Abstract

The study is focused on unconfined aquifer of Milan area, in the Po Plain; its vertical extension is about 80- 100 m depth..Using a water well data base TANGRAM, developed in the IDPA-CNR Milano, about 2200 data are stored, georeferenced and their stratigraphic logs are translated into alphanumeric codes. Textural well data have been translated and imported into GOCAD (Geological Object Computer Aided Design), where a 3-D grid of the aquifer volume (SGrid) is reconstructed with 3-D cells (Voxel). This grid is formed by 2.116.030 cells, 185 rows, 266 columns and 43 layers; each cell is a 250 m square, with a no constant thickness: it is between –1.8 m and 2-2 m. To each layers a permeability and porosity spatial value have been assigned and In MODFLOW the same dimensional stratigraphic grid have been created and the hydrogeological properties of each 43 layers imported: a value of top elevation, of bottom elevation, of permeability and of porosity define each cell. The model produces a piezometric distribution considering all input data used for the 3D reconstruction of real hydrogeological system and it reproduce correctly the aquifer gradient and flow. This is the first basically model our future research progress modelling in this area.

Abstract

GFLOW 2000, a groundwater flow simulation software, has been used to evaluate magnitude and direction of water exchanges between surface water and groundwater in a plain area (northern Italy). Piezometric levels for steady state conditions have been calculated starting from surface water levels measured in correspondence of 7 hydrometric station located along the considered rivers, using areal precipitation rates. Conjunctive surface water- groundwater solutions have also been computed using a stream network with calculated streamflow. Transitory condition scenarios have been simulated in order to test the code sensitivity in evaluating the effect of intense rainfall on piezometric levels (i.e. the flooding event of 13-16 October 2000) and on water exchanges within the system.

Abstract

Our paper is intended to assess the long-term impact of acid atmospheric deposition on the chemistry of ground waters in the Krušné hory Mts., and to explore to what extent forest cover participates in the development of chemical composition of relevant waters. The study was based on the results of long-term monitoring and assessment of mass flow in the Jezeří experimental catchment (1996-2007), and also on the results of investigation of a network of 26 experimental catchments in the Krušné hory Mts. that were affected by prolonged atmospheric deposition of high levels of sulfur and nitrogen. The monitoring of ground water quality and the health of forests at these catchments was not continuous. However, areal systematic sampling of waters characterizing the periods 1959-65, 1965-74, 1979-89 and 2000-02 enabled us to reconstruct the development of ground waters chemistry in the Krušné hory region during the last 40 years. The changes in chemical composition of waters were compared with data on atmospheric deposition. The obtained results proved the worst quality of ground waters to have occurred in the 1980s, which was manifested by the depletion of HCO3 contents, a decrease in pH and increase in SO4 and NO3 concentrations. Significant damage to the forest cover was recorded during this period of time. The results indicate that in the presence of a coniferous forest the negative impacts of acid atmospheric deposition on groundwater quality are greater. Simultaneously, the deterioration in the quality of groundwater, indicated by increased concentrations of aluminum, plays a part in damaging the health of the forest. [ABSTRACT FROM AUTHOR]

37. Detection of phase variation in expression of proteins involved in hemoglobin and hemoglobin-haptoglobin binding by nontypeable Haemophilus influenzae.

Abstract

Haemophilus influenzae can utilize different protein-bound forms of heme for growth in vitro. A previous study (I. Maciver, J. L. Latimer, H. H. Liem, U. Muller-Eberhard, Z. Hrkal, and E. J. Hansen. Infect. Immun. 64:3703-3712, 1996) indicated that nontypeable H. influenzae (NTHI) strain TN106 expressed a protein that bound hemoglobin-haptoglobin and was encoded by an open reading frame (ORF) that contained a CCAA nucleotide repeat. Southern blot analysis revealed that several NTHI strains contained between three and five chromosomal DNA fragments that bound an oligonucleotide probe for CCAA repeats. Three ORFs containing CCAA repeats were identified in NTHI strain N182; two of these ORFs were arranged in tandem. The use of translational fusions involving these three ORFs and the beta-lactamase gene from pBR322 revealed that these three ORFs, designated hgbA, hgbB, and hgbC, encoded proteins that could bind hemoglobin, hemoglobin-haptoglobin, or both compounds. Monoclonal antibodies (MAbs) specific for the HgbA, HgbB, and HgbC proteins were produced by immunizing mice with synthetic peptides unique to each protein. Both HgbA and HgbB were readily detected by Western blot analysis in N182 cells grown in the presence of hemoglobin as the sole source of heme, whereas expression of HgbC was found to be much less abundant than that of HgbA and HgbB. The use of these MAbs in a colony blot radioimmunoassay analysis revealed that expression of both HgbA and HgbB was subject to phase variation. PCR and nucleotide sequence analysis were used in conjunction with Western blot analyses to demonstrate that this phase variation involved the CCAA repeats in the hgbA and hgbB ORFs.

40. Binding of heme-hemopexin complexes by soluble HxuA protein allows utilization of this complexed heme by Haemophilus influenzae.

Abstract

Utilization of heme-hemopexin as a source of heme by Haemophilus influenzae type b is dependent on expression by this bacterium of the 100-kDa HxuA protein, which is both present on the bacterial cell surface and released into the culture supernatant (L. D. Cope, R. Yogev, U. Muller-Eberhard, and E. J. Hansen, J. Bacteriol. 177:2644-2653, 1995). Radioimmunoprecipitation analysis showed that the soluble HxuA protein present in H. influenzae type b culture supernatant bound heme-hemopexin complexes in solution. An isogenic H. influenzae type b hxuA mutant was unable to utilize soluble heme-hemopexin complexes for growth in vitro unless soluble HxuA protein was provided exogenously. Soluble HxuA protein secreted by a nontypeable H. influenzae strain also allowed growth of this H. influenzae type b hxuA mutant. These results indicated that the heme present in heme-hemopexin complexes is rendered accessible to H. influenzae when these complexes are bound by the soluble HxuA protein.

Abstract

Mean diameter of nucleolar bodies (nucleoli without the perinucleolar chromatin) per cell was studied in human leukemic myeloblasts represented by K 562 and Kasumi 1 cell lines which originated from chronic and acute myeloid leukaemia. The measurement of mean diameter of nucleolar bodies in specimens stained for RNA was very simple. Such approach eliminated the variability of the perinucleolar chromatin discontinuous shell which might influence the measured nucleolar size as suggested by earlier studies. Ageing of K 562 myeloblasts produced a significant decrease of cells in S+G2 phase of the cell cycle accompanied by a significant reduction of mean diameter of nucleolar bodies (MDNoBs) per cell. In contrast, treatment of Kasumi 1 myeloblasts with histone deacetylase inhibitor - Trichostatin A - produced a large incidence of resistant cells in S+G2 phase which were characterised by a large increase of MDNoBs. Thus, MDNoBs in leukemic myeloblasts might be a helpful tool to estimate the incidence of cells in the S+G2 phase at the single cell level in smear preparations when the number of cells is very small.
