Tibet orbivirus (TIBOV), a new candidate of Orbivirus genus, was initially isolated from mosquitoes in Tibet in 2009 and subsequently from both Culicoides and mosquitoes in several provinces of China and Japan. Little is known about the origin, genetic diversity, dissemination and pathogenicity of TIBOV, although its potential threat to animal health has been acknowledged. In this study, two viruses, V290/YNSZ and V298/YNJH, were isolated from the Culicoides and sentinel cattle in Yunnan Province. Their genome sequences, cell tropism in mammalian and insect cell lines along with pathogenicity in suckling mice were determined. Genome phylogenetic analyses confirmed their classification as TIBOV species; however, OC1 proteins of the V290/YNSZ and V298/YNJH shared maximum sequence identities of 31.5% and 33.9% with other recognized TIBOV serotypes (TIBOV-1 to TIBOV-4) and formed two monophyletic branches in phylogenetic tree, indicating they represented two novel TIBOV serotypes which were tentatively designated as TIBOV-5 and TIBOV-6. The viruses replicated robustly in BHK, Vero and C6/36 cells and triggered overt clinical symptoms in suckling mice after intracerebral inoculation, causing mortality of 100% and 25%. Cross-sectional epidemiology analysis revealed silent circulation of TIBOV in Yunnan Province with overall prevalence of 16.4% (18/110) in cattle, 10.8% (13/120) in goats and 5.5% (6/110) in swine. The prevalence patterns of four investigated TIBOV serotypes (TIBOV-1, -2, -5 and 6) differed from each one another, with their positive rates ranging from 8.2% (9/110) for TIBOV-2 in cattle to 0.9% (1/110) for TIBOV-1 and TIBOV-5 in cattle and swine. Our findings provided new insights for diversity, pathogenicity and epidemiology of TIBOV and formed a basis for future studies addressing the geographical distribution and the zoonotic potential of TIBOV.

Plumbago zeylanica L. (PZ) is a folk medicine for liver damage. To further explore its mechanism of action, a rat model of liver injury induced by carbon tetrachloride (CCl4) was used to evaluate the protective effect of PZ water extract (PZE). The results showed that PZE normalized the CCl4-induced serum levels of liver function index, hyaluronic acid (HA), superoxide dismutase (SOD), malondialdehyde (MDA), and decreased the hydroxyproline (Hyp) level in liver tissue. PZE also reduced the histological injuries induced by CCl4. In conclusion, these data demonstrated the hepatoprotective activities of PZE that may be related to its antioxidant properties.

Bluetongue is one of the most important vector-borne viral diseases that can lead to significant economic losses as a result of reduction of productivity and even death in some susceptible ruminants. However, epidemiological information on bluetongue virus (BTV) infection in cattle and goats is scarce in China. To determine the seropositive rate and risk factors of BTV infection in cattle and goats in Guangxi province, a subtropical region in Southern China, a total of 548 cattle serum samples and 6567 goat serum samples collected from 13 cities across Guangxi province during 2003-2015 were analyzed and found that the seroprevalence is 44.5% (244/548) in cattle and 28.0% (1837/6567) in goats and the main BTV serotypes are BTV-1, -2, -4, and -8. Climatic zone, age, and species are found to be the likely risk factors for BTV infection. To our knowledge, this is the first large-scale serological survey for BTV infection in domestic cattle and goats in Guangxi province, Southern China.

In 2013, seven virus strains were isolated from Culex tritaeniorhynchus and C. quinquefasciatus collected in Guangxi Province, China. The viruses caused cytopathological effects (CPE) only in C6/36 cells, and not in mammalian cells, e.g. BHK21 and Vero cells. Comparative genomic analysis revealed that the genomes of the seven virus strains ranged from 9589 to 9595 nt with G+C contents of 37.51-37.59%, and nucleotide sequence similarity > 99.8%. The nucleotide sequence homology of the seven virus strains with a Tanay virus isolate was 79.8-80.0%, while the corresponding amino acid homology of ORF1, ORF2, and ORF3 was 79.8-80, 73.9-4.6, and 93.4-94.3%, respectively. In phylogenetic analysis, based on the RdRP amino acid sequence, the seven virus strains were located on the same evolutionary branch, yet distinct from Tanay virus isolated from the Philippines. These results indicated that the seven virus strains isolated from Guangxi represent a new Tanay virus, "Tanay-like virus", within the genus Sandewavirus of the newly proposed taxon Negevirus. This was the first isolation of Tanay virus in China.

