The external store fitted to the aircraft may affect the flight characteristics and flight safety of the aircraft, which requires the analyses and testing on it. The purpose of this study is to identify and analyze types of failures that can affect the flight safety of aircraft due to the installation of external stores, and to check the flight safety of aircraft through dropping tests of the external stores. After identifying the types of failures that could affect the flight safety of the aircraft, the criticality was calculated to analyze the effect on the flight safety of the aircraft. Four types of failures were selected: unintentional dropping, failure of dropping, unintentional main wing deployment, and release of tail wing restraint of the external store, which are considered to affect the flight safety of the aircraft due to the operation of the external store. As a result of the aircraft's flight safety analysis on the failure types, the criticality requirements were met. Based on this, after obtaining the airworthiness certification, the drop test was successfully performed to confirm the flight safety of the aircraft by mounting an external store on the aircraft. However, in addition to the four hazards carried out in this study, the real external stores of the military aircraft may have various factors affecting the flight safety of the aircraft, so further research will be needed.

Two putative methylglyoxal synthases, which catalyze the conversion of dihydroxyacetone phosphate to methylglyoxal, from Oceanithermus profundus DSM 14,977 and Clostridium difficile 630 have been characterized for activity and thermal stability. The enzyme from O. profundus was found to be hyperthermophilic, with the optimum activity at 80 °C and the residual activity up to 59% after incubation of 15 min at 95 °C, whereas the enzyme from C. difficile was mesophilic with the optimum activity at 40 °C and the residual activity less than 50% after the incubation at 55 °C or higher temperatures for 15 min. The structural analysis of the enzymes with molecular dynamics simulation indicated that the hyperthermophilic methylglyoxal synthase has a rigid protein structure with a lower overall root-mean-square-deviation value compared with the mesophilic or thermophilic counterparts. In addition, the simulation results identified distinct regions with high fluctuations throughout those of the mesophilic or thermophilic counterparts via root-mean-square-fluctuation analysis. Specific molecular interactions focusing on the hydrogen bonds and salt bridges in the distinct regions were analyzed in terms of interatomic distances and positions of the individual residues with respect to the secondary structures of the enzyme. Key interactions including specific salt bridges and hydrogen bonds between a rigid beta-sheet core and surrounding alpha helices were found to contribute to the stabilisation of the hyperthermophilic enzyme by reducing the regional fluctuations in the protein structure. The structural information and analysis approach in this study can be further exploited for the engineering and industrial application of the enzyme.

The evaluation of the morphology and organization of collagen fibers is critical in understanding wound healing and tissue remodeling after a thermal injury of the skin. However, histological analysis conducted by pathologists is often labor-intensive and limited to qualitative evaluations and scoring within a narrow field of view. In this study, we propose a convolutional neural network (CNN) model to classify Masson’s trichrome (MT)-stained histology images of burn-induced scar tissue and to characterize the microstructures of normal tissue and scar tissue in a quantitative manner. The scar tissue is created on in vivo rodent models and prepared for MT-stained histology slides after wound healing. A CNN model is developed, trained, and tested with various sizes of the histology images for classification and characterization. The proposed model classifies both normal tissue (i.e., without burn, as the control) and scar tissue at various scales with over 97% accuracy. The features acquired from the proposed CNN model visually characterizes the density and directional variance of the collagen fibers distributed in the dermal layers from whole histology images. The proposed deep learning technique can provide an objective and reliable method to rapidly assess and quantify wound repair and remodeling.

