The present study aimed to identify novel immune-related markers of CAD.Methods: Three CAD-related datasets (GSE12288, GSE98583, GSE113079) were downloaded from the Gene Expression Integrated Database. Gene ontology annotation, Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis and weighted gene co-expression network analysis were performed on the common significantly differentially expressed genes (DEGs) of these three data sets, and the most relevant module genes for CAD obtained. The immune cell infiltration of module genes was evaluated with the CIBERSORT algorithm, and characteristic genes accompanied by their diagnostic effectiveness were screened by the machine-learning algorithm least absolute shrinkage and selection operator (LASSO) regression analysis. The expression levels of characteristic genes were evaluated in the peripheral blood mononuclear cells of CAD patients and healthy controls for verification.Results: A total of 204 upregulated and 339 downregulated DEGs were identified, which were mainly enriched in the following pathways: “Apoptosis”, “Th17 cell differentiation”, “Th1 and Th2 cell differentiation”, “Gl

Abstract

Particle-fluid flow in complex fractures plays a crucial role in the stimulation of unconventional reservoirs. In this paper, particle transport experiments were conducted in a vertical, nonplanar slot with two 90° bends and a straight slot. The corresponding simulation model was performed by a 3D coupled method of computational fluid dynamics (CFD) and discrete element method (DEM), which was calibrated and validated against experimental results. The effects of bending angle, fluid velocity and viscosity, particle density on particle placement are analyzed based on the experimental and simulation results. The studies reveal that the particle-fluid flow pattern is more complicated in the nonplanar slot than that of the straight slot. An irregular particle bed with two depressions around two bends would be formed. With the reduction of bending angle, the particle bed height and covered area decrease, while the sizes of the two depressions increase. The complicated flow induced by the bend increases the fluidization layer height. The high fluid velocity and viscosity can effectively suspend the low-density particles through two bends deeper into the complex fracture. A regression model with dimensionless numbers is developed for predicting the percentage of covered area. This study provides fundamental insight into understanding particle-fluid flow in a nonplanar fracture.

Abstract

Effective particle placement in complex fractures plays a crucial role in unconventional reservoirs. Non-planar fracture geometry poses a challenge to assess particle placement. Compared to the straight slot, particle-fluid flow in a curved fracture has not been fully understood. In this paper, particle-fluid flow in curved fractures is studied experimentally by laboratory size slots. The experimental results are compared with the published data, and they are consistent. After validation, the effect of bending angle, fluid velocity, particle size, particle density, and particle volume fraction on particle distribution is investigated. The results show that particle-fluid flow mechanisms in the curved slot are more complicated than that of the straight slot. With the decrease of the bending angle, the height and coverage area of the bed decrease. The flow redirection around the bend induces particle vortices, which change the transport mechanism from fluidization to resuspension. Although the bending section has a hindering effect on particle transport, resuspension promotes particles to flow through the bending section further into the slot and avoid bridging at the bend. The fluid velocity and particle size are two crucial factors impacting particle placement, and an irregular bed with two depressions would be formed in the curved slots. A correlation is developed for predicting the normalized coverage area. This study provides fundamental insight into understanding particle-fluid flow in a curved fracture.

Abstract

Na v 1.5 is the α-subunit of the cardiac sodium channel complex. Abnormal expression of Na v 1.5 on the cell surface because of mutations that disrupt Na v 1.5 trafficking causes Brugada syndrome (BrS), sick sinus syndrome (SSS), cardiac conduction disease, dilated cardiomyopathy, and sudden infant death syndrome. We and others previously reported that Ran-binding protein MOG1 (MOG1), a small protein that interacts with Na v 1.5, promotes Na v 1.5 intracellular trafficking to plasma membranes and that a substitution in MOG1, E83D, causes BrS. However, the molecular basis for the MOG1/Nav1.5 interaction and how the E83D substitution causes BrS remains unknown. Here, we assessed the effects of defined MOG1 deletions and alanine-scanning substitutions on MOG1's interaction with Na v 1.5. Large deletion analysis mapped the MOG1 domain required for the interaction with Na v 1.5 to the region spanning amino acids 146-174, and a refined deletion analysis further narrowed this domain to amino acids 146-155. Site-directed mutagenesis further revealed that Asp-148, Arg-150, and Ser-151 cluster in a peptide loop essential for binding to Na v 1.5. GST pulldown and electrophysiological analyses disclosed that the substitutions E83D, D148Q, R150Q, and S151Q disrupt MOG1's interaction with Na v 1.5 and significantly reduce its trafficking to the cell surface. Examination of MOG1's 3D structure revealed that Glu-83 and the loop containing Asp-148, Arg-150, and Ser-151 are spatially proximal, suggesting that these residues form a critical binding site for Na v 1.5. In conclusion, our findings identify the structural elements in MOG1 that are crucial for its interaction with Na v 1.5 and improve our understanding of how the E83D substitution causes BrS., (© 2018 Yu et al.)