Bluetongue (BT) is one of the most important insect-borne, non-contagious viral diseases of ruminants and can cause severe disease and death in sheep. Its pathogen, bluetongue virus (BTV) has a double-stranded RNA genome consisting of 10 segments that provides an opportunity for field and vaccine strains of different serotypes to reassort whilst simultaneously infecting the same animal. For the first time, we report the full-length genome sequence of a BTV strain of serotype 21 (5149E) isolated from sentinel cattle in Guangxi Province in China in 2015. Sequence analysis suggested that the isolate 5149E had undergone a reassortment incident and acquired seg-6 from an isolate of BTV-16 which originated from Japan. This study aims to provide more understanding as to the origin and epidemiology of BTV.

An epizootic hemorrhagic disease virus (EHDV) strain designated YN09-04 was isolated from sentinel cattle in China. The length of its complete genome was 19,344 bp in total, consisting of 10 segments ranging in size from 810 bp (S10) to 3942 bp (S1). Based on phylogenetic analysis of the S2 sequence, YN09-04 clusters with EHDV serotype 7 (EHDV-7) strains form a distinct, well-supported subgroup, indicating that YN09-04 belongs to EHDV-7. However, the origin of the YN09-04 genome is very complex. The S2 and S6 of YN09-04 cluster with those of Japanese EHDV-7 strains, whereas the S1, S3, S4, S5 and S7 of YN09-04 share high nucleotide sequence identity and a close relationship with those of Japanese Ibaraki viruses, and the S8, S9 and S10 nucleotide sequences of YN09-04 are more similar to those of some Australian EHDV strains than to those of other isolates. These results suggest that the genome of YN09-04 likely originated from a reassortment event between EHDV strains that were similar to the current Japanese and Australian strains and that YN09-04 and some EHDVs from Japan and Australia share the same ancestors. This is the first report of the isolation, identification and complete-genome phylogenetic analysis of an EHDV-7 strain from China.

Previous studies have pointed out that bluetongue virus (BTV) down-regulates the expression levels of type Ⅰ interferon (IFN-Ⅰ) and inhibits IFN-Ⅰ signaling by targeting on the Janus tyrosine kinase (JAK)-signal transducer and activator of transcription protein (STAT) pathway. However, individual viral protein could not effectively block IFN-Ⅰ signaling. There is a need to explore the underlying mechanisms by which viral proteins of BTV coordinate to antagonize the IFN-Ⅰ signaling. We investigated the coordinative role of BTV-1 nonstructural protein 3 (NS3) and NS4 in counteracting IFN-Ⅰ signaling in the JAK-STAT pathway by directly interacting with STAT1. The NS3 and NS4 targeted the SH2 domain of STAT1 to inhibit its phosphorylation, heterodimerization, nuclear translocation, as well as activation of downstream genes of the JAK-STAT pathway. NS3 and NS4 impaired STAT1 phosphorylation induced by IFN-Ⅰ in a dose dependent manner. Overall, this study confirmed that NS3 and NS4 of BTV participate in interfering with IFN-Ⅰ signaling process. Also, a new mechanism employed by BTV to evade host innate immune responses was revealed.

The full-genome sequence of bluetongue virus serotype 15 (BTV-15) strain B105/YN/1996 isolated in China was determined for the first time. Phylogenetic analyses demonstrate that it belongs to the major eastern BTV topotype. This work is the first to document the complete genomic sequence of a BTV-15 strain from China. The sequence information will help determine the geographic origin of Chinese BTV-15 and provide data to facilitate future analyses of the genetic diversity and phylogenetic relationships of BTV strains.

