A practical and facile strategy for the synthesis ofiortho/i-phosphated (hetero)arylamines from readily available arylhydroxylamines and dialkyl phosphites via cascade Atherton-Todd reaction/[3,3]-rearrangement was developed. This method is amenable to various arylhydroxylamines such as phenylhydroxylamines, naphthylhydroxylamines, and pyridylhydroxylamines, has mild reaction conditions, is oxidant-free, and has good functional-group compatibility and excellent regioselectivity.

In the absence of transition-metal catalysts and additional oxidants, arylhydroxylamines can undergo an unconventional O-sulfinylation/[2,3]-sigmatropic rearrangement/rearomatization cascade with trifluoromethylsulfinyl chloride to rapidly and efficiently synthesize versatile ortho-sulfonylated aromatic amines. A new strategy for the synthesis of fluorinated anilides in the absence of metals and oxidants has been developed. This deoxyfluorination of N-arylhydroxylamines with diethylaminosulfur trifluoride (DAST) proceeded smoothly under mild conditions, and the ortho- or para-fluorinated aromatic amine products were prepared in moderate to good yields. Structurally diverse fluorinated anilides, including heterocyclic and pharmaceutically relevant molecules, can be efficiently constructed by this protocol.

We developed herein a regioselective construction of non-C2 symmetrical NOBIN-type biaryls through a cascade N-arylation and [3,3]-sigmatropic rearrangement from O-arylhydroxylamines and diaryliodonium salts under mild conditions. The employment of copper salt could inhibit the further O-arylation of the newly formed biaryl products, otherwise, O-arylated NOBIN-type products were furnished in moderate to good isolated yields. The products of this protocol can be further converted into highly valuable functional molecules and heterocycles.

Herein, we disclosed a transition metal-free cascade protocol for the construction of 2-hydroxy-2′-amino-1,1′-biaryls that are difficult to prepare by employing conventional methods. The nucleophilic substitution of arylhydroxylamines to arylammonium salts at room temperature generates transient N,O-diarylhydroxylamines that could rapidly undergo a tandem process, including a [3,3]-sigmatropic rearrangement and rearomatization to afford NOBIN analogues.

A metal- and oxidant-free, practical and efficient method for the synthesis of highly versatile and synthetically useful ortho-trifluoromethanesulfonylated anilines from arylhydroxylamines and trifluoromethanesulfinic chloride was developed. This rapid transformation proceeded smoothly with good yields and excellent ortho-selectivity in the absence of any metals or ligands. Mechanistically, the reaction comprised a noncanonical O-trifluoromethanesulfinylation of the arylhydroxylamine, and the subsequent [2,3]-sigmatropic rearrangement to afford ortho-trifluoromethanesulfonylated aniline derivatives. The practical application of this reaction was demonstrated by further conversion into a series of functional molecules under different reaction conditions.

A transition-metal free synthesis of highly functionalized 2-hydroxy-2'-amino-1,1'-biaryls from N-arylhydroxylamines has been developed. This operationally simple and readily scalable approach relies on a cascade of reactions that initially generates transient N, O-diarylhydroxylamines, via direct O-arylation, which then undergo rapid [3,3]-sigmatropic rearrangement and subsequent rearomatization to form NOBIN-type products. Herein, we developed a copper-catalyzed O-vinylation of arylhydroxylamine using vinyliodonium salts as vinylation reagents to generate a transient O-vinyl-N-arylhydroxylamine that rapidly undergoes a [3,3]-sigmatropic rearrangement and subsequent cyclization/rearomatization to form a substituted indole. A wide range of highly substituted indoles and benzoindoles can be afforded in good yields. This approach is readily scalable, and the scope and application of this process are presented.

Existing research on PTFE-based reactive materials (RMs) mostly focuses on Al/PTFE RMs. To explore further possibilities of formulation, the reactive metal components in the RMs can be replaced. In this paper, Zr/PTFE and Ti/PTFE RMs were prepared by cold isostatic pressing and vacuum sintering. The static and dynamic compressive mechanical properties of Zr/PTFE and Ti/PTFE RMs were investigated at different strain rates. The results show that the introduction of zirconium powder and titanium powder can increase the strength of the material under dynamic loading. Meanwhile, a modified J-C model considering strain and strain rate coupling was proposed. The parameters of the modified J-C model of Zr/PTFE and Ti/PTFE RMs were determined, which can describe and predict plastic flow stress. To characterize the impact-induced reaction behavior of Zr/PTFE and Ti/PTFE RMs, a quasi-sealed test chamber was used to measure the over-pressure induced by the exothermic reaction. The energy release characteristics of both materials were more intense under the higher impact.

Herein, we present a transition-metal free direct O-arylation of arylhydroxylamines employing diaryliodonium salts as arylation reagents to form transient N,O-diarylhydroxylamines that could subsequently undergo [3,3]-sigmatropic rearrangement and re-aromatization to afford structurally diverse NOBIN analogs in good to excellent yields under mild conditions.

