Abstract The presence of antibiotic pollutants in water and wastewater can cause significant risks to the environment in different aspects. Therefore, antibiotics need to be removed from water. This study investigates the adsorption of nalidixic acid (NA), a common antibiotic, using bovine serum albumin nanoparticles (BSA NPs). These NPs were synthesized via desolvation technique and characterized using SEM, DLS, FT-IR, and UV-Vis spectroscopy. The effects of adsorbent dosage (0.02–0.9 mg), initial NA concentration (30–80 mg L− 1) and contact time (0.5–24 h) on adsorption efficiency were considered. Adsorption isotherms and kinetics were determined experimentally. The Freundlich isotherm best described the adsorption equilibrium, while the pseudo-second-order kinetic model accurately represented the adsorption process. Thermodynamic parameters confirmed the spontaneous and exothermic nature of NA adsorption onto BSA NPs. Under optimal conditions, BSA NPs achieved a removal efficiency of 75% for NA with a maximum adsorption capacity of 240 mg g− 1. These results demonstrate the potential of BSA NPs as an effective adsorbent for removing NA from aqueous solutions.

Abstract Radiation therapy and phototherapy are commonly used cancer treatments that offer advantages such as a low risk of adverse effects and the ability to target cancer cells while sparing healthy tissue. A promising strategy for cancer treatment involves using nanoparticles (NPs) in combination with radiation and photothermal therapy to target cancer cells and improve treatment efficacy. The synthesis of gold NPs (AuNPs) for use in biomedical applications has traditionally involved toxic reducing agents. Here we harnessed dopamine (DA)-conjugated alginate (Alg) for the facile and green synthesis of Au NPs (Au@Alg-DA NPs). Alg-DA conjugate reduced Au ions, simultaneously stabilized the resulting AuNPs, and prevented aggregation, resulting in particles with a narrow size distribution and improved stability. Injectable Au@Alg-DA NPs significantly promoted ROS generation in 4T1 breast cancer cells when exposed to X-rays. In addition, their administration raised the temperature under a light excitation of 808 nm, thus helping to destroy cancer cells more effectively. Importantly, no substantial cytotoxicity was detected in our Au@Alg-DA NPs. Taken together, our work provides a promising route to obtain an injectable combined radio enhancer and photothermally active nanosystem for further potential clinic translation.

Abstract Carbon nanotubes (CNTs) have the potential to serve as delivery systems for medicinal substances and gene treatments, particularly in cancer treatment. Co-delivery of curcumin (CUR) and Methotrexate (MTX) has shown promise in cancer treatment, as it uses fewer drugs and has fewer side effects. This study used MTX-conjugated albumin (BSA)-based nanoparticles (BSA-MTX) to enhance and assess the efficiency of CUR. In-vitro cytotoxicity tests, DLS, TEM, FTIR, UV/Vis, SEM, and DSC studies assessed the formulations' physical and chemical properties. The Proteinase K enzyme was used to severe amidic linkages between MTX and BSA. The findings demonstrated the efficacy of using ƒ-MWCNT-CUR-BSA-MTX as a vehicle for efficient co-delivery of CUR and MTX in cancer treatment. The MTT colorimetric method was used to evaluate the effect of chemical and medicinal compounds. Cell division was studied using the MTT method to investigate the effect of pure MWCNT, pure CUR, MTX-BSA, and ƒ-MWCNT-CUR-MTX-BSA. Studies on cell lines have shown that the combination of curcumin and MTX with CNT can increase and improve the effectiveness of both drugs against cancer. A combination of drugs curcumin and methotrexate simultaneously had a synergistic effect on MCF-7 cells, which indicated that these drugs could potentially be used as a strategy for both prevention and treatment of breast cancer. Also, ƒ-MWCNT-CUR-MTX-BSA was found to have a significant effect on cancer treatment with minimal toxicity compared to pure curcumin, pure MTX-BSA, MTX, and ƒ-MWCNT alone. Unique properties such as a high ratio of specific surface area to volume, high chemical stability, chemical adsorption ability, high capacity of drug and biomolecules of carbon nanotubes, as well as multiple drug loading at the same time The combination of ƒ-MWCNT-CUR-BSA MTX significantly impacts cancer therapy), are desirable as an alternative option for targeted drug delivery and high therapeutic efficiency.

Cancer, a prevalent disease across various societies, presents a significant challenge in treatment research. Studies show that combination therapies are one of the methods that can help in the effective treatment of cancer. Chemotherapy and radiation therapy are among the main cancer treatments and in this project, for combined chemoradiotherapy treatment, carbon nanotubes were used as improved carriers of chemotherapy in tumors, as well as a substrate for the preparation of radiation sensitizers for local radiation therapy. Following the synthesis of CNT-Platinum-Curcumin nanoparticles (CNT-Pt-CUR), a series of analyses were conducted to verify the successful production of these nanoparticles. Techniques such as Transmission Electron Microscopy (TEM), Dynamic Light Scattering (DLS), UV–Vis spectroscopy, Fourier Transform Infrared Spectroscopy (FTIR), and X-Ray Diffraction (XRD) were employed. The characterization data revealed a spherical shape Pt nanoparticle morphology with an 8.5 nm diameter on rod-shape CNT, as observed through TEM. Furthermore, FTIR analysis confirmed the successful loaded of the drug into the nanoparticles, highlighting the potential of this approach in cancer treatment. Then, hemolysis and (3(-4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) tests on normal cells were used to assess the biocompatibility of CNT-Pt-CUR nanoparticles. It also explored the anticancer efficacy of these nanoparticles at varying concentrations against cancer cells, both with and without exposure to X-rays. The research confirmed the successful synthesis of these nanoparticles and demonstrated their potential impact on cell viability. Specifically, breast cancer cells exhibited heightened susceptibility to toxicity when exposed to nanoparticles and X-rays. Further analysis revealed that the toxicity of nanoparticles is dose-dependent, and modifying the surface of carbon nanotube (CNT) nanoparticles with CUR significantly reduced blood toxicity. Interestingly, nanoparticle toxicity was significantly amplified in the presence of X-rays, suggesting mechanisms such as DNA damage and increased reactive oxygen species (ROS) levels within cells.