Chuzan virus (CHUV) belongs to the Palyam serogroup, causes bovine congenital disease, and is prevalent in Asia. To date, only one full Palyam virus (PALV) genome sequence, that of Japanese CHUV strain K47, has been reported. Sequence analysis indicates that PALV strains isolated from different geographical regions show significant diversity, which is mainly shaped by geographically independent evolution and genetic reassortment. Our understanding of the genetic characteristics of PALV is hampered by a very limited genomic sequence database. In this study, we report the complete genome sequence of CHUV strain SZ187, which was isolated for the first time in 2012 in mainland China. Sequence alignment and phylogenetic analysis demonstrate that SZ187 is closely related to other CHUV strains isolated in Taiwan and Japan, indicating that they may share a common ancestor. This new full-length CHUV genome sequence could help in the design of broader assays for epidemiological studies and facilitate the identification of new CHUV isolates in the future.

YNTBa is a rabbit-passaged attenuated strain of foot-and-mouth disease virus (FMDV) serotype O. Here, we announce the complete genome sequence of YNTBa, which provides data for further studies on replication, virulence, its determinants, and cell and host tropism of YNTBa.

An amino acid mutation (R127→I) in the 3A non-structural protein of an FMDV serotype Asia1 rabbit-attenuated ZB strain was previously found after attenuation of the virus. To explore the effects of this mutation on viral replication and infection, the amino acid residue isoleucine (I) was changed to arginine (R) in the infectious cDNA clone of the rabbit-attenuated ZB strain by sitedirected mutagenesis, and the R127-mutated virus was rescued. BHK monolayer cells and suckling mice were inoculated with the R127-mutated virus to test its growth property and pathogenicity, respectively. The effects of the R127 mutation on viral replication and virulence were analyzed. The data showed that there was a slight difference in plaque morphology between the R127-mutated and wild-type viruses. The growth rate of the mutated virus was lower in BHK-21 cells and its virulence in suckling mice was also attenuated. This study indicates that the R127 mutation in 3A may play an important role in FMDV replication in vitro and in pathogenicity in suckling mice.

This report presents a highly sensitive, poly(A)-stabilized gold nanoparticle-based assay with dual readouts (resonance light scattering and colorimetric) for detecting palmatine hydrochloride (PaH) in real samples. The detection mechanism is based on the fact that palmatine hydrochloride has strong affinity to poly(A), which can stabilize gold nanoparticles at high ionic strength, and cause the aggregation of poly(A)-stabilized AuNPs, resulting in the enhanced resonance light scattering (RLS). At the same time, the color change of poly(A)-stabilized AuNPs solution is from red to blue via purple. Thus a highly sensitive RLS assay for PaH has been developed with a linear range of 0.023–2.5 μg/mL. The limit of detection (LOD, 3 σ ) is 2.3 ng/mL. In this work, the reaction mechanism of this system was investigated by scanning electron microscope (SEM), dark-field light scattering images (DLSI), dynamic light scattering (DLS) and circular dichroism (CD). This proposed method was also applied successfully for the determination of PaH in pharmaceutical preparations and urine samples with RSD ⩽ 4.0%. The results are in good agreement with those from the official method.

The RNA genome sequence of the rabbit passage-attenuated strain of foot-and-mouth disease virus (FMDV) Asia 1, ZB/CHA/58(att), was determined to be 8165 nt in length excluding the poly(C) tract in the 5' UTR and the poly(A) tail at the 3' end. ZB/CHA/58(att) was most similar to the vaccine strain Asia 1/YNBS/58 in genome sequence and there were no deletions or insertions within the deduced polyprotein between ZB/CHA/58(att) and YNBS/58, but there were a total of 25 substitutions at the amino acid level and an extra 19-nt stretch in the 5' UTR was found in ZB/CHA/58(att). An infectious full-length cDNA clone of ZB/CHA/58(att) was developed. Infectious virus could be recovered in BHK-21 cells transfected with the synthetic viral RNA transcribed in vitro. The plaque morphology, growth kinetics and antigenic profile of the infectious clone-derived virus (termed tZB) were indistinguishable from those induced by the parental virus. Furthermore, the virulence properties of ZB/CHA/58(att) and tZB were found to be highly similar in the mouse model. The availability of genome sequence information and infectious cDNA clone of the FMDV ZB/CHA/58(att) lays a new ground for further investigation of FMDV virulence determinants and development of new potent vaccine to FMD.

