BackgroundYam production in sub-Saharan African countries faces challenges due to susceptibility to yam mosaic virus (YMV) disease, leading to significant yield losses. Dioscorea praehensilis, a semi-cultivated yam species known for its high yield and resistance to YMV, can be utilized as a new variety and a gene source to enhance tuber yield and YMV resistance of White Guinea yam. Investigating the genetic basis of tuber yield-related traits and YMV resistance in D. praehensilis through association mapping will facilitate the traits discovery and deployment into the yam breeding program.MethodologyA total of 132 D. praehensilis genotypes were evaluated in 2022 and 2023 seasons for tuber yield per plant, tuber size ratio and for mosaic virus severity response. The genotypes were sequenced using SNP markers generated from Diversity Array Technology Platform. The trait association analysis was conducted using mixed linear model implemented in GWASpoly package followed for putative genes analysis.ResultsPopulation structure and phylogeny analyses using 4,525 single nucleotide polymorphism (SNP) markers grouped the 132 D. praehensilis genotypes into three clusters. Using multiple gene action models which include dominant, additive, and general models, 27 SNP markers were significantly identified to be associated with the tuber yield, tuber size ratio and yam mosaic virus severity. Of these 27 SNPs, we identified three and two SNP markers for tuber yield and yam mosaic virus severity, respectively as stable markers across years. Using gene annotation, we identified 10 putative genes such as Serine/threonine-/dual specificity protein kinase, catalytic domain involved in starch biosynthesis and Ribosomal protein L5 involved in defense signaling against virus diseases. The marker effect revealed that alleles CC and TT were associated with high tuber yield, while AA and AC were linked with low tuber yield. For YMV severity response, alleles CC and CT were linked with low YMV severity, while TT was associated high YMV severity prediction.ConclusionThe results of this study offer valuable insights into comprehending the functional networks involved in developing strategies to enhance tuber yield and resistance to yam mosaic virus in Dioscorea spp. The markers and candidate genes discovered in this research will serve as crucial genomic resources for selection of D. praehensilis and other yam species.

IntroductionBush yam (Dioscorea praehensilis Benth.) is an important semi-domesticated food crop in West Africa. Limited information on the genetic architecture and its poor post-harvest tuber quality traits significantly hinder its use as food and source of income. Hence, dissecting the genetics underlying the expression of its post-harvest tuber quality traits is essential for establishing proper breeding schemes.MethodsIn this study, 138 D. praehensilis accessions collected in Ghana were sequenced using Diversity Array Technology (DArTSeq). The materials were profiled for dry matter content (DMC), tuber flesh oxidation (TBOXI) and for tuber flesh hardness (TBhard) during two cropping seasons.Results and discussionDiversity assessment using population structure, principal component analysis and hierarchical clustering methods revealed the presence of three major groups. Six genetic models were used for the trait association analysis using multiple random locus mixed linear model (MrMLM). Sixteen SNP markers distributed across the yam genome were identified to be associated with the evaluated traits. The associated SNP markers displayed a phenotypic variance ranged from 4.22% in TBHard to 16.92% in TBOXI. A total 25 putative candidate genes were identified around the SNP markers. The putative genes were identified to play key roles in tuber bulking, oxidative browning and starch hydroxylase. This study provides a valuable insight on the genetics underlying tuber quality traits in bush yam and opens avenues for developing genomic resources to improve D. praehensilis.

