Abstract

This article discusses the genetic determinants of atrial septal defect (ASD), a common congenital heart disease. The study identified five rare genetic variants in the FOXC1 gene in both familial and sporadic cases of ASD. Functional analysis confirmed that two of these variants, R173H and P79L, led to significant changes in FOXC1 transcriptional activity. Additionally, a mouse model with the R173H variant showed a thinning of the atrial septum, similar to the phenotype observed in humans. These findings provide new insights into the genetic causes of ASD and may contribute to improved diagnosis and counseling for affected individuals. [Extracted from the article]

Abstract

Trace elements selenium (Se) and cobalt (Co) are essential in the human body, and a correlation between Se and cardiac surgery has been suggested. We investigated the plasma concentrations of Se and Co during and after coronary artery bypass grafting (CABG) surgery under cardiopulmonary bypass (CPB). From December 2019 to January 2020, preoperative plasma samples from isolated first-time CABG patients (n=20; 10 males, 10 females) were prospectively collected post-anesthesia and before CPB (T1), 45 min after CPB started (T2), 90 min after CPB started (T3), and postoperative days 1 (T4), and day 4 (T5). Abstract

Hydrogen sulfide (H2S) represents the third and the youngest member of the gaseous transmitters family. The dominant effect of H2S on isolated vessels is vasodilation. As the mechanism of H2S-induced relaxation in human vessels remains unclear, the present study aimed to investigate the effects of H2S donor, sodium hydrosulfide (NaHS), on isolated human saphenous vein (HSV) and to determine the mechanism of action. Our results showed that NaHS (1 µM–3 mM) induced a concentration-dependent relaxation of endothelium-intact HSV rings pre-contracted by phenylephrine. Pre-treatment with L-NAME, ODQ and KT5823 significantly inhibited NaHS-induced relaxation, while indomethacin induced partial inhibition. Among K+ channel blockers, the combination of apamin and TRAM-34 significantly affected the relaxation produced by NaHS, while iberiotoxin and glibenclamide only reduced maximal relaxation of HSV. NaHS partially relaxed endothelium-intact rings pre-contracted by high K+, as well as phenylephrine-contracted rings in the presence of nifedipine. Additionally, the incubation of HSV rings with NaHS increased NO production. These results demonstrate that NaHS produces the concentration- and endothelium-dependent relaxation of isolated HSV. Vasorelaxation to NaHS probably involves activation of NO/cGMP/PKG pathway and partially prostacyclin. In addition, different K+ channels subtypes, especially SKCa and IKCa, as well as BKCa and KATP channels in high concentrations of NaHS, probably participate in the NaHS-induced vasorelaxation.

Abstract

The transient electromagnetic forward modeling of volcanogenic massive sulfide (VMS) ore deposits is to calculate the eddy currents electromagnetic response of a three dimensional body in the full space of deep-sea environment. We simulated the three dimensional transient electromagnetic response of the VMS deposits using full-domain vector finite element method. The ore body was discretized with brick rectangular elements. The finite element equation was deduced in frequency domain by employing Galerkin procedure, and the conversion to time domain was by inverse Fourier transform. We confirmed the validity of the full-domain vector finite element algorithm by comparing simulated results with analytical solutions of double half-space model. The results have a good agreement with each other, which indicates that the vector finite element method is capable of solving whole space problem. In order to demonstrate the ability of the numerical method in calculating the response of VMS deposits containing complex boundary conditions, we compared vector finite element solution of a three dimensional electrical model with physical experiment results according to electromagnetic physical scale modeling rules. The comparison suggests that for the complex electromagnetic boundary of seawater, ore body and country rocks, the transient electromagnetic response of VMS deposits calculated by full-domain vector finite element method has same features with the physical scale modeling result. The vector finite element method is simple and its results are precise with obvious and clear anomaly response.