Objective: To investigate the stability and efficacy of vitamin D3 and Bifidobacterium lactis nanoparticles coated with polycaprolactone-polyethylene glycol-polycaprolactone (PCL-PEG-PCL) in enhancing the nutritional value of Iranian white cheese and to assess their impact on the cheese's physicochemical and sensory properties. Methods: Bifidobacterium lactis and vitamin D3 were encapsulated using PCL-PEG-PCL. The particle size was analyzed using Dynamic Light Scattering (DLS). Five different cheese groups were produced and analyzed over two months. This included assessing microbial viability in simulated gastric and intestinal environments, quantifying vitamin D3 content, and evaluating physicochemical and sensory characteristics using a 9-point hedonic scale. Results: The Cheese with Bifidobacterium lactis coated with PCL-PEG-PCL and vitamin D3 nanoparticles (BCND3), showed a marked improvement in the stability of both Bifidobacterium lactis and vitamin D3. Specifically, the coated bacteria maintained a concentration of 106 cfu/g until day 45 in the intestinal model, demonstrating effective protection against the acidic gastric environment. The vitamin D3 nanoparticles displayed enhanced stability within the cheese matrix, contributing to the nutritional value and potentially extending the shelf life. Physicochemically, initial pH reduction due to lactic acid production stabilized by day 60, aligning with other groups. Sensory evaluation revealed a preference for the softer texture of cheese with vitamin D3 nanoparticles, suggesting a positive impact on consumer acceptability. Conclusion: The application of PCL-PEG-PCL coating significantly improved the stability of probiotics and vitamin D3 in cheese. This method proved effective in protecting Bifidobacterium lactis from harsh gastrointestinal conditions and in sustaining vitamin D3 content, with a notable impact on the cheese's sensory qualities. These findings highlight the potential of this coating technique in food fortification strategies.

Abstract To solve the traditional radiotherapy obstacles, and also to enhance the radiation therapy efficacy various radiosensitizers have been developed. Radiosensitizers are promising agents that under X-ray irradiation enhance injury to tumor tissue by accelerating DNA damage. In this report, silver-silver sulfide nanoparticles (Ag-Ag2S NPs) were synthesized via a facile, one-pot and environmentally friendly biomineralization method. Ag-Ag2S was coated with bovine serum albumin (BSA) in situ and applied as an X-ray sensitizer to enhance the efficiency of radiotherapy. Also, folic acid (FA) was conjugated to Ag-Ag2S@BSA to impart active targeting capability to the final formulation (Ag-Ag2S@BSA-FA). Prepared NPs were characterized by transmission electron microscopes (TEM), scanning electron microscope (SEM), dynamic light scattering (DLS), ultraviolet–visible spectroscopy (UV–Vis), X-ray diffraction analysis (XRD), and X-ray photoelectron spectroscopy (XPS) techniques. Results show that most of the NPs have well-defined uniform Janus structures. The biocompatibility of the NPs was then evaluated both in vitro and in vivo. A series of in vitro assays were performed on 4T1 cancer cells to evaluate the therapeutic efficacy of the designed NPs. In addition, the radio-enhancing ability of the NPs was tested on the 4T1 breast cancer murine model. MTT, live and dead cell staining, apoptosis, ROS generation, and clonogenic in vitro assays demonstrated the efficacy of NPs as radiosensitizers in radiotherapy. In vivo results as well as H&E staining tumor tissues confirmed tumor destruction in the group that received Ag-Ag2S@BSA-FA NPs and exposed to X-ray. The results showed that prepared tumor-targeted Ag-Ag2S@BSA-FA NPs could be potential candidates as radiosensitizers for enhanced radiotherapy.

