We assess the composition and geometry of four individual rock glaciers in Alaska, Wyoming and Colorado by measuring their radio wave speed and applying these results to ground-penetrating radar depth corrections and dielectric mixing models. Our method includes a correction for subsurface reflector dip angle, which we show can lead to an incorrect determination of wave speeds using common midpoint configurations. By observing the radar properties of the rock glaciers and their supraglacial debris, we find that some of the sites exhibit nearly pure ice cores, and all of the sites indicate volumetric ice fractions >50%. These results have implications for terrestrial glaciology and hydrology because the present ice volume is connected to past ice accumulation and subsurface ice preservation, which may affect the future availability of alpine water resources. An understanding of the processes that govern rock glacier evolution over a wide range of latitudes and elevations will also contribute to the exploration of planetary surfaces such as Mars, which hosts a significant population of debris-covered glaciers. Our subsurface composition and geometry estimates will inform simulations of rock glacier formation and evolution to test hypothesized ice origin mechanisms along with the preservation of climate signals.

An integrated seismic–stratigraphic study of the Lower Cretaceous Cromer Knoll Group was undertaken as part of a recent comprehensive analysis of the Upper Jurassic – lowermost Cretaceous petroleum system in the Danish Central Graben. This study of the basal group of the post-rift package yielded an updated regional assessment of the distribution of the Valhall, Tuxen, Sola and Rødby Formations. This is documented by four high resolution isochore maps (presented here) that record temporal shifts in subsidence patterns from the latest Ryazanian to the earliest Cenomanian. The distribution and thickness variation of the mud-dominated Valhall Formation (latest Ryazanian – early Hauterivian) at the base of the group attests to the progressive fill of inherited syn-rift morphology. The dominant depositional theme is thus ponding in, and onlap from, the main inherited depocentres, although growth faults and incipient inversion locally controlled stratigraphic architecture, and new depocentres were initiated in the east of the graben (Ål and Outer Rough Basins). The isochores for the succeeding, increasingly chalk-rich Tuxen, Sola and Rødby Formations (Hauterivian – earliest Cenomanian) document the regional weakening of syn-rift patterns but emphasize the shift in sedimentation patterns controlled by accelerating inversion activity in the east (Adda–Tyra area, Søgne Basin) and increased local subsidence. The latter sometimes coincided with syn-rift depocentres, such as the Roar Basin and the Arne-Elin Graben, but was also significant in the new Early Cretaceous depocentres in the west of the graben, particularly the Outer Rough Basin. The evolution of the Early Cretaceous basin recorded by this dataset reveals significant shifts in the subsidence pattern in the late Valanginian – early Hauterivian, in large part due to early inversion in the east, and during the late Aptian – early Albian when subsidence was focussed in central and western sub-basins, probably locally due to salt withdrawal. These events, in combination with sea-level change, had implications for the accumulation and preservation of Barremian and early Aptian reservoir chalks.

Enzymatic degradation of plastics is currently limited to the use of engineered natural enzymes. As of yet, all engineering approaches applied to plastic degrading enzymes retain the natural α/β-fold. While mutations can be used to increase thermostability, an inherent maximum likely exists for the α/β-fold. It is thus of interest to introduce catalytic activity toward plastics in a different protein fold to escape the sequence space of plastic degrading enzymes. Here, a method for designing highly thermostable enzymes that can degrade plastics is described. This has been used to design an enzyme that can catalyze the hydrolysis of polycarbonate, which no known natural enzymes can degrade. Rosetta enzyme design is used to introduce a catalytic triad into a set of thermostable scaffolds. Through computational evaluation, a potential PCase was selected and produced recombinantly in E. coli. CD spectroscopy suggests that the design has a melting temperature of >95°C. Activity towards a commercially used polycarbonate (Makrolon 2808) was confirmed using AFM, which showed that a PCase had been designed successfully.

Buried ice in the Martian mid-latitudes is of high interest for In-Situ Resource Utilization (ISRU) by future crewed missions, as well as for its scientific value as a record of climate history on another planet. ``Lobate debris aprons,'' widely considered to be debris-covered glaciers, constitute a large reservoir of such ice. Many have been probed by Mars Reconnaissance Orbiter's Shallow Radar (SHARAD) sounder, which imaged their base and constrained their internal composition to >80% water ice. The basal contact is not imaged by SHARAD for all aprons, however, which introduces the possibility that these features contain significantly less ice. In this study, we used SHARAD survey results in tandem with high-resolution digital terrain models to show that these ``non-detections'' can be attributed to surface roughness, which through scattering reduces the coherent signal that penetrates the aprons. We quantified this effect using a fractal backscattering model and found that the roughest surfaces coincided with the location of basal non-detections. There is thus no need for reduced ice content to explain nondetection of lobate debris apron bases with SHARAD. This result leads us to interpret all lobate debris aprons as likely debris-covered glaciers, due to the similarities in their gross morphology and surface textures to aprons with basal detections.

