To improve the comprehensive utilization value of the by-product residue of Pueraria thomsonii (kudzu), the extraction of conjugated phenolics from two common by-product residues (kudzu peel and kudzu dregs) was optimized. Infrared spectroscopy and scanning electron microscopy were used to compare the microstructure of the by-products of kudzu before and after extraction. The composition of the conjugated phenolics was characterized by LC-MS and the antioxidant activity of the two by-product residues was determined. The results showed that the optimal extraction conditions for the conjugated phenolics in the by-product kudzu dregs were as follows: Liquid to material ratio of 30:1 mL/g, hydrolysis time of NaOH solution of 4 h, and concentration of NaOH solution of 3 mol/L, and the yield of conjugated phenolics under these conditions was 2.54±0.070 mg/g. Accordingly, the optimal extraction conditions for conjugated phenolics in the by-product kudzu bark were 40:1 mL/g liquid/feed ratio, 5 h hydrolysis time in NaOH solution, and 3 mol/L concentration of NaOH solution, and the yield of conjugated phenolics under these conditions was 5.087±0.155 mg/g. The ester bond between phenolics and cellulose in the by-products, and the overall fiber structure was broken, which effectively facilitated the release of conjugated phenolics. The composition and content of conjugated phenolics in the two by-product residues differed greatly, with phenolic acids and flavonoids dominating the conjugated phenolics in kudzu peel, while flavonoids dominated the conjugated phenolics in kudzu dregs. The conjugated phenolics in both kudzu peel and kudzu dregs had strong antioxidant activity, and the antioxidant capacity of conjugated phenolics in kudzu peel was found better than that of kudzu dregs. In conclusion, the by-product residues of kudzu are rich in conjugated phenolics and have significant antioxidant activity, which is of good value for development and utilization.

Phenolics have various of physiological effects such as antioxidant, anti-inflammation, and improving intestinal health, as well as playing an important role in the growth of plant and the regulation of human health. Compared with the free phenolics that can be directly extracted, the conjugated phenolics that are bound to polysaccharides, proteins, lipids, and other components have a higher proportion and a wider distribution. The conjugated phenolics have potential application value and increasing attention. During the entire chain from plant growth to food processing and then to human digestion and utilisation, the conjugated phenolics undergo complex changes in composition, structure, migration transformation, and storage characteristics due to the influence of many factors. The study of its accumulation and release patterns is particularly important to enhance its biological activity and improve the utilisation rate of phenolics. Therefore, this paper provides a brief overview on the release of conjugated phenolics during plant growth, food processing, and human digestion, with a view to providing theoretical guidance for elucidating the release pattern of bound phenolics and improving their utilisation.

Abstract Background In the tumor microenvironment (TME), a bidirectional relationship exists between hypoxia and lactate metabolism, with each component exerting a reciprocal influence on the other, forming an inextricable link. The aim of the present investigation was to develop a prognostic model by amalgamating genes associated with hypoxia and lactate metabolism. This model is intended to serve as a tool for predicting patient outcomes, including survival rates, the status of the immune microenvironment, and responsiveness to therapy in patients with lung adenocarcinoma (LUAD). Methods Transcriptomic sequencing data and patient clinical information specific to LUAD were obtained from comprehensive repositories of The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO). A compendium of genes implicated in hypoxia and lactate metabolism was assembled from an array of accessible datasets. Univariate and multivariate Cox regression analyses were employed. Additional investigative procedures, including tumor mutational load (TMB), microsatellite instability (MSI), functional enrichment assessments and the ESTIMATE, CIBERSORT, and TIDE algorithms, were used to evaluate drug sensitivity and predict the efficacy of immune-based therapies. Results A novel prognostic signature comprising five lactate and hypoxia-related genes (LHRGs), PKFP, SLC2A1, BCAN, CDKN3, and ANLN, was established. This model demonstrated that LUAD patients with elevated LHRG-related risk scores exhibited significantly reduced survival rates. Both univariate and multivariate Cox analyses confirmed that the risk score was a robust prognostic indicator of overall survival. Immunophenotyping revealed increased infiltration of memory CD4 + T cells, dendritic cells and NK cells in patients classified within the high-risk category compared to their low-risk counterparts. Higher probability of mutations in lung adenocarcinoma driver genes in high-risk groups, and the MSI was associated with the risk-score. Functional enrichment analyses indicated a predominance of cell cycle-related pathways in the high-risk group, whereas metabolic pathways were more prevalent in the low-risk group. Moreover, drug sensitivity analyses revealed increased sensitivity to a variety of drugs in the high-risk group, especially inhibitors of the PI3K-AKT, EGFR, and ELK pathways. Conclusions This prognostic model integrates lactate metabolism and hypoxia parameters, offering predictive insights regarding survival, immune cell infiltration and functionality, as well as therapeutic responsiveness in LUAD patients. This model may facilitate personalized treatment strategies, tailoring interventions to the unique molecular profile of each patient’s disease.

