Abstract

Searches for high frequency gravitational waves using cavities based on the Gertsenshtein effect were recently proposed, building off existing axion dark matter experiments. In particular, the sensitivity of axion dark matter experiments using metamaterial plasmas (tunable plasma haloscopes) to gravitational waves has not been explored in detail. Here we perform a full analysis of gravitational wave detection in plasma haloscopes, showing that the baseline design of experiments such as ALPHA is several orders of magnitude less sensitive than previously thought. We show how simple changes to the experiment can recover that sensitivity and lead to a powerful gravitational wave detector in the order of $(10-50)$ GHz frequency range., Comment: 15 pages, 7 figures

Abstract

Understanding material failure is critical for designing stronger and lighter structures by identifying weaknesses that could be mitigated. Existing full-physics numerical simulation techniques involve trade-offs between speed, accuracy, and the ability to handle complex features like varying boundary conditions, grid types, resolution, and physical models. We present the first foundation model specifically designed for predicting material failure, leveraging large-scale datasets and a high parameter count (up to 3B) to significantly improve the accuracy of failure predictions. In addition, a large language model provides rich context embeddings, enabling our model to make predictions across a diverse range of conditions. Unlike traditional machine learning models, which are often tailored to specific systems or limited to narrow simulation conditions, our foundation model is designed to generalize across different materials and simulators. This flexibility enables the model to handle a range of material properties and conditions, providing accurate predictions without the need for retraining or adjustments for each specific case. Our model is capable of accommodating diverse input formats, such as images and varying simulation conditions, and producing a range of outputs, from simulation results to effective properties. It supports both Cartesian and unstructured grids, with design choices that allow for seamless updates and extensions as new data and requirements emerge. Abstract

BACKGROUND: HVTN 120 is a phase 1/2a randomized double-blind placebo-controlled human immunodeficiency virus (HIV) vaccine trial that evaluated the safety and immunogenicity of ALVAC-HIV (vCP2438) and MF59- or AS01B-adjuvanted bivalent subtype C gp120 Env protein at 2 dose levels in healthy HIV-uninfected adults. METHODS: Participants received ALVAC-HIV (vCP2438) alone or placebo at months 0 and 1. At months 3 and 6, participants received either placebo, ALVAC-HIV (vCP2438) with 200 μg of bivalent subtype C gp120 adjuvanted with MF59 or AS01B, or ALVAC-HIV (vCP2438) with 40 μg of bivalent subtype C gp120 adjuvanted with AS01B. Primary outcomes were safety and immune responses. RESULTS: We enrolled 160 participants, 55% women, 18-40 years old (median age 24 years) of whom 150 received vaccine and 10 placebo. Vaccines were generally safe and well tolerated. At months 6.5 and 12, CD4+ T-cell response rates and magnitudes were higher in the AS01B-adjuvanted groups than in the MF59-adjuvanted group. At month 12, HIV-specific Env-gp120 binding antibody response magnitudes in the 40 μg gp120/AS01B group were higher than in either of the 200 μg gp120 groups. CONCLUSIONS: The 40 μg dose gp120/AS01B regimen elicited the highest CD4+ T-cell and binding antibody responses. Clinical Trials Registration . NCT03122223.

Abstract

The 3C 220.3 system is a rare case of a foreground narrow-line radio galaxy ("galaxy A," $z_A = 0.6850$) lensing a background submillimeter galaxy ($z_{{\rm SMG1}} = 2.221$). New spectra from MMT/Binospec confirm that the companion galaxy ("galaxy B") is part of the lensing system with $z_B = 0.6835$. New three-color HST data reveal a full Einstein ring and allow a more precise lens model. The new HST images also reveal extended emission around galaxy A, and the spectra show extended [OII] emission with irregular morphology and complex velocity structure. All indications are that the two lensing galaxies are a gravitationally interacting pair. Strong [OII] emission from both galaxies A and B suggests current star formation, which could be a consequence of the interaction. This would indicate a younger stellar population than previously assumed and imply smaller stellar masses for the same luminosity. The improved lens model and expanded spectral energy distributions have enabled better stellar-mass estimates for the foreground galaxies. The resulting dark matter fractions are ~0.8, higher than previously calculated. Deeper Chandra imaging shows extended X-ray emission but no evidence for a point X-ray source associated with either galaxy. The detection of X-rays from the radio lobes of 3C 220.3 allows an estimate of ~3 nT for the magnetic fields in the lobes, a factor of ~3 below the equipartition fields, as typical for radio galaxies., Comment: 23 pages, 10 figures, 10 tables. Abstract

