1. SOWAHA as a Cancer Suppressor Gene Influence Metabolic Reprogramming

Abstract

SOWAHA is a protein-coding gene, also known as ANKRD43. Studies have indicated that SOWAHA can serve as a prognostic biomarker in colorectal cancer and pancreatic cancer. However, there are few reports about SOWAHA in other types of cancer and the specific mechanism of action of SOWAHA in cancer is also not clear. Based on National Center for Biotechnology Information (NCBI), The Cancer Genome Atlas (TCGA), Genotype-Tissue Expression Project (GTEx), cBioPortal, Human Protein Atlas (HPA), etc., we adopted bioinformatics methods to uncover the potential tumor genomic features of SOWAHA, including the correlation with prognosis, gene mutation, immune cell infiltration, and DNA methylation in different tumors and evaluated the association with tumor heterogeneity, stemness, chemokines chemokine receptors, and immunomodulators in pan-cancer. Besides, we knocked down SOWAHA in SW620 cells and performed RNA-seq analysis, then we conducted functional enrichment to uncover the biological significance of the gene set. SOWAHA has early diagnostic potential, and low expression of SOWAHA was associated with poor prognosis in was associated with poor prognosis in GBMLGG, PAAD, READ, etc. SOWAHA is associated with most tumor immune-infiltrating cells in pan-cancer. SOWAHA correlates with DNA methylation, tumor heterogeneity, and stemness in many epithelial carcinomas. Furthermore, SOWAHA is involved in many enzyme activity and metabolic pathways, mainly metabolic programming pathways in cancer. Additionally, we identified two potential transcription factors of SOWAHA, TBX4, and FOXP2, which are dysregulated in SW620 cells. Abstract

The all-digital massive multiple-input multiple-output (MIMO) system has a wide range of application prospects in mobile communication due to its advantages such as low cost, strong signal processing capability, and flexible configuration. On the basis of studying the one-bit sigma-delta modulation structure, the ergodic achievable spectral efficiency of the system was analyzed and compared with the one-bit structure through simulation. The results show that the one-bit sigma-delta modulation structure has higher spectral efficiency, is suitable for massive MIMO systems, and the analysis results are accurate.

Abstract

Rheumatoid arthritis (RA) is an idiopathic and chronic autoimmune disease for which there are currently no effective treatments. Oxypeucedanin hydrate (OXH) is a natural coumarin known for its potent anti-inflammatory properties. However, further investigations are needed to determine its therapeutic efficacy in treating RA. In this study, we evaluate the anti-inflammatory activity of OXH by treating LPS-induced RAW264.7 macrophages. Our results show that OXH treatment reverses the changes in iNOS, COX-2, IL-1β, IL-6, and TNF-α levels. Additionally, OXH reduces ROS production. Further analysis reveals that OXH suppresses the activation of the NF-κB/MAPK pathway. CETSA results show that OXH competes with LPS for binding to the TLR4/MD2 complex. MST experiments demonstrate the specific affinity of OXH for the TLR4/MD2 complex, with a Kd value of 33.7 μM. Molecular docking analysis suggests that OXH binds to the pocket of the TLR4/MD2 complex and interacts with specific amino acids, such as GLY-343, LYS-388, and PHE-345. Molecular dynamics simulations further confirm this conclusion. Finally, we investigate the potential of OXH in treating RA using a collagen-induced arthritis (CIA) model in rats. OXH effectively ameliorates the symptoms of CIA, including improving body weight, reducing swelling and redness, increasing talus volume, and decreasing bone erosion. OXH also decreases the mRNA levels of pro-inflammatory factors in synovial tissue. Transcriptome enrichment analysis and western blot analysis confirm that OXH suppresses the NF-κB/MAPK pathway, which is consistent with our in vitro findings.

Abstract

Objective To compare the effect of Traditional Chinese Medicine (TCM) hot compress and acupoint application on prevention of abdominal distension and constipation after multiple-level spinal fixation surgery. Methods This study was a prospective, randomized controlled clinical trial conducted at a single institution. From August 2020 to November 2021, patients who met the inclusion criteria were allocated to TCM hot compress group and acupoint application group (each for 30 patients). Another 30 more patients without intervention were recruited as control group. Primary outcomes included the time to first flatus and time to first defecation. Secondary outcomes included 3-day postoperative abdominal girth, 3-day postoperative abdominal distension and patient¡¯s satisfaction. Results Time to first flatus was (20. 80+-13. 00) h in TCM hot compress group, (21. 70+-13. 70)h in acupoint application group, both of which was lower than (38. 00+-14. 60)h of control group (P£¼0. 01; P£¼0. 01). Time to first defecation was (69. 10+-24. 90)h in TCM hot compress group, lower than(82. 80+-26. 50)h in acupoint application group (P£¼0. 05), both of which was lower than(100. 80+-28. 20)h in control group(P£¼0. 01; P£¼0. 05). The 3-day postoperative bloating score was(0. 23+-0. 43)in TCM hot compress group, lower than (0. 5+-0. 73) in acupoint application group (P£¼0. 05), both of them lower than (1. 07+-0. 87) in control group (P£¼0. 01; P£¼0. 01). The satisfaction rate was 100%(30/30) in Chinese medicine hot compress group, higher than 90. 00%(27/30) in acupoint application group and73. 33%(22/30) in control group (P£¼0. 01; P£¼0. 01). Conclusion Both TCM hot compress and acupoint application are effective in preventing abdominal distension and constipation after multiple-level spinal fixation surgery. TCM hot compress therapy shows a better therapeutic effect in aspects of shorter time to first defecation and relief of abdominal distension.
