Abstract Background Fritillaria ussuriensis is an endangered medicinal plant known for its notable therapeutic properties. Unfortunately, its population has drastically declined due to the destruction of forest habitats. Thus, effectively protecting F. ussuriensis from extinction poses a significant challenge. A profound understanding of its genetic foundation is crucial. To date, research on the complete mitochondrial genome of F. ussuriensis has not yet been reported. Results The complete mitochondrial genome of F. ussuriensis was sequenced and assembled by integrating PacBio and Illumina sequencing technologies, revealing 13 circular chromosomes totaling 737,569 bp with an average GC content of 45.41%. A total of 55 genes were annotated in this mitogenome, including 2 rRNA genes, 12 tRNA genes, and 41 PCGs. The mitochondrial genome of F. ussuriensis contained 192 SSRs and 4,027 dispersed repeats. In the PCGs of F. ussuriensis mitogenome, 90.00% of the RSCU values exceeding 1 exhibited a preference for A-ended or U-ended codons. In addition, 505 RNA editing sites were predicted across these PCGs. Selective pressure analysis suggested negative selection on most PCGs to preserve mitochondrial functionality, as the notable exception of the gene nad3 showed positive selection. Comparison between the mitochondrial and chloroplast genomes of F. ussuriensis revealed 20 homologous fragments totaling 8,954 bp. Nucleotide diversity analysis revealed the variation among genes, and gene atp9 was the most notable. Despite the conservation of GC content, mitogenome sizes varied significantly among six closely related species, and colinear analysis confirmed the lack of conservation in their genomic structures. Phylogenetic analysis indicated a close relationship between F. ussuriensis and Lilium tsingtauense. Conclusions In this study, we sequenced and annotated the mitogenome of F. ussuriensis and compared it with the mitogenomes of other closely related species. In addition to genomic features and evolutionary position, this study also provides valuable genomic resources to further understand and utilize this medicinal plant.

Abstract Background Variations exist in the response of patients with Crohn’s disease (CD) to ustekinumab (UST) treatment, but the underlying cause remains unknown. Our objective was to investigate the involvement of immune cells and identify potential biomarkers that could predict the response to interleukin (IL) 12/23 inhibitors in patients with CD. Methods The GSE207022 dataset, which consisted of 54 non-responders and 9 responders to UST in a CD cohort, was analyzed. Differentially expressed genes (DEGs) were identified and subjected to Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses. Least absolute shrinkage and selection operator (LASSO) regression was used to screen the most powerful hub genes. Receiver operating characteristic (ROC) curve analysis was performed to evaluate the predictive performances of these genes. Single-sample Gene Set Enrichment Analysis (ssGSEA) was used to estimate the proportions of immune cell types. These significantly altered genes were subjected to cluster analysis into immune cell-related infiltration. To validate the reliability of the candidates, patients prescribed UST as a first-line biologic in a prospective cohort were included as an independent validation dataset. Results A total of 99 DEGs were identified in the integrated dataset. GO and KEGG analyses revealed significant enrichment of immune response pathways in patients with CD. Thirteen genes (SOCS3, CD55, KDM5D, IGFBP5, LCN2, SLC15A1, XPNPEP2, HLA-DQA2, HMGCS2, DDX3Y, ITGB2, CDKN2B and HLA-DQA1), which were primarily associated with the response versus nonresponse patients, were identified and included in the LASSO analysis. These genes accurately predicted treatment response, with an area under the curve (AUC) of 0.938. T helper cell type 1 (Th1) cell polarization was comparatively strong in nonresponse individuals. Positive connections were observed between Th1 cells and the LCN2 and KDM5D genes. Furthermore, we employed an independent validation dataset and early experimental verification to validate the LCN2 and KDM5D genes as effective predictive markers. Conclusions Th1 cell polarization is an important cause of nonresponse to UST therapy in patients with CD. LCN2 and KDM5D can be used as predictive markers to effectively identify nonresponse patients. Rosiglitazone (RSG), as an insulin-sensitizing drug to treat type 2 diabetes mellitus (T2DM) is reported to decrease bone quality and increase bone fracture risk. The multiple off-target effects of Resveratrol (RSV), a natural specific agonist of Sirtuin1 (Sirt1) with pro-osteoblastogenesis and anti-adipogenesis effects, on bone loss in T2DM are still under discussion. In this study, successfully ovariectomized rats were fed with high-fat diet and STZ (HFD/STZ) to induced T2DM mice. RSV alone, RSG alone or co-administration of RSV and RSG were given orally to T2DM rats for 8 weeks to determine whether RSV administration had any prevention effect on T2DM osteoporosis. Bone mesenchymal stem cells (BMSCs) and bone marrow‑derived macrophages (BMMs) were cultured under high glucose condition and were induced to osteoblasts or adipocytes and osteoclasts, respectively. μCT and HE staining showed that in T2DM osteoporotic rats, RSV co-administration prevents RSG induced-bone loss. ELISA results confirmed that RSV suppressed osteoclast activity and promoted osteoblast activity in diabetic osteoporosis rats and RSG-administrated diabetic osteoporosis rats. In vitro study showed that RSV significantly reversed RSG induced inhibition on osteogenesis and promotion on adiopogenesis of BMSC under high glucose (HG). Moreover, RSV significantly reverse RSG induced osteoclast formation and mature under HG. Taken together, these findings uncover a previously unappreciated anti-osteoporosis effect of concomitant treatment with RSV in RSG-administrated diabetic rats, suggesting the clinical use of RSV as an adjuvant in the treatment of T2DM for preventing or reversing RSG administration-associated bone loss.

Abstract Sea cucumber is a morphologically diverse and ecologically important clade of echinoderms. The sea cucumber Apostichopus japonicus is the most economically valuable species of sea cucumber. The initial assembly of the A. japonicus genome was released in 2017. However, this genome assembly is fragmented and lacks relative position information of genes on chromosomes. In this study, we produced a high-quality chromosome-level genome of A. japonicus using Pacbio HiFi long-reads and Hi-C sequencing data. The assembled A. japonicus genome spanned 671.60 Mb with a contig N50 size of 17.20 Mb and scaffold N50 size of 29.65 Mb. A total of 99.9% of the assembly was anchored to 23 chromosomes. In total, 19,828 genes were annotated, and 97.2% of BUSCO genes were fully represented. This high-quality genome of A. japonicus will not only aid in the development of sustainable aquaculture practices, but also lay a foundation for a deeper understanding of their genetic makeup, evolutionary history, and ecological adaptation.

