To clarify the characteristic of follicular fluid in mares with granulose theca cell tumor (GTCT) 5 mares from five GTCT affected mares were obtained at the time of removal of affected ovary from different parts in Japan. The 9 ovaries from the 6 normal mares were obtained from the abattoir. The diameter of each follicle in the ovaries was measured and the follicles were classified according to the diameter. The concentration of estradiol -17 ß (E2), ir- INH, pro?-C and INH-A were significantly lower as compared with that of normal ovarian follicular fluid. But the average concentration of INH-B in affected animal was not significantly lower as compared with that of healthy ovarian follicular fluids. According to this study it seems that the large follicles in GTCT affected ovaries were not mature like a healthy follicle. As a result estradiol -17ß (E2), ir- INH, pro?-C and INH-A were significantly lower as compared with that of normal mares.

The structure of the equine ovary is different from that of other mammals in its extremely large size, the presence of ovarian fossa and the inverted location of its cortex and medulla. A three-dimensional internal structure microscopy (3D-ISM), which consists of a computer-controlled slicer, a CCD camera, a laser disc recorder and a PC, is very useful for the observation of the internal structures in equine ovaries. In addition, the three-dimensional images of follicles and corpus luteum (CL) reconstructed by the segmentation technique can clarify the spatial arrangement in the equine ovary. In this study, to understand the changes in the ovarian internal structures of the mare during the oestrous cycle, the size and numbers of follicles and luteal structures were analysed by 3D-ISM in addition to the concentrations of progesterone (P(4)) and oestradiol-17beta. As a result, many small follicles (10 mm in diameter) were detected. It was recognized that the luteal structures were distinguished into three types, such as the corpus haemorragicum (CH), which is formed by blood elements at the cavity after ovulation, CL and corpus albican (CA). There were some CHs and CL in the group, which had the concentration of P(4)1 ng/ml. CHs were also observed in the group, which had low level of P(4) (P(4)1 ng/ml). CAs were found regardless of the P(4) level. In conclusion, 3D-ISM enabled the internal observation of the ovarian structures in detail, and estimation of the stage of the ovarian cycle with complementary physiological information. The findings by 3D-ISM provide basic information for clinical applications.

黒毛和種の種雄牛および種雄牛候補牛計119頭を対象に、染色体分析を実施し、系統における転座保有牛の分布、繁殖能力に及ぼす影響について検討した。染色体分析の結果、雄牛119頭のうち10頭(8.4%)に1/29型(6頭)と7/21型(4頭)の2種類の染色体転座が認められた。系統別にみると5系統中3系統(C系、D系、E系)に転座保有牛の分布が認められ、転座型において、C系、D系では1/29型、D系、E系では7/21型が認められた。1/29型転座を保有する雄牛1頭と正常核型を示す雄牛4頭の精液性状を調べた結果、転座保有牛と正常牛との間に差異は認められなかった。また、試験交配時の受胎率を調べたところ、正常牛群で平均62.5%、転座保有牛では44.1%であり、X2検定の結果、両群間に有意な差(P＜0.05)が認められた。さらに転座保有牛と正常牛の精子の染色体分析を行った結果、不均衡な核型を持つ精子が正常牛では認められなかったのに対し、転座保有牛では114個中5個(4.4%)に認められた。また構造異常精子が正常牛で144個中22個(15.2%)に認められたのに対し、転座保有牛では114個中34個(29.8%)に認められ、X2検定の結果、両群間に有意な差(P＜0.01)が認めれた。以上の結果より、黒毛和種雄牛では、1/29型と7/21型の2種類のロバートソン型転座が広く分布していることが認められた。また各系統での転座保有牛の分布には偏りがみられ、黒毛和種での転座の起源は特定の系統に由来することが考えれた。繁殖能力に及ぼす影響については、精液性状、精子形成に異常が認められていないものの、受胎率の低下が明らかなことから、その原因は転座保有牛に不均衡型や構造異常を示す精子が高率に発生していることに関連していると思われた。