Hierarchical silica nanotubes with radially oriented mesoporous channels perpendicular to the central axis of the tube were synthesized by using self-assembled chiral anionic surfactant, co-structure directing agent (CSDA) and silica precursor. The average inner diameter and the wall thickness were ∼94, ∼62, and ∼62 nm and to ∼27, ∼33, and ∼45 nm, respectively, by manipulating the synthesis gel composition, while the diameter of the wall mesopores was kept constant at ∼4 nm. These materials with such a unique structure were produced only with chiral surfactant and achiral or racemic surfactant did not give rise to mesoporous silica nanotubes. The existence of helicity in the lipid bilayer template was confirmed by means of circular dichroism spectroscopy. The mesoporous penetrating from outside to inside of silica nanotubes are thought to originate from the initial formation of self-assembled lipid tubes with helical bilayers, which in turn re-assemble to form the mesopores in the wall of the nanotubes upon addition of co-structure directing agent and silica source.

Background Seadornavirus is a genus of viruses in the family Reoviridae, which consists of Banna virus, Kadipiro virus, and Liao ning virus. Banna virus is considered a potential pathogen for zoonotic diseases. Here, we describe a newly discovered Seadornavirus isolated from mosquitos (Culex tritaeniorhynchus) in Yunnan Province, China, which is related to Banna virus, and referred to as Mangshi virus. Methods and Results The Mangshi virus was isolated by cell culture in Aedes albopictus C6/36 cells, in which it replicated and caused cytopathic effects, but not in mammalian BHK-21 or Vero cells. Polyacrylamide gel analysis revealed a genome consisting of 12 segments of double-stranded RNA, with a “6–4–2” pattern in which the migrating bands were different from those of the Banna virus. Complete genome sequencing was performed by full-length amplification of cDNAs. Sequence analysis showed that seven highly conserved nucleotides and three highly conserved nucleotides were present at the ends of the 5′- and 3′-UTRs in each of 12 genome segments. The amino acid identities of Mangshi virus shared with Balaton virus varied from 27.3% (VP11) to 72.3% (VP1) with Banna virus varying from 18.0% (VP11) to 63.9% (VP1). Phylogenetic analysis based on amino acid sequences demonstrated that Mangshi virus is a member of the genus Seadornavirus and is most closely related to, but distinct from, Balaton virus and Banna virus in the genus Seadornavirus of the family Reoviridae. Conclusion Mangshi virus isolated from mosquitoes (C. tritaeniorhynchus) was identified as a newly discovered virus in the genus Seadornavirus and is phylogenetically close to Banna virus, suggesting that there is genetic diversity of seadornaviruses in tropical and subtropical areas of Southeast Asia.

Bluetongue virus (BTV) mainly infects sheep but can be transmitted to other domestic and wild ruminants, resulting in a considerable financial burden and trade restriction. Our understanding of the origin, movement, and distribution of BTV has been hindered by the fact that this virus has a segmented genome with the possibility of reassortment, the existence of 27 identified serotypes, and a lack of complete sequences of viruses isolated from different parts of the world. BTV serotype 7 is one of the prevalent BTV serotypes in Asia. Nonetheless, no complete genomic sequence of an Asian isolate of this serotype is available. In an effort to understand the molecular epidemiology of BTV infection in China, for the first time, we report here the complete genome sequence of a BTV serotype 7 strain, GDST008, which was isolated in 2014 in China. This sequence also represents the first complete genome sequence of a BTV serotype 7 from Asia and the third one in the world. Sequence analysis suggests that GDST008 consists of segments from BTV viruses of African lineage as well as those from China. Together, these results improve our understanding of the origin, emergence/re-emergence, and movement of BTV and thus can be applied in the development of vaccines and diagnostics.

Highly ordered mesoporous carbons with symmetry of three-dimensional (3d)-hexagonal P63/mmc and cubic Pm 3 ¯ n have been synthesized by using SBA-7 and SBA-1 silica as templates, respectively. The as-synthesized mesoporous silica templates without surfactant removal were treated by refluxing in boiling acid solution for a desired time, which was demonstrated effective for expanding the apertures and the cages as well, so that the introduction of sucrose solution was significantly improved and the mesostructure of silica template were replicated by carbon. To our best knowledge, it is the first time to report the synthesis of mesoporous carbon with 3d-hexagonal P63/mmc mesostructure.

