Abstract

A series of flavanols were synthesized to assess their biological activity against human non-small cell lung cancer cells (A549). Among the sixteen synthesized compounds, it was observed that compounds 6k (3.14 ± 0.29 µM) and 6l (0.46 ± 0.02 µM) exhibited higher potency compared to 5-fluorouracil (5-Fu, 4.98 ± 0.41 µM), a clinical anticancer drug which was used as a positive control. Moreover, compound 6l (4'-bromoflavonol) markedly induced apoptosis of A549 cells through the mitochondrial- and caspase-3-dependent pathways. Consequently, compound 6l might be developed as a candidate for treating or preventing lung cancer. [ABSTRACT FROM AUTHOR]

21. Using capillary electrophoresis--selective tandem mass spectrometry to identify pathogens in clinical samples

Abstract

Analysis of microbial mixtures in complex systems, such as clinical samples, using mass spectrometry can be challenging because the specimens may contain mixtures of several pathogens or both pathogens and nonpathogens. We have successfully applied capillary electrophoresis-selective MS/MS of unique peptide marker ions to the identification of common pathogens in clinical diagnosis. We searched the CE-MS/MS spectra acquired from the proteolytic digests of pure bacterial cell extracts against protein databases. The identified peptides that matched a protein associated with a particular pathogen were selected as marker ions to identify that bacterium in clinical specimens. Thirty-four clinical specimens, obtained from pus, wound, sputum, and urine samples, were analyzed using both biochemical and selective MS/ MS methods. The bacteria in these clinical samples were cultivated directly, without prior isolation of a pure colony, before performing the selective MS/MS analyses. The bacteria analyzed included both Gram-positive and -negative strains. The match with respect to the pathogens identified was good between the biochemical and the selective MS/MS methods; the matching rate was 91%. The rate was as high as 97% when not considering two specimens for which the bacteria were not grown successfully. Two of the specimens that we identified using the biochemical method as containing two bacterial species were confirmed also through selective tandem MS analysis.

Abstract

In this paper, we propose a new strategy for identifying specific bacteria in bacterial mixtures by using CE-selective MS/MS of peptide marker ions associated with the bacteria of interest. We searched the CE-MS/MS spectra acquired from the proteolytic digests of pure bacterial cell extracts against protein databases. The identified peptides that match the protein associated with the corresponding species were selected as marker ions for bacterial identification. Specific peptide marker ions were obtained for each of the following three pathogens: Pseudomonas aeruginasa, Staphylococcus aureus, and Staphylococcus epidermidis. To identify a bacterial species in a sample, we performed CE-MS/MS analysis of the selected marker ions in the proteolytic digest of the cell extract and then performed protein database searches. The selected peptides that we identified correctly from Xcorr values ranking at the top of the search results allowed us to identify the corresponding bacterial species present in the sample. We have applied this method successfully to the identification of various mixtures of the three pathogens. Even minor bacterial species present at a concentration of 1% can be identified with great confidence. This method for CE-MS/MS analysis of bacteria-specific marker peptides provides excellent selectivity and high accuracy when identifying bacterial species in complex systems. In addition, we have used this approach to identify P. aeruginasa in a saliva sample spiked with E. coli and P. aeruginasa.

32. Motion of a spacecraft with magnetic damper

Abstract

Three methods of numerically simulating the motion of a spacecraft with a magnetic damper are compared. Simulations of motion of the initial assembly stage of Space Station Freedom show that results obtained with the first approach are in general agreement with those based on the second approach, while results from the third method are incorrect unless the spacecraft is nearly at rest in a local-vertical-local-horizontal reference frame. Simulations based on the second method proceed much more quickly than simulations based on the first. An integral of equations of motion governing the behavior of a spacecraft, and a sphere, which is part of the damper is presented. The integral can be used to test the results of numerical integrations performed in connection with the first and second approaches.

34. Special class of nonlinear damping models in flexible space structures

Abstract

A special class of nonlinear damping models is investigated in which the damping force is proportional to the product of positive integer or the fractional power of the absolute values of displacement and velocity. For a one-degree-of-freedom system, the classical Krylov-Bogoliubov 'averaging' method is used, whereas for a distributed system, both an ad hoc perturbation technique and the finite difference method are employed to study the effects of nonlinear damping. The results are compared with linear viscous damping models. The amplitude decrement of free vibration for a single mode system with nonlinear models depends not only on the damping ratio but also on the initial amplitude, the time to measure the response, the frequency of the system, and the powers of displacement and velocity. For the distributed system, the action of nonlinear damping is found to reduce the energy of the system and to pass energy to lower modes.

Abstract

A convenient three-step strategy is proposed for the synthesis of mono and bis-indolo[1,2-c] quinazolines from 2-(2-aminophenyl)indole and various aryl aldehydes. The newly synthesized compounds were characterized by elemental analysis, IR, ¹H NMR, 13C NMR, and mass spectroscopic investigation. All the derivatives were screened for antibacterial (S. aureus, B. subtilis, S. pyogenes, S. typhimurium, E. coli, K. pneumonia) and antifungal (A. niger, C. albicans, T. viridae) activities by cup plate method. Among the compounds tested, mono- indolo[1,2-c] quinazolines (15-18) exhibited good antibacterial activities while 15 and 18 also showed notable antifungal activity. Especially, 19 and 20 exhibited stronger antibacterial as well as antifungal activity against all tested strains. Uma estratégia conveniente em três etapas é proposta para a síntese de mono e bis-indolo[1,2-c] quinazolinas, a partir de 2-(2-aminofenil)indol e aril aldeídos. Os novos compostos sintetizados foram caracterizados por análise elementar, IV, ¹H RMN, 13C RMN, e espectrometria de massa. Todos os derivados foram testados para avaliação das suas atividades antibacterial (S. aureus, B. subtilis, S. pyogenes, S. typhimurium, E. coli, K. pneumonia) e antifúngica (A. niger, C. albicans, T. viridae) usando o método cup plate. Dentre os compostos testados, as mono- indolo[1,2-c] quinazolinas (15-18) exibiram boas atividades antibacteriais, enquanto 15 e 18 também mostraram notável atividade antifúngica. Especialmente, 19 e 20 exibiram forte atividade antibacteriana e antifúngica contra todas as cepas testadas.

The emergence and spread of carbapenem-resistant Acinetobacter baumanniiposes a challenge for optimizing antibiotic therapies and preventing outbreaks. Traditional phenotypic assays such as the modified Hodge test (MHT) or polymerase chain reaction (PCR)-based detection of the carbapenemase genes are time-consuming and complicated. Therefore, new approaches for the efficient detection of carbapenemase-producing A. baumanniiare urgently required.