黒毛和種で認められる1/29型転座が同雌牛の繁殖効率に及ぼす影響について調べる目的で、1/29型転座をヘテロで保有している種雄牛の娘牛146頭を対象に染色体分析を行うとともに、繁殖成績の調査を行った。その結果、未経産のリピートブリーダー牛群で31頭中27頭(87.1%)、3回以内の授精で受胎した正常牛群では27頭中15頭(55.6%)に1/29型転座が認められ、この両群間の転座保有率には有意差(p＜0.05)があった。一方、経産牛では88頭中39頭(44.3%)に1/29型転座が認められ、未経産牛群と経産牛群との間には、転座保有率に有意な差(p＜0.01)があった。また、1/29型転座保有種雄牛からの娘牛で、転座を保有する経産牛34頭と正常核型を有する経産牛40頭、および対照として他の種雄牛からの娘牛128頭、計202頭の繁殖成績を調査したところ、転座保有牛群では正常核型を保有する牛群ならびに他の種雄牛からの娘牛群と比べて、受胎までの授精回数の増加と、受胎率の低下が認められた。以上の結果より、1/29型転座は黒毛和種雌牛の繁殖効率に影響を及ぼし、結果として未経産牛において不妊を主訴とする淘汰率の増加に影響を及ぼしていることが示唆された。また経産牛群における転座の保有率は約50%であること、および同群で1/29型転座と繁殖効率の間に明らかな相関が認められなかったことは、転座保有牛が未経産で受胎率低下の理由により淘汰されていることによると推測された。

In conclusion, this study demonstrated that ir-IGF-I is secreted during the oestrous phase of the cycle concomitant with the development of the future ovulatory dominant follicle, and it may act in synergy with other hormones for the selection and differentiation of the dominant follicle.

Aims This study aimed to investigate whether interleukin-18 (IL-18) gene polymorphisms are associated with the development of antibody against the 65-kDa isoform of recombinant human glutamic acid decarboxylase (GAD65Ab) in patients with Graves’ disease. Methods A total of 398 unrelated Japanese patients with Graves’ disease, with and without GAD65Ab, were recruited. Three single nucleotide polymorphisms in the IL-18 gene were examined and the polymorphic allele and the genotype and haplotype frequencies calculated. Results The frequency of the GG genotype at position −4675 of the IL-18 gene was significantly lower in Graves’ disease patients with GAD65Ab than those without (4% vs. 24%, P = 0.0126). The −4675C allele frequency was significantly greater in patients with GAD65Ab than those without (69% vs. 53%, P = 0.0168). The homozygous −4675G/−607A/−137G haplotype was less common in Graves’ disease patients with GAD65Ab than those without (4% vs. 23%, P = 0.0144). Conclusions These findings in a Japanese population indicate that Graves’ disease patients carrying the GG genotype at position −4657 of the promoter of the IL-18 gene or a gene in linkage disequilibrium with the −4675G/−607A/−137G haplotype have a low risk for the development of GAD65Ab in Graves’ disease.

In the proposed system, the disturbance observer is used for the inertia identification as well as for disturbance compensation. The inertia value is obtained by using the orthogonality relation among the torque components of the estimated disturbance torque. An inertia term of both the torque feedforward control and the disturbance observer is automatically readjusted by using the identification. The proposed motion control system consists of four parts: velocity feedback control, inertia torque feedforward control, disturbance observer. and inertia identification part. Experimental results show that precise control is achieved by the proposed motion control system. >

Early pregnancy factor (EPF) is an immunosuppressive protein detected in the early pregnancy serum. We have already reported that we developed the rosette inhibition test for mare EPF and detected EPF in thoroughbreds. The aim of this study was to determine whether or not our method could be used clinically.The rosette inhibition test for equine EPF was carried out on serum from six nonpregnant and six pregnant Shetland ponies, a female and a male Chinese pony, and four nonpregnant and 13 pregnant thoroughbred mares. In the thoroughbreds sera were collected during the pregnancy period. Furthermore, we measured progesterone and detected pregnant mare serum gonadotrophin (PMSG) in order to confirm pregnancy of the Chinese pony 3 and 6 months after mating.In the nonpregnant Shetland ponies, the rosette inhibition titre (RIT) was 6.0+/-1.0 and EPF was negative. In contrast, in the pregnant ponies, the RIT was 9.2+/-0.4 and EPF was positive. Based on these results, we diagnosed pregnancy of the Chinese pony. The RIT of the female Chinese pony (3 months after mating) was above 10 and EPF was positive. Furthermore, we detected PMSG and progesterone in the serum of this pony. EPF appeared in the maternal blood circulation at 24-72 hr after mating, it was detected until the second trimester, and after that it disappeared from the maternal serum.The pony's EPF was detected by using the same rosette inhibition test as in the thoroughbred and was present from 24 to 72 hr after mating until the second trimester. The results indicated that our method was useful for pregnancy diagnosis of Equine.

