High-frequency beam oscillation and synchronous wire-powder feeding were adopted for the laser-arc hybrid welding of a 2219-T6 high-strength aluminum alloy. Increasing the beam oscillation frequency from 0 to 250 Hz not only reduced the weld porosity from 2.64% to 0.16% but also promoted the transformation of columnar dendrites to fine equiaxed grains. In addition, the content of precipitated Al2CuMg increased with the refined size of the weld microstructure. The length of the columnar grains and the diameter of the equiaxed grains in the weld center were reduced by 40% and 50%, respectively. Owing to these improvements, the weld microhardness increased and became more uniform, and the variance decreased by 74%. The ultimate tensile strength and elongation of the welded joint reached 238.5 MPa and 4.3%, respectively, which were 26.5% and 105% higher than those of the non-oscillating hybrid weld, respectively. The weld tensile strength and the proportion of the area of the pore were negatively correlated. The porosity and microcracks of the welded joints were the main factors affecting the tensile strength of welded joints.

FeCoNiCrCu high-entropy alloy (HEA) and Cu foils were utilized as the intermediate layer to conduct laser welding of TC4 titanium alloy and Q345 steel. Welding is performed by adding single HEA and Cu/HEA double foils as interlayer respectively. We conducted in-depth studies on the microstructure and mechanical properties of the joint by stereomicroscopy, metallographic microscope, scanning electron microscope (SEM), micro-area X-ray diffraction (XRD), nanoindentation, electron backscatter diffraction (EBSD), and tensile testing. The results indicate that the position of the copper foil significantly affects the microstructure and performance of the joint. When the copper foil is on the TC4 side, its lower melting point causes a deeper keyhole, resulting in a narrower weld bead and then reduced content of Fe and Ti in the weld. Simultaneously, the increased proportion of Cu in the weld significantly enhances the content of Cu-rich phases. In the weld zone, we observed freely distributed Cu-rich phases and Ti-rich phases generated along the interface. Under tensile loads, cracks primarily initiate and propagate along the Cu-rich phases, leading to typical delamination on the fracture surface. With the copper foil on the TC4 side, due to the increased copper content in the microstructure, the hardness of the interface between the titanium alloy and the weld decreases, while the joint exhibits the highest tensile strength, reaching a maximum of 417 MPa.

Autoclave corrosion testing has been used for many years; however, few have studied the effect of its shortcomings on the formation of corrosion products and corrosion behaviour. In particular, understanding of these effects under high temperature and high pressure (HTHP) is rare. In this study, we highlighted the important parameters such as heating periods, flow rates and volume-to-surface ratios influencing carbon steel corrosion in the early stages. Especially, characterisation and protection behaviour of corrosion products associated with corrosion behaviour in the heating stage under the geothermal CO2-containing environment were systematically studied via mass loss, in-situ electrochemistry and surface analysis techniques. The roles of pCO2, volume-to-surface ratio and flow rate influence the formation of the corrosion products in the early stage are also evaluated.

Abstract Background A 3.0-mm ultrathin bronchoscope (UTB) with a 1.7-mm working channel provides better accessibility to peripheral bronchi. A 4.0-mm thin bronchoscope with a larger 2.0-mm working channel facilitates the use of a guide sheath (GS), ensuring repeated sampling from the same location. The 1.1-mm ultrathin cryoprobe has a smaller diameter, overcoming the limitation of the size of biopsy instruments used with UTB. In this study, we compared the endobronchial ultrasound localization rate and diagnostic yield of peripheral lung lesions by cryobiopsy using UTB and thin bronchoscopy combined with GS. Methods We retrospectively evaluated 133 patients with peripheral pulmonary lesions with a diameter less than 30 mm who underwent bronchoscopy with either thin bronchoscope or UTB from May 2019 to May 2023. A 3.0-mm UTB combined with rEBUS was used in the UTB group, whereas a 4.0-mm thin bronchoscope combined with rEBUS and GS was used for the thin bronchoscope group. A 1.1-mm ultrathin cryoprobe was used for cryobiopsy in the two groups. Results Among the 133 patients, peripheral pulmonary nodules in 85 subjects were visualized using r-EBUS. Abstract Background The plasma concentrations of acyl coenzyme A binding protein (ACBP, also known as diazepam-binding inhibitor, DBI, or ‘endozepine’) increase with age and obesity, two parameters that are also amongst the most important risk factors for cancer. Methods We measured ACBP/DBI in the plasma from cancer-free individuals, high-risk patients like the carriers of TP53 or BRCA1/2 mutations, and non-syndromic healthy subjects who later developed cancer. In mice, the neutralization of ACBP/DBI was used in models of non-small cell lung cancer (NSCLC) and breast cancer development and as a combination treatment with chemoimmunotherapy (chemotherapy + PD-1 blockade) in the context of NSCLC and sarcomas. The anticancer T cell response upon ACBP/DBI neutralization was characterized by flow cytometry and single-cell RNA sequencing. Results Circulating levels of ACBP/DBI were higher in patients with genetic cancer predisposition (BRCA1/2 or TP53 germline mutations) than in matched controls. In non-syndromic cases, high ACBP/DBI levels were predictive of future cancer development, and especially elevated in patients who later developed lung cancer. In preclinical models, ACBP/DBI neutralization slowed down breast cancer and NSCLC development and enhanced the efficacy of chemoimmunotherapy in NSCLC and sarcoma models. When combined with chemoimmunotherapy, the neutralizing monoclonal antibody against ACBP/DBI reduced the frequency of regulatory T cells in the tumor bed, modulated the immune checkpoint profile, and increased activation markers. Conclusion These findings suggest that ACBP/DBI acts as an endogenous immune suppressor. We conclude that elevation of ACBP/DBI constitutes a risk factor for the development of cancer and that ACBP/DBI is an actionable target for improving cancer immunosurveillance.

Abstract The influence of greenhouse warming on mesoscale air-sea interactions, crucial for modulating ocean circulation and climate variability, remains largely unexplored due to the limited resolution of current climate models. Additionally, there is a lack of theoretical frameworks for assessing changes in mesoscale coupling due to warming. Here, we address these gaps by analyzing eddy-resolving high-resolution climate simulations and observations, focusing on the mesoscale thermal interaction dominated by mesoscale sea surface temperature (SST) and latent heat flux (LHF) coupling in winter. Our findings reveal a consistent increase in mesoscale SST-LHF coupling in the major western boundary current regions under warming, characterized by a heightened nonlinearity between warm and cold eddies and a more pronounced enhancement in the northern hemisphere. To understand the dynamics, we develop a theoretical framework that links mesoscale thermal coupling changes to large-scale factors, which indicates that the projected changes are collectively determined by historical background wind, SST, and the rate of SST warming. Among these factors, the large-scale SST and its warming rate are the primary drivers of hemispheric asymmetry in mesoscale coupling intensification. This study introduces a simplified approach for assessing the projected mesoscale thermal coupling changes in a warming world.

