Abstract

In the Agricultural Internet of Things (AgIoT), Unmanned Aerial Vehicles (UAVs) can be used to collect sensor data. Thus, UAVs must plan the appropriate data collection paths so that sensors can collect the data under different positions and generate time-constrained data. Therefore, this paper proposes a UAV path planning algorithm based on Deep Reinforcement Learning (DRL), which jointly optimizes location, energy, and time deadline to maximize the data-energy ratio. The path planning process is modeled and decomposed into a Markov Decision Process (MDP), and then a Prioritized Experience Replay Double Deep Q Network (PER-DDQN) model is used to calculate the optimal solution. Furthermore, a time-constrained reward function and an improved adaptive upper confidence bound (UCB) exploration function are proposed to balance exploration and exploitation in the DRL algorithm, affording the developed algorithm to converge quickly and smoothly. The simulations demonstrate that compared with traditional methods, the proposed algorithm presents better path selection during the data collection process, lower execution time, and a higher data-energy ratio. Our algorithm promotes the use of UAV in AgIoT.

Abstract

We report on the detection of X-ray polarisation in the black-hole X-ray binary Swift J1727.8$-$1613 during its dim hard spectral state by the Imaging X-ray Polarimetry Explorer (IXPE). This is the first detection of X-ray polarisation at the transition from the soft to the hard state in an X-ray binary. We find an averaged 2$-$8 keV polarisation degree of (3.3 ${\pm}$ 0.4) % and a corresponding polarisation angle of 3{\deg} ${\pm}$ 4{\deg}, which matches the polarisation detected during the rising stage of the outburst, in September$-$October 2023, within 1${\sigma}$ uncertainty. The observational campaign complements previous studies of this source and enables comparison of the X-ray polarisation properties of a single transient across the X-ray hardness-intensity diagram. The complete recovery of the X-ray polarisation properties, including the energy dependence, came after a dramatic drop in the X-ray polarisation during the soft state. The new IXPE observations in the dim hard state at the reverse transition indicate that the accretion properties, including the geometry of the corona, appear to be strikingly similar to the bright hard state during the outburst rise despite the X-ray luminosities differing by two orders of magnitude., Comment: 8 pages, 6 figures. Abstract

XL-Calibur is a balloon-borne Compton polarimeter for X-rays in the $\sim$15-80 keV range. Using an X-ray mirror with a 12 m focal length for collecting photons onto a beryllium scattering rod surrounded by CZT detectors, a minimum-detectable polarization as low as $\sim$3% is expected during a 24-hour on-target observation of a 1 Crab source at 45$^{\circ}$ elevation. Systematic effects alter the reconstructed polarization as the mirror focal spot moves across the beryllium scatterer, due to pointing offsets, mechanical misalignment or deformation of the carbon-fiber truss supporting the mirror and the polarimeter. Unaddressed, this can give rise to a spurious polarization signal for an unpolarized flux, or a change in reconstructed polarization fraction and angle for a polarized flux. Using bench-marked Monte-Carlo simulations and an accurate mirror point-spread function characterized at synchrotron beam-lines, systematic effects are quantified, and mitigation strategies discussed. By recalculating the scattering site for a shifted beam, systematic errors can be reduced from several tens of percent to the few-percent level for any shift within the scattering element. The treatment of these systematic effects will be important for any polarimetric instrument where a focused X-ray beam is impinging on a scattering element surrounded by counting detectors., Comment: Submitted to Astroparticle Physics

Abstract

We present the results of the first X-ray polarimetric observation of the low-mass X-ray binary 4U 1957+115, performed with the Imaging X-ray Polarimetry Explorer in May 2023. The binary system has been in a high-soft spectral state since its discovery and is thought to host a black hole. The $\sim$571 ks observation reveals a linear polarisation degree of $1.9\% \pm 0.6\%$ and a polarisation angle of $-41^\circ.8 \pm 7^\circ.9$ in the 2-8 keV energy range. Spectral modelling is consistent with the dominant contribution coming from the standard accretion disc, while polarimetric data suggest a significant role of returning radiation: photons that are bent by strong gravity effects and forced to return to the disc surface, where they can be reflected before eventually reaching the observer. In this setting, we find that models with a black hole spin lower than 0.96 and an inclination lower than $50^\circ$ are disfavoured., Comment: 12 pages, 10 figures, 2 tables, accepted for publication in A&A