Percutaneous microwave coagulation therapy (PMCT) has recently been introduced as a new treatment for hepatocellular carcinoma (HCC) in Japan. This study was performed to evaluate its efficacy and safety.Thirteen patients with 17 nodules of unresectable HCC were subjected to PMCT under ultrasonic guidance. The tumors ranged from 1.2-4.4 cm in size. Assessment of the efficacy of PMCT was made by follow-up with dynamic computed tomography (CT).In the patients with small HCC (or = 2.0 cm), 8 of 10 nodules (80%) showed complete remission after PMCT. In small nodules located on the liver surface, 3 out of 4 nodules (75%) showed complete remission. However, in the patients with larger HCC (or = 2.1 cm), 5 out of 7 nodules developed local recurrence after PMCT. Regarding assessment of the necrotic area after PMCT, dynamic CT revealed enhancement that was possibly caused by congestion of the liver parenchyma surrounding the area of necrosis due to PMCT in the early phase of the treatment. Therefore, the necrotic area must be assessed carefully. Although a slight heat sensation and/or pain during microwave irradiation (a common effect of PMCT) occurred in all patients, there were no serious adverse effects.Complete remission of small HCC (or = 2 cm in diameter) can be achieved with PMCT alone, but there seem to be limitations to its effectiveness with larger HCC (or = 2.1 cm). There were no serious adverse effects from PMCT and the therapy can be safely carried out even in patients with poor liver function.

Two cases of severe acute pancreatitis associated with type V hyperlipoproteinemia are reported. A 39-year-old obese woman was hospitalized with continuous severe abdominal pain. The diagnosis was made on the day of admission to our hospital, and treatment using continuous regional arterial infusion of a protease inhibitor and an antibiotic was performed with good results. The other patient was a 35 year-old woman in the 35th week of pregnancy, and a diagnosis of gestational hyperlipidemic pancreatitis was made on the day of onset. She was treated supportively using intravenous hyperalimentation, protease inhibitors, and antibiotics. She recovered from the acute pancreatitis and delivered a healthy term infant. It is difficult to diagnose acute pancreatitis in patients with type V hyperlipoproteinemia, because even when serum amylase levels are high, the value is reduced by high serum triglycerides. Early diagnosis was achieved in both of the present cases, and early intensive therapy was performed, which may be of the utmost importance in saving the life of a patient.

We report a 67 year-old man with residual hepatocellular carcinoma after arterial embolization therapy, which was located on both the anterior and inferior surfaces of segment 6 of the liver. Percutaneous microwave coagulation therapy could be performed safely and the treated tumor became non-enhancing on contrast computed tomography. Two years after treatment, the tumor remains non-enhancing on contrast computed tomography and has decreased in size. Percutaneous microwave coagulation therapy appears to be useful even in patients who have superficial liver tumors associated with cirrhosis.

In a patient with liver cirrhosis complicated by solitary gastric fundal varices and portosystemic encephalopathy, Balloon-occluded retrograde transvenous obliteration (B-RTO) of the varices was performed. The gastric varices were decreased in size 2 weeks after treatment and had not recurred after 1 year. B-RTO successfully occluded the portosystemic shunt (gastrorenal shunt). Accordingly, the patient's blood ammonia levels, total bile acid level, and 15 min retention rate of indocyanine green decreased, and his hepatic encephalopathy improved. However, since consecutive increase in blood flow through the portal collateral vessels except for gastrorenal shunt vessel at 6 months and 1 year after B-RTO was noted, further careful follow-up may be required.

Two cases of unresectable pancreatic cancer underwent percutaneous ethanol injection (PEI). The first was a 65-year-old man with a 43 x 39 mm mass in the head of the pancreas. He received PEI (total 38.5 ml of ethanol) 6 times. Serum CA19-9 decreased from 3,800 U/ml to 1,700 U/ml. The patient was dead 6 months after PEI. Autopsy revealed coagulative necrosis at the site of ethanol injection. The second case was a 66-year-old man with a 70 x 50 mm mass in the body of the pancreas. He was treated by PEI (total 10 ml). After PEI, his abdominal pain was relieved considerably. He died 4 months after PEI treatment. These findings suggest that PEI may be useful not only for pain control but also for tumor reduction of unresectable pancreatic cancer.