A one-step QuEChERS automated detection technology for 22 pesticides in cowpeas and mangoes was established based on ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). The sample was extracted by acetonitrile containing 1% acetic acid, added with EN extraction salt (anhydrous Mg2SO4, sodium chloride, trisodium citrate dihydrate, disodium hydrogen citrate sesquihydrate) to remove water, and then purified with C18 and graphitized carbon black (GCB). The target analytes were separated by ACQUITY UPLC BEH C18 chromatography column, using 0.01% formic acid aqueous solution (containing 2 mmol/L ammonium formate)-0.01% formic acid methanol as the mobile phase, and determined in multiple reaction monitoring (MRM). The results showed that the 22 pesticides had good linearity in the range of 0.01~25.00 µg/L, with coefficient of determination (R2) higher than 0.994. The limits of detection (LOD, S/N=3) were 0.05~2 µg/kg, and the limits of quantification (LOQ, S/N=10) were 0.1~5 µg/kg. At the spiked levels of 1×LOQ, 2×LOQ, and 10×LOQ, the average recoveries for cowpeas were 70.9%~116.6%, 71.9%~118.3%, and 73.7%~113.7%, respectively, and for mangoes were 70.2%~114.6%, 71.2%~113.7%, and 74.3%~118.5%, respectively. The relative standard deviations for them were all within 20%. The method was successfully applied to the detection of 42 actual samples, and 33 samples detected pesticide residues. This method has high sensitivity and a satisfactory level of automation, effectively reducing human operation steps. It is suitable for the simultaneous detection and accurate quantification of 22 pesticides and their metabolites.

In this study, the keyhole dynamic behavior and molten pool flow were investigated to elucidate the effects of oscillating laser on porosity in narrow-gap oscillating laser-MIG hybrid welding of aluminum alloys. A weld with a porosity of 0.08% was produced at the laser oscillating frequency of 600 Hz and diameter of 1 mm, while 2.27% without oscillating laser. Compared to non-oscillating laser, keyhole opening size increased by about 2.3 times and keyhole depth fluctuation was suppressed. The forces acted on the interface between keyhole wall and molten pool became more balanced. The increased vapor recoil pressure overcame the combined closure effect of gravity and molten pool pressure. While applying oscillating laser, the vortex in molten pool was eliminated and the liquid metal flow was more stable. The combined impact force caused by droplets transfer and liquid metal convection due to constraining effect of narrow gap was buffered, which had less effect on molten pool and keyhole stability. The liquid metal tended to flow from the molten pool inside to surface, which favored the escape of bubbles from the molten pool.

A method of ultra-high performance liquid chromatography quadrupole electrostatic field Orbitrap mass spectrometry (UHPLC-Q/Orbitrap MS) was developed for rapid screening illegal additives in diet food. The efficacy of three extraction solvents for methanol, acetonitrile and acetonitrile containing 1% formic acid were evaluated for the extraction of illegal additives from green plums and jellies. Ultimately, acetonitrile is selected to extract illegally additives under ultrasonication. In order to achieve good separation of the additives by UHPLC in diet foods, four mobile phases were compared, including methanol (0.1% formic acid)-water (0.1% formic acid and 5 mmol/L ammonium acetate), acetonitrile (0.1% formic acid)-water (0.1% formic acid and 5 mmol/L ammonium acetate), acetonitrile-water (5 mmol/L ammonium acetate), acetonitrile-water. The effects of mobile phases and acidity levels on the chromatographic behavior of the additives were investigated, and acetonitrile-water containing 5 mmol/L ammonium acetate shows the best performance under appropriate gradient. Then, the MS data were acquired in Full MS/dd-MS2 mode on UHPLC-Q/Orbitrap MS with the matrix-matched external standard method for quantification. Based on the self-constructed database of illegal additives in diet foods, one unknown additive is detected in a diet food containing green plums and jelly. By characterizing the molecular ion peak, secondary fragment ion information, and isotopic information, the unknown compound is identified to be 4-chlorobisacetophenetidin, also an illegal additive, which was verified by computation of density functional theory and analysis of the standard substance. To validate the accuracy and reliability of the method, the limits of detection (LODs), the limits of quantification (LOQs), linear ranges, recoveries, relative standard deviations (RSDs) and matrix effect (ME) for eight compounds were determined. The LODs and LOQs of deacetylbisacodyl and bisacodyl are 0.001 and 0.005 mg/kg, respectively. The LODs and LOQs of other compounds are 0.05 and 0.1 mg/kg, respectively. The correlation coefficient (R2) is higher than 0.999. All of the compounds exhibit varying degrees of MEs from moderate to weak, and the recoveries is in the range of 71.02%-103.01% with the RSDs of 0.87%-13.24%. The approach was applied to diet food containing green plums and jelly. The 4-chlorobisacodyl are detected in 30% of the green plums, suggesting that this method is suitable for the rapid qualitative and quantitative analysis of illegal additives in diet food. It is also demonstrated that the established screening process plays a positive role in identifying potential risk substances. The combination of quantum chemistry computation and mass spectrometry provides a novel strategy for the identification of unknown additives in food.

Abstract Multiple classes of therapies targeting claudin-18 isoform 2 (CLDN18.2) are under development for the treatment of advanced gastroesophageal adenocarcinoma and other solid tumors. At the 2024 American Society of Clinical Oncology (ASCO) Annual Meeting, the final results of the phase 3 SPOTLIGHT trial were presented, demonstrating a significant survival benefit from the addition of the CLDN18.2-specific antibody zolbetuximab to chemotherapy in the first-line treatment of advanced gastroesophageal adenocarcinomas with ≥ 75% CLDN18.2 expression. Early-phase trial results presented at ASCO 2024 showed promising efficacy and safety of the afucosylated CLDN18.2-specific antibody FG-M108 in combination with chemotherapy in the first-line treatment of CLDN18.2-positive advanced gastroesophageal and pancreatic cancers. In addition, several early-phase trials presented at ASCO 2024 investigate other CLDN18.2-targeting approaches in CLDN18.2-positive refractory advanced solid tumors, including the CLDN18.2-targeting antibody–drug conjugates LM-302 and IBI343, the bispecific anti-CLDN18.2/CD3 antibody IBI38, and the chimeric antigen receptor T cell therapy satricabtagene autoleucel. These novel approaches could potentially expand the benefit of CLDN18.2-targeting therapies to a broader range of tumor types and to tumors expressing lower levels of CLDN18.2.

Abstract Ovarian granulosa cells are essential to gonadotrophin-regulated estrogen production, female cycle maintenance and fertility. The epithelial Na+ channel (ENaC) is associated with female fertility; however, whether and how it plays a role in ovarian cell function(s) remained unexplored. Here, we report patch-clamp and Na+ imaging detection of ENaC expression and channel activity in both human and mouse ovarian granulosa cells, which are promoted by pituitary gonadotrophins, follicle stimulating hormone (FSH) or luteinizing hormone (LH). Cre-recombinase- and CRISPR-Cas9-based granulosa-specific knockout of ENaC α subunit (Scnn1a) in mice resulted in failed estrogen elevation at early estrus, reduced number of corpus luteum, abnormally extended estrus phase, reduced litter size and subfertility in adult female mice. Further analysis using technologies including RNA sequencing and Ca2+ imaging revealed that pharmacological inhibition, shRNA-based knockdown or the knockout of ENaC diminished spontaneous or stimulated Ca2+ oscillations, lowered the capacity of intracellular Ca2+ stores and impaired FSH/LH-stimulated transcriptome changes for estrogen production in mouse and/or human granulosa cells. Together, these results have revealed a previously undefined role of ENaC in modulating gonadotrophin signaling in granulosa cells for estrogen homeostasis and thus female fertility.