Background Bioethanol is one of the most representative eco-friendly fuels developed to replace the non-renewable fossil fuels and is the most successful commercially available bio-conversion technology till date. With the availability of inexpensive carbon sources, such as cellulosic biomass, bioethanol production has become cheaper and easier to perform, which can facilitate the development of methods for converting ethanol into higher value-added biochemicals. In this study, a bioconversion process using Pseudomonas putida as a biocatalyst was established, wherein ethanol was converted to mevalonate. Since ethanol can be converted directly to acetyl-CoA, bypassing its conversion to pyruvate, there is a possibility that ethanol can be converted to mevalonate without producing pyruvate-derived by-products. Furthermore, P. putida seems to be highly resistant to the toxicity caused by terpenoids, and thus can be useful in conducting terpenoid production research. Results In this study, we first expressed the core genes responsible for mevalonate production (atoB, mvaS, and mvaE) in P. putida and mevalonate production was confirmed. Thereafter, through an improvement in genetic stability and ethanol metabolism manipulation, mevalonate production was enhanced up to 2.39-fold (1.70 g/L vs. 4.07 g/L) from 200 mM ethanol with an enhancement in reproducibility of mevalonate production. Following this, the metabolic characteristics related to ethanol catabolism and mevalonate production were revealed by manipulations to reduce fatty acid biosynthesis and optimize pH by batch fermentation. Finally, we reached a product yield of 0.41 g mevalonate/g ethanol in flask scale culture and 0.32 g mevalonate/g ethanol in batch fermentation. This is the highest experimental yield obtained from using carbon sources other than carbohydrates till date and it is expected that further improvements will be made through the development of fermentation methods. Conclusion Pseudomonas putida was investigated as a biocatalyst that can efficiently convert ethanol to mevalonate, the major precursor for terpenoid production, and this research is expected to open new avenues for the production of terpenoids using microorganisms that have not yet reached the stage of mass production.

The current study aims to evaluate the dependence of laser-induced optical breakdown (LIOB) on skin types by using 1064 nm picosecond laser with micro-lens arrays (MLA) and diffractive optical elements (DOE). Both black and white skin tissues were examined to comparatively assess the LIOB effects in the skin in terms of laser-induced vacuolization. The black skin irradiated at 3.0 J/cm2 demonstrated that MLA yielded a deeper distribution (180-400 μm) of laser-induced vacuoles with a size of 67 μm, compared to DOE (180-280 μm; 40 μm in size). However, the white skin presented that MLA created larger vacuoles (134 μm in size) in a smaller number at deeper distributions (125-700 μm) than MLA with the black skin. DOE generated no laser-induced vacuolization in the white skin. The white skin tissue with inherent higher scattering could be responsible for deeper vacuolization after the picosecond laser treatment. Further investigations are expected to determine the optimal treatment conditions for various skin types.

Carbon dioxide (CO2) emissions are related to global warming. However, CO2 can be used as an abundant and cheap carbon source for production of valuable chemicals using carbon capture and storage technology. Here, the genes related to carbon flux toward pyruvate biosynthesis in E. coli were deleted to enhance the incorporation of CO2 for succinate production. The codonoptimized carbonic anhydrase gene (SP(-)HCCA) derived from Hahella chejuensis KCTC 2396 and the phosphoenolpyruvate carboxylase gene (ppc) of E. coli W3110 were co-overexpressed to enhance carbon flux toward oxaloacetate synthesis in E. coli. Finally, we constructed SGJS134, which shows the highest production of succinate derived from CO2 compared with other strains. SGJS134 produced approximately 6.5 mM succinate from CO2 and yielded approximately 13.0 mM succinate per dry cell weight. These results may be useful for enhancing the incorporation of CO2 for succinate production in E. coli. Additionally, the metabolic engineering method used in this study will propose the potential of E. coli to convert CO2 to valuable chemicals.

