The impact of colonic fermentation on postileal absorption of Ca, Mg, P, Cu, Fe, Mn, and Zn was investigated in 8 ileally cannulated grower pigs (initial BW = 29.1 [+ or -] 1.6 kg) according to a double 4 x 4 Latin square. A semi-purified diet was supplemented with 5.20% low viscous, low fermentable cellulose (CEL), 6.25% high viscous, low fermentable carboxymethyl-cellulose (CMC), 8.95% low viscous, high fermentable oat [beta]-glucan (LG), or 9.25% high viscous, high fermentable oat [beta]-glucan (HG), resulting in 5% actual added nonstarch polysaccharides (NSP) in the diets. Because of the intrinsic mineral content in LG and HG, pigs receiving the LG and HG diets had a greater (P < 0.05) daily intake of Mg, P, Cu, Fe, Mn, and Zn, and also Ca for the HG diet compared with the CEL and CMC diets. Different amounts of minerals reached the large intestine for the 4 diets as indicated by the 60 to 86% less (P < 0.05) ileal flow of Ca, Mg, P, and Fe for CMC compared with CEL and HG. Apparent mineral retention was generally less (P < 0.05) for CEL compared with CMC. Regression analyses indicated that postileal flux of Ca, Cu, and Zn were related ([R.sup.2] = 0.24 to 0.99; P < 0.05) to short-chain fatty acid (SCFA) concentrations in feces. Postileal Ca absorption was negatively related ([R.sup.2] = 0.24; P < 0.05) to fecal total SCFA concentrations when SCFA concentrations were greater than 95.6 mmol/kg of DM. Furthermore, postileal Zn ([R.sup.2] = 0.99; P < 0.001) and Cu secretion ([R.sup.2] = 0.94; P < 0.001) decreased with increasing total SCFA concentrations in feces. Additionally, postileal secretion of Fe increased ([R.sup.2] = 0.20; P < 0.05) with increasing 16S rRNA gene copies of Enterobacteriaceae in feces, whereas the secretion of Cu decreased ([R.sup.2] = 0.25; P < 0.01) with increasing gene copies of Enterobacteriaceae. Overall, the apparent retention of Ca, Mg, and P was 27 to 85% less (P < 0.05) for CEL and HG than for CMC, whereas the apparent retention of Fe, Mn, and Zn was less (P < 0.05) for CEL than for CMC, LG, and HG. In conclusion, these data indicate that the stimulation of fermentation by dietary NSP affects net mineral flux in the large intestine that, in turn, can influence mineral excretion in feces. Additionally, negative effects of CEL on apparent retention may increase the daily requirement for minerals of grower pigs. An improved technique to study kinetics and quantitative absorption of nutrients in pigs is described. Three female pigs (35 kg of BW) were surgically modified with catheters in the hepatic portal vein and carotid artery and an ultrasonic flow probe around the portal vein. Catheter placement and patency was secured using distal modifications (rings and holes) and nonabsorbable suture. Catheters and flow probe cable were tunneled subcutaneously after exteriorization for further protection. Fibrosis and adhesions in the body cavity were minimized by avoiding excessive manipulation and drying of viscera. Pigs were supported during recovery by intravenous fluid therapy of AA and electrolytes until regular feeding resumed. Catheters were flushed daily with heparinized saline (200 IU/L). After 10 d, pigs were fed a diet based on wheat and soybean meal for 6 consecutive 7-d periods. On d 7, blood was collected postprandially every 15 min from -15 to 60 min, 30 to 240 rain, 60 to 480 min, and 120 to 720 rain. Blood flow was measured simultaneously. Plasma was analyzed for glucose, and net glucose absorption was calculated from plasma portal-arterial differences x plasma flow [blood flow x (1--hematocrit)]. The specific improvements for long-term use of this model are distal modifications of the catheters, postoperative treatment using parental nutrition and gut motility drug, prevention of infection of body cavity by further tunneling of catheters and blood flow probe cable, and use of ultrasonic blood flow probes and meter. Blood flow measurements using an ultrasonic blood flow probe was not changed after 52 d compared with 10 d post-surgery, indicating the reliability of this model. Carrasco et al. report a method that can predict the lipid nanoparticles (LNP) pKa from the structure of the ionizable lipid. They investigate the delivery efficiency for intramuscular and intravascular administration and propose design principles to limit off-target systemic distribution and expression for mRNA LNP vaccines.

