Background: Protists are essential contributors to eukaryotic diversity and exert profound influence on carbon fluxes and energy transfer in freshwaters. Despite their significance, there is a notable gap in research on protistan dynamics, particularly in the deeper strata of temperate lakes. This study aimed to address this gap by integrating protists into the well-described spring dynamics of Římov reservoir, Czech Republic. Over a 2-month period covering transition from mixing to established stratification, we collected water samples from three reservoir depths (0.5, 10 and 30 m) with a frequency of up to three times per week. Microbial eukaryotic and prokaryotic communities were analysed using SSU rRNA gene amplicon sequencing and dominant protistan groups were enumerated by Catalysed Reporter Deposition-Fluorescence in situ Hybridization (CARD-FISH). Additionally, we collected samples for water chemistry, phyto- and zooplankton composition analyses. Results: Following the rapid changes in environmental and biotic parameters during spring, protistan and bacterial communities displayed swift transitions from a homogeneous community to distinct strata-specific communities. A prevalence of auto- and mixotrophic protists dominated by cryptophytes was associated with spring algal bloom-specialized bacteria in the epilimnion. In contrast, the meta- and hypolimnion showcased a development of a protist community dominated by putative parasitic Perkinsozoa, detritus or particle-associated ciliates, cercozoans, telonemids and excavate protists (Kinetoplastida), co-occurring with bacteria associated with lake snow. Conclusions: Our high-resolution sampling matching the typical doubling time of microbes along with the combined microscopic and molecular approach and inclusion of all main components of the microbial food web allowed us to unveil depth-specific populations' successions and interactions in a deep lentic ecosystem. This study describes the seasonal course of denitrifying and nitrate-reducing bacteria in a dimictic mesotrophic lake (Lake Scharmützelsee, Brandenburg, Germany) within a three-year period from 2011 to 2013. The bacterial cell numbers were quantified by the fluorescence microscopy, most probable number (MPN) and PCR-dependent quantification of the chromosomal 16S rDNA and of the nirS and nirK gene copy number. The highest seasonal differences (up to three orders of magnitudes) have been measured using MPN in the epilimnion. This variation was not reflected by PCR-dependent approaches or direct microscopical enumeration. At adverse conditions (low temperature and/or low nitrate concentrations), the differences between MPN and gene copy numbers increased by up to five orders of magnitudes and decreased to one magnitude at favourable environmental conditions. These results can be explained best by an increasing ratio of viable but not cultivable (VBNC) cells or dead cells at impairing conditions. In the hypolimnion, the courses of MPN and nir gene copy numbers were similar. This can be explained by a higher feeding pressure and therefore smaller amounts of dormant cells. In the pelagial in general, the total cell numbers enumerated by either microscopical or molecular approaches were similar. In the sediment, more than 99% of the DNA was obviously not related to viable bacteria but was rather DNA in dead cells or adsorbed to particle surfaces. [ABSTRACT FROM AUTHOR]

Declining oxygen concentrations in the deep waters of lakes worldwide pose a pressing environmental and societal challenge. Existing theory suggests that low deep‐water dissolved oxygen (DO) concentrations could trigger a positive feedback through which anoxia (i.e., very low DO) during a given summer begets increasingly severe occurrences of anoxia in following summers. Specifically, anoxic conditions can promote nutrient release from sediments, thereby stimulating phytoplankton growth, and subsequent phytoplankton decomposition can fuel heterotrophic respiration, resulting in increased spatial extent and duration of anoxia. However, while the individual relationships in this feedback are well established, to our knowledge, there has not been a systematic analysis within or across lakes that simultaneously demonstrates all of the mechanisms necessary to produce a positive feedback that reinforces anoxia. Here, we compiled data from 656 widespread temperate lakes and reservoirs to analyze the proposed anoxia begets anoxia feedback. Lakes in the dataset span a broad range of surface area (1–126,909 ha), maximum depth (6–370 m), and morphometry, with a median time‐series duration of 30 years at each lake. Using linear mixed models, we found support for each of the positive feedback relationships between anoxia, phosphorus concentrations, chlorophyll a concentrations, and oxygen demand across the 656‐lake dataset. Likewise, we found further support for these relationships by analyzing time‐series data from individual lakes. Our results indicate that the strength of these feedback relationships may vary with lake‐specific characteristics: For example, we found that surface phosphorus concentrations were more positively associated with chlorophyll a in high‐phosphorus lakes, and oxygen demand had a stronger influence on the extent of anoxia in deep lakes. Taken together, these results support the existence of a positive feedback that could magnify the