Abstract

Transient electromagnetic method (TEM) is a commonly-used geophysical method of measuring the secondary electromagnetic field induced by transient pulse sources. Because of the inherent mutual-induction between TX and RX coils, the measured field always contains the mutual-induction field, which becomes stronger when TX coil becomes smaller and closer to RX coil. It influences the near-surface TEM systems, which utilize small TX coils to effectively resolve the near surface explorations. The mixture of the mutual-induction field with the secondary field at early period leads to a blind region in the shallow surface in TEM exploration. To solve this problem, we applied two concentric and parallel current coils in the TEM system. They are opposing coils, which means that the two coils are physically same, but the currents in them are of equal value and in reverse directions. The RX coil is equidistant to the two opposing coils. By means of this special arrangement, the mutual-induction field received at the RX coil is zero, which means that the mutual induction effect is efficiently eliminated, and pure secondary field of the conductive underground will be measured. It is proved by theory and model calculation that opposing coils transient electromagnetic method has advantages in lateral resolution. It is quite suitable for small coil TEM system aiming at shallow subsurface detection.

45. Effect of Benidipine in Human Internal Mammary Artery and Clinical Implications

Abstract

Background. Graft spasm remains challenging in coronary artery bypass grafting (CABG). Calcium antagonists are commonly used in patients with coronary artery disease. This study investigated the inhibitory effect of third-generation dihydropyridine calcium channel antagonist benidipine on the vasoconstriction induced by various vasoconstrictors in the human internalmammary artery(IMA). Methods. Isolated human IMA rings (N = 65, taken from 37 patients undergoing CABG) were studied in a myograph in 2 ways: the relaxing effect of benidipine on vasoconstrictor-induced precontraction by KCl and U46619 and the depressing effect of benidipine at plasma concentrations on the contraction. Enzyme-linked immunosorbent assay (ELISA) was used to measure the change of the protein related to the L-type calcium channel. Results. Benidipine caused more relaxation in KCl-contracted (86.7% +/- 3.3%; n = 12) than in U46619-contracted (63.8% +/- 5.3%; n = 8; p lt 0.001) IMA rings. Pretreatment of IMA with plasma concentrations of benidipine (-6.92 log M) significantly depressed subsequent contraction by KCl (from 17.3 +/- 2.7 mN to 7.4 +/- 1.2 mN; n = 6; p lt 0.05) but did not significantly affect the contraction caused by U46619. Benidipine also caused a decrease of caveolin (CaV) 1.2 protein content (0.55 +/- 0.02 versus 0.63 +/- 0.02 mg/mL; p lt 0.05). Conclusions. We conclude that in human IMA, the third-generation dihydropyridine calcium channel antagonist benidipine has a potent inhibitory effect on the vasoconstriction mediated by a variety of vasoconstrictors. Use of benidipine in patients undergoing CABG may provide vasorelaxant or antispastic effects in the grafts.

Abstract

Objectives: The 22q11.2 deletion syndrome is considered the most frequent chromosomal microdeletion syndrome in humans and the second leading chromosomal cause of congenital heart disease (CHD). We aimed to identify the prevalence and the detailed genetic characterisation of 22q11.2 region in children with CHD including simple defects and to explore the genotype-phenotype relationship between deletion/amplification type and clinical data.Methods: Patients with CHD for surgery were screened by multiplex ligation-dependent probe amplification and capillary electrophoresis methods. Universal Probe Library technology was applied for validation.Results: In 354 patients with CHD, 40 (11.3%) carried different levels of deletions/amplifications at the 22q11.2 region with various phenotypes. The affected genes at this region include CDC45 (15 patients), TBX1 (8), USP18 (8), RTDR1 (7), SNAP29 (6), TOP3B (6), ZNF74 (4) and other genes with less frequency. Among those, two patients carried 3 Mb typically deleted region from CLTCL1 to LZTR1 (low copy repeats A-D) or 1.5 Mb deletions from CLTCL1 to MED15 (low copy repeats A-C). Clinical facial manifestations were found in 12 patients.Conclusions: This study revealed an unexpected high prevalence of chromosome 22q11.2 variations in patients with CHD even in simple defects. The genotype-phenotype relationship analysis suggests that genetic detection of 22q11.2 may become necessary in all patients with CHD and that detection of unique deletions or amplifications may provide useful insight into personalised management in patients with CHD.
