In actual production, because increase of sows litter size, the management of piglets in split-suckling leads to intermittent neonatal maternal separation (NMS). Early lactation is a critical period of cognitive development in the piglets' brain. The experiment hypothesizes that intermittent NMS may affect piglets' neurological development and cognitive ability. To determine the extent of effects of NMS on the piglets' prefrontal cortex cognitive development, the control group (Con group, n=53) was set up with no NMS, the experimental group (MS group, n=53) was set up with MS for 4 h/day, and the piglets were milk supplemented during the separation period and weaned until 35d. Twelve piglets at postnatal day (PND) 35 were selected from each group for cognitive assessment. Three PND35 piglets from each group were slaughtered, and collected prefrontal cortex samples for reference transcriptome sequencing. Following bioinformatics analysis, GO enrichment and KEGG enrichment analysis as well as candidate gene screening were performed for differentially expressed genes. Finally, quantitative reverse transcription PCR (qRT-PCR) was used to verify the sequencing data were accurate. The results showed that, in the spatial holeboard discrimination task, the working memory and reference memory of piglets in the MS group were significantly lower than those in the Con group (P < 0.01). In the new object recognition task, the MS group was significantly lower than the Con group in the number of exposure to new objects and discrimination index (P < 0.01). In the prefrontal cortex of the MS group, 349 up-regulated, 151 down-regulated differential genes, and 584 significant GO entries were discovered. Screening for 9 genes associated with cognition at large differential multiples. qPCR validation results showed that MC4R, GRP, CYFIP2, NMB, CPEB1, ADCYAP1 were significantly downregulated and MAG, PLP1, CPEB4 were significantly upregulated. Light microscopy results showed changes in the morphological structure of neuronal cells and microglia. In summary, intermittent NMS may impair partial cognitive function in piglets by damaging the structure of the prefrontal cortex.

Ammonia is a significant pollutant in the livestock houses and the atmospheric environment, and excessive ammonia would harm the health of livestock and breeders. Previous studies have shown that ammonia exposure could damage the tissue structure of the nervous system, but the molecular mechanism of ammonia-induced hypothalamus damage was still unclear. The purpose of this study was to determine the role of excessive ammonia in abnormal autophagy of pig hypothalamus and whether selenomethionine would have a mitigating effect on ammonia toxicity. Twenty-four 18-week pigs were randomly divided into four groups: the control group (C group), the selenium group (Se group), the ammonia + selenium group (A + Se group), and the ammonia group (A group). In our study, the expression levels of NF-κB, IL-1β, iNOS, TNF-α, IKK-α, p-IKK-α, Nrf2, ATG5, ATG 10, ATG 12, LC3 I/II, HSP60, HSP70, and HSP90 were increased after ammonia exposure; meanwhile, IFN-γ, IKB-α, p-IKB-α, Keap1, P62, mTOR, AKT, p-AKT, PI3K, SQSTM, and Beclin1 showed decreasing trends. The results indicated that excessive ammonia inhalation inhibited the AKT/mTOR pathway to acclerated autophagy through oxidative stress-mediated inflammation in the porcine hypothalamus. L-selenomethionine could alleviate hypothalamus injury induced by ammonia exposure.

As a means of environmental enrichment, music environment has positive and beneficial effects on biological neural development. Kunming white mice (61 days old) were randomly divided into the control group (group C), the group of D-tone (group D), the group of A-tone (group A) and the group of G-tone (group G). They were given different tonal music stimulation (group A) for 14 consecutive days (2 h/day) to study the effects of tonal music on the neural development of the hippocampus and prefrontal cortex of mice in early life and its molecular mechanisms. The results showed that the number of neurons in the hippocampus and prefrontal cortex of mice increased, with the cell morphology relatively intact. In addition, the number of dendritic spines and the number of dendritic spines per unit length were significantly higher than those in group C, and the expressions of synaptic plasticity proteins (SYP and PSD95) were also significantly elevated over those in group C. Compared with group C, the expression levels of BDNF, TRKB, CREB, PI3K, AKT, GS3Kβ, PLCγ1, PKC, DAG, ERK and MAPK genes and proteins in the hippocampus and prefrontal cortex of mice in the music groups were up-regulated, suggesting that different tones of music could regulate neural development through BDNF and its downstream pathways. The enrichment environment of D-tone music is the most suitable tone for promoting the development of brain nerves in early-life mice. Our study provides a basis for screening the optimal tone of neuroplasticity in early-life mice and for the treatment of neurobiology and neurodegenerative diseases.