Sow productivity improvements are associated with high energetic demand due to increasing prolificity. The reproductive life and longevity of sows, and the readiness for weaning of the offspring may be impaired when sows loose significant body weight (BW) during lactation. The impact of a multicarbohydrase containing α-galactosidase on a low energy dense lactation diet was evaluated in this study. Two-hundred and eight sows (208 ± 25.2 kg) were blocked by parity and BW to one of four treatments, in which a corn-soybean meal diet was formulated to have varying levels of added fat (0, 1.5%, and 3%) to titrate an energy density model. A fourth treatment replicated the 0% added fat formulation with enzyme supplementation at 250 g/tonne. Sows were weighed individually on entry, post-farrow (by calculation) and at weaning. Daily feed intakes (ADFI) and caloric intake were used for calculation of sow feed efficiency (FE) and caloric efficiency. Litter performance was characterized at birth, and size was standardized within 24h of farrow and within treatment to ensure uniform litter sizes. Average wean weight and pre-weaning mortality were determined. Piglets were weighted individually to study litter weight distribution. Data was analyzed as a randomized completely block design, using sow as the experimental unit, treatment as the main effect, and standardized average weight and litter sizes as covariates where appropriate. BackgroundPerson-centred care (PCC) is being internationally recognised as a critical attribute of high-quality healthcare. The International Alliance of Patients Organisations defines PCC as care that is focused and organised around people, rather than disease. Focusing on delivery, we aimed to review and evaluate the evidence from interventions that aimed to deliver PCC for people with serious physical illness and identify models of PCC interventions.MethodsSystematic review of literature using Preferred Reporting Items for Systematic Reviews and Meta-Analyses guidelines. We searched AMED, CINAHL, Cochrane Library, Embase, Medline, PsycINFO, using the following key concepts: patient/person-centred care, family centred care, family based care, individualised care, holistic care, serious illness, chronic illness, long-term conditions from inception to April 2022. Due to heterogeneity of interventions and populations studied, narrative synthesis was conducted. Study quality was appraised using the Joanna Briggs checklist.ResultsWe screened n=6156 papers. Seventy-two papers (reporting n=55 different studies) were retained in the review. Most of these studies (n=47) were randomised controlled trials. Our search yielded two main types of interventions: (1) studies with self-management components and (2) technology-based interventions. We synthesised findings across these two models:Self-management component: the interventions consisted of training of patients and/or caregivers or staff. Some studies reported that interventions had effect in reduction hospital admissions, improving quality of life and reducing costs of care.Technology-based interventions: consisted of mobile phone, mobile app, tablet/computer and video. Although some interventions showed improvements for self-efficacy, hospitalisations and length of stay, quality of life did not improve across most studies.DiscussionPCC interventions using self-management have some effects in reducing costs of care and improving quality of life. Technology-based interventions improves self-efficacy but has no effect on quality of life. However, very few studies used self-management and technology approaches. Further work is needed to identify how self-management and technology approaches can be used to manage serious illness.PROSPERO registration numberCRD42018108302.

Introduction Osteoporosis is common in later life, leading to fragility fractures associated with increased mortality, disability, and costs. There is a surprising lack of data regarding the incidence of osteoporosis. We aimed to estimate the incidence of recorded diagnosis of osteoporosis, osteopenia, and fragility fracture in older people, explore time trends in diagnosis, and differences by age, sex, and social deprivation. Method We used de-identified patient data provided as part of routine primary care (IQVIA Medical Research Database (IMRD). All patients aged 50-99y registered with THIN (The Health Improvement Network) participating practices between 1/1/2000–31/12/2018 were included. Crude incidence rates (IR) were estimated per 10,000 person-years (PY). We used Poisson regression to calculate adjusted Incidence Rate Ratios (IRR) accounting for sex, age, calendar year and deprivation. Results The IR of osteoporosis was significantly higher in women, 84.32 (95%CI 83.81–84.83) vs. 16.66 (95%CI 16.43–16.90) in men per 10,000PY. In women, recorded IR of osteoporosis reached a peak in 2009. In the adjusted model, older men in most deprived areas had a higher IRR of osteoporosis [1.67 (95% 1.59–1.74)] compared to those in least deprived areas. Women were more likely to be diagnosed with osteopenia compared to men, at any age. Incidence of osteopenia diagnosis increased over time. In the adjusted model, men in most deprived areas had a higher IRR of osteopenia [1.44 (95%CI 1.35–1.53)] compared to least deprived areas. The IR of fragility fracture was higher in women, 84.97 (95%CI 84.45–85.48) vs. 31.15 (95%CI 30.83–31.48) in men per 10,000PY. In the adjusted model, men in most deprived areas had an increased IRR of fragility fracture [1.53 (95%CI 1.48–1.59)] compared to least deprived areas. Conclusion Community bone health interventions might be targeted at populations at higher risk of fragility fractures, including older men living in socially deprived areas.