An unprecedented copper-catalyzed intermolecular sulfur–sulfur bond insertion between aryldiazoacetates and benzenesulfonothioate has been successfully developed. Diverse α-disulfur functionalized esters were synthesized in good to excellent yields. The sulfur ylide formation and subsequent Stevens rearrangement are considered as the key steps for the discovered reaction.

A facile approach for synthesizing ortho-bromoanilides from readily available aryhydroxylamines and thionyl bromide is demonstrated in this work. Mild reaction conditions and broad scope of substrates ranging from heterocyclic structures to pharmaceutics-potential motifs are used in the reactions of this paper. Efficient bromination of ortho C H bonds of the aryhydroxylamines has been achieved. Ortho-bromoanilide products were obtained in good to excellent yields, and model scaled-up reactions of this synthetic approach are shown in this work.

Herein we disclose a novel method for the facile transfer of primary (–NH2) and secondary amino groups (–NHR) to heteroaryl- as well as arylcuprates at low temperature without the need for precious metal catalysts, ligands, excess reagents, protecting and/or directing groups. This one-pot transformation allows unprecedented functional group tolerance and it is well-suited for the amination of electron-rich, electron-deficient as well as structurally complex (hetero)arylmetals. In some of the cases, only catalytic amounts of a copper(I) salt is required.

International audience; A set of fifteen 2-naphthol-derived atropisomers were evaluated on three different cyclofructan-based chiral stationary phases (CSP). The cyclofructan CSPs were a dimethylphenyl-derivatized cyclofructan 7 (CF7-DMP), a isopropyl (CF6-P) and a R-naphthylethyl cyclofructan 6 (CF6-RN) derivative, all bonded to 5-mu m spherical fully porous silica particles packed into three 25cmx4.6mm columns (commercially available as Larihc columns). The 15 atropisomers were analyzed in the normal-phase mode with heptane/alcohol mobile phases. The CF7-DMP column was the most effective partially or fully separating 14 of the 15 compounds (93%). All 15 compounds could be separated by at least one cyclofructan column. Five compounds could be partially or fully separated by all three CSPs. The effect of ethanol, 2-propanol and butanol as 5 and 10% v/v polar modifiers in heptane was studied. A prototype 15cmx4.6mm column packed with superficially porous 2.7 mu m CF6-P bonded particles was tested with the same set of compounds and a standard HPLC system. The increased efficiency and solvent saving were significant.

Unadorned Aziridines Multiple catalytic methods have been developed to make aziridines—strained triangular carbon-nitrogen-carbon rings that function as versatile synthetic intermediates. However, the majority require protection of the nitrogen precursor with a sulfonyl group that is subsequently inconvenient to remove. Jat et al. (p. 61 ; see the Perspective by Türkmen and Aggarwal ) used a hydroxylamine derivative as the nitrogen source together with an established rhodium catalyst to prepare a wide range of unprotected aziridines, with nitrogen bonded simply to hydrogen or a methyl group.

Herein we disclose an organocatalytic aryl-aryl bond-forming process for the regio- and atroposelective synthesis of 2,2'-diamino-1,1'-binaphthalenes (BINAMs). In the presence of catalytic amounts of axially chiral phosphoric acids, achiral N,N'-binaphthyl hydrazines undergo a facile [3,3]-sigmatropic rearrangement to afford enantiomerically enriched BINAM derivatives in good to excellent yield. This transformation represents the first example of a metal-free, catalytic C(sp(2))-C(sp(2)) bond formation between two aromatic rings with concomitant de novo atroposelective installation of an axis of chirality. Density functional calculations reveal that, in the transition state for C-C bond formation, the phosphoric acid proton of the catalyst is fully transferred to one of the N-atoms of the substrate, and the resulting phosphate acts as a chiral counterion.

A transition-metal-free, regioselective direct aryl-aryl bond-forming process for the synthesis of halogenated 2-amino-2'-hydroxy-1,1'-biaryls that are currently either inaccessible or challenging to prepare using conventional methods is disclosed. The addition of ArMgX to an o-halonitrobenzene at low temperature generates a transient N,O-biarylhydroxylamine that rapidly undergoes either [3,3]- or [5,5]-sigmatropic rearrangement in one-pot to form a 2-amino-2'-hydroxy-1,1'-biaryl or 1-amino-1'-hydroxy-4,4'-biaryl, respectively. The periselectivity is controlled by the choice of solvent and temperature. This direct arylation process is also readily scalable (1-10 mmol). DFT calculations suggest that from the N,O-biarylhydroxylamine intermediate there is a low-energy stepwise pathway that involves initial Mg-mediated N-O bond cleavage followed by pathway branching toward either [3,3]- or [5,5]-rearrangement products via C-C bond formation and rearomatization.