With the rapid development of the transportation industry, permeable asphalt pavement has become widely used in the construction of expressways and urban roads due to its safety, comfort, and environmental protection characteristics. However, water-temperature coupling can damage the mechanical properties of permeable asphalt mixtures. The use of high-viscosity modified asphalt can improve the water damage resistance of the mixture to a certain extent, resulting in better mechanical properties. This paper focuses on the preparation of PAC-13 mixture specimens using high-viscosity modified asphalt and the study of its noise reduction and anti-skidding properties under water-temperature coupling. SMA-13 and AC-13 are used for comparative research. The influence of water-temperature coupling on the functional characteristics of permeable asphalt mixture is analyzed through standing wave tube test and pendulum test. The results show that PAC-13 exhibits better noise reduction and anti-skidding characteristics than SMA-13 and AC-13. As the temperature increases, the skid resistance of asphalt mixtures weakens, but the noise reduction properties are less affected. Furthermore, the noise reduction and anti-skidding properties of the three mixtures are significantly damaged by the action of water, with PAC-13 being the most affected. Nevertheless, PAC-13 still exhibits the best noise reduction and anti-skidding properties.

Summary: Background: Ustekinumab and vedolizumab are both effective for treating Crohn’s disease (CD). However, no head-to-head trials have been conducted thus far. We aimed to compare the effectiveness of ustekinumab and vedolizumab in CD patients either naïve or exposed to tumor necrosis factor-alpha inhibitors (TNFi). Methods: Patients treated with vedolizumab or ustekinumab for luminal CD were included from six centers in China from May 2020 to July 2023. Steroid-free remission, clinical remission, objective response, and remission at Weeks 26 and 52 were evaluated in a retrospective multicenter propensity score–weighted cohort. Findings: A total of 536 patients were included (386 ustekinumab, and 150 vedolizumab). After adjustment, ustekinumab showed higher rates of clinical remission (56.4% vs. 47.8%, P = 0.005), steroid-free remission (55.4% vs. 46.1%, P = 0.003), and objective response (67.8% vs. 42.7%, P

Objective: A liquid chromatography-tandem mass spectrometry coupled with immunoaffinity column clean-up method was established for the determination of 8 mycotoxins in aquatic products. Methods: The samples were extracted with acetonitrile water (84:16, V:V), and purified by multifunctional immunoaffinity columns. The chromatographic column was Agilent Proshell 120 SB·C18 column (2.1 mm×100 mm, 2.7 μm). Mobile phase was methanol and 5 mmol/L ammonium acetate (containing 0.1% formic acid), gradient elution. The flow rate was 0.4 mL/min, and the column temperature was 40 ℃. The electrospray was operated in both positive and negative mode at the same time, and the samples were detected by multiple reaction monitoring mode (MRM). Results: The linear relations of the eight mycotoxins were good within the concentration range of 1.0~50.0 ng/mL (R2>0.992). The limits of detection were between 0.05~0.50 μg/kg, and the limits of quantification were between 0.17~1.65 μg/kg. The average recoveries were 75.6%~106.3%, and the relative standard deviations (RSDs) were 3.5%~9.3%. Conclusion: The method is simple, sensitive and accurate for the rapid analysis of mycotoxin residues in aquatic products.

Introduction: Irregular bone defects of the humerus are common in clinical practice, but there are fewer reported cases of irregular humeral defects accompanied by shoulder joint “locking” dislocation and reverse Hill-Sachs injury caused by an electric shock. The choice of treatment for such cases is closely related to the extent of shoulder joint function recovery. This is a case report of a 60-year-old male patient who suffered from a shoulder joint “locking” dislocation with accompanying reverse Hill-Sachs injury due to muscle contraction after being electrically shocked at work. The patient was treated with a 3D-printed custom humeral head prosthesis for the treatment of the shoulder joint “locking” dislocation and reverse Hill-Sachs injury.Case presentation: A 60-year-old male patient, working as a construction worker, presented to our emergency department with right shoulder pain and restricted movement for more than 30 min after an electric shock. Right humeral CT revealed a comminuted fracture of the right humeral head. D-dimer levels were significantly elevated at 3239.00 ng/mL, and oxygen partial pressure was slightly decreased at 68 mmHg. Treatment included emergency wound debridement and dressing for the electrical injury, cardioprotective measures, anticoagulation, and symptomatic management. After stabilizing the patient’s condition, the patient underwent 3D-printed custom prosthesis-assisted partial replacement of the right humeral head and rotator cuff repair in the orthopedic department. Postoperatively, the patient’s right shoulder joint wound healed well, and mobility was restored.Conclusion: This case report demonstrates that the use of a 3D-printed custom prosthesis for the treatment of irregular humeral bone defects caused by specific injury mechanisms, especially cases involving shoulder joint “locking” dislocation and reverse Hill-Sachs injury, can achieve precise bone defect repair, minimize surgical trauma, and provide superior outcomes in terms of postoperative functional rehabilitation.

Persicaria perfoliata (L.) H. Gross is an herbal medicine with a long history of common use in China. In this study, we sequenced and assembled the complete chloroplast genome sequence of P. perfoliata and investigated its phylogenetic relationship in the family Polygonaceae. The total genome size is 160,585 bp in length with 37.96% GC content, consisting of a small single-copy (SSC) of 12,876 bp, a large single-copy (LSC) of 85,439 bp, and two inverted repeats (IRs) of 31,135 bp. The cp genome contains 128 genes, including 35 tRNA genes, eight rRNA genes, and 85 protein-coding genes. The phylogenetic tree showed that P. perfoliata was closely related to P. maackiana, and Persicaria exhibited a closer relationship with Bistorta in the family Polygonaceae. This work provides a molecular basis for investigating the evolutionary status, phylogenetic relationships, and population genetics of this species.

