Na última década, o gênero Hypericum ganhou repercussão mundial devido à utilização de Hypericum perforatum para obtenção de medicamentos antidepressivos. Por esta razão, a maioria dos estudos com outras espécies do gênero centra-se nesta atividade. Porém, um dos usos populares de espécies de Hypericum nativas do sul do Brasil é no tratamento de problemas gastrintestinais, inclusive como antiespasmódico. Neste trabalho, foi avaliado o efeito de uma das espécies de Hypericum nativas do Rio Grande do Sul, H. caprifoliatum, sobre as contrações induzidas por agonistas em íleo isolado de cobaio. (Continnuação) Foi investigado o efeito de um extrato ciclo-hexano purifi cado (isento de clorofi la e ceras), nas concentrações de 1, 3, 10 e 30 mg/mL, sobre curvas cumulativas de acetilcolina, histamina, potássio e serotonina (10-7 a 10-4 M). Na concentração de 30 mg/mL o extrato inibiu totalmente as contrações induzidas por todos os agonistas. Na concentração de 10 mg/mL, o extrato apresentou efeito antagonista nãocompetitivo de serotonina, reduzindo a contração máxima induzida por serotonina em cerca de 50 %. A resposta contrátil aos outros mediadores não foi alterada. Estes resultados indicam que espécies de Hypericum do sul do Brasil podem ser uma perspectiva interessante na busca de moléculas com atividade sobre a motilidade gastrintestinal. In the last decade the genus Hypericum has achieved worldwide recognition due to the therapeutic value of H. perforatum as an antidepressant drug. Consequently this activity is the most investigated one. However, species native to Brazil have other folk uses such as for the treatment of digestive disorders, including cramps. In this study we evaluated the effect of a purifi ed cyclohexane extract (chlorophyll and waxes free) (1,3,10 and 30 mg/mL) of H. caprifoliatum, a specie native to South Brazil, on isolated guinea pig ileum contractions induced by different mediators: serotonin, histamine, acetylcholine and potassium chloride (10-7 - 10-4 M). At 30 mg/mL all contractile responses were abolished. At 10 mg/mL only serotonin responses were altered: the extract reduced the maximal effect in 50%, which represents a non-competitive antagonism. At 1 and 3 mg/mL the extract was unable to modify all mediators response. These results point to native species of Hypericum as an interesting perspective in searching new molecules active on gastrointestinal motility.

The aim of the present study was to assess the analgesic activity of the aerial parts of two Hypericum species native to Southern Brazil, H. caprifoliatum and H. polyanthemum. The antinociceptive effect of the H. polyanthemum cyclohexane extract (POL; 180 mg/kg) and of the H. caprifoliatum methanol (MET) and cyclohexane (CH) extracts (90 mg/kg) was evaluated in the hot-plate (ip and po) and writhing (po) tests using male Swiss CF1 mice weighing 22-27 g (N = 10 per group). All extracts displayed antinociceptive effects in the hot-plate test (MET ip = 48%, MET po = 39%, CH ip = 27%, CH po = 50%, POL ip = 74%, and POL po = 49% compared to control). Pretreatment with naloxone (2.5 mg/kg, sc) abolished the effects of CH and POL, and partially prevented the analgesia induced by MET administered by the ip (but not by the po) route. POL and CH (po) significantly reduced the number of writhes induced by acetic acid, while MET was ineffective in this regard. We conclude that the antinociceptive effects of the H. caprifoliatum (CH) and H. polyanthemum (POL) hexane extracts seem to be mediated by the opioid system. Moreover, the antinociceptive activity of the H. caprifoliatum MET extract seems to depend on at least two chemical substances (or groups of substances) with distinct pharmacokinetic profiles and mechanisms of action. Only the naloxone-insensitive component of MET activity showed good bioavailability following oral administration.