The presence of various counteranions in the interfacial region of the silicate−surfactant mesophase introduces opportunities for manipulation of the phase structure. Well-ordered three-dimensional hexagonal P63/mmc, cubic Pm3n, two-dimensional hexagonal p6mm, and cubic Ia3d mesoporous materials have been synthesized with the same surfactant (cetyltriethylammonium bromide), depending on the kind of acids. The counteranions of acidic media have resulted in increasing surfactant packing parameter g in the order SO42- < Cl- < Br- < NO3-, which leads to different formation routes to the mesostructures. It has been found that the mesophases are always transformed from the lower curvature one into the higher curvature one in the acidic synthesis gel. The combination of X-ray diffraction patterns, scanning electron microscope images, and high-resolution transmission electron microscope images presented visible evidence for the mesostructural constructions. The full-genome sequence of the bluetongue virus serotype 1 (BTV-1) strain Y863, the first BTV-1 isolate of Eastern origin found in China, was determined. The virus was isolated from sheep during a severe outbreak of bluetongue in Shizhong County, Yunnan Province, southwest China, in 1979. The total size of the BTV-1 strain Y863 genome is 19,170 bp. Phylogenetic analyses demonstrate that it belongs to the major “Eastern” BTV topotype. The sequence information provided here will help in understanding the geographical origin and spread of this Chinese isolate of BTV-1, as well as aid in its comparison with global isolates of BTV-1 from sheep, cattle, and other host species origins.

Foot and mouth disease (FMD) outbreaks have been reported in China for many years. Recently, due to the rapid economic development, the price of meat and its demand have grown quickly. This trend has resulted in an increase in the number of livestock moving from south-east Asian countries into China. Foot and mouth disease is becoming one of the most important trans-boundary animal diseases affecting the livelihood of livestock owners in China. To contribute to the long term goal to control and eradicate FMD from China, the Chinese government has adopted a series of control measures which includes compulsory routine vaccination against the disease. In this paper, the surveillance results of the routine vaccination programme were systemically reviewed. The results from 28 published papers were combined and analysed through a meta-analysis approach. The results of the meta-analysis indicated that the vaccination programme has been very successful in China with more than 70% of animals protected against serotypes Asia-1 and O.

The 10kV distribution grid operates under an ‘open-ring operation’ mode. To avoid disturbance in HV power system due to the ring-opening or ring-closing current, ring-closing switching operation is forbidden on the LV (400V) bus-bar in distribution room. So, when switching operation on the LV bus-bar occurs, the customer should always endure a blackout, at least for several minutes. To solve this problem, the feasibility of ring-closing switching operation on the LV bus-bar is researched, through the digital simulation and field test. On this basis, a novel LV automatic bus transfer equipment (ABTE) based on PLC is developed and applied to distribution room in Beijing. By real time monitoring and detecting the ring-opening and ring-closing current, the function of ring current protection is integrated into the traditional ABTE, including function of automatic transferring and automatic resumption. Thus the LV customers can be automatic transferred to a standby resource without outage. This LV ABTE has been successfully applied to distribution grid in Beijing.

The complete genomic sequence of a bluetongue virus serotype 4 (BTV-4) strain (strain YTS-4), isolated from sentinel cattle in Yunnan Province, China, is reported here. This work is the first to document the complete genomic sequence of a BTV-4 strain from China. The sequence information will help determine the geographic origin of Chinese BTV-4 and provide data to facilitate future analyses of the genetic diversity and phylogenetic relationships of BTV strains.

In order to supply power to Olympic Games in 2008, OWTS(Oscillating Wave Test System) was introduced to Beijing. Many tests proved that OWTS is an effective way to find faults in 10kV cable joints that are newly installed. In 2011 Beijing Electric Power Corporation detected the state of 10kV cable lines sent by Power Supply Company in Beijing with OWTS(oscillating wave test system). We disassemble 10kV cable joints and found that Partial Discharge exceeded. We found many faults in 10kV cable joints, then analyzed 4 major factors that can influence the quality of cable joints in details, and made suggestions for revision.

