A new mobile application with a new semi-quantitative Food Frequency Questionnaire (FFQ) was developed as a quick dietary assessment tool. This study aimed to test the validation of food and nutrient intake measurements from an app-based FFQ, which included 49 dishes and food items, using non-consecutive two-day 24-hour diet recalls as the standard. A total of 179 women aged 19-70 years (mean (SD) = 33.7 (9.9)) from two rural areas of Kenya, Vihiga and Kitui counties, were included. Dietary assessment by the two dietary survey methods was conducted from November to December 2021. The intake of macro- and micronutrients, and food groups between the two methods were compared by Spearman's rank correlation coefficients, cross-classification agreement, and Bland-Altman plots for assessing the validation of the app-based FFQ. The median correlation coefficients between energy-adjusted intakes estimated by the app-based FFQ and the 24-h diet recall were 0.19 (0.10 (protein)-0.26 (fat)) for macronutrients, 0.24 (0.11 (sodium)-0.46 (vitamin B12)) for micronutrients, and 0.26 (0.07 (seasonings)-0.45 (pulses)) for 17-food groups. The median percentage of classification into the same quartiles between the two methods was 31% for macronutrient, 32% for micronutrient and 39% for food group. While 9% for macronutrient, 8% for micronutrient and 10% for the food group were misclassified into the opposite categories. Bland-Altman Plots showed acceptable agreement for macronutrients intake between the two methods. The app-based FFQ was shown to be able to rank or categorize individuals according to their intake of most of the nutrients and food groups we examined. This dietary assessment tool can be used for quantitative dietary data collection. Further research is needed to improve the validation of the app-based FFQ, as it has been shown that intakes of staple foods and sugar are underestimated and others are overestimated, and that estimating intakes of some specific nutrients, such as protein, is challenging. Radiological objective response (OR) was assessed using modified response evaluation criteria in solid tumors (mRECIST) guidelines.Results: Baseline demographic parameters were comparable between 126 (69.6%) patients with CP-A disease and 55 patients (30.4%) with CP-B disease. Frequency of lenvatinib-related adverse events, including decreased appetite (P=0.034), diarrhea (P=0.040), elevated serum bilirubin (P=0.016) and vomiting (P=0.009), were higher in CP-B than in CP-A patients. Relative dose intensity (RDI) was significantly higher in CP-A (0.69) than CP-B patients (0.50, P < 0.001). Furthermore, OR rate (44.0%) was markedly higher in CP-A5 patients as compared to CP-A6 (25.5%), CP-B7 (22.2%), and CP-B8 patients (5.3%), respectively (P=0.002). In multivariable analysis, performance status (0 vs 1, 2, P=0.026), CP class (A vs B, P=0.045) and RDI (≥ 0.7 vs < 0.7, P=0.034) were identified as factors associated with response to lenvatinib treatment. Overall survival (OS) at 12 months was significantly different between CP-A (66.3%) and CP-B patients (30.0%, P=0.002), and between CP 5– 7 (59.2%) and CP 8 patients (34.8%, P=0.003). In multivariable analysis, CP class (A vs B, P=0.007) and Barcelona clinic liver cancer (BCLC) stage (B vs C, P=0.002) were associated with OS following lenvatinib treatment.Conclusion: Lenvatinib treatment offers significant benefits in patients with good liver function in real-world practice. The various characteristics identified in this study might be helpful as clinical predictors of response to lenvatinib and survival in clinical practice. Pathogenic amyloid-β peptide precursor (APP) mutations clustered around position 693 of APP-position 22 of the Aβ sequence--are commonly associated with congophilic amyloid angiopathy (CAA) and intracerebral hemorrhages. In contrast, the Osaka (E693Δ) intra-Aβ APP mutation shows a recessive pattern of inheritance that leads to AD-like dementia despite low brain amyloid on in vivo positron emission tomography imaging. Here, we investigated the effects of the Osaka APP mutation on Aβ accumulation and deposition in vivo using a newly generated APP transgenic mouse model (E22ΔAβ) expressing the Osaka mutation together with the Swedish (K670N/M671L) double mutation. E22ΔAβ mice exhibited reduced α-processing of APP and early accumulation of intraneuronal fibrillar Aβ oligomers associated with cognitive deficits. In line with our in vitro findings that recombinant E22Δ-mutated Aβ peptides form amyloid fibrils, aged E22ΔAβ mice showed extracellular CAA deposits in leptomeningeal cerebellar and cortical vessels. In vitro results from thioflavin T aggregation assays with recombinant Aβ peptides revealed a yet unknown antiamyloidogenic property of the E693Δ mutation in the heterozygous state and an inhibitory effect of E22Δ Aβ42 on E22Δ Aβ40 fibrillogenesis. Moreover, E22Δ Aβ42 showed a unique aggregation kinetics lacking exponential fibril growth and poor seeding effects on wild-type Aβ aggregation. These results provide a possible explanation for the recessive trait of inheritance of the Osaka APP mutation and the apparent lack of amyloid deposition in E693Δ mutation carriers.

The periodontal ligament (PDL) contains mesenchymal stem cells (MSCs) that can differentiate into osteoblasts, cementoblasts, and fibroblasts. Nevertheless, the distribution and characteristics of these cells remain uncertain. Gli1, an essential hedgehog signaling transcription factor, functions in undifferentiated cells during embryogenesis. Therefore, in the present study, the differentiation ability of Gli1+ cells was examined using Gli1-CreERT2/ROSA26-loxP-stop-loxP-tdTomato (iGli1/Tomato) mice. In 4-wk-old iGli1/Tomato mice, Gli1/Tomato+ cells were only slightly detected in the PDL, around endomucin-expressing blood vessels. These cells had proliferated over time, localizing in the PDL as well as on the bone and cementum surfaces at day 28. However, in 8-wk-old iGli1/Tomato mice, Gli1/Tomato+ cells were quiescent, as most cells were not immunoreactive for Ki-67. These cells in 8-wk-old mice exhibited high colony-forming unit fibroblast activity and were capable of osteogenic, chondrogenic, and adipogenic differentiation in vitro. In addition, after transplantation of teeth of iGli1/Tomato mice into the hypodermis of wild-type mice, Tomato fluorescence indicating the progeny of Gli1+ cells was detected in the osteoblasts and osteocytes of the regenerated bone. These results demonstrate that Gli1+ cells in the PDL were MSCs and could contribute to the alveolar bone regeneration. [ABSTRACT FROM AUTHOR]

A study was conducted, using nuclear magnetic resonance (NMR) micro imaging, to examine the moisture distribution and oil distribution in tempura coating. The result reveals a three-layered fine structure of the tempura coating on these NMR images.