effects of climate change and other anthropogenic pressures driving the development of anoxia in lakes around the world. Difficulties in the localization of bodies of homicidal or drowning victims in natural water result in their submergence for weeks if not months. Water insects and microbes drastically change the body's appearance, which significantly changes the determination of a victim's identity. DNA analysis is commonly used for identifying the decedent; however, this PCR‐based approach is time‐consuming and destructive of the evidence. Considering that nearly half of the people in the world dye their hair with a variety of permanent and semi‐permanent dyes, one can expect that confirmatory identification of dyes on the body's hair can be used to shed light on the victim's identity. A growing body of evidence suggests that surface‐enhanced Raman spectroscopy (SERS) can be used to detect and identify hair dyes. In this study, we investigated the extent to which SERS could be used to detect black and blue, permanent and semi‐permanent dyes on hair submerged in hypolimnion water for up to twelve weeks. We found that SERS enabled 100% accurate identification of analyzed dyes on hair submerged in hypolimnion water for up to 8 weeks, whereas, on average, 87% accurate identification of the hair dyes could be achieved on hair exposed for 10 weeks and 50% for hair exposed 12 weeks in hypolimnion water. We also found that the aqueous environment caused progressive fading of some dyes, whereas other dyes showed substantial spectral transformations after prolonged submergence. Finally, we found that changes in the intensity of vibrational bands of dyes could be used to predict the duration of submergence of colored hair in hypolimnion water. [ABSTRACT FROM AUTHOR]

The central basin of Lake Erie is annually subject to hypolimnetic hypoxia, which affects biochemical processes and aquatic biota. Since European settlement, water quality in Lake Erie has degraded due to land-use changes within its catchment, cultural eutrophication, climate change, and the effects of invasive species. As limnological monitoring data for Lake Erie are largely unavailable prior to the 1970s, paleolimnological methods were used to infer long-term trends in hypolimnetic dissolved oxygen. Changes in lake water quality affect benthic community composition, and those communities can be used as indicators of long-term environmental changes, particularly those that affect hypolimnetic oxygen concentrations. Chironomid subfossil remains preserved in dated sediment cores indicated that the hypolimnion of the central basin was exhibiting low oxygen conditions prior to major Euro-American settlement impacts, and it has been mesotrophic to eutrophic since ∼1850. The disappearance of oxic-type taxa and a transition towards anoxic-tolerant chironomid communities by the 1950s suggest that the severity and duration of hypolimnetic anoxia has increased in recent decades. Variance partitioning analysis (VPA) indicated that both land use and climate variables were significant drivers of these changes in chironomid assemblages in the central basin. Lake Mead located in the Arizona–Nevada region of the Mohave Dessert is a unique and complex water system whose flow follows that of a warm monomictic lake. Although monomictic lakes experience thermal stratification for almost the entire year with a period of complete mixing, the lake on occasion deviates from this phenomenon, undergoing incomplete turnovers categorized with light stratifications every other year. The prolonged drought and growing anthropogenic activities have the potential to considerably impact the quality of the lake. Lake Mead and by extension the Boulder Basin receive cooler flow from the Colorado River and flow with varying temperatures from the Las Vegas Wash (LVW), which impacts its stratification and complete turnovers. This study analyzes four key water quality parameters (WQPs), namely, total dissolved solids (TDS), total suspended solids (TSS), temperature, and dissolved oxygen (DO), using statistical and spatial analyses to understand their variations in light of the lake stratifications and turnovers to further maintain its overall quality and sustainability. The study also evaluates the impacts of hydrological variables including in and out flows, storage, evaporation, and water surface elevation on the WQPs. The results produced from the analysis show significant levels of TDS, TSS, and temperature from the LVW and Las Vegas Bay regions compared with the Boulder Basin. LVW is the main channel for conveying effluents from several wastewater treatment facilities into the lake. We observed an increase in the levels of TDS, TSS, and temperature water quality in the epilimnion compared with the other layers of the lake. The metalimnion and the hypolimnion layer, however, showed reduced DO due to depletion by algal blooms. We observed statistically significant differences in the WQPs throughout various months, but not in the case for season and year, an indication of relatively consistent variability throughout each season and year. We also observed a no clear trend of influence of outflows and inflows on TDS, temperature, and DO. TSS concentrations in the lake, however, remained constant, irrespective of the inflows and outflows, possibly due to the settling of the sediments and the reservoir capacity.