Curcumin, derived from Curcuma longa, is a widely used natural compound in anticancer treatments. Single-walled carbon nanotubes (SWCNTs), a significant class of nanomaterials known for their unique physicochemical properties, serve as effective carriers for curcumin. This allows for the utilization of curcumin's antitumor capabilities while overcoming its conventional drawbacks of low aqueous solubility and instability under physiological conditions. In the current study, SWCNTs were cultivated without hydrogen gas, utilizing a Fe-Mo/Al2O3 catalyst. Once prepared by the Chemical Vapor Deposition (CVD) method under ideal conditions, they were combined with curcumin. Various techniques, including Transmission Electron Microscopy (TEM), Dynamic Light Scattering (DLS), UV–Vis, Fourier Transform Infrared (FTIR), Raman, and X-Ray Diffraction (XRD), were employed to characterize the system. The quality of the SWCNT was determined by the ID/IG ratio in Raman spectroscopy. The DLS technique revealed that the ζ-potential of SWCNT and SWCNT-Cur was approximately −13.9 and −25.5 mV, respectively. The Nano-formulation had a loading capacity of 14/87 %. The rate of hemolysis caused by SWCNT-Cur was less than 10 %, and the high rate of cell viability observed in HFF-2 cells following the MTT test indicates the high safety of the Nano-formulation. To validate the therapeutic effects of the SWCNT-Cur on the 4T1 cell line, both in the presence and absence of X-ray radiation, the MTT assay method was used in vitro. The results showed a significant increase in toxicity on breast cancer cells with increasing concentration.

Background and Objective: In medical sciences, identifying the anticancer properties of plumbagin is of special importance. For this reason, this study investigated the anticancer activity of polymeric nanoparticles loaded by plumbagin against breast cancer cells. Methods: In this descriptive study, the diblock copolymer mPEG–PCL was synthesized by ring-opening polymerization of caprolactone in the presence of mPEG as the initiator and Sn(oct)2 as the catalyst. The synthesized copolymers were characterized by Fourier-transform infrared spectroscopy, proton nuclear magnetic resonance, gel permeation chromatography, and differential scanning calorimetry. The nanoprecipitation method was used for preparing nanoparticles loaded with plumbagin. The characteristics of these nanoparticles were investigated by various techniques including dynamic light scattering. The cytotoxicity of plumbagin, copolymer, and the nanoparticles loaded with plumbagin on MCF7 and HFF2 cells was evaluated by MTT assay. Results: The average diameter of the nanoparticles was less than 115 nm. The loading capacity and encapsulation efficiencies were 15.4±0.13% and 79.1±0.65%, respectively. Drug release was slow, controlled, and almost dependent on pH. The results of the MTT assay showed strong and dose-dependent inhibition of cell growth by the plumbagin-loaded micelles compared with plumbagin alone in a way that the half maximal inhibitory concentration of this nanoparticle against MCF7 cells after 48 and 72 hours was 10.78 and 24.03 μM, respectively. Conclusion: The mPEG-PCL nanoparticles can be an efficient carrier for plumbagin, and plumbagin can be an effective drug on breast cancer cells, without toxicity on healthy cells.

Despite the over spatial separation and the ability to determine soft tissues, insufficient contrast is the shortcoming of magnetic resonance imaging (MRI) that could be circumvented by the use of contrast agents. The use of MRI contrast agents are widely applied to enhance the vision of internal body structures. Nano-sized contrast materials have unique application advantages compared to other contrast agents due to their size and shape. However, for contrast agents such as bare iron (II, III) oxide (Fe3O4) magnetic nanoparticles (NPs), aggregation and accumulation are the main shortcomings. Thus, surface modifications are necessary for their use in biopharmaceutical applications. Gold, Au, nanoparticles are of big interesting for use in biomedical purposes due to their chemical stability and oxidation resistance. In this study, we synthesized magnetic Fe3O4–Au hybrid NPs with a facile method and coated them with bovine serum albumin (BSA) to increase their chemical stability and biocompatibility. Afterwards, the hybrid nanosystem was characterized by some methods, and their potential to increase MRI contrast was investigated by the phantom MRI experiments. Our data showed that the signal intensity on MR images was significantly reduced, thus confirming the contrast ability of the formulated Fe3O4–Au-BSA NPs.

Background and purpose: Acinetobacter baumannii (A. baumannii) is known as a pathogen with antibiotic resistance, causing respiratory infections. PLGA has been approved for use in vaccines as well as drug delivery. This study was performed to evaluate PLGA nanoparticles containing the outer membrane proteins (OMPs) of A. baumannii in stimulating the mice’s immune system and improving pneumonia. Experimental approach: Double emulsion solvent evaporation technique was used. The properties of the obtained nanospheres were determined using a zetasizer, FTIR, and AFM devices. Nanoparticles were administered to mice BALB/c by applying the intramuscular route. ELISA was used to measure the amounts of immunoglobulins produced; also, an opsonophagocytic killing assay was used to measure the effectiveness of immunoglobulins. Immunized mice were then challenged with live A. baumannii through the lungs; their internal organs were also removed for bacteriological studies. Findings/Results: The prepared particles were 550 nm in diameter with a negative surface charge. The production of the OMPs specific IgG was much higher in the group receiving nanoparticles containing antigen as compared to those getting pure antigen. The immunoglobulins produced against nanoparticles were superior to those developed against pure antigens. Mice that received the new nanovaccine were more resistant to pneumonia caused by this bacterium than those that received pure antigen. Conclusion and implication: Overall, it can be said that PLGA nanoparticles could deliver their internal antigens (OMPs) well to the immune system of mice and stimulate humoral immunity in these animals, thus protecting them against pneumonia caused by A. baumannii.

