ABSTRACT: The legalization of industrial hemp by the 2018 Farm Bill in the United States has driven a sharp increase in its cultivation, including for cannabinoid extraction. Spent hemp biomass (SHB), produced from the extraction of cannabinoids, can potentially be used as feed for dairy cows; however, it is still illegal to do so in the United States, according to the US Food and Drug Administration Center for Veterinary Medicine, due to the presence of cannabinoids and the lack of data on the effect on animals. To assess the safety of this byproduct as feed for dairy cows, late-lactation Jersey cows (245 ± 37 d in milk; 483 ± 38 kg body weight; 10 multiparous and 8 primiparous) received a basal total mixed ration (TMR) diet plus 13% alfalfa pellet (CON) or 13% pelleted SHB for 4 wk (intervention period [IP]) followed by 4 wk of withdrawal period (WP), where all cows received only the basal TMR during WP. The dry matter intake (DMI), body weight, body condition score, milk yield, milk components, and fatty acid profile, blood parameters, N metabolism, methane emission, and activity were measured. Results indicated that feeding SHB decreased DMI mainly due to the low palatability of the SHB pellet, as the cows consumed only 7.4% of the total TMR with 13.0% SHB pellet offered in the ration. However, milk yield was not affected during the IP and was higher than CON during the WP, leading to higher milk yield/DMI. Milk components were not affected, except for a tendency in decreased fat percentage. Milk fat produced by cows fed SHB had a higher proportion of oleate and bacteria-derived fatty acids than CON. The activity of the cows was not affected, except for a shorter overall lying time in SHB versus CON cows during the IP. Blood parameters related to immune function were not affected. Compared with CON, cows fed SHB had a lower cholesterol concentration during the whole experiment and higher β-hydroxybutyric acid during the WP, while a likely low-grade inflammation during the IP was indicated by higher ceruloplasmin and reactive oxidative metabolites. Other parameters related to liver health and inflammatory response were unaffected, except for a tendency for higher activity of alkaline phosphatase during IP and a lower activity of gamma-glutamyl transferase during WP in the SHB group versus CON. The bilirubin concentration was increased in cows fed SHB, suggesting a possible decrease in the clearance ability of the liver. Digestibility of the dry matter and protein and methane emission were not affected by feeding SHB. The urea, purine derivatives, and creatinine concentration in urine was unaffected, but cows fed SHB had higher N use efficiency and lower urine volume. Altogether, our data revealed a relatively low palatability of SHB affecting DMI with minimal biological effects, except for a likely low-grade inflammation, a higher N use efficiency, and a possible decrease in liver clearance. Overall, the data support the use of SHB as a safe feed ingredient for lactating dairy cows.

The United States Department of Energy, the MH21-S Research Consortium of Japan, and the United States Geological Survey are collaborating to enable gas hydrate scientific drilling and extended-duration reservoir response testing on the Alaska North Slope. To feasibly execute such a test, a location is required that is accessible from existing roads and gravel pads and that can be occupied without disrupting ongoing industry operations. A review of potential locations meeting these criteria determined the likely occurrence of gas hydrate in two fine-grained marginal-marine sands of Tertiary age in the vicinity of the inactive “Kuparuk State 7-11-12” exploration pad in the western Prudhoe Bay Unit (PBU). Existing well and seismic data for that site were insufficient to preclude the potential for free gas occurrence within the deeper (and most prospective) target sand. Therefore, with support from the PBU Working Interest Owners, Alaska Department of Natural Resources, and Petrotechnical Resources Alaska, the Hydrate-01 Stratigraphic Test Well (STW) was drilled in December 2018 to confirm the suitability of the site for future gas hydrate scientific testing. The Hydrate-01 well was successfully drilled to −3290 ft (1003 m) subsea vertical depth at a bottom hole location of approximately 900 ft (∼275 m) east of the surface location. The drilling program featured acquisition of a full suite of logging while drilling data, the collection of side-wall pressure cores, and the installation of distributed temperature and distributed acoustic sensor fiber-optic cables. The log data acquired confirmed the occurrence of gas hydrate at high saturation in two target sands. Integrated evaluation of log and sidewall core data provide petrophysical and geomechanical property information that allow for potential reservoir response to depressurization to be simulated. The deeper “B1 sand” is deemed to be most favorable for reservoir response testing as a result of confirmed gas hydrate occurrence in sediments of high intrinsic permeability, location within 100 ft (30 m) of the base of gas hydrate stability, and minimal risk for direct communication with permeable water-bearing (hydrate-free) zones. The shallower “D1 sand” provides a secondary target that is differentiated by colder in situ temperatures and the interpreted direct hydraulic communication to a lower section of non-hydrate-bearing, water-saturated sand. The Hydrate-01 log data also confirm the occurrence of at least one sub-seismic fault in close proximity to the B1 sand reservoir. To better image the distribution of the gas-hydrate-bearing reservoir sections and associated faults, a three-dimensional (3D) vertical seismic profile was conducted in early 2019 using the distributed acoustic sensors installed as part of the Hydrate-01 STW completion. Detailed two-dimensional (2D) and 3D geologic models have been constructed to enable numerical simulations to inform the planning for potential future scientific tests of reservoir response to depressurization at the site.

