Abstract

Cell-penetrating peptides enter cells via diverse mechanisms, such as endocytosis, active transport, or direct translocation. For the design of orally delivered cell-penetrating peptides, it is crucial to know the contribution of these different mechanisms. In particular, the ability of a peptide to translocate through a lipid bilayer remains a key parameter for the delivery of cargos. However, existing approaches used to assess translocation often provide discrepant results probably because they have different sensitivities to the distinct translocation mechanisms. Here, we focus on the passive permeation of a range of hydrophobic cyclic peptides inspired by somatostatin, a somatotropin release-inhibiting factor. Using droplet interface bilayers (DIB), we assess the passive membrane permeability of these peptides and study the impact of the peptide cyclization and backbone methylation on translocation rates. Cyclization systematically improved the permeability of the tested peptides while methylation did not. By studying the interaction of the peptides with the DIB interfaces, we found membrane insertion and peptide intrinsic diffusion to be two independent factors of permeability. Compared to the industrial gold standard Caco-2 and parallel artificial membrane permeability assay (PAMPA) models, DIBs provide intermediate membrane permeability values, closer to Caco-2. Even for conditions where Caco-2 and PAMPA are discrepant, the DIB approach also gives results closer to Caco-2. Thereupon, DIBs represent a robust alternative to the PAMPA approach for predicting the permeability of peptides, even if the latter present extremely small structural differences.

Abstract

International audience; The phase diagram of the H2O-Zn(NO3)2-Al(NO3)3-Cu(NO3)2 quaternary system at 30 °C has been established by using the conductivity measurements. The solid-liquid equilibria of the H2O-Zn(NO3)2-Al(NO3)3, H2O-Zn(NO3)2-Cu(NO3)2, H2O-Al(NO3)3-Cu(NO3)2 ternary systems and two isoplethic sections were determined experimentally. The solid phases in equilibrium with the saturated solution are the tri- and hemipentahydrate of copper nitrate, the hexahydrate α and β of the zinc nitrate and the nonahydrate of aluminum nitrate. The copper and zinc nitrates are relatively soluble in opposition to the aluminum nitrate which presents some important precipitation domains.

Abstract

The study deals with the direct oxidation kinetics of micronic cobalt metal particles and its simulation for the complete transition from metal to ceramic. The simulation was also experimentally verified. All the three possible interfaces, Co/CoO, CoO/Co3O4 and Co3O4/O2 (air), have been taken into consideration for the simulation. The complete oxidation kinetics has been investigated from the thermogravimetric studies under isothermal conditions in the temperatures 973–1173 K. A quantitative interpretation based on the diffusion of Co or oxygen ions through the grown oxide layer has been proposed. The activation energy for the oxidation kinetics calculated from the Arrhenius law was 161 ± 20 kJ mol−1.

Abstract

Oxygen diffusion coefficients were determined in a commercial ZnO-based varistor by means of the gas-solid exchange method using the isotope 18O as the oxygen tracer. The diffusion annealings were performed at 892, 942, 992 and 1092oC, in an Ar + 18O2 atmosphere under an oxygen partial pressure of 0.2 atm. After the diffusion annealings, the 18O diffusion profiles were established by secondary ion mass spectrometry (SIMS). The results show an increase of the oxygen diffusion in the varistor, both in bulk and in grain boundaries, when compared to the oxygen diffusion in undoped ZnO. The increase of the oxygen bulk diffusion in the varistor agrees with an interstitial mechanism for the oxygen diffusion. The results also show that the grain boundary is a fast path for the oxygen diffusion in the varistor. However, the oxygen diffusion in the grain boundaries of the varistor seems to depend on several chemical and microstructural parameters and does not allow a simple explanation.

Abstract

International audience; The kinetics of the oxidation of N-aminopiperidine with chloramine was studied at different temperatures, with variable concentrations of the two reactants and at a pH ranging between 12 and 13.5. The reaction showed to be involving two steps: the first corresponded to the formation of a diazene intermediate, the second to the evolution of this intermediate into numerous compounds within a complex reactional chain. The rate law of the first step was determined by the Ostwald method and found to be first order with respect to each reactant. The rate constant was determined at pH = 12.89 and T = 25°C: k2 = 1.15  105 exp(-39/RT) M-1 s-1 (E2 in kJ mol-1) With decreasing pH value, the first step exhibited acid catalysis phenomena, and diazene was converted into azopiperidine particularly faster. This created overlapping UV-absorptions between chloramine and azopiperidine, also observed in HPLC. GC/MS analyses were used to identify some of the numerous by-products formed. Their proportions are dependent of both pH and the reactants' concentrations ratio. A reaction mechanism taking this relationship into account, was suggested.

