Population health research is increasingly focused on the genetic determinants of healthy ageing, but there is no public resource of whole genome sequences and phenotype data from healthy elderly individuals. Here we describe the first release of the Medical Genome Reference Bank (MGRB), comprising whole genome sequence and phenotype of 2570 elderly Australians depleted for cancer, cardiovascular disease, and dementia. We analyse the MGRB for single-nucleotide, indel and structural variation in the nuclear and mitochondrial genomes. MGRB individuals have fewer disease-associated common and rare germline variants, relative to both cancer cases and the gnomAD and UK Biobank cohorts, consistent with risk depletion. Age-related somatic changes are correlated with grip strength in men, suggesting blood-derived whole genomes may also provide a biologic measure of age-related functional deterioration. The MGRB provides a broadly applicable reference cohort for clinical genetics and genomic association studies, and for understanding the genetics of healthy ageing.

Combining extraction and scintillation properties within the same material is a relatively new approach in development of sensors for detection of radioactive elements. Structural organization of such materials at a nanoscale typically offers higher efficiency of detection and shorter response time. In this contribution, several new protocols are discussed for fabrication of stable extractive scintillating systems based on commercial Superlig 620 (SL) material with high affinity to radiostrontium. Application of hybrid organic–inorganic beads with SL particles used as core and halloysite clay nanotubes (HNT) modified with a polyvinyltoluene (PVT) brush as a permeable shell combines high-performance extracting properties of the SL material with efficient light emission properties of the polymer scintillator. The developed SL–HNT–PVT hybrid extractive scintillating material allows real-time detection of low-level concentrations of radiostrontium in water. Moreover, the suggested approach is not limited to detection of Sr but can find broader application in development of chemical, biological, or radioluminescent sensors and multifunctional materials. © 2018 WILEY-VCH Verlag GmbH & Co. In recognition of the fact that orang-utans (Pongo spp.) are severely threatened, a meeting of orang-utan experts and conservationists, representatives of national and regional governmental and non-governmental organizations, and other stakeholders, was convened in Jakarta, Indonesia, in January 2004. Prior to this meeting we surveyed all large areas for which orang-utan population status was unknown. Compilation of all survey data produced a comprehensive picture of orang-utan distribution on both Borneo and Sumatra. These results indicate that in 2004 there were c. 6,500 P. abelii remaining on Sumatra and at least 54,000 P. pygmaeus on Borneo. Extrapolating to 2008 on the basis of forest loss on both islands suggests the estimate for Borneo could be 10% too high but that for Sumatra is probably still relatively accurate because forest loss in orang-utan habitat has been low during the conflict in Aceh, where most P. abelii occur. When those population sizes are compared to known historical sizes it is clear that the Sumatran orang-utan is in rapid decline, and unless extraordinary efforts are made soon, it could become the first great ape species to go extinct. In contrast, our results indicate there are more and larger populations of Bornean orang-utans than previously known. Although these revised estimates for Borneo are encouraging, forest loss and associated loss of orang-utans are occurring at an alarming rate, and suggest that recent reductions of Bornean orang-utan populations have been far more severe than previously supposed. Nevertheless, although orang-utans on both islands are under threat, we highlight some reasons for cautious optimism for their long-term conservation

Evaluation of disease activity in systemic lupus erythematosus (SLE) nephritis is a challenge, and repeated renal biopsies are usually needed in order to confirm a suspicion of flare. In a previous cross-sectional study, we reported that serum soluble form of the interleukin-7 receptor (sIL7R) levels is strongly associated with nephritis in SLE patients. In the present study, we wanted to confirm the association between changes in serum sIL7R concentrations and renal disease activity in a large longitudinal cohort of SLE nephritis patients.

Objectives: Evaluation of disease activity in systemic lupus erythematosus (SLE) nephritis is a challenge, and repeated renal biopsies are usually needed in order to confirm a suspicion of flare. In a previous crosssectional study, we reported that serum soluble form of the interleukin-7 receptor (sIL7R) levels is strongly associated with nephritis in SLE patients. In the present study, we wanted to confirm the association between changes in serum sIL7R concentrations and renal disease activity in a large longitudinal cohort of SLE nephritis patients. Methods: Sera were harvested longitudinally in 105 SLE nephritis patients. Serum sIL7R cut-off value for the detection of SLE nephritis activity was determined as the mean sIL7R concentration in non-nephritis SLE patients + 2 SDs using data collected in our previous study. Patients with glomerular filtration rate (GFR) <60 mL/min/1.73 m2 (n=17) were excluded from the study due to persistently elevated serum sIL7R values. Results: Serum sIL7R concentrations above the renal cut-off value were observed in 25 (out of 88) patients with a normal GFR. These patients had significantly higher serum double-stranded DNA (dsDNA) Ab and urinary protein to creatinine (UPC) ratio. Strikingly, 12 of them developed a renal British Isles Lupus Assessment Group index (BILAG) A within the next 3 months, while this was only the case in four out of the 63 other patients (p<0.0001). The test had 75.0% sensitivity and 81.9% specificity for the detection of a renal BILAG A. Combination of serum sIL7R with any of the classical tests (anti-dsDNA Ab titres, UPC ratio, serum C3) resulted in an increased specificity for the detection of a renal flare. Administration of immunosuppressive therapy resulted in a significant decrease in serum sIL7R concentrations. Conclusions: Serum sIL7R is a sensitive and specific marker of renal disease activity in SLE. It has long been assumed that Sumatran forests are of higher quality for orangutans than Bornean forests, and that this is both the proximate and ultimate cause of many of the differences in socio-ecology between the two orangutan species. Yet this hypothesis has remained untested. This chapter presents data on the phenology and floristics of eight Bornean and three Sumatran forest sites where orangutans have been studied to examine the effects of floristic composition, habitat productivity, and seasonality on orangutan population density. The alternative hypotheses that higher orangutan densities in Sumatra are due to overall higher levels of plant productivity, the increased availability of preferred foods, the presence of more fallback foods, or differences in floristic composition between the two islands are tested empirically.

Orangutan populations are particularly susceptible to local extinction due to hunting, habitat loss, and fragmentation because they live at low population densities, grow slowly, and reproduce rarely. This chapter uses Population Viability Analysis (PVA) to consider the conservation implications of orangutan life history and population biology. First, a baseline model that incorporates the best available orangutan life-history data is presented. This model is then used to examine how plausible variation in model parameters, changes in the intensity of human-induced threats, and different conservation and management interventions would affect the probability of orangutan population persistence. The effects of existing threats on the extinction risk of specific orangutan populations on Borneo and Sumatra are also modelled. Finally, the conservation and management implications of this modeling exercise are considered.

The chapter examines differences in the activity budgets of wild orangutans (Pongo spp.) within and between a large number of study sites in Sumatra and Borneo. The authors of the chapter found that each orangutan population appeared to follow one of two distinct foraging strategies: either (1) ‘sit and wait’, in which orangutans aim to minimize their energy expenditure by spending long periods of time resting and relatively short periods feeding and travelling; or (2) ‘search and find’ in which orangutans aim to maximize their energy intake by resting little and mainly feeding or moving in search of food. Orangutans adopt the first strategy in mixed-dipterocarp forests characterized by mast-fruiting events and irregular fruit availability; and adopt the second strategy in swamp forests with a regular supply of fruit, or in dryland forests with high strangling-fig density. The chapter proposes that the determining factor for which strategy is adopted is the temporal availability of fruit in the habitat, as opposed to other possibilities such as orangutan taxonomy.