Bovine leukemia virus (BLV) is a retrovirus that causes malignant B-cell lymphoma in up to ten-percent of infected cattle. To date, the mechanisms of BLV linked to malignant transformation remain elusive. Although BLV-encoded miRNAs have been associated with the regulation of different genes involved in oncogenic pathways, this association has not been evaluated in cattle naturally infected with BLV. The objective of this study was to determine the relative expression of BLV-encoded miRNA blv-miR-b4-3p, the host analogous miRNA bo-miR-29a and a couple of potential target mRNAs (HBP-1 and PXDN, with anti-tumorigenic function in B-cells), in cattle naturally infected with BLV compared to uninfected animals (control group). We observed that PXDN was significantly downregulated in BLV-infected cattle (P = 0.03). Considering the similar expression of endogenous bo-miR-29a in both animal groups, the downregulation of PXDN in BLV-naturally infected cattle could be linked to blv-miR-b4-3p expression in these animals. Knowing that PXDN is involved in anti-tumoral pathways in B-cells, the results presented here suggest that blv-miR-b4-3p might be involved in BLV tumorigenesis during natural infection with BLV in cattle.

The ice-cored Galena Creek Rock Glacier, Wyoming, USA, has been the subject of a number of studies that sought to determine the origin of its ice. We present new observations of the rock glacier's internal structure from ground-penetrating radar to constrain ice and debris distribution and accumulation. We imaged dipping reflectors in the center of the glacier that are weak and discontinuous, in contrast to strong reflectors toward the edge of the cirque beneath large debris-avalanche chutes. These reflectors form a network of concave-up, up-glacier dipping layers. We interpret these as englacial debris bands formed by large debris falls buried by subsequent ice and snow accumulation. They are discontinuous where ice outpaces debris accumulation, but with sufficient debris accumulation an interleaved pattern of ice and debris layers can form. We propose a model in which the ice in these interleaved layers is snowfall preserved by debris-facilitated accumulation. Large debris falls that occur in early spring bury sections of the snowpack, which are then preserved through summer and incorporated into the rock glacier body over time. This study highlights the importance of sequential accumulation of ice and debris for understanding the dynamics of rock glaciers and debris-covered glaciers.

The marine, mudstone-dominated Hareelv Formation (Upper Jurassic) of Jameson Land, East Greenland is a representative of the widespread Kimmeridge Clay Formation equivalents, sensu lato, known from the greater North Atlantic region, western Siberia and basins off eastern Canada. These deposits constitute the most important petroleum source-rock succession of the region. The present study reports petroleum geochemical data from the 233.8 m thick succession penetrated by the fully cored Blokelv-1 borehole, and includes supplementary data from outcrop samples and other boreholes in Jameson Land. The succession consists of basinal mudstone intercalated with a significant proportion of gravity-flow sandstones, both in situ and remobilised as injectites. The mudstones are generally rich in organic carbon with values of TOC reaching nearly 19 wt% and high pyrolysis yields reaching values of S2 up to nearly 43 kg HC/ton. Hydrogen Indices are up to 363. The data presented herein demonstrate that weathering of abundant pyritic sulfur adversely affects the petroleum potential of the kerogen in outcrop samples. The succession is thermally immature to early mature, except where intrusions have locally heated adjacent mudstones. The documentation of rich gas/oil-prone Upper Jurassic successions in Jameson Land is important for the assessment of the regional petroleum potential, including the North-East Greenland continental shelf.