The potential of cowpea (Vigna unguiculata [L] Walp) to contribute to food security and livelihood sustenance of sub-Sahara Africans is constantly threatened by many biotic and abiotic stresses that are aggravated by climate change. To address these threats, cowpea breeding programs in the subregion prioritize climate-resilience traits and resistance to biotic stresses. However, before successful trait discovery and implementation, it is essential to characterize diversity and population structure of cowpea germplasm. To test the hypothesis that assembled cowpea germplasm exhibits limited and narrow genetic diversity with a well-defined population structure, we assessed the level of genetic variability and characterized the population structure of 188 cowpea genotypes using 5147 Single Nucleotide Polymorphism (SNP) markers. The structure results revealed five major genetic groups with moderate levels of genetic diversity and an admixture level of 17 %. Discriminant analysis and phylogenetic analysis supported this finding, indicating the presence of distinct groups within the cowpea population. The analysis of molecular variance (AMOVA) showed 27 % among population variance, 64 % within-population variance, and 9 % within individual variance. While considering the origin, the AMOVA showed 16 % among population variance, 75 % among individual variance, and 9 % within individual variance. This study provides valuable insights for future cowpea improvement programs by facilitating the selection of suitable progenitors for population development, and contributing in the conservation of cowpea genetic resources. Addressing these challenges and enhancing cowpea's diversity and resilience are crucial steps towards ensuring food security and sustainable livelihoods in sub-Saharan Africa.

The usage of biodegradable materials is gaining mounting applications owing to the environmental problems created by petroleum-based synthetic materials. Agro-waste materials are finding more scope as reinforcement materials in polymer composites, mainly because of their cost-effectiveness and availability. The main aim of this research work is to utilize agro-waste material in the form of mango seed waste (MSW) as a reinforcing material with polyvinyl alcohol (PVA) to form biocomposite. The biocomposites were fabricated using the solution casting technique. The research work focuses on the influence of varying ultrasonication periods (0–20 min at 5-min intervals, i.e., 0, 5, 10, 15, and 20 min) on the properties of PVA/MSW biocomposites. The fabricated biocomposites were subjected to tensile test, fracture morphology, moisture absorption (MA), Fourier transform infrared spectroscopy, and thermogravimetric analysis. The results revealed that the material with the highest tensile strength was 3.95 MPa obtained with the 20-min ultrasonication sample. This value is 49% higher than that obtained in the sample without ultrasonication. The scanning electron microscopy analysis shows that the 20-min ultrasonication sample indicated a good distribution of MSW fillers in the PVA contrasted with the rest of the samples. The thermal stability of the samples treated with ultrasonication was found to be significantly higher than that of the untreated samples, whereas the MA value decreased with increasing ultrasonication time. It can be observed from the results that biocomposites can be a potential alternative to the synthetic materials used in packaging applications.

Frozen Shoulder restricts daily life’s functional aspect, increasing morbidity. Hypertension and diabetes mellitus are risk factors that make an FS poor prognosis during treatment because of the diabetes glycation process and hypertension-enhanced vascularization. Prolotherapy injects an irritant solution into the tendon, joints, ligaments, and joint spaces to release growth factors and collagen deposition, reducing pain, restoring joint stability, and increasing the quality of life. We report 3 cases of patients with confirmed FS. Patient A with no comorbidity, patient B with diabetes mellitus, and patient C with hypertension, with all patient’s chief complaints of shoulder pain and limited ROM, and symptoms affected the general quality of daily life. This patient was provided injection with Prolotherapy treatment combined with physical therapy intervention. Patient A had significantly improved ROM to maximum after 6 weeks with relieved pain and improved shoulder function. Patients B and C showed increased ROM, still tiny, decreased pain, and improved shoulder function. In conclusion, prolotherapy demonstrated a beneficial effect in a patient with FS with comorbidities, although not to the maximum extent in patients without comorbidity.Keywords: protherapy, frozen shoulder, comorbidity, range of motion, quality of life

