Intercalation of very long molecules into the structure of multi-layered graphene oxide (GO) was studied using example of 1-hexadecanol (C16), an alcohol molecule with 16 carbon atoms. Brodie graphite oxide (BGO) immersed in excess of liquid C16 just above the melting point shows expansion of c-unit cell parameter from ~6 Å to ~48.76 Å forming a structure with two densely packed layers of C16 molecules in a perpendicular orientation relative to the GO planes (α-phase). Heating of the BGO-C16 α-phase in excess of C16 melt results in reversible phase transition into β-phase at 336–342K. The β-phase shows much smaller unit cell parameter of 29.83 Å (363K). Analysis of data obtained using vacuum-driven evaporation of C16 from the β-phase provides evidence for structure of β-phase consisting of five layers of C16 molecules in parallel to GO plane orientation. Therefore, the transition from α-to β-phase corresponds to change in orientation C16 molecules from perpendicular to parallel relative to GO planes and decrease in the amount of intercalated solvent. Cooling of the β-phase in absence of C16 melt is found to result in the formation of γ-phase with inter-layer distance of ~26.5 Å corresponding to one layer of C16 molecules intercalated perpendicularly relative to the GO planes. Structures with one and two layers of C16 molecules parallel to GO planes were identified in samples with rather small initial loading of C16. Surprisingly rich variety of structures revealed in the BGO-C16 system provides opportunities to create materials with precisely controlled GO inter-layer distance. Swelling is a property of hydrophilic layered materials, which enables the penetration of polar solvents into an interlayer space with expansion of the lattice. Here we report an irreversible swelling transition, which occurs in MXenes immersed in excess dimethyl sulfoxide (DMSO) upon heating at 362-370 K with an increase in the interlayer distance by 4.2 Å. The temperature dependence of MXene Ti3C2Tx swelling in several polar solvents was studied using synchrotron radiation X-ray diffraction. MXenes immersed in excess DMSO showed a step-like increase in the interlayer distance from 17.73 Å at 280 K to 22.34 Å above ∼362 K. The phase transformation corresponds to a transition from the MXene structure with one intercalated DMSO layer into a two-layer solvate phase. The transformation is irreversible and the expanded phase remains after cooling back to room temperature. A similar phase transformation was observed also for MXene immersed in a 2 : 1 H2O : DMSO solvent ratio but at a lower temperature. The structure of MXene in the mixed solvent below 328 K was affected by the interstratification of differently hydrated (H2O)/solvated (DMSO) layers. Above the temperature of the transformation, the water was expelled from MXene interlayers and the formation of a pure two-layer DMSO-MXene phase was found. No changes in the swelling state were observed for MXenes immersed in DMSO or methanol at temperatures below ambient down to 173 K. Notably, MXenes do not swell in 1-alcohols larger than ethanol at ambient temperature. Changing the interlayer distance of MXenes by simple temperature cycling can be useful in membrane applications, e.g. when a larger interlayer distance is required for the penetration of ions and molecules into membranes. Swelling is also very important in electrode materials since it allows penetration of the electrolyte ions into the interlayers of the MXene structure.

In this study, faceted mesocrystals have been assembled from the dispersion of truncated cubic-shaped iron oxide nanoparticles stabilized by oleic acid (OA) molecules using the non-solvent “gas phase diffusion technique” into an organic solvent. The effects of synthesis conditions as well as of the nanoparticle size and shape on the structure and morphogenesis of mesocrystals were examined. The interactions of OA capped iron oxide nanoparticles with solvent molecules were probed by analytical ultracentrifugation and double difference pair distribution function analysis. It was shown that the structure of the organic shell significantly depends on the nature and polarity of solvent molecules.

Visible light driven hydrogen (H2) production from water is a promising strategy to convert and store solar energy as chemical energy. Covalent organic frameworks (COFs) are front runners among different classes of organic photocatalyst, owing to their tunable porosity, crystallinity, optical and electronic properties. Photocatalytic activity of COFs depends on numerous factors such as band gap, crystallinity, porosity, exciton migration, charge separation and transport, stability etc. However, it is challenging to fine tune all these factors simultaneously to enhance the photocatalytic activity. Hence, in this report, we have prioritized the key factors for efficient photocatalytic H2 production through structure–property–activity relationship combined with microwave spectroscopy and first–principles calculations. Careful molecular engineering allowed us to tune the light absorption (i.e. band gap), crystallinity, porosity, layer stacking and charge carrier generation and transport of a series of isoreticular COFs. We have assessed how these properties and the interplay between them impact photocatalytic activity of studied COFs. From the structure–property–activity relationship, we found that light absorption and charge carrier generation and transport are the prime factors, which influence the photocatalytic H2 production of COFs in much greater extent than other factors.

The paper calls attention to the most symmetric interpenetration patterns of honeycomb layers. To the best of my knowledge, such patterns remained unknown so far. In my contribution a rigorous derivation of such patterns is given that makes use of a new approach to interpenetrating nets. The results are presented in a broad context of structural chemistry and crystal engineering.