In November 2021, 14 international travel-related severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) B.1.1.529 (omicron) variant of concern (VOC) patients were detected in South Korea. Epidemiologic investigation revealed community transmission of the omicron VOC. A total of 80 SARS-CoV-2 omicron VOC-positive patients were identified until December 10, 2021 and 66 of them reported no relation to the international travel. The current study aims to investigate the effects of micro-lens arrays (MLA) and diffractive optical elements (DOE) on skin tissue via intra-dermal laser-induced optical breakdown (LIOB) after irradiation of 1064-nm picosecond laser light at high energy settings. Irradiation with MLA and DOE was tested on dimming paper, tissue-mimicking phantom, and dark pigmented porcine skin to quantitatively compare distributions of micro-beams, micro-bubbles, and laser-induced vacuoles in the skin. DOE yielded more uniform distributions of the micro-beams on the paper and laser-induced micro-bubbles in the phantom, compared to MLA. The ex vivo skin test confirmed that the DOE-assisted irradiation accompanied more homogeneous generation of the micro-beams on the tissue surface (deviation of ≤ 3%) and a high density of small laser-induced vacuoles (∼78 µm) in the dermis than the MLA-assisted irradiation (deviation of ∼26% and ∼163 µm). The DOE-assisted picosecond laser irradiation may help to achieve deep and uniformly-generated vacuolization under the basal membrane after intra-dermal LIOB for effective fractional skin treatment.

Endoscopic submucosal dissection (ESD) is clinically used to remove early gastric cancer in stomach. The aim of the current study is to examine a therapeutic capacity of pulsed Ho:YAG laser for the development of laser-assisted ESD under various surgical parameters. Ex vivo porcine stomach tissue was ablated with 1-J Ho:YAG pulses at 10 Hz at different number of treatments (NT = 1, 2, and 3) and treatment speeds (TS = 0.5, 1, and 2 mm/s) without and with saline injection. Regardless of saline injection, straight tissue ablation showed that ablation depth increased with increasing NT and decreasing TS. At NT = 3 and TS = 0.5 mm/s, no saline injection yielded the maximum ablation depth (3.4 ± 0.3 mm), partially removing muscularis propria. However, saline injection confined the tissue ablation within a submucosal layer (2.1 ± 0.3 mm). Thermal injury was found to be 0.7~1.1 mm in the adjacent tissue with superficial carbonization. Circular tissue ablation (2 cm in diameter) at NT = 3 and TS = 0.5 mm/s presented that no saline injection yielded a reduction in the lesion area, whereas saline injection maintained the ablated lesion area. Histological analysis revealed that unlike no saline injection, saline injection ablated the entire mucosal layer without perforation in the muscular propria. The pulsed Ho:YAG laser can be a potential surgical tool for clinical ESD to incise a target lesion without adverse perforation. Further investigations will validate the efficacy and safety of the Ho:YAG laser-assisted ESD in in vivo porcine stomach models for clinical translation.

The conversion of ricinoleic acid from renewable sources to long-chain α,ω-dicarboxylic acids or w-hydroxyl carboxylic acids by microbial processes is constrained by toxicity issues. Here, we demonstrate the possible role of Corynebacterium glutamicum as a new microbial strategy for the biotransformation of fatty acids. The established strain Escherichia coli failed to grow at 5 mM n-heptanoic acid, while the specific growth rate of C. glutamicum declined by 28%. We partially constructed a previously designed multistep biocatalytic pathway in C. glutamicum, and confirmed that the C. glutamicum biocatalyst successfully converted ricinoleic acid to undec-9-enoic acid, heptyl ester via 12-keto-oleic acid. We investigated the effects of cultivation and reaction temperatures, and the type and concentration of non-ionic detergent on recombinant C. glutamicum whole-cell bioconversion. At a cultivation temperature of 30 °C and a reaction temperature of 35 °C, and in the presence of 0.09 g/L Triton X-100, the whole-cell C. glutamicum biocatalyst produced 0.8 mM undec-9-enoic acid, heptyl ester from 1.9 mM 12-ketooleic acid. It also generated 0.7 mM undec-9-enoic acid, heptyl ester from 5.5 mM ricinoleic acid. This is the first report of undec-9-enoic acid, heptyl ester production using a recombinant C. glutamicum-based biocatalyst.

