The molecular connectivity index (MCI) is offered as a predictor of chemical biotransfer (BTF) of organic chemicals from cattle diet into beef and milk. MCI can predict BTFs more easily and quickly than a measure of lipid/water solubility. Statistical comparison of model reliability linked to Kow and MCI as BTF predictors proves that MCI can be a more reliable BTF predictor than Kow.

Abstract

Baseline levels of mutagenic compounds associated with airborne particles that are 10 micrometers or less (PM10) in aerodynamic diameter and using low-volume air samplers may be measured over a range of ambient levels. This is necessary in evaluating incremental exposure to mutagenic/carcinogenic compounds in indoor areas. Mutagenic compounds associated with particles found outdoors were found to be a vital source of mutagenic activity indoors. In addition, mutagenic activity is associated with the fine particle fraction.

17. Trophic transfer of paralytic shellfish toxins from clams (Ruditapes philippinarum) to gastropods (Nassarius festivus)

Abstract

A local strain of the dinoflagellate Alexandrium tamarense (ATC101), which predominantly produces C2 toxin, was fed to the clams (Ruditapes philippinarum) under laboratory conditions. Concentrations of paralytic shellfish toxins (PSTs) in the dosed clams were determined by High Performance Liquid Chromatographic (HPLC) analyses, and the clams were homogenized and then fed to the gastropods (Nassarius festivus). In the toxin accumulation phase, which lasted for 42 days, concentrations of PSTs increased in the snails gradually, reaching a maximum of 1.10 nmole g(-1) at the end of the exposure period. The toxin content of the homogenized clams (food) was 13.18 nmole g(-1), which was about 12-fold higher than the PST content in the snails. Between day 43 and day 82, the snails were fed with non-toxic clams, and this period represented the depuration phase. Accumulation and depuration rates of PSTs in the snails, N. festivus, were determined by fitting the experimental data to user-defined parameters program using a one-compartment model. Two different modeling approaches were used to derive the accumulation and depuration rates. The first approach is to derive both values from the data for the toxin uptake. The second approach is to derive deputation rate from the depuration data and then to derive uptake rate, allowing for toxin depuration, from the data for toxin uptake. The first approach yielded more consistent results for the toxin concentration at the end of the uptake period, when compared with the experimental data. The toxin uptake and depuration rates were 1.64 (pmole of toxin into snail per day) per (nmole g(-1) of toxin in food) and 0.06 +/- 0.02 day(-1) (mean +/- SE), respectively. The toxin profiles of snails were similar to the clams, but different from the algae. Besides C toxins (C1 and C2), dcGTX2 and dcGTX3 were also detected in both clams and snails. Abstract

Web-based learning has become one of the emerging directions in higher education all over the world. The Web offers a vast number of resources and enormous opportunities for more effective learning. Yet, Web-based learning is in its infancy in many parts of the world including Hong Kong and many courses are still using a mixture of face-to-face and Web-based instruction. In September 2001, the Web-based course “ESCE 500 Introductory Environmental Chemistry and Microbiology” was introduced into the curriculum of the MSc Environmental Science and Engineering Program (ESCE Program) of the Hong Kong University of Science and Technology (HKUST). This course serves to bridge the knowledge gap for those MSc students who did not have sufficient background in chemistry, biochemistry and microbiology. “ESCE 500” has been a collaboration with the University’s Center for Enhanced Learning & Teaching (CELT) and implemented as a password-protected course through the WebCT learning platform. Initially, all the course content was delivered online while the final examination was still in a written format. With our previous experiences and the students’ feedback collected from the course evaluations, we have developed “ESCE 500” and now it is the first completely Web-based course in HKUST. The Secure Online Assessment System (SOAS) designed by CELT was employed for the final examination. Our experiences echoed the advantages of Web-based learning such as relieving the students from temporal and physical constraints (the course content can be accessed anytime and anywhere), the effectiveness of interactive learning (such as the use of self-assessed quizzes, pop-ups, and video clips), and the potential of the Web that provided extensive information on related topics. Abstract

Thrombopoietin receptor (TPOR) is a member of the cytokine receptor superfamily expressed primarily on hematopoietic cells. TPOR plays an important role in regulating platelet production. Due to its low expression level in human tissue, studies on the biochemical and biophysical properties of TPOR have been limited. In the present study, an extracellular domain of recombinant human TPOR (rh TPOR-EN) was expressed in Escherichia colias inclusion bodies. Purification was achieved by metal chelated chromatography under denaturing condition and was refolded by gel filtration chromatography. Far UV circular dichroism spectroscopy and surface plasmon resonance experiments were performed to demonstrate that the protein was in a refolded state and could bind with its ligand. Thus, a production and purification scheme was developed by which sufficient quantities of rh TPOR-EN can be made available for biochemical and biophysical characterizations.