Blockage of the human ether-à-go-go related gene (hERG) channel can result in life-threatening ventricular tachyarrhythmia. In an in vitro screening of herbal materials for hERG blockers using an automated two-microelectrode voltage clamp assay on Xenopus oocytes, an alkaloid fraction of Nelumbo nucifera Gaertn. (lotus) leaves induced ∼50% of hERG current inhibition at 100 μg/mL. Chromatographic separation resulted in the isolation and identification of (-)-asimilobine, 1, nuciferine, 2, O-nornuciferine, 3, N-nornuciferine, 4, and liensinine, 5. In agreement with in silico predicted ligand-target interactions, 2, 3, and 4 revealed distinct in vitro hERG blockages measured in HEK293 cells with IC50 values of 2.89, 7.91, and 9.75 μM, respectively. Because lotus leaf dietary weight loss supplements are becoming increasingly popular, the identified hERG-blocking alkaloids were quantitated in five commercially available products. Results showed pronounced differences in the content of hERG-blocking alkaloids ranging up to 992 μg (2) in the daily recommended dose.

Summary Type 1 diabetes is characterized by the destruction of pancreatic β cells, and generating new insulin-producing cells from other cell types is a major aim of regenerative medicine. One promising approach is transdifferentiation of developmentally related pancreatic cell types, including glucagon-producing α cells. In a genetic model, loss of the master regulatory transcription factor Arx is sufficient to induce the conversion of α cells to functional β-like cells. Here, we identify artemisinins as small molecules that functionally repress Arx by causing its translocation to the cytoplasm. We show that the protein gephyrin is the mammalian target of these antimalarial drugs and that the mechanism of action of these molecules depends on the enhancement of GABAA receptor signaling. Inhibition of the human ether-a-go-go-related gene channel is the single most important risk factor leading to acquired long QT syndrome. Drug-induced QT prolongation can cause severe cardiac complications, including arrhythmia, and is thus a liability in drug development. Considering the importance of the human ether-a-go-go-related gene channel as an antitarget and the daily intake of plant-derived foods and herbal products, surprisingly few natural products have been tested for channel blocking properties. In an assessment of possible human ether-a-go-go-related gene liabilities, a selection of widely used herbal medicines and edible plants (vegetables, fruits, and spices) was screened by means of a functional two-microelectrode voltage-clamp assay with Xenopus oocytes. The human ether-a-go-go-related gene channel blocking activity of selected extracts was investigated with the aid of a high-performance liquid chromatography-based profiling approach, and attributed to tannins and alkaloids. Major European medicinal plants and frequently consumed food plants were found to have a low risk for human ether-a-go-go-related gene toxicity.

GABAA receptor subtypes and TRPV1 receptors were expressed in Xenopus laevis oocytes. Modulation of GABA-induced chloride currents (IGABA) by piperine and SCT-66 and activation of TRPV1 was studied using the two-microelectrode-voltage-clamp technique and fast perfusion. Their effects on explorative behaviour, thermoregulation and seizure threshold were analysed in mice. Piperine acted with similar potency on all GABAA receptor subtypes (EC50 range: 42.8 ± 7.6 μM (α2β2)–59.6 ± 12.3 μM (α3β2)). IGABA modulation by piperine did not require the presence of a γ2S-subunit, suggesting a binding site involving only α and β subunits. Blockade of rapid delayed rectifier current (IKr) can lead to cardiac arrhythmia, which is a major concern in drug discovery and development. Drug safety tests involve the voltage clamp measurements on HEK cells expressing hERG potassium channels (Kv11.1) and high affinity hERG channel inhibition is widely accepted predictor of Torsade de Point arrhythmias (TdP). There is, however, evidence that multiple ion channel block prevent EADs and TdP arrhythmias. To address this question we estimated the IC50 values of the established hERG channel inhibitor dofetilide and 13 derivatives using a planar patch clamp system and HEK cells expressing hERG (Kv11.1) channels. Additionally we examined action potential prolongation in human induced pluripotent stem cell derived cardiomyocytes (hiPSC-CMs) in response to dofetilide and its derivatives. The arrhythmogenic potency was assessed by estimating the concentration required to prolong the action potential (AP) to 150% of control duration. In 11 out of 14 cases hERG inhibition was accompanied by a prolongation of AP the concentration of drugs inducing 150% increase of the AP duration correlated well with IC50 of hERG channels block (r = 0.94, p 200 µM). We conclude that the simultaneous block of depolarizing L-type calcium currents or cardiac sodium current offsets the effect of hERG block. Experimental data are supported by simulations of drug-induced changes in hiPSC-CMs action potentials. Our data support the new cardiac safety paradigm (CiPA) that discrimination between safe and unsafe drugs should address hERG inhibition and additionally their action on inward currents.

EtOAc extracts from two batches of Morus alba root bark (Sang bai pi) potentiated γ-aminobutyric acid (GABA)-induced chloride influx in Xenopus oocytes, which transiently expressed GABA (A) receptors of the subunit composition α₁β₂γ(2S). With the aid of HPLC-based activity profiling of the extract from the first batch, activity was traced to a peak subsequently identified as sanggenon G (3). The second batch had a different phytochemical profile, and HPLC-based activity profiling led to the identification of sanggenon C (4) and a stereoisomer of sanggenon D (2) as positive GABA (A) receptor modulators. The structurally related compound kuwanon L (1) was inactive. The sanggenons represent a new scaffold of positive GABA (A) receptor modulators.