Allograft bone fractures are critical complications in massive allograft bone transplantations. There are limited studies available on the application of 3D printing for massive allograft bone transplantation complications, and no related reports on the treatment of an allograft bone fracture with a complete biological intramedullary nail. A complex case of allograft bone fracture after massive bone transplantation for a right tibial osteosarcoma was treated with fixation of an individualized 3D printed biological tibial intramedullary nail. Prior to the operation, the intramedullary nail was designed and printed based on the results of computed tomography examination of the affected limb, and the surface of the intramedullary nail was treated with a hydroxyapatite coating. Intraoperatively, the intramedullary nail was implanted according to the preoperative 3D design plan. The intraoperative and postoperative examinations showed that the 3D printed intramedullary nail achieved good matching between the implant and the medullary cavity, and the biological coating integrated well with surrounding bone. The follow‐up results 44 months postoperatively showed that the patient was satisfied with the surgical results, where his ankle function met his daily needs, and the Musculoskeletal Tumor Society score was 24. 3D printing tibial intramedullary nail fixation can be successful in the treatment of allograft bone fractures and should be considered as a treatment of choice. In this case, the intramedullary nail matched the surrounding bone well, had good osseointegration, and the patient regained basic function.

Embryonic development, especially metamorphosis and settlement, has a major impact on the life history of marine invertebrates. Apostichopus japonicus is an economically important species of sea cucumber. In this study, we performed RNA sequencing on six key stages of A. japonicas development: fertilized eggs, blastula, gastrula, auricularia, doliolaria, and pentactula. A total of 32,353 genes were identified and annotated as a reference gene set for subsequent pairwise comparison analysis. After filtering out low-quality genes, the dynamic molecular responses to development were revealed by WGCNA. The results showed that of the 20 modules, genes in the blue, yellow, and darkslateblue modules were highly correlated with the gastrula, auricularia, and blastula stages, respectively. GO terms for “RNA” and “proteasome complex” were most significantly enriched in the blue module. In the darkslateblue and yellow module, receptors of signaling pathways and metabolic processes were significantly enriched, respectively. All DEGs were categorized into 34 terms, mainly associated with signal transduction and cellular immunity. The expression pattern of genes associated with adhesion, cell cycle, signal, transcription factor, extracellular matrix (ECM), and cytoskeleton was analyzed according to gene function. The results of this study facilitated a more comprehensive understanding of the molecular characteristics of sea cucumber embryonic development and will provide theoretical guidance for larva rearing in sea cucumber culture.

Aestivation is considered to be one of the “purest” hypometabolic states in nature, as it involves aerobic dormancy that can be induced and sustained without complex factors. Animals that undergo aestivation to protect themselves from environmental stressors such as high temperatures, droughts, and food shortages. However, this shift in body metabolism presents new challenges for survival, including oxidative stress upon awakening from aestivation, accumulation of toxic metabolites, changes in energy sources, adjustments to immune status, muscle atrophy due to prolonged immobility, and degeneration of internal organs due to prolonged food deprivation. In this review, we summarize the physiological and metabolic strategies, key regulatory factors, and networks utilized by aestivating animals to address the aforementioned components of aestivation. Furthermore, we present a comprehensive overview of the advancements made in aestivation research across major species, including amphibians, fish, reptiles, annelids, mollusks, and echinoderms, categorized according to their respective evolutionary positions. This approach offers a distinct perspective for comparative analysis, facilitating an understanding of the shared traits and unique features of aestivation across different groups of organisms.

Aflatoxin B1 (AFB1), with the strong toxicity and carcinogenicity, has been reported to great toxicity to the liver and other organs of animals. It cause huge economic losses to breeding industry, including the aquaculture industry. Chinese mitten crabs (Eriocheir sinensis), as one of important species of freshwater aquaculture in China, are deeply disturbed by it. However, the molecular and metabolic mechanisms of hepatopancreas and ovary in crabs underlying coping ability are still unclear. Hence, we conducted targeted injection experiment with or without AFB1, and comprehensively analyzed transcriptome and metabolomics of hepatopancreas and ovary. As a result, 210 and 250 DEGs were identified in the L-C vs. L-30 m and L-C vs. L-60 m comparison, among which 14 common DEGs were related to six major functional categories, including antibacterial and detoxification, ATP energy reaction, redox reaction, nerve reaction, liver injury repair and immune reaction. A total of 228 and 401 DAMs in the ML-C vs. ML-30 m and ML-C vs. ML-60 m comparison both enriched 12 pathways, with clear functions of cutin, suberine and wax biosynthesis, tyrosine metabolism, purine metabolism, nucleotide metabolism, glycine, serine and threonine metabolism, ABC transporters and tryptophan metabolism. Integrated analysis of metabolomics and transcriptome in hepatopancreas discovered three Co-enriched pathways, including steroid biosynthesis, glycine, serine and threonine metabolism, and sphingolipid metabolism. In summary, the expression levels and functions of related genes and metabolites reveal the regulatory mechanism of Chinese mitten crab (Eriocheir sinensis) adaptability to the Aflatoxin B1, and the findings contribute to a new perspective for understanding Aflatoxin B1 and provide some ideas for dealing with it.

Exosomes are small extracellular vesicles that contain nucleic acids such as microRNAs and may participate in important biological processes. We made the initial report of exosomes from sea cucumber Apostichopus japonicus, that were classically cup-shaped and had an average size of 74.65 nm, and identified specific exosome biomarkers (HSP70, TSG101, and CD9). We explored changes in the global expression of microRNAs in exosomes from the commercially important A. japonicus under normal conditions and heat-stressed conditions for 3 and 7 d. We found that heat stress increased exosome production and modified the expression profiles of the microRNAs that they contained. Novel_mir31, novel_mir132, novel_mir26, miR-92_1, and novel_mir27 were commonly found to be differentially expressed in three comparison groups, indicating their importance in the heat stress response. The microRNA expression levels were validated by qPCR. Function analysis of the target genes of these microRNAs indicated they were involved mainly in replication and repair in the initial response of A. japonicus to heat stress exposure. Conversely, during acclimation to the high temperature conditions, the target genes of the differentially expressed microRNAs were primarily involved in metabolism adjustments. Our results will contribute to a better understanding of the regulatory roles of exosomes in sea cucumber, and provide insights into the functions of sea cucumber exosome-shuttled microRNAs against environmental stresses exacerbated by global warming.