Este trabalho teve como objetivo avaliar o efeito dos derivados floroglucinóis HC1 e hiperbrasilol B, presentes em Hypericum caprifoliatum, sobre a neurotransmissão glutamatérgica, canais de Na+ sensíveis à voltagem e atividade da Na+, K+ ATPase, visando aprofundar o estudo da atividade antidepressiva de Hypericum caprifoliatum. O envolvimento da neurotransmissão glutamatérgica no efeito antidepressivo de HC1 foi estudado através de manipulação farmacológica com MK-801 (antagonista NMDA) no teste da natação forçada, e captação e liberação de [3H]-glutamato em sinaptossomas de cérebro total de ratos. Os resultados demonstraram que pelo menos em nível de receptores NMDA e captação e liberação de glutamato, a neurotransmissão glutamatérgica não está envolvida na atividade antidepressiva de HC1. A veratrina, um ativador de canais de Na+, preveniu o efeito de HC1 no teste da natação forçada e três dias de tratamento com HC1 aumentaram a atividade da enzima Na+, K+ ATPase em córtex total e hipocampo de camundongos quando medida ex vivo 2h depois da última administração. Porém esta inibição desapareceu quando os animais foram submetidos a uma sessão de 6 minutos de nado forçado. Assim como o HC1, o hiperbrasilol B também teve seu efeito prevenido pela veratrina no teste da natação forçada, e o tratamento por três dias com hiperbrasilol B aumentou a atividade da enzima Na+, K+ ATPase apenas no hipocampo, e não no córtex total de camundongos. Os resultados deste trabalho reforçam a utilidade das estruturas diméricas de floroglucinóis na pesquisa por novas moléculas com ação antidepressiva e indicam que o hiperbrasilol B pode possuir propriedades moduladoras do humor ou anticonvulsivantes, uma vez que altera a atividade cerebral da Na+, K+ ATPase em camundongos e tem seu efeito na natação forçada alterado pela veratrina. Aiming to narrow the study of the antidepressant activity of Hypericum caprifoliatum, this work evaluated the effect of the phloroglucinol derivatives HC1 and hyperbrasilol B, present in H. caprifoliatum on glutamatergic neurotransmission, Na+ channels and Na+, K+ ATPase activity. The involvement of glutamatergic neurotransmission in the antidepressant effect of HC1 was investigated by pharmacological manipulation with MK-801 (NMDA antagonist) in the forced swimming test, and [3H]-glutamate uptake and release in total rat brain synaptosomes. The results demonstrated that at least at level of NMDA receptors and glutamate uptake and release, the glutamatergic neurotransmission is not involved in the antidepressant activity of HC1. Veratrine, a Na+ channel activator, prevented the effect of HC1 in the forced swimming test and three days of treatment with HC1 increased the enzymatic activity of Na+, K+ ATPase in mice brain cortex and hippocampus. However, this inhibition disappeared when animals were subjected to a of 6-minute forced swim session. Like HC1, the effect of hyperbrasilol B on forced swimming session was also prevented by veratrine; and treatment for three days with hyperbrasilol B increased the activity of Na+, K+ ATPase in hippocampus, but not in brain cortex of mice. The results of this work reinforce the usefulness of the dimeric phloroglucinols structures in the research for new molecules with antidepressant action and suggest that hyperbrasilol B may possess anticonvulsant and mood stabilizing properties, since it alters mice brain Na+, K+ ATPase activity and has its effect on the forced swimming test affected by veratrine.

Espécies de Hypericum (Guttiferae) são mundialmente reconhecidas por seus efeitos farmacológicos, destacando-se H. perforatum pela atividade antidepressiva. O estudo com espécies brasileiras vem apresentando resultados promissores. Hypericum polyanthemum e H. caprifoliatum mostraram efeito antinociceptivo, enquanto H. caprifoliatum mostrou atividade do tipo antidepressiva. Além disso, verificou-se atividade de H. polyanthemum sobre o carrapato Rhipicephalus (Boophilus) microplus. Para essas espécies, é descrita a presença de benzopiranos, derivados de floroglucinol e flavonóides. Objetivos: Isolar compostos fenólicos lipofílicos de H. polyanthemum e o flavonóide hiperosídeo de H. caprifoliatum, assim como avaliar atividades biológicas de algumas dessas substâncias. Materiais e Métodos: Realizou-se a análise química através de técnicas cromatográficas (CC, CCD) e espectroscópicas (1H e 13C RMN). Hiperosídeo foi avaliado nos seguintes modelos comportamentais: potenciação do sono barbitúrico, atividade locomotora