Water yam (Dioscorea alata L.) is the most widely cultivated yam species with good agronomic attributes. However, several biotic and abiotic constraints and its lower food quality such as poor pound ability limit its production and use. Therefore, the identification of superior genotypes with suitable characteristics is needed for water yam improvement. This study aims to assess a panel of half-sib (progenies with one parent in common) and full-sib (progenies with the same male and female parents) progenies as well as their parents for selection of desirable ideotypes based on their agronomic and quality characteristics. A total of 280 progenies from bi-parental populations as well as five parents were evaluated, and a significant variation was observed (p < 0.01) in their performances for the eight traits used in the study. A moderate to high broad-sense heritability (30% < H2–H2 ≥ 60%) was observed for all traits except for tuber pound ability (H2 < 30%). Positive correlations were displayed between the traits, while the hierarchical clustering grouped genotypes into three clusters indicating the potential for selection of diverse genotypes for multiple traits from the four families under study. Plant vigor and number of tubers per plant contributed (p < 0.01) positively to the yield per plant in the path coefficient analysis. Using the multi-trait genotype–ideotype distance index (MGIDI), a total of 39 most promising genotypes were identified. These promising genotypes could be further used as progenitors in D. alata improvement programs targeting good agronomic and quality traits targeted for farmers and end users.

Abstract Yam (Dioscorea spp.) is cultivated in many villages of DR Congo as a means to sustain food security and alleviate poverty. However, the extent of the existing diversity has not been studied in details thus, considered as an orphan. A survey covering 540 farmers in 54 villages was conducted in six major yam growing territories covering three provinces in DR Congo to investigate the diversity, management and utilization of yam landraces using pre-elaborate questionnaires. Subject to synonymy, a total of 67 landraces from five different species were recorded. Farmers’ challenges limiting yam production were poor tuber qualities (69%), harvest pest attack (7%), difficulty in harvesting (6%), poor soil status (6%). The overall diversity was moderate among the recorded yam germplasm maintained at the household level (1.32) and variability exist in diversity amongst the territories and provinces. Farmers’ in territories of Tshopo and Mongala provinces maintained higher level of germplasm diversity (2.79 and 2.77) compared to the farmers in territories of Bas-Uélé (1.67). Some yam landraces had limited abundance and distribution due to loss of production interest in many villages attributable to poisons contained hence, resulting in possible extinction. Farmers’ most preferred seed source for cultivation were backyard (43%) and exchange with neighboring farmers (31%) with the objective of meeting food security and generating income. In villages where yam production is expanding, farmers are relying on landraces with good tuber qualities and high yield even though they are late maturing. This study revealed the knowledge of yam landrace diversity, constraints to production and farmers’ preferences criteria as a guide for collection and conservation of yam germplasm for yam improvement intervention.

The study assessed the levels of usage of conservation agriculture technology in Kisangani, Democratic Republic of Congo and Angonia, Mozambique. A structured questionnaire was randomly administered to 192 farmers in each study site to collect data through a multistage sampling process. The study employed descriptive statistics and multinomial logistic regression to examine possible relationships among the study parameters. The results showed that land size used for conservation agriculture was a significant predictor in both study sites. Farmers’ intention to adopt depended on the services of vulgarisation of conservation agriculture technology. The results further showed that Kisangani farmers do not use the three-conservation agriculture technology at the same time, but they use crop rotation (54%). However, farmers use the three technologies (30%) simultaneously and soil cover (38%) in Angonia. The results suggest that efforts to promote adoption in Kisangani should be based on equal provision of extension services in all locations, and that the current farmer field schools (FFS) approach should be redesigned for contextualisation. For both study sites, the results imply that the use of FFS should be adapted to the use of farmer-to-farmer extension services, which can improve the upscaling of conservation agriculture to increase food security in a sustainable manner

Dioscorea praehensilis Benth. is a semi-wild yam species and a valuable source of resistance trait genes. To access the agronomic and tuber quality performance, eleven quantitative phenotypic traits were used to discriminate and identify promising accessions among 162 accessions of D. praehensilis collected in Ghana. Significant and high genetic variability (p < 0.001) for all eleven quantitative traits was found among the evaluated accessions. Moderate broad-sense heritability (H2) (30–60%) was observed for all the evaluated quantitative traits except the response to YMV and tuber hardness. The accessions were clustered into three groups; each cluster displayed genotypes with good potentiality for the different traits evaluated. Path coefficient analysis revealed positive contributions (p < 0.01) of the number of tubers per plant, tuber length, tuber width, stem internode length, number of internodes, and tuber flesh hardness to the total tuber weight per plant. Through the multi-trait genotype–ideotype distance index (MGIDI), 24 accessions were identified from the 162 evaluated accessions as top-ranking and could be used as progenitors for trait introgression. The results of this study provide insight for future yam breeding and improvement programs in West Africa.

