14. Genomic Insights of a Methicillin-Resistant Biofilm-Producing Staphylococcus aureus Strain Isolated From Food Handlers.

Abstract

Methicillin-resistant Staphylococcus aureus (MRSA) is an important zoonotic pathogen associated with a wide range of infections in humans and animals. Thus, the emergence of MRSA clones poses an important threat to human and animal health. This study is aimed at elucidating the genomics insights of a strong biofilm-producing and multidrug-resistant (MDR) S. aureus MTR_BAU_H1 strain through whole-genome sequencing (WGS). The S. aureus MTR_BAU_H1 strain was isolated from food handlers hand swabs in Bangladesh and phenotypically assessed for antimicrobial susceptibility and biofilm production assays. The isolate was further undergone to high throughput WGS and analysed using different bioinformatics tools to elucidate the genetic diversity, molecular epidemiology, sequence type (ST), antimicrobial resistance, and virulence gene distribution. Phenotypic analyses revealed that the S. aureus MTR_BAU_H1 strain is a strong biofilm-former and carries both antimicrobial resistance (e.g., methicillin resistance; mecA, beta-lactam resistance; blaZ and tetracycline resistance; tetC) and virulence (e.g., sea, tsst, and PVL) genes. The genome of the S. aureus MTR_BAU_H1 belonged to ST1930 that possessed three plasmid replicons (e.g., rep16, rep7c, and rep19), seven prophages, and two clustered regularly interspaced short palindromic repeat (CRISPR) arrays of varying sizes. Phylogenetic analysis showed a close evolutionary relationship between the MTR_BAU_H1 genome and other MRSA clones of diverse hosts and demographics. The MTR_BAU_H1 genome harbours 42 antimicrobial resistance genes (ARGs), 128 virulence genes, and 273 SEED subsystems coding for the metabolism of amino acids, carbohydrates, proteins, cofactors, vitamins, minerals, and lipids. This is the first-ever WGS-based study of a strong biofilm-producing and MDR S. aureus strain isolated from human hand swabs in Bangladesh that unveils new information on the resistomes (ARGs and correlated mechanisms) and virulence potentials that might be linked to staphylococcal pathogenesis in both humans and animals.

We announce the genome sequence of the Staphylococcus gallinarum MTR_B001 strain isolated from the breast muscle of a chicken in 2022 in Bangladesh. This assembled genome had an estimated length of 2,889,393 bp (with 50× genome coverage), 15 contigs, 36 predicted antibiotic resistance genes, and 27 predicted virulence factor genes.

This announcement provides the genome sequence of the biofilm-forming methicillin-resistant Staphylococcus aureus MTR_V1 strain isolated from a ready-to-eat food sample in Bangladesh. Our assembled genome had a length of 2.8 Mb, 27 contigs, two CRISPR arrays, 38 predicted antibiotic resistance genes, and 66 predicted virulence factor genes.

Abstract

In this study, a series of nanocrystalline ferrites of Ni0.7Cu0.30AlxFe2-xO4 (x=0.00 to 0.10 with a step of 0.02) has been synthesized through the sol-gel auto combustion technique. The structural, morphological, dielectric, and electrical properties of the Ni-Cu spinel ferrite nanoparticles are analyzed due to the substitution of Al3+ content. The crystalline and structural characteristics of the prepared nanoparticles (NPs) have been studied employing the x-ray diffraction (XRD) spectra and FTIR analysis. The extracted XRD patterns assure the single-phase cubic spinel structure of all samples with homogeneity and no impurity, which indicates the yielding of high crystalline NPs. A slight decrease of average grain size with increment of Al3+ content is observed in the surface morphological study carried out by the field emission scanning electron microscopy (FESEM). The studied materials are found in semi-spherical shapes, showing the multi-domain grains separated by grain boundaries with some agglomerations. The chemical composition study for the synthesized NI-Cu spinel ferrites using energy dispersive x-ray (EDX) ensures the presence of each component in appropriate proportions in each sample.

Abstract

In the last years, electron density profile functions characterized by a linear dependence on the scale height showed good results when approximating the topside ionosphere. The performance above 800 km, however, is not yet well investigated. This study investigates the capability of the semi-Epstein functions to represent electron density profiles from the peak height up to 20,000 km. Electron density observations recorded by the Van Allen Probes were used to resolve the scale height dependence in the plasmasphere. It was found that the linear dependence of the scale height in the topside ionosphere cannot be directly used to extrapolate profiles above 800 km. We find that the dependence of scale heights on altitude is quadratic in the plasmasphere. A statistical model of the scale heights is therefore proposed. After combining the topside ionosphere and plasmasphere by a unified model, we have obtained good estimations not only in the profile shapes, but also in the Total Electron Content magnitude and distributions when compared to actual measurements from 2013, 2014, 2016 and 2017. Our investigation shows that Van Allen Probes can be merged to radio-occultation data to properly represent the upper ionosphere and plasmasphere by means of a semi-Epstein function.

43. A Conjectural Experiment to Observe the Effect of Conditional locked-down in an Epidemic

Abstract

In a pandemic like Covid-19, there are many countries of lower-earning cannot provide a complete locked-down within the duration of the detected case. The locked-down may result in famine throughout the region of underdeveloped countries after the outbreak. So, a conjectural setup of an epidemic has been studied by applying specific period of locked-down (30 days) in 5 different scenarios. The stochastic approach to the SEIR (Susceptible, Exposed, Infected and Recovered) model has been used to evaluate the dynamics and the effects of locked-down. It is observed that there exist a suitable period to apply locked-down where more susceptible escape from the infection. The effect of the early (as soon as the infected case detected) and late (with respect to the estimated peak of detected cases for no locked-down) implementation of the locked-down has also been studied and found that the late implementation of locked-down will take the least time to end the epidemic. 49. Predicting TED Talk Ratings from Language and Prosody

Abstract

We use the largest open repository of public speaking---TED Talks---to predict the ratings of the online viewers. Our dataset contains over 2200 TED Talk transcripts (includes over 200 thousand sentences), audio features and the associated meta information including about 5.5 Million ratings from spontaneous visitors of the website. We propose three neural network architectures and compare with statistical machine learning. Our experiments reveal that it is possible to predict all the 14 different ratings with an average AUC of 0.83 using the transcripts and prosody features only. Abstract

Automated prediction of public speaking performance enables novel systems for tutoring public speaking skills. We use the largest open repository---TED Talks---to predict the ratings provided by the online viewers. The dataset contains over 2200 talk transcripts and the associated meta information including over 5.5 million ratings from spontaneous visitors to the website. We carefully removed the bias present in the dataset (e.g., the speakers' reputations, popularity gained by publicity, etc.) by modeling the data generating process using a causal diagram. We use a word sequence based recurrent architecture and a dependency tree based recursive architecture as the neural networks for predicting the TED talk ratings. Our neural network models can predict the ratings with an average F-score of 0.77 which largely outperforms the competitive baseline method.
