Abstract Background Foot-and-mouth disease (FMD) is a devastating disease affecting cloven-hoofed animals, that leads to significant economic losses in affected countries and regions. Currently, there is an evident inclination towards the utilization of nanoparticles as powerful platforms for innovative vaccine development. Therefore, this study developed a ferritin-based nanoparticle (FNP) vaccine that displays a neutralizing epitope of foot-and-mouth disease virus (FMDV) VP1 (aa 140–158) on the surface of FNP, and evaluated the immunogenicity and protective efficacy of these FNPs in mouse and guinea pig models to provide a strategy for developing potential FMD vaccines. Results This study expressed the recombinant proteins Hpf, HPF-NE and HPF-T34E via an E. coli expression system. The results showed that the recombinant proteins Hpf, Hpf-NE and Hpf-T34E could be effectively assembled into nanoparticles. Subsequently, we evaluated the immunogenicity of the Hpf, Hpf-NE and Hpf-T34E proteins in mice, as well as the immunogenicity and protectiveness of the Hpf-T34E protein in guinea pigs. The results of the mouse experiment showed that the immune efficacy in the Hpf-T34E group was greater than the Hpf-NE group. The results from guinea pigs immunized with Hpf-T34E showed that the immune efficacy was largely consistent with the immunogenicity of the FMD inactivated vaccine (IV) and could confer partial protection against FMDV challenge in guinea pigs. Conclusions The Hpf-T34E nanoparticles stand out as a superior choice for a subunit vaccine candidate against FMD, offering effective protection in FMDV-infected model animals. FNP-based vaccines exhibit excellent safety and immunogenicity, thus representing a promising strategy for the continued development of highly efficient and safe FMD vaccines.

When False Data Injection (FDI) attacks inject false data into the controllers of a microgrid, it can lead the controllers to make decisions based on inaccurate information, ultimately resulting in system collapse. This study presents a novel approach to dynamically adjust output voltage in Campus Microgrids (CMG). Initially, we use feedback linearization techniques to address the inherent nonlinear complexities in Distributed Generation (DG) dynamics. Moreover, during voltage regulation, controllers and sensors may be susceptible to FDI attacks. To address this, an elastic follower observer is introduced to mitigate the impact of false information injection attacks on voltage coordination, thus ensuring the robustness and stability of the system. Notably, our method reduces dependence on global information and fault boundaries within the communication network, thereby enhancing system elasticity and efficiency. In conclusion, through comprehensive simulation experiments, we evaluate the effectiveness and performance of our proposed strategy. The simulation results validate the success of our approach in voltage regulation and its efficiency in fault management. These findings bear significant implications for the advancement of voltage regulation strategies in distributed generation systems.

African swine fever (ASF) is an infectious disease characterized by hemorrhagic fever, which is highly pathogenic and causes severe mortality in domestic pigs. It is caused by the African swine fever virus (ASFV). ASFV is a large DNA virus and primarily infects porcine monocyte macrophages. The interaction between ASFV and host macrophages is the major reason for gross pathological lesions caused by ASFV. Necroptosis is an inflammatory programmed cell death and plays an important immune role during virus infection. However, whether and how ASFV induces macrophage necroptosis and the effect of necroptosis signaling on host immunity and ASFV infection remains unknown. This study uncovered that ASFV infection activates the necroptosis signaling in vivo and macrophage necroptosis in vitro. Further evidence showed that ASFV infection upregulates the expression of ZBP1 and RIPK3 to consist of the ZBP1-RIPK3-MLKL necrosome and further activates macrophage necroptosis. Subsequently, multiple Z-DNA sequences were predicted to be present in the ASFV genome. The Z-DNA signals were further confirmed to be present and colocalized with ZBP1 in the cytoplasm and nucleus of ASFV-infected cells. Moreover, ZBP1-mediated macrophage necroptosis provoked the extracellular release of proinflammatory cytokines, including TNF-α and IL-1β induced by ASFV infection. Finally, we demonstrated that ZBP1-mediated necroptosis signaling inhibits ASFV replication in host macrophages. Our findings uncovered a novel mechanism by which ASFV induces macrophage necroptosis by facilitating Z-DNA accumulation and ZBP1 necrosome assembly, providing significant insights into the pathogenesis of ASFV infection.

Abdominal complications following cardiac surgery are rare events, but can be fatal if not diagnosed and managed in time. Aggressive treatments offer a better chance for survival in patients with risk factors. This case report describes the clinical presentation and details of 3 recently operated patients who suffered acute abdominal pain after cardiac surgery. The underlying causes were Crohn disease-associated ileal perforation, intestinal necrosis, and diverticulitis-associated colonic perforation. The presentation and management of these severe conditions are discussed.