In general, lakes provide various ecosystem services that support biotic habitats and human life. Lake Toba, the largest caldera volcanic lake in the world, has been functioning as a tourism destination and for fish farming, a fresh water source, and for power supplies. The lake has maximum depth of about 505 m. Stratification in the lake water column especially in a tropical region like Indonesia is a common phenomenon. Stratification of the lake is one important factor which will influence the next stage in the biological processes and water quality of the lake. The objective of the current study was to analyze and elucidate the stratification of Lake Toba through the variations of physical, chemical, and isotopic parameters. Water temperature, dissolved oxygen concentrations, water chemical contents, and isotopic parameters were observed periodically from 2016 until 2019. Fourteen sampling points evenly distributed across the surface of the lake were predetermined to represent the North, South, East, and West of the lake. A conductivity, temperature, and depth (CTD) device and Baro-divers were used to measure temperature and conductivity at various depths in the water column for each sampling point. The water samples for isotopic and chemical parameter measurements were collected from water depths of 0, 20, 40, 60, 80, and 100 m using a horizontal transparent acrylic water sampler at each sampling point. Some results from isotope analysis showed that all the water at all levels along the water column had undergone evaporation. Although there were some slight fluctuations, the chemical composition of the lake water was relatively homogenous up to 100 m of depth. The chemical pattern suggested that there was no other secondary process that altered the chemical composition of the lake water, thus the lake water and river water had the same facies. The stratification of Lake Toba was found to be permanent. The depth of the hypolimnion layer was consistently at about 80 m below the surface. However, the depth of the upper layer, the epilimnion, was strongly affected by the climate at the surface of the lake. [ABSTRACT FROM AUTHOR]

Widespread changes in dissolved organic carbon (DOC) concentrations in lakes of the Northern Hemisphere are well documented, and there is a need to understand the ecological implications for changes in cold‐water refugia. DOC controls thermal structure in small, wind‐sheltered lakes, thus influencing the volume and persistence of the hypolimnion and cold‐water habitat. We explored the relationship between the quantity and quality of DOC and the availability of cold‐water habitat over the course of summer stratification in small lakes of varying depths. The ratio of the hypolimnetic volume to total lake volume was calculated to assess the relative changes in cold‐water habitat availability in each lake over time. DOC concentration, maximum lake depth, and the interaction between DOC concentration and maximum lake depth explained 87% of the variability in percent change in hypolimnion volume ratio across studied lakes. Hypolimnion compression was greater in shallow, low DOC lakes, whereas deeper, intermediate DOC lakes had more persistent hypolimnion volumes over the course of summer stratification. These results demonstrate that depth moderates the effect of DOC on the volume and persistence of cold‐water refugia availability in north temperate lakes, advancing our understanding of differing lake sensitivity in a rapidly changing physical and chemical environment. Prokaryotic diversity in lakes has been studied for many years mainly focusing on community structure and how the bacterial assemblages are driven by physicochemical conditions such as temperature, oxygen, and nutrients. However, little is known about how the composition and function of the prokaryotic community changes upon lake stratification. To elucidate this, we studied Lake Cote in Costa Rica determining prokaryotic diversity and community structure in conjunction with physicochemistry along vertical gradients during stratification and mixing periods. Of the parameters measured, ammonium, oxygen, and temperature, in that order, were the main determinants driving the variability in the prokaryotic community structure of the lake. Distinct stratification of Lake Cote occurred (March 2018) and the community diversity was compared to a period of complete mixing (March 2019). The microbial community analysis indicated that stratification significantly altered the bacterial composition in the epi-meta- and hypolimnion. During stratification, the Deltaproteobacteria, Chloroflexi, Bacteroidetes, Nitrospirae, and Euryarchaeota were dominant in the hypolimnion yet largely absent in surface layers. Among these taxa, strict or facultative anaerobic bacteria were likely contributing to the lake nitrogen biogeochemical cycling, consistent with measurements of inorganic nitrogen measurements and microbial functional abundance predictions. In general, during both sampling events, a higher abundance of Alphaproteobacteria, Betaproteobacteria, Actinobacteria, and Cyanobacteria was found in the oxygenated layers. Lake Cote had a unique bacterial diversity, with 80% of Amplicon Sequence Variant (ASV) recovered similar to unclassified/uncultured strains and exhibits archetypal shallow lake physicochemical but not microbial fluctuations worthy of further investigation. This study provides an example of lake hydrodynamics impacts to microbial community and their function in Central American lakes with implications for other shallow, upland, and oligotrophic lake systems. Global change is modifying meteorological and hydrological factors that influence the thermal regime of water bodies. These modifications can lead to longer stratification periods with enlarged hypolimnetic anoxic periods, which can promote heterotrophic anaerobic processes and alter reservoir carbon cycling. Here, we quantified aerobic and anaerobic heterotrophic processes (aerobic respiration, denitrification, iron and manganese reduction, sulfate