Abstract

The Extracellular RNA Communication Consortium (ERCC) was launched to accelerate progress in the new field of extracellular RNA (exRNA) biology and to establish whether exRNAs and their carriers, including extracellular vesicles (EVs), can mediate intercellular communication and be utilized for clinical applications. Phase 1 of the ERCC focused on exRNA/EV biogenesis and function, discovery of exRNA biomarkers, development of exRNA/EV-based therapeutics, and construction of a robust set of reference exRNA profiles for a variety of biofluids. Here, we present progress by ERCC investigators in these areas, and we discuss collaborative projects directed at development of robust methods for EV/exRNA isolation and analysis and tools for sharing and computational analysis of exRNA profiling data.

Abstract

Until recently, estimation of β-amyloid plaque density as a key element for identifying Alzheimer's disease (AD) pathology as the cause of cognitive impairment was only possible at autopsy. Now with amyloid-positron emission tomography (amyloid-PET) neuroimaging, this AD hallmark can be detected antemortem. Practitioners and patients need to better understand potential diagnostic benefits and limitations of amyloid-PET and the complex practical, ethical, and social implications surrounding this new technology. To complement the practical considerations, Eli Lilly and Company sponsored a Bioethics Advisory Board to discuss ethical issues that might arise from clinical use of amyloid-PET neuroimaging with patients being evaluated for causes of cognitive decline. To best address the multifaceted issues associated with amyloid-PET neuroimaging, we recommend this technology be used only by experienced imaging and treating physicians in appropriately selected patients and only in the context of a comprehensive clinical evaluation with adequate explanations before and after the scan.

Abstract

A joint effort between the NASA Ames and Langley Research Centers was undertaken to analyze the Mach 0.745 variant of the Boeing Transonic Truss-Braced Wing (TTBW) Design. Two different flow solvers, LAVA and USM3D, were used to predict the TTBW flight performance. Sensitivity studies related to mesh resolution and numerical schemes were conducted to define best practices for this type of geometry and flow regime. Validation efforts compared the numerical simulation results of various modeling methods against experimental data taken from the NASA Ames 11-foot Unitary Wind Tunnel experimental data. The fidelity of the computational representation of the wind tunnel experiment, such as utilizing a porous wall boundary condition to model the ventilated test section, was varied to examine how different tunnel effects influence CFD predictions. LAVA and USM3D results both show an approximate 0.5o angle of attack shift from experimental lift curve data. This drove an investigation that revealed that the trailing edge of the experimental model was rounded in comparison to the CAD model, due to manufacturing tolerances, which had not been accounted for in the initial simulations of the experiment. Simulating the TTBW with an approximation of this rounded trailing-edge reduces error by approximately 60%. An accurate representation of the tested TTBW geometry, ideally including any wing twists and deflections experienced during the test under various loading conditions, will be necessary for more thorough validation of the CFD.

Abstract

Two Navier-Stokes codes were used to compute flow over the High-Lift Common Research Model (HL-CRM) in preparation for a wind tunnel test to be performed at the NASA Langley Research Center 14-by-22-Foot Subsonic Tunnel in fiscal year 2018. Both flight and wind tunnel conditions were simulated by the two codes at set Mach numbers and Reynolds numbers over a full angle-of-attack range for three configurations: cruise, landing and takeoff. Force curves, drag polars and surface pressure contour comparisons are shown for the two codes. The lift and drag curves compare well for the cruise configuration up to 10deg angle of attack but not as well for the other two configurations. The drag polars compare reasonably well for all three configurations. The surface pressure contours compare well for some of the conditions modeled but not as well for others.

Hunter (a) Keywords: C. elegans; lis-1; mdf-1; Programmed cell death; Spindle checkpoint Abstract: Deletion of the lissencephaly disease gene LIS-1 in humans causes an extreme disorganization of the brain associated with significant reduction in cortical neurons. Here we show that deletion or RNA interference (RNAi) of Caenorhabditis elegans lis-1 results in a reduction in germline nuclei and causes a variety of cellular, developmental, and neurological defects throughout development. Our analysis of the germline defects suggests that the reduction in nuclei number stems from dysfunctional mitotic spindles resulting in cell cycle arrest and eventually programmed cell death (apoptosis). Deletion of the spindle checkpoint gene mdf-1 blocks lis-1(lf)-induced cell cycle arrest and germline apoptosis, placing the spindle checkpoint pathway upstream of the programmed cell death pathway. These results suggest that apoptosis may contribute to the cell-sparse pathology of lissencephaly. Abstract

A new porous condition has been implemented in the PAB3D solver to simulate flow over porous surfaces. The newly added boundary condition is utilized to compute the flow field of a nonaxisymmetric, convergent-divergent nozzle incorporating porous cavities for shock-boundary layer interaction control. The nozzle has an expansion ratio (exit area/throat area) of 1.797 and a design nozzle pressure ratio (NPR) of 8.78. The flow fields for a baseline nozzle (no porosity) and for a nozzle with porous surfaces (10% porosity ratio) are computed for NPR varying from 2.01 to 9.54. Computational model results indicate that the overexpanded nozzle flow is dominated by shock-induced boundary-layer separation. Porous configurations are capable of controlling off-design separation in the nozzle by encouraging stable separation of the exhaust flow. Computational simulation results, wall centerline pressure, Mach contours, and thrust efficiency ratio are presented and discussed. Computed results are in excellent agreement with experimental data.

In C. elegans, loss-of-function mutations in ref-1, a distant nematode-specific member of this subfamily, were previously shown to cause ectopic neurogenesis from postembryonic lineages. However, while the vast majority of the nervous system in C. elegans is generated embryonically, the role of REF-1 in regulating these neural lineage decisions is unknown. Here, we show that mutations in ref-1 result in the generation of multiple ectopic neuron types derived from an embryonic neuroblast. In wild-type animals, neurons derived from this sublineage are present in a left/right symmetrical manner. However, in ref-1 mutants, while the ectopically generated neurons exhibit gene expression profiles characteristic of neurons on the left, they are present only on the right side. REF-1 functions in a Notch-independent manner to regulate this ectopic lineage decision. We also demonstrate that loss of REF-1 function results in defective differentiation of an embryonically generated serotonergic neuron type. These results indicate that REF-1 functions in both Notch-dependent and independent pathways to regulate multiple developmental decisions in different neuronal sublineages.