In seasonally frozen soil areas, high-steep rocky slopes resulting from open-pit mining and slope cutting during road construction undergo slow natural restoration, making ecological restoration generally challenging. In order to improve the problems of external soil attachment and long-term vegetation growth in the ecological restoration of high-steep rocky slopes in seasonally frozen areas, this study conducted a series of experiments through the combined application of polyacrylamide (PAM) and carboxymethyl cellulose (CMC) to assess the effects of soil amendments on soil shear strength, water stability, freeze–thaw resistance, erosion resistance, and vegetation growth. This study showed that the addition of PAM-CMC significantly increased the shear resistance and cohesion of the soil, as well as improving the water stability, freeze–thaw resistance, and erosion resistance, but the internal friction angle of the soil was not significantly increased after reaching a certain content. Moderate amounts of PAM-CMC can extend the survival of vegetation, but overuse may cause soil hardening and inhibit vegetation growth by limiting air permeability. It was observed by a scanning electron microscope (SEM) that the gel membrane formed by PAM-CMC helped to “bridge” and bind the soil particles. After discussion and analysis, the optimum application rate of PAM-CMC was 3%, which not only improved the soil structure but also ensured the growth of vegetation in the later stage under the optimum application rate. Field application studies have shown that 3% PAM-CMC-amended soil stably attaches to high-steep rocky slopes, with stable vegetation growth, and continues to grow after five months of freeze–thaw action, with no need for manual maintenance after one year.

In city-integrated energy systems containing electric–thermal multi-energy sources, the uncertainty of renewable energy sources and the fluctuation of loads challenge the safe, efficient, economic and stable operation of the integrated energy systems. This paper introduces a novel approach for the operation of a carbon capture plant/CHP with PV accommodation within a city-integrated energy system. The proposed strategy aims to maximize the utilization of photovoltaic (PV) power generation and carbon capture equipment, addressing issues related to small-scale CHP climbing constraints and short-term output regulation. Additionally, this paper presents a multi-timescale optimal scheduling strategy, which effectively addresses deviations caused by PV fluctuations and load changes. This was achieved through a detailed analysis of the CHP climbing constraints, carbon capture equipment operation and real-time characteristics of PV power generation. This paper introduces a fully distributed neural dynamics-based optimization algorithm designed to address multi-timescale optimization challenges. Utilizing rolling cycles, this algorithm computes both day-ahead and real-time scheduling outcomes for urban integrated energy systems. Theoretical analyses and numerical simulations were conducted to validate the precision and efficacy of the proposed model and algorithm. These analyses quantitatively evaluate the scheduling performance of PV power generation and carbon capture CHP systems across various timescales.