Yams (Dioscorea spp.) possess the potential to contribute to food security and poverty alleviation in DR Congo; however, yam production is limited by several constraints, including the lack of yam improvement programs to address challenges relating to yield improvement, resistance to foliar diseases, and post-harvest tuber quality. Identification of a superior genotype for these traits and reservoirs of genes for improvement would guide yams’ improvement. This study aims to evaluate and identify landraces with superior performance for farmers and consumers. We evaluated 191 accessions from six yam species, and significant variation in the performances was observed at p < 0.05. Accessions of D. alata were superior for tuber oxidative browning (−0.01), D. cayenensis for high yield potential (29 t/ha), D. bulbifera for yam mosaic virus (YMV) tolerance (AUDPC = 3.88), and D. rotundata for tuber dry matter content (37%). A high genotypic and phenotypic coefficient of variation (>40) was observed for tuber yield, number of tubers per plots, tuber flesh oxidative browning, and tuber flesh texture. High broad-sense heritability estimates (>60) were similarly observed for all the assessed parameters except number of tubers per plot. Tuber size was identified as the best predictor for tuber yield (b = 2.64, p < 0.001) and tuber dry matter content (b = 2.21, p < 0.001). The study identified twenty stable landrace accessions from three Dioscorea species (D. alata (7); D. cayenensis (2); D. rotundata (11)). These accessions combined high yield potential, high tuber dry matter, high tolerance to YMV and YAD, and low tuber flesh oxidation. The accessions could be considered for the establishment of a yam improvement program in DR Congo.

This research emphasizes eloquently on membrane technology for treatment of palm oil mill effluent (POME) as it is the Malaysia’s largest and most important agro based industry. Findings established significant quality improvement with an efficient recovery of water from palm oil mill via innovative membrane application. Conventional bio-methods, whilst adhering to the Department of Environment’s (DOE) discharge regulations, produces brownish liquid which pales in comparison to the crystal clear water obtained through membrane treatment. The pre-treatment process consists of coagulation-flocculation using green environmental coagulant bases such as Moringa oleifera (MO) seeds. The ultrafiltration polyvinylidene difluoride (PVDF) and thin film composite (TFC) reverse osmosis were vital for the membrane processes. The system gave 99% suspended solids reduction in suspended solid and 78% of water present was successfully recovered. This technology guarantees water recovery with drinking water quality; meeting the US Environmental Protection Agency (USEPA) standard or could be recycled into the plant with sludge utilization for palm oil estates, thus enabling the concept of zero discharge to be executed in the industries. In addition, green and healthy antioxidants such as oil and beta-carotene can be recovered from POME further demonstrate. Silica gel showed better performance in separation of carotenes from oil at temperature 40°C using adsorption chromatography with 1154.55 ppm. The attractiveness of this technology, enabling the utilization of reuse of agricultural waste into potentially value added products.

