Oil fields harbour a wide variety of microorganisms with different metabolic capabilities. To examine the microbial ecology of petroleum reservoirs, a molecular-based approach was used to assess the composition of bacterial communities in produced water of Diyarbakir oil fields in Turkey. Denaturing gradient gel electrophoresis (DGGE) of polymerase chain reaction (PCR)-amplified 16S rRNA gene fragments was performed to characterise the bacterial community structure of produced water samples and to identify predominant community members after sequencing of separated DGGE bands. The majority of bacterial sequences retrieved from DGGE analysis of produced water samples belonged to unclassified bacteria (50%). Among the classified bacteria, Proteobacteria (29.2%), Firmicutes (8.3%), Bacteroidetes (8.3%) and Actinobacteria (4.2%) groups were identified. Pseudomonas was the dominant genus detected in the produced water samples. The results of this research provide, for the first time, insight into the complexity of microbial communities in the Diyarbakir oil reservoirs and their dominant constituents.

The purpose of this study was to compare the efficacy of the filtration method with direct method (without filtration) for detection and enumeration of either aerobic heterotrophic bacteria or anaerobic sulphate reducing bacteria (SRB). In addition we compared the results of colony morphology types of heterotrophic bacteria performed by filtration-spread plate experiments with spread plate experiments (i.e. plating on R2A agar plates). We detected that there was no statistically significant difference in the mean of SRB and heterotrophic bacteria counts from unfiltered and filtered samples. The results of the experiments comparing the cultivation and enumeration success of the filtered samples with unfiltered samples clearly demonstrate that filtration method enables the cultivation and enumeration of SRB and heterotrophic bacteria. In addition, the numbers of distinct colony types recovered from filtered samples were usually higher than unfiltered samples.

Sülfat indirgeyen bakteriler (SRB), mikrobiyolojik olarak desteklenen korozyona (MIC)yol açan en önemli bakteri grubu olarak düşünülmektedir. SRB'nin metabolikaktiviteleri sonucu oluşan hidrojen sülfür (H2S), çelik, paslanmaz çelik, vb. metallericiddi bir şekilde korozyona uğratmakta ve büyük ekonomik kayıplara yol açmaktadır.Galvanizli çelik sac, korozyona ve biyofaulinge karşı dirençli olması nedeni ile sudepoları ve soğutma kulelerinin yapımında tercih edilen bir malzemedir. Özellikle açıktip soğutma kuleleri, mikroorganizmaların gelişmesine olanak sağlayan uygun koşullarasahiptirler.Bu tezdeki hedefimiz, soğutma kulesine benzer şekilde, suyun sürekli sirküle olduğumodel bir su sisteminde, SRB'nin galvanizli çelik sac üzerinde oluşturduğu korozyonuve miktarını saptamak ve ayrıca, biyolojik parametrelerin mikrobiyolojik korozyonüzerine etkilerini belirlemektir.Deneyler, galvanizli çelik kuponları içeren model su sirkülasyon sisteminde ve kontroldüzeneğinde gerçekleştirilmiştir. Deney kuponları ayda bir kez olmak üzere model susirkülasyon sisteminden çıkartılmış ve her deney süresinin sonunda SRB, aerobik veanaerobik heterotrofik bakteri sayısı (HBS) saptanmış, korozyon hızı hesaplanmış,hücre dışı polimerik maddelerin karbonhidrat miktarı ölçülmüş ve taramalı elektronmikroskobunda (SEM) incelemeleri yapılmıştır.SEM analizleri, çinkonun (Zn) birçok mikroorganizmaya karşı toksik etkisi olmasınarağmen aerobik ve anaerobik heterotrofik bakteriler ile birlikte anaerobik SRB'ningalvanizli çelik yüzeylere tutunabildiklerini ve biyofilm tabakası oluşturabildiklerinigöstermiştir. Çalışmamızdaki korozyon deneyleri sonucunda, mikroorganizmalarıngalvanizli çeliği korozyona uğratabildiği saptanmıştır. Ayrıca biyofilmdeki SRB sayısıarttıkça, deney kuponlarındaki ağırlık kaybının da arttığı belirlenmiştir (P< 0.05). Ekolarak deney kuponlarındaki ağırlık kaybı ile biyofilmdeki karbonhidrat miktarıarasında aynı yönde anlamlı bir ilişkinin varlığı tespit edilmiştir (P< 0.05). Sulphate reducing bacteria (SRB) were considered as the major bacterial group involvedin microbially influenced corrosion (MIC). As a result of their ability to generatehydrogen