20. Effect of trimetazidine on immune function in rats with Streptococcus pneumoniae pneumonia through cGAS-STING pathway.

Abstract

Objective: To investigate effect of trimetazidine (TMZ) on immune function of Streptococcus pneumoniae (SP) pneumonia rats through cyclic guanylate adenylate synthase (cGAS)-stimulator of interferon gene (STING) pathway. Methods: Rats were randomly separated into model group, TMZ group and DMXAA group (cGAS-STING signaling pathway activator). SP pneumonia rat model was replicated by intranasal instillation of SP, while rats in control group were infused with an equal amount of physiological saline. ELISA was applied to detect IL-1β and IL-10 levels. Flow cytometry was applied to detect CD4*T and CD8*T levels. Spleen and thymus indexes of rats in each group were compared. Transmission turbidity method was applied to detect IgG and IgA levels. HE staining was applied to observe pathological changes in lung tissue. Western blot was applied to detect cGAS-STING pathway proteins expressions. Results: Compared with control group, rats in model group showed an increase in alveolar wall and lung interstitium, accompanied by edema, bleeding and inflammatory cell infiltration, IL-1β, CD8*T cells levels, lung tissue pathological injury score, cGAS, STING expressions were increased, IL-10, CD4*T cells levels, CD4*T/CD8*T, spleen and thymus indexes, IgG, IgA levels were decreased (P<0.05). Compared with model group, rats in TMZ group showed reduced thickening of alveolar walls, great improvement in alveolar structure, and less infiltration of inflammatory cells, IL-1β, CD8*T cells levels, lung tissue pathological injury score, cGAS, STING expressions were decreased, IL-10, CD4*T cells levels, CD4*T/CD8*T, spleen and thymus indexes, IgG, IgA levels were increased (P<0.05). Compared with TMZ group, IL-1(3, CD8*T levels, pathological injury score of lung tissue, cGAS and STING expressions in DMXAA group were increased, while IL-10, CD4*T cells levels, CD4*T/CD8*T, spleen and thymus index, IgG and IgA levels were decreased (P<0.05). Conclusion: TMZ may inhibit inflammation by inhibiting cGAS-STING signaling pathway, and enhance immune function of rats with SP pneumonia. [ABSTRACT FROM AUTHOR]

43. Investigations on corrosion behaviors of super-strength sucker rod FG20 steel in high SO42− environment

Abstract

In some sour reservoirs and tertiary oil recovery blocks, SO42− in the well fluid can cause the corrosion and corrosion fatigue of the sucker rods. In this paper, the corrosion behaviors of super-strength sucker rod FG20 (16Mn2SiCrMoVTiA) steel in the well fluid are investigated by electrochemical measurements, and electron probe micro-analyzer (EPMA) analysis. The results show that FG20 steel has a favorable corrosion resistance in neutral solution. When the hydrogen ions increase, the hydrolysis of SO42− greatly accelerates the corrosion of FG20 steel. The energy dispersive X-ray (EDX) results demonstrate that the corrosion process of FG20 steel in neutral well fluid is an oxygen concentration process, and the protective FeCO3 and Fe2O3 on the surface of the samples can prevent the further corrosion. With the increase of the acidity in the well fluid, the corrosion process converts into a sulphide concentration process, and the sloppy FeS and mackinawite film cannot provide effective protection for the specimens, resulting in the increase of corrosion rate. 46. Limit state design of oil and gas well casings

Abstract

The casing is widely used as a protective conduit during all phases of drilling operations and production in the oil and gas industry. Traditionally casings were designed using working stress design, which had a number of shortcomings such as poor economics, inflexibility and uneven risk. This thesis was initiated by the increasing demand for an improved design of casings for the oil and gas industry. A new approach using the concept of limit state design is proposed to remedy the limitations of the present design code. Limit state design employs the probability of failure rather than the usage of a safety factor, from which the designer can gain an overall idea of the safety and adequacy of the design. The main objective of this thesis is to set up a set of limit state design equations for casings under different loadings. This objective is tackled by way of investigations into three fundamental casing failure modes, i. e. casing collapse, casing burst and casing axial tensile failure. Simple equations are proposed for the calculation of the load terms in the limit state design equations for the three failure modes. A comprehensive finite element methodology is developed to investigate the ultimate strength of casings with imperfections under different loadings. Extensive comparisons between finite element models and historical experimental data demonstrate that, if the variables are known, the ultimate strength of a casing can be predicted to a satisfactory degree of accuracy using the finite element method. Detailed parametric studies have been performed to investigate the effects of major factors (i. e., the ratio of outside diameter to wall thickness, ovality, eccentricity, material hardening, anisotropy and residual stress) on the casing strength. Existing design equations are assessed by means of full-scale test data, where they are found to be only accurate within a certain region. The investigations for new limit state design equations have been performed by employing a new concept of generalized material behaviour, which is constructed from experimental data and implemented in the finite element simulation. A set of limit state design equations are derived after regression analysis of the numerical results. Comparisons demonstrate that, the new design equations are capable of providing more accurate predictions of casing strength without compromising safety. The limit state design approach is provided in a structured way with a detailed design flow chart to enable a casing designer with a conventional engineering background to assess casing design using the limit state design methodology. It is anticipated that the implementation of a limit state design methodology in the design of casings will lay the foundation for an increased safety awareness whilst enhancing cost savings.