IntroductionMalignant and giant pelvic tumors are complex and rare, and hemipelvectomies are complex procedures performed for this malignant lesion. Only a few studies had been conducted on the survival and recurrence of pelvic sarcomas patients undergoing internal or external hemipelvectomy. In the present study, we compared internal with external hemipelvectomy in pelvic sarcomas on clinical outcomes by a meta-analysis.MethodsThe survival and recurrence rates of pelvic sarcomas patients were collected from research reports from CNKI, MEDLINE, EMBASE, the Cochrane Database, and Google Scholar until April 2022. The quality of included articles was evaluated by two independent reviewers. Differences between patients undergoing internal and external hemipelvectomy were analyzed based on postoperative survival and recurrence rates.ResultsFive articles were included according to selection criteria. There were 183 patients in total from these studies. Our results showed that there was no significant difference between limb salvage surgery and amputation according to survival; however, patients with internal hemipelvectomy had a lower recurrence rate.ConclusionsInternal hemipelvectomy results in a lower recurrence rate and similar survival rate, while not increasing the risk of metastasis and complications. This study provided more pieces of evidence to support internal hemipelvectomy as a favorable treatment of pelvic sarcomas.

The sea cucumber Apostichopus japonicus has high nutritional, medicinal, and economic value. However, factors such as overexploitation, climate change, and environmental pollution have resulted in serious germplasm degradation in both farmed and wild A. japonicus, and it has been listed as endangered on the IUCN (International Union for Conservation of Nature) Red List of Threatened Species. Cryopreservation is an important method to protect germplasm resources and solve the problem of germplasm degradation. Using a programmable freezer and computer-aided sperm analysis, we comprehensively studied and screened the factors that affect the post-thaw motility of A. japonicus sperm during cryopreservation. Based on our results, we propose the following optimal cryopreservation procedure for A. japonicus sperm: cryo-diluent composition of 12.5% dimethyl sulfoxide and 0.1 mol/L glucose, with filter-sterilized (the filter mesh size: 0.45μm) natural seawater (NSW) as the extender; 1:5 mixing ratio of sperm and cryo-diluent; cooling rate and thawing temperatures of 10°C/min and 20°C, respectively. The post-thaw motility of sperm treated using the optimal procedure was > 65%, the fertilization rate (in the blastocyst stage) was nearly 80%, and the hatching rate (in the early auricularia larva stage) was > 65%. Additionally, frozen sperm that had been cryopreserved for 1 year retained a considerable post-thaw motility and fertilization rate compared to recently cryopreserved sperm. We detected obvious differences in sperm freezability among individual A. japonicus, and cryopreservation caused some damage to the sperm structure. In conclusion, our optimized procedure make large-scale cryopreservation of A. japonicus sperm possible, and our results provide valuable information that could be applied to research and conservation of A. japonicus.

The sea cucumber Apostichopus japonicus has important nutritional and medicinal value. Unfortunately, we know little of the source of active chemicals in this animal, but the plentiful pigments of these animals are thought to function in intriguing ways for translation into clinical and food chemistry usage. Here, we found key cell groups with the gene activity predicted for the color morphology of sea cucumber body using single-cell RNA-seq. We refer to these cell populations as melanocytes and quinocytes, which are responsible for the synthesis of melanin and quinone pigments, respectively. We integrated analysis of pigment biochemistry with the transcript profiles to illuminate the molecular mechanisms regulating distinct pigment formation in echinoderms. In concert with the correlated pigment analysis from each color morph, this study expands our understanding of medically important pigment production, as well as the genetic mechanisms for color morphs, and provides deep datasets for exploring advancements in the fields of bioactives and nutraceuticals.

In this study, a ZSM−5/CLCA molecular sieve was prepared by the hydrothermal method using coal gangue as the raw material and cellulose aerogel (CLCA) as the green templating agent, which not only reduces the cost of traditional molecular preparation but also improves the comprehensive resource utilization rate of coal gangue. Through a series of characterization methods (XRD, SEM, FT-IR, TEM, TG, and BET), the crystal form, morphology, and specific surface area of the prepared sample were tested and analyzed. The performance of the adsorption process of malachite green (MG) solution was analyzed by adsorption kinetics and adsorption isotherm. The results show that the synthesized zeolite molecular sieve and the commercial zeolite molecular sieve are highly consistent. At a crystallization time of 16 h, a crystallization temperature of 180 °C, and an additive amount of cellulose aerogel of 0.6 g, the adsorption capacity of ZSM−5/CLCA for MG was up to 136.5 mg/g, much higher than that of commercially available ZSM−5. This provides an idea for the green preparation of gangue-based zeolite molecular sieves to remove organic pollutants from water. Moreover, the process of adsorbing MG on the multistage porous molecular sieve, which is spontaneous, conforms to the pseudo-second-order kinetic equation and Langmuir isothermal adsorption model.

Microorganisms colonize plastics in the aquatic environment but their composition on plastics used in aquaculture remains poorly studied. Microorganisms play a significant role in aquaculture in terms of water quality and the health of cultivated species. In the current study, we explored the composition of microorganisms on floating plastics and their surrounding water collected from ponds and open aquaculture areas. Using scanning electron microscopy, the diversity of microbial communities, primarily diatoms, and bacteria were identified on the plastic surfaces. Additionally, epifluorescence microscopy revealed that prokaryotes were colonized on all plastic samples from 0.1 to 29.27×103 cells/cm2, with a high abundance found in open aquaculture areas compared to ponds. Bacterial communities were characterized by 16S rRNA sequencing which showed that bacterial communities on plastics were dominated by Proteobacteria, Cyanobacteria, Bacteroidetes, and Actinobacteria. The level of these microbial communities on the plastics differed from those found in the surrounding seawater samples and the abundance of potentially pathogenic bacteria was higher in plastics than in seawater samples. Moreover, hydrocarbon-degrading bacteria were more abundant in the investigated plastic samples than in the water samples. This study contributes to the knowledge regarding the plastisphere community in aquaculture.