Seedling is a crucial stage in the growth and development of plants, and the expansion and persistence of plant populations can be achieved through seed regeneration. Sheep grazing, fertilization, light, soil moisture, vegetation diversity and biomass, and litter all have potential impacts on species regeneration. We measured vegetation diversity, annual net primary productivity (ANPP), litter, ground photosynthetically active radiation (PAR), and soil moisture of alpine meadows under sheep grazing and nitrogen addition treatments, and studied their effects on the dicotyledonous seedling abundance and diversity using linear regression models (LMs) and structural equation models (SEMs). We found that sheep grazing reduced ANPP, increased vegetation diversity and PAR, and decreased soil moisture. Fertilization increased ANPP and litter, decreased vegetation diversity and PAR, but had no effect on soil moisture. Sheep grazing and fertilization both reduced the abundance of dicotyledonous seedlings, and simultaneously fertilization can reduce the diversity of dicotyledonous seedlings, while sheep grazing had no effect on the diversity of dicotyledonous seedlings. LMs showed that vegetation diversity, ANPP, and litter, rather than light and soil moisture, affected dicotyledonous seedling abundance and diversity. SEMs revealed that sheep grazing and fertilization indirectly influenced seedling regeneration through vegetation diversity rather than ANPP and litter. Our research will increase our understanding of the dicotyledonous plant regeneration process in alpine grasslands and facilitate the development of strategies for management and protection of alpine grassland.

Abstract Plants can adapt to environmental changes by adjusting their functional traits and biomass allocation. The size and number of flowers are functional traits related to plant reproduction. Life history theory predicts that there is a trade‐off between flower size and number, and the trade‐off can potentially explain the adaptability of plants. Elevation gradients in mountains provide a unique opportunity to test how plants will respond to climate change. In this study, we tried to better explain the adaptability of the alpine plant Gentiana lawrencei var. farreri in response to climate change. We measured the flower size and number, individual size, and reproductive allocation of G. lawrencei var. farreri during the flowering period along an elevation gradient from 3200 to 4000 m, and explored their relationships using linear mixed‐effect models and the structural equation model. We found that with elevation increasing, individual size and flower number decreased and flower size increased, while reproductive allocation remained unchanged. Individual size positively affected flower number, but was not related to flower size; reproductive allocation positively affected flower size, but was not related to flower number; there is a clear trade‐off between flower size and number. We also found that elevation decreased flower number indirectly via directly reducing individual size. In sum, this study suggests that G. lawrencei var. farreri can adapt to alpine environments by the synergies or trade‐offs among individual size, reproductive allocation, flower size, and flower number. This study increases our understanding of the adaptation mechanisms of alpine plants to climate change in alpine environments.

