4. Survey of strain distribution and antibiotic resistance pattern of group B streptococci (Streptococcus agalactiae) isolated from clinical specimens

Abstract

Aim: The aims of the present study were to determine the antibiotic susceptibility profils with particular emphasis on susceptible or resistant strains to macrolides and lincosamids antibiotics and to determine possible antibiotic resistance mechanisms occurring in group B streptococci (GBS) strains using PCR assay and disk diffusion method.Methods: A total of 62 clinical GBS strains were investigated. Antibacterial susceptibility testing was performed using the disk diffusion method and inducible resistance test for clindamycin by standard double disk diffusion or D-zone test for all isolates to differentiate macrolide resistance phenotype (M), constitutive macrolide-lincosamide-streptogramin B phenotype (cMLS) and induced macrolide-lincosamide-streptogramin B phenotype (iMLS). In addition, minimum inhibitory concentrations (MIC) of penicillin were determined for all isolates. Finally, possible existence of antibiotic resistance genes for erythromycin , and ) and for clindamycin ) were examined among isolates using PCR assay.Results: All 62 isolates were susceptible to penicillin, ampicillin, linezolid, cefazoline and vancomycin. However, 93.5% (n=58) of isolates showed an increased MIC to penicillin. The overall rate of erythromycin resistance was 35.5% (n=22). All erythromycin-resistant isolates displayed the M phenotype (100%, n=22). All three erythromycin resistance genes (i.e. , and ) were found in erythromycin-resistant isolates.Conclusion: It was concluded that prescribing antibiotic without antibacterial susceptibility tests should be prevented because of the high prevalence of erythromycin-resistant GBS strains and the fact that erythromycin-resistant GBS strains has shown an increased MIC to penicillin, as the drug of choice for treating GBS infections.

14. Antibiotics-encapsulated nanoparticles as an antimicrobial agent in the treatment of wound infection.

Abstract

Disruption in the wound-healing process is caused by the presence of bacteria and leads to major problems and delays in wound healing. The limitations of commonly used medicines for treating wound infections include drug toxicity, insufficient microbial coverage, poor penetration, and increased resistance. This study aimed to determine the effect of ciprofloxacin loaded in solid lipid nanoparticles (Cip-SLN) on Pseudomonas aeruginosa and ampiciliin-vancomycin loaded in solid lipid nanoparticles (Amp-Van-SLN) on Staphylococcus aureus in wounds. Antibiotics were encapsulated in SLNs using the double emulsion method and were characterized. The in-vitro effect of antibiotic-loaded nanoparticles on P. aeruginosa and S. aureus was assessed using well diffusion and MIC methods. Finally, the topical antibacterial activity of these nanoparticles against bacterial wound infection was measured in a mouse model. MIC results showed that in the first 24 hours, the free drug had a greater effect on inhibiting bacteria, and in 72 hours, the inhibitory effect of nanoparticles increased. There was no toxicity effect of 400 µg/mL of nanoparticles on cells. According to the findings, the groups treated with Cip-SLN and Amp-Van-SLN were more effective than the control group (untreated) in different concentrations. In the wound healing process, the group treated with solid lipid nanoparticles (SLNs) exhibited a greater epithelial thickness, indicating enhanced healing, compared to the group treated with the free drug. The use of SLN can increase the accumulation of antibiotics at the site of infection with a slow release of the drug due to its fatty nature, which leads to a significant inhibitory effect on bacteria and also improves wound healing. Abstract