espontânea, placa quente, contorções induzidas por ácido acético e natação forçada. O efeito antinociceptivo de isobutiril-5,7-dimetóxi-2,2-dimetilbenzopirano (HP1), 7-hidróxi-6-isobutiril-5-metóxi-2,2-dimetil-benzopirano (HP2) e 5-hidróxi-6-isobutiril-7-metóxi-2,2-dimetil-benzopirano (HP3), foi investigado, assim como sua atividade acaricida através do teste de imersão de larvas. Resultados e Conclusões: Além dos benzopiranos HP1, HP2 e HP3, dois compostos não relatados para a espécie H. polyanthemum, a xantona 6- desoxijacareubina e um benzopirano de estrutura similar ao ácido eriostemóico, foram isolados de extrato obtido por fluido supercrítico. Hiperosídeo mostrou efeito depressor sobre o SNC, e efeito antiimobilidade, que parece ser mediado pelo sistema dopaminérgico, em ratos. HP1 apresentou efeito antinociceptivo mediado pela neurotransmissão opióide. Nos experimentos para investigação da atividade acaricida, os benzopiranos mostraram ser, pelo menos em parte, responsáveis pela toxicidade dos extratos sobre os ácaros. The pharmacological effects of some Hypericum species are worldwide recognized, mostly by the comproved efficacy of H. perforatum as antidepressant. The south Brazilian species have been showing promissing results. Hypericum polyanthemum and H. caprifoliatum presented antinociceptive effect, while an antidepressant-like activity of H. caprifoliatum was reported. In addition, extracts from H. polyanthemum showed to be highly toxic to Rhipicephalus (Boophilus) microplus cattle tick. The chemical characterization reveals that these species are rich in benzopyrans, flavonoids and phloroglucinol derivatives. Objectives: To isolate lipophilic compounds from H. polyanthemum and the flavonoid hyperoside from H. caprifoliatum, as well as to carry out a biological evaluation of some of the isolated substances. Material and Methods: Chromatography and NMR spectroscopy techniques were performed for isolation and structure elucidation. Hyperoside was applied on behavior models, as the open field, pentobarbital sleeping potentiation, hot plate, acetic acid-induced writhing and the forced swimming test. The antinociceptive effect and the acaricide potential of 6-isobutyryl-5,7-dimethoxy- 2,2-dimethyl-benzopyran (HP1), 7-hydroxy-6-isobutyryl-5-methoxy-2,2-dimethylbenzopyran (HP2) and 5-hydroxy-6-isobutyryl-7-methoxy-2,2-dimethylbenzopyran (HP3) were also investigated. Results and conclusions: A benzopyran similar to the eriostemoic acid and the xanthone 6-deoxyjacareubin were isolated from the extract obtained by supercritical fluid. Hyperoside showed a central depressor effect and no toxicity signs. The present study provides evidences for the involvement of D2-like dopaminergic neurotransmission on the mechanism of hyperoside antidepressant-like action in rats. HP1 presented opioid-mediated antinociceptive effect. On larvae immersion test, the benzopyrans showed to account for the acaricide properties of H. polyanthemum extracts.

International audience; Since depressive patients present alterations in the hypothalamo-pituitary-adrenal (HPA) axis that are normalised by antidepressants, this HPA axis has been considered as a target of their actions. We have investigated the mechanism of action of a cyclohexane extract of Hypericum caprifoliatum (HCP), which displays antidepressant like activity, by studying, in mice, the influence of HCP and of two established antidepressant drugs, imipramine and bupropion, administered either acutely or semi-chronically (once a day, three consecutive days), on serum and brain cortex corticosterone levels, either in basal conditions or shortly after a forced-swimming session (FSS). Administered acutely, imipramine (20 mg/kg, per os (p.o.)), bupropion (30 mg/kg, p.o.) and HCP (360 mg/kg, p.o.) significantly reduced the immobility time and had no effects on FSS-induced increase of serum and cortical corticosterone levels. Conversely, 3 days repeated treatment with imipramine or bupropion resulted in a significant reduction of immobility time and FSS-induced increase of serum and cortical corticosterone levels. In a different way, repeated treatment with HCP significantly reduced the immobility time and only cortical corticosterone levels in stressed mice. These results indicate that short-term treatments with antidepressants are sufficient to induce modifications in the HPA axis reactivity to stress; and that apparently HCP has an influence on corticosterone levels by a mechanism diverse from the other tested antidepressants.