The relative contribution of novel fibers such as polydextrose and soluble corn fiber (SCF) to the human gut microbiome and its association with host physiology has not been well studied. This study was conducted to test the impact of polydextrose and SCF on the composition of the human gut microbiota using 454 pyrosequencing and to identify associations among fecal microbiota and fermentative end-products. Healthy adult men (n = 20) with a mean dietary fiber (DF) intake of 14 g/d were enrolled in a randomized, double-blind, placebo-controlled crossover study. Participants consumed 3 treatment snack bars/d during each 21-d period that contained no supplemental fiber (NFC), polydextrose (PDX; 21 g/d), or SCF (21 g/d) for 21 d. There were no washout periods. Fecal samples were collected on d 16-21 of each period; DNA was extracted, followed by amplification of the V4-V6 region of the 16S rRNA gene using barcoded primers. PDX and SCF significantly affected the relative abundance of bacteria at the class, genus, and species level. The consumption of PDX and SCF led to greater fecal Clostridiaceae and Veillonellaceae and lower Eubacteriaceae compared with a NFC. The abundance of Faecalibacterium, Phascolarctobacterium, and Dialister was greater (P < 0.05) in response to PDX and SCF intake, whereas Lactobacillus was greater (P < 0.05) only after SCF intake. Faecalibacterium prausnitzii, well known for its antiinflammatory properties, was greater (P < 0.05) after fiber consumption. Principal component analysis clearly indicated a distinct clustering of individuals consuming supplemental fibers. Our data demonstrate a beneficial shift in the gut microbiome of adults consuming PDX and SCF, with potential application as prebiotics. [ABSTRACT FROM AUTHOR]

Central neuraxial blockade is a commonly performed anaesthetic technique for lower limb orthopaedic surgeries. Although it is considered a safe and reliable technique, occasionally an unexpectedly high or low level of block can be achieved due to accidental injection of local anaesthetic in a meningeal plane other than that desired. Here we report a young male who complained of aphasia and difficulty in breathing immediately after spinal anaesthesia. He had a high sensory block extending to C3 dermatome. His haemodynamics remained stable throughout the surgery. Speech returned to normal in 15 minutes. Subdural block was speculated as a cause for this unusual presentation. [ABSTRACT FROM AUTHOR]

Bronchospasm although is a rare event under spinal anaesthesia, has the potential to become an anaesthetic disaster if not recognized and managed properly. Ranitidine induced bronchospasm is a rare event in anaesthesia practice. Cystic hygroma is a benign tumor composed of large lymph containing cysts mainly in head and neck region. Perioperative management of such cases is a real challenge encountered by the anaesthesiologist in view of the difficult airway management and postoperative respiratory complications. We hereby report a case of 10 day old child presented with gross and huge swelling on the left side of the neck with difficulty in feeding. It was diagnosed to be cystic hygroma and a decision was made to excise the swelling, to enable the child to swallow and thrive better. Difficult intubation was anticipated and the child was intubated with inhalation induction maintaining spontaneous respiration. Complete heart block is a significant complication following surgical closure of ventricular septal defect. Pacemakers are being used for increased frequency for management of complete heart block in these patients. Cystic hygroma (CH) is a benign tumour composed of large lymph containing cysts. Lymphangiomas of head and neck region frequently present challenges to the anaesthesiologists due to extension in the neck, difficult airway and postoperative complications. We hereby describe the anaesthetic management of a one and a half month old baby presenting with large cystic hygroma on the left side of neck. This mass was large enough to disturb swallowing and breathing. Therefore, it was decided to excise it surgically. Difficult intubation was anticipated and inhalational induction with intubation in a spontaneously breathing child was planned. Antimicrobial resistance (AMR) is a significant threat that demands surveillance to identify and analyze trends of the emerging antibiotic resistance genes (ARGs) and potential microbial carriers. The influent of the wastewater treatment plants (WWTPs) reflects the microbes derived from the population and effluent being the source of dissemination of potential pathogenic microbes and AMR. The present study aimed to monitor microbial communities and antibiotic resistance genes in WWTPs employing a whole metagenome shotgun sequencing approach. The samples were collected from a sewage treatment plant (STP) and a common effluent treatment plant (CETP) in Delhi, India. The results showed the influent of STP to be rich in Bifidobacterium, Bacteroides, Escherichia, Arcobacter, and Pseudomonas residents of gut microbiota and known to cause diseases in humans and animals; whereas the CETP sample was abundant in Aeromonas, Escherichia, and Shewanella known to be involved in the degradation of different compounds. Interestingly, the effluent samples from both STPs and CETP were rich in microbial diversity, comprising organic and xenobiotic compound degrading and disease-causing bacteria, indicating the effluent being the source of dissemination of concerning bacteria to the environment. The functional profile at both sites displayed similarity with an abundance of housekeeping function genes as analyzed by Clusters of Orthologous Genes (COG), KEGG Orthology (KO), and subsystem databases. Resistome profiling by MEGARes showed the dominance of ARGs corresponding to beta-lactams having relative abundance ranging from 16% to 34% in all the metagenome datasets, followed by tetracycline (8%-16%), aminoglycosides (7%-9%), multi-drug (5%-9%), and