Synchronous chemotherapy and radiotherapy, termed chemoradiation therapy, is now an important standard regime for synergistic cancer treatment. For such treatment, nanoparticles can serve as improved carriers of chemotherapeutics into tumors and as better radiosensitizers for localized radiotherapy. Herein, we designed a Schottky-type theranostic heterostructure, Bi2S3–Au, with deep level defects (DLDs) in Bi2S3 as a nano-radiosensitizer and CT imaging contrast agent which can generate reactive free radicals to initiate DNA damage within tumor cells under X-ray irradiation. Methotrexate (MTX) was conjugated onto the Bi2S3–Au nanoparticles as a chemotherapeutic agent showing enzymatic stimuli-responsive release behavior. The designed hybrid system also contained curcumin (CUR), which cannot only serve as a nutritional supplement for chemotherapy, but also can play an important role in the radioprotection of normal cells. Impressively, this combined one-dose chemoradiation therapeutic injection of co-drug loaded bimetallic multifunctional theranostic nanoparticles with a one-time clinical X-ray irradiation, completely eradicated tumors in mice after approximately 20 days after irradiation showing extremely effective anticancer efficacy which should be further studied for numerous anti-cancer applications.

Cancer is among the most common causes of death in the world that affects a large number of people every year. Curcumin is one of the natural anticancer therapeutics with little or no negative effects. However, due to its hydrophobic nature, poor bioavailability, limited gastrointestinal uptake, and fast metabolism, its therapeutic applications are constrained. Therefore, the Bovine Serum Albumin-Coated Copper Sulfide anoparticles (CuS@BSA) for curcumin (CUR) drug delivery were synthesized and characterized, and then curcumin release from the nanosystem. Hemotoxicity, and cytotoxicity was investigated. This study involved the one-step synthesis of CuS@BSA nanoparticles first, followed by the addition of CUR. Then the synthesized nanoparticles were characterized employing Scanning Transient Electron Microscopy (STEM), Ultraviolet–visible spectroscopy (UV–vis) and Fourier-transform infrared spectroscopy (FT-IR) techniques. The Size and surface charge (zeta potential) of synthesized nanoparticles were determined by Dynamic Light Scattering (DLS) to be 120 nm and −13 eV, respectively. The results showed that the CUR loading was around 15% and also the release pattern of CUR was dependent on pH and increased in an acidic environment. The results of the hemolysis assay showed that the synthesized nanoparticles are not hemotoxic. The investigation of the cytotoxic effects of synthesized nanoparticles on cancer cells demonstrated that CuS@BSA nanoparticles did not exhibit any toxicity and therefore are an appropriate candidate for drug delivery.

Background: Nanotechnology has introduced valuable carriers for vaccine delivery. The success of vaccination depends on many factors, such as the intact and safe presentation of vaccine candidates to immune cells. We have conjugated branched PEI-2k and oleic acid (OL) as the building block of the cationic micelle. We aimed to introduce a novel carrier for vaccine candidates. Materials and Methods: We conjugated polyethyleneimine and OL (POA) to synthesize the building blocks of cationic micelles. The critical micelle concentration (CMC), size and zeta potential of micelles, and their stability in 60 days were determined. Loading, encapsulation efficiency, and in vitro release study were assessed using bovine serum albumin (BSA) as a protein model. Furthermore, the cytotoxicity and hemocompatibility of developed nanosized micelles were evaluated to ascertain the biocompatibility of fabricated micelles. Cell uptake of cationic micelles in the macrophage cell line was also followed up. Results: The conjugation of two polymer parts was confirmed by Fourier transform infrared spectroscopy and 1H nuclear magnetic resonance techniques. The CMC of the developed micelles was around 5.62 × 10 − 8 mg/ml, whereas the loading and encapsulation efficiencies were 16.5% and 70%, respectively. The size and zeta potential of the cationic micelles were 96.53 ± 18.53 nm and 68.3 mV, respectively. The release of BSA from POA micelles after 8 and 72 hours was 8.5% and 82%, respectively. Finally, fluorescence microscopy showed that the prepared micelles were successfully and effectively taken up by RAW264.7 cells. Conclusion: These results may provide a cutting-edge vaccine delivery solution and open up a new avenue for future vaccine research.

High atomic number Z, nanoparticles are able to enhance the photoelectric and Compton effects under X-Ray irradiation resulting the increase of radiation therapy efficacy. To achieve enhanced radiation therapy, Bi2S3 biocompatible particles coated with bovine serum albumin (BSA) (Bi2S3@BSA HNPs) were prepared through a BSA-mediated biomineralization procedure under green conditions. Then, to achieve improved chemo-radiation therapy against HT-29 cancer cells, curcumin (CUR) as natural anti-cancer therapy agent loaded on the Bi2S3@BSA (Bi2S3@BSA@CUR HNPs). Next, this synthesized nanodrug was evaluated for physical and chemical properties and in vitro cytotoxicity studies. Here, in vitro enhanced chemo-radiation combination therapy power was evaluated against HT-29 cell line under 2 Gy and 6 Gy X-ray irradiation doses. The Bi2S3@BSA HNPs without irradiation rarely affect cell viability which shown the non-toxicity of Bi2S3@BSA HNPs. The result of this study proved that Bi2S3@BSA@CUR HNPs can be used as both proficient vehicles for effective delivery of CUR and radiosensitizer in the treatment of cancer. In addition, the result of this study confirmed that the combination of high Z-element nanoradiosensitizer, Bi2S3@BSA HNPs, with a natural anti-cancer drug, CUR, enhanced therapeutic power against HT-29 cells.