The gamma-amino butyric acid (GABA) type A (GABA(A)) receptor represents a crucial target for clinical agents in the treatment of anxiety and insomnia. Using the two-microelectrode voltage clamp technique on recombinant α₁β₂γ(2S) GABA (A) receptors, effusol (1) and dehydroeffusol (2) were isolated in a bioactivity-guided approach from the pith of Juncus effusus L. Both compounds concentration-dependently enhanced GABA induced chloride currents (I(GABA)) by a maximum 188 ± 20 (1) and 239 ± 18 % (2), independent of the benzodiazepine (BZ) binding site. This activity on the GABA (A) receptor may explain the traditional use of J. effusus as a sedative and anxiolytic agent in Chinese medicine.

The coumarins imperatorin and osthole are known to exert anticonvulsant activity. We have therefore analyzed the modulation of GABA-induced chloride currents (I(GABA)) by a selection of 18 coumarin derivatives on recombinant α(1)β(2)γ(2S) GABA(A) receptors expressed in Xenopus laevis oocytes by means of the two-microelectrode voltage clamp technique. Osthole (EC(50)=14 ± 1 μM) and oxypeucedanin (EC(50)=25 ± 8 μM) displayed the highest efficiency with I(GABA) potentiation of 116 ± 4 % and 547 ± 56 %, respectively. I(GABA) enhancement by osthole and oxypeucedanin was not inhibited by flumazenil (1 μM) indicating an interaction with a binding site distinct from the benzodiazepine binding site. In general, prenyl residues are essential for the positive modulatory activity, while longer side chains or bulkier residues (e.g. geranyl residues) diminish I(GABA) modulation. Generation of a binary classification tree revealed the importance of polarisability, which is sufficient to distinguish actives from inactives. A 4-point pharmacophore model based on oxypeucedanin - comprising three hydrophobic and one aromatic feature - identified 6 out of 7 actives as hits. In summary, (oxy-)prenylated coumarin derivatives from natural origin represent new GABA(A) receptor modulators.

In a two-microelectrode voltage clamp with Xenopus laevis oocytes, a petroleum ether extract of Acorus calamus rhizomes enhanced the GABA-induced chloride current through GABA(A) receptors of the α₁β₂γ(2S) subtype by 277% ± 9.7% (100 μg/mL). β-Asarone (1), (+)-dioxosarcoguaiacol (2), (+)-shyobunone (3), and (+)-preisocalamenediol (4) were subsequently identified as main active principles through HPLC-based activity profiling and targeted isolation. The compounds induced maximum potentiation of the chloride current ranging from 588% ± 126% (EC₅₀: 65.3 ± 21.6 μM) (2) to 1200% ± 163% (EC(50): 171.5 ± 34.6 μM) (1), whereas (-)-isoshyobunone (5) and (-)-acorenone (6) exhibited weak GABA(A) modulating properties (5: 164% ± 42.9%; EC₅₀: 109.4 ± 46.6 μM and 6: 241% ± 23.1%; EC₅₀: 34.0 ± 6.7 μM). The relative configuration of 2 was established as 4R*8S*10R* by NOESY experiments and conformational analysis.

An EtOAc extract from the roots of Sophora flavescens (Kushen) potentiated γ-aminobutyric acid (GABA)-induced chloride influx in Xenopus oocytes transiently expressing GABA(A) receptors with subunit composition, α (1) β (2) γ (2S). HPLC-based activity profiling of the extract led to the identification of 8-lavandulyl flavonoids, kushenol I, sophoraflavanone G, (-)-kurarinone, and kuraridine as GABA(A) receptor modulators. In addition, a series of inactive structurally related flavonoids were characterized. Among these, kushenol Y (4) was identified as a new natural product. The 8-lavandulyl flavonoids are first representatives of a novel scaffold for the target.

An ethyl acetate extract of Biota orientalis leaves potentiated GABA-induced control current by 92.6% ± 22.5% when tested at 100 μg/mL in Xenopus laevis oocytes expressing GABA(A) receptors (α₁β₂γ(2S) subtype) in two-microelectrode voltage clamp measurements. HPLC-based activity profiling was used to identify isopimaric acid (4) and sandaracopimaric acid (5) as the compounds largely responsible for the activity. Sandaracopimaradienolal (3) was characterized as a new natural product. Compounds 4 and 5 were investigated for GABA(A) receptor subtype selectivity at the subtypes α₁β₁γ(2S), α₁β₂γ(2S), α₁β₃γ(2S), α₂β₂γ(2S), α₃β₂γ(2S), and α₅β₂γ(2S). Sandaracopimaric acid (5) was significantly more potent than isopimaric acid (4) at the GABA(A) receptor subtypes α₁β₁γ(2S), α₂β₂γ(2S), and α₅β₂γ(2S) (EC₅₀4: 289.5 ± 82.0, 364.8 ± 85.0, and 317.0 ± 83.7 μM vs EC₅₀5: 48.1 ± 13.4, 31.2 ± 4.8, and 40.7 ± 14.7 μM). The highest efficiency was reached by 4 and 5 on α₂- and α₃-containing receptor subtypes. In the open field test, ip administration of 5 induced a dose-dependent decrease of locomotor activity in a range of 3 to 30 mg/kg body weight in mice. No significant anxiolytic-like activity was observed in doses between 1 and 30 mg/kg body weight in mice.