L’objectif du présent travail était d’étudier les extraits cyclohexaniques de deux espèces d’Hypericum, natives du sud du Brésil, Hypericum caprifoliatum (HCP) et Hypericum polyanthemum (POL), qui joueraient un rôle antidépresseur et un rôle analgésique, afin de caractériser les substances responsables de ces activités, ainsi que leurs mécanismes d’action. Dans l’épreuve de la nage forcée, HCP et POL, ainsi que leurs dérivés phloroglucinols (respectivement nommés HC1 et HP4) réduisent de façon significative la durée d’immobilité des rats et des souris, indiquant un effet de type antidépresseur potentiel de ces extraits. Cet effet est totalement réversé par le SCH23390 et le sulpiride (antagonistes respectifs des récepteurs D1 et D2 de la dopamine), suggérant l’implication du système dopaminergique dans l’effet type antidépresseur de HCP et POL. D’autre part, leurs fractions riches en phloroglucinol (HC1 et HP4) inhibent la capture synaptosomale de la dopamine ([3 H]-DA), de la noradrénaline ([3 H]-NA) et de la sérotonine ([3 H]-5HT), avec une certaine primauté d’action sur la capture de la DA. Cependant, cet effet ne semble pas dépendre d’une action directe sur les transporteurs des monoamines, puisque HC1 et HP4 n’affectent ni la liaison du [3 H]-mazindol, ni celle de la [3 H]-nisoxétine et ni celle du [3 H]-citalopram, respectivement aux transporteurs de la DA, de la NA et de la 5-HT. Par ailleurs, nous avons évalué l’effet des extraits sur les adaptations fonctionnelles des récepteurs monoaminergiques et opioïdergiques, grâce à la liaison de [35S]-GTPγS stimulée respectivement par les monoamines et les opioïdes Nous avons montré qu’administrés à la dose unique ou après trois traitements pendant 24 heures, HCP et POL provoquent une augmentation significative de la liaison de [35S]-GTPγS stimulée par la DA, la NA et la 5-HT, alors qu’après cinq jours de traitement, à raison de deux administrations quotidiennes, ces extraits réduisent de façon significative la liaison de [35S]-GTPγS stimulée par les monoamines. Il est important de noter que quelque soit le traitement, ni HCP, ni POL n’affecte de manière significative la liaison de [35S]-GTPγS stimulée par les opioïdes. Ces résultats démontrent que HCP et POL provoquent une altération fonctionnelle des transmissions monoaminergiques. Nous nous sommes intéressés, par la suite, aux effets de HCP et de POL sur l’axe corticotrope, et avons montré qu’administré sur trois jours, à raison d’une administration quotidienne, HCP et POL réduisent de façon significative l’augmentation du taux de corticostérone corticale induite par le stress de la nage forcée, sans affectée de manière significative celle du taux de corticostérone sérique, ni les taux de corticostérone plasmatique et corticale basale chez les animaux non soumis aux stress Par ailleurs, nous avons montré, qu’aux doses où ils exercent un effet type antidépresseur, HCP et POL induisent, dans le test de la plaque chaude, des effets analgésiques réversés par la naloxone (antagoniste des récepteurs opioïdes). Cet effet pourrait être attribué soit à d’autres substances, autres que HC1 et HP4, présentes dans les extraits; soit à une action indirecte de HC1 et HP4 sur les récepteurs opioïdes, puisque ni HC1, ni HP4 n’affectent de manière significative la liaison de la [3 H]-naloxone. En conclusion, l’ensemble de ce travail indique que les extraits de H. caprifoliatum et H. polyanthemum développent des effets de type analgésique et antidépresseur, qui pourraient être attribués à leurs dérivés phloroglucinols. Ce dernier effet semble résulter de l’activation d’une transmission monoaminergique, selon un mécanisme d’action différent de celui des antidépresseurs de références. O objetivo deste trabalho foi estudar a ação antidepressiva e analgésica de extratos ciclo-hexano de Hypericum caprifoliatum (HCP) e Hypericum polyanthemum (POL) e seus principais constituintes químicos, os derivados floroglucinol, HC1 e uliginosina (HP4), respectivamente. HCP, POL, HC1 e HP4 reduziram significativamente a imobilidade de ratos e camundongos no teste de natação forçada (FST), indicando potencial atividade antidepressiva. Os efeitos de HCP e POL no FST foram prevenidos pelo pré-tratamento com SCH23390 e sulpirida, antagonistas de receptores D1 e D2 respectivamente. Os extratos, HC1 e HP4 inibiram de modo dose-dependente a recaptação sinaptossomal de dopamina ([3 H]-DA), noradrenalina ([3 H]-NA) e serotonina ([3 H]-5HT), com uma maior potência para a recaptação de DA. Entretanto, este efeito não parece ser dependente de uma ação direta das substâncias sobre o transportador de