BackgroundTGFB-induced factor homeobox 2 (TGIF2), a member of the Three-Amino-acid-Loop-Extension (TALE) superfamily, has been implicated in various malignant tumors. However, its prognostic significance in glioma, impact on tumor immune infiltration, and underlying mechanisms in glioma development remain elusive.MethodsThe expression of TGIF2 in various human normal tissues, normal brain tissues, and gliomas was investigated using HPA, TCGA, GTEx, and GEO databases. The study employed several approaches, including Kaplan-Meier analysis, ROC analysis, logistic regression, Cox regression, GO analysis, KEGG analysis, and GSEA, to explore the relationship between TGIF2 expression and clinicopathologic features, prognostic value, and potential biological functions in glioma patients. The impact of TGIF2 on tumor immune infiltration was assessed through Estimate, ssGSEA, and Spearman analysis. Genes coexpressed with TGIF2 were identified, and the protein-protein interaction (PPI) network of these coexpressed genes were constructed using the STRING database and Cytoscape software. Hub genes were identified using CytoHubba plugin, and their clinical predictive value was explored. Furthermore, in vitro experiments were performed by knocking down and knocking out TGIF2 using siRNA and CRISPR/Cas9 gene editing, and the role of TGIF2 in glioma cell invasion and migration was analyzed using transwell assay, scratch wound-healing assay, RT-qPCR, and Western blot.ResultsTGIF2 mRNA was found to be upregulated in 21 cancers, including glioma. High expression of TGIF2 was associated with malignant phenotypes and poor prognosis in glioma patients, indicating its potential as an independent prognostic factor. Furthermore, elevated TGIF2 expression positively correlated with cell cycle regulation, DNA synthesis and repair, extracellular matrix (ECM) components, immune response, and several signaling pathways that promote tumor progression. TGIF2 showed correlations with Th2 cells, macrophages, and various immunoregulatory genes. The hub genes coexpressed with TGIF2 demonstrated significant predictive value. Additionally, in vitro experiments revealed that knockdown and knockout of TGIF2 inhibited glioma cell invasion, migration and suppressed the epithelial-mesenchymal transition (EMT) phenotype.ConclusionTGIF2 emerges as a potential biomarker for glioma, possibly linked to tumor immune infiltration and EMT.

Abstract Photo‐assisted uranium reduction from uranium mine wastewater is expected to overcome the competition between impurity ions and U(VI) in the traditional process. Here, B‐TiO2@Co2P‐X S‐scheme heterojunction with metal‐oxygen‐hydrogen (M‐O‐H) is developed insitu modification for photo‐assisted U(VI) (hexavalent uranium) reduction. Relying on the DFT calculation and Hard‐Soft‐Acid‐Base (HSAB) theory, the introduction of metal‐oxygen‐hydrogen (M‐O‐H, hard base) metallic bonds in the B‐TiO2@Co2P‐X is found to enhance the hydrophilicity and the capture capability for uranyl ion (hard acid). Accordingly, B‐TiO2@Co2P‐500 hybrid nanosheets exhibit excellent U(VI) reduction ability (>98%) in the presence of competing ions. By self‐consistent energy band calculations and in‐situ KPFM spectral analysis, the formation of the internal electric field between B‐TiO2 and Co2P at the heterojunction is proven, offering a strong driving force and atomic transportation highway for accelerating the S‐scheme charge carriers directed migration and promoting the photocatalytic reduction of uranium. This work provides a valuable route to explore the functionally modified photocatalyst with high‐efficiency photoelectron separation for U(VI) reduction.