Objective Nowadays, with the increasing proportion of osteoporosis in patients with lumbar degenerative diseases, doctors are facing the choice of intraoperative internal fixation methods. The purpose of this study was to compare and assess the clinical results of posterior bilateral pedicle screw fixation and lateral fixation in the extreme lateral interbody fusion (XLIF) in patients with osteopenia or osteoporosis. Methods The retrospective review was performed on 67 degenerative lumbar diseases patients with osteopenia or osteoporosis who underwent XLIF in our hospital from January 2018 to July 2021. Patients in this study were classified into lateral screw (LS) group, lateral self‐locking plate (LP) group, and bilateral pedicle screw (BPS) group. The functional evaluation factors containing Japanese Orthopaedic Association (JOA) score, visual analogue scale (VAS) of leg pain, and VAS of low back pain, radiological factors such as disc height (DH), lumbar lordotic (LL) angle, segmental lordotic (SL) angle, cage subsidence degree and interbody fusion degree were compared. Results Primary outcomes: no differences were observed with regards to the incidence of complications among LS, LP and BS group (P 0.05). When comparing VAS of leg pain and JOA scores, no differences were identified among LS, LP and BPS groups (P > 0.05). There are four thigh sensory complaint, one hip flexor weakness and one thigh pain occurred and no death was observed. Abstract Objective To assess and compare the pathological and radiological outcomes of multifidus degeneration in scoliosis and lumbar disc herniation patients. Methods We performed a retrospective review on 24 patients with scoliosis and 26 patients with lumbar disc herniation (LDH) in the Third Hospital of Hebei Medical University from January 2017 to March2021. The patients were divided into scoliosis group and LDH group according to the treatment. The MRI fatty infiltration rate (FIR) of multifidus and strength of back muscle were calculated to evaluate muscle condition. Multifidus biopsy samples were obtained during surgery in the affected side at L4 or L5 segment in LDH group and on the concavity side of apical vertebrae in scoliosis group. The biopsy fatty infiltration degree (FID) and FIR in two groups, the FIR of affected and unaffected side in LDH group, and the FIR of concavity and convexity side in scoliosis group were compared. The correlation between concavity-convexity FIR difference and cobb angle in scoliosis group, back muscle strength and FIR in LDH group, FID and FIR in both groups was calculated respectively. Results The FIR was higher in scoliosis group than in LDH group, higher in concavity side than convexity side in scoliosis group (both P 0.05). ABSTRACT African swine fever virus (ASFV) causes acute hemorrhagic infectious disease in pigs. The ASFV genome encodes various proteins that enable the virus to escape innate immunity; however, the underlying mechanisms are poorly understood. The present study found that ASFV MGF-360-10L significantly inhibits interferon (IFN)-β-triggered STAT1/2 promoter activation and the production of downstream IFN-stimulated genes (ISGs). ASFV MGF-360-10L deletion (ASFV-Δ10L) replication was impaired compared with the parental ASFV CN/GS/2018 strain, and more ISGs were induced by the ASFV-Δ10L in porcine alveolar macrophages in vitro. We found that MGF-360-10L mainly targets JAK1 and mediates its degradation in a dose-dependent manner. Meanwhile, MGF-360-10L also mediates the K48-linked ubiquitination of JAK1 at lysine residues 245 and 269 by recruiting the E3 ubiquitin ligase HERC5 (HECT and RLD domain-containing E3 ubiquitin protein ligase 5). The virulence of ASFV-Δ10L was significantly lower than that of the parental strain in vivo, which indicates that MGF-360-10L is a novel virulence factor of ASFV. Our findings elaborate the novel mechanism of MGF-360-10L on the STAT1/2 signaling pathway, expanding our understanding of the inhibition of host innate immunity by ASFV-encoded proteins and providing novel insights that could contribute to the development of African swine fever vaccines. IMPORTANCE African swine fever outbreaks remain a concern in some areas. There is no effective drug or commercial vaccine to prevent African swine fever virus (ASFV) infection. In the present study, we found that overexpression of MGF-360-10L strongly inhibited the interferon (IFN)-β-induced STAT1/2 signaling pathway and the production of IFN-stimulated genes (ISGs). Furthermore, we demonstrated that MGF-360-10L mediates the degradation and K48-linked ubiquitination of JAK1 by recruiting the E3 ubiquitin ligase HERC5. ABSTRACT Senecavirus A (SVA) is a type of nonenveloped single-stranded, positive-sense RNA virus. The VP2 protein is a structural protein that plays an important role in inducing early and late immune responses of the host. However, its antigenic epitopes have not been fully elucidated. Therefore, defining the B epitopes of the VP2 protein is of great importance to revealing its antigenic characterization. In this study, we analyzed B-cell immunodominant epitopes (IDEs) of the VP2 protein from the SVA strain CH/FJ/2017 using the Pepscan approach and a bioinformatics-based computational prediction method. The following four novel IDEs of VP2 were identified: IDE1, 41TKSDPPSSSTDQPTTT56; IDE2, 145PDGKAKSLQELNEEQW160; IDE3, 161VEMSDDYRTGKNMPF175; and IDE4, 267PYFNGLRNRFTTGT280. Most of the IDEs were highly conserved among the different strains. To our knowledge, the VP2 protein is a major protective antigen of SVA that can induce neutralizing antibodies in animals. Here, we analyzed the immunogenicity and neutralization activity of four IDEs of VP2. Consequently, all four IDEs showed good immunogenicity that could elicit specific antibodies in guinea pigs. A neutralization test in vitro showed that the peptide-specific guinea pig antisera of IDE2 could neutralize SVA strain CH/FJ/2017, and IDE2 was identified as a novel potential neutralizing linear epitope. This is the first time VP2 IDEs have been identified by using the Pepscan method and a bioinformatics-based computational prediction method. These results will help elucidate the antigenic epitopes of VP2 and clarify the basis for immune responses against SVA. IMPORTANCE The clinical symptoms and lesions caused by SVA are indistinguishable from those of other vesicular diseases in pigs. SVA has been associated with recent outbreaks of vesicular disease and epidemic transient neonatal losses in several swine-producing countries. Due to the continuing spread of SVA and the lack of commercial vaccines, the development of improved control strategies is urgently needed. The VP2 protein is a crucial antigen on the capsids of SVA particles. Furthermore, the latest research showed that VP2 could be a promising candidate for the development of novel vaccines and diagnostic tools. Hence, a detailed exploration of epitopes in the VP2 protein is necessary. In this study, four novel B-cell IDEs were identified using two different antisera with two different methods. IDE2 was identified as a new neutralizing linear epitope. Our findings will help in the rational design of epitope vaccines and further understanding of the antigenic structure of VP2.

Objectives To compare the clinical efficacy of a new retractor‐assisted Wiltse transforaminal lumbar interbody fusion (TLIF), minimally invasive TLIF (MIS‐TLIF), and traditional posterior lumbar interbody fusion (PLIF) in treating single‐level lumbar degenerative diseases. Methods A retrospective study was conducted by analyzing the clinical and imaging data of consecutive patients with single‐level lumbar degenerative diseases who underwent the new retractor‐assisted Wiltse TLIF, MIS‐TLIF, or traditional PLIF. This study enrolled 87 concurrent patients between June 2016 and December 2019 (Wiltse TLIF 29 cases; MIS‐TLIF 28 cases; PLIF 30 cases). Abstract. Both empyema and type A aortic dissection are life-threatening conditions. The combination, though rare, can cause serious complications and death. There have been rare cases of empyema caused by a co-infection with a Gram-positive bacterium, Granulicatella adiacens, which is difficult to identify, and a Gram-negative bacterium, Escherichia coli. In this case, a 50-year-old man was referred to the emergency department due to sudden chest pain and a suspected diagnosis of type A aortic dissection. The patient was subsequently found to have co-morbid empyema and a fissure in the esophagus when the cause of the disease was investigated. Considering the possibility that the patient's infection was of digestive origin, a combination of Gram-positive and Gram-negative antibiotics was given immediately. Etiological examination later confirmed the presence of Granulicatella adiacens and Escherichia coli in chest drainage fluid, and the patient was successfully treated with antimicrobial therapy and conservative treatment.