monoaminas, uma vez que diferentes concentrações de HC1 e HP4 não afetaram a ligação de [3 H]-mazindol, [3 H]-nisoxetina e [3 H]- citalopram aos sítios de recaptação de DA, NA e 5-HT, respectivamente. Para avaliação do efeito dos extratos sobre os receptores de DA, NA, 5-HT e opióides, foi utilizada a técnica de ligação de [35S]-GTPγS estimulada por agonistas. O tratamento agudo de ratos com HCP ou POL aumentou significativamente a ligação de [35S]-GTPγS estimulada por DA, NA e 5-HT, enquanto 5 dias de tratamento diminuiram esta ligação Nenhum dos regimes de tratamento afetou a ligação estimulada por DAMGO, agonista opióide. Estes resultados demonstram que os tratamentos com HCP e POL resultam em alterações na transmissão monoaminérgica. Porém estas não se devem a efeitos diretos de HC1 e HP4 sobre os receptores, pois a incubação com HC1 ou HP4 não alterou a ligação de [35S]-GTPγS em nenhuma preparação estudada. Além do estudo sobre o sistema monoaminérgico, foi verificada a ação de HCP e POL sobre o eixo HPA através da medida dos níveis séricos e plasmáticos de corticosterona em camundongos submetidos ou não ao estresse da natação forçada. Três dias de tratamento com HCP e POL reduziram significativamente o aumento do nível cortical de corticosterona induzido pelo nado forçado, sem afetar o nível sérico, evidenciando que o tratamento repetido com os extratos afeta repostas hormonais ao estresse. Em relação à atividade analgésica, HCP e POL apresentaram efeito antinociceptivo na placa quente, o qual foi prevenido pelo pré-tratamento com naloxona.Entretanto, HC1 e HP4 não inibiram a ligação da [3 H]-naloxona, o efeito antinociceptivo de HP4 não foi prevenido pela naloxona, e o tratamento com os extratos, assim como a incubação direta HC1 ou HP4 não modificou a ligação de [35S]-GTPγS ao receptor opióide. Os resultados apresentados confirmam que HCP e POL têm potencial atividade antidepressiva e analgésica, provavelmente com um mecanismo de ação diferente dos fármacos atuais, sendo os derivados floroglucinol os prováveis responsáveis por estes efeitos. This work aimed to study the antidepressant and analgesic effect of Hypericum caprifoliatum (HCP) and H. polyanthemum (POL) cyclohexane extracts, as well as the substances and mechanisms involved on their activities. HCP, POL and their phloroglucinol derivatives (HC1 and HP4, respectively) significantly reduced rats and mice immobility time in the forced swimming test (FST), indicating potential antidepressant effect. The extracts showed some selectivity for activating the dopaminergic system. The antidepressant-like effect of HCP and POL was inhibited by D1 and D2 antagonists; and their extracts, HC1 and HP4, inhibited dopamine ([3 H]-DA), noradrenaline ([3 H]-NA) and serotonin ([3 H]-5HT) uptake, more potently of dopamine. However, this effect is not related to the binding to monoamine transporters, since they did not affect the binding of [3 H]-mazindol, [3 H]-nisoxetine and [3 H]- citalopram to DA, NA and 5-HT transporters, respectively. In order to investigate the functional role of DA, NA, 5-HT and opioid receptors in the effects of HCP and POL, we evaluated the extracts effect on monoamine and opioid receptor-stimulated [35S]-GTPγS binding. We demonstrated that acute or 3 treatments within 24h increases [35S]-GTPγS binding to DA, NA and 5-HT sites, while after 5 days of treatment the binding is reduced. Non treatment regimen affected binding to opioid sites. Hence, the results demonstrate that the treatment with HCP and POL induce alterations in monoaminergic transmission. As seen at the binding to monoamine transporters, these effects are not due to HC1 or HP4 interaction with the receptors, since direct incubation of HC1 and HP4 did no affect [35S]-GTPγS binding to membranes Besides studying HCP and POL effects on the monoaminergic system we investigated their activity on other system related to depression, the hypothalamus-pituitaryadrenal axis. We demonstrated that 3 days treatment with the extract reduce cortical corticosterone but not serum levels increased by FST. Regarding to the analgesic activity, HCP and POL antinociceptive effect in the hot plate were prevented by naloxone. However, HC1 and HP4 did not inhibited [3 H]-naloxone binding to opioid receptors. In conclusion, HCP and POL have potencial antidepressant and analgesic activities and the phloroglucinol derivatives are very likely to be the substances responsible for them. Even though the mechanism of action involves the monoaminergic system, the manner by which the extracts influence this system is different from classical antidepressants.

