The fruit-piercing moth (FPM) Eudocima phalonia is widely present in the tropics, causing major damages to the citrus industry. In its ancestral distribution range, e.g. Australia, FPM develops on vines from the Menispermaceae family. But in more recently colonized islands, e.g. New Caledonia, FPM populations have shifted to new host plants of the Fabaceae family, Erythrina spp.. To understand this host shift, we studied the chemical ecology of FPM as a mechanism driving host plant acceptance. We collected volatile headspace samples of Erythrina species and Menispermaceae and compared their chemical spectra. We assessed the electrophysiological responses of FPM populations from the two countries to the plant chemical extracts and identified bioactive compounds. The volatile profiles from each species were quite different between and within each plant family. However, five compounds common across the two families triggered electrophysiological responses in both FPM populations. Those common bioactive compounds could have facilitated the host shift to completely different plant family. Furthermore, the diverging history between the two FPM populations may explain differences in electrophysiological sensitivity to other specific compounds. Diplofornicis scoliius gen. and sp. nov. is described from adult female and male mites riding on the scoliid wasp Aelocampsomeris variegata (Fabricius). These wasps represent an unusual phoretic carrier for Diplogyniidae and these mites present unusual features such as a highly humped idiosoma, raised ventromedial region of the sternal and ventrianal shield, and spiny processes on the female’s latigynal shields. We discuss possible generic relationships and regard the Neotropical genera Brachysternopsis Hunter, Brachysternum Trägårdh, and Crenamargo Hicks as the most likely closest relatives. The emergence of new diseases is an urgent concern, but hosts can also vary in resistance to pathogens that are novel to them, facilitating evolutionary rescue. However, little is known about the genetic source for polymorphic resistance to novel pathogens or its relationship to defences against endemic diseases. With anther‐smut disease from wild plant populations, we used selection experiments and genetic analyses to show that resistances to novel and endemic pathogens are genetically independent, despite being positively correlated in nature. Moreover, novel‐pathogen resistance presented a much simpler genetic basis and more rapid response to selection. We demonstrate that polymorphic resistance to a newly introduced disease is genetically determined and not an extension of defences against the related endemic pathogen, challenging the conventional view of nonhost resistance. We recently found a population of the leaf‐footed cactus bug, Narnia femorata (Hemiptera: Coreidae), a historical cactus specialist, living and feeding on Cirsium thistle. We also found adults breeding and males using their enlarged hind legs (i.e., weapons) in male–male combat on thistle. When we compared this thistle population with a population feeding on cactus, we found that both populations had similar body and weapon sizes as well as weapon composition. However, the population living on thistle had longer mouthparts than the population found on cactus, although this difference only occurred at larger body sizes. This difference in adult mouthpart size is likely a result of the juvenile rearing environment (i.e., thistle or cactus). However, genetic differences may also affect this trait. Morphological plasticity may be induced by altering host plants and commonly it is sex‐specific in phytophagous insects. It is hypothesized that stress in insects caused by a host shift leads to morphological changes and developmental destabilization, which may be identified by fluctuating asymmetry. We performed reciprocal transplant experiments in eight replicated populations of the seed beetle Acanthoscelides obtectus (Say) (Coleoptera: Chrysomelidae, Bruchinae) adapted to common bean (Phaseolus vulgaris L., Fabaceae) as its ancestral host and chickpea (Cicer arietinum L., Fabaceae) as a suboptimal host. Using methods of geometric morphometrics we assessed the effects of the short‐ and long‐term host shift on size and shape variation of beetles of both sexes. We also tested the hypothesis that fluctuating asymmetry is lower when the beetles develop within seeds of the ancestral host. Our results showed that females respond more plastically related to their body size and shape of the abdomen, particularly during a short‐term host shift, suggesting that females have a greater importance in maintaining the population on a new host. The level of fluctuating asymmetry is lowest in males that have evolved for generations on bean indicating that they have the most canalized development, so we suggested that symmetry in A. obtectus is maintained by sexual selection. Even if the beetles are adapted to a suboptimal host over many generations, they stabilize their development after returning to the ancestral host. The rabies virus (RABV) is the exclusive lyssavirus affecting both wild and domestic mammalian hosts in the Americas, including humans. Additionally, the Americas stand out as the sole region where bat rabies occurs. While carnivore rabies is being increasingly managed across the region, bats are emerging as significant reservoirs of RABV infection for humans and domestic animals. Knowledge of the bat species maintaining rabies and comprehending cross-species transmission (CST) and host shift processes are pivotal for directing surveillance as well as ecological research involving wildlife reservoir hosts. Prior research indicates that bat RABV CST is influenced by host genetic similarity and geographic overlap, reflecting host adaptation. In this study, we compiled and analyzed a comprehensive nucleoprotein gene dataset representing bat-borne RABV diversity in Argentina and the broader Americas using Bayesian phylogenetics. We examined the association between host genus and geography, finding both factors shaping the global phylogenetic structure. Utilizing a phylogeographic approach, we inferred CST and identified key bat hosts driving transmission. Consistent with CST determinants, we observed monophyletic/paraphyletic clustering of most bat genera in the RABV phylogeny, with stronger CST evidence between host genera of the same family. Background: Throughout the Americas, Lyssavirus rabies (RV) perpetuates as multiple variants among bat and mesocarnivore species. Interspecific RV spillover occurs on occasion, but clusters and viral host shifts are rare. The spillover and host shift of a big brown bat (Eptesicus fuscus) RV variant Ef-W1 into mesocarnivores was reported previously on several occasions during 2001–2009 in Flagstaff, Arizona, USA, and controlled through rabies vaccination of target wildlife. During autumn 2021, a new cluster of Ef-W1 RV cases infecting striped skunks (Mephitis mephitis) was detected from United States Department of Agriculture enhanced rabies surveillance in Flagstaff. The number of Ef-W1 RV spillover cases within a short timeframe suggested the potential for transmission between skunks and an emerging host shift. Materials and Methods: Whole and partial RV genomic sequencing was performed to evaluate the phylogenetic relationships of the 2021–2023 Ef-W1 cases infecting striped skunks with earlier outbreaks. Additionally, real-time reverse-transcriptase PCR (rtRT-PCR) was used to opportunistically compare viral RNA loads in brain and salivary gland tissues of naturally infected skunks. Results: Genomic RV sequencing revealed that the origin of the 2021–2023 epizootic of Ef-W1 RV was distinct from the multiple outbreaks detected from 2001–2009. Naturally infected skunks with the Ef-W1 RV showed greater viral RNA loads in the brain, but equivalent viral RNA loads in the mandibular salivary glands, compared to an opportunistic sample of skunks naturally infected with a South-Central skunk RV from northern Colorado, USA. Conclusion: Considering a high risk for onward transmission and spread of the Ef-W1 RV in Flagstaff, public outreach, enhanced rabies surveillance, and control efforts, focused on education, sample characterization, and vaccination, have been ongoing since 2021 to mitigate and prevent the spread and establishment of Ef-W1 RV in mesocarnivores. While there are documented host shifts in many bacterial plant pathogens, the genetic foundation of host shifts is largely unknown. Xylella fastidiosa is a bacterial pathogen found in over 600 host plant species. Two parallel host shifts occurred-in Brazil and Italy-in which X. fastidiosa adapted to infect olive trees, whereas related strains infected coffee. Using 10 novel whole-genome sequences from an olive-infecting population in Brazil, we investigated whether these olive-infecting strains diverged from closely related coffee-infecting strains. Several single-nucleotide polymorphisms, many derived from recombination events, and gene gain and loss events separated olive-infecting strains from coffee-infecting strains in this clade. The olive-specific variation suggests that this event was a host jump with genetic isolation between coffee- and olive-infecting X. fastidiosa populations. Next, we investigated the hypothesis of genetic convergence in the host shift from coffee to olive in both populations (Brazil and Italy). Each clade had multiple mutations and gene gain and loss events unique to olive, yet no overlap between clades. Using a genome-wide association study technique, we did not find any plausible candidates for convergence. Overall, this work suggests that the two populations adapted to infect olive trees through independent genetic solutions.