The fabrication of composite materials is an effective way to improve the performance of a single material and expand its application range. In recent years, graphene-based materials/polymer composite aerogels have become a hot research field for preparing high-performance composites due to their special synergistic effects in mechanical and functional properties. In this paper, the preparation methods, structures, interactions, properties, and applications of graphene-based materials/polymer composite aerogels are discussed, and their development trend is projected. This paper aims to arouse extensive research interests in multidisciplinary fields and provide guidance for the rational design of advanced aerogel materials, which could then encourage efforts to use these new kinds of advanced materials in basic research and commercial applications.

Electrochemical conversion of CO2 and NO3− waste (EC-CO2/NO3−) into valuable urea is a promising method for fertilizer production and environmental remediation, but its practical application is currently limited by the low efficiency of electrocatalytic processes. Here, we report a novel In2O3 nanotube (In2O3-NT) material derived from a metal–organic framework (MOF) which functions as an electrocatalyst for durable and efficient urea synthesis via EC-CO2/NO3−. The obtained In2O3-NT-500 with a porous structure and rich oxygen vacancies (Vo) is more conducive to the target reaction system, reaching a urea formation rate of 1441 μg mgcat−1 h−1 with a high faradaic efficiency of 60.3%, exhibiting the top-level performance toward urea synthesis via the current route. In situ attenuated total reflection Fourier transform infrared spectroscopy verified that In2O3-NT with enriched Vo could stabilize the *CO2NH2 intermediate, thus accelerating the rate-determining step (RDS). The DFT simulation demonstrated that the transformation of *COOHNH2 to *CONH2 is the RDS for urea formation. The defect-engineered In2O3-NT catalyst significantly lowers the energy barrier for this step, thus boosting the overall efficiency of urea synthesis. This work provides an example showing that the defect engineering of In2O3-NT is highly capable of activating CO2 and NO3− waste molecules for urea synthesis, and is conceptually versatile for other value-added chemical production methods. Cis-regulatory sequences, such as enhancers, control development and physiology by regulating gene expression. The identification and functional characterization of these distal regulatory elements remains challenging, especially in non-model aquatic organisms like sea cucumbers. Apostichopus japonicus, an important echinoderm species, exhibits diverse body colour. Pigmentation is an important stage of growth and development in this species, but the regulatory mechanism remains poorly understood, especially the extent to which epigenetic changes regulate pigmentation. Herein, we globally profiled chromatin accessibility using the assay for transposase-accessible chromatin sequencing technique during pigmentation in green, white and purple sea cucumbers. We have got 27, 817 peaks in total and discovered 1427 peaks showing differentially accessed DNA sites across the genome, which were categorized into four distinct clusters via unsupervised hierarchical clustering. Among three colour morphs, white sea cucumber exhibited the most extensive changes and the greatest differences from the other two colour morphs. Global increases in chromatin accessibility occur in the late pigmentation stage suggesting that active function in the late pigmentation stage drives body colour formation. The screened key cis-regulatory elements KIT, RPS6KB2 and IRF8 may contributed to the albinism, slow growth and poor disease resistance of white sea cucumber. In conclusion, our study provides a framework for understanding the accessible chromatin dynamics of body colour diversity during echinoderm pigmentation process and a new avenue to unravel the transcriptional regulatory mechanisms that facilitate the occurrence of relevant molecular events.

Gonads play a critical role in the reproduction of sea cucumbers, and their development is regulated by a large number of genes. To date, the ideal reference genes (RGs) for studying sea cucumber gonadal development have not been reported. In this study, we selected ten RGs and conducted quantitative real-time PCR analysis to identify the most stable among Apostichopus japonicus gonads at four different stages of gonadal development. We used ∆Cq, GeNorm, NormFinder, BestKeeper, and RefFinder to evaluate the stability of candidate genes. EF1β and Grb2 together were the preferred combination of stable RGs among all of the gonad samples and for the ovaries alone. UBQ combined with RPS4 was the best combination of stable RGs in the testes. This study aimed to provide a basis for analysis of differences in gene expression related to gonadal development in sea cucumbers, and the results provide valuable information for further studies of the molecular mechanisms involved in sea cucumber gonadal development.

There is a need for synthetic grafts to reconstruct large bone defects using minimal invasive surgery. Our previous study showed that incorporation of Sr into bioactive borate glass cement enhanced the osteogenic capacity in vivo. However, the amount of Sr in the cement to provide an optimal combination of physicochemical properties and capacity to stimulate bone regeneration and the underlying molecular mechanism of this stimulation is yet to be determined. In this study, bone cements composed of bioactive borosilicate glass particles substituted with varying amounts of Sr (0 mol% to 12 mol% SrO) were created and evaluated in vitro and in vivo. The setting time of the cement increased with Sr substitution of the glass. Upon immersion in PBS, the cement degraded and converted more slowly to HA (hydroxyapatite) with increasing Sr substitution. The released Sr2+ modulated the proliferation, differentiation, and mineralization of hBMSCs (human bone marrow mesenchymal stem cells) in vitro. Osteogenic characteristics were optimally enhanced with cement (designated BG6Sr) composed of particles substituted with 6mol% SrO. When implanted in rabbit femoral condyle defects, BG6Sr cement supported better peri-implant bone formation and bone-implant contact, comparing to cements substituted with 0mol% or 9mol% SrO. The underlying mechanism is involved in the activation of Wnt/β-catenin signaling pathway in osteogenic differentiation of hBMSCs. These results indicate that BG6Sr cement has a promising combination of physicochemical properties and biological performance for minimally invasive healing of bone defects.