A petroleum ether extract of Kadsura longipedunculata enhanced the GABA-induced chloride current (I(GABA)) by 122.5±0.3% (n=2) when tested at 100 μg/ml in Xenopuslaevis oocytes expressing GABA A receptors (α(1)β(2)γ(2S) subtype) in two-microelectrode voltage clamp measurements. Thirteen compounds were subsequently identified by HPLC-based activity profiling as responsible for GABA A receptor activity and purified in preparative scale. 6-Cinnamoyl-6,7-dihydro-7-myrceneol and 5,6-dihydrocuparenic acid were thereby isolated for the first time. The determination of the absolute stereochemistry of these compounds was achieved by comparison of experimental and calculated ECD spectra. All but one of the 13 isolated compounds from K. longipedunculata potentiated I(GABA) through GABA A receptors composed of α(1)β(2)γ(2S) subunits in a concentration-dependent manner. Potencies ranged from 12.8±3.1 to 135.6±85.7 μM, and efficiencies ranged from 129.7±36.8% to 885.8±291.2%. The phytochemical profiles of petroleum ether extracts of Kadsura japonica fruits (114.1±2.6% potentiation of I(GABA) at 100 μg/ml, n=2), and Schisandra chinensis fruits (inactive at 100 μg/ml) were compared by HPLC-PDA-ESIMS with that of K. longipedunculata.

A plant extract library was screened for GABA(A) receptor activity making use of a two-microelectrode voltage clamp assay on Xenopus laevis oocytes. An ethyl acetate extract of black pepper fruits [Piper nigrum L. (Piperaceae) 100 microg/mL] potentiated GABA-induced chloride currents through GABA(A) receptors (composed of alpha(1), beta(2), and gamma(2S) subunits) by 169.1 +/- 2.4%. With the aid of an HPLC-based activity profiling approach, piperine (5) was identified as the main active compound, together with 12 structurally related less active or inactive piperamides (1-4, 6-13). Identification was achieved by on-line high-resolution mass spectrometry and off-line microprobe 1D and 2D NMR spectroscopy, using only milligram amounts of extract. Compound 5 induced a maximum potentiation of the chloride currents by 301.9 +/- 26.5% with an EC(50) of 52.4 +/- 9.4 microM. A comparison of the modulatory activity of 5 and other naturally occurring piperamides enabled insights into structural features critical for GABA(A) receptor modulation. The stimulation of chloride currents through GABA(A) receptors by compound 5 was not antagonized by flumazenil (10 microM). These data show that piperine (5) represents a new scaffold of positive allosteric GABA(A) receptor modulators targeting a benzodiazepine-independent binding site.

Background and purpose: Heterologous expression of a1, b2 and g2S(g1) subunits produces a mixed population of GABAA receptors containing a1b 2o ra1b2g2S(g1) subunits. GABA sensitivity (lower in receptors containing g 1o rg2S subunits) and the potentiation of GABA-activated chloride currents (IGABA) by benzodiazepines (BZDs) are dependent on g2S(g1) incorporation. A variable g subunit incorporation may affect the estimation of IGABA potentiation by BZDs. We propose an approach for estimation of BZD efficiency that accounts for mixed population of a1b2 and a1b2g2S(g1) receptors. Experimental approach: We investigated the relation between GABA sensitivity (EC50) and BZD modulation by analysing triazolam-, clotiazepam- and midazolam-induced potentiation of IGABA in Xenopus oocytes under two-microelectrode voltage clamp.

An approach for rapid HPLC-based profiling for new GABA (A) ligands of natural origin has been developed. Active extracts are separated by a single injection of 3-10 mg of extract onto a semi-preparative (150 x 10 mm i. d.) HPLC column with gradient elution and time-based fractionation. The microfractions are tested in an automated two-microelectrode voltage-clamp assay on Xenopus oocytes expressing recombinant GABA (A) channels composed of alpha (1), beta (2) and gamma (2S) subunits. The protocol has been validated by spiking experiments with inactive extract and the GABA (A) receptor ligand magnolol, and by profiling of active extracts such as valerian extract containing the known GABA (A) receptor ligand valerenic acid. For dereplication of GABA containing extracts, we established a rapid and simple procedure by which GABA is analyzed as OPA derivative by reversed-phase HPLC. This dereplication protocol was validated with plant and fungal extracts which had been previously tested active or inactive in the oocyte assay and with spiking experiments.