reduction, and methanogenesis) on dissolved inorganic carbon (DIC) production in the hypolimnion of a Mediterranean reservoir (El Gergal, Spain) under two contrasting hydrological conditions: a wet year with heavy direct rainfall and frequent water inputs from upstream reservoirs, and a dry year with scarce rainfall and negligible water inputs. During the wet year, water inputs and rainfall induced low water column thermal stability and earlier turnover. By contrast, thermal stratification was longer and more stable during the dry year. During wet conditions, we observed lower DIC accumulation in the hypolimnion, mainly due to weaker sulfate reduction and methanogenesis. By contrast, longer stratification during the dry year promoted higher hypolimnetic DIC accumulation, resulting from enhanced methanogenesis and sulfate reduction, thus increasing methane emissions and impairing reservoir water quality. Aerobic respiration, denitrification and metal reduction produced a similar amount of DIC in the hypolimnion during the two studied years. All in all, biological and geochemical (calcite dissolution) processes explained most of hypolimnetic DIC accumulation during stratification regardless of the hydrological conditions, but there is still ~ 30% of hypolimnetic DIC production that cannot be explained by the processes contemplated in this study and the assumptions made. Previous studies in Lake Sevan during summer stratification have shown that the highest zooplankton abundance and biomass was found at the lower boundary of the epilimnion. At the same time, whitefish (Coregonus lavaretus), being main planktivorous species in the lake, preferred the hypolimnion, but was leaving this layer if dissolved oxygen concentrations decreased to 1–5 mg/L. The study was conducted in Bolshoy Sevan (the southern part of Lake Sevan) on July 29–30, 2019. The vertical profiles of water temperature and dissolved oxygen concentration, as well as distribution of major taxonomic groups of aquatic organisms were assessed four times a day: in the dark (night time), in the light (day time), and in twilight (morning and evening). Fish population density was assessed using a “Simrad EK80” echosounder. Vertical distribution of zooplankton and fish observed within the study period did not correspond to that described earlier. Maximal zooplankton biomass during the study was recorded in the hypolimnion, and minimal biomass, in the epilimnion. Most of fish (89–100%) located in the metalimnion. At night, about 10% of the fish population descended to the hypolimnion, despite the low dissolved oxygen concentrations found there (3.0–3.1 mg/L). At dawn, about 7% of the fish ascended to the epilimnion, reaching a 7-m depth, where the water temperature was 20.8 °C. The water temperature range preferred by most fish individuals varied during the observation period and did not correspond to previously reported ranges.

Under-ice community dynamics are barely understood. Temporal and spatial studies are needed to fully understand the consequences of a declining ice cover on microbial biodiversity. Here, bacterial communities of different years (2015, 2017–2021) and layers (upper and lower euphotic layer, euphotic layer, hypolimnion) were assessed by Illumina sequencing of the 16S rRNA gene. Alpha- and beta-diversity of summer and under-ice hypolimnetic communities were similar, and a seasonal difference was found only when excluding summer hypolimnetic communities. Similarly, in non-metric multidimensional scaling (NMDS), summer and under-ice communities were different even though hypolimnetic communities were similar. Investigating under-ice conditions, the year 2017 showed highest under-ice light and chlorophyll-a while 2021 showed no under-ice light and lowest chlorophyll-a. Under-ice communities were not linked to layer differences implying that a spatial distinction under ice was less important than in summer, especially in years with little or no under-ice light. Most under-ice bacterial classes and ASVs showed direct and indirect dependencies on light availability and primary production. Similarly in NMDS with only under-ice communities, light transparency and primary production were important. In the future, ice conditions with less snow cover might lead to bacterial communities similar to that of high-light years (2017, 2018, 2020). [ABSTRACT FROM AUTHOR]

In 1984, a limnic eruption occurred in Lake Monoun, Cameroon, and the CO2 gas released from the lake surface resulted in casualties in the neighboring communities. Subsequent scientific research revealed that the CO2 gas released from the lake surface was CO2 of magmatic origin dissolved in the lake water; however, the mechanism of that limnic eruption remains unclear. In this study, we analyzed in detail the lake-bottom bathymetry of the eastern basin, i.e., one of the three basins in Lake Monoun, to understand the mechanism of the 1984 limnic eruption. We discovered two significant depressions at the lake bottom near the scarp and obtained vertical profiles of several parameters of the lake water at the depression locations. The northeastern depression (D1) was ∼ 1.2 m deeper than the lake bottom and contained water with higher temperature and electrical conductivity and lower pH relative to the lake water. Conversely, the southern depression (D2) was ∼ 2.2 m deeper than the lake bottom, and there were no anomalies regarding its water parameters. Although the warm water discharged from the bottom of D1 was not saturated with dissolved CO2, bubbles likely existed at the bottom of D1, influenced by the partial pressure of dissolved CH4 in the lake water. Our results suggest that just before the 1984 limnic eruption, water containing high concentrations of dissolved CO2 was discharged from D1; this water would have reached the lake surface with bubbles. According to earlier numerical simulations of the limnic eruption, rising bubbles could have induced the limnic eruption. The rising bubbles entrained the surrounding lake water containing high concentrations