Parasites exhibit high diversity in various ecosystems. Habitat selection has played an important role in the diversification of parasites by facilitating intra-host speciation and repeated shifts to a single host species by multiple parasite taxa. In this study, we investigated habitat selection in two gastropod species, Melanella cf. acicula and Peasistilifer nitidula (Caenogastropoda: Vanikoroidea: Eulimidae), that are parasitic on the black sea cucumber Holothuria atra in southwestern Japan. The number and position of parasites were observed for 335 hosts; in total, 89 individuals of M. cf. acicula and 194 of P. nitidula were found. Melanella cf. acicula exhibited strong habitat selection, with 70 individuals (78.7%) attached around the host's mouth, whereas P. nitidula had relatively low infection site specificity. This suggests that the two parasite species have different feeding habits. Molecular phylogenetic analysis verified their non-monophyletic relationship, indicating that they likely made independent host shifts to H. atra. The observed difference in habitat preference, as well as the abundance of suitable habitats, may have contributed to the repeated host shifts and sympatric distribution of M. cf. acicula and P. nitidula. Our phylogenetic analysis also revealed the non-monophyly of eulimids parasitising holothuroids, as in asteroid and ophiuroid parasitic taxa. [ABSTRACT FROM AUTHOR]

The parasitoid way of life (parasitizing and finally killing a single arthropod host) is one of the most successful lifeways in the animal kingdom, sparking an explosive diversification and accompanying numerous parasitoid strategies in insects, especially in the order Hymenoptera. Amongst parasitoid wasps, the Polysphincta group of genera has evolved a highly distinctive parasitoid mode of life, as solitary koinobiont ectoparasitoids of spiders (Chelicerata: Araneae). Some species of polysphinctine wasps have a remarkable ability to control spiders' web-building behaviour (host web manipulation) to protect the vulnerable wasp cocoons. The group currently consists of 25 genera and 294 extant species worldwide, with 14 genera known to manipulate their host spiders. This study reviews the current species composition, distribution, host utilization and biology of all genera belonging to the Polysphincta group and their ancestral genera of spider egg mass pseudo-parasitoids, highlighting specific offensive approaches for subjugating spider hosts for oviposition and web manipulation against specific spider hosts. There must still be many more unique behaviours to be discovered, given that the life histories of several polysphinctine genera are poorly known or unknown. A tidy correspondence between lineages of polysphinctines and host spiders was recognized, implying the evolutionary history of polysphinctines. Based on the integrated information on behavioural data and host utilization, we suggest some hypotheses determining triggers for host shifts and discuss the possibility of adaptive radiation driven by divergent natural selection on host differentiation. Abstract The Microsporum canis complex consists of one zoophilic species, M. canis, and two anthropophilic species, M. audouinii and M. ferrugineum. These species are the most widespread zoonotic pathogens causing dermatophytosis in cats and humans worldwide. To clarify the evolutionary relationship between the three species and explore the potential host shift process, this study used phylogenetic analysis, population structure analysis, multispecies coalescent analyses, determination of MAT idiomorph distribution, sexual crosses, and macromorphology and physicochemical features to address the above questions. The complex of Microsporum canis, M. audouinii and M. ferrugineum comprises 12 genotypes. MAT1-1 was present only in M. canis, while the anthropophilic entities contained MAT1-2. The pseudocleistothecia were yielded by the mating behaviour of M. canis and M. audouinii. Growth rates and lipase, keratinolysis and urea hydrolytic capacities of zoophilic M. canis isolates were all higher than those of anthropophilic strains; DNase activity of M. ferrugineum exceeded that of M. canis. The optimum growth temperature was 28 °C, but 22 °C favoured the development of macroconidia. Molecular data, physicochemical properties and phenotypes suggest the adaptation of zoophilic M. canis to anthropophilic M. ferrugineum, with M. audouinii in an intermediate position.

Abstract In phytophagous insects, adaptation to a new host is a dynamic process, in which early and later steps may be underpinned by different features of the insect genome. Here, we tested the hypothesis that early steps of this process are underpinned by a shift in gene expression patterns. We set up a short-term artificial selection experiment (10 generations) for the use of an alternative host (Cicer arietinum) on populations of the bean beetle Zabrotes subfasciatus. Using Illumina sequencing on young adult females, we show the selected populations differ in the expression of genes associated to stimuli, signalling, and developmental processes. Particularly, the “C. arietinum” population shows upregulation of histone methylation genes, which may constitute a strategy for fine-tuning the insect global gene expression network. Using qPCR on body regions, we demonstrated that the “Phaseolus vulgaris” population upregulates the genes polygalacturonase and egalitarian and that the expression of an odorant receptor transcript variant changes over generations. Moreover, in this population we detected the existence of vitellogenin (Vg) variants in both males and females, possibly harbouring canonical reproductive function in females and extracellular unknown functions in males. This study provides the basis for future genomic investigations seeking to shed light on the nature of the proximate mechanisms involved in promoting differential gene expression associated to insect development and adaptation to new hosts.