Long-period radio transients are an emerging class of extreme astrophysical events of which only three are known. These objects emit highly polarised, coherent pulses of typically a few tens of seconds duration and minutes to hour-long periods. While magnetic white dwarfs and magnetars, either isolated or in binary systems, have been invoked to explain these objects, a consensus has not emerged. Here we report on the discovery of ASKAP J193505.1+214841.0 (henceforth ASKAPJ1935+2148) with a period of 53.8 minutes exhibiting three distinct emission states - a bright pulse state with highly linearly polarised pulses with widths of 10-50 seconds; a weak pulse state which is about 26 times fainter than the bright state with highly circularly polarised pulses of widths of approximately 370 milliseconds; and a quiescent or quenched state with no pulses. The first two states have been observed to progressively evolve over the course of 8 months with the quenched state interspersed between them suggesting physical changes in the region producing the emission. A constraint on the radius of the source for the observed period rules out a magnetic white dwarf origin. Unlike other long-period sources, ASKAPJ1935+2148 is the first to exhibit drastic variations in emission modes reminiscent of neutron stars. Abstract

Permutation Entropy and statistiCal Complexity Analysis for astRophysics (PECCARY) is a computationally inexpensive, statistical method by which any time-series can be characterized as predominately regular, complex, or stochastic. Elements of the PECCARY method have been used in a variety of physical, biological, economic, and mathematical scenarios, but have not yet gained traction in the astrophysical community. This study introduces the PECCARY technique with the specific aims to motivate its use in and optimize it for the analysis of astrophysical orbital systems. PECCARY works by decomposing a time-dependent measure, such as the x-coordinate or orbital angular momentum time-series, into ordinal patterns. Due to its unique approach and statistical nature, PECCARY is well-suited for detecting preferred and forbidden patterns (a signature of chaos), even when the chaotic behavior is short-lived or when working with a relatively short duration time-series or small sets of time-series data. A variety of examples are used to demonstrate the capabilities of PECCARY. These include mathematical examples (sine waves, varieties of noise, sums of sine waves, well-known chaotic functions), a double pendulum system, and astrophysical tracer particle simulations with potentials of varying intricacies. Since the adopted timescale used to diagnose a given time-series can affect the outcome, a method is presented to identify an ideal sampling scheme, constrained by the overall duration and the natural timescale of the system. 7. An Image-Based Search for Pulsar Candidates in the MeerKAT Bulge Survey

Abstract

We report on the results of an image-based search for pulsar candidates toward the Galactic bulge. We used mosaic images from the MeerKAT radio telescope, that were taken as part of a 173 deg**2 survey of the bulge and Galactic center of our Galaxy at L band (856-1712 MHz) in all four Stokes I, Q, U and V. The image root-mean-square noise levels of 12-17 uJy/ba represent a significant increase in sensitivity over past image-based pulsar searches. Our primary search criterion was circular polarization, but we used other criteria including linear polarization, in-band spectral index, compactness, variability and multi-wavelength counterparts to select pulsar candidates. We first demonstrate the efficacy of this technique by searching for polarized emission from known pulsars, and comparing our results with measurements from the literature. Our search resulted in a sample of 75 polarized pulsar candidates. Bright stars or young stellar objects were associated with 28 of these sources, including a small sample of highly polarized dwarf stars with pulsar-like steep spectra. Comparing the properties of this sample with the known pulsars, we identified 30 compelling candidates for pulsation follow-up, including two sources with both strong circular and linear polarization. The remaining 17 sources are either pulsars or stars, but we cannot rule out an extragalactic origin or image artifacts among the brighter, flat spectrum objects., Comment: 28 pages, 10 figures, 6 tables. Abstract