of dissolved CO2, which amplified the flow rate of CO2 degassing from the lake water and resulted in a limnic eruption. The limnic eruption that occurred just above D1 displaced lake water on the eastern shore. It is estimated that the impact of the displaced water eroded the scarp and deposited sediment as a mound near D1. A similar mound also exists near D2, suggesting that D2 is a trace of another limnic eruption that occurred earlier than 1984. Of the three basins that make up Lake Monoun, the two smaller basins to the west have high concentrations of dissolved CO2 in their deep waters. This dissolved CO2 was not supplied from the bottom of the basins but is likely a remnant of the dissolved CO2 that existed in 2003 before the start of artificial CO2 degassing. Our results suggest that another limnic eruption occurred before 1984. Lake Monoun may have experienced several limnic eruptions in the past. If the artificial degassing of CO2 is not continued, the water released from D1, containing high concentrations of dissolved CO2, will increase the concentration of dissolved CO2 in the lake water, and the bubbles rising from D1 will cause another limnic eruption. In the future, the flux of CO2 supplied from D1 may increase and exceed the flux of CO2 removed by the artificial degassing, potentially increasing the amount of CO2 accumulated in the lake water. The regular monitoring of the CO2 amount in lake water should be also continued.

In this study, we report on turbulent mixing observed during the annual stratification cycle in the hypolimnetic waters of Lake Michigan (USA), highlighting stratified, convective, and transitional mixing periods. Measurements were collected using a combination of moored instruments and microstructure profiles. Observations during the stratified summer showed a shallow, wind‐driven surface mixed layer (SML) with locally elevated dissipation rates in the thermocline (ϵ≈10−7W/kg) potentially associated with internal wave shear. Below the thermocline, turbulence was weak (ϵ≈10−9W/kg) and buoyancy‐suppressed (Q=ϵ/νN2 < 8.5), with low hypolimnetic mixing rates (Kz≈10−7m2/s) limiting benthic particle delivery. During the convective winter period, a diurnal cycle of radiative convection was observed over each day of measurement, where temperature overturns were directly correlated with elevated turbulence levels throughout the water column (ϵ≈10−7W/kg; Kz≈10−1m2/s). A transitional mixing period was observed for spring conditions when surface temperatures were near the temperature of maximum density (TMD≈3.98°C) and the water column began to stably stratify. While small temperature gradients allowed strong mixing over the transitional period (KZ≈10−3m2/s), hypolimnetic velocity shear was overwhelmed by weakly stable stratification (N2≈10−6s−2; Ri≈0.2), limiting the development of the SML. These results highlight the importance of radiative convection for breaking down weak hypolimnetic stratification and driving energetic, full water column mixing during a substantial portion of the year (>100 days at our sample site). Ongoing surface water warming in the Laurentian Great Lakes is significantly reducing the annual impact of convective mixing, with important consequences for nutrient cycling, primary production, and benthic‐pelagic coupling. Plain Language Summary: Turbulent mixing is a fundamental physical process in aquatic systems, where it plays a role in transferring momentum and heat, regulating water quality, and controlling the vertical transport of nutrients and dissolved gases. Mixing rates in large, temperate lakes are especially dynamic, since annual changes in atmospheric forcing (e.g., air temperature, wind speed, daylight hours, etc.) have dramatic consequences for both the magnitude and structure of turbulence throughout the water column. In this study, we investigate annual changes in deep water mixing as observed at a 55 m deep site in a large, freshwater lake (Lake Michigan, USA). Observations are used to define three distinct mixing regimes, including (a) the stratified period, where strong density gradients suppressed turbulent overturns; (b) the transitional period, where the water column was nearly isothermal and wind‐driven mixing was only marginally affected by density stratification; and (c) the convective period, where radiative heating generated density instabilities that enhanced mixing across the water depth. Results are used to describe the effects of recent surface warming on reducing deep‐water mixing. This work represents one of the most comprehensive studies of offshore mixing in the Laurentian Great Lakes, with important implications for biophysical modeling and invasive species management. Abstract The release of water from the reservoir hypolimnion, lower concentration of oxygen and the anthropogenic regulation of the river flow, could affect the reproduction of fish, especially migratory species. However, little is known about the effects of these changes in water on non-migratory species. In this sense, the reproduction of Acestrohynchus lacustris was evaluated in two sections of São Francisco River, Minas Gerais, Brazil. Section 1, located immediately downstream from Três Marias Dam (18°09’31.65”S and 45°13’36.00”W) and section 2, located at the confluence of the São Francisco and the Abaeté Rivers (18°02’47.78”S and 45°10’57.95”W). For this, we obtained the physico-chemical parameters of water of each study section. Additionally, biometric data and biological indices of all specimens were measured. Fecundity and follicles diameters were measured in females. Temperature, dissolved oxygen and flow showed lower values in section 1. Fish captured in this section, had lower values of GSI in both sexes, and females presented decreased values of fecundity and follicles diameter. This species showed reproductive activity in the two sections analyzed, however, in section 1 where the temperature and dissolved oxygen presented significant lower values, the reproductive capacity of A. lacustris, was negatively affected.