Disulfidptosis, a newly revealed form of cell death, regulated by numerous genes that has been recently identified. The exact role of disulfidptosis in lung adenocarcinoma (LUAD) still uncertain. Objective of this study was to explore potential prognostic markers among disulfidptosis genes in LUAD. By combining transcriptomic information from Gene Expression Omnibus databases and The Cancer Genome Atlas, we identified differentially expressed and prognostic disulfidptosis genes. By conducting least absolute shrinkage and selection operator with multivariate Cox regression, four disulfidptosis genes were selected to create the prognostic signature. The implementation of the signature separated the training and validation cohorts into groups with high- and low-risk. Subsequently, the model was verified by conducting an independent analysis of receiver operating characteristic (ROC) curves. Further comparisons were made between the two risk-divided groups with regards the tumor microenvironment, immune cell infiltration, immunotherapy response, and drug sensitivity. The signature was constructed using four disulfidptosis-related genes: SLC7A11, SLC3A2, NCKAP1, and GYS1. According to ROC curves, the signature was effective for predicting LUAD prognosis. In addition, the prognostic signature correlated with sensitivity to chemotherapeutic agents and the efficacy of immunotherapy in LUAD. Finally, through external validation, we showed that NCKAP1 are correlated with tumor migration, proliferation, and invasion of LUAD cells. GYS1 affects immune cell, especially M2 macrophage infiltration in the tumor microenvironment. The disulfidptosis four-gene model can reliably predict the prognosis of patients diagnosed with LUAD, thereby providing valuable information for clinical applications and immunotherapy.

To investigate the digestion and absorption of wall-broken Ganoderma lucidum spores in the oral and gastrointestinal tracts and the effects on the intestinal environment. In this study, wall-broken Ganoderma lucidum spore (GLS) by mechanical milling, microwave and ultrasonic methods were subjected to in vitro simulation of human oral, gastric and small intestinal digestive systems and dialysis models, followed by in vitro fermentation of the digested substrate. The loss rate, release of polysaccharides and triterpenoids, bioacceptability and dialysis rate of Ganoderma lucidum spore at each stage of digestion as well as the pH changes of the small intestine digested substrate during 0, 6, 12, 24 and 48 h in vitro fermentation were measured. The results showed that the stomach and small intestine were the main digestion sites of Ganoderma lucidum spores, and the average mass loss rate of the broken wall group reached 23.84%, among which the mechanically milled group was more easily digested with a mass loss rate of 29.46%. 97% of the polysaccharides were dissolved in the gastrointestinal fluid, and the bioacceptability of polysaccharides in the mechanically milled group had the highest level of 87.33%. Triterpenes were only minimally soluble in the simulated digestion of the small intestine in the wall-breaking group, averaging about 1.50±0.04 mg/g, and 95.2% of the triterpenes entered the colon with the precipitation. The in vitro fermentation results showed that the pH value of the fermentation broth decreased continuously from 0 to 12 h and reached the end of fermentation at 12 h. Comparing the digestive characteristics of the three types of wall-broken Ganoderma lucidum spores, the quality of the mechanical milling method of wall-broken was considerable, and it was a preferable choice for enterprise scale wall-broken production. In conclusion, wall-breaking was conducive to increasing the release and bioacceptability of the active ingredients of Ganoderma lucidum spores in the human digestive environment, promoting colonic fermentation and acid production, and helping to regulate the human intestinal environment.

Resistant starch has shown great advantages in maintaining intestinal ecology, regulating glycolipid metabolism, and protecting against cardiovascular diseases due to its anti-digestibility and probiotic function. Pueraria, lotus seeds, yam root and other edible medicinal substances that rich in resistant starch have gradually become a hot spot for researchers. In this paper, the research progress on the physicochemical properties and physiological functions of resistant starch derived from edible medicinal substances is reviewed. Its processing adaptability and application scenarios as raw food materials have also been discussed. Resistant starch derived from edible medicinal substances is widely used in food industry due to its good physical and chemical processing properties. It can improve the quality of food and has a positive impact on disease prevention and treatment, with physiological functions such as regulating intestinal flora, improving the intestinal barrier, reducing inflammatory factors, regulating the body's glucose metabolism and inhibiting fat accumulation. The physicochemical properties, physiological functions and application scenarios of resistant starch from different sources differ significantly and the underlying mechanisms need to be investigated in depth. Whether resistant starch encapsulate and carry specific functional components from edible medicinal substances to perform specific physiological functions also needs to be further explored. This paper may provide some basis and ideas for the further development of resistant starch in starch-based edible medicinal substances.

