13. Genetic Variation of Native Perilla Germplasms Collected from South Korea Using SSR Markers and Morphological Characteristics

Abstract

Using morphological characteristics and SSR markers, we evaluated the morphological and genetic variation of 200 Perilla accessions collected from the five regions of South Korea and other region. In morphological characteristics analysis, particularly, leaf color, stem color, degree of pubescence, leaf size were found to be useful for distinguishing the characteristics of native Perilla accessions cultivated in South Korea. A total of 137 alleles were identified in the 20 simple sequence repeat (SSR) markers, and the number of alleles per locus ranged from 3 to 13, and the average number of alleles per locus was 6.85. The average gene diversity (GD) was 0.649, with a range of 0.290-0.828. From analysis of SSR markers, accessions from the Jeolla-do and Gyeongsang-do regions showed comparatively high genetic diversity values compared with those from other regions in South Korea. In the unweighted pair group method with arithmetic mean (UPGMA) analysis, the 200 Perilla accessions were found to cluster into three major groups and an outgroup with a genetic similarity of 42%, and did not showed a clear geographic structure from the five regions of South Korea. Therefore, it is believed that landrace Perilla seeds are frequently exchanged by farmers through various routes between the five regions of South Korea. The results of this study are expected to provide useful information for conservation of these genetic resources and selection of useful resources for the development of varieties for seeds and leafy vegetables of cultivated var. frutescens of Perilla crop in South Korea.

Abstract

Eggplant is one of the most economically and nutritionally important vegetables worldwide. The study of the association of phenotypic traits with genetic factors is vital for the rapid and efficient identification and selection of eggplant genetic resources for breeding purposes with desired traits. The eggplant resources (587) collected from different countries, including Korea, were used for establishing the core collection. A total of 288 accessions were selected from 587 Solanum accessions based on 52 single nucleotide polymorphisms (SNPs) markers together with 17 morphological traits. This core collection was further used to analyze the genetic associations of eggplant morphological variations. A large variation was found among the evaluated eggplant accessions for some agro-morphological traits. Stem prickles and leaf prickles showed a significant positive correlation (r = 0.83***), followed by days to flowering and days to maturity (r = 0.64***). A total of 114,981 SNPs were filtered and used for phylogenetic tree analysis, population structure analysis, and genome-wide association study (GWAS). Among the agro-morphological traits, significantly associated SNPs were found for six traits. A total of 377 significantly associated SNPs with six agro-morphological traits were identified. These six traits and the number of SNPs were: days to maturity (51), flower size (121), fruit width (20), harvest fruit color (42), leaf prickles (38), and stem prickles (105). The largest fraction of significant SNPs (11.94%) was obtained on chromosome Ch01, followed by Ch07 and Ch06 with 11.67% and 10.08%, respectively. This study will help to develop markers linked to the most important agro-morphological traits of eggplant genetic resources and support the selection of desirable traits for eggplant breeding programs. Abstract

Seed macro‐ and micro‐morphology were analyzed to evaluate their capacity to discriminate species in the genus Vicia (Fabaceae). To assess the interspecific variation of the taxa in the genus Vicia, 41 accessions were obtained from the USDA‐ARS germplasm collection in the USA and 19 accessions were collected from Korea. Seed morphological characteristics such as shape, color, mottling, finish, length, width, diameter, hilum shape, hilum color, hilum length, and lens distance from the hilum were examined under a stereomicroscope. Testa texture characteristics such as testa pattern, papillae type, density, height, ribbing, surface deposits, and peaks topped with wax were examined under scanning electron microscopy. Various gross‐morphological traits of seeds of Vicia species have been analyzed and compared. The present study revealed significant variation in testa traits. Testa were papillose and papillose with mounds, the latter being observed only in Vicia lathyroides. Abstract

A variation of temperature by sunlight acting on a RC roof slab causes a change of stress in concrete since it expands during summer and is compressed during winter. This behavior repeats annually and affects structural capacity of member for both serviceability and ultimate level. In this paper, a variation of cyclic temperature loading is calculated by analyzing the weather data of Korea for 20 years. In addition, an experimental work is planned to find the long term effect of temperature change. Four RC slabs are made with same dimension. Test parameters are loading duration (10, 20, 30 years). Observation of stiffness variations according to cyclic loading duration shows that the serious stiffness drop happens after 10 year's cyclic loading at summer while after 30 year's loading at winter. From the failure test, maximum strength of specimen that experienced repeated preloading was approximately 12% less than standard specimen without any repeated preloading.

Abstract

An experimental study was performed to evaluate seismic reduction performance and applicability of 3-dimensional floor isolation system to the main control room of nuclear power plant. A friction pendulum system(FPS) and air spring were designed and fabricated for 3-dimensional floor isolation system. Two kind of the partial experimental model of a main control room attached to the FPS and air spring were tested on the shaking table. The experimental model consisted of a control panel, a access floor, four FPS and air springs. The artificial time histories based on the vertical and horizontal floor response spectrums(OBE, SSE) of the main control room were used as the earthquake input signals in the test. Compared to non-isolated system, the seismic response of experimental models using 3-dimensional floor isolation system were shown considerable seismic reduction performance.

Abstract

Three RC slab specimens were designed and manufactured to investigate the fatigue behavior of RC slab retrofitted with carbon fiber mesh (CFM) and mortar. Among three slab specimens, two slabs were pre-loaded to introduce damage prior to the fatigue loading tests. Fatigue loading was applied to the top of slab maintaining 40% of the ultimate strength of specimen at 2Hz loading speed. The fatigue test was terminated when the loading cycle exceeded 1,000,000 cycles. From the test results, the effectiveness of the retrofitting using CFM was evaluated. Test result showed that the specimens retrofitted after either severe or light damages had very similar strength and stiffness to those of the specimens retrofitted without damage. It was found that the strength was even slightly improved in the specimen retrofitted with CFM.