sulfide (H2S), SRB cause corrosion of various metals including steels,stainless steels, etc. and severe economic damages.Galvanized steel is frequently used in the construction of cooling towers and watercontainers, owing to its good corrosion resistance and resistance to biofouling.Especially open cooling towers have suitable conditions for growing ofmicroorganisms.The aim of this study is to determine corrosion and its amount on galvanized steelcaused by SRB in a model recirculating water system which is similar to cooling towerand, also establish the effects of biological parameters on MIC.The experiments were carried out in a model recirculating water system and controlsystem with galvanized steel coupons. Assay coupons were removed from the modelrecirculating water system monthly for enumeration of SRB, aerobic and anaerobicheterotrophic bacteria, corrosion rate measurement, extracellular polysaccharidesubstances extraction, carbohydrate analysis and scanning electron microscope (SEM)observation.SEM analysis showed that SRB, aerobic and anaerobic heterotrophic bacteria attachedand formed a biofilm layer on the galvanized steel surfaces, even though zinc is toxic toa variety of microorganisms. Our results showed that microorganisms were responsiblefor the corrosion of galvanized steel. Also weight loss of galvanized steel was becorrelated with the number of sessile SRB cells (P< 0.05). In addition, there was acorrelation between weight loss of assay coupons and carbohydrate amount (P< 0.05). 93

ÖZET `Soğutma Kulelerinin Suyunda Sülfat İndirgeyen Bakterilerin Araştırılması' Bu çalışmada, İstanbul civarındaki otel ve iş merkezlerinden oluşan 30 farklı binanın soğutma kulesi sulan, sülfat indirgeyen bakteriler bakımından incelenmiştir. Sülfat indirgeyen bakteriler ortamda bulunan sülfat iyonlarım son elektron alıcısı olarak kullanarak hidrojen sülfür (H2S) oluşturmakta ve korozyona (aşınmaya) neden olmaktadırlar. Bu problem, soğutma kuleleri de dahil olmak üzere bir çok endüstriyel çevreyi olumsuz yönde etkilemektedir. Sülfat indirgeyen bakterilerin neden olduğu düşünülen korozyon, büyük ekonomik kayıplara yol açtığından son yıllarda bu bakterilerin soğutma kulesindeki işlevlerinin araştırılması oldukça önem kazanmıştır. Bu nedenle bakterinin soğutma kulelerindeki bulunma sıklığını ve ekolojisini öğrenmek amacıyla, 30 farklı binanın soğutma kulesinden 36 su örneği alınmıştır. İncelenen binaların 14'üne ait (% 46,6) soğutma kulesinden sülfat indirgeyen bakteri susu izole edilmiştir. En düşük sülfat indirgeyen bakteri sayısı mililitrede 10 iken, en yüksek değer mililitrede 104 bakteri olarak saptanmıştır. Sülfat indirgeyen bakterilerle benzer ekolojik istekleri olan ve Lejyoner hastalığına yol açan Legionella cinsi bakterilerin, bu bakterilerle aynı ortamda bulunabilecekleri düşünülmüştür. Bu nedenle, çalışma esnasında sülfat indirgeyen bakterilerin yam sıra Legionella cinsi bakterilerin analizi de yapılmıştır. Legionella cinsi bakteriler bakımından incelenen 25 binanın 10'una ait (% 40) soğutma kulesi suyundan Legionella pneumophila bakterisi izole edilmiş ve izolatlarm tümü serogurup 1 olarak tanımlanmıştır. En düşük Legionella pneumophila sayısı mililitrede 240.000 iken, en yüksek sayı mililitrede 109 bakteri olarak tespit edilmiştir. İncelenen soğutma kuleleri içinde sadece iki su örneğinde (% 8) aynı anda sülfat indirgeyen bakterilere ve Legionella pneumophila bakterisine rastlanmıştır. Örneklerin birinde sülfat indirgeyen bakteri sayısı mililitrede 10 iken, Legionella pneumophila sayısı mililitrede 5.048.000 bakteri olarak saptanmıştır. Diğer su örneğinde ise, elde edilen sülfat indirgeyen bakteri sayısı mililitrede yine 10 iken, Legionella pneumophila sayısı 109 gibi çok yüksek bir değerde bulunmuştur. Sülfat indirgeyen bakterilerin varlığı ile Legionella pneumophila sayısı arasında anlamlı bir ilişkinin olmadığı saptanmıştır.Bu çalışmada incelenen binaların soğutma kulesi suyu sıcaklıklarının 12°C ile 33°C arasında değiştiği gözlenmiştir. Yapılan istatistiksel çalışmada, soğutma kulesi suyunun sıcaklık değerleri ile sülfat indirgeyen bakteri sayısı arasında herhangi bir korelasyonun olmadığı saptanmıştır (P>0,05). Legionella pneumophila bakterisinin 12°C ile 28°C