Air pollution sources and the hazards of high particulate matter 2.5 (PM2.5) concentrations among air pollutants have been well documented. Shipping emissions have been identified as a source of air pollution, therefore, it is necessary to predict air pollutant concentrations to manage seaport air quality. However, air pollution prediction models rarely consider shipping emissions. Here, the PM2.5 concentrations of the Busan North and Busan New Ports were predicted using a recurrent neural network and long short-term memory model by employing the shipping activity data of Busan Port. In contrast to previous studies that employed only air quality and meteorological data as input data, our model considered shipping activity data as an emission source. The model was trained from 1 January 2019 to 31 January 2020 and predictions and verifications were performed from 1–28 February 2020. Verifications revealed an index of agreements (IOA) of 0.975 and 0.970 and root mean square errors of 4.88 and 5.87 µg/m3 for Busan North Port and Busan New Port, respectively. Regarding the results based on the activity data, a previous study reported an IOA of 0.62–0.84, with a higher predictive power of 0.970–0.975. Thus, the extended approach offers a useful strategy to prevent PM2.5 air pollutant-induced damage in seaports.

Here, Corynebacterium glutamicum ATCC13032 expressing Baeyer–Villiger monooxygenase from Pseudomonas putida KT2440 was designed to produce 9-(nonanoyloxy) nonanoic acid from 10-ketostearic acid. Diverse parameters including cultivation and reaction temperatures, type of detergent, and pH were found to improve biotransformation efficiency. The optimal temperature of cultivation for the production of 9-(nonanoyloxy) nonanoic acid from 10-ketostearic acid using whole cells of recombinant C. glutamicum was 15 °C, but the reaction temperature was optimal at 30 °C. Enhanced conversion efficiency was obtained by supplying 0.05 g/L of Tween 80 at pH 7.5. Under these optimal conditions, recombinant C. glutamicum produced 0.28 mM of 9-(nonanoyloxy) nonanoic acid with a 75.6% (mol/mol) conversion yield in 2 h. This is the first report on the biotransformation of 10-ketostearic acid to 9-(nonanoyloxy) nonanoic acid with a recombinant whole-cell C. glutamicum-based biocatalyst and the results demonstrate the feasibility of using C. glutamicum as a whole-cell biocatalyst.

Over the past decade, nanofluidic diodes that rectify ionic currents (i.e. greater current in one direction than in the opposite direction) have drawn significant attention in biomolecular sensing, switching and energy harvesting devices. To obtain current rectification, conventional nanofluidic diodes have utilized complex nanoscale asymmetry such as nanochannel geometry, surface charge density, and reservoir concentration. Avoiding the use of sophisticated nano-asymmetry, micro/nanofluidic diodes using microscale asymmetry have been recently introduced; however, their diodic performance is still impeded by (i) low (even absent) rectification effects at physiological concentrations over 100 mM and strong dependency on the bulk concentration, and (ii) the fact that they possess only passive predefined rectification factors. Here, we demonstrated a new class of micro/nanofluidic diode with an ideal perm-selective nanoporous membrane based on ion concentration polarization (ICP) phenomenon. Thin side-microchannels installed near a nanojunction served as mitigators of the amplified electrokinetic flows generated by ICP and induced convective salt transfer to the nanoporous membrane, leading to actively controlled micro-scale asymmetry. Using this device, current rectifications were successfully demonstrated in a wide range of electrolytic concentrations (10-5 M to 3 M) as a function of the fluidic resistance of the side-microchannels. Noteworthily, it was confirmed that the rectification factors were independent from the bulk concentration due to the ideal perm-selectivity. Moreover, the rectification of the presenting diode was actively controlled by adjusting the external convective flows, while that of the previous diode was passively determined by invariant nanoscale asymmetry.