The study attempted to understand the effect of the host plants on the call parameters of native tree cricket, Oecanthus indicus (Order: Orthoptera, Sub-order: Ensifera, Family: Gryllidae) while calling from native (Justicia adhatoda) and non-native host plant species (Lantana camara and Hyptis suaveolens). The study was conducted at four locations across India. Calls of O. indicus were recorded on these host plants in the field and spectral and temporal parameters of calls were analysed. The results suggested that the peak frequency varied among the two non-native plant species while the difference in temporal pattern between the native and non-native host plants was observed only in the syllable period. The study also quantified the choice of calling positions of insects from the three-host species. The native O. indicus chose non-native H. suaveolens leaves extensively as a preferable site to baffle (37%). Differences in the call parameters and choice of the host plant by insects may ultimately affect the preference and performance of insects on invasive plants. Branched broomrape, Phelipanche ramosa (L.) Pomel, is a globally distributed parasitic weed of economic importance. In Europe, where it is native, it can infest several crops, notably tomato, tobacco, and hemp. In western France, it has recently adapted to a new host crop, oilseed rape, causing substantial damage. The aim of this study was to investigate the evolutionary relationships and genetic differentiation among P. ramosa populations infesting different hosts. We collected 1611 P. ramosa samples from 109 fields cultivated with six different crops (oilseed rape, tobacco, hemp, tomato, lentil, and celery) and distributed among six European countries. All samples were genotyped for ten microsatellite loci and a subset of samples was sequenced for two nuclear genes and two chloroplast genes. Genetic differentiation among populations was high (FST = 0.807) and mainly driven by differentiation among different host crops, with no significant geographic structure. Genetic structure analysis identified up to seven biologically meaningful clusters that matched with host crops of origin. Reconstructed networks of sequence haplotypes and multilocus SSR genotypes showed a large genetic divergence between samples collected on oilseed rape and samples collected on other crops. The phylogeny inferred from DNA sequences placed samples collected from oilseed rape as a basal lineage. Approximate Bayesian Computations were used to compare different evolutionary scenarios of divergence among the three main genetic clusters, associated, respectively, with oilseed rape, tobacco, and hemp as host crops. The best‐supported scenario indicated that P. ramosa infesting oilseed rape derived recently from an ancient, unknown lineage. Our results suggest that a more complete description of the genetic diversity of P. ramosa is still needed to uncover the likely source of the recent adaptation to oilseed rape and to anticipate future new host shifts. Biotic interactions can modulate the responses of organisms to environmental stresses, including diet changes. Gut microbes have substantial effects on diverse ecological and evolutionary traits of their hosts, and microbial communities can be highly dynamic within and between individuals in space and time. Modulations of the gut microbiome composition and their potential role in the success of a species to maintain itself in a new environment have been poorly studied to date. Here we examine this question in a large wood-boring beetle Cacosceles newmannii (Cerambycidae), that was recently found thriving on a newly colonized host plant. Using 16S metabarcoding, we assessed the gut bacterial community composition of larvae collected in an infested field and in "common garden" conditions, fed under laboratory-controlled conditions on four either suspected or known hosts (sugarcane, tea tree, wattle, and eucalyptus). We analysed microbiome variation (i.e. diversity and differentiation), measured fitness-related larval growth, and studied host plant lignin and cellulose contents, since their degradation is especially challenging for wood-boring insects. We show that sugarcane seems to be a much more favourable host for larval growth. Bacterial diversity level was the highest in field-collected larvae, whereas lab-reared larvae fed on sugarcane showed a relatively low level of diversity but very specific bacterial variants. Bacterial communities were mainly dominated by Proteobacteria, but were significantly different between sugarcane-fed lab-reared larvae and any other hosts or field-collected larvae. We identified changes in the gut microbiome associated with different hosts over a short time frame, which support the hypothesis of a role of the microbiome in host switches. [ABSTRACT FROM AUTHOR]

Dendrolimus houi Lajonquière is one of the most prevalent pine caterpillars in southern China, causing overwhelming forest infestation. It can overcome the negative impact of host shift from the original host, Cryptomeria fortune, to other tree species to complete its life cycle. In this work, D. houi larvae (1st to 3rd instar), originally feeding on C. fortunei needles, were used to determine the changes in the growth and development after they moved to needles of Cupressus funebris and Pinus yunnanensis during their 4th to 5th instar. Metabonomics and transcriptomics were conducted to evaluate the effects of the host shift on metabolite accumulation and gene expression of D. houi larvae. The results showed that the host shift significantly inhibited the pest growth and development by extending developmental duration and minifying the body length, body weight and head shell width. Besides, we found that the host shift influenced steroid hormone biosynthesis in D. houi larvae, which might lead to their abnormal development. The results may help to understand the response of D. houi larvae to host shift, and their adaptation mechanism to different hosts during multiple life cycles, providing a new plantation mode for mixed forests to suppress D. houi as well. Introduction: Host shift of parasites may have devastating effects on the novel hosts. One remarkable example is that of the ectoparasitic mite Varroa destructor, which has shifted its host from Eastern honey bees (Apis cerana) to Western honey bees (Apis mellifera) and posed a global threat to apiculture. Objectives: To identify the genetic factors underlying the reproduction of host-shifted V. destructor on the new host. Methods: Genome sequencing was conducted to construct the phylogeny of the host-shifted and non-shifted mites and to screen for genomic signatures that differentiated them. Artificial infestation experiment was conducted to compare the reproductive difference between the mites, and transcriptome sequencing was conducted to find differentially expressed genes (DEGs) during the reproduction process. Results: The host-shifted and non-shifted V. destructor mites constituted two genetically distinct lineages, with 15,362 high-FST SNPs identified between them. Oogenesis was upregulated in host-shifted mites on the new host A. mellifera relative to non-shifted mites. The transcriptomes of the host-shifted and non-shifted mites differed significantly as early as 1h post-infestation. The DEGs were associated with nine genes carrying nonsynonymous high-FST SNPs, including mGluR2-like, Lamb2-like and Vitellogenin 6-like, which were also differentially expressed, and eIF4G, CG5800, Dap160 and Sas10, which were located in the center of the networks regulating the DEGs based on protein-protein interaction analysis. Conclusions: The annotated functions of these genes were all associated with oogenesis. These genes appear to be the key genetic determinants of the oogenesis of host-shifted mites on the new host. Further study of these candidate genes will help elucidate the key mechanism underlying the success of host shifts of V. destructor.