We report the first successful demonstration ofoptical parametric oscillation in periodically poled RbTiOAsO4(PPRTA). We have achieved >400mW ofsignal output power with 3.0W of 1.O64tm pump at 20kHz. The structure of the Y chromosome was investigated by FISH and G banding in 3 cases of bovine XY females shown by PCR to lack the SRY gene. Although 2 different repeat-DNA sequences (BC1.2 and btDYZ-1) hybridized to the short arm of the Y chromosome in a normal bull, they hybridized to both arms of the Y chromosome in the XY females. In the Y chromosome from XY females, only the centromeric region was darkly stained by G banding, in comparison with dark staining on the long arm of a normal Y chromosome. From the results obtained, it seemed that the Y chromosome from the XY females was structurally abnormal, namely an isochromosome with an identical short arm. Our results also confirm that SRY is located on the long arm of the Y chromosome in bovine.

We describe five sporadic cases of the EEC syndrome (ectrodactyly, ectodermal dysplasia, and cleft lip). There are only a few reports of the syndrome being inherited in Japan. Two of the five patients had genitourinary anomalies and one mental retardation. Four of the five patients had clefts in the primary and secondary palate. All had deformities of the hands, polydactyly, syndactyly, and camptodactyly. Syndactyly could be a component of the syndrome.

In 6 sterile heifers which were female in appearance and in genital organs, all tissues derived from blood, skin, spleen and kidney had a single 60, XY type of metaphase plate. Although the SRY gene was detected by PCR in normal bull, it was not detected in normal cow or in 3 60, XY cases studied.

Average power scaling of mid-infrared Optical Parametric Oscillators are presently limited by nonlinear material bulk absorption and resulting thermal gradients and thermally induced focusing and higher order aberrations. We discuss an optical correction scheme that corrects higher order aberrations typically present in gaussian intensity profile beams used for pumping OPO's to reduce the nonlinear dephasing by one- half. While correction of thermal focus has been shown to improve the power scaling in an OPO, further improvements can be made by correcting the remaining higher order aberrations. Such reduction in dephasing should significantly improve the power scaling potential of solid state lasers and OPO's.

A minute fragment was found in the metaphase plates obtained from one Holstein bull carrying Robertsonian translocation. The pedigree investigation indicated that three out of 34 offspring had the same minute fragment as their sire. Fluorescence in situ hybridization (FISH) studies with bovine satellite DNA revealed that the minute fragment was hybridized with this DNA and was part of the bovine centromere. We propose that the minute fragment is the product of centric fusion and is transmitted to offspring without being lost in the subsequent cell divisions.

A cw-lamp-pumped Nd:YAG injection-mode-locked seeded master oscillator-amplifier beamline is used to pump various optical parametric oscillators (OPOs) using KTP, KNbO3, AgGaS2, and AgGaSe2 crystals as the nonlinear materials. A comparison of these materials is presented and limitations on their ultimate power scaling are identified. This program has demonstrated tunable output in the 3 - 5 mm wavelength spectrum using a mode- locked and Q-switched (MLQS) pulse waveform at low power. One way of achieving high-power diode-end pumped lasers is to angularly multiplex several diodes on each end of the laser rod. We have successfully multiplexed four 15 W diode arrays on each end of a 6.3 mm diameter X 7.5 mm long Nd:YAG rod to produce an approximately equals 2 mm diameter pump spot. Higher laser power was achieved by adding a second laser rod pumped at both ends. The addition of the second rod facilitates thermally induced birefringence compensation by introducing a quartz polarization rotator between the rods. In addition, it was necessary to add an aspheric lens to compensate the thermal aberration induced at these high, nonuniform pump powers. With this arrangement, > 90 W has been extracted multimode, and > 60 W in a near-diffraction-limited beam.

A 200-Hz, 3-W Q-switched Nd:YAG oscillator is end-pumped by four quasi-cw diode arrays. Operation that is insensitive to the diode wavelength, and fifth harmonic generation are described.

We have demonstrated 62% energy conversion efficiency in a 2.12-μm degenerate KTP optical paramebic oscillator pumped with a 1.06-μm mode locked and Q-switched pulse. The oscillator is synchronously-pumped and has very high output coupling.