Carbon dioxide (CO2) is an abundant and cheap carbon source that is partly responsible for global warming in the atmosphere. The objective of this study was to construct a recombinant E. coli strain that can show enhanced production of succinate derived from CO2. In this study, we confirmed the enhancement of utilization by analyzing succinate containing one carbon-13 (13C) derived from 13CO2. Firstly, the carbonic anhydrase gene (SP(-)HCCA) derived from Hahella chejuensis KCTC 2396 was over-expressed to enhance carbon flux toward bicarbonate ion (HCO3-) synthesis in E. coli. The phosphoenolpyruvate carboxylase gene (ppc) was over-expressed to enhance the production of oxaloacetate by enhancing the carbon flux. Compared with the control strain, the percentage of the succinate containing one 13C (succinate119) to total succinate was enhanced by approximately 2.80-fold and the amount of succinate119 also increased by approximately 4.09-fold in SGJS120. Secondly, the lactate dehydrogenase gene (ldhA) was deleted to re-direct the utilization of the carbon source from glucose to enhance succinate production in SGJS120. However, ldhA deletion did not increase CO2 utilization in SJGS120. Finally, the phosphotransferase system gene (ptsG) and pyruvate kinase F gene (pykF) were deleted to increase the amount of phosphoenolpyruvate (PEP). SGJS126 (pykF deletion strain) showed the highest increase, which was 6.05-fold higher than the control strain. From the results, SP(-)HCCA overexpression and pykF deletion may be useful for enhancing CO2 utilization in E. coli. Additionally, engineered strains showed the potential to reduce the cost of succinate production by using an industrially cheaper carbon source such as CO2 and converting CO2 to a valuable chemical.

Simple Summary Colorectal cancer is one of the most common cancers and the third leading cause of cancer-related deaths in the United States. As a non- or minimally invasive cancer treatment, photothermal therapy (PTT) has been widely used to generate irreversible thermal injuries in tumors. However, conventional PTT employs an end-firing flat fiber to deliver laser energy, leading to the incomplete removal of tumor tissues due to an uneven beam distribution over the tumor surface. Multi-lens arrays (MLA) generate multiple micro-beams to uniformly distribute laser energy on the tissue surface. Therefore, the application of MLA for PTT in cancer affords a spatially enhanced distribution of micro-beams and laser-induced temperature in the tumor. The purpose of the current study is to computationally and experimentally demonstrate the therapeutic benefits of MLA-assisted fractional PTT on colorectal cancer, in comparison to flat fiber-based PTT. Abstract Conventional photothermal therapy (PTT) for cancer typically employs an end-firing flat fiber (Flat) to deliver laser energy, leading to the incomplete treatment of target cells due to a Gaussian-shaped non-uniform beam profile. The purpose of the current study is to evaluate the feasibility of multi-lens arrays (MLA) for enhanced PTT by delivering laser light in a fractional micro-beam pattern. Computational and experimental evaluations compare the photothermal responses of gelatin phantoms and aqueous dye solutions to irradiations with Flat and MLA. In vivo colon cancer models have been developed to validate the therapeutic capacity of MLA-assisted irradiation. MLA yields 1.6-fold wider and 1.9-fold deeper temperature development in the gelatin phantom than Flat, and temperature monitoring identified the optimal treatment condition at an irradiance of 2 W/cm2 for 180 s. In vivo tests showed that the MLA group was accompanied by complete tumor eradication, whereas the Flat group yielded incomplete removal and significant tumor regrowth 14 days after PTT. The proposed MLA-assisted PTT spatially augments photothermal effects with the fractional micro-beams on the tumor and helps achieve complete tumor removal without recurrence. Further investigations are expected to optimize treatment conditions with various wavelengths and photosensitizers to warrant treatment efficacy and safety for clinical translation.

The study was conducted for optimal design of hybrid power generation system (HPS) in which diesel generators and renewable energy generators are incorporated to supply electricity to islands isolated from the national grid. In order to optimally design capacity and operation planning of the hybrid system, hourly electricity demand data is needed over 8760 h of a year. The required hourly data was simulated from monthly electricity consumption reports and EnergyPlus-based building models. An optimization method that matches hourly supply and demand may encounter problems of exhaustive computing time and ‘out of memory’ because size of associated matrices increases in multiples of 8760 × 8760. The problems were resolved with sparse matrices and the linear programming algorithm. Linear constraints necessary for the linear programming were developed in this study. According to the simulation result with the target island (Deokjeok Island), the optimization method recommends a hybrid generation system rather than a conventional diesel only system due to high fuel cost. And the capacity design reflects renewable energy resources abundant in the island. And designed operation planning illustrates good hourly matching of demand and supply over 8760 h with minimal excess electricity, which validates integrity of the proposed design method.