This review comprehensively examines the influence of LncRNA on the inflammatory processes driving OA progression, focusing on their role in regulating gene expression, cellular activities, and inflammatory pathways. Notably, LncRNAs such as MALAT1, H19, and HOTAIR are upregulated in OA and exacerbate the inflammatory milieu by modulating key signaling pathways like NF-κB, TGF-β/SMAD, and Wnt/β-catenin. Conversely, LncRNA like MEG3 and GAS5, which are downregulated in OA, show potential in dampening inflammatory responses and protecting against cartilage degradation by influencing miRNA interactions and cytokine production. By enhancing our understanding of LncRNA’ roles in OA inflammation, we can better leverage them as potential biomarkers for the disease and develop innovative therapeutic strategies for OA management. Abstract

Gamma rays from HESS J1849$-$000, a middle-aged TeV pulsar wind nebula (PWN), are observed by the Tibet air shower array and the muon detector array. The detection significance of gamma rays reaches $4.0\, \sigma$ and $4.4\, \sigma$ levels above 25 TeV and 100 TeV, respectively, in units of Gaussian standard deviation $\sigma$. The energy spectrum measured between $40\, {\rm TeV} < E < 320\, {\rm TeV}$ for the first time is described with a simple power-law function of ${\rm d}N/{\rm d}E = (2.86 \pm 1.44) \times 10^{-16}(E/40\, {\rm TeV})^{-2.24 \pm 0.41}\, {\rm TeV}^{-1}\, {\rm cm}^{-2}\, {\rm s}^{-1}$. The gamma-ray energy spectrum from the sub-TeV ($E < 1\, {\rm TeV}$) to sub-PeV ($100\, {\rm TeV} < E < 1\, {\rm PeV}$) ranges including the results of previous studies can be modeled with the leptonic scenario, inverse Compton scattering by high-energy electrons accelerated by the PWN of PSR J1849$-$0001. On the other hand, the gamma-ray energy spectrum can also be modeled with the hadronic scenario in which gamma rays are generated from the decay of neutral pions produced by collisions between accelerated cosmic-ray protons and the ambient molecular cloud found in the gamma-ray emitting region. The cutoff energy of cosmic-ray protons $E_{\rm p\, cut}$, cut is estimated at ${\rm log}_{10}(E_{\rm p,\, cut}/{\rm TeV}) = 3.73^{+2.98}_{-0.66}$, suggesting that protons are accelerated up to the PeV energy range. Our study thus proposes that HESS J1849$-$000 should be further investigated as a new candidate for a Galactic PeV cosmic-ray accelerator, PeVatron., Comment: 10 pages, 2 figures, Accepted for publication from the Astrophysical Journal

Abstract

HESS J1843$-$033 is a very-high-energy gamma-ray source whose origin remains unidentified. This work presents, for the first time, the energy spectrum of gamma rays beyond $100\, {\rm TeV}$ from the HESS J1843$-$033 region using the data recorded by the Tibet air shower array and its underground muon detector array. A gamma-ray source with an extension of $0.34^{\circ} \pm 0.12^{\circ}$ is successfully detected above $25\, {\rm TeV}$ at $(\alpha,\, \delta) = (281.09^{\circ}\pm 0.10^{\circ},\, -3.76^{\circ}\pm 0.09^{\circ})$ near HESS J1843$-$033 with a statistical significance of $6.2\, \sigma$, and the source is named TASG J1844$-$038. The position of TASG J1844$-$038 is consistent with those of HESS J1843$-$033, eHWC J1842$-$035, and LHAASO J1843$-$0338. The measured gamma-ray energy spectrum in $25\, {\rm TeV} < E < 130\, {\rm TeV}$ is described with ${\rm d}N/{\rm d}E = (9.70\pm 1.89)\times 10^{-16} (E/40\, {\rm TeV})^{-3.26\pm 0.30}\, {\rm TeV}^{-1} {\rm cm}^{-2} {\rm s}^{-1}$, and the spectral fit to the combined spectra of HESS J1843$-$033, LHAASO J1843$-$0338, and TASG J1844$-$038 implies the existence of a cutoff at $49.5\pm 9.0\, {\rm TeV}$. Associations of TASG J1844-038 with SNR G28.6$-$0.1 and PSR J1844-0346 are also discussed in detail for the first time., Comment: 11 pages, 4 figures, 1 table

Abstract

The Super $\tau$-Charm facility (STCF) is an electron-positron collider proposed by the Chinese particle physics community. It is designed to operate in a center-of-mass energy range from 2 to 7 GeV with a peak luminosity of $0.5\times 10^{35}{\rm cm}^{-2}{\rm s}^{-1}$ or higher. The STCF will produce a data sample about a factor of 100 larger than that by the present $\tau$-Charm factory -- the BEPCII, providing a unique platform for exploring the asymmetry of matter-antimatter (charge-parity violation), in-depth studies of the internal structure of hadrons and the nature of non-perturbative strong interactions, as well as searching for exotic hadrons and physics beyond the Standard Model. The STCF project in China is under development with an extensive R\&D program. This document presents the physics opportunities at the STCF, describes conceptual designs of the STCF detector system, and discusses future plans for detector R\&D and physics case studies.