The nearby elliptical galaxy M87 contains one of the only two supermassive black holes whose emission surrounding the event horizon has been imaged by the Event Horizon Telescope (EHT). In 2018, more than two dozen multi-wavelength (MWL) facilities (from radio to gamma-ray energies) took part in the second M87 EHT campaign. The goal of this extensive MWL campaign was to better understand the physics of the accreting black hole M87*, the relationship between the inflow and inner jets, and the high-energy particle acceleration. Understanding the complex astrophysics is also a necessary first step towards performing further tests of general relativity. The MWL campaign took place in April 2018, overlapping with the EHT M87* observations. We present a new, contemporaneous spectral energy distribution (SED) ranging from radio to very high energy (VHE) gamma-rays, as well as details of the individual observations and light curves. We also conduct phenomenological modelling to investigate the basic source properties. We present the first VHE gamma-ray flare from M87 detected since 2010. The flux above 350 GeV has more than doubled within a period of about 36 hours. We find that the X-ray flux is enhanced by about a factor of two compared to 2017, while the radio and millimetre core fluxes are consistent between 2017 and 2018. We detect evidence for a monotonically increasing jet position angle that corresponds to variations in the bright spot of the EHT image. Our results show the value of continued MWL monitoring together with precision imaging for addressing the origins of high-energy particle acceleration. While we cannot currently pinpoint the precise location where such acceleration takes place, the new VHE gamma-ray flare already presents a challenge to simple one-zone leptonic emission model approaches, and emphasises the need for combined image and spectral modelling., Comment: 46 pages, 23 figures, accepted by Astronomy & Astrophysics on August. 29, 2024

Abstract

Dynamic 2-[18F] fluoro-2-deoxy-D-glucose positron emission tomography (dFDG-PET) for human brain imaging has considerable clinical potential, yet its utilization remains limited. A key challenge in the quantitative analysis of dFDG-PET is characterizing a patient-specific blood input function, traditionally reliant on invasive arterial blood sampling. This research introduces a novel approach employing non-invasive deep learning model-based computations from the internal carotid arteries (ICA) with partial volume (PV) corrections, thereby eliminating the need for invasive arterial sampling. We present an end-to-end pipeline incorporating a 3D U-Net based ICA-net for ICA segmentation, alongside a Recurrent Neural Network (RNN) based MCIF-net for the derivation of a model-corrected blood input function (MCIF) with PV corrections. The developed 3D U-Net and RNN was trained and validated using a 5-fold cross-validation approach on 50 human brain FDG PET datasets. The ICA-net achieved an average Dice score of 82.18% and an Intersection over Union of 68.54% across all tested scans. Furthermore, the MCIF-net exhibited a minimal root mean squared error of 0.0052. The application of this pipeline to ground truth data for dFDG-PET brain scans resulted in the precise localization of seizure onset regions, which contributed to a successful clinical outcome, with the patient achieving a seizure-free state after treatment. These results underscore the efficacy of the ICA-net and MCIF-net deep learning pipeline in learning the ICA structure's distribution and automating MCIF computation with PV corrections. This advancement marks a significant leap in non-invasive neuroimaging.