Modern lactating sows are under high energetic demand due to increasing prolificity. Significant body weight (BW) losses during lactation may impair sows’ reproductive life and longevity, and the readiness for weaning of the offspring. This study aimed at evaluating the impact of a multicarbohydrase containing alpha-galactosidase (CAG; AlphaGal™280P, Kerry) on a low energy dense lactation diet. Two-hundred and eight sows (218 ± 25.2kg) were blocked by parity and BW to one of 4 treatments, in which a corn-soybean meal diet was formulated to have graded levels of added fat (0, 1.5% and 3%) to titrate an energy density model (T1–T3). T4 replicated the 0% added fat formulation with CAG supplementation at 250 g/MT. Sows were weighed individually on entry, post-farrow (by calculation) and at weaning. Daily feed intakes (ADFI) were used for calculation of sow feed conversion ratio (FCR). Litter performance was characterized at birth, and size was standardized within 24h of farrow and within treatment to ensure uniform litter sizes (LS). Average wean weight (WWt) and pre-weaning mortality were determined. Litter weight distribution was also evaluated via individually weighed piglets, with consideration for pigs < 4.1kg BW. Data were analyzed as a RCBD, using sow as the experimental unit, treatment as the main effect, and standardized average weight and LS as covariates where appropriate. CAG tended to increase sow ADFI (P < 0.10) and yielded significant improvements in sow FCR (P < 0.01), comparable to T3. Although CAG group had lower standardized LS (P < 0.001), WWt was higher and equivalent to T3 upon use of standardized LS as a covariate. CAG tended to reduce the proportion of light pigs within the litter, when compared to T1. CAG improved the efficiency of sows, while increasing WWt of the offspring, suggesting an improvement in nutrient digestion and/or post absorption metabolic efficiency from typical lactation diets.

Mars Polar Science is an integrated, compelling system that serves as a nearby analogue to numerous other planets, supports human exploration, and habitability. Mars possesses the closest and most easily accessible layered ice deposits outside of Earth, and accessing those layers to read the climate record would be a triumph for planetary science.

This paper emphasizes the importance of using terrestrial analogues to improve our understanding of the role of ice on Mars through its associated landforms. We discuss terrestrial regions and techniques that can help understand Martian icy environments, and highlight the necessity to explore the Martian cryosphere as the next natural step.

Multiple nations and private entities are pushing to make landing humans on Mars a reality. The majority of proposed mission architectures envision ‘living off the land’ by leveraging Martian water-ice deposits for fuel production and other purposes. Fortunately for mission designers, water ice exists on Mars in plentiful volumes. The challenge is isolating accessible ice deposits within regions that optimize other preferred landing-site conditions. Here we present the first results of the Mars Subsurface Water Ice Mapping (SWIM) project, which has the aim of searching for buried ice resources across the mid-latitudes. Through the integration of orbital datasets in concert with new data-processing techniques, the SWIM project assesses the likelihood of ice by quantifying the consistency of multiple, independent data sources with the presence of ice. Concentrating our efforts across the majority of the northern hemisphere, our composite ice-consistency maps indicate that the broad plains of Arcadia and the extensive glacial networks across Deuteronilus Mensae match the greatest number of remote-sensing criteria for accessible ice-rich, subsurface material situated equatorwards of the contemporary ice-stability zone. The Mars Subsurface Water Ice Mapping (SWIM) project aims at determining the regions where near-surface ice is most likely to be present, according to the combination of all the available datasets. Focusing on the northern mid-latitudes, they identify in particular Deuteronilus Mensae and Arcadia Planitia as promising sites.

Primiparous sows may have difficulty consuming sufficient feed during lactation to account for energy lost in milk production, development, and body maintenance. Addition of higher energy and/or higher standardized ileal digestible (SID) Lys may improve gilt and litter performance. One hundred seventy-two primiparous DNA F1 females were randomly allotted within breeding/farrowing group, previous treatment, and entry weight at farrowing to one of four dietary treatments. Treatments were arranged in a 2 x 2 factorial with energy (3186 and 3332 kcal/kg ME) and SID lysine (1.15 and 1.30%). Sows were weighed upon entry and exit of the farrowing room. Feed disappearance was calculated as the total amount of feed provided to the sow during lactation. A post-farrow weight model was utilized to assess true weight loss in lactation. Litters were standardized within 24 h of birth, and individual pig birth and weaning weights were recorded. The wean to estrus interval was also measured. Data were analyzed as a randomized complete block design with the fixed effect of treatment and the random effect of replicate. Addition of high energy and/or high lysine did not impact (P > 0.10) piglet wean weight or sow average daily feed intake. High energy diets reduced (P < 0.05) sow weight loss post-farrow by 15.5% from 8.75 to 7.39 kg. High SID lysine diets reduced (P < 0.05) number of sows not expressing estrus by day 9 post-wean from 13.0 to 7.7% of sows eligible for insemination. The high SID lysine diets targeted 70 g/d and led to an increase (P < 0.05) of 10.3% of sows displaying estrus on day 4 and 5. These data show that higher levels of SID lysine to target 70 g/d in lactation can improve wean to estrus interval, and potentially decrease non-productive sow days.