Abstract

The increase in luminosity, and consequent higher backgrounds, of the LHC upgrades require improved rejection of fake tracks in the forward region of the ATLAS Muon Spectrometer. The New Small Wheel upgrade of the Muon Spectrometer aims to reduce the large background of fake triggers from track segments that are not originated from the interaction point. The New Small Wheel employs two detector technologies, the resistive strip Micromegas detectors and the "small" Thin Gap Chambers, with a total of 2.45 Million electrodes to be sensed. The two technologies require the design of a complex electronics system given that it consists of two different detector technologies and is required to provide both precision readout and a fast trigger. It will operate in a high background radiation region up to about 20 kHz/cm$^{2}$ at the expected HL-LHC luminosity of $\mathcal{L}$=7.5$\times10^{34}$cm$^{-2}$s$^{-1}$. The architecture of the system is strongly defined by the GBTx data aggregation ASIC, the newly-introduced FELIX data router and the software based data handler of the ATLAS detector. The electronics complex of this new detector was designed and developed in the last ten years and consists of multiple radiation tolerant Application Specific Integrated Circuits, multiple front-end boards, dense boards with FPGA's and purpose-built Trigger Processor boards within the ATCA standard. The New Small Wheel has been installed in 2021 and is undergoing integration within ATLAS for LHC Run 3. It should operate through the end of Run 4 (December 2032). Nevertheless, the therapeutic efficacy of GO nanoparticles is commonly hindered by their inherent drawback of low photothermal conversion efficiency (PCE).Methods: Herein, we engineer the Ag/GO-GelMA platform by growing the Ag on the surface of GO and encapsulating the Ag/GO nanoparticles into the GelMA hydrogels.Results: The resulting Ag/GO-GelMA platform demonstrates a significantly enhanced PCE (47.6%), surpassing that of pure GO (11.8%) by more than fourfold. As expected, the Ag/GO-GelMA platform, which was designed to integrate the benefits of Ag/GO nanoparticles (high PCE) and hydrogel (slowly releasing Ag+ to exert an inherent antibacterial effect), has been shown to exhibit exceptional antibacterial efficacy. Furthermore, transcriptome analyses demonstrated that the Ag/GO-GelMA platform could significantly down-regulate pathways linked to inflammation (the MAPK and PI3K-Akt pathways) and had the ability to promote cell migration.Discussion: Taken together, this study presents the design of a potent photothermal antibacterial platform (Ag/GO-GelMA) aimed at enhancing the healing of infectious wounds. The platform utilizes a handy method to enhance the PCE of GO, thereby making notable progress in the utilization of GO nano-PHTAs. The purpose of this study was to investigate the protective effect of rosmarinic acid (RA) on RIPF in mice as well as the mechanism involved.Methods: m7G-tRNA-seq and tRNA-seq analyses were conducted to identify m7G-modified tRNAs. Western blotting, immunohistochemistry, northwestern blotting, northern blotting, immunofluorescence, wound-healing assays and EdU experiments were performed to explore the molecular mechanism by which RA regulates fibroblast-to-myofibroblast transformation (FMT) by affecting the exosomes of lung epithelial cells. Ribo-seq and mRNA-seq analyses were used to explore the underlying target mRNAs. Seahorse assays and immunoprecipitation were carried out to elucidate the effects of RA on glycolysis and FMT processes via the regulation of 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 3 (PFKFB3) acetylation.Results: We found that RA had an antifibrotic effect on the lung tissues of RIPF model mice and inhibited the progression of FMT through exosomes derived from lung epithelial cells. Mechanistically, RA reduced the transcription and translation efficiency of sphingosine kinase 1 in lung fibroblasts by decreasing N7-methylguanosine modification of tRNA, downregulating the expression of tRNAs in irradiated lung epithelial cell-derived exosomes, and inhibiting the interaction between sphingosine kinase 1 and the N-acetyltransferase 10 protein in fibroblasts. Furthermore, the acetylation and cytoplasmic translocation of PFKFB3 were reduced by exosomes derived from irradiated lung epithelial cells, which following RA intervention. This suppression of the FMT process, which is triggered by glycolysis, and ultimately decelerating the progression of RIPF.Conclusion: These findings suggest that RA is a potential therapeutic agent for RIPF.Keywords: rosmarinic acid, exosomes, tRNA, N7-methylguanosine, radiation-induced pulmonary fibrosis