Background/Aims Crohn’s disease (CD) primarily affects young female adults of reproductive age. Few studies have been conducted on this population’s ovarian reserve status. The aim of study was to investigate potential risk factors associated with low ovarian reserve, as reflected by serum anti-Müllerian hormone (AMH) in women of reproductive age with CD. Methods This was a case-control study. Cases included 87 patients with established CD, and healthy controls were matched by age, height and weight in a 1:1 ratio. Compressed hyperspectral imaging is a powerful technique for satellite-borne and airborne sensors that can effectively shift the complex computational burden from the resource-constrained encoding side to a presumably more capable base-station decoder. Reconstruction algorithms play a pivotal role in compressive imaging systems. Traditional model-based reconstruction approaches are computationally burdensome and achieve limited success. Deep learning-based approaches, while improving in reconstruction accuracy and speed, depend heavily on data, which is a major challenge for satellite-borne hyperspectral compressed imaging. In this article, we combine the respective advantages of model-based and learning-based approaches in a distributed compressed hyperspectral sensing framework, employing linear mixed model assumptions and spectral library learning to simultaneously improve the reconstruction speed and accuracy without using a large amount of additional hyperspectral data. First, the relationship between the CS band and the key band is learned from the spectral library to ensure that the key band endmembers can be accurately predicted. Then, the joint horizontal and vertical difference operators are proposed to enhance the estimation of the initial values of abundance. Finally, the CS band endmembers and residuals are updated in the reconstruction module to deal with the endmember and abundance mismatch. Extensive experimental results on five real hyperspectral datasets demonstrate that the proposed spectral library learning, abundance initialization, and reconstruction strategy can effectively improve the compressed sensing reconstruction accuracy, outperforming the existing state-of-the-art methods.

Parental nutrient reserves are directly related to reproductive performance in sea cucumbers. This study focused on the lipid requirements of male and female sea cucumbers Apostichopus japonicus during the reproductive stage and analyzed their physiological responses to a high-fat diet (HFD). The intestinal lipid metabolites and microbiome profile changed significantly in animals fed with the HFD, as given by an upregulation of metabolites related to lipid metabolism and an increase in the predominance of Proteobacteria in the microbiome, respectively. The metabolic responses of male and female sea cucumbers to the HFD differed, which in turn could have triggered sex-related differences in the intestinal microbiome. These results suggest that the lipid content in diets can be differentially adjusted for male and female sea cucumbers to improve nutrition and promote reproduction. This data contributes to a better understanding of the reproductive biology and sex differences of sea cucumbers.

Some sea cucumbers are economically and ecologically important, but they are threatened by thermal and hypoxic stress in changing oceanographic conditions. We construct circRNAs profiles, reveal circRNAs characters, and illustrate the potential regulatory roles of circRNAs in one commercially important species of sea cucumber, Apostichopus japonicus. Reads are distributed in intergenic (44.14%), exonic (48.26%) and intronic (7.60%) regions of the genome. A total of 1684 circRNAs were identified, and the most common spliced length is 269 nt in the present study. In three treatments (HT [thermal stress], LO [hypoxic stress], and HL [combined thermal and hypoxic stress]), 24, 27 and 27 differentially expressed (DE) circRNAs were identified, respectively. Five novel DE-circRNAs commonly occur in these treatments (novel_circ_0003311, novel_circ_0000229, novel_circ_0003944, novel_circ_0001458 and novel_circ_0000707), and based on them, potential circRNA-miRNA binding pairs were predicted. Sanger sequencing, RNase R treatment experiment and qPCR validation identified the accuracy of the circRNAs. Key circRNAs identified in the present study were covalently closed and were more stable under RNase R treatment than linear RNAs. Based on function analysis, circRNAs could regulate metabolic process, signal transduction, and ion responses in A. japonicus when exposed to thermal and hypoxic stress, and ‘regulation of response to stimulus’ is a common gene ontology (GO) term that is significantly enriched in each treatment; GO terms for ‘DNA’ and ‘stress’ are commonly enriched in heat-related treatments (HT and HL); and GO terms for ‘protein’ are commonly enriched in hypoxia-related treatments (LO and HL). When environmentally stressed, ‘metabolism,’ ‘transport and catabolism,’ ‘membrane transport,’ and ‘signal transduction’ were significantly responded in sea cucumber based on KEGG analysis. We provide insights into circRNA functions in stress regulation and lay a foundation for invertebrate circRNA research.

Melanin, which is a pigment produced in melanocytes, is an important contributor to sea cucumber body color. MITF is one of the most critical genes in melanocyte development and melanin synthesis pathways. However, how MITF regulates body color and differentiation in sea cucumbers is poorly understood. In this study, we analyzed the expression level and location of MITF in white, purple, and green sea cucumbers and identified the genes regulated by MITF using chromatin immunoprecipitation followed by sequencing. The mRNA and protein expression levels of MITF were all highest in purple morphs and lowest in white morphs. In situ hybridization indicated that MITF mRNA were mainly expressed in the epidermis. We also identified 984, 732, and 1191 peaks of MITF binding in green, purple, and white sea cucumbers, which were associated with 727, 557, and 887 genes, respectively. Our findings suggested that MITF contributed to the body color differentiation of green, purple, and white sea cucumbers through expression differences and regulation of downstream genes. These results provided a basis for future studies to determine the mechanisms underlying body color formation and provided insights into gene regulation in sea cucumbers.

Behavioral plasticity in animals allows for moment-by-moment behavioral adjustments to biotic and abiotic uncertainties in the environment. For most aquatic animals, reproduction involves gonadal development and other physiological changes, causing increasing demands in nutrients and energy for females. The aim of this report was to determine how the female sea cucumber, Apostichopus japonicus, accommodates increasing energy demands during reproduction by adopting a behavioral energy conservation strategy. Dynamic changes in feeding activity, total body mass, locomotor activity, energetic condition, and metabolic performance of the females were measured from the non-breeding stage up to the mature stage. Routine metabolic rate analysis showed that reproduction caused a significant increase in energy demands in the adult. However, reproduction also suppressed the appetite of adults resulting in decreased energy intake. Interestingly, combining time-lapse camera and behavioral analysis software, the results showed that sea cucumbers down-regulated their locomotor activities in order to conserve energy effectively. Energy budget analysis and unchanged cortisol level revealed that the reduction in motility offset the increased energy demand for reproduction while helping to maintain energy homeostasis in the maternal body. Altogether, our study suggests that plasticity in the locomotor behavior allows sea cucumbers to cope with the high energy demands of reproduction.