Assessment of thermal maturity by solid bitumen reflectance measurements in vitrinite-lean or vitrinite-barren shales is widely used in unconventional shale gas and shale oil plays but the method is commonly challenged by the complexity in the measurements. The Upper Jurassic marine Vaca Muerta shale in the Neuquén Basin in western Argentina is both a prolific conventional source rock and a primary unconventional reservoir for oil and gas. The shale is rich in solid bitumen and lean or barren in vitrinite and offers an excellent opportunity to gain insight into thermal maturity assessment of a shale with a complex organic matter composition. The kerogen composition of a set of immature to highly mature Vaca Muerta shale samples was investigated by organic petrography and reflectance measurements were taken on vitrinite and solid bitumen. The results were integrated with available Rock-Eval data and stable carbon isotope values of shale gas (ethane and propane) collected at the same depth as the rock samples. The maceral composition of immature Vaca Muerta shale is highly sapropelic and oil-prone with Hydrogen Index values of 600 mg HC/g TOC or higher. The liptinite macerals consist primarily of alginite with minor amounts of inertinite and/or oxidized vitrinite. Indigenous vitrinite is generally scarce and in some samples absent. Vitrinite reflectance values range from 0.79%Ro to more than 1.94%Ro corresponding to early peak oil to dry gas maturity. Solid bitumen is abundant in all mature samples. Two general types of solid bitumen morphology were observed: (i) BR I, a dark reflecting porous variant disseminated in the clayey mineral matrix and (ii) BR II, a lighter reflecting more ‘dense’ textured variant. Bands of denser solid bitumen were commonly seen ‘flowing’ in more porous or weak zones in the matrix and around mineral grains. Degassing pores were frequently observed. Solid bitumen reflectance measurements on BR I and BR II provided two populations with different reflectance ranges. The correlation between vitrinite and BR II reflectances is considered to have highest confidence. The correlation can be expressed by the equation VReqv = (BR II + 0.3043)/1.026. The established VReqv-BR II correlation yields results comparable to other published correlations but also demonstrates significant discrepancy to others. This warrants caution when assessing the maturity of complex unconventional shale gas and oil plays by translating solid bitumen reflectance values into vitrinite reflectance equivalents using correlations not calibrated to that specific shale play. Both Hydrogen Index values and ethane and propane stable carbon isotope values of Vaca Muerta shale gas show strong correlations to VReqv derived from BR II. Thus, in cases with absence of vitrinite and questionable solid bitumen measurements reasonable maturities (vitrinite reflectance equivalents) can be inferred from these relationships.

The end-Triassic mass extinction is considered to have been caused by voluminous and repeated emissions of CO2 and/or methane and other gases from magmatic activity in the Central Atlantic Magmatic Province. Despite improved geochronological dating and correlation between the magmatic activity and the extinctions, exactly how the biotic crisis commenced remains poorly understood. Here, we compile palynological and palaeobotanical data, bulk organic δ13C, biomarkers, mercury and other geochemical proxies, charcoal, and sedimentology, from a Rhaetian terrestrial succession in southern Sweden. Our results provide an insight into the climatic, environmental and ecosystem changes that took place at the onset of the mass extinction event.

Background Diagnosis of inflammatory Bowel disease (IBD) can be challenging and time consuming. There is substantial variation in the reported time to diagnosis and the impact of diagnostic delay on disease course. We therefore conducted a systematic review to evaluate the length of time to diagnosis and a meta-analysis of the impact of delay on the disease course of IBD. Methods The Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines were followed, and final papers were assessed for risk of bias. We searched MEDLINE and Embase for retrospective and prospective studies, published up to December 2020, that reported diagnostic interval, from the time of symptom onset or first consultation to a confirmed diagnosis of IBD, in adults (age ≥18 years). We extracted data from published studies and calculated the weighted median and interquartile range (IQR) of the reported diagnosis interval. Delay was defined as a time period greater than the 75th centile. From eligible studies, we extracted risk estimates and, using random-effects meta-analysis, calculated the pooled odds ratios (OR) and 95% confidence intervals (CI) of studies that examined the association of diagnostic delay with subsequent clinical outcomes, including disease phenotype and surgical intervention. Statistical heterogeneity was assessed, I2 >50% indicating substantial heterogeneity. Results Of 1,533 studies identified, 28 fulfilled the eligibility criteria, including 25,764 patients diagnosed with IBD: Crohn’s disease (CD) = 13,206 and ulcerative colitis (UC) = 12,558. In CD, the pooled weighted median for the diagnostic interval was 8.8 months (IQR 3.0–61.1 months). In UC, the pooled weighted median was 15.2 months (IQR 2.4–62.8). Considering studies from high income countries alone, the pooled weighted median was 6.4 months (IQR 1.7–46.7) in CD, and 2.5 months (IQR 0.3–23.0) in UC. 11 studies were eligible for inclusion in the meta-analysis, representing 6,403 patients diagnosed with IBD (CD; n = 4,902 and UC; n = 1,501). In CD, pooled analysis showed an association between delayed diagnosis and stricturing disease (OR:1.77, CI 1.28–2.46; I2 =61%), penetrating disease (OR: 1.48, CI 1.21–1.82; I2=0%) and intestinal surgery (OR:1.88, CI 1.45–2.44; I2 =34%; figure 1a). In UC, delayed diagnosis was associated with an increased risk of colectomy (OR:4.13, CI 1.04–16.40; I2 =0%; figure 1b). Conclusion Measures are required to reduce delay in the diagnosis of IBD, which is associated with an increase in stricturing and penetrating disease phenotype in CD and a substantially increased risk of surgery in both CD and UC.