Gastrointestinal dysfunction can cause a few chronic diseases or other health problems. Maintaining the health of the gastrointestinal tract is essential to improve human health and quality of life. Medicinal and edible substances as main raw materials have shown greater benefits in improving gastrointestinal tract function. This paper summarizes the medicinal and edible substances and ancient prescriptions containing medicinal and edible substances, and analyzes the mechanism of medicinal and edible substances and compound prescriptions for improving gastrointestinal tract function from three aspects: Anti-Helicobacter pylori, digestion promotion and gastric mucosa protection. Finally, we analyze the efficacy and formulae of existing food products for improving gastrointestinal tract on the market, in order to provide some ideas for further development of dietary resources and research of dietary therapy products with improving gastrointestinal tract functions.

Abstract Three different methods, including autoclaving, autoclaving–debranching, and purification, were used to prepare Kudzu‐resistant starch (KRS) from Kudzu starch (KS). The physicochemical properties, such as thermodynamic properties, pasting properties, solubility, swelling, and coagulability, as well as the in vitro digestive characteristics of the three kinds of KRS were studied. The results showed that the morphology of starch granules of KRS prepared by autoclave, autoclave enzymatic hydrolysis, and purification methods was changed and the relative crystallinity was significantly decreased compared with the original starch. X‐ray diffraction (XRD) showed that KRS exists in the form of C and C+V crystalline form. There was a significant increase in the pasting temperature and a remarkable decrease in the peak viscosity and the expansion degree of the KRS prepared by all three methods. The solubility of the resistant starch (RS) obtained by autoclaving–debranching and that by purification were both increased compared to that of native KS, while the solubility of the RS obtained by autoclaving was decreased. Meanwhile, the retrogradation of the three RS was also improved to varying degrees. The contents of RS in the samples were: P‐KRS (71%) > DA‐KRS (43%) > A‐KRS (42%) > KS (9%). Simulated human in vitro digestion experiments showed that RS has stronger antidigestibility properties than native starch. Among them, the RS prepared by the purification method has stronger antidigestive properties, and it is predicted that it may have a better potential value in regulating blood glucose. These results indicated that the processing properties of KRS, especially the digestibility, are significantly improved and can be used as a new functional food ingredient, which deserves thorough study.

Non-alcoholic fatty liver disease (NAFLD) is a hepatic manifestation of metabolic syndrome with a global prevalence. Impaired gut barrier function caused by an unhealthy diet plays a key role in disrupting the immune-metabolic homeostasis of the gut-liver axis (GLA), leading to NAFLD. Therefore, dietary interventions have been studied as feasible alternative therapeutic approaches to ameliorate NAFLD. Resistant starches (RSs) are prebiotics that reduce systemic inflammation in patients with metabolic syndrome. The present review aimed to elucidate the mechanisms of the GLA in alleviating NAFLD and provide insights into how dietary RSs counteract diet-induced inflammation in the GLA. Emerging evidence suggests that RS intake alters gut microbiota structure, enhances mucosal immune tolerance, and promotes the production of microbial metabolites such as short-chain fatty acids (SCFAs) and secondary bile acids. These metabolites directly stimulate the growth of intestinal epithelial cells and elicit GPR41/GPR43, FXR, and TGR5 signaling cascades to sustain immune-metabolic homeostasis in the GLA. The literature also revealed the dietary-immune-metabolic interplay by which RSs exert their regulatory effect on the immune-metabolic crosstalk of the GLA and the related molecular basis, suggesting that dietary intervention with RSs may be a promising alternative therapeutic strategy against diet-induced dysfunction of the GLA and, ultimately, the risk of developing NAFLD.

