Rationale: Air pollution has been associated with increased prevalence of type 2 diabetes; however, the mechanisms remain unknown. We have shown that acute ozone exposure in rats induces release of stress hormones, hyperglycemia, leptinemia, and glucose intolerance that are associated with global changes in peripheral glucose, lipid, and amino acid metabolism.Objectives: To examine ozone-induced metabolic derangement in humans using serum metabolomic assessment, establish human-to-rodent coherence, and identify novel nonprotein biomarkers.Methods: Serum samples were obtained from a crossover clinical study that included two clinic visits (n = 24 each) where each subject was blindly exposed in the morning to either filtered air or 0.3 parts per million ozone for 2 hours during 15-minute on-off exercise. Serum samples collected within 1 hour after exposure were assessed for changes in metabolites using a metabolomic approach.Measurements and Main Results: Metabolomic analysis revealed that ozone exposure markedly increased serum cortisol and corticosterone together with increases in monoacylglycerol, glycerol, and medium- and long-chain free fatty acids, reflective of lipid mobilization and catabolism. Additionally, ozone exposure increased serum lysolipids, potentially originating from membrane lipid breakdown. Ozone exposure also increased circulating mitochondrial β-oxidation-derived metabolites, such as acylcarnitines, together with increases in the ketone body 3-hydroxybutyrate. These changes suggested saturation of β-oxidation by ozone in exercising humans.Conclusions: As in rodents, acute ozone exposure increased stress hormones and globally altered peripheral lipid metabolism in humans, likely through activation of a neurohormonally mediated stress response pathway. Many potential applications in nanotechnology envisage the use of better-dispersed and functionalized preparations of carbon nanotubes. Single-walled carbon nanotubes (SWCNTs) were treated with 1:1 mixtures of concentrated nitric and sulfuric acids for 3 min in a microwave oven under 20 psi pressure followed by extensive dialysis to remove the acids. This treatment resulted in acid functionalized SWCNTs (AF-SWCNTs) that had high negative charge (Zeta potential -40 to -60 mV) and were well dispersed (98% of the particles <150 nm) in aqueous suspensions. In vitro and in vivo toxic effects of SWCNTs and AF-SWCNTs were compared. AF-SWCNTs exerted a strong cytotoxic effect on LA4 mouse lung epithelial cells in culture that could be blocked by prior treatment of the nanotubes with poly L-lysine which neutralized the electric charge and promoted re-agglomeration. AF-SWCNT, but not the unmodified SWCNT preparations, strongly inhibited cell cycling of LA4 cells. Both SWCNTs and AF-SWCNTS were however equally effective in inducing apoptotic responses in LA4 cells as examined using an Annexin V binding assay. Oro-pharyngeal aspiration of AF-SWCNT preparation induced a strong acute inflammatory response in the lungs of CD1 mice, compared to control SWCNTs which caused only a marginal effect. Taken together the results indicate that unlike pristine SWCNTs, acid-functionalized SWCNT preparations exert strong toxic effects in vitro and in vivo and these effects can be reversed by neutralizing their surface charge. Introduction: As a lifestyle factor, poor sleep status is associated with increased cardiovascular morbidity and mortality and may be influenced by environmental stressors, including air pollution., Methods: To determine whether exposure to air pollution modified cardiovascular effects of sleep disruption, we evaluated the effects of single or repeated (twice/wk for 4 wks) inhalation exposure to eucalyptus wood smoke (ES; 964 μg/m 3 for 1 h), a key wildland fire air pollution source, on mild sleep loss in the form of gentle handling in rats. Blood pressure (BP) radiotelemetry and echocardiography were evaluated along with assessments of lung and systemic inflammation, cardiac and hypothalamic gene expression, and heart rate variability (HRV), a measure of cardiac autonomic tone., Results and Discussion: GH alone disrupted sleep, as evidenced by active period-like locomotor activity, and increases in BP, heart rate (HR), and hypothalamic expression of the circadian gene Per2 . A single bout of sleep disruption and ES, but neither alone, increased HR and BP as rats transitioned into their active period, a period aligned with a critical early morning window for stroke risk in humans. These responses were immediately preceded by reduced HRV, indicating increased cardiac sympathetic tone. In addition, only sleep disrupted rats exposed to ES had increased HR and BP during the final sleep disruption period. These rats also had increased cardiac output and cardiac expression of genes related to adrenergic function, and regulation of vasoconstriction and systemic blood pressure one day after final ES exposure. There was little evidence of lung or systemic inflammation, except for increases in serum LDL cholesterol and alanine aminotransferase.