Expansion of the host range in phytophagous insects, followed by the specialisation on novel hosts, encompasses changes in many aspects of insects' behaviour, physiology, and the interaction between their life‐history features.Here, we analyse the roles of insects' developmental plasticity in the process of host shift. Using laboratory populations of the seed beetle (Acanthoscelides obtectus), which have evolved on both optimal (common beans) and suboptimal (chickpea) plant hosts for more than 35 years, we experimentally replicated the process of host shift and analysed the patterns of short‐term and long‐term life‐history responses to host variation.In order to test whether selection for increased plasticity has an effect on host shifting processes, we used existing bean and chickpea adapted populations to establish new populations in which the host plant offered for insect development was changed each generation (for 13 generations). To test the potential for a short‐term plastic response, beetles from each laboratory population were raised on both hosts for one generation.Results showed that, in contrast to the populations that evolved on beans, which maintained high levels of developmental plasticity, long‐term host switching to chickpeas was accompanied with specialisation of pre‐adult viability with a simultaneous increase in fecundity. Populations evolved on alternate plant hosts that revealed similar plasticity patterns as their ancestral populations.These results suggest that short‐term plastic responses could determine the paths of long‐term evolution of life‐history plasticity. Host‐shifts, where pathogens jump from an ancestral host to a novel host, can be facilitated or impeded by standing variation in disease resistance, but only if resistance provides broad‐spectrum general resistance against multiple pathogen species. Host resistance comes in many forms and includes both general resistance, as well as specific resistance, which may only be effective against a single pathogen species or even genotype. However, most evolutionary models consider only one of these forms of resistance, and we have less understanding of how these two forms of resistance evolve in tandem. Here, we develop a model that allows for the joint evolution of specific and general resistance and asks if the evolution of specific resistance drives a decrease in the evolution of general resistance. We also explore how these evolutionary outcomes affect the risk of foreign pathogen invasion and persistence. We show that in the presence of a single endemic pathogen, the two forms of resistance are strongly exclusionary. Critically, we find that specific resistance polymorphisms can prevent the evolution of general resistance, facilitating the invasion of foreign pathogens. We also show that specific resistance polymorphisms are a necessary condition for the successful establishment of foreign pathogens following invasion, as they prevent the exclusion of the foreign pathogen by the more transmissible endemic pathogen. Abstract Branched broomrape, Phelipanche ramosa (L.) Pomel, is a globally distributed parasitic weed of economic importance. In Europe, where it is native, it can infest several crops, notably tomato, tobacco, and hemp. In western France, it has recently adapted to a new host crop, oilseed rape, causing substantial damage. The aim of this study was to investigate the evolutionary relationships and genetic differentiation among P. ramosa populations infesting different hosts. We collected 1611 P. ramosa samples from 109 fields cultivated with six different crops (oilseed rape, tobacco, hemp, tomato, lentil, and celery) and distributed among six European countries. All samples were genotyped for ten microsatellite loci and a subset of samples was sequenced for two nuclear genes and two chloroplast genes. Genetic differentiation among populations was high (FST = 0.807) and mainly driven by differentiation among different host crops, with no significant geographic structure. Genetic structure analysis identified up to seven biologically meaningful clusters that matched with host crops of origin. Reconstructed networks of sequence haplotypes and multilocus SSR genotypes showed a large genetic divergence between samples collected on oilseed rape and samples collected on other crops. The phylogeny inferred from DNA sequences placed samples collected from oilseed rape as a basal lineage. Approximate Bayesian Computations were used to compare different evolutionary scenarios of divergence among the three main genetic clusters, associated, respectively, with oilseed rape, tobacco, and hemp as host crops. The best‐supported scenario indicated that P. ramosa infesting oilseed rape derived recently from an ancient, unknown lineage. Our results suggest that a more complete description of the genetic diversity of P. ramosa is still needed to uncover the likely source of the recent adaptation to oilseed rape and to anticipate future new host shifts.

Virus spillover from managed to unmanaged bees and vice versa may be one mechanism driving colony losses of the former and declines of the latter. There is clear evidence that the ubiquitous Deformed wing virus (DWV) is a major driver of honey bee (Apis mellifera) colony mortality. Although DWV has been detected in the solitary bee Osmia bicornis, data on DWV infectivity and virulence from solitary bees are scarce. Here, we used microinjection to investigate whether DWV genotype A (DWV-A) obtained from honey bees can replicate in O. bicornis. DWV-A titers and intermediate strand analyses suggest that DWV-A does not replicate in O. bicornis and thus is probably not infectious for this solitary bee species. Interestingly, the data demonstrate that DWV-A recovered from O. bicornis 16 days post-microinjection remains infectious for A. mellifera. Therefore, despite the lack of apparent virulence of DWV in this solitary bee species, O. bicornis has the potential to act as a virus spillover host and may contribute to increased colony losses of managed honey bees and declines in populations of other managed or unmanaged bee species.

Simple Summary: Pentastiridius leporinus is a planthopper that transmits two bacterial pathogens to sugar beet plants: Arsenophonus and Stolbur. These bacteria cause an economically important disease known as syndrome basses richesses (SBR), characterized by deformed yellow leaves and low beet yields. Having observed potato fields in Germany infested with planthoppers and showing signs of leaf yellowing, we set out to identify the planthoppers and any transmitted pathogens. We identified P. leporinus by comparisons of COI and COII partial sequences and detected both bacterial pathogens in planthoppers, as well as in beet and potato plants. We found that P. leporinus nymphs and adults can transmit the bacteria and that long, warm summers allow two generations of planthoppers to breed, probably increasing the population size and threat of disease transmission in the following year. We conclude that the planthopper P. leporinus has expanded its host range to potato, and that further studies are needed, to facilitate the development of control strategies that minimize losses in sugar beet and potato crops. Having observed potato fields in Germany infested with cixiid planthoppers and showing signs of leaf yellowing, we used morphological criteria and COI and COII as molecular markers, to identify the planthoppers (adults and nymphs) primarily as P. leporinus. We analyzed planthoppers, potato tubers, and sugar beet roots and detected both pathogens in all sample types, confirming that P. leporinus adults and nymphs can transmit the bacteria. This is the first time that P. leporinus has been shown to transmit Arsenophonus to potato plants. We also found that two generations of P. leporinus were produced in the warm summer of 2022, which will probably increase the pest population size (and thus the prevalence of SBR) in 2023. We conclude that P. leporinus has expanded its host range to potato, and can now utilize both host plants during its developmental cycle, a finding that will facilitate the development of more efficient control strategies. To identify the genetic factors underlying the reproduction of host-shifted V. destructor on the new host. Genome sequencing was conducted to construct the phylogeny of the host-shifted and non-shifted mites and to screen for genomic signatures that differentiated them. Artificial infestation experiment was conducted to compare the reproductive difference between the mites, and transcriptome sequencing was conducted to find differentially expressed genes (DEGs) during the reproduction process. The host-shifted and non-shifted V. destructor mites constituted two genetically distinct lineages, with 15,362 high- F ST SNPs identified between them. Oogenesis was upregulated in host-shifted mites on the new host A. mellifera relative to non-shifted mites. The transcriptomes of the host-shifted and non-shifted mites differed significantly as early as 1h post-infestation. The DEGs were associated with nine genes carrying nonsynonymous high- F ST SNPs, including mGluR2-like , Lamb2-like and Vitellogenin 6-like , which were also differentially expressed, and eIF4G , CG5800 , Dap160 and Sas10 , which were located in the center of the networks regulating the DEGs based on protein-protein interaction analysis. The annotated functions of these genes were all associated with oogenesis. These genes appear to be the key genetic determinants of the oogenesis of host-shifted mites on the new host. Sagamiscintilla thalassemicola (Bivalvia: Galeommatoidea: Galeommatidae) is a rare ectocommensal bivalve that lives on the proboscis of echiuran worms, Anelassorhynchus spp. (Annelida: Thalassematidae: Thalassematinae: Thalassematini), and has been known only from the temperate zones of Japan. In this study, we found S. thalassemicola on the proboscis of the large echiuran Ochetostoma sp. (Thalassematidae: Thalassematinae: Thalassematini) on intertidal flats of three islands of the Ryukyu Archipelago, southern Japan. These are the first records of S. thalassemicola on non- Anelassorhynchus hosts and also from the subtropical regions. Additionally, we also collected S. thalassemicola from an intertidal flat of Kushimoto, Wakayama, Kii Peninsula, Japan, which is an update of the easternmost record of this species. The genetic differences in the mitochondrial cytochrome c oxidase subunit I and nuclear internal transcribed spacer 2 genes among S. thalassemicola , including those with Ochetostoma sp. from the subtropical region and with Anelassorhynchus spp. from the temperate region, can be considered within the intraspecific variation. Baylisascaris schroederi, a roundworm (ascaridoid) parasite specific to the bamboo-feeding giant panda (Ailuropoda melanoleuca), represents a leading cause of mortality in wild giant panda populations. Here, we present a 293-megabase chromosome-level genome assembly of B. schroederi to infer its biology, including host adaptations. Comparative genomics revealed an evolutionary trajectory accompanied by host-shift events in ascaridoid parasite lineages after host separations, suggesting their potential for transmission and rapid adaptation to new hosts. Genomic and anatomical lines of evidence, including expansion and positive selection of genes related to the cuticle and basal metabolisms, indicate that B. schroederi undergoes specific adaptations to survive in the sharp-edged bamboo-enriched gut of giant pandas by structurally increasing its cuticle thickness and efficiently utilizing host nutrients through gut parasitism. Additionally, we characterized the secretome of B. schroederi and predicted potential drug and vaccine targets for new control strategies. Overall, this genome resource provides new insights into the host adaptation of B. schroederi to the giant panda as well as the host-shift events in ascaridoid parasite lineages. Our findings on the unique biology of B. schroederi will also aid in the development of prevention and treatment measures to protect giant panda populations from roundworm parasitism.