Objective To explore the precautions of pre-transfusion examination in patients with antibodies to erythrocyte protective solution, discrepant ABO blood typing results, and positive unexpected antibodies, so as to ensure the safety of blood transfusion. Methods The screen cells were divided into two groups according to the presence or absence of washing reagent red blood cells in normal saline. One group had untreated forward typing cells, antibody screening cells and identification panel, and the other group had saline-washed reverse typing cells, antibody screening cells and identification panel. The experiments were carried out by microcolumn gel method, saline medium method and polyamine method to analyze the effect of red blood cell preservation solution on serum agglutination reaction of specific patients. Results Among the 8 patients, forward typing was AB (+ ) in 1 patient, B (+ ) in 4, and A(+ ) in 3, and the reverse typing were interfered. The plasma of 8 patients agglutinated with unwashed reverse typing cells (saline tube method), screen cells and identification panels (saline tube method plus cassette method), while not agglutinated with the polybrene method. The interference was eliminated as using washed reverse typing cells (salinetube method), screen cells and identification panels (saline tube method plus cassette method). Conclusion The erythrocyte preservation solution affected patients’ blood group typing, but not affected the outcome of blood transfusion as no adverse reactions occurred.

At present, the global fishery catch is approaching the peak situation, and the development of aquaculture is facing huge environmental pressure. Organic aquaculture can not only meet the needs of consumers for high-quality protein, but also effectively alleviate the ecological problems and ensure the quality and safety of food. Organic aquaculture has become the consensus of all countries in the world. To a certain extent, fishery developed countries have formed a relatively perfect organic aquaculture system. Based on the current situation and trend of global aquaculture development, this paper studied the relevant experience of green aquaculture in European Union, the United States and Japan, and finally put forward relevant policy suggestions combined with the development status of aquaculture in China.

Collapses are one of the most common geological disasters in mountainous areas, which easily damage buildings and infrastructures and bring huge property losses to people’s production and life. This paper uses Huinan County as the study area, and with the help of a geographic information system (GIS) based on the formation principle of natural disaster risk, the information content method (ICM), the analytical hierarchy process (AHP), and the analytical hierarchy process–information content method (AHP-ICM) model are applied to hazard mapping, and the analytical hierarchy process-entropy weight method (AHP-EWM) model is applied to exposure, vulnerability and emergency responses, and recovery capability mapping. A risk mapping model for collapse disasters was also constructed using these four elements. Firstly, an inventory map of 52 landslides was compiled using remote sensing interpretation, field verification, and comprehensive previous survey data. Then, the study area mapping units were delineated using the curvature watershed method in the slope unit, and 21 indicators were used to draw the collapse disaster risk zoning map by considering the four elements of geological disaster risk. The prediction accuracy of the three hazard mapping models was verified using the receiver operating characteristic (ROC) curve, and the area under the curve (AUC) results of the AHP, ICM, and AHP-ICM models were 80%, 85.7%, and 87.4%, respectively. After a comprehensive comparison, the AHP-ICM model is the best of the three models in terms of collapse hazard mapping, and it was applied to collapse risk mapping with the AHP-EWM model to produce a reasonable and reliable collapse risk zoning map, which provides a basis for collapse management and decision making.

Objective To assess and compare the therapeutic effects of Anterior Cervical Discectomy and Fusion (ACDF) and Cervical Laminectomy and Fusion (CLF) in the treatment of 4‐level cervical. Methods We performed a retrospective review on 39 patients with 4‐level CSM who underwent ACDF or CLF in the Third Hospital of Hebei Medical University from January 2010 to December 2018. The patients were divided into ACDF group and CLF group according to the treatment. The operative index was evaluated based on intraoperative blood loss and operation time. The functional outcomes including Japanese Orthopedic Association (JOA) score and visual analogue scale (VAS) of axial pain were compared. The Cobb angle, Cobb angle improvement rate, range of motion (ROM) and ROM loss ratio were measured for radiographic evaluation. Results No major complications or deaths occurred. The average age at baseline was 55 years. There was no significant difference between the ACDF and CLF group in follow‐up time (26.29 months, 25.39 months, P > 0.05). The frequent occurrence of urban flooding in recent years has resulted in significant damage to ground-level infrastructure and poses a substantial threat to the metro system. As the central city’s core transportation network for public transit, this threat can have unpredictable consequences on travel convenience and public safety. Therefore, assessing the risk of urban flooding in the metro system is of utmost importance. This study is the first of its kind to employ comprehensive natural disaster risk assessment theory, establishing an assessment database with 22 indicators. We propose a GIS-based method combined with the analytical hierarchy process (AHP) and an improved entropy weight method to comprehensively evaluate the urban flood risk in Changchun City’s metro systems in China. This study includes a total of nine metro lines, including those that are currently operational as well as those that are in the planning and construction phases, situated in six urban areas of Changchun City. In this study, we utilize the regional risk level within the 500 m buffer zone of the metro lines to represent the flood risk of the metro system. The proposed method assesses the flood risk of Changchun’s rail transit system. The results reveal that over 30% of Changchun’s metro lines are located in high-risk flood areas, mainly concentrated in the densely populated and economically prosperous western part of the central city. To validate the risk assessment, we vectorized the inundation points and overlaid them with the regional flood risk assessment results, achieving a model accuracy of over 90%. As no large-scale flood events have occurred in the Changchun rail transit system, we employed receiver operating characteristic (ROC) curves to verify the accuracy of the flood risk assessment model, resulting in an accuracy rate of 91%. These findings indicate that the present study is highly reliable and can provide decision makers with a scientific basis for mitigating future flood disasters.