Apostichopus japonicus achieves intestinal regeneration in a short period after evisceration, and multiple genes are involved in this process. The transcriptome of A. japonicus was screened for regeneration-associated protein (Aj-Orpin), a gene that is specifically upregulated during intestinal regeneration. The expression and function of Aj-Orpin were identified and investigated in this study. The 5′ and 3′ RACE polymerase chain reaction (PCR) was used to clone the full-length cDNA of Aj-Orpin. The open reading frame codes for a 164 amino-acid protein with an EF-hand_7 domain and overlapping signal peptides and transmembrane regions. Moreover, Aj-Orpin mRNA and protein expression during intestinal regeneration was investigated using real-time quantitative PCR and Western blot. The expression pattern of Aj-Orpin in the regenerating intestine was investigated using immunohistochemistry. The results showed that Aj-Orpin is an exocrine protein with two EF-hand-like calcium-binding domains. Expression levels were higher in the regenerating intestine than in the normal intestine, but protein expression changes lagged behind mRNA expression changes. Aj-Orpin was found to play a role in the formation of blastema and lumen. It was primarily expressed in the serosal layer and submucosa, suggesting that it might be involved in proliferation. These observations lay the foundation for understanding the role of Orpin-like in echinoderm intestinal regeneration.

Abstract In the process of picking, actinidia arguta has difficulty in image recognition and occlusion problems, and there are few studies on kiwifruit recognition. Based on this, this study uses the color model to perform image basic processing and uses frequency domain enhancement to process the image. Simultaneously, in the frequency domain of the image, this study applied filtering to the original image of kiwifruit orchard, used homomorphic filtering to enhance the image of actinidia arguta orchard, highlighted the characteristics of actinidia arguta trunk, and reduced the influence of background noise on the recognition of actinidia arguta trunk. In addition, this study used a binocular stereo vision system for fruit location recognition to improve recognition accuracy. Finally, the effectiveness of the research method is verified by experimental research. The results show that the proposed algorithm performs well and can provide theoretical references for subsequent related research.

Most species of the class Holothuroidea are able to regenerate most of their internal organs following a typical evisceration process, which is a unique mechanism that allows sea cucumbers to survive in adverse environments. In this review, we compare autotomy among different type of sea cucumber and summarize the histocytological events that occur during the five stages of intestinal regeneration. Multiple cytological activities, such as apoptosis and dedifferentiation, take place during wound healing and anlage formation. Many studies have focused on the molecular regulation mechanisms that underlie regeneration, and herein we describe the techniques that have been used as well as the development-related signaling pathways and key genes that are significantly expressed during intestinal regeneration. Future analyses of the underlying mechanisms responsible for intestinal regeneration should include mapping at the single-cell level. Studies of visceral regeneration in echinoderms provide a unique perspective for understanding whole-body regeneration or appendage regeneration.

Microplastics (MPs) in the form of microfibres (MFs) are of great concern because of their size and increasing abundance, which increase their potential to interact with or be ingested by aquatic organisms. Although MFs are the dominant shape of MPs ingested by sea cucumbers in habitats, their effect on sea cucumbers remains unclear. This study examined the effect of dietary exposure to MFs on the growth and physiological status of both juvenile and adult Apostichopus japonicus sea cucumbers. MFs were mixed into the diet of sea cucumbers for 60 d at environmentally relevant concentrations of 0.6 MFs g−1, 1.2 MFs g−1 and 10 MFs g−1. Dietary exposure to MFs, with concentrations at or above those commonly found in the habitats, did not significantly affect the growth and faecal production rate of either juvenile or adult sea cucumbers. However, a disruption in immunity indices (acid phosphatase and alkaline phosphatase activity) and oxidative stress indices (total antioxidant capacity and malondialdehyde content) was observed in juvenile and adult sea cucumbers, indicating that these indices might be useful as potential biomarkers of the exposure to MF ingestion in sea cucumbers. This study provides insights into the toxicity mechanism of MF ingestion in a commercially and ecologically important species.

A novel sulfonic-cellulose succinate half ester (S-CSHE) dispersant for coal–water slurry (CWS) was successfully synthesized using cotton linters, sulfamic acid and succinic anhydride in DMF by a one-pot synthesis. The effects of the synthetic condition of S-CSHE as a dispersant for CWS were studied. An S-CSHE with a maximum degree of substitution of 0.98 was obtained under these optimized conditions. The synthesized samples were characterized by GPC, FT-IR spectroscopy, 13C-NMR, and SEM. The molecular weight was from 2.2 × 103 to 1.2 × 104, revealed by GPC; FT-IR spectra analysis revealed characteristic absorptions of the sulfonic-cellulose succinate half ester; SEM images showed smooth cellulose structures, while the S-CSHE had a compact surface. Effects of S-CSHE on the fluidity of application as the dispersants for the CWS prepared from Chinese Zhundong coal were studied further. The CWS application performance investigations showed that S-CSHE can most effectively reduce CWS viscosity, and perform excellent dispersity and stability. When the dosage of S-CSHE was up to 0.5 wt.%, the maximum coal content of CWS may reach 70 wt.%, and the apparent viscosity of CWS was 487 mPa·s. The CWS prepared using S-CSHE (0.38 wt.%) for Zhundong coal showed the rheological characteristics of shear-thinning, and is consistent with the Herschel–Bulkley model. This work found a new route for utilizing cotton linters cellulose and enlarged the selecting range of the dispersant for CWS. It has a positive significance for efficient and clean utilization of Xinjiang Zhundong coal.