So the synergistic administration of chemotherapeutic drug DOX and JQ1 is expected to increase the sensitivity of tumors to immune checkpoint therapy and jointly enhance the body’s own immunity, thus effectively killing tumor cells. Therefore, a drug delivery system loaded with DOX and JQ1 was devised in this study.Methods: Polydopamine nanoparticles (PDA NPs) were synthesized through spontaneous polymerization. Under appropriate pH conditions, DOX and JQ1 were loaded onto the surface of PDA NPs, and the release of DOX and JQ1 were measured using UV-Vis or high performance liquid chromatography (HPLC). The mechanism of fabricated nanocomplex in vitro was investigated by cell uptake experiment, cell viability assays, apoptosis assays, and Western blot analysis. Finally, the tumor-bearing mouse model was used to evaluate the tumor-inhibiting efficacy and the biosafety in vivo.Results: JQ1 and DOX were successfully loaded onto PDA NPs. PDA-DOX/JQ1 NPs inhibited the growth of prostate cancer cells, reduced the expression of apoptosis related proteins and induced apoptosis in vitro. The in vivo biodistribution indicated that PDA-DOX/JQ1 NPs could accumulated at the tumor sites through the EPR effect. In tumor-bearing mice, JQ1 delivered with PDA-DOX/JQ1 NPs reduced PD-L1 expression at tumor sites, generating significant tumor suppression. Furthermore, PDA-DOX/JQ1 NPs could reduce the side effects, and produce good synergistic treatment effect in vivo.Conclusion: We have successfully prepared a multifunctional platform for synergistic prostate cancer therapy. Despite receiving treatment, patients with the middle and late stage of OA have poor survival outcomes. Therefore, within the framework of predictive, preventive, and personalized medicine (PPPM/3PM), early personalized diagnosis of OA is particularly prominent. PPPM aims to accurately identify disease by integrating multiple omic techniques; however, the efficiency of currently available methods and biomarkers in predicting and diagnosing OA should be improved. Disulfidptosis, a novel programmed cell death mechanism and appeared in particular metabolic status, plays a mysterious characteristic in the occurrence and development of OA, which warrants further investigation.Methods: In this study, we integrated three public datasets from the Gene Expression Omnibus (GEO) database, including 26 OA samples and 20 normal samples. Via a series of bioinformatic analysis and machine learning, we identified the diagnostic biomarkers and several subtypes of OA. Moreover, the expression of these biomarkers were verified in our in-house cohort and the single cell dataset.Results: Three significant regulators of disulfidptosis (NCKAP1, OXSM, and SLC3A2) were identified through differential expression analysis and machine learning. And a nomogram constructed based on these three regulators exhibited ideal efficiency in predicting early- and late-stage OA. Furthermore, based on the expression of three regulators, we identified two disulfidptosis-related subtypes of OA with different infiltration of immune cells and personalized expression level of immune checkpoints. Notably, the expression of the three regulators was demonstrated in a single-cell RNA profile and verified in the synovial tissue in our in-house cohort including 6 OA patients and 6 normal people. Finally, an efficient disulfidptosis-mediated diagnostic model was constructed for OA, with the AUC value of 97.6923% in the training set and 93.3333% and 100% in two validation sets.Conclusion: Overall, with regard to PPPM, this study provided novel insights into the role of disulfidptosis regulators in the personalized diagnosis and treatment of OA.Keywords: osteoarthritis, disulfidptosis, perspective of predictive, preventive and personalized medicine, immune microenvironment, diagnostic model