Abstract

PurposeTo compare rates of retinal nerve fiber layer change over time in healthy, eyes with nonprogressing glaucoma and eyes with progressing glaucoma using single wide-field (SWF) and optic nerve head (ONH) cube scan optical coherence tomography (OCT) images.MethodsForty-five eyes of 25 healthy individuals and 263 eyes of 161 glaucoma patients from the Diagnostic Innovations in Glaucoma Study were included. All eyes underwent 24-2 visual field testing and OCT (Spectralis SD-OCT) ONH and macular imaging. SWF images (up to 43° × 28°) were created by stitching together ONH cube scans centered on the optic disc and macular cube scans centered on the fovea. Visual field progression was defined as guided progression analysis likely progression and/or a significant (P < 0.01) mean deviation slope of less than -1.0 dB/year. Mixed effects models were used to compare rates of change. Highly myopic eyes were included.ResultsThirty glaucomatous eyes were classified as progressing. In eyes with glaucoma, mean global rate of change was -1.22 µm/year (P < 0.001) using SWF images and -0.83 µm/year (P = 0.003) using ONH cube scans. Rate of change was significantly greater in eyes with progressing glaucoma compared with eyes with nonprogressing glaucoma (-1.51 µm/year vs. -1.24 µm/year; P = 0.002) using SWF images and was similar using ONH cube scans (P = 0.27).ConclusionsIn this cohort that includes eyes with and without high axial myopia, the mean rate of retinal nerve fiber layer thinning measured using SWF images was faster in eyes with progressing glaucoma than in eyes with nonprogressing glaucoma. Wide-field OCT images including the ONH and macula can be effective for monitoring glaucomatous progression in patients with and without high myopia.

Abstract

IntroductionPalbociclib is a widely used treatment for advanced breast cancer in older adults. However, the existing evidence regarding its safety and tolerability in this age group is inconsistent and limited to retrospective subgroup or pooled analyses.Materials and methodsWe conducted a prospective single-arm multicenter phase 2 study to evaluate the safety and tolerability of palbociclib in participants aged 70 years or older with advanced hormone receptor-positive breast cancer. Participants were given palbociclib in combination with their physician's choice of endocrine therapy (letrozole or fulvestrant). The primary endpoint was the incidence of grade 3+ adverse events (AEs) by six months. Secondary endpoints included AE-related dose delays, dose reductions, early discontinuations, and hospitalizations. Additionally, we compared these endpoints by age groups (70-74 and ≥ 75 years).ResultsOf the 90 participants (median age 74 years [70-87]) enrolled, 75.6% (95% confidence interval [CI], 65.4-84.0) had grade 3+ AEs by six months. The most frequent grade 3+ AEs were neutropenia (61%), fatigue (4%), and nausea (3%). Febrile neutropenia was uncommon (1.1%). Due to AEs, 36% had dose delays, 34% had dose reductions, 10% had early discontinuations, and 10% had hospitalizations. Compared to those aged 70-74 years, participants aged ≥75 years had higher rates of early discontinuations (5.9% vs 15.9%, a difference of 9.5% [95% CI 3.5%-22.5%]).DiscussionPalbociclib has an overall favorable safety profile in adults aged ≥70 with advanced breast cancer. However, adults ≥75 years had a trend toward higher rates of AE-related early discontinuations compared to those 70-74 years. Abstract

We perform a set of flow and reactive transport simulations within three-dimensional fracture networks to learn the factors controlling mineral reactions. CO$_2$ mineralization requires CO$_2$-laden water, dissolution of a mineral that then leads to precipitation of a CO$_2$-bearing mineral. Our discrete fracture networks (DFN) are partially filled with quartz that gradually dissolves until it reaches a quasi-steady state. At the end of the simulation, we measure the quartz remaining in each fracture within the domain. We observe that a small backbone of fracture exists, where the quartz is fully dissolved which leads to increased flow and transport. However, depending on the DFN topology and the rate of dissolution, we observe a large variability of these changes, which indicates an interplay between the fracture network structure and the impact of geochemical dissolution. In this work, we developed a machine learning framework to extract the important features that support mineralization in the form of dissolution. In addition, we use structural and topological features of the fracture network to predict the remaining quartz volume in quasi-steady state conditions. As a first step to characterizing carbon mineralization, we study dissolution with this framework. We studied a variety of reaction and fracture parameters and their impact on the dissolution of quartz in fracture networks. We found that the dissolution reaction rate constant of quartz and the distance to the flowing backbone in the fracture network are the two most important features that control the amount of quartz left in the system. Abstract