Artemisinin is used as an antimalarial and anticancer agent with minimal toxic effects on the host body. Biotin-PEG-PCL polymers have been used for targeted drug delivery to cancer, as well as to improve the pharmacokinetics of the drug and reduce its effects. In this study, biotin-conjugated copolymers were fabricated with polymerization of the ring opening method and the properties of copolymer and nanoparticles were investigated using various techniques. The toxicity of artemisinin and its nanoparticles have been investigated on MCF-7 and normal HFF2 cells. The results showed that the encapsulation efficacy of artemisinin in nanoparticles was 45.5 ± 0.41%. The release profile of the drug indicates that the release is slow and controlled and is approximately pH dependent. The results of artemisinin cell culture on human breast cancer cells showed that biotin-PEG-PCL nanoparticles had an inhibitory effect on MCF-7 cells and had no toxic effects on HFF2 cells. Anticancer activity in vivo in the 4T1 breast cancer model showed that tumour volumes were decreased up 40 mm3 by ART-loaded micelles and 76 mm3 by free ART, compared to the control group (2150 mm). In vivo results showed that this formulation significantly increases the accumulation of substances in the tumours. Therefore, the molecular formulation of ART-based copolymers can be a desirable process for cancer treatment purposes.

In this study, we developed a green and facile top–down approach for the production of nano-sized graphene oxide (GO) like flakes from natural sources by sonochemistry. The long carbon flakes produced by the pyrolysis of waste onion sheathing were then converted into uniform nano-sized GO-like flakes (6.6 ​± ​2.4 ​nm) by sonication in deionized water. The effect of time on the size of GO-like nanostructures obtained by ultrasonication of bio-sourced carbon flakes has been extensively investigated. Transmission electron microscopy (TEM) and atomic force microscopy (AFM) were used to determine the size of these nanostructures. The changes in the surface chemical composition of the nanostructures during the sonochemical treatment were analyzed by X-ray photoelectron spectroscopy (XPS). Furthermore, cytotoxicity and hemocompatibility of nano-sized GO-like flakes were evaluated. Pyrolysis of waste onion sheathing followed by sonication could therefore be explored as a highly cost-effective, eco-friendly, uncomplicated, green, and efficient way of producing large-scale nano-sized GO-like carbon flakes.

An open-label, single-dose, randomized, 2-way crossover study was conducted in fasted healthy Iranian male volunteers. Eligible participants randomly assigned in a 1:1 ratio to be given one tablet of the test or reference formulation, followed by a 2-week washout period and administration of the exchange formulation. A quick and susceptible Liquid Chromatography–tandem Mass Spectrometry (LC-MS) method for the evaluation of amlodipine and atorvastatin in tablets.Revealing of analysts was achieved by tandem mass spectrometry with ElectroSpray Ionization (ESI) interface in positive ion mode was operated under the multiple-reaction monitoring mode. The assay results ascertain the presence and compendia quality of amlodipine and atorvastatin in all these products. The validation tests on the developed method indicated an acceptable degree of linearity, sensitivity, precision, accuracy and recovery for the method. The intra-day and inter-day precision and accuracy results were well within the acceptable limits. The method is rapid, simple, very stable and specific for the partition, assignment, evaluation of amlodipine and atorvastatin healthy Iranian adult male volunteers.

The present study aimed to synthesize triacetin-microemulsion (T-ME) and T-ME hybridized with bovine serum albumin nanoparticles (T-BSA-ME) having narrow particle size distribution and versatile carrier systems as a novel microemulsion system. The suggested ME system was characterized by Fourier Transform Infrared spectroscopy (FTIR), Differential Scanning Calorimetry (DSC), and Atomic Force Microscopy (AFM). The physicochemical properties of microemulsion system including particle size, PDI and ζ-potential, refractive index, Conductivity, %Transmittance, pH, and rheological behavior were also evaluated. In vivo biocompatibility was done using Median Lethal Dose (LD 50) calculated and trialed to evaluate the acute toxicity. In Addition, hemolysis and leukocyte proliferation assay were characterized to evaluate in-vitro biocompatibility of the suggested MEs systems. Moreover, cytotoxicity of MEs systems was also investigated on HFF-2 and HEK-293 cells. The presence of BSA NPs as a macromolecular biomaterial hybridized with T-ME reduced the cytotoxicity. The properties of the suggested MEs system proposed the T-ME hybridized with BSA-NPs as a promising candidate for co-delivery and multifunctional biomedicine applications.

Ezetimide belongs to a class of lipid lowering compounds that selectively inhibits intestinal absorption of cholesterol and related phytosterols. The purpose of this study is to establish a reliable and quick method for the assignment of ezetimibe in tablets form by high performance liquid chromatography with ultraviolet detection (HPLC-UV). A rapid and sensitive HPLC method has been developed for determination of ezetimibe in tablets formulation. Mobile phase was composed of acetonitrile-ammonium acetate (10 mM, pH 3.0), 75:25 (v/v) with a flow rate of 1 ml/min. The eluted peaks were detected by a UV detector was set at wavelength of 240 nm. The method results in excellent separation with good resolution of analyte. Standard curves were linear (r = 0.996) over the wide ezetimibe concentration range of 10-60.0 µg mL-1 with acceptable accuracy and precision. The limits of detection (LOD) and quantitation (LOQ) of the method were 5 and 10 µg/ml, respectively. The average drug recovery was 95.3% throughout the linear concentration range. Statistical assessment of various in vitro dissolution parameters and assay results was also conducted to establish if there were any significant difference among them. The validated HPLC method has been used successfully to study ezetimibe. Due to simplicity, rapidity and accuracy of the method, we believe that the method will be useful for routine quality control analysis.