This study investigates the extra UV fluence needed to meet the International Maritime Organisation's ballast water discharge standards for the 10–50 μm size-class using the approved vital stain (VS) method compared to the Most Probable Number (MPN) method for organism viability assessment. Low- and medium pressure UV collimated beam treatments were applied to natural algae collected in temperate and tropical water environments and analysed using both methods. About 10 times higher UV fluence was required to meet discharge standards when using VS compared to MPN. Implementing a dark-hold period after UV treatments decreased algal viability. Length of dark-hold period to meet discharge standards decreased with increasing UV fluence. No significant differences between temperate and tropical samples were observed. The onshore Mangyshlak Basin, located in western Kazakhstan along the eastern coastline of the Caspian Sea, is a prolific hydrocarbon province and home to numerous oil and gas fields. The primary reservoirs are Jurassic clastic sandstones, but the Dunga Field contains hydrocarbons in Lower Cretaceous (Valanginian, Barremian, Aptian, Albian) clastic and carbonate reservoirs. The general perception is that these petroleum accumulations were charged from marine source rocks of likely Middle Triassic age. A detailed organic geochemical investigation of oils from the Cretaceous reservoirs of Dunga Field suggests that all were generated from the same lacustrine source rock facies (organofacies C) in mature kitchens in the offshore Mangyshlak Basin, followed by a complex filling history. Weichteilsarkome sind mit weniger als 1 % aller Krebserkrankungen selten. In Deutschland wurden in den letzten Jahren zwischen 3500 und 3750 Neuerkrankungen pro Jahr registriert. Es lassen sich grob 2 Gruppen unterteilen: Entitaten mit einzelnen, gut definierten Treibermutationen und Sarkome mit einem komplexen Genotyp. Als Risikofaktoren gelten eine genetische Pradisposition wie die Neurofibromatose, radioaktive Strahlung, infektiose Agenzien, Umweltgifte und chronische Entzundungen. Daneben konnen insbesondere fibroblastische Tumoren mit einem Trauma assoziiert sein. Die vorliegende Ubersichtsarbeit basiert auf der mehrjahrigen Erfahrung des Erstautors als Leiter des Referenzzentrums fur Weichteiltumoren am Institut fur Pathologie des Universitatsklinikums Jena und prasentiert aktuelle Daten zur Epidemiologie sowie zur pathologischen, genetischen und epigenetischen Klassifikation von Sarkomen. Daruber hinaus werden die technischen Entwicklungen der Sarkomdiagnostik mittels Methylom- und Genkopienzahlanalytik beleuchtet sowie die Moglichkeit illustriert, wie sich Sarkome anhand ihres Methylierungsmusters klassifizieren lassen. Diese Methodik wird voraussichtlich zusammen mit der massiven parallelen DNA-Sequenzierung Einzug in die Sarkomdiagnostik halten. Dem Pathologen wird zukunftig eine wichtige Rolle in der Interpretation dieser Daten im Kontext seiner konventionellen, auf histomorphologischen und immunhistologischen Befunden basierenden Klassifikation zukommen. Neben der Kenntnis der vielen Tumorentitaten bedarf es dabei eines Verstandnisses der molekularen und zellularen Mechanismen im Sinne einer Pathway-Pathologie. Es ist zu hoffen, dass sich daraus neue translationale Ansatze bei der Behandlung dieser aktuell nur schwer medikamentos therapierbaren Tumoren ergeben werden.