A hydrogeochemical study was performed on 45 lakes spread across six islands of the Azores archipelago (Portugal). Water temperature of the studied lakes is cold (mean = 16.2 °C), and the deeper lakes depict a monomictic behaviour, with water column stratification during summer resulting in higher carbon dioxide concentrations in the hypolimnion. Samples present an acid to neutral pH (mean = 7.04), and in general correspond to diluted and weakly mineralized waters (mean electrical conductivity = 107 μS/cm). The prevailing hydrogeochemical facies are of the Na–Cl type, which derives from atmospheric marine salts contamination, and, albeit not to the same extent, Na–HCO 3 , suggesting a cumulative effect of water-rock interaction. Water from Furna do Enxofre Lake exhibits a dissimilar Mg–HCO 3 type. The neutralization of water acidity is promoted by water-rock interaction, which also leads to a bicarbonate enrichment in many lakes in the Azores. The HCO 3 − enrichment shows that other processes are controlling the water composition, such as the dissolution of CO 2 from various origins, of which CO 2 -rich gas emissions from volcanic sources. A fraction of the emitted CO 2 at Furna do Enxofre Lake (Graciosa) and Furnas Lake (on São Miguel) is of deep origin as showed by the δ13C content. In the remaining lakes, where these isotopic analyses (δ13C) were carried out, the emitted CO 2 has a biogenic origin, derived from the decomposition of organic matter and eutrophication, a process that affects several lakes in the Azores. The study of other stable isotopic contents (δ18O and δ2H) allowed also the confirmation of the meteoric origin of studied waters. Excluding Furna do Enxofre lake, classified in class 5 (i.e. low activity lake), all lakes are classified as no activity (class 6). Thermal stratification is considered to be the most important limnological feature of deep lake ecosystems affecting a water column's chemical characteristics thereby directly connected with lake management. The lake is also attracting the attention of management authorities, being a growing tourist destination with sustainable environmental management plan. Lake Manasbal is only true warm monomictic urban lake in Kashmir and remains thermally stratified for 8–9 months, with separate layers of epilimnion, metalimnion and hypolimnion.We noticed that in the lower part of the hypolimnion, the process of stratification is characterized by an oxygen deficit (O2 < 1 mg/L). The rate of oxygen depletion reported in the lake's bottom layers is indicative of the water body's more eutrophic nature. A clinograde type of pH curve with a wide range of fluctuation from top to bottom layers of water was observed. Thermal stratification of the lake allows ammonium and phosphates to pool in the hypolimnion for much of the stratification period, resulting in concentrations higher than in the epilimnion. Consequently, the lake was characterized by a nutrient-enriched hypolimnion and an epilimnion deprived of nutrients. With the advent of the lake stratification, the thermocline was formed on 15th March between 2 and 3 m and on 7th April between 4 and 6 m, which gradually moved down to 10 m on 4th July and remained at that depth until circulation. The concentration of phosphorus and nitrogen in Lake Manasbal showed an increase towards the bottom, thus revealing an inverse clinograde depth distribution. Due to stratification phenomena in this lake, the internal release and subsequent distribution of nutrients throughout water column especially phosphorus from sediment water interface after mixing is a cause of concern for management authorities as it has the potential to diminish the ecological and aesthetics of the lake.Therefore, we argue that this study will provide some valuable insights on how stratification is affecting the nutrient dynamics and will offer inputs for better management accordingly both from ecological and economic perspectives. A toolbox of methods must be available for the remediation of lakes and water bodies suffering from eutrophication. One method suggested is hypolimnetic withdrawal based on a closed-circuit system. Prior to the start of a pilot-scale test at Lake Hönsan, Sweden, a laboratory trial with containers filled with water and bottom sediment from this lake was performed. A peristaltic pump distributed equal bottom water volume to four columns, two filled with glass beads and two with the filter material Polonite, and then back to the surface of the containers. The reactive filter medium (RFM) removed phosphate (PO4-P) efficiently (98.6%), despite the relatively low influent concentration (390 µg L−1). The control column filled with glass beads, removed 2.9% of the PO4-P. The anoxic sediment, containing 2.47 mg P g−1, released PO4-P, which was indicated by the increased concentration in near-bottom water. The redirected water after RFM filtration had high pH (x¯=11.1); however, an equalization took place in the water mass to a lower but still increased pH value (x¯=8.7) compared to the control (x¯=7.02). This article reports the pros and cons of a full-scale system using the proposed method.