Abstract Puerariae Lobatae Radix (PLR) is the dried root of the leguminous plant Pueraria lobata and is a common component of health products and medicines. Although it is considered safe, some studies have reported that PLR has hepatotoxicity and estrogen‐like effects. In this study, the safety of high doses of PLR water extract administered to adolescent SD rats for 30 days was evaluated by biochemical, histopathological, and metabolomic analyses. Overall, there were no significant differences between the low‐dose and blank control groups in parameter values, including organ wet weight, organ coefficient, routine blood indicators, serum biochemical indexes of liver and renal function, levels of estradiol and testosterone, histopathological parameters, and primary differential metabolite profiles. Compared with the blank control group, the high‐dose group may have a certain effect on the liver. These effects might be mediated by abnormal phenylalanine, tyrosine, and tryptophan biosynthesis or phenylalanine metabolism. However, histopathological analyses did not show differences in the liver, kidney, breast, uterus, ovary, testis, and epididymis between the control group and the group treated with a high dose of PLR water extract. PLR water extract did not significantly promote the precocity of male and female sexual organs. Overall, PLR water extract is relatively safe for adolescent SD rats.

Background. In recent years, carbapenem-resistant Klebsiella pneumoniae (CRKP) has emerged rapidly in China with the abuse and overuse of antibiotics, and infections caused by CRKP pose a serious threat to global public health safety. The present study aimed to explore the epidemiological characteristics of CRKP isolates in Northern China and to elucidate their drug resistance mechanisms. Methods. 45 CRKP strains were consecutively collected at a teaching hospital from March 1st, 2018 to June 30th, 2018. Antimicrobial susceptibility was determined by the VITEK2 compact system and microbroth dilution method. Polymerase chain reaction (PCR) and sequencing were used to analyze multilocus sequence typing (MLST), drug resistance determinants, and plasmid types. The transfer of resistance genes was determined by conjugation. All statistical analysis was performed using SPSS 22.0 software. Results. All 45 isolates showed multidrug resistance (MDR). MLST analysis showed ST11 (48.9%, 22/45) was the most frequent type. All of the 45 CRKP isolates contained carbapenemase genes, extended-spectrum β-lactamase (ESBL) genes, and plasmid-mediated quinolone resistance (PMQR) genes. For carbapenemase genes, KPC-2 (93.3%, 42/45) was the main genotype, and followed by GES (37.8%, 17/45) and NDM-1 (11.1%, 5/45). Plasmid typing analysis showed that IncFII and IncFIB were the most prevalent plasmids. The carbapenem resistance rate of K.pneumoniae was 11.4% and ICU was the main CRKP infection source. Conclusions. ST11 is the most frequent sequence type and KPC-2 is the predominant carbapenemase of CRKP strains in Northern China. KPC-2-ST11 are representative clonal lineages.

In view of the problem of how to determine the position of the sealing hole and increase the concentration of gas extraction, this article proposes the use of a high-pressure gas attenuation method to test the key sealing position and the comparison test of the sealing method using the new sealing position and the traditional sealing position, proposes a method to determine the range of the coal seam roadway loosening belt by measuring the pressure attenuation, and develops a set of devices for rapid determination of the range of the loose belt. Through the field test in Zhangcun Coal Mine, it is shown that when the sealing range is 7.5 m∼18 m, the best effect can be achieved, and when the sealing length is greater than the loose belt, the extraction concentration is higher, and the concentration attenuation and long drilling life are long.

The interaction of protein carrier and polyphenol is variable due to their environmental sensitivity. In this study, the interaction between resveratrol and whey protein isolate (WPI), sodium caseinate (SC) and soy protein isolate (SPI) during storage were systematically investigated from the aspects of polyphenol loading, antioxidant activity and oxidability. It was revealed that resveratrol loaded more in the SPI core and existed both in the core of SC micelles and on the particle surface, while WPI and resveratrol mainly formed in complexes. The loading capacity of the three proteins ranked in order SC > SPI > WPI. ABTS assay showed that the antioxidant activity of the protein carriers in the initial state was SC > SPI > WPI. The results of sulfhydryl, carbonyl and amino acid analysis showed that protein oxidability was SPI > SC > WPI. WPI, with the least oxidation, improved the storage stability of resveratrol, and the impact of SC on resveratrol stability changed from a protective to a pro-degradation effect. Co-oxidation occurred between SPI and resveratrol during storage, which refers to covalent interactions. The data gathered here suggested that the transition between the antioxidant and pro-oxidative properties of the carrier is the primary factor to investigate its protective effect on the delivered polyphenol.

