1. Gestalt Perception: A Game Designed to Explore Players' Gameplay Self-Efficacy and Anxiety Reflected in Their Learning Effects

Abstract

Gestalt perception relates to inferring a holistic scene from separate elements. Using this theory, an application game named Gestalt Puzzle was designed for students to play by recognizing a few parts of an image to reason the whole image of a particular object. Cognitive style can be divided into field independence (FI) and field dependence (FD) depending on individuals' ability to identify minor components of a whole image. To understand the gameplay learning effect, this study explored how players' field-independent cognitive style (FI-CS) interacted with their gameplay performance as the two types of cognitive styles were mediated by their self-efficacy and gameplay anxiety in a competition setting. Data from 112 sixth-grade students were collected for confirmatory factor analysis with structural equation modeling. First, participants completed a trial to familiarize themselves with the functions and mechanism of the game; second, all of the participants were divided into teams to compete against each other for 15 minutes by identifying Chinese paintings; they then completed a questionnaire. The results of this study indicated that FI-CS was positively related to gameplay self-efficacy, but negatively related to gameplay anxiety; perceived utility value could be positively predicted by gameplay self-efficacy, but negatively predicted by gameplay anxiety; and perceived value was positively related to game performance. Moreover, FI-CS was positively related to perceived value mediated by gameplay self-efficacy and gameplay anxiety. The implication of this study is that the Gestalt perception game may engage FI-CS learners with higher levels of gameplay self-efficacy, perceived value, and performance, but a lower level of gameplay anxiety. The present study findings imply that teachers can use Gestalt perception games to promote students' learning of Taiwanese culture.

Abstract

Specifying the primitive streak (PS) guides stem cell differentiation in vitro; however, much remains to be learned about the transcription networks that direct anterior and posterior PS cells (APS and PPS, respectively) to differentiate to distinct mesendodermal subpopulations. Here, we show that APS genes are predominantly induced in YAP1-/- human embryonic stem cells (hESCs) in response to ACTIVIN. This finding establishes the Hippo effector YAP1 as a master regulator of PS specification, functioning to repress ACTIVIN-regulated APS genes in hESCs. Moreover, transient exposure of wild-type hESCs to dasatinib, a potent C-SRC/YAP1 inhibitor, enables differentiation to APS-derived endoderm and cardiac mesoderm in response to ACTIVIN. Importantly, these cells can differentiate efficiently to normal beating cardiomyocytes without the cytoskeletal defect seen in YAP1-/- hESC-derived cardiomyocytes. Overall, we uncovered an induction mechanism to generate APS cells using a cocktail of ACTIVIN and YAP1i molecules that holds practical implications for hESC and induced pluripotent stem cell differentiation into distinct mesendodermal lineages.

6. Correlation Between Low Cytoplasmic Expression of XBP1 and the Likelihood of Surviving Hepatocellular Carcinoma

Abstract

BACKGROUND/AIM: Our objectives in this study were to (i) evaluate the clinical significance of X-box-binding protein 1 (XBP1) expression in cases of hepatocellular carcinoma (HCC) and (ii) assess the potential of XBP1 to be used as a prognostic biomarker. PATIENTS AND METHODS: The expression of XBP1 protein in 267 HCC tissue specimens was measured using immunohistochemistry in order to characterize the associations among XBP1 expression, clinicopathological factors and survival outcomes. Survival analysis using follow-up data was used to assess the prognostic value of XBP1 in cases of HCC. Immunohistochemistry revealed a significant decrease in cytoplasmic XBP1 protein expression in HCC tumor tissue. RESULTS: Immunoreactivity results showed that low cytoplasmic XBP1 expression was significantly associated with vascular invasion, as well as poor 5-year overall survival and long-term disease-specific (DSS) and disease-free (DFS) survival rates. Kaplan-Meier survival curves further confirmed a significant association between low cytoplasmic XBP1 protein expression and poor DSS and DFS. Univariate and multivariate analyses revealed that XBP1 expression, tumor differentiation, vascular invasion, tumor stage, and the rate of recurrence were linked to DSS, while low cytoplasmic XBP1 expression remained an independent predictor of poor DSS. Our analysis also revealed that XBP1 expression, tumor differentiation, vascular invasion, and T classification were linked to DFS, while low cytoplasmic XBP1 expression remained an independent predictor of poor DFS. CONCLUSION: Low cytoplasmic XBP1 protein expression may play an important role in the pathogenesis of HCC, which suggests that XBP1 could potentially be targeted to benefit therapeutic strategies for HCC.

Abstract

This paper proposes a novel passive micromixer design for mixing enhancement by forming a large three-dimensional (3-D) flow vortex in a counterflow microfluidic system. The counterflow fluids are self-driven by surface tension to perform mixing in an open chamber. The chamber design consists of two rectangular bars to house the chamber and to form two opening inlets from opposite directions. The best design is selected from various versions of mixing chambers. The mixing effectiveness is tremendously increased by folds of contacting surface between two fluids induced and enhanced due to the stretching of two fluid contacting interfaces by the formation of a 3-D large size vortex structure inside the mixing chamber itself with unaccountable numbers of fluid layers. Both numerical simulations and experiments are performed and compared to identify the design parameters for maximum utilization in this microfluidic system, such as the length of rectangular bar, microchannel wall height, and mixing chamber size. Compared to traditional micromixers operated by two-dimensional (2-D) vortex, this passive mixer can greatly enhance mixing efficiency and reduce mixing time by tenfold from around 10 s to less than 10 ms by 3-D effective chaotic flow structures in a more compact size. This mixing chamber is also suitable for an H-shape digital fluidic system for parallel mixing process in different mixing ratio simultaneously as a lab-on-a-chip system. 45. Severe ulcerative esophagitis and colitis induced by crizotinib therapy: A case report

Abstract

The expression of anaplastic lymphoma kinase (ALK), a target of tyrosine kinase is altered in several kinds of cancer, including in non‐small cell lung cancer (NSCLC). For this reason, the use of crizotinib to treat locally advanced or metastatic ALK‐positive NSCLC was approved by the US Food and Drug Administration in 2011. The reported adverse events during clinical trials with crizotinib included esophageal disorders in 11% of the patients, although none of them experienced severe events. Drug‐induced esophagitis or more severe ulcerations are considerably rare according to the available case reports. We report a patient who presented with severe esophageal ulcers after 2‐year treatment with crizotinib for metastatic, ALK‐positive NSCLC. These clinical conditions improved after education on proper eating habits and an additional prescription of a proton pump inhibitor. Several months later, the patient developed colitis with severe diarrhea, which was successfully controlled with mesalazine without reductions in the dosage of crizotinib. To the best of our knowledge, this is the first published case of successfully treated, consecutive esophageal and colonic ulcers related to crizotinib use. Crizotinib is a widely used targeted therapy for locally advanced or metastatic ALK‐positive NSCLC, and clinicians should keep in mind its severe drug‐related adverse effects such as esophagitis and colitis. Treatment with mesalazine, esomeprazole, and slightly decreasing the dose of crizotinib to 200 mg twice daily will be effective.