Abstract

The nonlinear electrical properties of CoO-doped SnO2 ceramics were characterized. A second phase is observed at the surface of the microstructure of the specimens. Energy-dispersive X-ray analysis reveals that the second phase is composed of tin, cobalt and oxygen with an approximate atomic ratio estimated for Co and Sn: 2:1. The phase was identified by X-ray diffraction and unambiguously establish the identity of the phase: Co2SnO4. Electrical characterization of the binary CoO–SnO2 was carried out on a very large range of current from 10−9 up to 100 A cm−2 showing that the doping of CoO increases drastically the resistivity of the ceramic from 104 to 1010 Ω cm. This value appears to be a threshold for which the nonlinear effect appears.

Abstract

International audience; The use of ab initio and DFT methods to calculate the enthalpies of formation of solid ionic compounds is described. The results obtained from the calculations are then compared with those from experimental measurements on nitrogen-rich salts of the 2,2-dimethyltriazanium cation (DMTZ) synthesized in our laboratory and on other nitrogen-rich ionic compounds. The importance of calculating accurate volumes and lattice enthalpies for the determination of heats of formation is also discussed. Furthermore, the crystal structure and hydrogen-bonding networks of the nitroformate salt of the DMTZ cation is described in detail. Lastly, the theoretical heats of formation were used to calculate the specific impulses (I-sp) of the salts of the DMTZ cation in view of a prospective application in propellant formulations.

Abstract

International audience; the kinetics of the Cl-transfert reaction between chloramine and piperidine in a Raschig medium was studied at various temperatures, with variable concentrations of the two reactants. The influence of the pH on the interaction was examined at a pH ranging between 8.25 and 12.89. The Cl-transfert reaction resulted in the formation of 1-chloropiperidine, which, in the presence of NaOH, underwent a dehydrohalogenation leading to an endocyclic imine derivative: 2,3,4,5-tetrahydropyridine. The kinetics of the dehydrohalogenation was also studied at different temperatures, with variable concentrations of 1-chloropiperidine and NaOH. Kinetic and thermodynamic parameters were determined.

Abstract

International audience; Isoplethic thermal analysis was used to determine the solid-liquid-liquid equilibria in the ternary system water-sodium sulfate-piperidine. The changes in state observed on the thermogram recorded during the displacement in a quasi-binary section permit the identification of the different phases and the delimitation of the corresponding equilibrium domains. Two isotherms were established at 25°C and 40°C because these temperatures frame the peritectic decomposition of the sodium sulfate decahydrate. Miscibility gaps were completely delimited and each critical point was calculated. This study permitted to determine the optimal conditions for separating the organic phase by liquid-liquid extraction.

Abstract

International audience; Lithium oxide in form of Li2CO3 solution is added with contents of 0–200 ppm to two ZnO-based varistors standard formulations, once with Sb2O3 and the other without. According to Li2CO3 concentration, both threshold field and energy absorption capability evolution are studied. It is found that with the benefit of antimony, the lithium allows reaching high threshold field but concomitantly, low energy absorption capability. Without antimony, threshold fields up to 300 V/mm are attained, associated with a fair energy absorption capability. With 100 ppm of Li2CO3, optimum couple of values (315 V/mm; 115 J/cm3) is achieved. With 200 ppm of Li2CO3, threshold field exceeds 500 V/mm but energy absorption capability falls below 50 J/cm3. Correlations with SEM microstructures observations suggest that lithium increases voltage barrier height by decreasing donor density and that spinel phases (Zn7Sb2O12) have detrimental effects on the electrical absorption capability by limiting the density of current, reducing the effective current path from one ZnO grain to another.