The purpose of this study was to evaluate agreement in grading levels between primary and secondary graders and to calculate their sensitivity and specificity for identifying sight-threatening disease in an optometry-based retinopathy screening program.Methods: This was a retrospective study using data from 8,977 patients registered in the North Nottinghamshire retinal screening program. In all cases, the ophthalmology diagnosis was used as the arbitrator and considered to be the gold standard. Kappa statistics were used to evaluate the level of agreement between graders.Results: Agreement between primary and secondary graders was 51.4% and 79.7% for detecting no retinopathy (R0) and background retinopathy (R1), respectively. For preproliferative (R2) and proliferative retinopathy (R3) at primary grading, agreement between the primary and secondary grader was 100%. Where there was disagreement between the primary and secondary grader for R1, only 2.6% (n=41) were upgraded by an ophthalmologist. The sensitivity and specificity for detecting R3 was 78.2% and 98.1%, respectively. None of the patients upgraded from any level of retinopathy to R3 required photocoagulation therapy. The observed kappa between the primary and secondary grader was 0.3223 (95% confidence interval 0.2937–0.3509), ie, fair agreement, and between the primary grader and ophthalmology for R3 was 0.5667 (95% confidence interval 0.4557–0.6123), ie, moderate agreement. Conclusion: These data provide information on the safety of a community optometry-based retinal screening program for screening as a primary and as a secondary grader. The level of agreement between the primary and secondary grader at a higher level of retinopathy (R2 and R3) was 100%. Sensitivity and specificity for R3 were 78.2% and 98.1%, respectively. Abstract Obstructive sleep apnoea (OSA) is a cardio-metabolic disorder. Whether metabolic syndrome (MS), insulin resistance (IR) and albuminuria are independently associated with OSA is unclear, but defining the interactions between OSA and various cardiovascular (CV) risk factors independent of obesity facilitates the development of therapeutic strategies to mitigate their increased CV risks. We prospectively recruited 38 subjects with OSA and 41 controls. Anthropometric measurements, glucose, lipids, insulin and blood pressure (BP) were measured after an overnight fast. IR state was defined as homeostasis model assessment (HOMA) value >3.99 and MS diagnosed according to the International Diabetes Federation (IDF) criteria. Subjects with OSA were more obese, more insulin resistant, more hyperglycaemic, had higher Epworth score (measure of day time somnolence) and systolic blood pressure levels. The prevalence of MS was higher in OSA compared with non-OSA subjects (74% vs 24%, p < 0.001). The prevalence of microalbuminuria in both groups was negligible. Logistic regression adjusted for age, BMI and smoking showed that the patient with OSA was 5.9 (95% CI 2.0–17.6) times more likely to have MS than non-OSA patient. Triglyceride (p = 0.031), glucose (0.023) and Epworth score (0.003) values were independently associated with OSA after adjusting for BMI and other covariates whilst IR status was found not to be significant. Using the ROC curve analysis, we found that a waist circumference of >103 cm would predict MS in patients with OSA at 75–78% sensitivity and 61–64% specificity. The agreement between MS and IR state in this cohort is poor. Thus, OSA is associated with MS independent of obesity predominantly due to increased triglyceride, glucose and Epworth score values but not IR or microalbuminuria status. This observation suggests an alternative pathogenic factor mediating the increased cardiovascular risk in patients with OSA and MS, other than that due to IR. The independent link between Epworth score and MS in patients with OSA implicates the role of daytime sleepiness and chronic hypoxia as a potential mediator. Given the discordant between MS and IR state, measurement of waist is useful for predicting mainly MS but not insulin resistance status in patients with OSA. Appropriate pharmacological intervention targeting these independent factors is important in reducing the increased CV risks among patients with OSA.

Yam is an important tuber crop with enormous potential to enhance rural sustenance and livelihood in DRC. However, studies to enhance its genetic improvement are very far from sufficient with only a handful of information available on the crop. Yam has been treated as an orphan crop compared to contemporary crops such as cassava and sweet potato which have adapted to different cropping systems and become widespread in production. The lack of research attention to address the major production challenges has further decreased the value and potential of the crop compared to its contemporaries. These production constraints include lack of adequate quality planting materials, low yield potential, poor resistance/tolerance to yam mosaic and anthracnose diseases and ultimately poor tuber quality attributes focusing on tuber taste, flesh oxidation and dry matter contents of the majority of the farmers preferred varieties. In this review, we evaluated the status of yam in DRC and presented the needful activities to be incorporated for its improvement. Diversity has however been maintained mainly through ennoblement efforts in house gardens and small farmlands using traditional farming methods. Studies from other nations where yam has been successful with prominence in characterization and genetic improvement brought to light the need for DRC to consider yam as a staple carbohydrate food source, even to the extent of modifications in food public policy.