Aquatic N-fixation is generally associated with the growth and mass development of Cyanobacteria in nitrogen-deprived photic zones. However, sequenced genomes and environmental surveys suggest active aquatic N-fixation also by many non-cyanobacterial groups. Here, we revealed the seasonal variation and genomic diversity of potential N-fixers in a humic bog lake using metagenomic data and nif gene clusters analysis. Groups with diazotrophic operons were functionally divergent and included Cholorobi, Geobacter, Desulfobacterales, Methylococcales, and Acidobacteria. In addition to nifH (a gene that encodes the dinitrogenase reductase component of the molybdenum nitrogenase), we also identified sequences corresponding to vanadium and iron-only nitrogenase genes. Within the Chlorobi population, the nitrogenase (nifH) cluster was included in a well-structured retrotransposon. Furthermore, the presence of light-harvesting photosynthesis genes implies that anoxygenic photosynthesis may fuel nitrogen fixation under the prevailing low-irradiance conditions. The presence of rnf genes (related to the expression of H+/Na+-translocating ferredoxin: NAD+ oxidoreductase) in Methylococcales and Desulfobacterales suggests that other energy-generating processes may drive the costly N-fixation in the absence of photosynthesis. The highly reducing environment of the anoxic bottom layer of Trout Bog Lake may thus also provide a suitable niche for active N-fixers and primary producers. While future studies on the activity of these potential N-fixers are needed to clarify their role in freshwater nitrogen cycling, the metagenomic data presented here enabled an initial characterization of previously overlooked diazotrophs in freshwater biomes.

Kinetoplastid flagellates are generally abundant in the deep sea and recently they were even found to be dominant in the hypolimnion of a deep freshwater lake. Therefore, to understand the distribution of kinetoplastids in deep freshwater lakes, we have collected vertical samples from five lakes in Japan. The abundance of kinetoplastids was enumerated by Catalyzed Reporter Deposition-Fluorescence in situ Hybridization, and the diversity was determined by 18S amplicon sequencing using universal eukaryote and kinetoplastid-specific primers. Kinetoplastids were abundant in the deep waters of all the lakes, contributing up to 53.6% of total nanoeukaryotes. Despite this significant contribution, kinetoplastids remain undetected by amplicon sequencing using universal primers that are widely used in eukaryotic diversity studies. However, they were detected with specific primers, and the communities were characterized by both ubiquitous and lake-specific unique OTUs. Oligotyping of a ubiquitous and dominant OTU revealed the presence of lake-specific sequence types (oligotypes). Remarkably, we also detected diplonemids (a sister group of kinetoplastids and considered to be specific in the marine habitat) using kinetoplastid-specific primers, showing their presence in freshwaters. Underestimation of kinetoplastids and diplonemids using universal primers indicates that euglenozoan flagellates are overlooked in diversity studies worldwide. The present study highlighted the importance of kinetoplastids in the hypolimnion of deep lakes, thereby indicating their role in material cycling in deep waters.

—Studies of epilimnion, metalimnion, and hypolimnion are of great importance for understanding the vertical structure and functioning of aquatic ecosystems. The present article reports new facts related to the stratification of freshwater ecosystems. In particular, the concentration of dissolved oxygen, temperature, and hydrobiological parameters at depths from 0 to 30 m of Glubokoe Lake (Moscow oblast) were determined. The metalimnion (thermocline) which is characterized by a high vertical temperature gradient was found to accumulate a large amount of detritus and contain increased amounts of bacteria and algae cells (phytoplankton). Calculation of the minimum oxygen concentration in the hypolimnion of lakes based on the balance approach was used to estimate the level of production processes corresponding to the emergence of anoxia in the hypolimnion. Data from regular monitoring of the dissolved oxygen regime in 16 lakes and reservoirs of the world were processed to obtain the linear regression relationships of annual primary production values at which anoxia may appear in the hypolimnion of a lake. An inverse linear dependence of the critical primary production on the lake’s shape factor was established and tested using data from independent observations of the oxygen regime in lakes.

