Background/Aims There have been numerous efforts to reduce mother-to-child transmission (MTCT) of hepatitis B virus (HBV) with antiviral agents during pregnancy. However, there are limited data regarding the outcomes of pregnant women after delivery. This study was performed to evaluate the efficacy of antiviral agents in preventing MTCT of HBV and maternal long-term outcomes. Methods The HBV-infected pregnant women treated with antiviral agents to prevent MTCT were retrospectively reviewed. Forty-one pregnant women who received telbivudine or tenofovir during late pregnancy (28-34 week) were analyzed. Hepatitis B virus surface antibody (HBsAb) positivity was tested in 43 infants after 7 months of birth. Eleven mothers were followed >1 year after delivery. Results The mean HBV DNA titer before antiviral therapy was 8.67 (6.60–9.49) log copies/mL, and the median age at delivery was 32 years (range, 22–40). Eleven patients were treated with tenofovir and 30 with telbivudine. The median duration was 57 days (range, 23–100), and the median HBV DNA titer at birth was 5.06 log copies/mL (range, 2.06–6.50). Antiviral treatments were associated with significant HBV DNA reduction (P12 months, and an antiviral agent was administered. Conclusions Antiviral treatment during late pregnancy effectively reduced MTCT. Long-term follow-up should be required in such cases. In addition, given that maternal biochemical flare occurred in 18% of mothers, re-administration of antiviral agents might be required.

2-Deoxy-scyllo-inosose (DOI) has been a valuable starting natural product for the manufacture of pharmaceuticals or chemical engineering resources such as pyranose catechol. DOI synthase, which uses glucose-6-phosphate (Glc6P) as a substrate for DOI biosynthesis, is indispensably involved in the initial stage of the biosynthesis of 2-deoxystreptamine-containing aminoglycoside antibiotics including butirosin, gentamicin, kanamycin, and tobramycin. A number of metabolically engineered recombinant strains of Bacillus subtilis were constructed here; either one or both genes pgi and pgcA that encode Glc6p isomerase and phosphoglucomutase, respectively, was (or were) disrupted in the sugar metabolic pathway of the host. After that, three different DOI synthase–encoding genes, which were artificially synthesized according to the codon preference of the B. subtilis host, were separately introduced into the engineered recombinants. The expression of a natural btrC gene, encoding DOI synthase in butirosin-producing B. circulans, in the heterologous host B. subtilis (BSDOI-2) generated approximately 2.3 g/L DOI, whereas expression of an artificially codon-optimized tobC gene, derived from tobramycin-producing Streptomyces tenebrarius, into the recombinant of B. subtilis (BSDOI-15) in which both genes pgi and pgcA are disrupted significantly enhanced the DOI titer: up to 37.2 g/L. Fed-batch fermentation by the BSDOI-15 recombinant using glycerol and glucose as a dual carbon source yielded the highest DOI titer (38.0 g/L). The development of engineered microbial cell factories empowered through convergence of metabolic engineering and synthetic biology should enable mass production of DOI. Thus, strain BSDOI-15 will surely be a useful contributor to the industrial manufacturing of various kinds of DOI-based pharmaceuticals and fine chemicals.

Purpose: To identify the average turning point by comparing the learning curves of two surgeons learning to perform strabismus surgery. Materials and Methods: Patients who underwent procedures to correct exotropia between January 2010 and December 2014 followed for at least 3 months were retrospectively assessed. The first 70 patients on whom each of two ophthalmologists (A and B) performed surgery to treat strabismus were divided into 7 cohorts comprising 10 patients each based on the chronological order of the surgery. Factors, including patient age, preoperative angle of deviation, operative time, and success or failure of the operation, were compared between the two surgeons. Learning curves were calculated based on changes in operative time and operation success rate. Operation success was determined by measuring the angle of deviation at a distance of 5 m 3 months after the operation. Results: A turning point was observed after 40 cases for Surgeon A and 50 cases for Surgeon B based on the operative time learning curve. No turning point was observed in the operation success rate learning curve based on the absence of a specific trend. Success rate by cohort was not significantly different between the two surgeons (P > 0.05). Surgeon B had a significantly longer mean operative time than Surgeon A (P = 0.045). Conclusions: Approximately 50 cases are required for an ophthalmologist to reach a turning point in strabismus surgery. This outcome can be used as a guideline when training surgeons to perform strabismus surgery.