Abstract Background The Synotis (C. B. Clarke) C. Jeffrey & Y. L. Chen is an ecologically important genus of the tribe Senecioneae, family Asteraceae. Because most species of the genus bear similar morphology, traditional morphological identification methods are very difficult to discriminate them. Therefore, it is essential to develop a reliable and effective identification method for Synotis species. In this study, the complete chloroplast (cp.) genomes of four Synotis species, S. cavaleriei (H.Lév.) C. Jeffrey & Y.L. Chen, S. duclouxii (Dunn) C. Jeffrey & Y.L. Chen, S. nagensium (C.B. Clarke) C. Jeffrey & Y.L. Chen and S. erythropappa (Bureau & Franch.) C. Jeffrey & Y. L. Chen had been sequenced using next-generation sequencing technology and reported here. Results These four cp. genomes exhibited a typical quadripartite structure and contained the large single-copy regions (LSC, 83,288 to 83,399 bp), the small single-copy regions (SSC, 18,262 to 18,287 bp), and the inverted repeat regions (IR, 24,837 to 24,842 bp). Each of the four cp. genomes encoded 134 genes, including 87 protein-coding genes, 37 tRNA genes, 8 rRNA genes, and 2 pseudogenes (ycf1 and rps19). The highly variable regions (trnC-GCA-petN, ccsA-psaC, trnE-UUC-rpoB, ycf1, ccsA and petN) may be used as potential molecular barcodes. The complete cp. genomes sequence of Synotis could be used as the potentially effective super-barcode to accurately identify Synotis species. Phylogenetic analysis demonstrated that the four Synotis species were clustered into a monophyletic group, and they were closed to the Senecio, Crassocephalum and Dendrosenecio in tribe Senecioneae. Conclusions This study will be useful for further species identification, evolution, genetic diversity and phylogenetic studies within this genus Synotis and the tribe Senecioneae.

Abstract Background Pseudomonas juntendi is a newly identified opportunistic pathogen, of which we have limited understanding. P. juntendi strains are often multidrug resistant, which complicates clinical management of infection. Methods A strain of Pseudomonas juntendi (strain L4326) isolated from feces was characterized by MALDI-TOF-MS and Average Nucleotide Identity BLAST. This strain was further subject to whole-genome sequencing and Maximum Likelihood phylogenetic analysis. The strain was phenotypically characterized by antimicrobial susceptibility testing and conjugation assays. Results We have isolated the novel P. juntendi strain L4236, which was multidrug resistant, but retained sensitivity to amikacin. L4236 harbored a megaplasmid that encoded bla OXA−1 and a novel bla IMP−1 resistance gene variant. P. juntendi strain L4236 was phylogenetically related to P. juntendi strain SAMN30525517. Conclusion A rare P. juntendi strain was isolated from human feces in southern China with a megaplasmid coharboring bla IMP−1−like and bla OXA−1. Antimicrobial selection pressures may have driven acquisition of drug-resistance gene mutations and carriage of the megaplasmid.

Selenium (Se) is an essential micronutrient for human body that plays an important physiological function in life. Compared with inorganic selenium, organic selenium has high bioavailability and low toxicity, making it a superior source of selenium supplementation. Bioactive selenium-containing peptides are important forms of organic selenium, mainly be isolated from selenium-enriched plants and obtained from synthetic pathway, which have become a research hotspot in recent years due to their multiple healthful biological activities. In this review, the isolation and artificial chelating preparation, and the common sources of plant-derived bioactive selenium-containing peptides, as well as their various bioactivities such as antioxidant, hepatoprotective, immunomodulatory, anticancer, hypotensive are summarized. This review would provide a theoretical reference for the development and utilization of plant-derived bioactive selenium-containing peptides in functional foods and medicines.

Obesity is primarily caused by excessive intake as well as absorption of sugar and lipid. Postprandial surge in distention pressure and intestinal motility accelerates the absorption of nutrients. The response of intestinal epithelial cells to mechanical stimulation is not fully understood. Piezo1, a mechanosensitive ion channel, is widely expressed throughout the digestive tract. However, its function in intestinal nutrient absorption is not yet clear. In our study, excessive lipid deposition was observed in the duodenum of obese patients, while duodenal Piezo1–CaMKK2–AMPKα was decreased when compared to normal-weight individuals. Under high-fat diet condition, the Piezo1iKO mice exhibited abnormally elevated sugar and lipid absorption as well as severe lipid deposition in the duodenum and liver. These phenotypes were mainly caused by the inhibition of duodenal CaMKK2–AMPKα and the upregulation of SGLT1 and DGAT2. In contrast, Yoda1, a Piezo1 agonist, was found to reduce intestinal lipid absorption in diet induced obese mice. Overexpression of Piezo1, stretch and Yoda1 inhibited lipid accumulation and the expression of DGAT2 and SGLT1, whereas knockdown of Piezo1 stimulated lipid accumulation and DGAT2 in Caco-2 cells. Our study reveals a previously unexplored mechanical regulation of nutrient absorption in intestinal epithelial cells, which may shed new light on the therapy of obesity.

Optimization of process conditions for biosynthesis of pinoresinol (Pin), pinoresinol-4-O-β-D-glucopyranoside (PMG) and pinoresinol diglucoside (PDG) by Phomopsis sp. XP-8 assisted Eucommia ulmoides Oliv. was performed. The effects of the cultured method, the addition of Eucommia ulmoides Oliv. and the time of resting on the production of Pin, PMG and PDG by Phomopsis sp. XP-8 assisted Eucommia ulmoides Oliv. were studied. The antioxidative, anticancer properties of the related products were also investigated. The results showed that the optimal yield of Pin, PMG and PDG was obtained at the condition with the PDB medium contained 3% Eucommia ulmoides Oliv. with resting treatment for 5 d. Under the optimal condition, the free radical scavenging ability for DPPH and ABTS+ of metabolite liquid was significantly better than that of the blank culture medium. This indicated that the antioxidative substances were generated during the fermentation process. The hepatocellular carcinoma HepG-2 proliferation was suppressed by the extract solution. The pure product verifications of activity of the standard samples (Pin, PMG and PDG) were performed. It indicated that Pin was identified as the main active ingredient that suppressed HepG-2. The suppressed effect of the metabolite liquid on HepG-2 was a synergistic effect.

The paper analyses the spatial and temporal characteristics of extreme precipitation in the upper reaches of the Yangtze River through extreme precipitation indicators based on the trend method, the Mann–Kendall trend test, and the rescaled extreme deviation extreme deviation using daily precipitation data from 1961 to 2021. The following conclusions were obtained: The overall precipitation in the upper reaches of the Yangtze River is reduced, and the number of rainy days is reduced. The frequency of extreme precipitation is generally reduced, but the spatial difference in the intensity of extreme precipitation is greater, which makes the occurrence of extreme precipitation more concentrated and more destructive. Extreme precipitation indicators showed relatively large fluctuations after 2000, especially in terms of extreme precipitation intensity. The frequency of extreme precipitation in the upper reaches of the Yangtze River is the highest in the main stream of the Yangtze River Basin and the Wujiang River Basin, the intensity of extreme precipitation is in the Jialing River and the Wujiang River Basin, and the accumulation of extreme precipitation is the highest in the Jialing River and the Wujiang River Basin, whereas the maximum value of the station extreme precipitation intensity and frequency is in the Minjiang River Basin. HIGHLIGHTS The overall extreme precipitation characteristics of the upper reaches of the Yangtze River under climate change were analysed.; Differences in extreme precipitation characteristics of the first-tier tributaries of the upper Yangtze River were analysed.; Changes in water resources in the upper reaches of the Yangtze River have a major impact on China's economy, food security, and energy resources.;