In this paper, electrical tree structures once after initiation are presented, and the initiation time of electrical tree at different electric field strength is recorded by the stereomicroscope under continuous voltage. Applied voltage and electrode separation are changed to obtain different field strength. Numerical calculations based on curve fitting show a certain degree of field dependence of the electrical tree initiation time, which leads to a threshold field strength at this experiment system. However, initiation time shows increased dispersion by the decrease of field strength. Even when the field strength is close to the threshold, initiation becomes random. It is inferred that threshold field strength should be considered as an approximate value based on the available experiment results.

The complete genome of foot-and-mouth disease virus (FMDV) vaccine candidate strain O/YM/YN/2000 was sequenced and analyzed. Seven overlapping cDNA fragment which covered the O/YM/YN/2000 genome were amplified by RT-PCR and sequenced. The results showed that the complete genome of the strain O/YM/YN/2000 has 8 133 nucleotides (nt) in length including a 1 053 nt 5′- untranslated region (UTR) (including 18 nt poly C), a 6 969 nt open reading frame (ORF), a 93 nt 3′-UTR and at least 18 nt poly A tail. The homologous comparison and phylogenetic analysis of the nucleotide and deduced amino acid (aa) sequences among O/YM/YN/2000 and other FMDV strains available deposited in GenBank were performed. Phylogenetic analysis based on the complete genome and VP1 revealed that that the strain O/YM/YN/2000 is clustered in the FMDV serotype O cathay topotype. Fragment deletions mutation in O/YM/YN/2000 were revealed, deletions involving 4 nt in the S fragment of 5′-UTR, 43 nt which possibly contributed to the loss of pseudo-knot domain II of 5′-UTR and 10 aa residues in 3A protein. The results in this research might provide some information for screening FMDV vaccine strain

In China, the length of power cable tunnel exceeds 13,500 km and more than 80% of power cable lines rated voltage from 110 kV up to 500 kV were installed in the tunnel. Research and development of the smart telemonitor system for controlling and managing power cable tunnel in real time is emergency and necessary because the HV and EHV cable lines are so important for main power network. The smart telemonitor system, which has been put into operation in Beijing Power Corporation since 2007, was integrated in Command Center, and it has four representative characters, i.e. information or data transmission, command exchange and intelligent telecontrol in real time as well as grade interactive management. This smart system is designed for monitoring and controlling status of the power cable tunnel in real time based on sensors, cameras, optic-fiber and computer. The tunnel status includes cable surface temperature, air temperature of tunnel or fire or smoke, concentration of harmful gas, ground water level, cable partial discharge, manhole covers, person entering, lighting and ventilating, etc.. The principle and framework of this smart telemonitor system are described and discussed in this paper and its functions may be extended and improved continuously according to new requirements.

Waterproof surge protective grounding devices for high voltage power cable lines were design and developed by theory analysis and validating tests in this paper. The devices are made up of three main parts including high strength polymeric framework, copper bus-bars and over voltage protectors. All parts were sealed in one piece of EPR body which molded by continuously injecting rubber. These devices improve disadvantages of conventional protective grounding devices such as protection failure due to water intruded, dew coagulated and bus-bars incorrectly connected on site, etc. Type test was used for validating operation reliability and production quality of the protective device in the paper. The results of application research and type test had been proved that this waterproof surge protective device can satisfy the technical requirements of the standards and meet the practical demands of protective grounding and over voltage protecting of HV power cable lines in field.

A new type surge protective grounding device for high voltage power cable line was design and developed by theory analysis and validating tests in this paper. This device is made up of three main parts including high strength polymeric framework, copper bus-bar and over voltage protector. All parts were sealed in one piece of EPR body which molded by continuously injecting rubber. This device combined cable metallic shield grounding and cable sheath over voltage protecting techniques with waterproof technique to overcome defects of conventional protective grounding device such as protection failure due to water intruding, coagulated dew and incorrect connection etc. This protective grounding device not only protects power cable line from over voltage damaged and/or circulating current affected effectively, but also its weight and volume were decreased much more. Furthermore, type test was used for validating operation reliability and production quality of the protective device in the paper. The results of application research and type test had been proved that this new type surge protective device can satisfy the technical requirements of the standards and meet the practical demands of protective grounding and over voltage protecting of power cable lines in field.