Analyzing the relationship between BMI and percentage of excess weight loss (%EWL) by using Spearman correlation analysis and linear regression analysis.Results: The %EWL at 12 months after the surgical procedure was (104.26± 16.41)%, (90.36± 9.98)%, and (78.30± 14.64)% for patients with Class I, II, and III obesity, respectively, P< 0.05. Spearman correlation coefficients between %EWL and BMI at 1, 3, 6, and 12 months after surgery were R=− 0.334 (P< 0.001), R=− 0.389 (P< 0.001), and R=− 0.442 (P< 0.001), R=− 0.641 (P< 0.001), respectively. The remission of hypertension, diabetes and dyslipidaemia did not differ significantly between groups (P> 0.05).Conclusion: Individuals with obesity for varying BMI can experience favorable outcomes following LSG surgery. The purpose of this study was to explore the drug–drug interaction (DDI) of multiple drugs combined with olanzapine and to recommend the optimal administration of olanzapine in schizophrenia patients.Methods: In this study, we obtained olanzapine concentrations from therapeutic drug monitoring (TDM) database. In addition, related medical information, such as physiological, biochemical indexes, and concomitant drugs was acquired using medical log. Sixty-five schizophrenia patients were enrollmented for analysis using population pharmacokinetic model by means of nonlinear mixed effect (NONMEM).Results: Weight and combined use of aripiprazole significantly affected olanzapine clearance. Without aripiprazole, for once-daily olanzapine administration dosages, 0.6, 0.5 mg/kg/day were recommended for 40– 70, and 70– 100 kg schizophrenia patients, respectively; for twice-daily olanzapine administration dosages, 0.6, 0.5 mg/kg/day were recommended for 40– 60, and 60– 100 kg schizophrenia patients, respectively. With aripiprazole, for once-daily olanzapine administration dosages, 0.4, 0.3 mg/kg/day were recommended for 40– 53, and 53– 100 kg schizophrenia patients, respectively; for twice-daily olanzapine administration dosages, 0.4 mg/kg/day was recommended for 40– 100 kg schizophrenia patients, respectively.Conclusion: Aripiprazole significantly affected olanzapine clearance, and when schizophrenia patients use aripiprazole, the olanzapine dosages need adjust. Signature genes and ferroptosis phenotypes were verified through in vivo animal experiments and clinical samples. The expression levels of non-coding RNAs in samples from patients with OA were determined using qRT-PCR. ceRNA network analysis results were confirmed using dual-luciferase assays.Results: JUN, ATF3, and CDKN1A were identified as OA- and ferroptosis-associated signature genes. GSEA analysis demonstrated an enrichment of these genes in immune and inflammatory responses, and amino acid metabolism. The CIBERSORT algorithm showed a negative correlation between T cells and these signature genes in the cartilage, and a positive correlation in the synovium. Moreover, RP5-894D12.5 and FAM95B1 regulated the expression of JUN, ATF3, and CDKN1A by competitively binding to miR-1972, miR-665, and miR-181a-2-3p. In vivo, GPX4 was downregulated in both OA cartilage and synovium; however, GPX4 and GSH were downregulated, while ferrous ions were upregulated in patient OA cartilage and synovium samples, indicating that ferroptosis was involved in the pathogenesis of OA. Furthermore, JUN, ATF3, and CDKN1A expression was downregulated in both mouse and human OA synovial and cartilage tissues. qRT-PCR demonstrated that miR-1972, RP5-894D12.5, and FAM95B1 were differentially expressed in OA tissues. Targeted interactions between miR-1972 and JUN, and a ceRNA regulatory mechanism between RP5-894D12.5, miR-1972, and JUN were confirmed by dual-luciferase assays.Conclusion: This study identified JUN, ATF3, and CDKN1A as possible diagnostic biomarkers and therapeutic targets for joint synovitis and OA. Abstract

Material extrusion (ME) technique has been one of the most popular additive manufacturing techniques because of its environmental friendliness, low cost, simple operation, and so forth. However, significant surface defects occur on ME parts due to the discontinuity between adjacent layers. For this reason, a new method, using ultrasonic-vibration machining, was innovatively proposed to improve the ME parts' surface quality. A theoretical surface roughness (SR) model of the ME parts processed by using ultrasonic vibration was established. An ultrasonic-vibrating ME equipment was then set up by refitting an ordinary one and the samples built without and with ultrasonic-vibration machining utilized were prepared, respectively. With the laser microscope, the built parts' SR was tested and determined. Compared the predicted SR results with the measured ones, the theoretical model was verified. The influence of different ultrasonic vibration on the surface quality of ME parts was also discussed, clarifying the corresponding mechanism. Abstract

We report the ultrafast optical pump-probe spectroscopy measurements on the recently discovered quantum critical ferromagnet CeRh$_6$Ge$_4$. Our experimental results reveal the two-stage development of the hybridization between localized $f$ moments and conduction electrons with lowering temperature, as evidenced by (1) the presence of hybridization fluctuation for temperatures from $\sim$85 K ($T^*$) to $\sim$140 K ($T^\dagger$), and (2) the emergence of collective hybridization below the coherence temperature, $T^*$, marked by the opening of an indirect gap of 2$\Delta$ $\approx$12 meV. We also observe three coherent phonon modes being softened anomalously below $T^*$, reflecting directly their coupling with the emergent coherent heavy electrons. Our findings establish the universal nature of the hybridization process in different heavy fermion systems.

