Modulation of mast-cell activation may provide novel ways to control allergic diseases. Here, we show that protein tyrosine phosphatase epsilon (PTPepsilon; Ptpre) plays key regulatory roles during mast-cell activation mediated by the high-affinity IgE receptor (FcepsilonRI). Bone marrow-derived mast cells (BMMC) from Ptpre(-/-) mice exhibited enhanced FcepsilonRI-induced Ca(2+) mobilization and mitogen-activated protein kinase (MAPK) (JNK and p38) activation, and showed corresponding enhancement of evoked degranulation and cytokine production, but not leukotriene production. Examination of proteins linking tyrosine kinase activation and Ca(2+) mobilization revealed that the absence of PTPepsilon leads to increased phosphorylation of the linker for activation of T cells and SH2 domain-containing leucocyte phosphoproteins of 76 kDa, but not Grb2-associated binder-2 (Gab2). Because Gab2 is considered to be situated downstream of Fyn kinase, we reasoned that Fyn may not be a target of PTPepsilon. In the event, Syk but not Lyn was hyperphosphorylated in PTPepsilon-deficient BMMC. Thus, PTPepsilon most likely exerts its effects at the level of Syk, inhibiting downstream events including phosphorylation of SLP-76 and linker of activated T cells and mobilization of Ca(2+). Consistent with the in vitro data, antigen- and IgE-mediated passive systemic anaphylactic reactions were augmented in Ptpre(-/-) mice. Given that the number of mast cells is unchanged in these mice, this observation most likely reflects alterations of mast cell-autonomous signalling events. These data suggest that PTPepsilon negatively regulates FcepsilonRI-mediated signalling pathways and thus constitutes a novel target for ameliorating allergic conditions.

Experimental studies on in-beam γ-ray spectroscopy using a 12Be radioactive beam are presented. Inelastic scattering of the neutron-rich 12Be nucleus on 208Pb, 12C, and (CH2)n targets has been studied by measuring de-excitation γ-rays in coincidence with scattered particles. The level schemes and transition probabilities are determined for low-lying excited states in 12Be. The present paper presents a brief review of the spectroscopic results, which may be associated with the N = 8 shell quenching near the drip line.

Inelastic scattering of the neutron-rich nucleus 12 Be on lead and carbon targets has been studied by measuring de-excitation γ rays in coincidence with scattered 12 Be. The strong γ -ray transition from the state at E x =2.68(3) MeV following E1 Coulomb excitation was observed for the lead target, leading to an assignment of J π =1 − for the excited state. The low excitation energy of this intruder 1 − state and the deduced large B (E1;0 g.s. + →1 − ) value of 0.051(13) e 2 fm 2 provide a consistent picture of the N =8 shell melting in 12 Be.

Inelastic proton scattering exciting the 2 + 1 states in the neutron-rich beryllium isotopes 10,12 Be has been studied in inverse kinematics. From a coupled-channel analysis, the deformation lengths for the 2 + 1 states in 10 Be and 12 Be were determined to be 1.80±0.25 fm and 2.00±0.23 fm respectively, indicating that a tendency towards strong quadrupole deformation is preserved for these nuclei and that the singly-closed shell structure does not prevail in 12 Be. A quantitative analysis based on shell model calculations supports this picture.

The α-decay energy and halflife of 195mAt were determined to be 6960±20 keV and 385+69 −51 ms respectively, on the basis of genetic correlations in the 169Tm(36Ar, α6n)195At reaction, while those of 195gAt measured simultaneously were 7105±30 keV and 146+21 −17 ms respectively, reconfirming the previously reported values. A new isotope 199Fr was also produced and identified in the same way in the 169Tm(36Ar, 6n)199Fr reaction, yielding Eα= 7655±40 keV and T1/2= 12+10 −4 ms.

In the neutron-rich region around N = 8, experimental evidence is being to accumulate that several neutron-rich nuclei, such as 11Li and 11Be, are strongly influenced by lowlying intruder orbitals [1,2]. In this neighborhood, 12Be is a typical neutron-rich nucleus with N = 8 , where the effects of shell quenching are most likely to be manifested. Since properties of low-lying states in even-even nuclei are sensitive to the modification of the nuclear shell structure, we have studied the low-lying E1 and E2 transitions of 12Be via inelastic scattering.

We have performed an experiment of Coulomb excitation of a doubly magic nucleus 56Ni to its first excited 2+ state at 2.7 MeV in inverse kinematics. Radioactive 56Ni particles at an incident energy 70.7 MeV/nucleon with 1500 s−1 was obtained via fragmentation of 58Ni at RIPS facility of RIKEN. The E2 transition probability B(E2) between the 0+ ground state to the first 2+ state was deduced to be 580±70 e2fm4 by a coupled channel analysis assuming Coulomb excitation mechanism. The present result agrees with the value deduced from the 56Ni+p inelastic scattering experiment at GSI.

A novel, rapid, cell-counting method for unfixed frozen brains comprehensively quantifies at least four neural cell populations

A giant protein of apparent molecular weight (Mr) 2000 kDa, as determined by SDS-PAGE, was isolated and partially purified, under denaturing conditions, from the detergent-resistant cytomatrix of unfertilized sea urchin egg. Immunoblot analysis and indirect immunofluorescence microscopy observations indicated that this high-molecular-weight protein cross-reacted with the immunospecific serum raised against chicken breast muscle beta-connectin. However, rotary-shadowing electron microscopy images of the protein revealed short threadlike structures which appear morphologically different from beta-connectin structure. Indirect immunofluorescence localization of the protein with anti-beta-connectin serum showed a distribution throughout the whole unfertilized egg cytomatrix. This immunofluorescence pattern seems to change upon egg fertilization, since at metaphase the fluorescence stain appears to be excluded from the mitotic apparatus region as revealed by the double immunolabeling with anti-beta-connectin serum and monoclonal anti-alpha-tubulin antibody. Moreover, when egg cortical fragments were double-labeled with anti-beta-connectin serum and rhodamin-conjugated phalloidin, it was observed that the microfilaments assembled after fertilization seem to be in close association with the protein at the cleavage furrow and other locations. The possible significance of this sea urchin egg connectin(titin)-like protein is discussed.