The rate at which zoonotic viruses spill over into the human population varies significantly over space and time. Remarkably, we do not yet know how much of this variation is attributable to genetic variation within viral populations. This gap in understanding arises because we lack methods of genetic analysis that can be easily applied to zoonotic viruses, where the number of available viral sequences is often limited, and opportunistic sampling introduces significant population stratification. Here, we explore the feasibility of using patterns of shared ancestry to correct for population stratification, enabling genome-wide association methods to identify genetic substitutions associated with spillover into the human population. Using a combination of phylogenetically structured simulations and Lassa virus sequences collected from humans and rodents in Sierra Leone, we demonstrate that existing methods do not fully correct for stratification, leading to elevated error rates. We also demonstrate, however, that the Type I error rate can be substantially reduced by confining the analysis to a less-stratified region of the phylogeny, even in an already-small dataset. Using this method, we detect two candidate single-nucleotide polymorphisms associated with spillover in the Lassa virus polymerase gene and provide generalized recommendations for the collection and analysis of zoonotic viruses.

The spatial distribution of parasitoids is closely linked to the distribution and ecological requirements of their hosts. Several studies have documented changes in the fauna composition of parasitoids in response to elevation, but data on parasitoids associated with spiders are missing. The koinobiont ichneumonid wasp Zatypota anomala is strictly specialised on spiders of the genus Dictyna (Dictynidae) in Europe. We examined the distribution of spiders of the family Dictynidae in forest ecotones in central Europe across a broad elevation gradient (110-1466 m a.s.l.). We checked the spiders for parasitism by Z. anomala. It was most abundant at the mid-elevations (median 712 m a.s.l., range 179-870 m a.s.l.). We identified four dictynid spider species as Z. anomala hosts. These were Dictyna arundinacea, Dictyna uncinata, Nigma flavescens, and Nigma walckenaeri. All four species and the genus Nigma were recorded as hosts for the first time. The parasitoids strongly preferred juvenile instars of their hosts. The body length differed between parasitised Dictyna and Nigma spiders (medians: 1.95 mm and 2.55 mm, respectively). The distribution of Dictyna and Nigma spiders overlapped along the elevation gradient, but parasitism incidence significantly differed between spider genera along the elevation gradient. Nigma was parasitized at lower elevations between 179-254 m a.s.l. and Dictyna at higher elevations between 361-870 m a.s.l. The phenology of Z. anomala is closely tied to the univoltine life strategy of its host spiders. The parasitoid female oviposits in autumn, and its offspring overwinter as larvae on the host, reach adulthood during spring, and pass the summer as an adult. [ABSTRACT FROM AUTHOR]

Clustered regularly interspaced short palindromic repeat (CRISPR)-Cas systems are bacterial defences that target bacteriophages and mobile genetic elements. How these defences evolve in novel host environments remains largely unknown. We studied the evolution of the CRISPR-Cas system in Mycoplasma gallisepticum (also named Mycoplasmoides gallisepticum ), a bacterial pathogen of poultry that jumped into a passerine host ~30 years ago. Over the decade following the host shift, all isolates displaying a functional CRISPR-Cas system were found not only to harbour completely new sets of spacers, but the DNA protospacer adjacent motif recognized by the main effector M. gallisepticum Cas9 (MgCas9) was also different. These changes in CRISPR-Cas diversity and specificity are consistent with a change in the community of phages and mobile elements infecting M. gallisepticum as it colonized the novel host. In the years following the host shift, we also detected a gradual rise in isolates displaying non-functional MgCas9. After 12 years, all circulating isolates harboured inactive forms only. This loss of CRISPR-Cas function comes at a time when the passerine host is known to have evolved widespread resistance, which in turn drove the evolution of increasing M. gallisepticum virulence through antagonistic coevolution. Such striking concordance in the rise of inactivated forms of CRISPR-Cas and the evolution of host resistance suggests that the inactivation of the CRISPR-Cas system was necessary for enabling adaptive bacterial responses to host-driven selection. We highlight the need to consider both host and pathogen selection pressures on bacteria for understanding the evolution of CRISPR-Cas systems and the key factors driving the emergence of a pathogenic bacterium in a novel host.

The Actinomycetota (formerly Actinobacteria) genus Clavibacter includes phytopathogens with devasting effects in several crops. Clavibacter michiganensis , the causal agent of tomato bacterial canker, is the most notorious species of the genus. Yet, its origin and natural reservoirs remain elusive, and its populations show pathogenicity profiles with unpredictable plant disease outcomes. Here, we generate and analyse a decade-long genomic dataset of Clavibacter from wild and commercial tomato cultivars, providing evolutionary insights that directed phenotypic characterization. Our phylogeny situates the last common ancestor of C. michiganensis next to Clavibacter isolates from grasses rather than to the sole strain we could isolate from wild tomatoes. Pathogenicity profiling of C. michiganensis isolates, together with C. phaseoli and C. californiensis as sister taxa and the wild tomato strain, was found to be congruent with the proposed phylogenetic relationships. We then identified gene enrichment after the evolutionary event, leading to the appearance of the C. michiganesis clade, including known pathogenicity factors but also hitherto unnoticed genes with the ability to encode adaptive traits for a pathogenic lifestyle. The holistic perspective provided by our evolutionary analyses hints towards a host shift event as the origin of C. michiganensis as a tomato pathogen and the existence of pathogenic genes that remain to be characterized.