Flavonoids are well known for their extensive health benefits. However, few studies compared the differences between flavonoid O-glycoside and C-glycoside. In this work, flavonoid O-glycoside (isoquercitrin), C-glycoside (orientin), and their aglycones (quercetin and luteolin) were chosen to compare their differences on antioxidant activities and metabolism during in vitro digestion and in vivo. In vitro digestion, the initial antioxidant activity of the two aglycones was very high; however, they both decreased more sharply than their glycosides in the intestinal phase. The glycosidic bond of flavonoid O-glycoside was broken in the gastric and intestinal stage, while the C-glycoside remained unchanged. In vivo, flavonoid O-glycoside in plasma was more elevated than C-glycoside on the antioxidant activity; however, flavonoid C-glycoside in urine was higher than O-glycoside. These results indicate that differences of flavonoid glycosides and their aglycones on antioxidant activity are closely related to their structural characteristics and metabolism in different samples. Aglycones possessed higher activity but unstable structures. On the contrary, the sugar substituents reduced the activity of flavonoids while improving their stability and helping to maintain antioxidant activities after digestion. Especially the C-glycoside was more stable because the stability of the C–C bond is higher than that of the C–O bond, which contributes to the difference between flavonoid O-glycoside and C-glycoside on the absorption and metabolism in vivo. This study provided a new perspective for comparing flavonoid O-glycoside, flavonoid C-glycoside, and their aglycones on their structure–activity relationship and metabolism.

N6-methyladenosine (m6A) is one of the most prevalent epigenetic modifications of eukaryotic RNA. The m6A modification is a dynamic and reversible process, regulated by three kinds of regulator, including m6A methyltransferases, demethylases and m6A-binding proteins, and this modification plays a vital role in many diseases, especially in cancers. Accumulated evidence has proven that this modification has a significant effect on cellular biological functions and cancer progression; however, little is known about the effects of the m6A modification in non-small cell lung cancer (NSCLC). In this review, we summarized how various m6A regulators modulate m6A RNA metabolism and demonstrated the effect of m6A modification on the progression and cellular biological functions of NSCLC. We also discussed how m6A modification affects the treatment, drug resistance, diagnosis and prognosis of NSCLC patients.

The development of diabetes mellitus is related to oxidant stress induced by a high carbohydrate/high-fat diet (HFD). Quercetin, as a major bioactive component in Toona sinensis leaves (QTL), is a natural antioxidant. However, the exact mechanism by which QTL ameliorate diabetes mellitus is still unknown. In this study, we investigated the hypoglycemic effects and hepatocytes protection of QTL on HFD and alloxan induced diabetic mice. Intragastric administration of QTL significantly reduced body weight gain, serum glucose, insulin, total cholesterol, triglyceride, low density lipoprotein-cholesterol, alanine aminotransferase, and aspartate aminotransferase serum levels compared to those of diabetic mice. Furthermore, it significantly attenuated oxidative stress, as determined by lipid peroxidation, nitric oxide content, and inducible nitric oxide synthase activity and as a result attenuated liver injury. QTL also significantly suppressed the diabetes-induced activation of the p65/NF-κB and ERK1/2/MAPK pathways, as well as caspase-9 and caspase-3 levels in liver tissues of diabetic mice. Finally, micrograph analysis of liver samples showed decreased cellular organelle injury in hepatocytes of QTL treated mice. Taken together, QTL can be viewed as a promising dietary agent that can be used to reduce the risk of diabetes mellitus and its secondary complications by ameliorating oxidative stress in the liver.