BackgroundRocuronium has been well known to produce withdrawal response in 50-80% patients when administered intravenously. Several drugs are administered prior injection of rocuronium to prevent the withdrawal response. We compared the preventive effect of lidocaine, ketamine, and remifentanil on the withdrawal response of rocuronium.MethodsA total of 120 patients undergoing various elective surgeries were enrolled. Patients were allocated into 4 groups according to the pretreatment drugs (Group N, normal saline; Groups L, lidocaine 40 mg; Group K, ketamine 0.5 mg/kg; Group R, remifentanil 1 µg/kg). Patients received drugs prepared by dilution to 3 ml volume before injection of rocuronium. Withdrawal responses after injection of rocuronium were graded on a 4-point scale. Hemodynamic changes were observed before and after administration of pretreatment drugs and after endotracheal intubation.ResultsIncidence of withdrawal response was significantly lower in group L (20%), group K (30%), and group R (0%), than group N (87%). Severe withdrawal response was observed in 5 of the 30 patients (17%) in group L, and in 9 of the 30 patients (30%) in group K. There was no severe withdrawal response in group R. Mean blood pressure and heart rate were significantly decreased in group R compared to other groups.ConclusionsIt seems that remifentanil (1 µg/kg intravenously) was the strongest and most effective in prevention of withdrawal response after rocuronium injection among the 3 drugs.

BackgroundHypoxemia during one-lung ventilation (OLV) remains a major concern. The present study compared the effect of alveolar recruitment strategy (ARS) on arterial oxygenation during OLV at varying tidal volumes (Vt) with or without positive end-expiratory pressure (PEEP).MethodsIn total, 120 patients undergoing wedge resection by video assisted thoracostomy were randomized into four groups comprising 30 patients each: those administered a 10 ml/kg tidal volume with or without preemptive ARS (Group H and Group H-ARS, respectively) and those administered a 6 ml/kg tidal volume and a 8 cmH2O PEEP with or without preemptive ARS (Group L and Group L-ARS, respectively). ARS was performed using pressure-controlled ventilation with a 40 cmH2O plateau airway pressure and a 15 cmH2O PEEP for at least 10 breaths until OLV began.ResultsPreemptive ARS significantly improved the PaO2/FiO2 ratio compared to the groups that did not receive ARS (P < 0.05). The H-ARS group showed a highest PaO2/FiO2 ratio during OLV, the L-ARS and H groups showed similarly improved arterial oxygenation, which was significantly higher than in group L (P < 0.05). The plateau airway pressure in group H-ARS was significantly higher than in group L-ARS (P < 0.05).ConclusionsPreemptive ARS can improve arterial oxygenation during OLV. Furthermore, a 6 ml/kg tidal volume combined with 8 cmH2O PEEP after preemptive ARS may reduce the risk of pulmonary injury caused by high tidal volume during one-lung ventilation in patients with normal pulmonary function.

Pluripotency transcription programs by core transcription factors (CTFs) might be reset during M/G1 transition to maintain the pluripotency of embryonic stem cells (ESCs). However, little is known about how CTFs are governed during cell cycle progression. Here, we demonstrate that the regulation of Oct4 by Aurora kinase b (Aurkb)/protein phosphatase 1 (PP1) during the cell cycle is important for resetting Oct4 to pluripotency and cell cycle genes in determining the identity of ESCs. Aurkb phosphorylates Oct4(S229) during G2/M phase, leading to the dissociation of Oct4 from chromatin, whereas PP1 binds Oct4 and dephosphorylates Oct4(S229) during M/G1 transition, which resets Oct4-driven transcription for pluripotency and the cell cycle. Aurkb phosphor-mimetic and PP1 binding-deficient mutations in Oct4 alter the cell cycle, effect the loss of pluripotency in ESCs, and decrease the efficiency of somatic cell reprogramming. Our findings provide evidence that the cell cycle is linked directly to pluripotency programs in ESCs.