Abstract Polyphagous aphids often consist of host‐specialized lineages, which have greater fitness on their native hosts than on others. The underlying causes are important for understanding of the evolution of diet breadth and host shift of aphids. The cotton‐melon aphid Aphis gossypii Glover is extremely polyphagous with many strict host‐specialized lineages. Whether and how the lineage specialized on the primary host hibiscus shifts to the secondary host cucumber remains elusive. We found that the hibiscus‐specialized lineage suffered high mortality and gave birth to very few nymphs developing into yellow dwarfs on fresh cucumber leaves, and did not inflict any damage symptoms on cucumber plants. The poor performance did not improve with prolonged exposure to cucumber; however, it did significantly improve when the cucumber leaves were pre‐infected with a biotrophic phytopathogen Pseudoperonospora cubensis. More importantly, the hibiscus‐specialized lineage with two‐generation feeding experience on pre‐infected cucumber leaves performed as well as the cucumber‐specialized lineage did on fresh cucumber leaves, and inflicted typical damage symptoms on intact cucumber plants. Electrical penetration graph (EPG) indicated that the hibiscus‐specialized lineage did not ingest phloem sap from fresh cucumber leaves but succeeded in ingesting phloem sap from pre‐infected cucumber leaves, which explained the performance improvement of the hibiscus‐specialized lineage on pre‐infected cucumber leaves. This study revealed a new pathway for the hibiscus‐specialized lineage to quickly acclimate to cucumber under the assistance of the phytopathogen. We considered that the short feeding experience on pre‐infected cucumber may activate expression of effector genes that are related to specific host utilization. We suggest to identify host‐specific effectors by comparing proteomes or/and transcriptomes of the hibiscus‐specialized lineage before and after acclimating to cucumber.

ABSTRACT Bats are reservoirs of diverse coronaviruses (CoVs), including progenitors of severe acute respiratory syndrome CoV (SARS-CoV) and SARS-CoV-2. In the Americas, there is a contrast between alphacoronaviruses (alphaCoVs) and betaCoVs: while cospeciation prevails in the latter, alphaCoV evolution is dominated by deep and recent host switches. AlphaCoV lineages are maintained by two different bat family groups, Phyllostomidae and Vespertilionidae plus Molossidae. In this study, we used a Bayesian framework to analyze the process of diversification of the lineages maintained by Molossidae and Vespertilionidae, adding novel CoV sequences from Argentina. We provide evidence that the observed CoV diversity in these two bat families is shaped by their geographic distribution and that CoVs exhibit clustering at the level of bat genera. We discuss the causes of the cocirculation of two independent clades in Molossus and Tadarida as well as the role of Myotis as the ancestral host and a major evolutionary reservoir of alphaCoVs across the continent. Although more CoV sampling efforts are needed, these findings contribute to a better knowledge of the diversity of alphaCoVs and the links between bat host species. IMPORTANCE Bats harbor the largest diversity of coronaviruses among mammals. In the Americas, seven alphacoronavirus lineages circulate among bats. Three of these lineages are shared by members of two bat families: Vespertilionidae and Molossidae. Uncovering the relationships between these coronaviruses can help us to understand patterns of cross-species transmission and, ultimately, which hosts are more likely to be involved in spillover events. We found that two different lineages cocirculate among the bat genera Molossus and Tadarida, which share roosts and have common viral variants. The bat genus Myotis functions as a reservoir of coronavirus diversity and, as such, is a key host. Although there were some spillovers recorded, there is a strong host association, showing that once a successful host jump takes place, it is transmitted onward to members of the same bat genus.

Abstract The diamondback moth, Plutella xylostella, is an important agricultural pest that severely damages cruciferous vegetables. Although previously considered a threat only to Brassica species, P. xylostella has been observed to feed on noncruciferous vegetables. Here, we established a population of P. xylostella on the pea Pisum sativum (PxP population). We compared this PxP population's performance on the pea host plant to a population (PxR) reared on the original host plant radish (Raphanus sativus) for several generations using an age‐stage, two‐sex life table and analyzed the correlations between different fitness parameters. In the 1st generation of the PxP population, survival rate of immature stage was 17%, while the survival rate of PxR was 68%; the duration of the 4th larval instar (5.30 d) and mortality (25%) of this generation were significantly longer (2.8 d) and higher (1%) than that of PxR, respectively (both p

ABSTRACT Bats harbor the largest number of coronavirus (CoV) species among mammals, serving as major reservoirs of alphaCoVs and betaCoVs, which can jump between bat species or to different mammalian hosts, including humans. Bat-CoV diversity is correlated with host taxonomic diversity, with the highest number of CoV species found in areas with the highest levels of bat species richness. Although the Americas harbor a unique and distinctive CoV diversity, no cross-species transmission (CST) or phylogeographic analysis has yet been performed. This study analyzes a large sequence data set from across the Americas through a Bayesian framework to understand how codivergence and cross-species transmission have shaped long-term bat-CoV evolution and ultimately identify bat hosts and regions where the risk of CST is the highest. Substantial levels of CST were found only among alphaCoVs. In contrast, cospeciation prevailed along the evolution of betaCoVs. Brazil is the center of diversification for both alpha and betaCoVs, with the highest levels of bat species richness. The bat family Phyllostomidae has played a key role in the evolution of American bat-CoVs, supported by the highest values of host transition rates. Although the conclusions drawn from this study are supported by biological/ecological evidence, it is likely that novel lineages will be discovered, which could also reveal undetected CSTs given that sequences are available from 11 of the 35 countries encompassing the Americas. The findings of this study can be useful for conducting targeted discovery of bat-CoVs in the region, especially in countries of the Americas with no reported sequences. Abstract While contemporary changes in feeding preferences have been documented in phytophagous insects, the mechanisms behind these processes remain to be fully clarified. In this context, the insect gut microbiome plays a central role in adaptation to novel host plants. The cucurbit frugivorous fruit fly Zeugodacus cucurbitae (Diptera, Tephritidae) has occasionally been reported on “unconventional” host plants from different families, including Solanaceae. In this study, we focus on wild parental (F0) adults and semiwild first filial (F1) larvae of Z. cucurbitae from multiple sites in La Réunion and explore how the gut microbiome composition changes when this fly is feeding on a noncucurbit host (Solanum melongena). Our analyses show nonobvious gut microbiome responses following the F0–F1 host shift and the importance of not just diet but also local effects, which heavily affected the diversity and composition of microbiomes. We identified the main bacterial genera responsible for differences between treatments. These data further stress the importance of a careful approach when drawing general conclusions based on laboratory populations or inadequately replicated field samples.