It is an increased interest in the therapeutic potential of Cannabis sativa L. (marijuana) for treatment of multiple sclerosis and HIV neuropathy. Because of limitation in cultivation of this plant, an efficient hairy root induction system for Cannabis sativa L. was developed in the present study. Agrobacterium rhizogenes mediated transformation performed by two different co-cultivation mediums and four different bacterial strains including A4, ATCC15834, MSU440, and A13 (MAFF-02-10266). Genomic DNA from putative transgenic hairy root lines and the control root was extracted using a modified CTAB protocol. Molecular analysis of transformed root lines was confirmed by polymerase chain reaction using specific primers of the rolB gene. The transformation frequency by Murashige and Skoog co-cultivation medium resulting in hairy root induction frequencies of 42.3%, 46.3%, 68.6% and 39.3% by A4, ATCC15834, MSU440, and A13 strains, respectively. There was no significant difference between MS or ½ MS co-cultivation mediums. This study established a reliable protocol for induction of hairy roots of C. sativa. The best A. rhisogenes strain was MSU440. It was observed no significant difference between MS and ½ MS co-cultivation mediums on transformation frequency.

Tolterodine tartrate, is a new, potent and competitive muscarinic receptor antagonist in clinical development for the treatment of urge incontinence and other symptoms of unstable bladder. The purpose of this study is to establish a reliable and quick method for the assignment of tolterodine tartrate by high performance liquid chromatography with ultraviolet detection (HPLC-UV). A rapid and sensitive high performance liquid chromatographic (HPLC) method has been developed for determination of tolterodine tartrate. Mobile phase was composed of phosphate acetate 0.1 M (pH 2.5)-acetonitrile (50:50 v/v) with a flow rate of 1.2 ml/min. The eluted peaks were detected by a UV detector was set at wavelength of 285 nm. The method was validated in the range of tolterodine tartrate concentrations from 10 to 100 µg/ml. The limits of detection (LOD) and quantitation (LOQ) of the method were 5 and 10 µg/ml, respectively. The average drug recovery was 98.20 % throughout the linear concentration range. The average within-run and between-run accuracy values of 98.56 % and 99.11 % respectively. Statistical assessment of various in vitro dissolution parameters and assay results was also conducted to establish if there were any significant difference among them. The validated HPLC method has been used successfully to study tolterodine tartrate.

Cefixime is essential member of orally energetic third generation cephalosporin and has tremendous activity aligned with many pathogens. The virtual bioavailability of a newly industrial dispersible tablet as compared with a recognized identified formulation. A simple and available reversed-phase HPLC method with UV detection has been urbanized and validate for cefixime evaluate in human plasma using a C18 analytical column and a mobile phase of tetrabutylammonium hydroxide (pH 6.5)-acetonitrile (3:1 v/v). The detection wavelength was 280 nm. To method observed major linear response-concentration association all through the cefixime concentration range of 15-100 ng/ml, with the average accuracy within-run and between-run values of 97.29% and 99.27%. The average drug recovery from plasma was 98.2% throughout the linear concentration range. The limits of detection (LOD) and quantitation (LOQ) of the method were 5 and 15 ng/ml, respectively. The method is quick, easy, very steady and precise for the partition, assignment, evaluation of cefixime in human plasma.

A simple and sensitive high performance liquid chromatography-electrospray ionization mass spectrometry method has been evaluated for the assignment of clonidine hydrochloride in human plasma. The mobile phase composed of acetonitrile–water 60:40 (v/v), and 0.2% formic acid 20 µL of sample was chromatographically analyzed using a repacked ZORBAX-XDB-ODS C18 column (2.1 mm×30 mm, 3.5 micron). Detection of analytes was achieved by tandem mass spectrometry with electrospray ionization (ESI) interface in positive ion mode was operated under the multiple-reaction monitoring mode (m/z 230.0 → 213). Sample pretreatment involved in a one-step protein precipitation (PPT) with methanol and percholoric acid (HClO4) of 0.15 mL plasma. Standard curve was linear (r = 0.998) over the concentration range of 0.01-10.0 ng/ml and showed suitable accuracy and precision. The limit of quantification (LOQ) was 0.01 ng/ml. The method is rapid, simple, very steady and precise for the separation, assignment, evaluation of clonidine healthy in human plasma.

