggerint que la agregació d'aquests nou compostos segueix una estructura helicoïdal. Finalment, els nous gelificadors s'han complexat amb alguns ions metàl·lics, però els complexos resultants no son capaços de gelificar cap dissolvent. Aquests resultats confirmen que el disseny racional de metal·logelificadors és un gran repte. 2) Diferents tripètids hídrids que contenen dos unitats de prolina i un aminoàcid ciclobutànic s'han sintetitzat i estudiat com a organocatalitzadors per a la reacció aldòlica. Notablement, la enantioselectivitat de la reacció aldòlica va ser invertida en la presència d'aigua. Els resultats s'han racionalitzat amb estudis conformacionals i mecanístics utilitzant RMN, dicroisme circular i càlculs teòrics. Els rendiments gairebé quantitatius i les bones enantioselectivitats aconseguides confereixen a aquests pèptids amb propietats interessants ha ser utilitzats en reaccions aldòliques i a ser estudiats més profundament en altres processos químics. 3) Dos nous lligands que contenen una diamina ciclobutànica s'han sintetitzat i complexat amb diferents ions metàl·lics paramagnètics. La estabilitat termodinàmica, la inertesa cinètica i el numero de hidratació d'aquests complexos s'ha investigat utilitzant diferent tècniques. Finalment, s'han estudiat com a potencials agents de contrast per a ressonància magnètica d'imatge utilitzant diferent metodologies de RMN. Globalment, e, In this thesis, different systems containing a cyclobutane-based scaffold were synthesized and studied as gelators, catalysts or contrast agents. Results of this thesis are divided in three chapters. 1) Two peptide-based low molecular weight gelators previously studied in our group were functionalized with a terpyridine derivative. The influence of this added moiety was determined in the final gelation behavior. This study was performed by tube inversion test, scanning electron microscopy, circular dichroism and theoretical calculations, suggesting that the aggregation of both compound followed a helical-like structure. These new gelators were complexed with some metal ions in order to obtain metallogelators, but the obtained complexes were not able to gelate any solvent, confirming that the rational design of metallogelators is still a big challenge. 2) Different new hybrid tripeptides containing two proline units and a cyclobutane-based amino acid were synthesized and studied as organocatalysts for aldol reactions. Noteworthy, the enantioselectivity in aldol reactions was reversed in the presence of water. Results were rationalized by conformational and mechanistic studies using NMR, circular dichroism and theoretical calculations. The almost quantitative yields and good enantioselectivities achieved under easy reaction conditions, confers these peptide catalysts with interesting properties to be employed in aldol reactions and to be further explored in other chemical processes. 3) Two new linear ligands containing a cyclobutane-based diamine were synthesized and complexed with different paramagnetic metal ions. The thermodynamic stability, kinetic inertness, and the hydration number of these complexes were investigated using different techniques. They were studied as potential contrast agents for magnetic resonance imaging (MRI) using different NMR methodologies. Overall, results suggested that one of them is a good and safe candidate to be used as contrast agent

La unidad estructural de anillo ciclobutánico es una importante plataforma que se encuentran en numerosos compuestos bioactivos y está presente en intermedios clave para la síntesis química de compuestos funcionalizados con interesantes propiedades fisicoquímicas y con posibles aplicaciones en los campos de la biomedicina y de los materiales. Los compuestos ciclobutánicos 1,2- y 1,3-difuncionalizados, además de presentar una alta versatilidad sintética, exhiben ciertas ventajas estructurales debido a la presencia del anillo de ciclobutano como un elemento de restricción conformacional y de dos centros quirales de configuración conocida. Haciendo uso de la versatilidad sintética de los compuestos ciclobutánicos 1,2- y 1,3-difuncionalizados y empleando el anillo de ciclobutano y su quiralidad, se prepararon tres grupos de compuestos ópticamente activos. ·Tensioactivos ciclobutánicos polifuncionales: Se prepararon cuatro nuevos compuestos anfifílicos quirales. Al mismo tiempo, se realizó el estudio fisicoquímico y la caracterización de las propiedades de superficie del tensioactivo zwiteriónico derivado de amida. Como resultado se consiguió un tensioactivo en forma de mezcla del derivado de amida y su respectiva sal de trifluoroacetato, el cual se comporta como un tensioactivo eficiente con agregación de tipo vesícula oligolamelar. · Triaminas ciclobutánicas conjugadas con Gd(III)-DOTA: Se llevó a cabo la síntesis estereoselectiva de cuatro complejos, los cuales presentan un núcleo ciclobutánico conjugado con Gd-DOTA y fueron estudiados como posibles Agentes de Contrastes (ACs) en Resonancia Magnética de Imagen (RMI). Los resultados de RMI y los cálculos teóricos sugieren que existe una amplia influencia de las sustituciones N-H y N-Me y de los grupos protectores de la función amina en la capacidad de contraste. El complejo bis(acetamida) presentó una relajatividad mayor que el compuesto de referencia DOTAREM, lo que lo convierte en un posible candidato a ser usado com, The structural unit of cyclobutane ring is an important platform present in several bioactive compounds and key intermediates for the chemical synthesis of funtionalized compounds with interesting physicochemical properties and possible applications in fields such as biomedicine and materials. The 1,2- and 1,3-difunctionalized compounds exhibit a high synthetic versatility and some structural advantages due to the presence of both the cyclobutane ring, as a conformational constraining element, and two chiral centers of known configuration. Because of the synthetic versatility of the 1,2- and 1,3-difunctionalized compounds and the chirality of the cyclobutane ring three groups of optically active compounds were prepared. • Polyfunctional cyclobutane surfactants: Four new chiral amphiphilic compounds were prepared. Then, a physicochemical study and the characterization of the surface properties of the amide derived zwiterionic surfactant were carried out. As a result, a surfactant as a mixture of the amide derivative and its corresponding trifluoroacetate salt was achieved. This mixture behaves as an efficient surfactant with oligolamellar vesicle type aggregation. • Triamine cyclobutane cores conjugated to Gd(III)-DOTA: A stereoselective synthesis of four compounds was carried out. They exhibit a cyclobutane core conjugated to Gd-DOTA and were studied as possible Contrast Agents (CAs) in Magnetic Resonance Imaging (MRI). The MRI results and the theoretical calculations suggest a wide influence of the N-H and N-Me substitutions and the protecting groups of the amine function in the contrast capacity.