Abstract Background Wolbachia is an endosymbiont common to most invertebrates, which can have significant evolutionary implications for its host species by acting as a barrier to gene flow. Despite the importance of Wolbachia, still little is known about its prevalence and diversification pattern among closely related host species. Wolbachia strains may phylogenetically coevolve with their hosts, unless horizontal host-switches are particularly common. We address these issues in the genus Erebia, one of the most diverse Palearctic butterfly genera. Results We sequenced the Wolbachia genome from a strain infecting Erebia cassioides and showed that it belongs to the Wolbachia supergroup B, capable of infecting arthropods from different taxonomic orders. The prevalence of Wolbachia across 13 closely related Erebia host species based on extensive population-level genetic data revealed that multiple Wolbachia strains jointly infect all investigated taxa, but with varying prevalence. Finally, the phylogenetic relationships of Wolbachia strains are in some cases significantly associated to that of their hosts, especially among the most closely related Erebia species, demonstrating mixed evidence for phylogenetic coevolution. Conclusions Closely related host species can be infected by closely related Wolbachia strains, evidencing some phylogenetic coevolution, but the actual pattern of infection more often reflects historical or contemporary geographic proximity among host species. Multiple processes, including survival in distinct glacial refugia, recent host shifts in sympatry, and a loss of Wolbachia during postglacial range expansion seem to have jointly shaped the complex interactions between Wolbachia evolution and the diversification of its host among our studied Erebia species.

Climate-driven geographic range shifts have been associated with transitions between dietary specialism and generalism at range margins. The mechanisms underpinning these often transient niche breadth modifications are poorly known, but utilization of novel resources likely depends on phenological synchrony between the consumer and resource. We use a climate-driven range and host shift by the butterfly Aricia agestis to test how climate-driven changes in host phenology and condition affect phenological synchrony, and consider implications for host use. Our data suggest that the perennial plant that was the primary host before range expansion is a more reliable resource than the annual Geraniaceae upon which the butterfly has become specialized in newly colonized parts of its range. In particular, climate-driven phenological variation in the novel host Geranium dissectum generates a narrow and variable 'window of opportunity' for larval productivity in summer. Therefore, although climatic change may allow species to shift hosts and colonise novel environments, specialization on phenologically limited hosts may not persist at ecological margins as climate change continues. We highlight the potential role for phenological (a)synchrony in determining lability of consumer-resource associations at range margins and the importance of considering causes of synchrony in biotic interactions when predicting range shifts. Chaitophorinae is a predominantly Northern Hemisphere aphid subfamily characterized by numerous setae on the body. Two constituent tribes are associated with different host plants, with Chaitophorini feeding on deciduous trees and shrubs and Siphini colonizing grasses. Based on data from multiple genes (COI, COII, Cytb and EF‐1α), geographical distribution and host association, this study investigated the phylogeny and macroevolution of Chaitophorinae using phylogenetic reconstruction, molecular dating, model‐based ancestral area and character estimations and diversification rate calculation. Our results support the monophyly of Chaitophorinae and two tribes, indicate that Sipha and the two largest genera Chaitophorus and Periphyllus are not monophyletic, and suggest a need for a change in the taxonomic status of Lambersaphis, which was nested within Chaitophorus in the phylogenetic tree. We recovered an origin of Chaitophorinae on Acer plants from eastern Asia during the Late Cretaceous to early Palaeocene, followed by multiple dispersals into other areas that were responsible for its contemporary distribution. The origins of Siphini and Chaitophorus + Lambersaphis coincided with colonizations of novel host plants. An increase in diversification rate occurred within Chaitophorus in the Miocene and was associated with range expansion and switching onto new host plants, highlighting the roles of dispersal and host shift in aphid diversification. BACKGROUND: The general principle of using microbes from one species to manage a different pest species has a clear precedent in the large‐scale release of mosquitoes carrying a Wolbachia bacterium derived from Drosophila flies. New technologies will facilitate the discovery of microbes that can be used in a similar way. Previously, we found three novel partiti‐like viruses in the African armyworm (Spodoptera exempta). To investigate further the utility and consistency of host shift of insect viruses as a potential pest management tool, we tested the interaction between the partiti‐like viruses and another novel host, the Egyptian cotton leafworm (Spodoptera littoralis). RESULT: We found that all three partiti‐like viruses appeared to be harmful to the novel host S. littoralis, by causing increased larval and pupal mortality. No effect was observed on host fecundity, and partiti‐like virus infection did not impact host susceptibility when challenged with another pathogen, the baculovirus SpliNPV. Transcriptome analysis of partiti‐like virus‐infected and noninfected S. littoralis indicated that the viruses could impact host gene‐expression profiles of S. littoralis, but they impact different pathways to the two other Spodoptera species through effects on pathways related to immunity (Jak–STAT/Toll and Imd) and reproduction (insulin signaling/insect hormones). CONCLUSION: Taken together with the previous findings in the novel host S. frugiperda, these results indicate a parasitic relationship between the partiti‐like viruses and novel insect hosts, suggesting a possible use and novel pest management strategy through the artificial host shift of novel viruses. © 2021 Society of Chemical Industry. During a parasitological survey of goldfish Carassius auratus (Linnaeus, 1758) in China, two myxosporeans were collected in the present study including a Thelohanellus species and a Myxobolus species. To identified the two myxosporeans, a morphological, histological and molecular study was performed. The Thelohanellus species was characterized by round, white plasmodia in fins of goldfish. Compared with all described Thelohanellus species, spore morphology of the Thelohanellus species in the present study was distinct from them. BLAST search showed that SSU rDNA sequence of this Thelohanellus species did not match any sequence in GenBank database. Phylogenetically, the Thelohanellus species obtained in the present study was sister to Thelohanellus nikolskii Achmerov, 1955. So, the present Thelohanellus species was identified as a novel species and named Thelohanellus pseudonikolskii n. sp. The Myxobolus species was characterized by a number of round or irregularly shaped, white plasmodia in gills of goldfish. When compared with all reported Myxobolus species, spore morphology of the present Myxobolus species was most similar to Myxobolus koi Kudo, 1920. Histological analysis showed that plasmodia of this Myxobolus species simultaneously developed in stratified epithelium of filament and in lamellae, which was same to M. koi. Moreover, BLAST search showed that SSU rDNA sequence of the present Myxobolus species was identical to M. koi. Thus, the morphological, histological and molecular analysis demonstrated that the present Myxobolus species was conspecific with M. koi. Due to M. koi consistently described from gills of common carp Cyprinus carpio Linnaeus, 1758 and koi carp C. carpio in the previous researches, M. koi was first found to infect gills of goldfish in the present study. Considering koi carp and goldfish both as ornamental fish frequently cultured together in China, the possible host shift of M. koi from koi carp to goldfish is of interest for further investigation.