The Volgian–Ryazanian shale interval in the Norwegian Central Graben, North Sea, is assigned to the Mandal Formation. Despite shales with similar lithological and log characteristics are being present in the Danish Central Graben, the formation is not defined for this part of the Central Graben. This paper presents a new lithostratigraphic subdivision of the Upper Volgian–Upper Ryazanian interval in the Danish Central Graben, where the Mandal Formation is defined and replaces the uppermost part of the Farsund Formation in the ‘old’ lithostratigraphy. The Mandal Formation is recognized from its log character and is correlatable across the Danish Central Graben. The formation typically has high gamma-ray (GR) and resistivity readings accompanied by low velocity and density values. In basin centres it normally consists of a lower part showing an upward increase in GR readings (‘warming-up’ interval), an overlying interval with conspicuous high GR readings (Bo Member), and an upper heterogeneous shale interval. The base of the Mandal Formation can be seismically mapped in most of the Danish Central Graben. The upper boundary of the formation, corresponding to the Base Cretaceous Unconformity (BCU) at the transition to the Lower Cretaceous, can likewise be seismically mapped throughout the basin. The Mandal Formation is developed with thick successions in basin depocentres, while it is thin or absent along inversion structures and on structural highs. The organic-rich shales of the Mandal Formation were deposited in a restricted, low-energy, oxygen-deficient/anoxic marine environment distal from any significant terrestrial influence. Along the eastern Coffee Soil Fault in the Danish Central Graben, siliciclastic deposits constituting the gravity-flow sandstones of the Poul Member interfinger with the Mandal Formation. Gravity-flow deposits formed by turbidite currents are also common throughout the Mandal Formation in basin centres. The Mandal Formation and particularly the Bo Member are highly oil-prone and contain the best source rock quality of the entire Upper Jurassic – lowermost Cretaceous shale succession in the Danish Central Graben. Interbedded potential reservoir sandstones suggest the Mandal Formation may in places be considered as an interval with hydrocarbon prospectivity, especially along the Coffee Soil Fault.

The Jude-1 well is located in the central part of the Danish Central Graben, North Sea, and was drilled into immature black shales of the Upper Jurassic–lowermost Farsund Formation, a world-class marine source rock. The well was drilled with oil-based drilling mud (OBM). The lithofacies consists of argillaceous shale, calcareous shale and dolomite stringers as confirmed by cuttings and core. A core was taken in the upper part of the formation at the beginning of a ‘hot’ argillaceous shale interval characterized by elevated TOC and HI values. Cuttings and core samples were investigated to unravel kerogen composition, source rock quality and the contamination effect of OBM on organic geochemical analyses. The organic facies of the source rocks corresponds to Type II kerogen or Organofacies B (sensu Pepper and Corvi, 1995). Considerable fluctuations in HI values through the Farsund Formation reflect pronounced variations in source rock quality, although a faint lamination of the shales and overall high TOC contents testify to prevailing oxygen-deficient depositional conditions preventing significant bioturbation and reworking of the sediment and organic matter. Petrography of core samples reveals a relatively homogenous sapropelic kerogen composition dominated by a groundmass of yellowish fluorescing amorphous organic matter and liptodetrinite intimately associated with the mineral matrix. Telalginite is less abundant, but Tasmanites- and in particular Leiosphaerida-type telalginites were observed. Minor but varying amounts of detrital terrigenous macerals suggest a considerable distance to land areas and thus limited supply of land plant-derived organic matter. The argillaceous shale lithofacies generally is more oil-prone and organic-rich than the more gas-prone calcareous shale facies. The uppermost section includes the ‘hot’ argillaceous shales, which together with two deeper argillaceous shale intervals are highly organic-rich and oil-prone. Average TOC of the ‘hot’ argillaceous shales is ~ 7 wt% and HI reaches > 500 mg HC/g TOC. The total Ultimate Expulsion Potential (UEP) of the over 853 m (2799 ft) thick Farsund Formation is ~ 142 mmboe/km2. Results from non-contaminated samples are thus consistent with the Farsund Formation being a world-class highly oil-prone marine source rock. The predicted oil composition corresponds to ‘paraffinic low wax oils’. The adverse effect of OBM contamination on geochemical analyses is demonstrated by Rock-Eval data and measured bulk kinetics of contaminated core samples. The measured kinetics has a considerable proportion of low activation energies related to OBM contamination which is also supported by high Production Indices. Further, gas chromatograms of the saturated fraction of extracts from contaminated cores clearly show evidence of OBM in the nC11–nC14 range. Rock-Eval S2 peaks have a ‘shoulder’ showing that the low Ea-peaks result from carry over from the S1 peak, caused by the OBM contamination. As a consequence Hydrogen Index values are increased.