Background. In gastric carcinogenesis, changes of DNA methylation appear to be an early molecular event, and the genome-wide methylation state is closely correlated with the level of long interspersed nucleotide element-1 (LINE-1) methylation. In this study, we measured LINE-1 methylation level according to genetic instability and evaluated the effect of Helicobacter pylori infection on genetic instability in gastric epithelial dysplasia. Methods. Total 100 tissue samples of gastric epithelial dysplasia were analyzed. Seven loci that linked to tumor suppressor genes were used to identify significant structural chromosomal aberrations. Microsatellite status was investigated for two different microsatellite marker loci (BAT25 and BAT26). Also, we measured LINE-1 methylation level by combined bisulfite restriction analysis (COBRA-LINE-1) method. Results. There were no significant differences of LINE-1 methylation level according to chromosomal/microsatellite instability and H. pylori state. Abstract An operational scheme for predicting the symmetric R30 and R50 of tropical cyclones (TCs) in the western North Pacific was developed using a statistical regression method and track pattern clustering (four clusters). The statistical–dynamical model employs multiple linear regressions of two to eight variables at each cluster and forecast lead time. The dependent variable for prediction was the change in the 5‐kt wind radius (R5)—a proxy of TC size—relative to the initial time. The performance of the model was compared for the training (2008–2016) and testing (2017–2018) periods. The effect of clustering on TC size prediction was evaluated by comparing the performance of the non‐clustering and clustering models. The clustering model improved the prediction of TC size by 3%–24% at all lead times during the training period, especially with a significant improvement of up to 43% in Cluster 2. In Cluster 2, because most TCs tend to develop strongly and continue to increase in size, it greatly reduced the variability in TC size through clustering, allowed for smarter predictor selection, and ultimately improved TC size prediction. In the real‐time R30 and R50 predictions for the 2017 and 2018 TCs, the error of the clustered model was 18%–19% less than that of the non‐clustered model. The analysis results revealed that the real‐time prediction errors of the current model increase when the TC tracks are difficult to classify into specific clusters, the predicted environments and TC tracks are inaccurate, and the size and intensity of a TC rapidly increase.

Abstract Background Megakaryocytes (MKs) are platelet precursors, which arise from hematopoietic stem cells (HSCs). While MK lineage commitment and differentiation are accompanied by changes in gene expression, many factors that modulate megakaryopoiesis remain to be uncovered. Replication initiation determinant protein (RepID) which has multiple histone-code reader including bromodomain, cryptic Tudor domain and WD40 domains and Cullin 4-RING E3 ubiquitin ligase complex (CRL4) recruited to chromatin mediated by RepID have potential roles in gene expression changes via epigenetic regulations. We aimed to investigate whether RepID-CRL4 participates in transcriptional changes required for MK differentiation. Methods The PCR array was performed using cDNAs derived from RepID-proficient or RepID-deficient K562 erythroleukemia cell lines. Correlation between RepID and DAB2 expression was examined in the Cancer Cell Line Encyclopedia (CCLE) through the CellMinerCDB portal. The acceleration of MK differentiation in RepID-deficient K562 cells was determined by estimating cell sizes as well as counting multinucleated cells known as MK phenotypes, and by qRT-PCR analysis to validate transcripts of MK markers using phorbol 12-myristate 13-acetate (PMA)-mediated MK differentiation condition. Interaction between CRL4 and histone methylation modifying enzymes were investigated using BioGRID database, immunoprecipitation and proximity ligation assay. Alterations of expression and chromatin binding affinities of RepID, CRL4 and histone methylation modifying enzymes were investigated using subcellular fractionation followed by immunoblotting. RepID-CRL4-JARID1A-based epigenetic changes on DAB2 promoter were analyzed by chromatin-immunoprecipitation and qPCR analysis. Results RepID-deficient K562 cells highly expressing MK markers showed accelerated MKs differentiation exhibiting increases in cell size, lobulated nuclei together with reaching maximum levels of MK marker expression earlier than RepID-proficient K562 cells. Recovery of WD40 domain-containing RepID constructs in RepID-deficient background repressed DAB2 expression. CRL4A formed complex with histone H3K4 demethylase JARID1A in soluble nucleus and loaded to the DAB2 promoter in a RepID-dependent manner during proliferation condition. RepID, CRL4A, and JARID1A were dissociated from the chromatin during MK differentiation, leading to euchromatinization of the DAB2 promoter. Conclusion This study uncovered a role for the RepID-CRL4A-JARID1A pathway in the regulation of gene expression for MK differentiation, which can form the basis for the new therapeutic approaches to induce platelet production. Sedimentable solids generated in aquaponic systems are mainly composed of organic waste, presenting molecules such as phytate, which can be a potential source of inorganic nutrients through mineralization. This work aimed to isolate and identify phytase-producing bacteria and evaluate the inoculation effects of pure strains on mineralization and nutrient release from solid waste generated in aquaponic systems at different oxygen and temperature conditions. The bacteria were isolated from the settleable solids of a commercial aquaponic system and molecularly identified by amplifying the 16S rRNA gene. Subsequently, two tests were carried out: 1. Test for the biochemical identification of phytase-producing bacteria; 2. In vitro mineralization test, where the ability to mineralize phytate and release nutrients under different oxygen conditions [0 rpm (2.1 mg L−1) and 200 rpm (7.8 mg L−1)] and temperatures (24 and 37 °C) were evaluated. Our findings show that two pure strains of Lysinibacillus mangiferihumi can mineralize phytate under conditions of 200 rpm and 24 °C, mainly increasing the mineralization of PO4- and Ca, a property that has not yet been reported for this species. On the other hand, at 0 rpm and 24 °C, an increase in K was observed (control conditions), while the conditions of 200 rpm and 24 °C, regardless of bacterial inoculation, favored a rise in S, Mg, and Fe. The Lysinibacillus strains obtained in this investigation are of great importance due to their application in agriculture and the optimization of mineralization in aquaponic systems. A proper combination of oxygen and temperature will lead to a greater availability of nutrients for the growth and development of vegetables.