Valeriana Officinalis L . is a traditionally used sleep remedy, however, the mechanism of action and the substances responsible for its sedative and sleep-enhancing properties are not fully understood. As we previously identified valerenic acid as a subunit-specific allosteric modulator of GABAA receptors, we now investigated the relation between modulation of GABAA receptors by Valerian extracts of different polarity and the content of sesquiterpenic acids (valerenic acid, acetoxyvalerenic acid). All extracts were analysed by HPLC concerning the content of sesquiterpenic acids. GABAA receptors composed of alpha 1, beta 2 and gamma 2S subunits were expressed in Xenopus laevis oocytes and the modulation of chloride currents through GABAA receptors (IGABA) by Valerian extracts was investigated using the two-microelectrode voltage clamp technique. Apolar extracts induced a significant enhancement of IGABA, whereas polar extracts showed no effect. These results were confirmed by fractionating a highly active ethyl acetate extract: again fractions with high contents of valerenic acid exhibited strong receptor activation. In addition, removal of sesquiterpenic acids from the ethyl acetate extract led to a loss of I (GABA) enhancement. In conclusion, our data show that the extent of GABAA receptor modulation by Valerian extracts is related to the content of valerenic acid.

In traditional Asian medicinal systems, preparations of the root and stem bark of Magnolia species are widely used to treat anxiety and other nervous disturbances. The biphenyl-type neolignan honokiol together with its isomer magnolol are the main constituents of Magnolia bark extracts. We have previously identified a nitrogen-containing honokiol derivative (3-acetylamino-4'-O-methylhonokiol, AMH) as a high efficient modulator of GABAA receptors. Here we further elucidate the structure-activity relation of a series of nitrogenated biphenyl-neolignan derivatives by analysing allosteric modulation and agonistic effects on α1β2γ2S GABAA receptors. The strongest IGABA enhancement was induced by compound 5 (3-acetamido-4'-ethoxy-3',5-dipropylbiphenyl-2-ol, Emax: 123.4±9.4% of IGABA-max) and 6 (5'-amino-2-ethoxy-3',5-dipropylbiphenyl-4'-ol, Emax: 117.7±13.5% of IGABA-max). Compound 5 displayed, however, a significantly higher potency (EC50=1.8±1.1 μM) than compound 6 (EC50=20.4±4.3 μM). Honokiol, AMH and four of the derivatives induced significant inward currents in the absence of GABA. Strong partial agonists were honokiol (inducing 78±6% of IGABA-max), AMH (63±6%), 5'-amino-2-O-methylhonokiol (1) (59±1%) and 2-methoxy-5'-nitro-3',5-dipropylbiphenyl-4'-ol (3) (52±1%). 3-N-Acetylamino-4'-ethoxy-3',5-dipropyl-biphenyl-4'-ol (5) and 3-amino-4'-ethoxy-3',5-dipropyl-biphenyl-4'-ol (7) were less efficacious but even more potent (5: EC50=6.9±1.0 μM; 7: EC50=33.2±5.1 μM) than the full agonist GABA.

Here we report on technology developments implemented into the Graphene Flagship European project for the integration of graphene and graphene-related materials (GRMs) into energy application devices. Many of the technologies investigated so far aim at producing composite materials associating graphene or GRMs with either metal or semiconducting nanocrystals or other carbon nanostructures (e.g., CNT, graphite). These composites can be used favourably as hydrogen storage materials or solar cell absorbers. They can also provide better performing electrodes for fuel cells, batteries, or supercapacitors. For photovoltaic (PV) electrodes, where thin layers and interface engineering are required, surface technologies are preferred. We are using conventional vacuum processes to integrate graphene as well as radically new approaches based on laser irradiation strategies. For each application, the potential of implemented technologies is then presented on the basis of selected experimental and modelling results. It is shown in particular how some of these technologies can maximize the benefit taken from GRM integration. The technical challenges still to be addressed are highlighted and perspectives derived from the running works emphasized.

GABA(A) receptors composed of alpha(1), beta(2), gamma(1) subunits are expressed in only a few areas of the brain and thus represent interesting drug targets. The pharmacological properties of this receptor subtype, however, are largely unknown. In the present study, we expressed alpha(1)beta(2)gamma(1)-GABA(A) receptors in Xenopus laevis oocytes and analyzed their modulation by 21 ligands from 12 structural classes making use of the two-microelectrode voltage-clamp method and a fast perfusion system. Modulation of GABA-induced chloride currents (I(GABA)) was studied at GABA concentrations eliciting 5 to 10% of the maximal response. Triazolam, clotiazepam, midazolam, 2-(4-methoxyphenyl)-2,3,5,6,7,8,9,10-octahydro-cyclohepta-(b)pyrazolo[4,3-d]pyridin-3-one (CGS 20625), 2-(4-chlorophenyl)-pyrazolo[4,3-c]quinolin-3-one (CGS 9896), diazepam, zolpidem, and bretazenil at 1 microM concentrations were able to significantly (>20%) enhance I(GABA) in alpha(1)beta(2)gamma(1) receptors. Methyl-6,7-dimethoxy-4-ethyl-beta-carboline-3-carboxylate, 3-methyl-6-[3-trifluoromethyl-phenyl]-1,2,4-triazolo[4,3-b]pyridazine (Cl 218,872), clobazam, flumazenil, 5-(6-ethyl-7-methoxy-5-methylimidazo[1,2-a]pyrimidin-2-yl)-3-methyl-[1,2,4]-oxadiazole (Ru 33203), 2-phenyl-4-(3-ethyl-piperidinyl)-quinoline (PK 9084), flurazepam, ethyl-7-methoxy-11,12,13,13a-tetrahydro-9-oxo-9H-imidazo[1,5-a]pyrrolo[2,1-c] [1,4]benzodiazepine-1-carboxylate (l-655,708), 2-(6-ethyl-7-methoxy-5-methylimidazo[1,2-a]pyrimidin-2-yl)-4-methyl-thiazole (Ru 33356), and 6-ethyl-7-methoxy-5-methylimidazo[1,2-a]pyrimidin-2-yl)phenylmethanone (Ru 32698) (1 microM each) had no significant effect, and flunitrazepam and 2-phenyl-4-(4-ethyl-piperidinyl)-quinoline (PK 8165) inhibited I(GABA). The most potent compounds triazolam, clotiazepam, midazolam, and CGS 20625 were investigated in more detail on alpha(1)beta(2)gamma(1) and alpha(1)beta(2)gamma(2S) receptors. The potency and efficiency of these compounds for modulating I(GABA) was smaller for alpha(1)beta(2)gamma(1) than for alpha(1)beta(2)gamma(2S) receptors, and their effects on alpha(1)beta(2)gamma(1) could not be blocked by flumazenil. CGS 20625 displayed the highest efficiency by enhancing at 100 microM I(GABA) (alpha(1)beta(2)gamma(2)) by 775 +/- 17% versus 526 +/- 14% I(GABA) (alpha(1)beta(2)gamma(1)) and 157 +/- 17% I(GABA) (alpha(1)beta(2)) (p < 0.05). These data provide new insight into the pharmacological properties of GABA(A) receptors containing gamma(1) subunits and may aid in the design of specific ligands for this receptor subtype.