Niosomes (the nonionic surfactant vesicles), considered as novel drug delivery systems, can improve the solubility and stability of natural pharmaceutical molecules. They are established to provide targeting and controlled release of natural pharmaceutical compounds. Many factors can influence on niosome construction such as the preparation method, type and amount of surfactant, drug entrapment, temperature of lipids hydration, and the packing factor. The present review discusses about the most important features of niosomes such as their diverse structures, the different preparation approaches, characterization techniques, factors that affect their stability, their use by various routes of administration, their therapeutic applications in comparison with natural drugs, and specially the brain targeting with niosomes-ligand conjugation. It also provides recent data about the various types of ligand agents which make available active targeting drug delivery to the central neuron system. This system has an optimistic upcoming in pharmaceutical uses, mostly with the improving availability of innovative schemes to overcome blood-brain barrier and targeting the niosomes to the brain.

Purpose: A simple and available reversed-phase high performance liquid chromatography (HPLC) method with UV detection has been developed and validated for mycophenolic acid (MPA) assay in human plasma. Methods: MPA was extracted from plasma with protein precipitation method by acetonitrile: percholeric acid: methanol (75:5:20 v/v/v). The drug separation was achieved using a C8 analytical column and a mobile phase of 0.1M triethylammonium phosphate (pH=5.4)-acetonitril (65:35, v/v), with a flow rate of 1.5 ml/min. The detection wavelength was 304 nm. Limit of detection (LOD) of the method was determined as the lowest MPA concentration producing a signal-to-noise (S/N) ratio of about 3. Limit of quantitation (LOQ) was determined as the lowest MPA concentration capable of being quantitated with enough accuracy and precision. Results: The method showed significant linear response-concentration relationship throughout the MPA concentration range of 0.2-10 µg/ml. A typical linear regression equation of the method was: y = 8.5523 x + 0.094, with x and y representing MPA concentration (in µg/ml) and peak height respectively, and the regression coefficient (r) of 0.9816. The average within-run and between-run variations of 7.81 and 4.78 percent. The average drug recovery from plasma was 95.24 percent throughout the linear concentration range. The limits of detection (LOD) and quantitation (LOQ) of the method were 0.05 and 0.2 µg/ml, respectively. The practical applicability of the method was proven throughout a bioequivalence study. Conclusion: The results showed the acceptable degree of linearity, sensitivity, precision, accuracy and recovery for the method. The method was used successfully for quantitation of MPA in plasma samples of healthy volunteers throughout a bioequivalence study.

A rapid and sensitive liquid chromatography–tandem mass spectrometry (LC-MS) method was developed for the determination of enalapril and enalaprilat in human plasma. Detection of analytes was achieved by tandem mass spectrometry with electrospray ionization (ESI) interface in positive ion mode which was operated under the multiple-reaction monitoring mode. Sample pretreatment was involved in a one-step protein precipitation (PPT) with per chloric acid of plasma. The reconstituted samples were chromatographed on C18 column by pumping methanol: water: acid formic 74:24:2 (v/v) at a flow rate of 0.2 mL/min. Each plasma sample was chromatographed within1.25 min. The standard curves were found to be linear in the range of 0.1–20 ng/mL of enalapril and enalaprilat with mean correlation coefficient of ≥0.999 for each analyte. The intra-day and inter-day precision and accuracy results were well within the acceptable limits. The limit of quantification (LOQ) was 0.1ng/ml for enalapril and enalaprilat. The lower limit of detection (LOD) was 0.08 ng/ml for enalapril and enalaprilat.

Phenylalanine (Phe) is the most reliable indicator for the diagnosis of phenylketonuria (PKU). The purpose of this study is to establish a reliable and quick method for the assignment of Phe in peripheral capillary blood from newborns and children by high performance liquid chromatography with ultraviolet detection (HPLC-UV). PKU is an inborn error of metabolism characterized by the inability of the body to use Phe. A rapid and sensitive high performance liquid chromatographic (HPLC) method has been developed for determination of Phe in plasma. The method uses a protein precipitation step with sulfosalicilic acid for sample preparation by separation on a Nova-pack C18 column using sodium acetate buffer and acetonitrile (94: 6 v/v) adjusted to pH 6.5 with glacial acetic acid. The eluted peaks detected by a UV detector was set at wavelength of 215 nm. The method was validated in the range of Phe concentrations from 0.1 to 20 µg/ml. The limits of detection (LOD) and quantitation (LOQ) of the method were 0.05 and 0.1 µg/ml, respectively. The average drug recovery from plasma was 88.60 percent throughout the linear concentration range., with the average within-run and between-run accuracy values of 103.3 and 115.350, respectively. The method is quick, easy, very steady and precise for the screen, assignment, and evaluation of Phe in human plasma by HPLC, which is particularly a useful way for screening and diagnosis of PKU and monitoring of a diet therapy.

