Highlights • Vanadium history and industrial applications. • Recent research on metal vanadium complexes with amino acids. • Vanadium complexes with amino acids and their properties. • Vanadium complexes are promising candidates as metallopharmaceuticals., In the last 30 years, since the discovery that vanadium is a cofactor found in certain enzymes of tunicates and possibly in mammals, different vanadium-based drugs have been developed targeting to treat different pathologies. So far, the in vitro studies of the insulin mimetic, antitumor and antiparasitic activity of certain compounds of vanadium have resulted in a great boom of its inorganic and bioinorganic chemistry. Chemical speciation studies of vanadium with amino acids under controlled conditions or, even in blood plasma, are essential for the understanding of the biotransformation of e.g. vanadium antidiabetic complexes at the physiological level, providing clues of their mechanism of action. The present article carries out a bibliographical research emphaticizing the chemical speciation of the vanadium with different amino acids and reviewing also some other important aspects such as its chemistry and therapeutical applications of several vanadium complexes.

Graphical abstract, Highlights • Naturally-occurring antibodies were more resistant to degradation than monoclonal antibodies. • Monoclonal sIgA was more resistant to degradation than IgG and IgA. • Monoclonal antibodies may need to be provided at a higher dose to compensate for digestive losses., To help rationally design an antibody for oral administration, we examined how different isotypes (IgG, IgA and sIgA) with the same variable sequence affect antibody stability across digestion. We compared the degradation of recombinant palivizumab (IgG1), and recombinant IgA and sIgA versions of palivizumab spiked in human milk to the degradation of naturally-occurring anti-respiratory syncytial virus (RSV) sIgA/IgA and IgG in human milk from four donors across gastric and intestinal phases of an in vitro model of infant digestion via a validated RSV F protein ELISA. Palivizumab IgG and IgA formats were less stable than the sIgA version after complete simulated gastrointestinal digestion: palivizumab IgG, IgA and sIgA decreased across complete simulated gastrointestinal digestion by 55%, 48% and 28%, respectively. Naturally-occurring RSV F protein-specific IgG was stable across digestion, whereas naturally-occurring sIgA/IgA was stable in the gastric phase but decreased 33% in the intestinal phase of simulated digestion.

In vitro culture conditions were determined to induce an anti-Salmonella abortusovis antibody synthesis from lymph node leucocytes of three immunized sheep and two unprimed lambs maintained in culture in the presence of heat-inactivated bacteria for 2 weeks. Humoral immune responses were assessed by enumerating specific antibody-secreting cells using ELISASPOT and by titrating immunoglobulins secreted into culture supernatants using ELISA techniques. Optimal secondary antibody response was observed from cultures performed with fetal calf serum (compared with horse serum) and with an antigen concentration of one to ten bacteria per cell. This kind of antigenic stimulation allowed induction of numerous antibody-secreting cells without adsorption of the secreted antibodies. Maximal numbers of antibody-secreting cells could reach a rate of 1% of the sheep leucocytes initially put into culture. Kinetic profiles of antibody production from boosted lymph node cells were characterized by an ascending phase from the sixth to the twelfth day of culture and then showed a plateau phase until Day 14. Most of the responses were composed of IgM and IgG1 antibodies, traces of IgG2 being detected at the end of experiments. From the twelfth day of antigenic stimulation, the IgM isotype was preferentially expressed with high antigen concentration (100 bacteria per cell), whereas the highest amounts of IgG1 were detected at lower concentration (one to ten bacteria per cell). While anti-Salmonella IgM appeared to be mainly specific for the lipopolysaccharide (LPS) cell wall fraction, some IgG1 recognized other bacterial antigens. Kinetic profiles and magnitudes of primary antibody responses induced in vitro from lamb lymph node cells did not differ from those defined in cultures of sheep boosted leucocytes. But these immune reactions were mainly made up of anti-LPS IgM. Few anti-Salmonella IgG1 were detected from the tenth day of culture. So these in vitro assays allowed induction of an antibody synthesis from either in vivo sensitized or unprimed sheep lymph node leucocytes. This methodology would permit achievement of more detailed studies on interactions between Salmonella and lymph node leucocytes, leading to a better understanding of the mechanisms controlling bacterial dissemination through the lymphoid tissue.