Abstract

The kinetics of complete oxidation of different samples of zinc powder by air has been investigated by thermogravimetric measurements under isothermal conditions in the range 973–1,173 K. Particles size was in the 63–80 μm range. We succeeded in carrying out the full oxidation of the powders far above the zinc-metal melting point (692.6 K). This phenomenon is linked to the presence of a thin ZnO layer which confines the liquid metal during the oxidation process. Two kinetics models have been verified. The apparent activation energies obtained from the Arrhenius law were 128 kJ mol−1 and 129 kJ mol−1, respectively.

Abstract

International audience; After an exhaustive study of the system ammonia-dimethylchloramine in liquid ammonia, it was interesting to compare the reactivity of this system in liquid ammonia with the same systeme in an aqueous medium. Dimethylchloramine prepared in a pure state, undergoes dehydrohalogenation in an alkaline medium: the principal products formed are N-methylmethanimine, 1,3,5-trimethylhexahydrotriazine, formaldehyde and methylamine. The kinetics of this reaction was studied by UV, GC and HPLC as a function of temperature, initial concentrations of sodium hydroxide and chlorinated derivative. The reaction is of the second order and obeys an E2 mechanism (k1 = 4.2 x 10-5 M-1 s-1, ΔH°# = 82 kJ mol-1, ΔS°# = - 59 J mol-1 K-1). The oxidation of unsymmetrical dimethylhydrazine by dimethylchloramine involves two consecutive processes. The first step follows a first-order law with respect to the haloamine and the hydrazine, leading to the formation of an aminonitrene intermediate (k2 = 150 x 10-5 M-1 s-1). The second step corresponds to the conversion of aminonitrene into formaldehyde dimethylhydrazone at pH=13). This reaction follows a first-order law (k3 = 23.5 x 10-5 s-1). The dimethylchloramine/ammonia interaction corresponds to a SN2 bimolecular mechanism (k4 = 0.9 x 10-5 M-1 s-1, pH = 13 and T = 25°C). The kinetic model formulated on the basis of the above reactions shows that the formation of the hydrazine in an aqueous medium comes under strong competition from the dehydrohalogenation of DMC and the oxidation of the hydrazine formed by the original chlorinated derivative. A global model which explains the mechanisms both in an anhydrous and in an aqueous medium was elaborated.

L'oxydation Directe d'un Alliage Précurseur permet de préparer des poudres céramiques homogènes par atomisation et trempe d'un alliage homogène fondu. En utilisant cette approche, de nouvelles varistances à base de dioxyde d'étain ont été préparées. Ces composants présentent des propriétés électriques similaires voire supérieures aux varistances à base de monoxyde de zinc et ceci malgré un dopage de la matrice 10 fois plus faible.

Abstract

HexaazahexaNitroIsoWurtzitane (or HNIW) hexanitred derivate, still called CL20, is a new highly energy molecule, which presents four polymorphic forms : α, β, γ and ε. Is the form ε most active and marketing stipulates to provide a product of which content of form ε that is to say higher than 95%, with estimate of the contents of the other forms as well as majority im-purity (pentanitred derivative). The aim of this work is on the one hand to characterize the four formes polymorphes, and on the other hand to develop, to optimize and validate a new quantitative analytical method to measure polymorphic composition of the CL20 by coupling IRTF (MIR) spectrometry/chemometric, respecting the technical and commercial require-ments, and transferable on factory site. The chemometric methods are used to conceive the experimentation and to process the experimental data necessary (calibration and validation).; L'HexaazahexaNitroIsoWurtzitane (ou HNIW), dérivé hexanitré, encore appelé CL20, est une nouvelle molécule hautement énergétique, qui présente quatre formes polymorphes : α, β, γ et ε. La forme ε est la plus active et la commercialisation stipule de fournir un produit dont la teneur en forme ε soit supérieure à 95%, avec estimation des teneurs des autres formes ainsi que de l'impureté majoritaire (dérivé pentanitré). L'objet du présent travail est d'une part de caractériser les différentes formes polymorphes, et d'autre part de mettre au point, d'optimiser et de valider une méthode d'analyse quantitative du CL20 polymorphe par couplage spectro-métrie IRTF/chimiométrie, respectant les impératifs techniques et commerciaux et transférable sur site industriel. Les méthodes chimiométriques sont utilisées pour concevoir l'expérimentation et traiter l'information expérimentale nécessaire (étalonnage et validation).