The demand for high-quality strawberries continues to grow, emphasizing the need for innovative agricultural practices to enhance both yield and fruit quality. In this context, the utilization of natural products, such as biostimulants, has emerged as a promising avenue for improving strawberry production while aligning with sustainable and eco-friendly agricultural approaches. This study explores the influence of a bacterial filtrate (BF), a vegetal-derived protein hydrolysate (PH), and a standard synthetic auxin (SA) on strawberry, investigating their effects on yield, fruit quality, mineral composition and metabolomics of leaves and fruits. Agronomic trial revealed that SA and BF significantly enhanced early fruit yield due to their positive influence on flowering and fruit set, while PH treatment favored a gradual and prolonged fruit set, associated with an increased shoot biomass and sustained production. Fruit quality analysis showed that PH-treated fruits exhibited an increase of firmness and soluble solids content, whereas SA-treated fruits displayed lower firmness and soluble solids content. The ionomic analysis of leaves and fruits indicated that all treatments provided sufficient nutrients, with heavy metals within regulatory limits. Metabolomics indicated that PH stimulated primary metabolites, while SA and BF directly affected flavonoid and anthocyanin biosynthesis, and PH increased fruit quality through enhanced production of beneficial metabolites. This research offers valuable insights for optimizing strawberry production and fruit quality by harnessing the potential of natural biostimulants as viable alternative to synthetic compounds.

Abstract The importance of vesicular monoamine transporter 2 (VMAT2) in dopamine regulation, which is considered crucial for neuropsychiatric disorders, is currently being studied. Moreover, the development of disease treatments using histone deacetylase (HDAC) inhibitors (HDACi) is actively progressing in various fields. Recently, research on the possibility of regulating neuropsychiatric disorders has been conducted. In this study, we evaluated whether VMAT2 expression increased by an HDACi can fine‐tune neuropsychotic behavior, such as attention deficit hyperactivity disorder (ADHD) and protect against the cell toxicity through oxidized dopamine. First, approximately 300 candidate HDACi compounds were added to the SH‐SY5Y dopaminergic cell line to identify the possible changes in the VMAT2 expression levels, which were measured using quantitative polymerase chain reaction. The results demonstrated, that treatment with pimelic diphenylamide 106 (TC‐H 106), a class I HDACi, increased VMAT2 expression in both the SH‐SY5Y cells and mouse brain. The increased VMAT2 expression induced by TC‐H 106 alleviated the cytotoxicity attributed to 6‐hydroxydopamine (6‐OHDA) or 1‐methyl‐4‐phenylpyridinium (MPP+) and free dopamine treatment. Moreover, dopamine concentrations, both intracellularly and in the synaptosomes, were significantly elevated by increased VMAT2 expression. These results suggest that dopamine concentration regulation by VMAT2 expression induced by TC‐H 106 could alter several related behavioral aspects that was confirmed by attenuation of hyperactivity and impulsivity, which were major characteristics of animal model showing ADHD‐like behaviors. These results indicate that HDACi‐increased VMAT2 expression offers sufficient protections against dopaminergic cell death induced by oxidative stress. Thus, the epigenetic approach could be considered as therapeutic candidate for neuropsychiatric disease regulation.