Purpose: Among the potent anticancer agents, curcumin is known as a very efficacious against many different types of cancer cells, but its clinical applications has been limited because of hydrophobicity, low gastrointestinal absorption, poor bioavailability and rapid metabolism. In this way, a novel micellar delivery system with mPEG–PCL was synthesized and the release profile of the curcumin from the drug-loaded micelles was evaluated. Methods: In this study, curcumin was encapsulated within monomethoxypoly(ethylene glycol)-poly(ε-caprolactone) (mPEG-PCL) micelles through a single-step nano-precipitation method, leading to creation of curcumin-loaded mPEG-PCL (Cur/mPEG-PCL) micelles. Di-block mPEG-PCL copolymers were synthesized and used to prepare micelles. mPEG-PCL copolymer was characterized in vitro by HNMR, FTIR, DSC and GPC techniques. Then, mPEG–PCL copolymers with curcumin were self-assembled into micelles in aqueous solution. The resulting micelles were characterized further by various techniques such as dynamic light scattering (DLS) and atomic force microscopy (AFM). Results: The findings showed the successful formation of smooth and spherical curcumin-loaded micelles. The encapsulation efficiency of curcumin was 88 ± 3.32%. The results of AFM revealed that the micelles have spherical shapes with size of 73.8 nm. The release behavior of curcumin from micelles was compared in different media. In vitro release of curcumin from curcumin-entrapped micelles was followed remarkably sustained profile. The sustained release of drug was hypothetically due to the entrapment of curcumin in core of micelles. Conclusion: The results indicate the successful formulation of curcumin loaded m-PEG/PCL micelles. From the results, iIt can be concluded that curcumin m-PEG-PCL micelles may be considered as an effective treatment strategy for cancer in the future.

Methoxypoly(ethylene glycol) Poly(caprolactone) (mPEG–PCL) copolymers are important synthetic biomedical materials with amphiphilicity, controlled biodegradability, and great biocompatibility. They have great potential application in the fields of nanotechnology, tissue engineering, pharmaceutics, and medicinal chemistry. mPEG–PCL copolymers are biodegradable, biocompatible, and semi-crystalline copolymers having a very low glass transition temperature. Due to their slow degradation, mPEG–PCLs are ideally suitable for long-term delivery extending over a period of more than one year. This has led to its application in the preparation of different delivery systems in the form of microspheres, nanospheres, micelles, polymersomes, nanogels, and implants. Various categories of drugs have been encapsulated in mPEG–PCL for targeted drug delivery and for controlled drug release. As a research acmes incurrent years, mPEG–PCL copolymers and their end group-derived nanoparticles can progress the drug loading of hydrophobic drugs, enhancement bioavailability, escape being engulfed by phagocytes, reduce the burst release, and augment the circulation time of drugs in blood. Furthermore, these nanoparticles can assemble in inflammation or target locations to improve drug efficiency and diminish toxicity because of their smaller particle size and modified surface. Recent advances in mPEG–PCL block copolymer nanoparticles, counting the synthesis of mPEG–PCL and the preparation of mPEG–PCL nanoparticles, were discussed in this study, especially the drug release and adjustable characteristics of mPEG–PCL nanoparticles and their application in pharmaceutical preparations.

The Clavulanic acid (CLV) is a suicide inhibitor of bacterial beta-lactamase enzymes from Streptomyces clavuligerus. In the present study, a reliable drug delivery system using poly (ε-caprolactone)-poly (ethylene glycol)-poly (ε-caprolactone) (PCL-PEG-PCL) was synthesized and the release profile of the CLV from the drug-loaded polymersomes was evaluated. In this study, CLV was encapsulated within PCL-PEG-PCL nanoparticles by a double emulsion technique (w/o/w), leading to creation of CLV-loaded PCL-PEG-PCL (CLV/PCL-PEG-PCL) polymersomes. Characterization, stability of polymersomes, the particle size was determined by DLS. The release profile of the CLV from the polymersomes which ready by the drug-loaded copolymer, was evaluated. Our studies resulted in a successful establishment of uniformity and spherical CLV-loaded PCL-PEG-PCL polymersomes. The loading efficiency of CLV was 16.00 ± 1.45%. The results of DLS show that the polymersomes have spherical shapes with size of 113 nm. In vitro release of CLV from CLV-entrapped polymersomes was remarkably sustained. The results indicate the successful formulation of curcumin loaded PCL-PEG-PCL polymersomes.

A rapid and selective liquid chromatography–mass spectrometry (LC–MS) method was developed for the evaluation of amlodipine and atorvastatin in human plasma. Detection of analytes was assigned by tandem mass spectrometry with electrospray ionization interface in positive ion mode was applied under the multiple-reaction monitoring mode (MRM). Analytes were extracted from plasma by simple liquid–liquid extraction technique using ethyl acetate. The reconstituted samples were chromatographed on C18 column by pumping acetonitrile–water (10 mM CH3COONH4, pH 3.0) = 70:30 (v/v) at a flow rate of 0.15 mL/min. The standard curves were assigned to be linear in the range of 0.2–20 ng/mL for atorvastatin and 0.1–10 ng/mL for amlodipine with mean correlation coefficient of ≥0.999 for each analyte. The intra-day and inter-day precision and accuracy results were well within the acceptable limits. The urbanized evaluate method was successfully applied to validation of amlodipine and atorvastatin in human plasma.

The method uses a protein precipitation step with methanol. Mobile phase was composed of phosphate buffer 0.1 M (pH 6.5)-acetonitrile (70:30 v/v) with a flow rate of 1 ml/min. The eluted peaks were detected by a UV detector was set at wavelength of 205 nm. The method was validated in the range of selegiline concentrations from 10 to 100 ng/ml. The limits of detection and quantitation of the method were 5 and 10 ng/ml, respectively. The average drug recovery from plasma was 98.20% throughout the linear concentration range. The average within-run and between-run accuracy values of 99.24 and 98.78, respectively. The method is quick, easy, very steady, and precise for the screen, assignment, and evaluation of selegiline in human plasma by HPLC.
