Apical parts of stems of Brassica napus L. var. oleifera cv. Gorczanski (winter rape) and cv. Mlochowski (spring rape), grown in vitro, were subjected to direct electric current (DC) of different polarity, duration and voltage. The positive orientation of DC, i.e. anode attached to the apical part and cathode to the medium, markedly enhanced the differentiation of the apical meristem in winter rape. The reverse polarity was without effect. DC treatment of positive polarity resulted in spring rape in transition of all explants to generative state while 70 % of non-treated plants remained at vegetative stage. Even negative orientation of DC brought about a rise in percentage of flowering plants with regard to control. The contents of endogenous free and conjugated polyamines, putrescine (Put) and spermidine (Spd), were determined during 9 week of vernalization (at 5 °C) in winter wheat seedlings cultivated on Murashige and Skoog media without (MS0) and with 2 mg dm-3 zearalenone (MSZEN). At the 4th week of chilling treatment, which is sufficient to induce generative development in 30 % of plants, the marked increase in free and conjugated forms of Put and free Spd were observed. The presence of ZEN in medium significantly accelerated the vernalization. About 20 % of plants treated with ZEN flowered already after 2 weeks and 40 % after 3 weeks of chilling. Significantly higher content of free Put was determined in roots grown on MSZEN compared with MS0 during the first 5 weeks of vernalization with maximum at the 4th week. After germination, a marked decrease in free Spd content was observed both in plants grown on MS0 and MSZEN. Application of ZEN significantly slowed down the Spd decline in leaves and roots during the first and second week of vernalization. The content of Spd and its conjugates decreased in vernalized plants after 1 week of cultivation at 20 °C.

The cultivated Brassica species include numerous vegetable and oil crops of global importance. Three genomes (designated A, B and C) share mesohexapolyploid ancestry and occur both singly and in each pairwise combination to define the Brassica species. With organizational errors (such as misplaced genome segments) corrected, we showed that the fundamental structure of each of the genomes is the same, irrespective of the species in which it occurs. This enabled us to clarify genome evolutionary pathways, including updating the Ancestral Crucifer Karyotype (ACK) block organization and providing support for the Brassica mesohexaploidy having occurred via a two-step process. We then constructed genus-wide pan-genomes, drawing from genes present in any species in which the respective genome occurs, which enabled us to provide a global gene nomenclature system for the cultivated Brassica species and develop a methodology to cost-effectively elucidate the genomic impacts of alien introgressions. Our advances not only underpin knowledge-based approaches to the more efficient breeding of Brassica crops but also provide an exemplar for the study of other polyploids.

The role of ethylene and auxin in stigma-to-ovule signalling was investigated in maize (Zea mays L.). Maturation of the egg cells in an ear was stimulated before actual fertilization by the application of fresh pollen grains or quartz sand to fully receptive stigmas. Ethylene emission by maize ears increased in response to those treatments. Silks and ovaries were involved in ethylene synthesis after pollen or sand was shed over the silks. The content of ethylene precursor [1-aminocyclopropane-1-carboxylic acid (ACC)] increased in both pistil parts soon after pollination. ACC rise was delayed by 4 h in the ovaries, and by 8 h in the silks after mock-pollination with sand. The auxin level increased rapidly in the silks and ovaries after pollination, and it was very high in the pollinated silks due to the high indole-3-acetic acid (IAA) content of pollen grains. IAA rise also appeared in the silks and ovaries after treatment with sand but it was delayed by 8 h. Application of ACC (10 microM) or IAA (6 microM) solutions to non-pollinated silks stimulated maturation of the egg cells. Moreover, the response of the egg cells to pollination was cancelled by l-alpha-(2-aminoethoxyvinyl)-glycine, alpha-aminoisobutyric acid or 2,3,5-triiodobenzoic acid applied to the silks before pollination. Thus ethylene synthesis and polar auxin transport in the silks pollinated with fresh pollen were necessary to evoke accelerated differentiation of the egg cells in maize ovules. Differences in pistil responses found between true- and mock-pollination suggest that signalling pathways are at least partially different for the reception of pollen grains and sand crystals on maize stigma.

Ethylene formation was studied in 5- to 6-d-old Chenopodium rubrum seedlings under the following light regimes: continuous light (CL), continuous darkness (CD), and alternating light/darkness (12 h of each). No significant regular oscillations in ethylene formation were found in either the CL or CD groups. In the light/dark regime, pronounced diurnal fluctuations in ethylene formation were observed. Activity of 1-aminocyclopropane-1-carboxylic acid (ACC) oxidase was transiently increased on transfer from light to dark and vice versa. In CL, ACC oxidase activity did not change significantly, whereas in CD, it decreased continuously after the initial increase. The in vivo levels of ACC and N-malonyl-ACC (MACC) were constant for the first few hours of darkness, then decreased dramatically, but increased again in the light. In constant darkness, the level of ACC displayed endogenous rhythm, with minimum values at h 12 and 44, and a maximum value at h 32 to 36. The level of MACC in both shoots and roots decreased in the CD group until h 12, and then remained constant until h 30 before decreasing continuously. We conclude that the photoperiodic regime affects both ACC and MACC levels, as well as the conversion of ACC to ethylene. Correlation of the described changes in ethylene formation to photoperiodic flower induction is discussed.