The goal of this study was to design, experimentally validate, and apply a virtual screening workflow to identify novel hERG channel blockers. The hERG channel is an important antitarget in drug development since cardiotoxic risks remain as a major cause of attrition. A ligand-based pharmacophore model collection was developed and theoretically validated. The seven most complementary and suitable models were used for virtual screening of in-house and commercially available compound libraries. From the hit lists, 50 compounds were selected for experimental validation through bioactivity assessment using patch clamp techniques. Twenty compounds inhibited hERG channels expressed in HEK 293 cells with IC50 values ranging from 0.13 to 2.77 μM, attesting to the suitability of the models as cardiotoxicity prediction tools in a preclinical stage.

Inhibition of the cardiac human ether-a-go-go-related gene channel is a problematic off-target pharmacological activity and, hence, a major safety liability in clinical practice. Several non-cardiac drugs have been restricted in their use, or even removed from the market due to this potentially fatal adverse effect. Comparatively little is known about the human ether-a-go-go-related gene inhibitory potential of plant-derived compounds. In the course of an ongoing human ether-a-go-go-related gene in vitro study, a total of 32 structurally diverse alkaloids of plant origin as well as two semi-synthetically obtained protoberberine derivatives were screened by means of an automated Xenopus oocyte assay. Protopine, (+)-bulbocapnine, (+)-N-methyllaurotetanine, (+)-boldine, (+)-chelidonine, (+)-corynoline, reserpine, and yohimbine reduced the human ether-a-go-go-related gene current by ≥ 50 % at 100 µM, and were submitted to concentration-response experiments. Our data show that some widely occurring plant-derived alkaloids carry a potential risk for human ether-a-go-go-related gene toxicity.

Piperine activates TRPV1 (transient receptor potential vanilloid type 1 receptor) receptors and modulates γ-aminobutyric acid type A receptors (GABAAR). We have synthesized a library of 76 piperine analogues and analyzed their effects on GABAAR by means of a two-microelectrode voltage-clamp technique. GABAAR were expressed in Xenopus laevis oocytes. Structure–activity relationships (SARs) were established to identify structural elements essential for efficiency and potency. Efficiency of piperine derivatives was significantly increased by exchanging the piperidine moiety with either N,N-dipropyl, N,N-diisopropyl, N,N-dibutyl, p-methylpiperidine, or N,N-bis(trifluoroethyl) groups. Potency was enhanced by replacing the piperidine moiety by N,N-dibutyl, N,N-diisobutyl, or N,N-bistrifluoroethyl groups. Linker modifications did not substantially enhance the effect on GABAAR. Compound 23 [(2E,4E)-5-(1,3-benzodioxol-5-yl)-N,N-dipropyl-2,4-pentadienamide] induced the strongest modulation of GABAA (maximal GABA-induced chloride current modulation (IGABA-max = 1673% ± 146%, EC50 = 51.7 ± 9.5 μM), while 25 [(2E,4E)-5-(1,3-benzodioxol-5-yl)-N,N-dibutyl-2,4-pentadienamide] displayed the highest potency (EC50 = 13.8 ± 1.8 μM, IGABA-max = 760% ± 47%). Compound 23 induced significantly stronger anxiolysis in mice than piperine and thus may serve as a starting point for developing novel GABAAR modulators.