Abstract Obesity increases the risk of chronic kidney disease. We have previously demonstrated the benefits of preconception maternal weight loss on fertility and pregnancy outcomes in a mouse model of maternal obesity. Here, we elucidate if preconception weight loss, either by diet modification or the glucose-like peptide 1 agonist liraglutide, used in the treatment of diabetes and obesity, improves maternal kidney outcomes in late gestation. C57BL/6 female mice were fed either a high-fat-diet (HFD) or a chow (control) diet for 8 weeks. To induce pre-pregnancy weight loss, HFD-fed dams were switched to chow diet (HFD-C) or administered liraglutide (0.3 mg/kg subcutaneous) whilst continuing on HFD (HFD-L). Liraglutide was discontinued one week prior to mating. HFD-V mice continued on HFD, with saline injections. A group of HFD-fed dams were ‘diet switched’ to chow after conception (post-conception, HFD-PC). Maternal body weight and glucose tolerance were measured: (1) preconception and (2) during late gestation followed by blood, urine and kidney collection. Serum creatinine, urinary creatinine and albumin, kidney tissue gene expression and protein were measured. Abstract Objective To investigate the current situation of emergency preparation and emergency drill in the CSSD, and analyze its influence on the nurses’ emergency attitude and ability. Methods This study employed a multicenter stratified sampling method, conducted from January to June 2023 using the online survey, participants completed the general data, emergency preparedness and drill questionnaire, public health emergency response questionnaire and emergency capacity scale. An independent samples t test or Kruskal-Wallis test was used to analyse differences in nurses’ emergency capacity and attitudes. Results The data from 15 provinces 55 hospitals in China. Overall, 77.58% of participants’ institutions set up emergency management teams, 85.45% have an emergency plan and revise it regularly. 92.12% store emergency supplies. All survey staff participated in the emergency drill, which predominantly consisted of individual drills (51.52%), with 90.30% being real combat drills, 49.09% of participants engaging in drills every quarter, and 91.52% of the drill’s participants exceeding 50%. The respondents’ emergency attitude score was (29.346 ± 6.029), their emergency ability score was (63.594 ± 10.413), and those with rescue experience showed a more positive attitude (Z = -2.316, P = 0.021). The species of Carpesium lipskyi C.Winkl. 1998 is an important traditional Chinese medicine in China. In this study, the complete chloroplast (cp) genome of C. lipskyi was determined and analyzed. The result showed that the complete cp genome of C. lipskyi was 151,244 bp in length, consisting of a large single-copy (LSC) region of 82,908 bp, a small single-copy (SSC) region of 18,430 bp, and a pair of inverted repeats (IRs) of 24,953 bp. The overall GC content of the C. lipskyi is 37.68%. The species of C. lipskyi possessed 127 genes, including 83 protein-coding genes, 36 transfer RNA genes, and eight ribosomal RNA genes. The present study found that Inula is sister groups with the closest genetic relationship. The obtained knowledge could provide useful information for future phylogenetic, taxonomic, and evolutionary studies on Inuleae.

Abstract The collective modes of the superconducting order parameter fluctuation can provide key insights into the nature of the superconductor. Recently, a family of superconductors has emerged in non-magnetic kagome materials AV3Sb5 (A = K, Rb, Cs), exhibiting fertile emergent phenomenology. However, the collective behaviors of Cooper pairs have not been studied. Here, we report a distinct collective mode in CsV3-x Ta x Sb5 using scanning tunneling microscope/spectroscopy. The spectral line-shape is well-described by one isotropic and one anisotropic superconducting gap, and a bosonic mode due to electron-mode coupling. With increasing x, the two gaps move closer in energy, merge into two isotropic gaps of equal amplitude, and then increase synchronously. The mode energy decreases monotonically to well below $${{2}}{{\Delta }}$$ 2 Δ and survives even after the charge density wave order is suppressed. We propose the interpretation of this collective mode as Leggett mode between different superconducting components or the Bardasis-Schrieffer mode due to a subleading superconducting component.

Abstract. Background:. Acute respiratory distress syndrome (ARDS) is a common cause of respiratory failure in many critically ill patients. Although inflammasome activation plays an important role in the induction of acute lung injury (ALI) and ARDS, the regulatory mechanism of this process is still unclear. When cells are stimulated by inflammation, the integrity and physiological function of mitochondria play a crucial part in pyroptosis. However, the underlying mechanisms and function of mitochondrial proteins in the process of pyroptosis are largely not yet known. Here, we identified the 18-kDa translocator protein (TSPO), a mitochondrial outer membrane protein, as an important mediator regulating nucleotide-binding domain, leucine-rich repeat, and pyrin domain-containing protein 3 (NLRP3) inflammasome activation in macrophages during ALI. Methods:. TSPO gene knockout (KO) and lipopolysaccharide (LPS)-induced ALI/ARDS mouse models were employed to investigate the biological role of TSPO in the pathogenesis of ARDS. Murine macrophages were used to further characterize the effect of TSPO on the NLRP3 inflammasome pathway. Activation of NLRP3 inflammasome was preformed through LPS + adenosine triphosphate (ATP) co-stimulation, followed by detection of mitochondrial membrane potential, reactive oxygen species (ROS) production, and cell death to evaluate the potential biological function of TSPO. Comparisons between two groups were performed with a two-sided unpaired t-test. Results:. TSPO-KO mice exhibited more severe pulmonary inflammation in response to LPS-induced ALI. TSPO deficiency resulted in enhanced activation of the NLRP3 inflammasome pathway, promoting more proinflammatory cytokine production of macrophages in LPS-injured lung tissue, including interleukin (IL)-1β, IL-18, and macrophage inflammatory protein (MIP)-2. Mitochondria in TSPO-KO macrophages tended to depolarize in response to cellular stress. The increased production of mitochondrial damage-associated molecular pattern led to enhanced mitochondrial membrane depolarization and pyroptosis in TSPO-KO cells. Conclusion:. TSPO may be the key regulator of cellular pyroptosis, and it plays a vital protective role in ARDS occurrence and development.

Abstract Cadmium (Cd) pollution is a serious threat to food safety and human health. Minimizing Cd uptake and enhancing Cd tolerance in plants are vital to improve crop yield and reduce hazardous effects to humans. In this study, we designed three Cd concentration stress treatments (Cd1: 0.20 mg·kg−1, Cd2: 0.60 mg·kg−1, and Cd3: 1.60 mg·kg−1) and two foliar silicon (Si) treatments (CK: no spraying of any material, and Si: foliar Si spraying) to conduct pot experiments on soil Cd stress. The results showed that spraying Si on the leaves reduced the Cd content in brown rice by 4.79–42.14%. Si application increased net photosynthetic rate (Pn) by 1.77–4.08%, stomatal conductance (Gs) by 5.27–23.43%, transpiration rate (Tr) by 2.99–20.50% and intercellular carbon dioxide (CO2) concentration (Ci) by 6.55–8.84%. Foliar spraying of Si significantly increased the activities of superoxide dismutase (SOD) and peroxidase (POD) in rice leaves by 9.84–14.09% and 4.69–53.09%, respectively, and reduced the content of malondialdehyde (MDA) by 7.83–48.72%. In summary, foliar Si spraying protects the photosynthesis and antioxidant system of rice canopy leaves, and is an effective method to reduce the Cd content in brown rice.