31. Relativistic light synthesis of femtosecond sawtooth pulses

Abstract

Formation of femtosecond sawtooth pulses by high harmonic generation from relativistic oscillating mirrors is studied. For oblique incidence of $p$-polarized laser pulse, one can efficiently control the intensity of the first few harmonics by modifying the plasma density gradient at the plasma-vacuum interface. With appropriate choice of the laser amplitude and incidence angle, the generated harmonics can form a millijoule femtosecond sawtooth pulse. The scheme does not rely on phase manipulation and/or pulse synchronization as in the state-of-art methods. It is also shown that such a sawtooth pulse can boost the generation of ultrashort terahertz pulses in laser-plasma interaction by one order of magnitude, as compared with that from the conventional two-color laser pulse., Comment: 5 pages, 4 figures

However, the comprehensive analysis of the CBX family in head and neck squamous cell carcinoma (HNSC) is lacking.Methods: In this work, we used multiple online databases and tools to investigate the roles of CBX family in aspects of gene expression, prognostic evaluation, genetic alteration, immune micro-environment of tumor, and status of methylation.Results: The mRNA expression levels of CBX1, CBX3, and CBX5 were aberrantly increased in patients with HNSC, while CBX7 was aberrantly decreased. Higher expression of CBX7 was significantly associated with longer OS. Within the 5– 11% of genetic alteration rate of CBXs, CBX3 ranked the highest and CBX5/7 ranked the lowest. SPRR1B, S100A7, CASP14, CDSN, LCE3D were the top 5 neighbor genes with the strongest association with CBXs in HNSC patients. Signaling pathways such as epidermal cell differentiation, cornification, and peptide cross-linking were demonstrated to have a strong association with CBX genes. The profiles of immune cell infiltration had high similarity for the group of HNSC patients stratified by expression of CBXs. The methylation levels of CBX1 and CBX5 significantly decreased, while that of CBX7 significantly increased in HNSC samples when compared with normal tissue.Conclusion: In conclusion, the CBX family showed its valuation for further investigation in HNSC. DSB burden was assessed based on answers to two questions on DSBs. Participants who answered “sometimes” or “often/usually” to one of them were considered to have poor DSB burden, or otherwise were considered not having any. A composite cognition score was computed by averaging the available standardized individual test results from four neurocognitive tests: ie, a reaction time test for information processing speed, a pairs matching test for visual episodic memory, a fluid intelligence test for reasoning, and a prospective memory test. Mixed-effects models with adjustment for the variables used in extracting matched uninfected controls were performed to test the hypotheses.Results: Having poor DSB burden was associated with a 0.15 – standard deviation (SD) decrease in cognitive performance (p = 0.006). People living with HIV infection (PLWH) also performed worse on the cognitive tasks than uninfected controls, with an effect size similar to that of having poor DSB burden (p = 0.003). HIV infection significantly modified the negative association between DSB burden and cognition (p for interaction: 0.008). Specifically, the association between DSB burden and cognition was not statistically significant in uninfected controls, whereas PLWH who reported having poor DSB burden had a 0.28 – SD decrease in cognitive performance compared to PLWH who did not.Conclusion: HIV infection significantly increased the adverse association between DSBs and cognitive performance. Weighted gene co-expression network analysis (WGCNA) was used to further identify important mRNAs. The negatively regulatory network construction of miRNA–inflammation, oxidative stress and prognosis-related mRNAs was performed, followed by protein–protein interaction (PPI) and functional analysis of mRNAs.Results: A total of three pairs of negatively regulatory network of miRNA–inflammation and prognosis-related mRNAs (hsa-miR-636/hsa-miR-491-3p/hsa-miR-188-5p/hsa-miR-188-3p-AQP9, hsa-miR-518a-3p-C5AR1 and hsa-miR-509-3-5p/hsa-miR-127-5p-PLAUR), two pairs of negatively regulatory network of miRNA–oxidative stress and prognosis-related mRNAs (hsa-miR-604-TLR4 and hsa-miR-139-5p-CXCL1) and three pairs of negatively regulatory network of miRNA-inflammation, oxidative stress and prognosis-related mRNA (hsa-miR-634/hsa-miR-591-TLR2, hsa-miR-938-NFKBIA and hsa-miR-520h/hsa-miR-450b-3p-ADM) were identified. In this study, the mechanism of YHT in LUAD recurrence was investigated.Methods: Firstly, the bioactive compounds and targets of YHT, as well as related targets of LUAD recurrence, were collected from public databases. The protein–protein interaction network, Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses were performed to find the pivotal compounds, hub genes, functional annotation and main pathways. Subsequently, RNA sequencing of recurrent tumor tissues from Lewis lung carcinoma mice treated with YHT was used to explore the main pathways. At the same time, pathways screened by network pharmacology and RNA sequencing analysis were considered the most important pathways. Finally, liquid chromatography mass spectrometry was used to validate the pivotal active ingredients. Molecular docking technology was performed to validate the binding association between the hub genes and the pivotal active ingredients. PCR and WB analysis were used to validate the main pathways.Results: There were 128 active compounds and 419 targets interacting with YHT and LUAD recurrence. Network analysis identified 4 pivotal compounds, 28 hub genes and 30 main pathways. Sphingolipid signaling pathway was the common main pathway in network pharmacology and RNA sequencing results. The hub gene related to the sphingolipid signaling pathway was S1PR5. Qualitative phytochemical analysis confirmed the presence of 3 pivotal compounds, namely stigmasterol, nootkatone and ergotamine. The molecular docking verified that the pivotal compounds could good affinity with S1PR5. The PCR and WB analysis verified YHT suppressed Lewis lung cancer cells proliferation and migration by inhibiting the sphingolipid signaling pathway.Conclusion: The potential mechanism and therapeutic effect of YHT against the recurrence of LUAD may be ascribed to inhibition of the sphingolipid signaling pathway.Graphical Abstract: Keywords: sphingolipid signaling pathway, traditional Chinese medicine, S1PR5, stigmasterol, nootkatone, ergotamine