Carbapenem-resistant (CR) Gram-negative bacteria have become a significant public health problem in the last decade. In recent years, the prevalence of CR bacteria has increased. The resistance to carbapenems could result from different mechanisms such as loss of porin, penicillin-binding protein alteration, carbapenemase, efflux pump, and biofilm community. Additionally, genetic variations like insertion, deletion, mutation, and post-transcriptional modification of corresponding coding genes could decrease the susceptibility of bacteria to carbapenems. In this regard, scientists are looking for new approaches to inhibit CR bacteria. Using bacteriophages, natural products, nanoparticles, disulfiram, N-acetylcysteine, and antimicrobial peptides showed promising inhibitory effects against CR bacteria. Additionally, the mentioned compounds could destroy the biofilm community of CR bacteria. Using them in combination with conventional antibiotics increases the efficacy of antibiotics, decreases their dosage and toxicity, and resensitizes CR bacteria to antibiotics. Therefore, in the present review article, we have discussed different aspects of non-antibiotic approaches for managing and inhibiting the CR bacteria and various methods and procedures used as an alternative for carbapenems against these bacteria. In this study, we introduce an innovative semiconductor quantum well (SQW) nanostructure, which exhibits a unique four-level configuration arranged in an inverted Y-shape. This configuration enables interaction with three laser fields and demonstrates the intriguing nonlinear phenomenon of parametric generation. Our investigation delves into a wide range of parametric conditions, aiming to understand the profound impact of various system parameters on the generation and propagation of new light fields within this groundbreaking nanostructure. By employing a combination of analytical and numerical methods, we not only reveal the potential for precise manipulation of coherence among energy levels but also showcase its practical implications — high-efficiency and tunability in the parametric generation process. This pioneering scheme represents a significant advancement in the field of SQW structures, bridging the gap between theoretical exploration and practical application. Abstract

The association of CETP Taq1B polymorphism with some metabolic traits is still controversial. The interaction of adherence to dietary indices with this polymorphism on the severity of coronary artery stenosis and serum lipid parameters needs to be investigated. This study aimed to test this hypothesis. This cross‐sectional study included 453 patients who were referred from Afshar Hospital of Yazd and undergoing coronary angiography from 2020 to 2021. Dietary intake was evaluated by a 178‐item validated and reliable dietary questionnaire. Dietary indices such as dietary antioxidant index (DAI), dietary antioxidant quality score (DAQS), and dietary phytochemical index (DPI) are determined according to dietary guidelines. The Taq1B variant was genotyped by the polymerase chain reaction‐restriction fragment length polymorphism method (PCR‐RFLP). Two‐way ANOVA was used to test the interaction between Taq1B polymorphism and dietary indices. The results of the frequency analysis of Taq1B genotypes showed that 10.4% were B1B1, 72.4% B1B2, and 17.2% B2B2. No significant interaction was found between the Taq1B variant with high adherence to DAQS, DAI, and DPI on total cholesterol (TC), high‐density lipoprotein cholesterol (HDL), low‐density lipoprotein cholesterol (LDL), triglyceride (TG) levels, and Gensini score (GS) and Syntax score (SS). In high‐adherence dietary indices, lipid profile and coronary artery stenosis scores did not differ significantly in Taq1B genotypes. Due to the insignificant results in this research, further studies are needed to investigate the role of Taq1B SNP in modulating dyslipidemia and the severity of the CAD in interaction with dietary indices. Abstract

Inertial navigation system (INS) is the most common positioning system in the underwater vehicle due to the limitations of penetration of the global navigation system. Amendment of INS cumulative error is made possible using data of auxiliary sensors such as Doppler velocity log (DVL). Due to the environmental conditions of the seabed, access to full DVL beams is impractical. Moreover, the statistical characteristics of measuring noise are unknown. These dramatically reduce the performance of the navigation system. Therefore, a loosely coupled (LC) approach is used to fuse partial DVL measurements in this study. This approach benefits from a virtual beam containing the velocity obtained in the last INS step instead of inaccessible DVL beams. To deal with the unknown statistical characteristics of the measurement and the limitations of access to DVL beams, an adaptive robust Kalman filter (ARKF) based on the Huber cost function is proposed. The performance of the proposed integrated navigation system for a remotely operated vehicle is investigated in the presence of observation noise and outliers. Results show that the proposed ARKF is robust against vigorous maneuvers, improves the estimation accuracy effectively, and can effectively reject measurement outliers. [ABSTRACT FROM AUTHOR]