The objective was to determine a dietary strategy for gilt development that would improve reproductive performance. One thousand eighty-three gilts (DNA) were blocked by weight (average initial weight = 27.5 ± 4.8 kg) and allocated to one of four dietary treatments (267-273 gilts per treatment). Treatments included: 1) standard grower/finisher (SGF) diet; 2) maximize bone growth (MB) diet with increased available P and Ca; 3) maximize growth (MG) diet with increased energy and AA levels, or 4) maximize both bone and growth (MBG) diet. Each treatment had five phases with all gilts fed a common gestation diet after insemination. At 20 weeks of age, gilts fed MG and MBG diets were heavier (P < 0.05) than gilts fed other diets. Gilts fed the MG diet were heavier (116.9 kg; P < 0.05) at 23 weeks (start of boar exposure) than gilts fed other diets (SGF, 112 kg; MB, 110.3 kg; MBG, 114.6 kg). Gilts fed the MG diet were heavier (143.8 kg; P < 0.05) at mating (29 weeks of age) than gilts fed other diets (SGF, 139.0 kg; MB, 136.8 kg; MBG, 139.6 kg). Upon entering farrowing stalls, gilts fed the MG diet continued to be heavier (P < 0.05). More gilts farrowed (P < 0.05) when fed the MG diet (MG, 89.8%; MBG, 86.8%; MB, 82.2%; SGF, 81.9%). Dietary treatment had no impact on number of pigs born alive per litter, total pigs born per litter, or piglet birth and weaning weights. There was a tendency (P < 0.10) for gilts to remain in the sow herd to farrow their second litter when fed either the MG or MBG diet. Overall, feeding higher energy and AA levels to maximize growth increased gilt development weights, improved the number of gilts that farrowed, and tended to increase sow retention.

Understanding of the processes of petroleum source rock (SR) accumulation in lacustrine rift basins and the behavior of lake systems as long-term carbon sinks is fragmentary. Investigation of an 800 m thick (500 m core and ~ 300 m outcrop), deep-lacustrine, Oligocene section in Vietnam, provides a rare insight into the controls and deposition of organic carbon (OC) and SR formation in continental rift basins. A multidisciplinary dataset, combining elemental data, inorganic and organic geochemistry with sedimentology, shows that the richest alginite-prone, sapropelic SR developed during periods of relative tectonic quiescence characterized by moderate primary productivity in a mainly dysoxic lacustrine basin. Increased rift activity and further development of graben morphology intensified water column stratification and anoxia, which hindered nutrient recycling. Sapropelic organic matter (OM) continued to accumulate, but with increasing amorphous OM content and decreasing total OC values. Periods of increased seasonality were characterized by thermocline weakening, enhanced mixing of water columns, increased primary productivity and diatom blooming. The results suggest that a change from dysoxia towards anoxia or extreme primary productivity does not necessarily enhance OC burial and SR quality. External nutrient input from a phosphate-rich hinterland is sufficient for sapropel formation, whereas the main limiting factor is methanogenesis.

To challenge ballast water treatment system (BWTS) efficacy for organisms in the size-class 10–50 μm, intake concentration during tests must reach certain minimum requirements. Often, natural concentrations are too low to meet intake requirements and standard test organisms (STOs) are added. We tested the robustness of Tetraselmis suecica and Odontella sp. to a range of UV-treatments to explore fluences needed to meet the IMO discharge standard (−1) evaluated using two viability assessment methods. To meet discharge standards, fluences of >1000 mJ cm−2 were required using vital stain whereas 135–500 mJ cm−2 were needed using regrowth assays. Besides, results suggest that T. suecica and Odontella sp. were at least as robust as natural algae towards UV-treatments. We suggest the advantageous use of these species as STOs in test water to support intake water requirements and to obtain more conservative validation of UV-based BWTS to ensure more environmental protective procedures.

The ESCRT-III membrane fission machinery maintains the integrity of the nuclear envelope. Although primary nuclei resealing takes minutes, micronuclear envelope ruptures seem to be irreversible. Instead, micronuclear ruptures result in catastrophic membrane collapse and are associated with chromosome fragmentation and chromothripsis, complex chromosome rearrangements thought to be a major driving force in cancer development. Here we use a combination of live microscopy and electron tomography, as well as computer simulations, to uncover the mechanism underlying micronuclear collapse. We show that, due to their small size, micronuclei inherently lack the capacity of primary nuclei to restrict the accumulation of CHMP7–LEMD2, a compartmentalization sensor that detects loss of nuclear integrity. This causes unrestrained ESCRT-III accumulation, which drives extensive membrane deformation, DNA damage and chromosome fragmentation. Thus, the nuclear-integrity surveillance machinery is a double-edged sword, as its sensitivity ensures rapid repair at primary nuclei while causing unrestrained activity at ruptured micronuclei, with catastrophic consequences for genome stability. acceptedVersion