sıcaklık değerleri arasında ürediği görülmüştür. Bakterinin en yüksek sayıda izole edildiği su sıcaklık değerlerinin ise, 18 - 28°C arasında olduğu saptanmıştır. En düşük bakteri sayısmm izole edildiği su sıcaklık derecesi ise 22°C olarak tespit edilmiştir. Buna göre, soğutma kulesi suyunun sıcaklık değerleri ile Legionella pneumophila sayısı arasında anlamlı bir korelasyonun olmadığı saptanmıştır (P>0,05). İncelenen soğutma kulesi sularının pH'sının, 7,5 - 9,4 değerleri arasında değiştiği gözlenmiştir. Ayrıca soğutma kulelerinde hem sülfat indirgeyen bakterilerin hem de Legionella pneumophila baterisinin mevsimlere göre dağılımı incelenmiş ve her iki bakterinin bulunuşunun da mevsimlere bağlı olmadığı saptanmıştır. VI SUMMARY `Investigation of Sulfate Reducing Bacteria in Water Samples of Cooling Towers` In this study, water samples taken from cooling towers of 30 different buildings, such as hotels and business centres in Istanbul, were examined with regard to sulfate reducing bacteria. Sulfate reducing bacteria generate H2S (hydrogen sulfide) using sulfate ions in the environment as final electron acceptor and cause corrosion. This process affects most of the industrial environment especially the cooling towers. Recently, the investigation of hazards of these bacteria in cooling towers is getting gradually important due to corrosion which is thought to be caused by the sulfate reducing bacteria results in great economical losses. In this study, therefore, 36 water samples of cooling towers from 30 different buildings were collected to find out the presence, the abundance and the ecology of bacterium. Sulfate reducing bacteria were isolated from 14 cooling towers out of 30 (46.6 %) buildings. While the lowest concentration of sulfate reducing bacteria was 10 bacteria/ml, the highest concentration was determined as 104 bacteria/ml. It is considered that the bacteria belonging to the Genus Legionella can occur in the same environment with sulfate reducing bacteria due to the similar ecological needs. For this reason, apart from the sulfate reducing bacteria, also the analysis of the bacteria of Genus Legionella was made. 25 buildings were examined to find out the Genus Legionella. Legionella pneumophila was isolated from 10 cooling towers of 25 (40 %) buildings. All of the isolates were defined as serogroup 1. While the lowest concentration of Legionella pneumophila was 240.000 bacteria /ml, the highest concentration was 109 bacteria/ml. In cooling towers, the numerical values of bacteria were found higher than the permitted value (>1000/ml). Therefore, it can be concluded that Legionella pneumophila positive cooling towers have high level of Legionnaires' disease risk. Among the cooling towers examined, both sulfate reducing bacteria and Legionella pneumophila were isolated simultaneously from only two of the water samples. While the number of sulfate reducing bacteria detenriined in one of these samples was 10 bacteria/ml, the number of Legionella pneumophila was determined as 5.048.000 per millilitre. In the other sample, while the number of sulfate reducing bacteria was the same (10/ml), the number of Legionella pneumophila was higher such as 109 per millilitre. No relationship was found between the numbers of sulfate reducing bacteria and Legionella pneumophila. vnIn this study, the recorded temperature of water samples taken from cooling towers was between 12 - 33°C. However, there was no correlation between the water temperature and the number of sulfate reducing bacteria, statistically (P>0,05). It was found that Legionella pneumophila generated between the water temperatures of 12 - 28°C. The water temperature in which the highest number of Legionella pneumophila isolated was between 18 - 28°C, while the lowest number of Legionella pneumophila was isolated at 22°C. Thus, it was concluded that there was no correlation between the water temperature and the number of Legionella pneumophila (P>0,05). The pH of water samples was between 7.5 and 9.4. In addition, the distribution of both sulfate reducing bacteria and Legionella pneumophila throughout the seasons was observed and found that the presence of these bacteria in cooling towers didn't depend on the seasons. vm 39