Title compounds are synthesized in one step by new gold-catalyzed tandem bicyclization from alkynylalkenones and styrylindoles under mild conditions.

A dramatic substituent effect was found in the silver(I)-catalyzed cyclization reaction of ortho-alkynylaryl aldehyde oxime derivatives. When R is an alkyl group, the Ag(I)-catalyzed reaction in dimethylacetamide at 110 °C (conditions A) affords isoquinolines in good to excellent yields, in contrast, isoquinolin-1(2 H)-ones were produced in moderate to high yields under conditions B (dimethylformamide, room temperature) when R is an acetyl group. A plausible mechanism was proposed for this product selectivity control reaction (PSCR) by subtle structure modification.

Synthesis of 5,7-phenylquinoline from the Skraup reaction of m-terphenyl-amine and glycerol in the presence of acid is reported. Further reaction of 5,7-diphenyl-quinoline with phenyl lithium prepared in situ led to the formation of 2,5,7-triphenyl-quinoline. All of the products and their intermediates were characterized and the UV-Vis and photo-luminescence (PL) spectra of m-terphenylamine, 5,7-diphenylquinoline and 2,5,7-triphenylquinoline are also reported.

An organic acid catalyzed direct arylation of aromatic C(sp(2))-H bonds in phenols and naphthols for the preparation of 1,1'-linked functionalized biaryls was developed. The products are non-C2-symmetrical, atropoisomeric, and represent previously untapped chemical space. Overall this transformation is operationally simple, does not require an external oxidant, is readily scaled up (up to 98 mmol), and the structurally diverse 2,2'-dihydroxy biaryl (i.e., BINOL-type), as well as 2-amino-2'-hydroxy products (i.e., NOBIN-type) are formed with complete regioselectivity. Density-functional calculations suggest that the quinone and imino-quinone monoacetal coupling partners are exclusively arylated at their α-position by an asynchronous [3,3]-sigmatropic rearrangement of a mixed acetal species which is formed in situ under the reaction conditions.

A broad spectrum of O-aryloximes is readily available on treatment of inexpensive oximes with symmetrical diaryliodonium salts in the absence of a transition metal.

The synthesis of fused N-heterocycles starting from readily available 2-nitrobiaryls and phenylmagnesium bromide is disclosed.

O-aryloximes, generated from readily available and inexpensive oximes through transition-metal-free O-arylation, can either be hydrolyzed to O-arylhydroxylamines or conveniently converted to structurally diverse benzo[b]furans through an environmentally benign, one-pot [3,3]-sigmatropic rearrangement/cyclization sequence.

Normal phase chiral HPLC methods are presented for the enantiomeric separation of 30 biaryl atropisomers including 18 new compounds recently produced via a novel synthetic approach. Three new cyclofructan based chiral stationary phases were evaluated. Separations were achieved for all but six analytes and the LARIHC™ CF6-P alone provided 15 baseline separations. Effects of polar modifiers and temperature effects also were studied. Apparent thermodynamic parameters were determined by van't Hoff plots. Preparative scale methods were developed and employed resulting in the first ever isolation of these novel atropisomers in their pure enantiomeric form. Insights into the mechanism of retention and chiral discrimination are presented.

We disclose an efficient and operationally simple protocol for the preparation of fused N-heterocycles starting from readily available 2-nitrobiaryls and PhMgBr under mild conditions. More than two dozen N-heterocycles, including two bioactive natural products, have been synthesized using this method. A stepwise electrophilic aromatic cyclization mechanism was proposed by DFT calculations.

The reaction proceeds with complete regioselectivity to afford the target compounds, which are difficult to prepare using other methods.

We disclose a facile, aerobic, transition-metal-free, direct, and regiospecific mono-α-arylation of ketones to yield aryl benzyl and (cyclo)alkyl benzyl ketones with substitution patterns that are currently inaccessible or challenging to prepare using conventional methods. The transformation is operationally simple, scalable, and environmentally friendly. There is no need for pre-functionalization (i.e., α-halogenation or silyl enol ether formation) or the use of specialized arylating agents (i.e., diaryliodonium salts). DFT calculations suggest that the in situ-generated enolate undergoes direct C-C bond formation with the nitroarene followed by regioselective O2-mediated C-H oxidation.

The present new method to synthesize highly substituted cyclopenta[c]furans is applicable to different alkenylalkenones and a wide range of alkynes (III).

The exo/endo-selectivity in the tandem heterocyclization/formal [4 + 3] cycloaddition of alkynes (I) with furan (II) is shown to be switchable by subtle choice of a gold(I) or silver(I) catalyst, in general.

A novel catalytic exo/endo selectivity-controllable tandem heterocyclization/formal [4+3] cycloaddition was developed, which provides rapid, efficient and stereoselective access to highly fused polyheterocycles from readily available 2-(1-alkynyl)-2-alken-1-ones and 1,3-diphenylisobenzofuran under mild conditions.