Short-term probabilistic forecasts of the trajectory of the COVID-19 pandemic in the United States have served as a visible and important communication channel between the scientific modeling community and both the general public and decision-makers. Forecasting models provide specific, quantitative, and evaluable predictions that inform short-term decisions such as healthcare staffing needs, school closures, and allocation of medical supplies. Starting in April 2020, the US COVID-19 Forecast Hub (https://covid19forecasthub.org/) collected, disseminated, and synthesized tens of millions of specific predictions from more than 90 different academic, industry, and independent research groups. A multimodel ensemble forecast that combined predictions from dozens of groups every week provided the most consistently accurate probabilistic forecasts of incident deaths due to COVID-19 at the state and national level from April 2020 through October 2021. The performance of 27 individual models that submitted complete forecasts of COVID-19 deaths consistently throughout this year showed high variability in forecast skill across time, geospatial units, and forecast horizons. Two-thirds of the models evaluated showed better accuracy than a naïve baseline model. Forecast accuracy degraded as models made predictions further into the future, with probabilistic error at a 20-wk horizon three to five times larger than when predicting at a 1-wk horizon. This project underscores the role that collaboration and active coordination between governmental public-health agencies, academic modeling teams, and industry partners can play in developing modern modeling capabilities to support local, state, and federal response to outbreaks.

A considerable amount of research has been performed by various groups with the goal of detecting objects of interest in various environments. These objects can be above ground, partially buried, or fully buried. One of the detection methods that has piqued the interest of researchers is Automated Target Recognition (ATR) algorithms that can pick out the locations of such objects from infrared (IR) sensor imagery once it was recognized that such objects absorb and emit heat at rates that distinguish them from their surrounding environments. In order to obtain training data for ATRs, one option is to gather large volumes of field data from many locations, but such efforts are resource and labor intensive. Another option is to generate synthetic training data that simulates heat flux in real environments. Several research groups in the past have implemented a combination of both, using field data to inform and verify simulation tools that model heat flux, energy absorption, and IR emission from various parts of the environment, as well developing simulation workflows that model real environments. Such workflows are often developed for the purpose of simulating a single environment. Our goal is to modify and generalize such a workflow so that simulation parameters and the location of object of interest can be changed by non-experts or automated processes. In addition; this study is also focused on developing an efficient methodology to simulate a large variety of scenarios to produce varied sets of synthetic training data for the ATRs. The HPC simulations involved in generating the training data for ATRs require the exchange of data between several high-fidelity physics models. Efficient techniques have been implemented to ensure that the inputs to each component are consistent and correct. A more generalized workflow has been developed that is able to simulate complex scenarios with new LiDAR data, weather data, vegetation placements, target placements, and as output produce synthetic IR imagery useful for training ATRs. The synthetic IR images are automatically tagged with bounding boxes so that minimum post processing is required when the images are used for training the ATRs. The synthetic IR imagery was passed on to researchers developing ATRs and used to train the algorithms. The parts of the workflow that are streamlined can now be executed with relative ease on a high-performance computing environment and components of the workflow can now be more easily updated and modified in the future. During May-October 2018, four patients from three states experienced sepsis after transfusion of apheresis platelets contaminated with Acinetobacter calcoaceticus-baumannii complex (ACBC) and Staphylococcus saprophyticus; one patient died. ACBC isolates from patients' blood, transfused platelet residuals, and two environmental samples were closely related by whole genome sequencing. S. saprophyticus isolates from two patients' blood, three transfused platelet residuals, and one