For the oil and gas pipeline operating in the permafrost regions, buckling deformation is usually caused by frost heaving and thaw settlement of the foundation soil, which affects the safety and stability of pipeline seriously. With 4 typical types of pipeline foundation soil along China–Russia Eastern Gas Pipeline located in the permafrost region of Greater Khingan Range as the object of study, the relationship of the coefficient of thaw settlement and frost heaving ratio with other influencing factors of various soil samples was analyzed through the frost heaving test and thaw settlement test, and the sensitivity of the influencing factors of the coefficient of thaw settlement and frost heaving ratio was also analyzed with gray correlation method. The results indicate that the coefficient of thaw settlement and frost-heaving ratio of the soil samples are greatly influenced by water content and dry density. In addition, superplastic water content, mud content and loss on ignition also have significant effect on the silty soil, sandy soil and peat soil. According to the grey correlation analysis, water content is the main influencing factor of frost heaving and thaw settlement, but it is not absolute. For some soil, the mud content and superplastic water content may play a major role in frost heaving and thaw settlement, to which great attention shall be paid during design and construction. The study results can provide scientific support and theoretical reference to the design, construction, operation, maintenance and prevention and control of frost damage of buried pipelines in the permafrost regions.

African swine fever (ASF) has brought excellent barriers to swine production in China and the world. Studies have shown that extracellular vesicles mediate the RNA and protein spread of pathogenic microorganisms and RNA and proteins. After infection by pathogenic microorganisms causes significant differences in the proteins contained within extracellular vesicles. Based on the above studies, the extracellular vesicles were extracted from ASF virus (ASFV)-infected swine plasma. And qPCR, western blot, and confocal experiment were carried out. The research shows that extracted extracellular vesicles significantly promote the replication of ASFV in susceptible and non-susceptible cells Proteomics analysis of the extracellular vesicle proteins revealed that ASFV infection could cause significant differences in the protein profile. This study demonstrates that extracellular vesicles play a critical role in ASFV replication and transmission and cause significant differences in the protein profile encapsulated in extracellular vesicles.

Energy management of virtual power plants (VPPs) directly affects operators’ operating profits and is also related to users’ comfort and economy. In order to provide a reasonable scheme for scheduling each unit of the VPP and to improve the operating profits of the VPP, this study focuses on the optimization of VPP energy management under the premise of ensuring the comfort of flexible load users. First, flexible loads are divided into time-shiftable load (TL), power-variable load (PL), and interruptible load (IL), and their accurate power consumption models are established, respectively. Then, aiming at maximizing the operation profits of a VPP operator, an optimization model of VPP energy management considering user comfort is proposed. Finally, the improved cooperative particle swarm optimization (ICPSO) algorithm is applied to solve the proposed VPP energy management optimization model, and the optimal scheduling scheme of VPP energy management is obtained. Taking a VPP in the coastal area of China as an example, results show that the optimization model proposed in this article has the advantages of good economy and higher user comfort. Meanwhile, the ICPSO algorithm has the characteristics of faster optimization speed and higher accuracy when solving the problem with multiple variables.

ABSTRACT African swine fever (ASF)—an aggressive infectious disease caused by the African swine fever virus (ASFV)—is significantly unfavorable for swine production. ASFV has a complex structure and encodes 150–167 proteins; however, the function of most of these proteins is unknown. This study identified ASFV MGF360-9L as a negative regulator of the interferon (IFN)-β signal. Further evidence showed that MGF360-9L interacts with signal transducer and activator of transcription (STAT) 1 and STAT2 and degrades STAT1 and STAT2 through apoptosis and ubiquitin–proteasome pathways, respectively. Subsequently, the activation of IFN-β signaling was inhibited. Naturally isolated or genetically manipulated live attenuated viruses are known to protect against the virulent parental ASFV strains. Therefore, through homologous recombination, we deleted MGF360-9L from the virulent ASFV CN/GS/2018 strain to construct a recombinant strain, ASFV-Δ360-9L. Compared with the parent ASFV CN/GS/2018 strain, the replication level of ASFV-Δ360-9L decreased in primary porcine alveolar macrophage cultures at 24 h postinfection, but the difference is unlikely to be biologically relevant. Notably, ASFV-Δ360-9L was partially attenuated in pigs. To our knowledge, this study is the first to uncover the function of MGF360-9L during ASFV infection. MGF360-9L inhibits IFN-β signaling through the targeted degradation of STAT1 and STAT2. Furthermore, MGF360-9L is a key virulence gene of ASFV. Our findings reveal a new mechanism by which ASFV inhibits host antiviral response; this might facilitate the development of live attenuated ASFV vaccines. IMPORTANCE African swine fever—an acute, febrile, hemorrhagic, highly contacting, and highly lethal disease caused by African swine fever virus (ASFV)—jeopardizes the global pig industry. Understanding the mechanism ASFV employs to evade host defense during infection is essential for developing targeted drugs and vaccines against ASFV. To our knowledge, this study identifies the mechanism of innate immunity against by MGF360-9L and the effect of MGF360-9L on ASFV pathogenicity. The results showed that MGF360-9L may help ASFV escape the host immunity by degrading STAT1 and STAT2 and thus inhibiting IFN-β signaling. MGF360-9L is also an important virulence factor of ASFV. The deletion of MGF360-9L reduces ASFV virulence in pigs. This study explored a new mechanism of ASFV against innate immunity and identified a new ASFV virulence factor; these findings may guide the development of live attenuated ASFV vaccines.