In this study, phosphoenolpyruvate carboxylase (PEPC) derived from Oceanimonas smirnovii (OS) was expressed as a soluble protein in Escherichia coli BL21(DE3). We isolated OS-PEPC (a recombinant PEPC protein) by his-tag purification. The purified protein showed a single band upon analysis with SDS-PAGE, and it had an apparent molecular mass of 98 kDa. Pufied OS-PEPC showed a specific activity value of 21.8 ± 0.495 U/mg protein. Especially, OS-PEPC showed the enzymatic activity between 40 and 50 °C. It maintained enzymatic activity in basic pH conditions (pH value, 9-10). We also measured OS-PEPC PEP and HCO3 (-) saturation kinetics and confirmed the effect of divalent cation on OS-PEPC activity.

To produce 10-ketostearic acid from oleic acid.Oleic acid was converted to 10-ketostearic acid by a recombinant Corynebacterium glutamicum ATCC 13032 expressing oleate hydratase from Stenotrophomonas maltophilia and a secondary alcohol dehydrogenase from Micrococcus luteus under the control of a synthetic constitutive promoter. Optimal conditions for 10-ketostearic acid production were pH 7.5 and 30 °C with 5 g cells l(-1) and 2.5 g oleic acid l(-1). Under these conditions, the cells produced 1.96 g 10-ketostearic acid l(-1) from oleic acid in 6 h, with a conversion yield of 78 % (w) and a maximum volumetric productivity of 1.67 g l(-1) h(-1).This is the first report of 10-ketostearic acid production using a recombinant C. glutamicum.

The use of planning support systems in urban planning and management provides objective information that is beneficial in the decision-making stage. The existing planning support system, however, lacks in the consideration of ecological properties. In this study, a planning support system development that is capable of reflecting ecological elements was set as the ultimate goal. This system can support planning processes ranging from ecological potential anaylsis to alternatives designing and stimulation. Additionally, factors such as soil, water, climate, biotope, ecological network and recreation can be handled according to their natural potentials. The establishment and visualization of land use planning that addresses ecological characteristics based on the analyzed results are possible. Therefore, this system could operate as a framework to assist planners and decision makers through a computer-based system to provide useful ecological information and to allow effective decision-making by analyzing, displaying and visualizing ecological spatial data.

Carbon dioxide (CO

The biocatalytic efficiency of recombinant Corynebacterium glutamicum ATCC 13032 expressing the secondary alcohol dehydrogenase of Micrococcus luteus NCTC2665 was studied. Recombinant C. glutamicum converts ricinoleic acid to a product, identified by gas chromatography/mass spectrometry as 12-ketooleic acid (12-oxo-cis-9-octadecenoic acid). The effects of pH, reaction temperature, and non-ionic detergent on recombinant C. glutamiucm whole cell bioconversion were examined. The determined optimal conditions for production of 12-ketooleic acid are pH 8.0, 35°C, and 0.05 g/l Tween80. Under these conditions, recombinant C. glutamicum produces 3.3 mM 12-ketooleic acid, with a 72% (mol/mol) maximum conversion yield, and 1.1 g/l/h volumetric productivity in 2 h; and 3.9 mM 12- ketooleic acid, with a 74% (mol/mol) maximum conversion yield, and 0.69 g/l/h maximum volumetric productivity in 4 h of fermentation. This study constitutes the first report of significant production of 12-ketooleic acid using a recombinant Corynebacterium glutamicum-based biocatalyst.