We analyzed long-term oxygenation dynamics in 3 waterbodies: Spring Hollow Reservoir (SHR) and Carvins Cove Reservoir (CCR), which are water supply reservoirs, and North Twin Lake (NTL), a natural lake. Bubble plume line diffuser hypolimnetic oxygenation system (HOS) operation was for 13, 10, and 7 yr for SHR, CCR, and NTL, respectively. Water column profiles were measured in all 3 waterbodies. Additionally, remotely deployed sensors were used to monitor dissolved oxygen (DO) in NTL year-round, specifically to capture winter conditions during ice cover. Hypolimnetic oxygen demand (HOD) was observed to decrease after several years of HOS operation in all 3 waterbodies. Annual HOD decreased more for HOS operated year-round compared to recovery-based operation where HOS was started after DO decreased below 5.0 mg/L. Additionally, HOD throughout the summer stratified period was lower following maintenance of higher DO during spring prior to onset of stratification. An analogous result was observed during winter ice cover relative to DO conditions following fall turnover. Prior work showed increased HOD during HOS operation compared to pre-HOS. The increased HOD during HOS operation was termed diffuser-induced oxygen demand and implied the HOS was inducing oxygen demand. However, the current work shows HOS operation is not actually inducing oxygen demand; rather, it is addressing the existing oxygen demand already in the sediments. Our data suggest that a HOS is an effective tool to maintain hypolimnion DO in stratified lakes and reservoirs. Hypolimnetic oxygenation of water supply reservoirs improves water quality by preventing anoxia. This article summarizes the operational results using linear bubble plume diffuser hypolimnetic oxygenation systems installed in water supply reservoirs. The results obtained for 8 sites demonstrate that diffuser technology was effective at increasing hypolimnetic dissolved oxygen (DO) and spreading it to blanket sediments. The diffuser systems maintained increased DO in the hypolimnion during successive years of operation at every site. Improved oxygen levels reduced anoxic products and nutrients to mitigate the causes of taste and odor in the water supplied to water treatment facilities. This yielded additional treatment capacity, reduced treatment costs, and provided alternatives to water treatment plant modifications. The diffuser systems provide a simple and effective hypolimnetic oxygenation system, with 19 in operation. The hypolimnetic oxygenation systems (HOS) functions without a water pump and can obtain favorable oxygen transfer efficiencies. The diffusers are installed and maintained from the surface without divers. Diffusers provide an economical means to distribute oxygen input over large areas of the reservoir hypolimnion. Diffuser system installation costs run between $0.5M and $2.5M ($40 to $800 per hectare meter), with annual operating costs between $30K and $140K ($5 to $36 per hectare meter). Capital and operational costs vary depending on site specific conditions. With diffuser oxygenation, customer complaints were reduced and, in some cases, substantial monetary savings were realized by reduced treatment costs. This study compares performance of hypolimnetic aeration (HA) with hypolimnetic oxygenation (HO) from 1988 to 2018, over multiple project phases and a baseline period 1984 to 1986. Saint Paul Regional Water Services, Minnesota, pumps 170 MLD (45 MGD) from the Mississippi River through a chain of 4 lakes before treatment. Two, Pleasant Lake and Vadnais Lake, were equipped with partial-lift HA later replaced by linear diffuser HO. Ferric chloride was injected into inflows and into Vadnais at the HA or HO system. HA failed to relieve hypolimnetic hypoxia in Pleasant (median DO 1.7 mg/L) or Vadnais (median DO 2.7 mg/L). Median HO values were aerobic (Pleasant 4.2 mg/L, Vadnais 8.5 mg/L). Both HA and HO raised hypolimnetic oxidation reduction potential to high positive values from negative values associated with Mn and Fe solubilization. Compared to HA, HO reduced median hypolimnetic total Mn concentrations by 75% in Pleasant and in 93% in Vadnais, and hypolimnetic total Fe by 84% in Pleasant and 81% in Vadnais. HA significantly lowered median hypolimnetic TP from baseline, 0.552 mg/L to 0.146 mg/L in Pleasant, and 0.533 mg/L to 0.045 mg/L in Vadnais. HO median hypolimnetic TP was significantly lower at 0.053 mg/L in Pleasant, 0.030 mg/L in Vadnais. Superior performance of HO occurred at similar oxygen delivery rates: 1775 kg/d in Pleasant and 970 kg/d in Vadnais, versus HA of 1200 kg/d in each lake.