Objective For elderly femoral neck fracture patients, anemia is one of the most common complications, increasing the risk of postoperative adverse events. Tranexamic acid (TXA) has been widely applied to the perioperative blood management. However, the optimal route of TXA administration in elderly femoral neck fracture remains unclear. The aim of this study is to evaluate the efficacy and safety of oral and intravenous (IV) application of TXA in elderly patients with femoral neck fracture undergoing total hip arthroplasty (THA) and hemiarthroplasty (HA). Methods All elderly patients aged over 65 years old diagnosed with femoral neck fracture admitted to the trauma orthopedics from August 1, 2020 to February 28, 2022 were enrolled in this prospective cohort study. Participants were divided into three groups: oral group: TXA 2g orally 2 h before incision; IV group: intravenous infusion of TXA 1g 15 min before incision; and control group: usual hemostatic method. The primary outcomes were total blood loss, allogeneic transfusion rate, and postoperative thromboembolic events. SPSS 23.0 (IBM, Armonk, NY, USA) was used for statistical analysis, and p ≤ 0.05 was considered statistically significant. Results A total of 100 patients were enrolled, including 32 cases in the oral group, 34 cases in the IV group and 34 cases in the control group. Compared with the control group, the total perioperative blood loss in the oral and IV groups was significantly decreased (763.92 ± 358.64 mL vs 744.62 ± 306.88 mL vs 1250.60 ± 563.37 mL, p = 0.048). No significant difference was identified between the oral and IV groups (p = 0.970). The rate of allogeneic transfusion was lower in the oral and IV groups than in the control group, but the difference had no statistical significant (6 vs 5 vs 12, p = 0.108), However, subgroup analysis showed that the IV and oral groups in patients who underwent THA have significant lower transfusion rate compared with the control group (1 vs 3 vs 7, p = 0.02). During 6 months follow‐up, no thromboembolic events were identified. Two patients (one from the oral group and one from the control group) died of respiratory failure. Since Epstein-Barr virus (EBV) was discovered in 1964, it has been reported to be associated with various malignancies as well as benign diseases, and the pathogenicity of EBV has been widely studied. Several databases have been established to provide comprehensive information on the virus and its relation to diseases and introduce convenient analysis tools. Although they have greatly facilitated the analysis of EBV at the genome, gene, protein, or epitope level, they did not provide enough insight into the genomic variants of EBV, which have been suggested as relevant to diseases by multiple studies. Here, we introduce dbEBV, a comprehensive database of EBV genomic variation landscape, which contains 942 EBV genomes with 109,893 variants from different tissues or cell lines in 24 countries. The database enables the visualization of information with varying global frequencies and their relationship with the human health of each variant. It also supports phylogenetic analysis at the genome or gene level in subgroups of different characteristics. Information of interest can easily be reached with functions such as searching, browsing, and filtering. In conclusion, dbEBV is a convenient resource for exploring EBV genomic variants, freely available at http://dbebv.omicsbio.info.

Background: Deep convolutional neural networks have garnered considerable attention in numerous machine learning applications, particularly in visual recognition tasks such as image and video analyses. There is a growing interest in applying this technology to diverse applications in medical image analysis. Automated three-dimensional Breast Ultrasound is a vital tool for detecting breast cancer, and computer-assisted diagnosis software, developed based on deep learning, can effectively assist radiologists in diagnosis. However, the network model is prone to overfitting during training, owing to challenges such as insufficient training data. This study attempts to solve the problem caused by small datasets and improve model detection performance. Methods: We propose a breast cancer detection framework based on deep learning (a transfer learning method based on cross-organ cancer detection) and a contrastive learning method based on breast imaging reporting and data systems (BI-RADS). Results: When using cross organ transfer learning and BIRADS based contrastive learning, the average sensitivity of the model increased by a maximum of 16.05%. Conclusion: Our experiments have demonstrated that the parameters and experiences of cross-organ cancer detection can be mutually referenced, and contrastive learning method based on BI-RADS can improve the detection performance of the model.

Abstract Background Previous studies have demonstrated a strong association between depression and job burnout among healthcare professionals, but the results have been inconsistent, and there is a lack of in-depth exploration of such a relationship among different healthcare professions. The present study aims to investigate the interrelationships between depression and burnout among Chinese healthcare professionals and whether there are differences in the networks of these symptoms between doctors and nurses. Methods The Maslach Burnout Inventory-General Survey and the 2-item Patient Health Questionnaire were employed to assess job burnout and depression among 3,684 healthcare professionals. The translation has been refined to ensure accuracy and academic suitability. Subsequently, network analysis was conducted on 2,244 participants with a higher level of job burnout to identify core symptoms and explore the associations between job burnout and depression. Results The present study showed a network association between lack of interest and pleasure in things and being exhausted from work, excessive tiredness facing work, tendency to collapse at work, and lack of passion for work than before among healthcare professionals, as well as a notable difference in the network association between lack of interest and pleasure in things and lack of passion for work than before between nurses and doctors. Conclusions The depression-burnout network structures differ between doctors and nurses, highlighting the need for targeted intervention measures for both groups.

Abstract Background Drug use disorders (DUDs) have emerged as one of the most significant public health crises, exerting a substantial influence on both community health and socio-economic progress. The United States (US) also suffers a heavy burden, it is necessary to figure out the situation from multiple perspectives and take effective measures to deal with it. Therefore, using the data from the Global Burden of Diseases, Injuries, and Risk Factors (GBD) 2021, we evaluated this topic. Methods Annual data on DUDs-related burden were collected from the GBD study 2021. We calculated the indicator of estimated annual percentage change (EAPC) to evaluate the changing trend of burden. The Bayesian model for age-period-cohort was introduced to forecast the burden. Results In 2021, the number and age-standardized rate of prevalence were particularly prominent, with 12,146.95 thousand and 3821.43 per 100,000, respectively. Higher burden was also observed in males, 15–45 years old populations, and opioid use disorders subtype. From 1990 to 2021, the DUDs-related burden increased in the US and all states, especially in West Virginia; and the national death-related burden with the highest increase (EAPC = 7.96). Other significant inverse associations were seen between EAPC, age-standardized rates, and socio-demographic index (SDI). Moreover, in the next 14 years, the projected DUDs burden remains exigent. Conclusions The burden of DUDs in the US is heavy and has been enlarging. This study proposes that greater attention should be paid to the strategies in males, the younger population, opioid use disorders, and low-SDI states implemented by decision-makers to achieve goals such as reducing burden.