The Yellow Sea (YS) is exposed to various weather systems, such as typhoons, monsoon activities, and extratropical cyclones, which can pose a major threat to the adjacent coastal regions through the development of energetic oceanic surface waves. Unusually severe surface wave events in the YS occur with considerable frequency during the boreal spring (March-April-May), but have received less attention compared to winter and summer. This study focuses on the characteristics of spring extreme wave height (EWH) events in the YS during 1979-2022, based on observational and long-term reanalysis datasets. Our analysis shows that extreme waves, i.e. daily maximum heights in the top 5% of all spring days, take about 12 h to build up to the peaks, while they decay more slowly after the peaks. During the extreme events, the Siberian High is found to extend anomalously eastward compared to spring climatology. Such an anomalous extension contributes to the increase of the sea level pressure gradient and the intensification of the surface wind speed in the YS. Meanwhile, in the range of 6 ∼ 24 h following the peaks of the EWH events in the YS, swells induced by strong northerly winds begin to have an impact in the YS. These swells contribute to maintaining higher wave energy levels in the YS for longer after the atmospheric source has been removed. We further explore the large-scale environmental conditions that could provide the predictability of extreme waves in the basin developed by these findings. This study presents implications for assessing the risks associated with extreme waves in coastal regions and for improving coastal management strategies in the YS.

The prevalence of attention deficit hyperactivity disorder (ADHD), a developmental neuropsychiatric disorder, is high among children and adolescents. The pathogenesis of ADHD is mediated with genetic, biological, and environmental factors. Most therapeutic drugs for ADHD have so far targeted biological causes, primarily by regulating catecholaminergic neurotransmitters. However, ADHD drugs that are clinically treated have various problems in their addictiveness and drug stability; thus, it is recommended that efficacy and safety should be secured through simultaneous prescription of multiple drugs rather than a single drug treatment. Accordingly, it is necessary to develop drugs that newly target pathogenic mechanisms of ADHD. In this study, we attempt to confirm the possibility of developing new drugs by reviewing dopamine-related developmental mechanisms of neurons and their correlation with ADHD. Histone deacetylase inhibitors (HDACi) can regulate the concentration of intracellular dopamine in neurons by expressing vesicular monoamine transporter 2 and inducing the exocytosis of neurotransmitters to the synaptic cleft, thereby promoting the development of neurons and signal transmission. This cellular modulation of HDACi is expected to treat ADHD by regulating endogenous catecholamines such as dopamine. Although studies are still in the preclinical stage, HDAC inhibitors clearly have potential as a therapeutic agent with low addictiveness and high efficacy for ADHD treatment.

pH is the major issue that concerns all producers in aquaponics, as the main three organisms (aquatic animal, plant, and microbes) have different preferences. Additional C is a potential approach to amend the growing environment and improve shrimp and plant growth, and microbe establishment. Aquaponics under saline conditions has, however, not been studied in detail in regard to the effect of pH and additional C. In this study, we evaluate the impact of pH and additional C on the growth of Pacific whiteleg shrimp and five edible plants (three halophytes and two glycophytes) in marine aquaponic systems using nutrient film technique (NFT). The results indicated that plants grow better in both pH 6.5 treatments; however, additional C improved the growth in pH 7.5 + C treatment and had similar yield to lower pH treatments. The results indicated both pH and additional C had little impact on shrimp growth. In conclusion, adding C can be a practical solution to the pH conundrum for marine aquaponics. Appling additional C was suggested for the operation of marine aquaponic food production system when the pH is high.

Plant-derived protein hydrolysates (PHs) are gaining global interest for their sustainability and positive effects on crops under abiotic stresses. However, the long-term effects of PHs on the yield and quality of greenhouse crops have not been described. Romaine lettuce (Lactuca sativa L.) and Micro-Tom tomato (Solanum lycopersicum L.) were grown with commercial growing media in 1 L pots and fertigated with four N levels (2, 5, 10, and 15 mM). PH (0 or 3 g/L) was applied as a foliar spray (PH-F) or root drench (PH-R) once every week. Compared to PH-F, PH-R effectively stimulated the yield, photosynthesis, water-use efficiency, chlorophyll contents, and antioxidant activities, and compounds regardless of N levels and species. Increasing the N level led to a total dry weight gain, and PH-R enhanced the lettuce shoot dry weight (+31%) and tomato fruit dry weight (+22%). PH-R also increased the fresh marketable yield of lettuce (+21%) and tomato (+32%). The increasing N level decreased antioxidant parameters in lettuce leaves, not in tomato fruits, whereas PH-R improved them in both species. PH-R significantly enhanced the N use and uptake efficiency. Taken together, our results suggested that the addition of PH-R effectively increased the N uptake and subsequently increased the lettuce and tomato yield and quality regardless of N levels.