OBJETIVO: Desenvolver e validar a técnica de CLAE-DE para o doseamento de dopamina (DA), ácido diidrofenilacético (DOPAC), ácido homovanílico (HVA) e 3-metoxitiramina (3-MT); validar a técnica de microdiálise (MD) cerebral em animais conscientes e avaliar o efeito do tratamento agudo com um extrato lipofílico das partes aéreas de Hypericum caprifoliatum (HCP) sobre os níveis cerebrais de DA e seus metabólitos. MATERIAIS E MÉTODOS: A validação analítica foi realizada conforme o preconizado pela ANVISA, com análise dos parâmetros linearidade, precisão, exatidão, especificidade e limite inferior de quantificação. A técnica de microdiálise foi realizada de forma clássica: guias de sonda foram implantadas nas regiões cerebrais de interesse por cirurgia estereotáxica; os dialisados foram obtidos, 48 horas após a cirurgia, através da perfusão da sonda com líquido cérebroespinhal artificial (1 μL/min) e coleta durante três horas, em intervalos de 20 min; as três primeiras coletas foram utilizadas para determinação da linha de base e só então foram administrados os tratamentos. Para a validação da MD a sonda foi implantada no estriado (A: -0,3; L: + 3,2; P: - 4,5) e o tratamento foi sulfato de anfetamina (2 mg/kg, s.c.). Para a avaliação do efeito de HCP, a sonda foi implantada no núcleo acumbens (A: +2,2; L: -1,5; P: -5,8) e o tratamento foi uma dose de 270 mg/kg do extrato (v.o.). RESULTADOS E DISCUSSÃO: Os parâmetros avaliados para validar o método analítico ficaram dentro dos limites especificados pela ANVISA. O sulfato de anfetamina produziu um aumento nos níveis extracelulares de DA em 160% e uma redução de DOPAC e de HVA em 80% e 50%, respectivamente. O extrato HCP não alterou o conteúdo intersticial de DA, DOPAC e HVA no núcleo acumbens. Os valores basais encontrados estão de acordo com dados relatados na literatura. Não foi possível quantificar 3-MT. CONCLUSÃO: A metodologia analítica e as condições experimentais de microdiálise utilizadas permitiram a mensuração de mudanças induzidas por anfetamina nos níveis extracelulares de DA e seus principais metabólitos demonstrando que os fundamentos da técnica estão estabelecidos em nossas condições. O regime de tratamento empregado com HCP não foi suficiente para alterar os níveis de DA e seus principais metabólitos no núcleo acumbens de ratos. OBJECTIVE: The objectives of this work were to validate the HLPC-ED technique for measuring dopamine (DA), dihydroxyphenylacetic acid (DOPAC), homovanillic acid (HVA) and 3- methoxytyramine (3-MT); to put into operation the brain microdialysis technique (MD) in freely moving rats; to evaluate, by using MD, the effect of a single treatment with lipophilic extract from aerial parts of Hypericum caprifoliatum (HCP) on DA, DOPAC, HVA and 3-MT brain levels. MATERIALS AND METHODS: In order to validate the analytical technique we followed the parameters established by ANVISA for bioanalytical samples. The parameters were: precision, accuracy, specificity and also the lower limit of quantitation. The microdialysis technique was carried out following classical procedures: male Wistar rats were submitted to stereotaxic surgery for the guide cannula implantation in the brain regions of interest; 48 hours later, the microdiaysis probe was inserted and perfused with cerebrospinal fluid (1 μL/min). After baseline samples collecting (1 h), the treatments were administrated and samples were collected every 20 min, during 2h. To accomplish MD technique validation the animals were treated with amphetamine sulfate (2 mg/kg, s.c.) and the extracellular levels of DA, DOPAC e HVA were measured in striatum (A: -0,3; L: + 3,2; P: - 4,5). The effect of HCP acute treatment (270 mg/kg, p.o.) was evaluated in nucleus accumbens (A: +2,2; L: -1,5; P: -5,8). RESULTS AND DISCUSSION: The analytical parameters values found were within the acceptable ranges for bio-analytical limits specified by ANVISA. As expected, amphetamine sulfate administration significantly increased DA (160%) extracellular levels and decreased DOPAC (80%) and HVA (50%) levels. The quantification of 3-MT was not possible. Acute HCP administration did not affected DA, DOPAC and HVA levels in the nucleus accumbens. CONCLUSION: The HPLC-ED methodology and microdialysis procedures employed allowed the detection of amphetamineinduced changes in extracellular levels of DA and its metabolites demonstrating that the technical starting point was acceptable. The acute administration of 270 mg/kg, p.o. of HCP to rats was not sufficient to modify the extracellular levels of DA, DOPAC and HVA in the nucleus accumbens.