However, there still remain unanswered questions about the relationship between overlap syndrome (OVS), where OSA and COPD coexist, and polycythemia. Here, we aimed to establish the prevalence of polycythemia in OVS patients and to explore the impact of OSA on polycythemia.Patients and Methods: Patients with COPD underwent overnight polysomnography (PSG), pulmonary function tests, echocardiography, and complete blood counts. All patients were ethnic Han Chinese and free of prolonged oral corticosteroid use, hematological system disease, severe systemic disease, and other sleep-disordered breathing. OVS was defined as COPD patients with an apnea–hypopnea index ≥ 15 events/h, and polycythemia was defined as an Hb > 165 g/L in men and > 160 g/L in women.Results: Eight-hundred and eighty-six patients with COPD were included in the analysis. The prevalence of polycythemia was significantly higher in OVS patients than COPD-alone patients (6.4% vs 2.9%, p < 0.05). The prevalence of polycythemia increased with OSA severity (χ2 = 7.885, p = 0.007), but not in GOLD grade 3– 4 COPD patients (χ2 = 0.190, p = 0.663). After adjusting for confounders, percentage of total sleep time with SaO2 < 90% (TS90) remained independently associated with an increased odds of polycythemia (OR 1.030, 95% CI 1.015– 1.046) and, with an increase in TS90, the hemoglobin increased, especially in GOLD grade 1– 2 patients (p < 0.05).Conclusion: Patients with OVS have a higher prevalence of polycythemia than those with COPD alone, and TS90 is an independent factor for polycythemia, especially in GOLD1-2 COPD patients.Keywords: chronic obstructive pulmonary disease, obstructive sleep apnea, overlap syndrome, polycythemia