Abstract A statistical model for typhoon‐induced accumulated rainfall (TAR) prediction over the Korean Peninsula using track, intensity and rainfall data of 91 typhoons affecting the peninsula during the period 1977–2014 is developed. The statistical estimation of the TAR consists of three steps: (1) estimating the TAR at 56 observational weather stations for the 91 typhoons; (2) selecting typhoons whose tracks are similar to that of the target typhoon within the area of 32–40 ° N and 120–138 ° E using a fuzzy C‐mean clustering method; and (3) calculating the mean TAR for the 16 selected typhoons based on track similarity after an intensity correction of the TAR using a linear regression between the TAR and intensity anomaly. To validate the model, real case predictions were performed on typhoons Chan‐hom and Goni in 2015 and compared with the observed TARs as well as with those from local and global operational models. The result showed that when the best‐track data are used, the present statistical model can predict the TAR with the accuracy of mean root mean square errors (RMSEs) of 33 mm (Chan‐hom) and 29 mm (Goni) at the 56 stations, which were much less than the results of the local model. With the predicted track and intensity data for the two typhoons, the present model also showed an overall good performance with RMSEs of 30–34 mm (Chan‐hom) and 29–49 mm (Goni), depending on the accuracy of the predicted track and intensity, which were generally less than the results of the global model.

Climate change: tricking the trees Warmer winter temperatures suppress flowering of litchi trees, threatening fruit production, but researchers have found another way to trigger litchi trees to reproduce. In litchi, spring buds contain both flower and leaf primordia, and cold cues the plants to shed the young leaves, allowing the flower primordia to develop. With recent warmer winters, the leaves are crowding out the flowers. Biyan Zhou at South China Agricultural University investigated whether spraying with compounds that make the trees produce stress-signaling molecules, such as nitrous oxide, would mimic the effects of a cold winter. Following treatment, the trees shed their rudimentary leaves and flowers developed. Microscopic examination of buds showed that the leaf shedding process looked similar to cold-triggered shedding. This study offers a method to induce litchis to flower and fruit, even in a changing climate.

Agronomic trial revealed that SA and BF significantly enhanced early fruit yield due to their positive influence on flowering and fruit set, while PH treatment favored a gradual and prolonged fruit set, associated with an increased shoot biomass and sustained production. Fruit quality analysis showed that PH-treated fruits exhibited an increase of firmness and soluble solids content, whereas SA-treated fruits displayed lower firmness and soluble solids content. The ionomic analysis of leaves and fruits indicated that all treatments provided sufficient nutrients, with heavy metals within regulatory limits. Metabolomics indicated that PH stimulated primary metabolites, while SA and BF directly affected flavonoid and anthocyanin biosynthesis, and PH increased fruit quality through enhanced production of beneficial metabolites. This research offers valuable insights for optimizing strawberry production and fruit quality by harnessing the potential of natural biostimulants as viable alternative to synthetic compounds. Aquaponics is a circulating and sustainable system that combines aquaculture and hydroponics and forms a symbiotic relationship between fish, plants, and microorganisms. We hypothesized that feed alone could support plant growth, but the symbiosis with fish adds some beneficial effects on plant growth in aquaponics. In this study, we created three closed culture systems, namely, aquaponics, hydroponics without nitrogen (N) and phosphorus (P), and aquaculture, and added the same amount of feed containing N and P to all the treatments in order to test the hypothesis. Accumulation of NO3− and PO43− was alleviated in aquaponics and hydroponics as a result of plant uptake. Lettuce plants grown in aquaponics grew vigorously until 2 weeks and contained a constant level of N in plants throughout the production period, whereas those in hydroponics grew slowly in the early stage and then vigorously after 2 weeks with a late increment of N concentration. These results suggest that catfish help with the faster decomposition of the feed, but, in hydroponics, feed can be slowly dissolved and decomposed owing to the absence of the fish. The bacterial community structures of the culture solution were investigated using 16S rRNA gene amplicon sequencing. At the class level, Actinobacteria, Alphaproteobacteria, Betaproteobacteria, and Gammaproteobacteria were the major microbial groups in the solutions. Aquaponics prevented the pollution of tank solution and maintained a higher water quality compared with hydroponics and aquaculture, suggesting that aquaponics is a more sustainable cultivation system even in a small-scale system.