Abstract. Background:. Cardiovascular disease (CVD) has emerged as the leading cause of death from prostate cancer (PCa) in recent decades, bringing a great disease burden worldwide. Men with preexisting CVD have an increased risk for major adverse cardiovascular events when treated with androgen deprivation therapy (ADT). The present study aimed to explore the prevalence and risk evaluation of CVD among people with newly diagnosed PCa in China. Methods:. Clinical data of newly diagnosed PCa patients were retrospectively collected from 34 centers in China from 2010 to 2022 through convenience sampling. CVD was defined as myocardial infarction, arrhythmia, heart failure, stroke, ischemic heart disease, and others. CVD risk was estimated by calculating Framingham risk scores (FRS). Patients were accordingly divided into low-, medium-, and high-risk groups. χ2 or Fisher’s exact test was used for comparison of categorical variables. Results:. A total of 4253 patients were enrolled in the present study. A total of 27.0% (1147/4253) of patients had comorbid PCa and CVD, and 7.2% (307/4253) had two or more CVDs. The enrolled population was distributed in six regions of China, and approximately 71.0% (3019/4253) of patients lived in urban areas. With imaging and pathological evaluation, most PCa patients were diagnosed at an advanced stage, with 20.5% (871/4253) locally progressing and 20.5% (871/4253) showing metastasis. Most of them initiated prostatectomy (46.6%, 1983/4253) or regimens involving ADT therapy (45.7%, 1944/4253) for prostate cancer. In the present PCa cohort, 43.1% (1832/4253) of patients had hypertension, and half of them had poorly controlled blood pressure. With FRS stratification, as expected, a higher risk of CVD was related to aging and metabolic disturbance. However, we also found that patients with treatment involving ADT presented an originally higher risk of CVD than those without ADT. This was in accordance with clinical practice, i.e., aged patients or patients at advanced oncological stages were inclined to accept systematic integrative therapy instead of surgery. Among patients who underwent medical castration, only 4.0% (45/1118) received gonadotropin releasing hormone antagonists, in stark contrast to the grim situation of CVD prevalence and risk. Conclusions:. PCa patients in China are diagnosed at an advanced stage. A heavy CVD burden was present at the initiation of treatment. Patients who accepted ADT-related therapy showed an original higher risk of CVD, but the awareness of cardiovascular protection was far from sufficient.

Abstract Background Triple Negative Breast Cancer (TNBC) is a particular type of breast cancer with the highest mortality rate. Essential oils are concerned more and more as potential anti-cancer drugs. Methods TNBC cells were treated with different concentrations of navel orange peel essential oil (NOPEO), and then a variety of experiments were performed to investigate the changes in the growth and progression of TNBC cells. MTT assay was performed to detect the proliferation of TNBC cells. The changes of cell cycle and apoptosis were analyzed by FACS. In order to explored the migration of TNBC cells, scratch wound assay was carried out. Western blotting and qPCR were used to examine the expression of proteins and mRNA of related genes. Furthermore, RNA-seq was used to analyze the altered genes and explored the possible signal pathway. Results NOPEO demonstrated dose- and time-dependent suppression of TNBC cell growth. TNBC cells showed an increased percentage of G2/M-phase cells and the protein levels of CyclinB1 and CyclinD1 were decreased after NOPEO treatment. The apoptotic cells were increased in the NOPEO treated TNBC cells. The migration mobility was significantly inhibited by NOPEO. In total, 1376 genes were found to be up-regulated and 1335 genes were down-regulated after NOPEO treatment. According to KEGG and GO pathways, the differentially expressed genes were related to MAPK, Jak/stat and FoxQ signaling pathways. Conclusion This investigation explored the bio-activity and molecular mechanisms of NOPEO against TNBC cells. These results indicated that NOPEO could suppress TNBC growth and migration perhaps via the MAPK and Jak/stat signaling pathways, which may provide theoretical reference for anticancer drug development. NOPEO may be a potential natural product for the chemotherapeutic of TNBC.

Abstract Background Depressive disorders have been identified as a significant contributor to non-fatal health loss in China. Among the various subtypes of depressive disorders, dysthymia is gaining attention due to its similarity in clinical severity and disability to major depressive disorders (MDD). However, national epidemiological data on the burden of disease and risk factors of MDD and dysthymia in China are scarce. Methods This study aimed to evaluate and compare the incidence, prevalence, and disability-adjusted life-years (DALYs) caused by MDD and dysthymia in China between 1990 and 2019. The temporal trends of the depressive disorder burden were evaluated using the average annual percentage change. The comparative risk assessment framework was used to estimate the proportion of DALYs attributed to risk factors, and a Bayesian age-period-cohort model was applied to project the burden of depressive disorders. Results From 1990 to 2019, the overall age-standardized estimates of dysthymia in China remained stable, while MDD showed a decreasing trend. Since 2006, the raw prevalence of dysthymia exceeded that of MDD for the first time, and increased alternately with MDD in recent years. Moreover, while the prevalence and burden of MDD decreased in younger age groups, it increased in the aged population. In contrast, the prevalence and burden of dysthymia remained stable across different ages. In females, 11.34% of the DALYs attributable to depressive disorders in 2019 in China were caused by intimate partner violence, which has increasingly become prominent among older women. From 2020 to 2030, the age-standardized incidence, prevalence, and DALYs of dysthymia in China are projected to remain stable, while MDD is expected to continue declining. Conclusions To reduce the burden of depressive disorders in China, more attention and targeted strategies are needed for dysthymia. It’s also urgent to control potential risk factors like intimate partner violence and develop intervention strategies for older women. These efforts are crucial for improving mental health outcomes in China.

Abstract Background C. Oleifera is among the world’s largest four woody plants known for their edible oil production, yet the contribution rate of improved varieties is less than 20%. The species traditional breeding is lengthy cycle (20–30 years), occupation of land resources, high labor cost, and low accuracy and efficiency, which can be enhanced by molecular marker-assisted selection. However, the lack of high-quality molecular markers hinders the species genetic analysis and molecular breeding. Results Through quantitative traits characterization, genetic diversity assessment, and association studies, we generated a selection population with wide genetic diversity, and identified five excellent high-yield parental combinations associated with four reliable high-yield ISSR markers. Early selection criteria were determined based on kernel fresh weight and cultivated 1-year seedling height, aided by the identification of these 4 ISSR markers. Specific assignment of selected individuals as paternal and maternal parents was made to capitalize on their unique attributes. Conclusions Our results indicated that molecular markers-assisted breeding can effectively shorten, enhance selection accuracy and efficiency and facilitate the development of a new breeding system for C. oleifera.

Abstract Objective To investigate the role of BTG2 in periodontitis and diabetic kidney disease (DKD) and its potential underlying mechanism. Methods Gene expression data for periodontitis and DKD were acquired from the Gene Expression Omnibus (GEO) database. Differential expression analysis identified co-expressed genes between these conditions. The Nephroseq V5 online nephropathy database validated the role of these genes in DKD. Pearson correlation analysis identified genes associated with our target gene. We employed Gene Set Enrichment Analysis (GSEA) and Protein-Protein Interaction (PPI) networks to elucidate potential mechanisms. Expression levels of BTG2 mRNA were examined using quantitative polymerase Chain Reaction (qPCR) and immunofluorescence assays. Western blotting quantified proteins involved in epithelial-to-mesenchymal transition (EMT), apoptosis, mTORC1 signaling, and autophagy. Additionally, wound healing and flow cytometric apoptosis assays evaluated podocyte migration and apoptosis, respectively. Results Analysis of GEO database data revealed BTG2 as a commonly differentially expressed gene in both DKD and periodontitis. BTG2 expression was reduced in DKD compared to normal conditions and correlated with proteinuria. GSEA indicated enrichment of BTG2 in the EMT and mTORC1 signaling pathways. The PPI network highlighted BTG2’s relevance to S100A9, S100A12, and FPR1. Immunofluorescence assays demonstrated significantly lower BTG2 expression in podocytes under high glucose (HG) conditions. Reduced BTG2 expression in HG-treated podocytes led to increased levels of EMT markers (α-SMA, vimentin) and the apoptotic protein Bim, alongside a decrease in nephrin. Lower BTG2 levels were associated with increased podocyte mobility and apoptosis, as well as elevated RPS6KB1 and mTOR levels, but reduced autophagy marker LC3. Conclusion Our findings suggest that BTG2 is a crucial intermediary gene linking DKD and periodontitis. Modulating autophagy via inhibition of the mTORC1 signaling pathway, and consequently suppressing EMT, may be pivotal in the interplay between periodontitis and DKD.

