The effect of arsenite exposure on cell viability, protein synthesis, energy metabolism and the expression of genes coding for cytoplasmic (hsp70) and endoplasmic reticulum (ER; gadd153, grp78, grp94) stress proteins was investigated in primary neuronal cell cultures. Furthermore, signs of ER stress were evaluated by investigating xbp1 mRNA processing. Arsenite levels of 30 and 100 μM induced severe cell injury. Protein synthesis was reduced to below 20% of control in cultures exposed to 30 and 100 μM arsenite for 1 h, and it remained markedly suppressed until 24 h of exposure. Arsenite induced a transient inhibition of energy metabolism after 1 h of exposure, but energy state recovered completely after 3 h. Arsenite exposure affected the expression and translation of genes coding for HSP70 and GRP78, GRP94, GADD153 to different extents. While hsp70 mRNA levels rose drastically, approximally 550-fold after 6 h exposure, HSP70 protein levels did not change over the first 6 h. On the other hand, gadd153 mRNA levels rose only approximately 14-fold after 6 h exposure, while GADD153 protein levels were markedly increased after 3 and 6 h exposure. HSP70 protein levels were markedly increased and GADD153 protein levels decreased to almost control levels in cultures left in arsenite solution for 24 h, i.e. when only a small fraction of cells had escaped arsenite toxicity. Arsenite exposure of neurons thus induced an imbalance between pro-apoptotic and survival-activating pathways. Despite the marked increase in gadd153 mRNA levels, we did not observe signs of xbp1 processing in arsenite exposed cultures, indicating that arsenite did not produce ER stress. To study the changes in gene expression in senescent cells we applied the suppression subtractive hybridization of two cDNA pools isolated from human parenchymal kidney cells in the phase of exponential growth and cellular senescence in vitro. In addition to several genes known to be associated with cellular senescence, we identified a new gene, which is overexpressed in senescent kidney parenchymal cells. The full-length cDNA consists of 5226 nucleotides with an open reading frame (ORF) encoding 701 amino acids (Accession number: AJ306929). The gene product has a predicted molecular mass of 77.31 kDa. The ORF of the new gene shows significant homology to P-type ATPase family gene products and therefore was called AFURS1 (ATPase family homolog up-regulated in senescence cells). The consensus sequence phosphorylation site (DKTGTLT) is highly conserved. According to the GenBank database AFURS1 is mapped to the sequence segment NT_005535.3 at chromosomal region 3q26.32 and has 18 exons. The AFURS1 gene might have a role in cellular aging and tumor suppression as well. [Copyright &y& Elsevier]

Hepatocellular carcinoma and cholangiocarcinoma are the most common primary liver tumours, whose incidence and associated mortality have increased over recent decades. Liver cancer is often diagnosed late when curative treatments are no longer an option. Characterising new molecular determinants of liver carcinogenesis is crucial for the development of innovative treatments and clinically relevant biomarkers. Recently, circular RNAs (circRNAs) emerged as promising regulatory molecules involved in cancer onset and progression. Mechanistically, circRNAs are mainly known for their ability to sponge and regulate the activity of microRNAs and RNA-binding proteins, although other functions are emerging ( e.g. transcriptional and post-transcriptional regulation, protein scaffolding). In liver cancer, circRNAs have been shown to regulate tumour cell proliferation, migration, invasion and cell death resistance. Their roles in regulating angiogenesis, genome instability, immune surveillance and metabolic switching are emerging. Importantly, circRNAs are detected in body fluids. Due to their circular structure, circRNAs are often more stable than mRNAs or miRNAs and could therefore serve as promising biomarkers - quantifiable with high specificity and sensitivity through minimally invasive methods.