Abstract Paravalvular leakage (PVL) after mitral valve replacement is a troublesome complication that may lead to severe symptoms and reoperation. Previous case reports on total thoracoscopic cardiac surgery without aortic cross-clamping for repairing late PVL are rare. We describe a 64-year-old man who had undergone aortic and mitral valve replacement via median sternotomy eight years earlier, and who recently developed cardiac failure due to severe tricuspid regurgitation (TR) and PVL in the posterior mitral annulus. During total thoracoscopic surgery with using the beating heart technique, direct closure of the PVL was achieved via pledgeted mattress sutures, and tricuspid valvuloplasty was routinely performed to treat TR. This case indicated that total thoracoscopic surgery on a beating heart may be an excellent option for treating PVL concomitant with TR.

Parameter identification of the proton exchange membrane fuel cell (PEMFC) is a good way of increasing their efficiency in the next designs. In this study, an optimized improved Elman neural network based on a new hybrid optimization algorithm is proposed for this purpose. The proposed algorithm is a hybrid algorithm based on a combination of two newly algorithms, the world cup optimization (WCO) and the fluid Search Optimization (FSO) algorithms. The proposed method is applied to improve the method efficiency for estimating the PEMFC model parameters. The method is then validated by four different operational conditions. The optimization algorithm efficiency is also analyzed by comparison with some popular algorithms. Simulation results showed that using the designed method gives higher accuracy forecast for the PEMFC model parameters. An improved variational inequality strategy for dealing with variational inequality in a Hilbert space is proposed in this article as an alternative; if Hilbert space is used as the domain of interest, the original extra-gradient method is proposed for resolving variational inequality. This improved variational inequality strategy can be used as a substitute for the original extra-gradient method in some situations. Mann’s mean value method, coupled with the widely used sub-gradient extra-gradient strategy, makes it possible to update all of the previous iterations in a single step, thus saving time and effort. All of this is made feasible via the use of Mann’s mean value technique in conjunction with the convex hull of all prior iterations of the algorithm. It is guaranteed that the mean value iteration will result in an acceptable resolution of a variational inequality issue as long as one or more of the criteria for the averaging matrix are fulfilled. Numerous experiments were performed in order to demonstrate the correctness of the theoretical conclusion obtained.

Clean groundwater resources are vital to human health. To evaluate groundwater quality in the Songnen Plain (Jilin), a field investigation sampling test, multivariate statistical analysis, and spatial analysis were conducted based on a geographic information system. The main substances exceeding the standard were screened out, and the main controlling factors affecting groundwater quality were discussed. The research result showed that nine components exceeded groundwater standards by approximately 10%: Al, total hardness (TH), total dissolved solids (TDS), Mn, As, NO3−, Fe, F−, and BaP. The over-standard of TDS and TH in groundwater are mainly distributed in the geological environment conditions and unreasonable exploitation and utilization of groundwater in this area. The results of the multi-index evaluation showed that the most important factors affecting groundwater quality were general chemical indices, followed by inorganic toxicology and heavy metals. Controlling the overexploitation of water resources, controlling agricultural activities and sewage discharge, and implementing water conservation systems are the main pathways to improve water quality in the study area. The research results can provide a reference for groundwater pollution control and water resource protection in the Songnen Plain (Jilin).

Senecavirus A (SVA) is a newly porcine virus that has been detected in many countries since its first detection in pigs in Canada in 2007, and it remains endemic in many countries in Asia and America, which has become a substantial problem for the pig industry. Vaccination is a potentially effective strategy for the prevention and control of SVA infection. Our lab has developed a SVA vaccine candidate previously. In this study, the antibody response to the prepared vaccine in sows and their offspring was evaluated. Vaccination of sows with inactivated SVA vaccines during pregnancy elicited SVA-specific virus-neutralizing antibodies. Vaccination with a high dose of SVA vaccine followed a booster immunization contributed to a long-term duration of the persistence of maternally derived neutralizing antibodies (MDAs) in the milk of the sows (>14 days). In contrast, vaccination with a single low dose of SVA vaccine resulted in a short-term persistence of MDAs in the milk (2–7 days). The MDAs could be efficiently transferred from the sows to their offspring through the colostrum/milk but not the umbilical cord blood. The antibody titers and the duration of the persistence of MDAs in the offspring are highly associated with the antibody levels in the milk from the sows. Vaccination of sows with a booster dose of SVA vaccine resulted in a longer-lasting MDAs in their offspring (persisted for at least 90 days). However, vaccination with the single low dose of vaccine only brought about 42 days of MDAs persistence in their offspring. The effect of MDAs on active immunization with SVA vaccine in offspring was further evaluated, which showed that vaccination of the SVA vaccine in the presence of MDAs at the titer of ≈1:64 or less could overcome the MDAs’ interference and give rise to effective antibody response. This will help for establishing the optimal times and schedules for SVA vaccination in pigs.