To address the challenges in tracking complex maneuvering extended targets, an effective maneuvering extended target tracking method was proposed for irregularly shaped star-convex using a Transformer network. Initially, the alpha-shape algorithm was used to model the variations in the star-convex shape. In addition, a recursive approach was proposed to estimate the irregular shape of an extended target by detailed derivation in the Bayesian filtering framework. This approach accurately estimated the shape of a static star convex extended target. Moreover, through the structural redesign of the target state transition matrix and the real-time estimation of the maneuvering extended target’s state transition matrix using a transformer network, the accurate tracking of complex maneuvering targets was achieved. Furthermore, the real-time tracking of star convex maneuvering extended targets was achieved by fusing the estimated shape contours with motion states. This study focused on constructing certain complex maneuvering extended target tracking scenarios to assess the performance of the proposed method and the comprehensive estimation capabilities of the algorithm considering both shapes and motion states using multiple performance indicators.

This paper analyzes the species and characteristics of marine polysaccharides from different sources. It also summarizes the preparation methods of marine polysaccharide-based probiotic microcapsules, and expounds the application of marine polysaccharide-based probiotic microcapsules in food industry, biomedicine, aquaculture feed and other fields. It is expected to provide some references for the research direction of probiotic microcapsules and the high-value utilization of marine resources.

Ultra-high-temperature ceramic nanowires have shown increasing potential for use as thermal structural components. Herein, novel single-crystal Hf0.5Ta0.5C solid solution nanowires were synthesized and incorporated with a HfC coating to construct a robust structure with Hf0.5Ta0.5C solid solution nanowires uniformly distributed and interconnected within the coating. The novel Hf0.5Ta0.5C solid solution nanowires could effectively hinder crack propagation through crack tip pinning and crack deflection. This mechanism substantially enhanced the elastic modulus and fracture toughness of the HfC coating by 53.29% and 59.67%, respectively. The toughened HfC coating displayed superior fracture toughness and good interfacial binding strength with the substrate to resist severe oxidation and scouring. Additionally, the high thermal conductivity of the toughened HfC coating promoted heat transmission. Thus, in comparison to the pure HfC coating, the toughened HfC coating displayed smaller mass and linear ablation rates of −0.35 mg·s−1 and −0.46 μm·s−1, which decreased by 39.66% and 36.98%, respectively. Our work not only simultaneously enhances the mechanical properties and ablation resistance of HfC-coated carbon/carbon (C/C) composites but also provides novel prospects for advanced ultrahigh-temperature ceramic nanowires under extreme conditions.

Abstract Background We describe a case in which bilateral optic nerve infiltration and leukemic retinopathy were the initial signs of disease relapse in a patient with Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+-ALL) with central nervous system (CNS) involvement. Case presentation A 65-year-old Asian female with Ph+-ALL in complete remission presented at our institution with symptoms of visual disturbance, central scotoma and pain with eye movement in both eyes for a 1-month duration. Ophthalmic examination revealed remarkable optic disc swelling with multiple flame-shaped peripapillary hemorrhages, retinal venous dilation and retinal hemorrhages in both eyes. She was subsequently referred to the treating oncologist and diagnosed with Ph+-ALL relapse with multiple relapsed diseases involving the bone marrow and CNS. After intrathecal (IT) therapy, her visual acuity dramatically improved, and her leukemic infiltrates decreased. Conclusions To the best of our knowledge, this is the first case report of ALL relapse with CNS involvement presenting as bilateral optic nerve infiltration and leukemic retinopathy in an adult. Hence, we highlight the priority and sensitivity of ophthalmic examinations, as they are noninvasive methods for detecting leukemia relapse.

Ti6Al4V is widely used in industrial fields due to its many excellent properties, but it still has the disadvantages of poor wear resistance. In order to broaden its application range, it needs to be connected with other materials with complementary properties. The method of sealing glass to connect dissimilar materials is widely used because of its low cost and low difficulty. In this study, Ti6Al4V was pre-oxidized by thermal oxidation method, and it was found that the oxidation temperature and holding time had a significant effect on the surface morphology and properties of the alloy. The wetting behavior between bismuthate sealing glass and Ti6Al4V under atmospheric conditions was investigated by sessile drop method. The effect of pre-oxidation temperature on the wettability of the alloy surface was studied by high temperature contact angle measuring instrument during the dynamic heating process. Combined with interface analysis, it was found that the sealing glass reacted with the Ti6Al4V alloy to form Bi4Ti3O12, and the reaction degree of Ti4+ and Bi3+ was the main reason for the wetting of the glass on the alloy surface. It was concluded that the higher the proportion of TiO2 on the surface, the better the wettability.

Abstract El Niño-Southern Oscillation (ENSO) is the dominant mode of interannual climate variability in the tropical Pacific, whose nature nevertheless may change significantly in a warming climate. Here, we show that the predictability of ENSO may decrease in the future. Across the models in the Coupled Model Intercomparison Project Phase 6 (CMIP6), we find a robust decrease of the persistence and predictability for the Central Pacific (CP) ENSO under global warming, notably in passing through the boreal spring. The strength of spring predictability barrier will be increased by 25% in the future. The reduced predictability of CP ENSO is caused by the faster warming over surface ocean in tropical Pacific and, in turn, the enhanced thermodynamical damping rate on CP ENSO in response to global warming. In contrast, the predictability of Eastern Pacific ENSO will not change. Our results suggest that future greenhouse warming will make the prediction of CP ENSO more challenging, with far-reaching implications on future climate predictions.

Abstract N6-methyladenosine (m6A) stands as the most prevalent modified form of RNA in eukaryotes, pivotal in various biological processes such as regulating RNA stability, translation, and transcription. All members within the YT521-B homology (YTH) gene family are categorized as m6A reading proteins, capable of identifying and binding m6A modifications on RNA, thereby regulating RNA metabolism and functioning across diverse physiological processes. YTH domain-containing 2 (YTHDC2), identified as the latest member of the YTH family, has only recently started to emerge for its biological function. Numerous studies have underscored the significance of YTHDC2 in human physiology, highlighting its involvement in both tumor progression and non-tumor diseases. Consequently, this review aims to further elucidate the pathological mechanisms of YTHDC2 by summarizing its functions and roles in tumors and other diseases, with a particular focus on its downstream molecular targets and signaling pathways.

Abstract Research on the development of cognitive selectivity predominantly focuses on attentional selection. The present study explores another facet of cognitive selectivity—memory selection—by examining the ability to filter attended yet outdated information in young children and adults. Across five experiments involving 130 children and 130 adults, participants are instructed to use specific information to complete a task, and then unexpectedly asked to report this information in a surprise test. The results consistently demonstrate a developmental reversal-like phenomenon, with children outperforming adults in reporting this kind of attended yet outdated information. Furthermore, we provide evidence against the idea that the results are due to different processing strategies or attentional deployments between adults and children. These results suggest that the ability of memory selection is not fully developed in young children, resulting in their inefficient filtering of attended yet outdated information that is not required for memory retention.