Neuropeptides are endogenous active substances that are present in nervous tissues and participate in behavioral and physiological processes of the animal system. Locomotor behavior is basic to predation, escape, reproduction in animals, and neuropeptides play an important role in locomotion. In this study, the function of pedal peptide-type neuropeptide (PDP) in the process of locomotor behavior of the sea cucumber Apostichopus japonicus was evaluated. The locomotor behavior of A. japonicus was recorded by infrared camera before and after PDP administration, and muscle physiology was studied by ultra performance liquid chromatography and quadrupole time-off-light mass spectrometry (UPLC-Q-TOF-MS) to clarify the potential physiological mechanisms. The results showed that PDP enhanced the cumulative duration of moving significantly at the 7th h after injection, and reduced the mean and maximum velocity by 16.90 and 14.22% in A. japonicus. The data of muscle metabolomics suggested that some significantly changed metabolites were related to locomotor behavior of sea cucumbers. The decreases of phosphatidylethanolamine (PE) and phosphatidylcholine (PC) might result in the increases of lysophosphatidylcholines (lysoPC) and lysophosphatidylethanolamine (lysoPE), and suggested the change of fluidity and permeability in the muscle cell membrane, which would affect the physiology and function of muscle cells, and finally alter the locomotor behavior. In addition, the increased level of arachidonic acid (ARA) might activate K+ ion channels and then affect the signaling of muscle cells, or promote the sensitivity of muscle cells to Ca2+ and then result in the contractility of longitudinal muscles in sea cucumbers. ARA was also involved in the linoleic acid metabolism which was the only pathway that disturbed significantly after PDP administration. In conclusion, PDP participated in the regulation of locomotor behavior in the sea cucumber, and the decreased PE and PC, increased lysoPC, lysoPE and ARA might be the potential physiological mechanisms that responsible for behavioral effects of PDP in A. japonicus.

Physiological plasticity allows animals to adjust their physiological function to abiotic and biotic variations. It has been mostly studied in the context of response to external factors and not much is known on how animals adjust their physiology to cope with variations in internal conditions. The process of reproduction implies gonadal maturation and other internal changes, bringing various challenges to the animal such as an increased demand for energy and oxygen. Here, the capacity of the sea cucumber, Apostichopus japonicus to adjust its respiratory function and physiological mechanisms during reproduction was studied using a time-lapse videography and metabolomics approach. The results showed that reproduction caused a significant increase in oxygen consumption in A. japonicus. Interestingly, breeding sea cucumbers can accommodate the high oxygen demand by accelerating respiratory rate. However, to maintain a necessary high level of respiratory activity during reproduction, sea cucumbers need consume large amounts of adenosine triphosphate (ATP). In addition, the metabolomic data suggests that oxidative stress and hormone regulation are the physiological mechanisms linking reproduction and respiratory function. Altogether, these findings suggest that plasticity of respiratory function is an effective tactic to cope with high oxygen demand during reproduction.

Genetic linkage maps have become an indispensable tool for genetics and genomics research. Sea cucumber (Apostichopus japonicus), which is an economically important mariculture species in Asia, is an edible echinoderm with medicinal properties. In this study, the first SNP-based high-density genetic linkage map was constructed by sequencing 132 A. japonicus individuals (2 parents and 130 offspring) according to a genotyping-by-sequencing (GBS) method. The consensus map was 3181.54 cM long, with an average genetic distance of 0.52 cM. A total of 6144 SNPs were assigned to 22 linkage groups (LGs). A Pearson analysis and QTL mapping revealed the correlations among body weight, body length, and papillae number. An important growth-related candidate gene, protein still life, isoforms C/SIF type 2 (sif), was identified in LG18. The gene was significantly highly expressed during the larval developmental stages. Its encoded protein reportedly functions as a guanine nucleotide exchange factor. These results would facilitate the genetic analysis of growth traits and provide valuable genomic resources for the selection and breeding of new varieties of sea cucumbers with excellent production traits.

The sea cucumber, an important economic species, has encountered high mortality since 2013 in northern China because of seasonal environmental stress such as hypoxia, high temperature, and low salinity. MicroRNAs (miRNAs) are important in regulating gene expression in marine organisms in response to environmental change. In this study, high-throughput sequencing was used to investigate alterations in miRNA expression in the sea cucumber under different levels of dissolved oxygen (DO). Nine small RNA libraries were constructed from the sea cucumber respiratory trees. A total of 26 differentially expressed miRNAs, including 12 upregulated and 14 downregulated miRNAs, were observed in severe hypoxia (DO 2 mg/L) compared with mild hypoxia (DO 4 mg/L) and normoxic conditions (DO 8 mg/L). Twelve differentially expressed miRNAs were clustered in severe hypoxia. In addition, real-time PCR revealed that 14 randomly selected differentially expressed miRNAs showed significantly increased expressions in severe hypoxia and the expressions of nine miRNAs, including key miRNAs such as Aja-miR-1, Aja-miR-2008, and Aja-miR-184, were consistent with the sequencing results. Moreover, gene ontology and pathway analyses of putative target genes suggest that these miRNAs are important in redox, transport, transcription, and hydrolysis under hypoxia stress. Notably, novel-miR-1, novel-miR-2, and novel-miR-3 were specifically clustered and upregulated in severe hypoxia, which may provide new insights into novel “hypoxamiR” identification. These results will provide a basis for future studies of miRNA regulation and molecular adaptive mechanisms in sea cucumbers under hypoxia stress.

Host-intestine microbiota interactions have been widely studied in aquatic animals, but these interactions in the intestine regeneration process of the sea cucumber Apostichopus japonicus have been rarely investigated. To understand how intestine regeneration impacts the developing intestinal microbiome composition and function, we performed a case study to characterize the intestinal microbial composition and functional genes of A. japonicus during intestine regeneration stages. High-throughput 16S rRNA gene sequencing revealed significantly different intestine microbiota compositions in different regeneration stages. The phylogenetic diversity and composition of the intestinal microbiota changed significantly in the early regeneration stage and tended to recover in the end stage. During the regeneration process, the abundance of Bacteroidetes and Rhodobacterales increased significantly. A network analysis revealed that Rhodobacteraceae and Flavobacteriaceae may function as keystone taxa in the intestinal microbial community of A. japonicus during intestine regeneration. Metagenomic analyses of representative samples revealed that the microbiomes of regenerating intestines were enriched in genes facilitating cell proliferation, digestion and immunity. The increased abundance of Bacteroidetes elevated the enrichment of genes associated with carbohydrate utilization. Some functional features in the subsystem category changed in a pattern that was consistent with the changing pattern of microbiota composition during intestine regeneration. Our results revealed that seemingly regular alterations in the intestinal microbiome composition and function are associated with intestine regeneration stages. Intestinal microbiota can increase the abundance of beneficial bacterial members and upregulate related functional genes to adapt to intestine regeneration and reconstruct a stable community structure. This study provides a new insight into the mechanism of the host-microbiota interaction response to organ regeneration.