An integrated zircon geochronological, elemental geochemical, and Sr–Nd–Hf isotopic investigation was carried out on a suite of dioritic–granitic rocks at Zalute in the southern Great Xing’an Range (SGXR), NE China, in order to probe the source and petrogenesis of these granitoid rocks and further constrain the geodynamical setting of early Early Cretaceous magmatism. The results of Sensitive High-Resolution Ion Micro Probe (SHRIMP) zircon U–Pb dating reveal that the Zalute dioritic–granitic rocks have a consistent crystallization age of ca. 137–136 Ma, consisting of quartz diorite (136 ± 1.4 Ma), monzogranite (136 ± 0.8 Ma), and granite porphyry (137 ± 1.3 Ma), which record an early Early Cretaceous magmatic intrusion. Geochemically, the quartz diorites, monzogranites, and granite porphyries are mostly high-K calc-alkaline and show features of typical I-type affinity. They possess uniform and depleted Sr–Nd–Hf isotopic compositions (e.g., initial 87Sr/86Sr ratios of 0.7035 to 0.7049, εNd(t) of −0.02 to +2.61, and εHf(t) of +6.8 to +9.6), reflecting a common source, whose parental magma is best explained as resulting from the partial melting of juvenile source rocks in the lower crust produced by underplating of mantle-derived mafic magma, with minor involvement of ancient crustal components. Evidence from their close spatio–temporal relationship, common source, and the compositional trend is consistent with a magmatic differentiation model of the intermediate-felsic intrusive suite, with continued fractional crystallization from quartz diorites, towards monzogranites, then to granite porphyries. Combined with previously published data in the SGXR, our new results indicate that the Zalute intermediate-felsic intrusive suite was formed during the post-collisional extension related to the closure of the Mongol–Okhotsk Ocean and subsequent slab break-off.

With the rapid development of industry, more fossil energy is consumed to generate electricity, which increases carbon emissions and aggravates the burden of environmental protection. To reduce carbon emissions, traditional centralized power generation networks are transforming into distributed renewable generation systems. However, the deployment of distributed generation systems can affect power system economy and stability. In this paper, under different time scales, system economy, stability, carbon emissions, and renewable energy fluctuation are comprehensively considered to optimize battery and super-capacitor installation capacity for an off-grid power system. After that, based on the genetic algorithm, this paper shows the optimal system operation strategy under the condition of the theoretical best energy storage capacity. Finally, the theoretical best capacity is tested under different renewable energy volatility rates. The simulation results show that by properly sizing the storage system’s capacity, although the average daily costs of the system can increase by 10%, the system’s carbon emissions also reduce by 42%. Additionally, the system peak valley gap reduces by 23.3%, and the renewable energy output’s fluctuation range and system loss of load probability are successfully limited in an allowable range. Lastly, it has less influence on the theoretical best energy storage capacity if the renewable energy volatility rate can be limited to within 10%.

This paper experimentally analyzes the critical current degradation and AC (alternating current) losses of second-generation (2G) high-temperature superconductor (HTS) tape during the impregnation process. Two impregnation materials were utilized: Gallium-Indium-Tin (GaInSn), and an epoxy resin, Araldite. The critical current of the impregnation materials was measured after different thermal cycles and compared with the tape with no impregnation process. The experimental results show that the critical current of Yttrium Barium Copper Oxide (YBCO) short samples varies between differently impregnated materials. The resin, Araldite, degraded the critical current; however, the GaInSn showed no degradation. Two degradation patterns with Araldite were identified due to the impregnation process, and the corresponding causes were analyzed. We further measured the AC losses of tapes impregnated with liquid metal at different frequencies, up to 600 Hz. Based on the experimental results, GaInSn liquid metal should be the most suitable impregnation material in terms of critical current degradation.

This paper studies the critical current of second generation, high temperature superconducting coils under an external magnetic field experimentally and numerically. Two identical coils with different coated conductors are fabricated and tested under a direct current (DC) magnetic field along the axis of the coil. Then, a numerical model in cylindrical coordinates based on a sheet current model is built by taking the measured magnetic field dependency to analyze the current distribution and magnetic field distribution. The simulated critical currents of the coils under the DC magnetic field have good agreement with the measured results. We find that under the in-phase field, the critical current decreases as the magnetic field in the innermost turn is enhanced by the external field. Meanwhile, the anti-phase external field increases the critical current a bit at first, then decreases the critical current. We further discuss the critical current criteria of the coils, showing that the parallel field plays a more important role in critical current determination.

With the rapid growth of domestic electric vehicle charging loads, the peak-valley gap and power fluctuation rate of power systems increase sharply, which can lead to the increase of network losses and energy efficiency reduction. This paper tries to regulate network loads and reduce power system transmission loss by optimizing domestic electric vehicle charging loads. In this paper, a domestic electric vehicle charging loads model is first developed by analyzing the key factors that can affect users’ charging behavior. Subsequently, the Monte Carlo method is proposed to simulate the power consumption of a cluster of domestic electric vehicles. After that, an optimal electric vehicle charging strategy based on the 0-1 integer programming is presented to regulate network daily loads. Finally, by taking the IEEE33 distributed power system as an example, this paper tries to verify the efficacy of the proposed optimal charging strategy and the necessity for considering seasonal factors when scheduling electric vehicle charging loads. Simulation results show that the proposed 0-1 integer programming method does have good performance in reducing the network peak-valley gap, voltage fluctuation rate, and transmission loss. Moreover, it has some potential to further reduce power system transmission loss when seasonal factors are considered.