In this work, the issues of broadband tuning in the design of a high average power Ti:Sapphire amplifier for a chromatic scanning lidar system are addressed. A novel pump delay and staging scheme is used to maintain high extraction efficiency with flat temporal and spatial pulse profiles while tuning over a 700-900 nm range. The effects of the chromatic scanning scheme on ASE suppression and beam pointing are discussed in relation to the amplifier optical system. The average power scaling for three power amplifier geometries (zigzag slabs, parallel plates, and active mirrors) was investigated to determine the limitations of each. Pump light is provided by diode laser-pumped Nd:GGG slab lasers which make up over 60 percent of the system weight. Predicted electrical to light laser efficiency was 3-4 percent over the wavelength tuning range.

The corneal endothelial cells in 6 cases of mitochondrial myopathies were examined by specular microscope, and electron microscopic studies of the cornea were made in one autopsy case. The cornea was clear and the thickness was within the normal range. The endothelial cell mean size was larger in 3 out of 10 eyes than among controls and the variance of endothelial cell size was larger in all of 10 eyes than among controls. The difference was found to be statistically significant. Electron microscopical findings of the cornea revealed mitochondrial abnormalities. Inclusions were observed in the stromal keratocyte.

We used a cw-pumped, Q-switched, and mode-locked Nd:YAG oscillator and a cw-pumped, multipass traveling-wave amplifier with a face-pumped total-internal-reflection Nd:YAG slab as the gain element to produce an 80-W, TEM00 diffraction-limited beam at a Q-switching rate of 10 kHz (50 W at 3 kHz). The output of this device was used to synchronously pump a degenerate optical parametric oscillator that utilized six KTiOPO4 crystals as nonlinear converters. A maximum average power of 14 W at 2.128 μm was generated with a conversion efficiency of 25% at 4.25 kHz.

Es wird eine modifizierte Trypsin-Giemsa-Bandtechnik fur Chromosomen beschrieben, die beim Schwein eine deutliche Banderung zur Chromosomenidentifikation ergibt. Die Banderung wird erzielt durch eine Verkarzung der Kolchizineinwirkung auf 30 Minuten und eine Behandlung der 3–5 Tage alten Objekttrager mit einer 0,1 % Trypsinlosung (Sorensenpuffer pH 6,8) uber eine Dauer von 10–20 Minuten bei 4°C und anschlieβender Farbung mit 2 % Giemsalosung (Phosphatpuffer pH 6,8) fur 7–10 Minuten. Mit dieser Technik konnten die 18 autosomalen Chromosomen und die beiden Geschlechtschromosomen des Schweines identifiziert werden. Einzelheiten der G-Bandzeichnung eines jeden Chromosoms dieser Spezies werden beschrieben und in einem Karyotypogramm photographisch und schematisch belegt. Das beschriebene Verfahren kann fur Zytogenetiker der Human- und Tiermedizin von Bedeutung sein. Contents In the present paper the authors present a modified trypsin-giemsa chromosomal banding technique showing clear banding patterns for the identification of pig chromosomes. The banding patterns were induced by the reduction of colchicine density or trypsin treatment at a critical temperature of 4°C. Identification of 18 pairs of autosomes and a pair of sex-chromosomes of pig were made successfully. Details of the G-banding patterns of each chromosome were described and illustrated both in a karyotypical photograph and in a schematic diagram. The methods may be of importance to veterinary cytogenetics and genetical pathology, as well as to human medicine.

In order to elucidate the relationship between taste exhibition and chemical structure of bitter peptide BPIa, di–BPIa, Gly–Gly–BPIa, BPIa–Gly–Gly, and Gly–Gly–BPIa–Gly–Gly were synthesized. All of the peptides possessed a strong bitter taste of the same level as that of BPIa. In addition, the CD curves of the peptides were similar to that of BPIa. The results suggested that their spatial structures are essentially similar and that the whole molecular shape of BPIa contributes to its bitterness.

A performance mapping test of a 1 kW radiatively heated resistojet demonstrated high specific impulse and efficiency with hydrogen, nitrogen and ammonia as propellants. The resistojet was based on an unmodified augmentation section of the flight-qualified Rocket Research Company Augmented Catalytic Thruster. Peak specific impulse values of 686.9 sec for hydrogen, 191.6 sec for nitrogen, and 342 sec for ammonia were demonstrated. A maximum specific impulse efficiency of 94.3 percent was demonstrated with hydrogen. Testing covered a thrust range of 27 to 112 mlbf with augmentation powers ranging from 300 to 1000 watts. A total of 83 runs accumulating 25 hours of operation were conducted with the three propellants.