hospital environmental sample formed two whole genome sequencing clusters. This whole genome sequencing analysis indicated a potential common source of bacterial contamination; investigation into the contamination source continues. All platelet donations were collected using apheresis cell separator machines and collection sets from the same manufacturer; two of three collection sets were from the same lot. One implicated platelet unit had been treated with pathogen-inactivation technology, and two had tested negative with a rapid bacterial detection device after negative primary culture. Because platelets are usually stored at room temperature, bacteria in contaminated platelet units can proliferate to clinically relevant levels by the time of transfusion. Clinicians should monitor for sepsis after platelet transfusions even after implementation of bacterial contamination mitigation strategies. Recognizing adverse transfusion reactions and reporting to the platelet supplier and hemovigilance systems is crucial for public health practitioners to detect and prevent sepsis associated with contaminated platelets.

There is a lack of standardized measurement tools globally to assess knowledge, attitudes, and perceptions of expecting women toward prenatal screening. The purpose of this systematic review was to identify reasons women pursue or decline prenatal screening and compare the strengths and limitations of available measurement tools used to assess pregnant women's perceptions, knowledge, and attitudes toward prenatal screening. This review followed the five-step York methodology by Arksey and O'Malley and incorporated recommendations from the Preferred Reporting Items for Systematic Reviews and Meta-Analysis checklist for the extraction, analysis, and presentation of results. The five steps consisted of: (1) identification of the research questions; (2) searching for relevant studies; (3) selection of studies relevant to the research questions; (4) data charting; and (5) collation, summarization, and reporting of results. Four online databases (PubMed, Embase, Web of Science, and Cochrane Library) were selected after the librarian's development of a detailed search strategy. The Rayyan platform was used between June 2023 and August 2023 to epitomize the articles produced from our search. A total of 68 eligible studies were included in the analysis. The top five major reasons for declining prenatal screening uptake included (1) being unsure of the risk of prenatal screening and harm to the baby or miscarriage (n = 15), (2) not considering action such as termination of pregnancy for prenatal screening to be considered as necessary (n = 14), (3) high cost (n = 12), (4) lack of knowledge about testing procedures and being anxious about the test (n = 10), and (5) being worried about probability of false negative or false positive results (n = 6). Only 32 studies utilized scientifically validated instruments. Difficulties in capturing representative, adequately sized samples inclusive of diverse ethnicities and demographics were pervasive. Findings highlight the need for rigorous validation of research measurement methodologies to ensure the accuracy and applicability of resulting data regarding the assessment of prenatal screening perceptions, knowledge, and attitudes across diverse female populations. In the United States, a generic Type A medicated article product can gain the FDA approval by demonstrating bioequivalence (BE) to the pioneer product by successfully conducting a blood level, pharmacodynamic, or clinical BE study. A biowaiver can be granted based on several criteria, assuming the dissolution of the test and reference products represents the only factor influencing the relative bioavailability of both products. Monensin is practically insoluble in H2O per the USP definition. Previously published data from a comparison study of monensin dissolution profiles from the pioneer product and four generic products using biorelevant media showed that generic monensin products demonstrated different dissolution profiles to the pioneer product in these USP biorelevant rumen media. This follow‐up study compared the solubility profiles in simulated intestinal fluid (cFaSSIF, pH 7.5) for the pioneer product and four generic products. The generic monensin products demonstrated different in vitro dissolution profiles to the pioneer product in biorelevant media. The differences demonstrated in solubility and dissolution profiles are of concern regarding the potential efficacy of generic monensin in cattle. There are also additional concerns for the potential development of Eimeria resistance in cattle receiving a sub‐therapeutic dose of monensin from a less soluble generic product.