BackgroundHepatitis B virus (HBV) infection is common with major clinical consequences. In Asian Americans, the HBsAg carrier rate ranges from 2% to 16% which approximates the rates from their countries of origin. Similarly, HBV is the most important cause of cirrhosis, hepatocellular carcinoma (HCC) and liver related deaths in HBsAg positive Asians worldwide.AimTo generate recommendations for the management of Asian Americans infected with HBV.MethodsThese guidelines are based on relevant data derived from medical reports on HBV from Asian countries as well as from studies in the HBsAg positive Asian Americans. Therefore, deriving a prognostic signature by integrating hypoxia- and immune infiltrating cell-related genes (H&IICRGs) may add value over prognostic indices derived from genes driving either process alone.Methods: Differentially expressed H&IICRGs (DE-H&IICRGs) were identified in The Cancer Genome Atlas transcriptomic data using limma, CIBERSORT, weighted gene co-expression network analysis, and intersection analysis. A stepwise Cox regression model was constructed to identify prognostic genes and to produce a gene signature based on DE-H&IICRGs. The potential biological functions associated with the gene signature were explored using functional enrichment analysis. The prognostic signature was externally validated in a separate cohort from the Gene Expression Omnibus database.Results: Five prognostic genes associated with overall survival in LUAD were used in the DE-H&IICRG-based prognostic signature. Patients in the high-risk group had an inferior prognosis, which was validated in an independent external cohort, and had lower expression of most immune checkpoint genes. In multivariate analysis, only risk score and T stage were independent prognostic factors. Differentially expressed genes (DEGs) associated with the risk score were enriched for pathways related to cell cycle, hypoxia regulation, and immune response. TIDE analyses showed that low-risk LUAD patients might also respond better to immunotherapy.Conclusion: This study establishes and validates a prognostic profile for LUAD patients that combines hypoxia and immune infiltrating cell-related genes. This study aimed to compare the therapeutic effects of two agents on osteoarthritis (OA) and investigate underlying mechanism using proteomics.Methods: In vitro, the proliferation and migration abilities of chondrocytes treated with hUC-MSCs or hUC-MSC-sEVs were detected by Cell Counting Kit-8 assay and scratch wound assay. In vivo, hUC-MSCs (a single dose of 5 × 105) or hUC-MSC-sEVs (30 μg/time) were injected into the knee joints of anterior cruciate ligament transection-induced OA model. Hematoxylin and eosin, Safranin O/Fast Green staining were used to observe cartilage degeneration. The levels of cartilage matrix metabolic molecules (Collagen II, MMP13 and ADAMTS5) and macrophage polarization markers (CD14, IL-1β, IL-10 and CD206) were assessed by immunohistochemistry. Finally, proteomics analysis was performed to characterize the proteinaceous contents of two agents.Results: In vitro data showed that hUC-MSC-sEVs were taken up by chondrocytes. A total of 15 μg/mL of sEVs show the greatest proliferative and migratory capacities among all groups. In the animal study, hUC-MSCs and hUC-MSC-sEVs alleviated cartilage damage. This effect was mediated via maintaining cartilage homeostasis, as was confirmed by upregulation of the COL II and downregulation of the MMP13 and ADAMTS5. Moreover, the M1 macrophage markers (CD14) were significantly reduced, while the M2 macrophage markers (CD206 and IL-10) were increased in the hUC-MSCs and hUC-MSC-sEVs relative to the untreated group. Mechanistically, we found that many proteins connected to cartilage repair were more abundant in sEVs. Notably, compared to hUC-MSCs, the upregulated proteins in sEVs were mostly involved in the regulation of immune effector process, extracellular matrix organization, PI3K-AKT signaling pathways, and Rap1 signaling pathway.Conclusion: Our study indicated that hUC-MSC-sEVs protect cartilage from damage and many cartilage repair-related proteins are probably involved in the restoration process. Their morphology, particle size, zeta potential, entrapment efficiency, release profiles, and targeting effects were studied. The antitumor effects of the docetaxel-loaded nano-micelles were investigated in a MDA-MB-231 cell model in vitro and a MDA-MB-231 xenograft model in vivo.Results: The prepared RGD-modified docetaxel-loaded lipid-core micelles were spherical with a particle size of 16.44± 1.35 nm, zeta potential of − 19.24± 1.24 mV, and an encapsulation efficiency of 96.52± 0.43%. The drug delivery system showed sustained release properties and could significantly enhance docetaxel uptake by MDA-MB-231 tumor cells in vitro, which was proved to be a caveolae pathway mediated process requiring ATP, Golgi apparatus, and acid lysosomes. The results of the pharmacokinetic study displayed that the area under the curve of the targeted micelles was 3.2-times higher than that of docetaxel commercial injections. Furthermore, in a MDA-MB-231 tumor-bearing mice model, a higher antitumor efficacy than docetaxel commercial injections was displayed, and the safety experiments showed that the micellar material did not cause major organ damage after intravenous administration in mice.Conclusion: The novel RGD-modified PEGylated lipid-core micelle delivery system significantly improved the antitumor effects and reduced the side-effects of docetaxel, providing a promising therapeutics for the treatment of TNBC.Keywords: docetaxel, lipid-core micelles, RGD peptide, breast cancer therapy

Physical cellular microenvironment is of critical importance for stem cells differentiation and influences the function of other biological/chemical factors to differentiation.Methods: Based on adjusting the mechanical/interfacial properties of hydrogels, multicellular spheroids (MCSs) of DPSCs generated through self-organization. The spheroids were characterized by immunofluorescent staining and flow cytometry. Quantitative real-time polymerase chain reaction, alkaline phosphatase (ALP) activity assay, ALP staining and Alizarin Red S staining were performed to evaluate the osteogenic/odontogenic differentiation of DPSCs with or without magnetic iron oxide nanoparticles (IONPs) induction.Results: MCSs of DPSCs exhibited a significant upregulation of E-cadherin and N-cadherin and enriched CD146 positive subpopulation, along with a stronger osteogenic/odontogenic differentiation ability. Moreover, DPSCs spheroids showed more substantial osteogenic differentiation tendency than the classical two-dimensional cultured DPSCs under the stimulation of magnetic IONPs.Conclusion: Three-dimensional spheroids culture of DPSCs based on composite viscoelastic materials combined with mechanical/magnetic stimulation may provide a theoretical basis for the subsequent development of dentin or bone regeneration technology.Keywords: tunable mechanical properties, dental pulp stem cells, magnetic nanomaterials, osteogenic/odontogenic differentiation, multicellular spheroids