Our previous study reported that fresh produce grown in aquaponic and hydroponic systems can pose potential food safety hazards due to an accidental introduction of contaminated fish and cross-contamination between the systems. In this study, we examined the effects of plant species and age on the likelihood and level of internalization of Shiga toxin-producing Escherichia coli (STEC) in aquaponic and hydroponic systems. Four plant species, basil (Ocimum basilicum L. cv. Genovese), cilantro (Coriandrum Sativum L.), lettuce (Lactuca sativa cv. Cherokee), and kale (Brassica oleracea var. sabellica), received root damage treatment as seedlings before transplanting or mature plants at three weeks after transplanting by cutting off 1-cm tips of one-third of the roots. Enrichments and selective media were used for the isolation, and presumptive positive colonies were confirmed by PCR for the presence of stx1 gene in plant tissues, recirculating water, and fish feces collected at four weeks after transplanting. In hydroponic systems, STEC was found neither in the solution nor in the roots and leaves of all four plant species, possibly through improved sanitation and hygiene practices. However, consistent with our previous findings, STEC was found in the water, on the plant roots, and in the fish feces in aquaponic systems, even after thorough sanitation prior to the study. Regardless of plant age, STEC was internalized in the roots of all plant species when the roots were damaged, but there was no difference in the degree of internalization with STEC among plant species. STEC was present in the leaves only when seedlings received root damage treatment and were grown to maturity, indicating that root damage allows STEC to internalize in the roots within a week, but a longer period is required for STEC to internalize into the leaves. We concluded that root damage on seedlings can cause the internalization of E. coli O157:H7 in the edible parts of leafy vegetables and herbs in soilless production systems.

Plant-derived protein biostimulants exhibit hormone-like activities promoting plant growth and yield, yet detailed investigations on hormonal function have remained limited. This study was conducted to investigate the effects of vegetal-derived-biostimulant on morphological and metabolic changes in cuttings of three herbaceous species demonstrating different rooting ability, basil (Ocimum basilicum L.), tomato (Solanum lycopersicum L.), and chrysanthemum (Chrysanthemum indicum L.), in comparison to auxin. Unrooted cuttings were applied with or without biostimulant (100, 1000, 5000, and 10,000 mg L−1) or auxin [1% indole-3-butyric acid (IBA) plus 0.5% 1-naphthaleneacetic acid (NAA); 100, 200, 300, and 500 mg L−1] as a basal quick-dip, stuck into inert media, and evaluated at 20 days after placement under intermittent mist. Both compounds increased adventitious rooting in all cuttings. Biostimulant required a significantly higher threshold for a series of adventitious rooting responses than auxin, and the maximum effectiveness was achieved at 5000 mg L−1 for biostimulant and 100, 200, and 300 mg L−1 for auxin in basil, tomato, and chrysanthemum, respectively. Adventitious rooting responses (dry mass and length) to biostimulant showed a gradual logarithmic rise as a function of increasing dosages, which was not in agreement with biphasic dose-response of auxin. Biostimulant significantly increased or tended to increase fine roots in all tested cuttings, which was not consistent with auxin. Relatively high levels of endogenous brassinosteroids (BRs) were present in non-treated cuttings of basil, tomato, and chrysanthemum in decreasing order. Both compounds had no effects or concomitantly increased or decreased BR levels in plant tissues, with fewer effects on basil and tomato, containing high BR levels, but more prominent effects on chrysanthemum, containing relatively low BR levels. Contrasting effects of biostimulant and auxin were found in antioxidant activities, which were promoted by biostimulant but inhibited by auxin either in roots or shoots. These results indicate that the hormonal effects of vegetal-derived biostimulant are primarily exerted by BR-mediated processes while involving interaction with auxin. Both the biostimulant-derived BRs and auxin were suggested to modulate endogenous BR pool via overlapping and interdependent regulatory functions, inducing morphological and metabolic changes during adventitious rooting of cuttings in a plant species-specific manner.
