21. Theranostics through Utilizing Cherenkov Radiation of Radioisotope Zr-89 with a Nanocomposite Combination of TiO2and MnO2

Abstract

Cherenkov radiation (CR) derived from the decay of diagnostic and therapeutic radionuclides is currently being studied by the scientific community to determine if these emissions can be harnessed for cancer detection and therapy. While Cherenkov luminescence imaging (CLI) has been studied in the preclinical and clinical settings, Cherenkov radiation-induced cancer therapy (CRICT) is a relatively new area of research that harnesses the emitted photons to kill cancer cells through free radical generation and DNA damage. Nanoparticles seem well suited for developing a theranostic platform that would allow researchers to visualize therapy delivery and also generate the reactive oxygen species necessary to kill cancer cells. Herein, we report the preparation of an 89Zr–TiO2–MnO2nanocomposite that incorporates transferrin onto the nanoparticle surface to enhance cancer cell growth inhibition. The incorporation of the positron emission tomography (PET) radioisotope 89Zr (half-life: 3.3 days) allowed for the detection of the nanoparticle using PET and for the creation of Cherenkov emissions that interacted with the nanoparticle surface to generate free radicals for therapy delivery. After preparation, these systems were observed to be stable in various media and provided excellent tumor growth control after being intratumorally injected into mice bearing CT-26 tumors. These results demonstrate that a therapeutically efficient CRICT platform can be generated using commercially available and affordable materials.

22. Radiation Monitoring System of 30 MeV Cyclotron

Abstract

A state-of-the-art radiation monitoring system was implemented at KAERI for a 30-MeV cyclotron. This system consists of several types of radiation measuring systems for ambient dose equivalent rate measurements of outside photon and neutron areas as well as inside the cyclotron, and monitors the alpha and beta particulates released from a stack, as well as the results of worker contamination at the portal of the cyclotron. In addition, an automatic alarm system is also mounted if there are alarms in the measuring systems.

Abstract

89Zr is an emerging radionuclide with promising application in nuclear medicine for the non-invasive diagnosis of various cancers with PET imaging. This paper would discuss optimization efforts in the production of 89Zr carried out at Accelerator Radioisotope Development Team, Korea Atomic Energy Research Institute. Optimized conditions reveal high production yields of 89Zr ~ 37 ± 1.4 MBq/µA using 89Y(p,n)89Zr, ~ 12.7 MeV of proton, and 35 µA using yttrium coin target with indigenously constructed 30 MeV Cyclotron (RFT-30). Besides, the production optimization, quality control, and biological behavior of [89Zr]Zr-oxalate, chloride and, DFO demonstrated promising results. [ABSTRACT FROM AUTHOR]

Abstract

Successful construction of the control system is an important problem in the accelerator. The RFT-30 cyclotron is 30 MeV cyclotron for radioisotope production and fundamental researches. To operate the RFT-30 cyclotron for beam irradiation, the human operators should carefully manipulate the control parameters. If the control does not function properly, it becomes difficult to handle the cyclotron and cannot perform the accurate operations for the control. In this work, we propose a deep learning based model predictive control approach for the RFT-30 cyclotron. The proposed approach is composed of two steps: system identification and a control design. In the system identification procedure, the proposed approach constructs the predictive model of the accelerator using the deep learning approach. In the control design stage, the controller finds the optimal control inputs by solving the optimization problem. Abstract

Beamline tuning is time consuming and difficult work in accelerator system. In this work, we propose a neural generalized predictive control (NGPC) approach for the RFT-30 cyclotron beamline. The proposed approach performs system identification with the NN model and finds the control parameters for the beamline. Abstract

RFT-30 30 MeV cyclotron has been developed for the production of radioisotopes and their applications. Fluorine-18, which is a widely-used positron emitter, has been produced regularly since 2015. In addition, research on the production of generator radioisotopes has been performed using this cyclotron. A generator means a device used to extract the positron-emitting daughter radioisotope from a source of the decaying parent radioisotope such as Ti-44 and Ge-68. In this research, gold-coated and natural Sc targets were proton-irradiated in order to produce Ti-44. Abstract

The solid target of the RFT-30 30 MeV cyclotron in KAERI was designed to produce the metalic radioisotopes, such as Zn-62, Cu-67, Ge-68, Pd-{103}, and In-111. The target control system should provide high reliability to prevent any kind of failure. Moreover, the operating procedures and maintenance cycle should be optimized and well organized to cover the unexpected situations. In this study, a simulation of the control system for the solid target in KAERI was carried out to confirm the operability of the solid target transport system. The receiving and irradiation stations are connected each other through square tube, and the control software was also checked.