rifampin (3%-9%). Also, AMR genes oxa, ant3-DPRIME, and rpoB, which are of clinical importance were predominantly and most prevalently present in all the samples. The presence of AMR in effluents from both types of treatment plants indicates that wastewater from both sources contributes to the spread of pathogenic bacteria and resistance genes, increasing the environmental AMR burden and therefore requires tertiary treatment before discharge. Pyrolysis, a thermochemical conversion approach of transforming plastic waste to energy has tremendous potential to manage the exponentially increasing plastic waste. However, understanding the process kinetics is fundamental to engineering a sustainable process. Conventional analysis techniques do not provide insights into the influence of characteristics of feedstock on the process kinetics. Present study exemplifies the efficacy of using machine learning for predictive modeling of pyrolysis of waste plastics to understand the complexities of the interrelations of predictor variables and their influence on activation energy. The activation energy for pyrolysis of waste plastics was evaluated using machine learning models namely Random Forest, XGBoost, CatBoost, and AdaBoost regression models. Feature selection based on the multicollinearity of data and hyperparameter tuning of the models utilizing RandomizedSearchCV was conducted. Random forest model outperformed the other models with coefficient of determination (R 2 ) value of 0.941, root mean square error (RMSE) value of 14.69 and mean absolute error (MAE) value of 8.66 for the testing dataset. The explainable artificial intelligence-based feature importance plot and the summary plot of the shapely additive explanations projected fixed carbon content, ash content, conversion value, and carbon content as significant parameters of the model in the order; fixed carbon > carbon > ash content > degree of conversion. Recent terrorist assaults have demonstrated the need for the exploration and design of sustainable and stable chemical sensors with quick reaction times combined with great sensitivity. Among several classes of chemical warfare agents, nerve agents have been proven to be the most hazardous. Even short-term exposure to them can result in severe toxic effects. Human beings inadvertently face the after-effects of these chemicals even several years after these chemicals were used. Due to the extreme toxicity and difficulty in handling, dimethyl methylphosphonate (DMMP), a simulant of nerve agents with much lesser toxicity, is frequently used in laboratories as a substitute. Having a chemical structure almost identical to those of nerve agents, DMMP can mimic the properties of nerve agents. Through this paper, authors have attempted to introduce the evolution of several chemical sensors used to detect DMMP in recent years, including field-effect transistors, chemicapacitors, chemiresistors, and mass-sensitive sensors. Antiferromagnets hosting real-space topological textures are promising platforms to model fundamental ultrafast phenomena and explore spintronics. However, they have only been epitaxially fabricated on specific symmetry-matched substrates, thereby preserving their intrinsic magneto-crystalline order. This curtails their integration with dissimilar supports, restricting the scope of fundamental and applied investigations. Here we circumvent this limitation by designing detachable crystalline antiferromagnetic nanomembranes of α-Fe 2 O 3 . First, we show-via transmission-based antiferromagnetic vector mapping-that flat nanomembranes host a spin-reorientation transition and rich topological phenomenology. Second, we exploit their extreme flexibility to demonstrate the reconfiguration of antiferromagnetic states across three-dimensional membrane folds resulting from flexure-induced strains. Finally, we combine these developments using a controlled manipulator to realize the strain-driven non-thermal generation of topological textures at room temperature. The integration of such free-standing antiferromagnetic layers with flat/curved nanostructures could enable spin texture designs via magnetoelastic/geometric effects in the quasi-static and dynamical regimes, opening new explorations into curvilinear antiferromagnetism and unconventional computing., (© 2024. Groundwater, a vital global resource, is essential for sustaining life and various human activities. However, its quality and availability face increasing threats from both natural and human-induced factors. Widespread contamination, arising from both natural origins and human activities such as agriculture, industry, mining, improper waste disposal, and wastewater release, poses significant risks to human health and water security. India, known for its dense population and pronounced groundwater challenges, serves as a prominent case study. Notably, in most of its regions, groundwater resources have been found to be severely contaminated by various chemical, biological, and radioactive contaminants. This review presents an examination of contamination disparities across various states of semi-arid and cold regions, encompassing diverse assessment methods. The studies conducted in semi-arid regions of North, South, West, and East India highlight the consistent presence of fluorides and nitrates majorly, as well as heavy metals in some areas, with values exceeding the permissible limits recommended by both the Bureau of Indian Standards (BIS) and the World Health Organization (WHO). These contaminants pose skeletal and dental threats, methemoglobinemia, and even cancer. Similarly, in cold regions, nitrate exposure and pesticide residues, reportedly exceeding BIS and WHO parameters, pose gastrointestinal and other waterborne health concerns. The findings also indicated that the recommended limits of several quality parameters, including pH, electrical conductivity, total dissolved solids (TDS), total hardness, and total alkalinity majorly surpassed.