This paper presents a novel control algorithm for optimising operational costs of a combined domestic micro-CHP (combined heat and power), battery and heat storage system. Using a minute by minute basic time-step, this work proposes a simple and computationally efficient rule based whole-system management, developed from empirical study of realistic simulated domestic electricity and heat loads. The CHP availability is considered in two binary states which, together with leveraging storage effectively, maximises CHP efficiency, and gives the algorithm increased real world feasibility. In addition, a novel application of a dual battery system is proposed to support the micro-CHP with each battery supplying just one of the distinctive morning and evening electrical load peaks, and thus inherently improving overall battery system lifetime. A case study is presented where the algorithm is shown to yield approximately 23% energy cost savings above the base case, almost 3% higher savings than that of the closest previous work, and 96.8% of the theoretical minimum cost. In general, the algorithm is shown to always yield better than 88% of the theoretical minimum cost, a ratio that will be considerably higher when real-world CHP limitations are factored into the theoretical minimum calculation.

Complex power electronic conversion devices, most of which have high transmission performance, are important power conversion units in modern aircraft power systems. However, these devices can also affect the stability of the aircraft power system more and more prominent due to their dynamic and nonlinear characteristics. To analyze the stability of aircraft power systems in a simple, accurate and comprehensive way, this paper develops a unified large signal model of aircraft power systems. In this paper, first the Lyapunov linearization method and the mixed potential theory are employed to analyze small signal and large signal stability, respectively, and then a unified stability criterion is proposed to estimate small and large signal stability problems. Simulation results show that the unified large signal model of aircraft power systems presented in this paper can be used to analyze the stability problem of aircraft power systems in an accurate and comprehensive way. Furthermore, with simplicity, universality and structural uniformity, the unified large signal model lays a good foundation for the optimal design of aircraft power systems.

With the rapid growth of thermostatically controlled loads, active power fluctuation and peak demand growth within an autonomous micro-grid become serious problems. This paper tries to suppress power fluctuation and shave peak demand for a micro-grid through optimizing domestic electric water heaters (controllable load). In this paper, domestic electric water heater models are first built to optimize power flow within a single water heater. Subsequently, the Monte Carlo method is proposed to simulate power consumption of a cluster of domestic electric water heaters. After that, the temperature state priority list method is presented to suppress power flow and shave peak demand for a given micro-grid. Optimization results show that the proposed temperature state priority list method can reduce peak demand by 12.5%. However, it has a wider active power fluctuation range and needs a longer reaction time compared with the simplified temperature state priority list method. In addition, the optimization results show that by increasing the number of controllable loads participating in load scheduling, active power fluctuation can be reduced and the maximum active power of the given micro-grid can be cut. However, to achieve this, about 1.2% of extra electrical energy needs to be generated by the external grid.

The average daily benefit to cost ratio of a building energy storage system is mainly constrained by the battery lifetime. This paper aims to minimize the average daily cost of a hybrid energy storage system (HESS) (comprised of a battery and supercapacitor) by optimizing the battery capacity. A novel optimization model is proposed with the objective to find the minimum average daily investment cost of the HESS. The objective function has two parts: (1) the investment cost formula for the battery is derived as a function of the battery capacity, which has an interdependence with the minimum state of charge (SOC) and the maximum discharge current; (2) the investment cost formula for the supercapacitor is also established as a function of battery capacity by matching the maximum battery power with that of the supercapacitor. Case studies demonstrate several ways to increase the average daily benefit to cost ratio: (1) adopting a suitable control strategy to avoid capacity saturation; (2) reducing the battery SOC to increase the threshold for the maximum discharge current (MDC) saturation; and (3) increasing MDC to raise the threshold for the SOC saturation. Results show that the average daily benefit to cost ratio is doubled compared to previous work.

African swine fever, one of the major viral diseases of swine, poses an imminent threat to the global pig industry. The high-efficient replication of the causative agent .African swine fever virus (ASFV) in various organs in pigs greatly contributes to the disease. However, how ASFV manipulates the cell population to drive high-efficient replication of the virus in vivo remains unclear. Here, we found that the spleen reveals the most severe pathological manifestation with the highest viral loads among various organs in pigs during ASFV infection. By using single-cell-RNA-sequencing technology and multiple methods, we determined that macrophages and monocytes are the major cell types infected by ASFV in the spleen, showing high viral-load heterogeneity. A rare subpopulation of immature monocytes represents the major population infected at late infection stage. ASFV causes massive death of macrophages, but shifts its infection into these monocytes which significantly arise after the infection. The apoptosis, interferon response, and antigen-presentation capacity are inhibited in these monocytes which benefits prolonged infection of ASFV in vivo. Until now, the role of immature monocytes as an important target by ASFV has been overlooked due to that they do not express classical monocyte marker CD14. The present study indicates that the shift of viral infection from macrophages to the immature monocytes is critical for maintaining prolonged ASFV infection in vivo. This study sheds light on ASFV tropism, replica-tion, and infection dynamics, and elicited immune response, which may instruct future research on antiviral strategies. [ABSTRACT FROM AUTHOR]