The most prominent determinant of acquired long QT syndrome is inhibition of the hERG potassium channel. Drug-induced QT prolongation can cause undesirable cardiac side effects and has led to several drug withdrawals in the past. In contrast to synthetic drugs, little is known about hERG channel inhibitors of natural origin. For assessing the risk of natural products on hERG channel inhibition, extracts obtained from frequently consumed spices, food, and medicinal plants were submitted to a broad in vitro screening. We established an HPLC-based activity profiling approach for this target by combining HPLC-microfractionation with on-line and off-line spectroscopy, and an automated two-microelectrode voltage clamp assay with transfected Xenopus laevis oocytes. Among the extracts tested, the methanolic extract of the TCM herbal drug Evodiae Fructus (Evodia rutaecarpa [Juss.] Benth., Rutaceae) reduced the peak tail hERG current by 60.9±6.9% at 100µg/mL. HPLC-based activity profiling of the crude extract led to the identification of dehydroevodiamine and hortiamine, two indoloquinazoline alkaloids, as the active principles. First information on structure activity relationships revealed that both the methyl group at position 14 and the double bond between C(13b) and N(14) are essential for the inhibitory effect of this compound class. We developed a method for removal of both inhibitory alkaloids by filtration over a cation exchange resin (Lewatit® MonoPlus SP 112). The resulting extract was devoid of hERG channel blocking activity. Moreover, we determined the dehydroevodiamine content in different commercial batches of Evodiae Fructus and in various processed TCM products. Several Chinese herbal medicines (CHMs) are used in the treatment of insomnia, restlessness, or anxiety. However, mechanisms underlying this effect and scientific proof for their traditional use is scarce. In the present study CHMs were screened for their ability to modulate GABA-induced chloride currents (I(GABA)), and active principles were isolated thus providing scientific evidence for their use as sedative and/or anxiolytic agents in CM. Herbal drugs were extracted successively with petroleum ether, ethyl acetate, methanol and water and further fractionated according to their bioactivity. The obtained extracts, fractions and finally pure compounds were tested for their ability to potentiate I(GABA) using the two-microelectrode voltage clamp technique on recombinant α₁β₂γ(2S) GABA(A) receptors expressed in Xenopus laevis oocytes. From all tested extracts the petroleum ether extract of Atractylodes macrocephala Koidz. rhizomes showed the strongest I(GABA) potentiation and was studied in more detail. This led to the isolation of the main components atractylenolide II and III, which seem to be responsible for the observed positive modulation of I(GABA) (166±12%, n=3 and 155±12%, n=3, respectively) in vitro. They were more active than the analogous compound atractylenolide I (96±3%, n=3) which differs in an additional double binding in position 9, 9a. Furthermore it could be shown that this effect is mediated independently of the benzodiazepine (BZ) binding site. In conclusion, A. macrocephala exerts its in vitro activity on recombinant GABA(A) receptors mainly through the two sesquiterpene lactones atractylenolide II and III (Fig. 1). This positive allosteric modulation of I(GABA) may partially be responsible for the traditional ethnopharmacological use of this herbal drug as a sedative.

Inhibition of the hERG channel delays repolarization and prolongs the QT interval and cardiac action potential which can lead to sudden death. Several drugs have been withdrawn from the market due to hERG channel inhibition. In the search of hERG channel inhibitors of natural origin, we established an HPLC-based profiling approach which combines HPLC-microfractionation and bioactivity testing on Xenopus laevis oocytes. The methanolic extract of the TCM herbal drug Coptidis rhizoma (Coptis chinensis Franch., Ranunculaceae) reduced the peak tail hERG current by 31.7 ± 2.0% at 100 µg/mL. HPLC-based activity profiling pointed towards berberine as the active constituent. However, hERG inhibition by 100 µM of a reference sample of berberine (16.3 ± 1.6%) was less pronounced than previously reported. Subsequent LC-PDA-MS analysis showed that berberine collected by microfractionation of the Coptis extract had been, in part, transformed to active dihydroberberine. Formic acid added to the HPLC mobile phase to reduce peak tailing of protoberberine alkaloids acted as a reducing reagent according to the mechanism of the Leuckart-Wallach reaction. Among other structurally related protoberberines tested, dihydroberberine (30.1 ± 10.1% at 100 µM) was the most potent hERG inhibitor.

Nematode infections cause human morbidity and enormous economic loss in livestock. Since resistance against currently available anthelmintics is a worldwide problem, there is a continuous need for new compounds. The cyclooctadepsipeptide PF1022A is a novel anthelmintic that binds to the latrophilin-like transmembrane receptor important for pharyngeal pumping in nematodes. Furthermore, PF1022A binds to GABA receptors, which might contribute to the anthelmintic effect. Like other cyclodepsipeptides, PF1022A acts as an ionophore. However, no correlation between ionophoric activity and anthelmintic properties was found. This is the first study describing the effect of PF1022A on mammalian cells and tissues. While channel-forming activity was observed already at very low concentrations, changes in intracellular ion concentrations and reduction of contractility in isolated guinea pig ileum occurred at multiples of anthelmintically active concentrations. PF1022A did not induce necrotic cell death indicated by complete lack of cellular lactate dehydrogenase release. In contrast, apoptosis induction via the mitochondrial pathway was suggested for long-term drug treatment at high concentrations due to numerous apoptotic morphological changes as well as mitochondrial membrane depolarisation. Short time effects were based on cell cycle blockade in G0/G1 phase. Additionally, the cell cycle and apoptosis regulating proteins p53, p21 and bax, but not Bcl-2 were shown to impact on PF1022A-induced cytotoxicity. However, since PF1022A-induced cytotoxicity was found at drug concentrations higher than those used in anthelmintic treatment, it can be suggested that PF1022A intake might not impair human or animal health. Thus, PF1022A seems to be a safe alternative to other anthelmintic drugs.