Large-scale discrete fracture network (DFN) simulators are standard fare for studies involving the sub-surface transport of particles since direct observation of real world underground fracture networks is generally infeasible. While these simulators have seen numerous successes over several engineering applications, estimations on quantities of interest (QoI) - such as breakthrough time of particles reaching the edge of the system - suffer from a two distinct types of uncertainty. A run of a DFN simulator requires several parameter values to be set that dictate the placement and size of fractures, the density of fractures, and the overall permeability of the system; uncertainty on the proper parameter choices will lead to some amount of uncertainty in the QoI, called epistemic uncertainty. Furthermore, since DFN simulators rely on stochastic processes to place fractures and govern flow, understanding how this randomness affects the QoI requires several runs of the simulator at distinct random seeds. The uncertainty in the QoI attributed to different realizations (i.e. different seeds) of the same random process leads to a second type of uncertainty, called aleatoric uncertainty. In this paper, we perform a Sensitivity Analysis, which directly attributes the uncertainty observed in the QoI to the epistemic uncertainty from each input parameter and to the aleatoric uncertainty. We make several design choices to handle an observed heteroskedasticity in DFN simulators, where the aleatoric uncertainty changes for different inputs, since the quality makes several standard statistical methods inadmissible. Abstract

Despite its high prevalence among dementias, Lewy body dementia (LBD) remains poorly understood with a limited, albeit growing, evidence base. The public-health burden that LBD imposes is worsened by overlapping pathologies, which contribute to misdiagnosis, and lack of treatments. For this report, we gathered and analyzed public-domain information on advocacy, funding, research outputs, and the therapeutic pipeline to identify gaps in each of these key elements. To further understand the current gaps, we also conducted interviews with leading experts in regulatory/governmental agencies, LBD advocacy, academic research, and biopharmaceutical research, as well as with funding sources. We identified wide gaps across the entire landscape, the most critical being in research. Many of the experts participated in a workshop to discuss the prioritization of research areas with a view to accelerating therapeutic development and improving patient care. This white paper outlines the opportunities for bridging the major LBD gaps and creates the framework for collaboration in that endeavor. HIGHLIGHTS: A group representing academia, government, industry, and consulting expertise was convened to discuss current progress in Dementia with Lewy Body care and research. Consideration of expert opinion,natural language processing of the literature as well as publicly available data bases, and Delphi inspired discussion led to a proposed consensus document of priorities for the field.

Abstract

BackgroundElicitation of broad immune responses is understood to be required for an efficacious preventative HIV vaccine. This Phase 1 randomized controlled trial evaluated whether administration of vaccine antigens separated at multiple injection sites vs combined, fractional delivery at multiple sites affected T-cell breadth compared to standard, single site vaccination.MethodsWe randomized 90 participants to receive recombinant adenovirus 5 (rAd5) vector with HIV inserts gag, pol and env via three different strategies. The Standard group received vaccine at a single anatomic site (n = 30) compared to two polytopic (multisite) vaccination groups: Separated (n = 30), where antigens were separately administered to four anatomical sites, and Fractioned (n = 30), where fractions of each vaccine component were combined and administered at four sites. All groups received the same total dose of vaccine.FindingsCD8 T-cell response rates and magnitudes were significantly higher in the Fractioned group than Standard for several antigen pools tested. CD4 T-cell response magnitudes to Pol were higher in the Separated than Standard group. T-cell epitope mapping demonstrated greatest breadth in the Fractioned group (median 8.0 vs 2.5 for Standard, Wilcoxon p = 0.03; not significant after multiplicity adjustment for co-primary endpoints). IgG binding antibody response rates to Env were higher in the Standard and Fractioned groups vs Separated group.InterpretationThis study shows that the number of anatomic sites for which a vaccine is delivered and distribution of its antigenic components influences immune responses in humans.FundingNational Institute of Allergy and Infectious Diseases, NIH.
