Abstract

BACKGROUND: Hypertension is a major cause of death worldwide. Although arsenic exposure has been associated with the risk of hypertension, this association appears nonuniform due to inconsistent results from studies conducted in different populations. Moreover, hypertension is a complex condition with multiple underlying mechanisms and factors. One factor is impaired production and bioavailability of vascular nitric oxide (NO). However, the implications of the effects of arsenic exposure on circulating NO and its association with hypertension in humans are largely unknown. OBJECTIVE: We investigated the dose–response relationship between arsenic exposure and hypertension with vascular NO levels as a potential mediator of arsenic-related hypertension in individuals exposed to a broad range of arsenic. METHODS: A total of 828 participants were recruited from low- and high-arsenic exposure areas in Bangladesh. Participants’ drinking water, hair, and nail arsenic concentrations were measured by inductively coupled plasma mass spectroscopy. Hypertension was defined as a systolic blood pressure (SBP) value of = 140 and a diastolic (DBP) value of = 90 mmHg. Serum NO levels reflected by total serum nitrite concentrations were measured by immunoassay. A formal causal mediation analysis was used to assess NO as a mediator of the association between arsenic level and hypertension. RESULTS: Increasing concentrations of arsenic measured in drinking water, hair, and nails were associated with the increasing levels of SBP and DBP. The odds of hypertension were dose-dependently increased by arsenic even in participants exposed to relatively low to moderate levels (10–50 µg/L) of water arsenic [odds ratios (ORs) and 95% confidence intervals (CIs): 2.87 (95% CI: 1.28, 6.44), 2.67 (95% CI: 1.27, 5.60), and 5.04 (95% CI: 2.71, 9.35) for the 10–50 µg/L, 50.01–150 µg/L, and >150 µg/L groups, respectively]. Causal mediation analysis showed a significant mediating effect of NO on arsenic-related SBP, DBP, and hypertension. CONCLUSION: Increasing exposure to arsenic was associated with increasing odds of hypertension. The association was mediated through the reduction of vascular NO bioavailability, suggesting that impaired NO bioavailability was a plausible underlying mechanism of arsenic-induced hypertension in this Bangladeshi population. Abstract

Ras‐association domain family 6 (RASSF6) is a tumor suppressor that interacts with MDM2 and stabilizes p53. Caenorhabditis elegans unc‐119 encodes a protein that is required for normal development of the nervous system. Humans have 2 unc‐119 homologues, UNC119 and UNC119B. We have identified UNC119 as a RASSF6‐interacting protein. UNC119 promotes the interaction between RASSF6 and MDM2 and stabilizes p53. Thus, UNC119 induces apoptosis by RASSF6 and p53. UNC119 depletion impairs DNA repair after DNA damage and results in polyploid cell generation. These findings support that UNC119 is a regulator of the RASSF6‐MDM2‐p53 axis and functions as a tumor suppressor. [ABSTRACT FROM AUTHOR]

Abstract

Elevated exposure to arsenic has been suggested to be associated with atherosclerosis leading to cardiovascular disease (CVD). However, biochemical events underlying the arsenic-induced atherosclerosis have not yet been fully documented. The aim of this study was to investigate the associations of circulating molecules involved in atherosclerosis with arsenic exposure in the individuals exposed to arsenic in Bangladesh. A total of 324 study subjects, 218 from arsenic-endemic areas and 106 from nonendemic areas in Bangladesh, were recruited. Drinking water, hair, nail, and blood samples were collected from the study subjects for analysis. Total cholesterol (TC), low-density lipoprotein (LDL), and high-density lipoprotein (HDL) levels were lower in arsenic-endemic subjects than those of nonendemic subjects. Oxidized LDL (Ox-LDL), C-reactive protein (CRP), intercellular adhesion molecule-1 (ICAM-1), and vascular cell adhesion molecule-1 (VCAM-1) levels were significantly higher in arsenic-endemic subjects than those in nonendemic subjects. All these circulating molecules showed significant correlations with arsenic exposure (water, hair, and nail arsenic concentrations), and all these relations were significant before and after adjusting for relevant covariates. Among the circulating molecules tested in this study, HDL, Ox-LDL, and CRP showed dose-response relationships with arsenic exposure. Ox-LDL/ HDL ratios were increased with the increasing concentrations of arsenic in the water, hair, and nails. Furthermore, non-HDL cholesterol and TC/ HDL ratios were significantly correlated with arsenic exposure before and after adjusting for relevant covariates. Abstract

Lead (Pb), a common toxicant is ubiquitously present in the environment. Chronic Pb exposure affects almost every organ system of human body including liver. Clerodendrum viscosum is a medicinal plant and its leaves are known to have hepatoprotective, anti‐inflammatory, and anti‐hyperglycemic activities. However, the protective effect of C. viscosum leaves against Pb‐induced hepatotoxicity is yet to be studied. Therefore, this study was designed to assess the protective effect of the aqueous extract of C. viscosum leaf (Cle) against Pb‐induced hepatotoxicity in experimental mice. Pb‐acetate was given to Pb and Pb + Cle groups interperitoneally, and Cle was supplemented to Cle and Pb + Cle groups by oral gavage. Serum biomarkers of liver function—butyrylcholinesterase (BChE), alkaline phosphatase (ALP), aspartate aminotransferase (AST), and alanine transaminase (ALT), antioxidant enzyme activities in hepatic tissue—superoxide dismutase (SOD), reduced glutathione reductase (rGR) and catalase (CAT), levels of transcription factor—nuclear factor erythroid 2‐related factor 2 (Nrf2), and inflammatory marker—interleukin‐6 (IL‐6) were analyzed. Additionally, histological analyses of hepatic tissues of all groups of experimental mice were performed. Pb‐treatment significantly increased ALP, AST, and ALT activities and decreased BChE activity compared to control mice. The antioxidant enzyme (SOD, rGR, and CAT) activities and expression of Nrf2 level were significantly (p < .05) decreased, while IL‐6 level was significantly (p < .05) increased in the hepatic tissue homogenates of Pb‐treated mice compared to the control group. Furthermore, histological examination revealed the disruption of hepatic tissue integrity in Pb‐treated mice. Notably, supplementation of Cle provided significant protection against the changes in the activities of liver function biomarkers and antioxidant enzymes, levels of Nrf2 and IL‐6, and disruption of hepatic tissue by Pb.