Valerian is a commonly used herbal medicinal product for the treatment of anxiety and insomnia. Here we report the stimulation of chloride currents through GABA(A) receptors (I(GABA)) by valerenic acid (VA), a constituent of Valerian. To analyse the molecular basis of VA action, we expressed GABA(A) receptors with 13 different subunit compositions in Xenopus oocytes and measured I(GABA) using the two-microelectrode voltage-clamp technique. We report a subtype-dependent stimulation of I(GABA) by VA. Only channels incorporating beta(2) or beta(3) subunits were stimulated by VA. Replacing beta(2/3) by beta(1) drastically reduced the sensitivity of the resulting GABA(A) channels. The stimulatory effect of VA on alpha(1)beta(2) receptors was substantially reduced by the point mutation beta(2N265S) (known to inhibit loreclezole action). Mutating the corresponding residue of beta(1) (beta(1S290N)) induced VA sensitivity in alpha(1)beta(1S290N) comparable to alpha(1)beta(2) receptors. Modulation of I(GABA) was not significantly dependent on incorporation of alpha(1), alpha(2), alpha(3) or alpha(5) subunits. VA displayed a significantly lower efficiency on channels incorporating alpha(4) subunits. I(GABA) modulation by VA was not gamma subunit dependent and not inhibited by flumazenil (1 microM). VA shifted the GABA concentration-effect curve towards lower GABA concentrations and elicited substantial currents through GABA(A) channels at > or = 30 microM. At higher concentrations (> or = 100 microM), VA and acetoxy-VA inhibit I(GABA). A possible open channel block mechanism is discussed. In summary, VA was identified as a subunit specific allosteric modulator of GABA(A) receptors that is likely to interact with the loreclezole binding pocket.

Fast ('concentration jump') applications of neurotransmitters are crucial for screening studies on ligand-gated ion channels. In this paper, we describe a method for automated fast perfusion of neurotransmitters (or drugs) during two-microelectrode voltage-clamp experiments on Xenopus oocytes. The oocytes are placed in a small bath chamber that is covered by a glass plate with two channels for the microelectrodes that are surrounded by a quartz funnel serving as a reservoir for test solutions. The oocytes are perfused in a vertical direction via the two channels in the plate. Automation of compound delivery is accomplished by means of a programmable pipetting workstation. A mean rise time for 10-90% current increase through muscle-type nACh channels of 55.0+/-1.3 ms (30 muM acetylcholine) was estimated. Automation, fast perfusion rates, and economical use of compounds ( approximately 100 mul/data point) make the system suitable for screening studies on ligand- and voltage-gated ion channels.

localized sources that are possible targets for radiofrequency ablation (RFA). However, accurate identification of AF drivers in the 3D atrial structure may require simultaneous epi/endocardial (Epi/Endo) mapping. Methods: Coronary perfused human right atria (RA, n 1⁄4 6, 35–72 years) were optically mapped simultaneously by 3 high-resolution CMOS cameras (2 aligned Epi/Endo views, 3.3 3.3 cm, and 1 entire Endo view, 9 9 cm) with near-infrared dye. Sustained AF was induced by burst pacing during 10– 100 mM pinacidil (n 1⁄4 6). The AF driver was defined in the area of highest dominant frequency as a stable repetitive source of fibrillatory conduction. Results: Dual-sided intramural mapping revealed the common mechanism of sustained AF (430 minutes) in all 6 RA was a single, spatially and temporally stable, leading reentrant driver in the pectinate muscle region. Reentrant circuits (11.7 5 3.1 2 5.0 3 mm, 93 44 ms cycle length [CL]) had 2 limbs and intramural pivot points or “U turns” in conduction, creating stable patterns seen from sub-Endo but not sub-Epi. Endo RFA through pivot points consecutively organized (þ10% CL, n 1⁄4 5) and/or terminated AF (n 1⁄4 11). Attempts to reinduce AF resulted in slower AF drivers in different locations (0–2 per RA) or macroreentrant AT around the ablation lesion that was terminated by further ablation (n 1⁄4 3). Conclusions: Our integrative mapping study demonstrates, for the first time, that stable intramural reentry may drive AF in the human heart. Furthermore, this study represents a proof of concept that targeted ablation of localized drivers may successfully treat AF.

Background Heterologous expression of α1, β2 and γ2S(γ1) subunits produces a mixed population of GABAA receptors containing α1β2 or α1β2γ2S(γ1) subunits. GABA sensitivity (lower in receptors containing γ1 or γ2S subunits) and the potentiation of GABA-activated chloride currents (IGABA) by benzodiazepines (BZDs) are dependent on γ2S(γ1) incorporation [1]. A variable γ subunit incorporation may affect the estimation of IGABA potentiation by BZDs. We propose an approach for estimation of BZD efficiency that accounts for a mixed population of α1β2 and α1β2γ2S(γ1) receptors.
