ヘムはポルフィリン環に二価鉄が配位した鉄化合物であり、酸素の運搬や呼吸、活性酸素の除去などに働くタンパク質の補因子としてあらゆる生物にとって重要である。大腸菌でも、ヘムは電子伝達系のシトクロムや活性酸素の除去に働くカタラーゼおよびペルオキシダーゼの補因子などとして重要な働きをしている。ヘム合成に関与する酵素の遺伝子は多くの生物で保存されているが、ヘム合成遺伝子の変異株では、ヘムが合成されないために生育が悪くなるのに加えて、蓄積したヘム合成の中間体が生育を阻害することが知られている。大腸菌ではヘム合成遺伝子hemH が欠損すると可視光照射下では生育できなくなることが報告されていて、ヘム合成経路の最後の中間体であるprotoporphyrinⅨの蓄積が原因であると考えられている。この光感受性の原因は酸化ストレスであろうと推測されているが、実験的に示されてはいなかった。また大腸菌のhemH 遺伝子以外の一連のヘム合成遺伝子の変異株の光感受性については明らかになっていなかった。そこで本研究ではヘム合成遺伝子群の変異株の光感受性について解析を行った。ヘム合成遺伝子は欠失させると好気条件下での生育が著しく低下し、サプレッサー変異などが起こることによって遺伝学的解析が難しくなる可能性があるため、高温感受性欠失株を利用した。まず一連のヘム合成遺伝子変異株について高温での暗条件と光条件での生育を調べたところ、hemH 遺伝子以外にもポルフィリン中間体が蓄積するhemD 遺伝子, hemE 遺伝子, hemF とhemN 遺伝子、hemG 遺伝子の変異株は、ポルフィリン中間体が蓄積しないhemB 遺伝子, hemC 遺伝子の変異株と比べて、光条件での生育が悪く、特にhemF とhemN 変異株とhemH 変異株は光感受性が強いことがわかった。またヘム合成の前駆体である5-アミノレブリン酸を添加すると光感受性が強くなることもわかった。これらの変異株の光感受性の原因が酸化ストレスである可能性について調べるために、まず嫌気条件での生育について調べたところ、好気条件に比べて嫌気条件では光感受性が弱くなることが分かった。またhemH 変異株でさらに活性酸素の除去に働く酵素の遺伝子を欠失させた株の光感受性について調べたところ、ペルオキシダーゼをコードするahpCF 遺伝子の欠失株では、暗条件と光条件の両方で生育が低下することが分かり、ヘム合成遺伝子群の変異株の光感受性が、蓄積されたポルフィリン中間体によって生じた活性酸素種による酸化ストレスによって引き起こされることが示唆された。本研究によって大腸菌のヘム合成遺伝子群の変異株の光感受性について新たな知見を得ることができた。これらの知見をもとに、今後実際に蓄積されているポルフィリン中間体を調べることによって、それぞれの中間体の性質やそれらの毒性を回避するシステムの可能性などについて調べていくことができると考えられる。, Heme is a compound, in which divalent iron is coordinated to the porphyrin ring, and is important for all organisms as a cofactor of proteins involved in oxygen transportation, respiration, and removal of reactive oxygen species. In E. coli, heme was known to play an important role as a cofactor of cytochrome, a component of electron transport system, and as that of catalase and peroxidase for oxidative stress tolerance. The hem genes required for heme synthesis are conserved in many organisms. Their mutant cells are often deficient in cell growth especially under aerobic condition due to the loss of hemes and the accumulation of the toxic intermediates of the heme synthesis. In E. coli, the hemH mutant was reported to be sensitive to visible light irradiation possibly due to the accumulation of protoporphyrinⅨ, which is the final intermediate of the heme synthesis pathway. However, the photosensitivity of other hem mutants had not been reported. And the photosensitivity is supposed to be caused by oxidative stress, but it has not been confirmed experimentally. In this work, I studied the photosensitivity of other hem mutants of E. coli. To avoid the effects of suppressor mutations etc, I used the temperature-sensitive heme deletion mutants, which lose the complementing hem plasmids at high temperature. First, I examined the cell growth of the hem mutants under dark or light conditions. The hemD, hemE, hemF-hemN, hemG mutants, which accumulate porphyrin intermediates, grow poorly under light condition as well as the hemH mutant. Especially, the hemF-hemN double mutant and the hemH mutant were remarkably photosensitive. Furthermore their photosensitivity was enhanced in the presence of 5-aminolevulinic acid, a precursor of heme synthesis. But the growth of the hemB and hemC mutants, which does not accumulate porphyrin intermediates, was not inhibited. To confirm that the photosensitivity of these mutants is due to oxidative stress, the cell growth under anaerobic condition was examined. The photosensitivity was not remarkable under anaerobic condition. And the effect of the mutation of the ahpCF gene encoding a peroxidase involved in oxidative stress tolerance was studied. The cell growth of the ahpCF derivatives was inhibited under both dark and light conditions. These results suggested that the photosensitivity of those mutants is caused by reactive oxygen species derived from the accumulated porphyrin intermediates. In this work, I investigated the photosensitivity of the mutants of the hem genes.