ABSTRACT Because parasites have an inextricable relationship with their host, they have the potential to serve as viral reservoirs or facilitate virus host shifts. And yet, little is known about viruses infecting parasitic hosts except for blood-feeding arthropods that are well-known vectors of zoonotic viruses. Herein, we uncovered viruses of flatworms (phylum Platyhelminthes, group Neodermata) that specialize in parasitizing vertebrates and their ancestral free-living relatives. We discovered 115 novel viral sequences, including 1 in Macrostomorpha, 5 in Polycladida, 44 in Tricladida, 1 in Monogenea, 15 in Cestoda, and 49 in Trematoda, through data mining. The majority of newly identified viruses constitute novel families or genera. Phylogenetic analyses show that the virome of flatworms changed dramatically during the transition of neodermatans to a parasitic lifestyle. Most Neodermata viruses seem to codiversify with their host, with the exception of rhabdoviruses, which may switch hosts more often, based on phylogenetic relationships. Neodermata rhabdoviruses also have a position ancestral to vertebrate-associated rhabdo viruses, including lyssaviruses, suggesting that vertebrate-associated rhabdoviruses emerged from a flatworm rhabdovirus in a parasitized host. This study reveals an extensive diversity of viruses in Platyhelminthes and highlights the need to evaluate the role of viral infection in flatworm-associated diseases. IMPORTANCE Little is known about the diversity of parasite-associated viruses and how these viruses may impact parasite fitness, parasite-host interactions, and virus evolution. The discovery of over a hundred viruses associated with a range of free-living and parasitic flatworms, including parasites of economic and clinical relevance, allowed us to compare the viromes of flatworms with contrasting lifestyles. The results suggest that flatworms acquired novel viruses after their transition to a parasitic lifestyle and highlight the possibility that they acquired viruses from their hosts and vice versa. An interesting example is the discovery of flatworm rhabdoviruses that have a position ancestral to rabies viruses and other vertebrate-associated rhabdoviruses, demonstrating that flatworm-associated viruses have emerged in a vertebrate host at least once in history. Therefore, parasitic flatworms may play a role in virus diversity and emergence. The roles that parasite-infecting viruses play in parasite-associated diseases remain to be investigated.

A parasite's lifestyle is characterized by a critical dependency on its host for feeding, shelter and/or reproduction. The ability of parasites to exploit new host species can reduce the risk associated with host dependency. The number of host species that can be infected by parasites strongly affects their ecological and evolutionary dynamics along with their pathogenic effects on host communities. However, little is known about the processes and the pathways permitting parasites to successfully infect alternative host species, a process known as host shift. Here, we tested whether molecular plasticity changes in gene expression and in molecular pathways could favor host shift in parasites. Focusing on an invasive parasite, Tracheliastes polycolpus, infecting freshwater fish, we conducted a transcriptomic study to compare gene expression in parasites infecting their main host species and two alternative host species. We found 120 significant differentially expressed genes (DEGs) between parasites infecting the different host species. A total of 90% of the DEGs were identified between parasites using the main host species and those using the two alternative host species. Only a few significant DEGs (seven) were identified when comparing parasites from the two alternative host species. Molecular pathways enriched in DEGs and associated with the use of alternative host species were related to cellular machinery, energetic metabolism, muscle activity and oxidative stress. Abstract Parasite host shifts can impose a high selective pressure on novel hosts. Even though the coevolved systems can reveal fundamental aspects of host–parasite interactions, research often focuses on the new host–parasite relationships. This holds true for two ectoparasitic mite species, Varroa destructor and Varroa jacobsonii, which have shifted hosts from Eastern honey bees, Apis cerana, to Western honey bees, Apis mellifera, generating colony losses of these pollinators globally. Here, we study infestation rates and reproduction of V. destructor and V. jacobsonii haplotypes in 185 A. cerana colonies of six populations in China and Thailand to investigate how coevolution shaped these features. Reproductive success was mostly similar and low, indicating constraints imposed by hosts and/or mite physiology. Infestation rates varied between mite haplotypes, suggesting distinct local co‐evolutionary scenarios. The differences in infestation rates and reproductive output between haplotypes did not correlate with the virulence of the respective host‐shifted lineages suggesting distinct selection scenarios in novel and original host. The occasional worker brood infestation was significantly lower than that of drone brood, except for the V. destructor haplotype (Korea) from which the invasive lineage derived. Whether mites infesting and reproducing in atypical intraspecific hosts (i.e., workers and queens) actually predisposes for and may govern the impact of host shifts on novel hosts should be determined by identifying the underlying mechanisms. In general, the apparent gaps in our knowledge of this coevolved system need to be further addressed to foster the adequate protection of wild and managed honey bees from these mites globally.

Host shifting and host expansion of fungal plant pathogens increases the rate of emergence of new pathogens and the incidence of disease in various crops, which threaten global food security. Magnaporthe species cause serious disease in rice, namely rice blast disease, as well as in many alternative hosts, including wheat, barley, and millet. A severe outbreak of wheat blast due to Magnaporthe oryzae occurred recently in Bangladesh, after the fungus was introduced from South America, causing great loss of yield. This outbreak of wheat blast is of growing concern, because it might spread to adjacent wheat-producing areas. Therefore, it is important to understand the host range and population structure of M. oryzae and related species for determining the evolutionary relationships among Magnaporthe species and for managing blast disease in the field. Here, we collected isolates of M. oryzae and related species from various Poaceae species, including crops and weeds surrounding rice fields, in Korea and determined their phylogenetic relationships and host species specificity. Internal transcribed spacer-mediated phylogenetic analysis revealed that M. oryzae and related species are classified into four groups primarily including isolates from rice, crabgrass, millet and tall fescue. Based on pathogenicity assays, M. oryzae and related species can infect different Poaceae hosts and move among hosts, suggesting the potential for host shifting and host expansion in nature. These results provide important information on the diversification of M. oryzae and related species with a broad range of Poaceae as hosts in crop fields.

Guam’s established population of non-native coconut rhinoceros beetle (CRB, Oryctes rhinoceros L.) began creating burrows in stem apices of several cycad species in a managed garden. We conducted an island-wide survey to determine the spatial patterns of CRB burrowing of stems of in situ Cycas micronesica. We also measured starch of healthy and unhealthy coconut leaf tissue and compared this with starch of cycad stem tissue. The starch concentration of the central unexpanded leaf in healthy Philippine coconut trees was ≈90 mg·g−1, and that of unhealthy Guam coconut trees was ≈40 mg·g−1. The starch concentration of the tissue within the CRB burrow locations on C. micronesica trees was 145 mg·g−1. Burrowing of C. micronesica was restricted to female CRB adults and was found throughout the full latitudinal gradient of Guam. Our findings indicate Guam’s unhealthy coconut trees are no longer nutrient-dense, and the female CRB population may have exhibited a phylogenetically distant host shift to the abundant C. micronesica plants for a starch-rich diet within the concepts of the ‘ecological fitting’ hypothesis. We add proximity to coconut tree habitats as a new threat to Guam’s endangered C. micronesica population.
