10. Investigating the Potential of River Sediment Bacteria for Trichloroethylene Bioremediation.

Abstract

Trichloroethylene (TCE) is a prevalent groundwater contaminant detected worldwide, and microbes are sensitive indicators and initial responders to these chemical contaminants causing disturbances to their ecosystem. In this study, microbes isolated from San Marcos River sediment were screened for their TCE degradation potential. Among the twelve isolates (SAN1-12), five isolates demonstrated TCE degradation within 5 days at 25 °C and 40 mg/L of TCE concentration in the following order: SAN8 (87.56%), SAN1 (77.31%), SAN2 (76.58%), SAN3 (49.20%), and SAN7 (3.36%). On increasing the TCE concentration to 80 mg/L, the degradation efficiency of these isolates declined, although SAN8 remained the prominent TCE degrader with 75.67% degradation. The prominent TCE-degrading isolates were identified as Aeromonas sp. SAN1, Bacillus sp. SAN2, Gordonia sp. SAN3, and Bacillus proteolyticus SAN8 using 16S rRNA sequencing. The TCE degradation and cell biomass of Bacillus proteolyticus SAN8 were significantly improved when the incubation temperature was increased from 25 °C to 30 °C. However, both slightly acidic and alkaline pH levels, as well as higher TCE concentrations, lowered the efficacy of TCE degradation. Abstract

Biochar has proven effective in the remediation of excess nitrogen from soil and water. Excess nitrogen from agricultural fields ends up in aquatic systems and leads to reduced water quality and the proliferation of invasive species. This study aimed to assess the efficiency of chemically surface-modified biochar produced from invasive Pistia stratiotes L. for the adsorption of inorganic nitrogen (NH4+ and NO3−). Biochar structure was investigated using scanning electron microscopy, energy-dispersive X-ray analysis, X-ray photoelectron spectroscopy, Fourier-transform infrared spectroscopy, and inductively coupled plasma mass spectrometry. The results from adsorption experiments indicate that NH4+ removal was optimal (0.8–1.3 mg N g−1) at near-neutral pH levels (6.0–7.5), while NO3− removal was optimal (0.4–0.8 mg N g−1) under acidic pH conditions (4.8–6.5) using the modified biochar. These findings highlight the significance of solution pH, biochar morphology, and surface chemistry in influencing the adsorption of NH4+ and NO3−. However, further studies are necessary to assess the potential oxidative transformation of NH4+ to NO3− by biochar, which might have contributed to the reduction in NH4+ in the aqueous phase. 24. Exopolysaccharide production using pinewood hydrolysate as a substrate for psychrotrophic bacterium isolated from Svalbard glacier soil

Abstract

The Arctic region harbors elite extremophiles skilled in producing novel exopolysaccharides (EPS) to cope with extreme environmental conditions. In the present study, psychrotrophic microorganisms isolated from Svalbard glacier soil were investigated for EPS production. Pseudomonas frederiksbergensisBBL-P-9 proved higher EPS production under different substrate-feeding strategies. On feeding with pure glucose (30 g L−1) and xylose (10 g L−1), this bacterium produced 12.67 g L−1and 6.38 g L−1of EPS, respectively. Interestingly, on examining different lignocellulosic biomass hydrolysates as the substrates, P. frederiksbergensisBBL-P-9 produced 9.42 g L−1of EPS using pinewood hydrolysate (glucose content adapted to 30 g L−1). Furthermore, this bacterium evidenced tolerance to the plant biomass pre-treatment-derived side products in the following order: acetate>furfural>vanillin>5-hydroxymethylfurfural. The composition of EPS was altered with the type and concentration of the carbon substrate fed. However, β-D-glucopyranose (20~34%), β-D-mannopyranose (15~22%), and β-D-galactopyranose (17~27%) were the dominant monosaccharides detected in P. frederiksbergensisBBL-P-9 EPS.

41. Remediation of RDX-contaminated water using alkaline hydrolysis

Abstract

The objective of this study was to assess the effectiveness of alkaline hydrolysis as an alternative ex situ technology for remediating groundwater contaminated with hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX). Treatment in both batch reactor and continuous stirred tank reactor (CSTR) was investigated. RDX reactivity was strongly dependent on the reaction pH investigated (11-13). The batch system achieved pseudo-first-order RDX reaction rates in the range of (0.8-27.7) x [10.sup.-3] [min.sup.-1], corresponding to half-life periods of 17.9 to 0.5 h, respectively. In the CSTR system operated at the initial RDX concentration of 4.5 x [10.sup.-3] mM, 99% RDX removal was achieved with the hydraulic retention time of 2 days and the reaction pH of 11.9. Formate and nitrite were produced as the major hydrolysates in the CSTR system, indicating a simultaneous reaction mechanism involving RDX ring cleavage and elimination of the ring nitrogen. The net OH- demand used only for RDX removal in the CSTR was found to be 1.5, 390, and 130 M O[H.sup.-]/M RD[X.sub.removed] at pH values of 11.9, 11.5, and 11.0, respectively. A conceptual cost analysis indicated that the expense of alkaline treatment may be comparable to the expense of granular activated carbon treatment for long treatment periods (30 years or more), due to the potentially lower annual operational cost of alkali treatment.