Abstract Abnormal granulosa cell (GC) death contributes to cyclophosphamide (CTX) induced primary ovarian insufficiency (POI). To investigate the contribution of GCs to POI, gene profiles of GCs exposed to CTX were assessed using RNA-Seq and bioinformatics analysis. The results showed the differentially expressed genes (DEGs) were enriched in the ferroptosis-related pathway, which is correlated with upregulated heme oxygenase 1 (HO-1) and downregulated glutathione peroxidase-4 (GPX4). Using CTX-induced cell culture (COV434 and KGN cells), the levels of iron, reactive oxygen species (ROS), lipid peroxide, mitochondrial superoxide, mitochondrial morphology and mitochondrial membrane potential (MMP) were detected by DCFDA, MitoSOX, C11-BODIPY, MitoTracker, Nonylacridine Orange (NAO), JC-1 and transmission electron microscopy respectively. The results showed iron overload and disrupted ROS, including cytoROS, mtROS and lipROS homeostasis, were associated with upregulation of HO-1 and could induce ferroptosis via mitochondrial dysfunction in CTX-induced GCs. Moreover, HO-1 inhibition could suppress ferroptosis induced GPX4 depletion. This implies a role for ROS in CTX-induced ferroptosis and highlights the effect of HO-1 modulators in improving CTX-induced ovarian damage, which may provide a theoretical basis for preventing or restoring GC and ovarian function in patients with POI.

The influence of three-stage heat treatment on the microstructure and mechanical properties of pulsed laser-arc hybrid welded TC4 titanium alloy joints was studied by experiments and molecular dynamics calculations. The results showed that after the three-stage heat treatment, a large number of fine and diffusely distributed secondary α phases were found in the welded joint, which increased the content of the phase boundary. The TEM characterization and calculation results show that the phase boundary had a strong hindering effect on the dislocation slip. As a result, under three-stage heat treatment conditions, the hardness of the welded joint was increased by 50%, and the yield strength and tensile strength of the welded joints were increased by 24.3% and 13.9%, respectively. The impact toughness of the base metal significantly improved after heat treatment, which was 31.6% higher than that before heat treatment due to the increase in the equiaxed α phase from the weld metal to the base metal. A large number of stacking faults were generated during the tensile deformation of the welded joint before heat treatment due to the low stacking fault energy of the α phase, which could effectively release the stress and hinder the dislocation slip. After heat treatment, the stacking fault of the welded joint decreased owing to the obvious increase in stacking fault energy. The micro-cracks propagated rapidly along the lamellar α phase to fracture. Therefore, the plasticity of the welded joint decreased, although the strength was improved after heat treatment.

Abstract The need for tumor postoperative treatments aimed at recurrence prevention and tissue regeneration have raised wide considerations in the context of the design and functionalization of implants. Herein, an injectable hydrogel system encapsulated with anti-tumor, anti-oxidant dual functional nanoparticles has been developed in order to prevent tumor relapse after surgery and promote wound repair. The utilization of biocompatible gelatin methacryloyl (GelMA) was geared towards localized therapeutic intervention. Zeolitic imidazolate framework-8@ceric oxide (ZIF-8@CeO2, ZC) nanoparticles (NPs) were purposefully devised for their proficiency as reactive oxygen species (ROS) scavengers. Furthermore, injectable GelMA hydrogels loaded with ZC NPs carrying doxorubicin (ZC-DOX@GEL) were tailored as multifunctional postoperative implants, ensuring the efficacious eradication of residual tumor cells and alleviation of oxidative stress. In vitro and in vivo experiments were conducted to substantiate the efficacy in cancer cell elimination and the prevention of tumor recurrence through the synergistic chemotherapy approach employed with ZC-DOX@GEL. The acceleration of tissue regeneration and in vitro ROS scavenging attributes of ZC@GEL were corroborated using rat models of wound healing. The results underscore the potential of the multifaceted hydrogels presented herein for their promising application in tumor postoperative treatments.

Abstract Ovarian metastasis is one of the major causes of treatment failure in patients with gastric cancer (GC). However, the genomic characteristics of ovarian metastasis in GC remain poorly understood. In this study, we enroll 74 GC patients with ovarian metastasis, with 64 having matched primary and metastatic samples. Here, we show a characterization of the mutation landscape of this disease, alongside an investigation into the molecular heterogeneity and pathway mutation enrichments between synchronous and metachronous metastasis. We classify patients into distinct clonal evolution patterns based on the distribution of mutations in paired samples. Notably, the parallel evolution group exhibits the most favorable prognosis. Additionally, by analyzing the differential response to chemotherapy, we identify potential biomarkers, including SALL4, CCDC105, and CLDN18, for predicting the efficacy of paclitaxel treatment. Furthermore, we validate that CLDN18 fusion mutations improve tumor response to paclitaxel treatment in GC with ovarian metastasis in vitro and vivo.

In this paper, the relationship between mechanical properties and microstructure of steels with different Ti contents (0 wt%, 0.044 wt%, 0.087 wt% and 0.140 wt%) is systematically investigated. The results show that the addition of trace amounts of Ti can improve the strength and plasticity of steel by grain refinement, change in precipitation behavior, and obstruction of dislocation motion. The observation of the interaction behavior of TiC with different sizes and dislocations shows that dislocations can shear the small precipitates, but form dislocation pile-up at the boundary of the large precipitates. The difference between the predicted and tested values of yield strength gradually increases with increasing Ti content. The synergistic effect of the formation of precipitates (TiN and its composites) and the coarsening of TiC under a high content of Ti can make the steel more susceptible to cracking and fracture during the tensile process. This study provides a theoretical basis and technical guidance for the design and application of Ti-microalloyed steels by investigating the strengthening and fracture mechanisms of different Ti content.

Laser additive manufacturing (LAM) has attracted significant interest in developing Al alloys due to its potential to fabricate structural components at low cost and high efficiency. Nevertheless, LAM Al alloys still suffer many issues, including heterogeneous microstructure, various defects, and poor mechanical properties, significantly hindering the advancement of LAM Al alloys. This article seeks to provide a comprehensive overview of the current advancements made in the field of LAM Al alloys and attempts to establish an in-depth relationship between microstructural characteristics, defects, and performances. Development status and limitations of LAM technologies are described, microstructural characteristics of Al alloys in LAM are introduced, mechanisms of formed defects and corresponding mitigation measures are discussed, and current quality improvement strategies are summarized. Following the research of previous work, this review will provide an outlook on overcoming the existing challenges and provide a helpful resource and reference for the development of LAM Al alloys.

Keyhole instability and high porosity are prone to occur in laser-arc hybrid welding (LAHW) with an increase in aluminum alloy thickness. In this study, a high-power laser up to 8.0 kW at different oscillating frequencies (f) was used to achieve acceptable hybrid welding of a 10-mm-thick 6082 aluminum alloy. According to the observations of the laser keyhole from a high-speed camera, increasing f from 0 to 100 Hz increased the opening diameter from 1.3 to 1.6 mm and decreased the collapse frequency from 642 to 353 Hz. With the improved keyhole stability, the weld porosity decreased from 4.5% to 1.8%. In addition, beam oscillation refined the weld microstructure by 15% and transferred the tensile fracture from the weld center to the heat-affected zone (HAZ). Although the change in the ultimate tensile strength (UTS) was not obvious, the elongation increased by 25%. Beam oscillation promoted the formation of turbulent flow inside the molten pool, homogenized the melt flow, and reduced the necking tendency of the keyhole caused by the melt flow, thus greatly improving the keyhole stability.