Background Cancer patients who receive anthracycline-based chemotherapy regimens often discontinue chemotherapy due to cardiotoxicity. Preventing and reducing anthracycline-induced cardiotoxicity (ACT) is a hot topic in cardio-oncology research. Network pharmacology is a new discipline that integrates pharmacology, bioinformatics, and systems biology. It can be used to analyze the mechanism of action of drugs in the body from a holistic perspective by constructing a "disease-gene-drug" network, providing a new method to explore compounding mechanisms of Chinese medicine. Based on network pharmacology, this study explored the mechanism of the reduction of cardiotoxicity of anthracyclines by Qishen Huanwu Capsule. Methods The active ingredients of Qishen Huanwu Capsule and their targets were screened based on the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform and Chemistry Database. The target genes of ACT were screened through the PharmGkb, GeneCards, Online Mendelian Inheritance in Man (OMIM), Genetic Association Database (GAD), and Therapeutic Target Database (TTD). The Venny2.1 online analysis tool was used to construct a Venn diagram to obtain the common targets of ACT and Qishen Huanwu Capsule. The STRING platform was used to construct the protein-protein interactions (PPI) among the common targets; ClueGO software was used to perform Gene Ontology (GO) biological process enrichment analysis for the common targets; the R language was used to perform Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis; and the results were visualized using Cytoscape software. Results The predictions indicate that Qishen Huanwu Capsule has 35 main active ingredients capable of reducing the cardiotoxicity of anthracyclines and that there are 36 common targets of ACT and Qishen Huanwu Capsule that are enriched in 133 biological processes and 27 signaling pathways. Conclusions Qishen Huanwu Capsule regulates phosphatidylinositol 3‑kinase/protein kinase B (PI3K/Akt), mitogen-activated protein kinase (MAPK), forkhead box class O (FoxO) and other signaling pathways by regulating targets such as RAC-alpha serine/threonine protein kinase (Akt1), mitogen-activated protein kinase 1 (MAPK1), and mitogen-activated protein kinase 8 (MAPK8) and thereby inhibits oxidative stress and regulates apoptosis and autophagy to reduce the cardiotoxicity of anthracyclines.

Chlorpyrifos is a poisonous pesticide that is highly toxic to fish and aquatic organisms. However, there are fewer reports about how chlorpyrifos influences the redox balance of immune cells. Herein, the head kidney tissue treated with chlorpyrifos to do transcriptome analysis and TCR γ was screened out. Subsequently, we established TCR γ knockdown and overexpression carp head kidney lymphocyte models, respectively, by using RNA interference and pcDNA3.1. Real-time PCR, fluorescent staining, oxidation and antioxidant kit were used to detect the related factors. We found that TCR γ knockdown significantly increased the mRNA expression of HSP70 and HSP90 and decreased the mRNA expression of SOD and CAT. Meanwhile, TCR γ knockdown reduced the activities of GSH, GSG-PX, T-AOC, CAT and SOD and increased the content of MDA and H2 O2 and activities of iNOS. Adverse results were obtained in TCR γ overexpression group. Additionally, TCR γ knockdown significantly increased the mRNA expression of IFN-γ, IL-1β, IL-8, IL-10, Nrf2 and NF-κB, but relieved TCR γ overexpression, in which the process of inflammation was activated. Our results reported here indicated that chlorpyrifos induces redox imbalance-dependent inflammation in common carp lymphocyte through dysfunction of T-cell receptor γ, and HSPs play potential protective role in entire process.

The enrichment of the social environment during lactation alleviates the stress of weaned piglets. It is significant to understand how the enriched social environment improves the weaning stress of piglets. RNA sequencing (RNA-seq) of colonic mucosa, 16S rRNA sequencing of feces, and short-chain fatty acids (SCFAs) of colonic content were used to determine the effects of social contact during lactation. In this study, thirty litter lactating piglets were divided into intermittent social contact (ISC) group that contacted with neighbors intermittently, continuous social contact (CSC) group that contacted with neighbors starting at day (D) 14 after birth, and control (CON) group in which piglets were kept in their original litter. The piglets were weaned at D35 and regrouped at D36. The colonic mucosal RNA-seq, fecal microbes, and SCFAs of colonic contents of 63-day-old piglets were analyzed. The results of RNA-seq showed that compared with the CON group, the pathways of digestion and absorption of minerals, protein, and vitamins of piglets were changed in the ISC group, whereas the pathways of retinol metabolism and nitrogen metabolism in the colonic mucosal were affected and stimulated the immune response in the CSC group. Compared with the CON group, the abundances of pernicious microorganisms (Desulfovibrio, Pseudomonas, Brevundimonas, etc.) in the CSC group and pernicious microorganisms (Desulfovibrio, Neisseria, Sutterella, etc.) and beneficial bacteria (Bifidobacterium, Megamonas, and Prevotella_9) in the ISC group were significantly higher (p < 0.05). The abundances of proinflammatory bacteria (Coriobacteriaceae_unclassified, Coprococcus_3, and Ruminococcus_2) in the CSC group were significantly increased (p < 0.05), but the abundances of SCFAs producing bacteria (Lachnospiraceae_UCG-010, Parabacteroides, Anaerotruncus, etc.) and those of anti-inflammatory bacteria (Eubacterium, Parabacteroides, Ruminiclostridium_9, and Alloprevotella) were significantly reduced (p < 0.05) in the CSC group. Compared with the CON group, the concentrations of microbial metabolites, acetate, and propionate in the colonic contents were reduced (p < 0.05) in the ISC group, whereas the concentration of acetate was reduced (p < 0.05) in the CSC group. Therefore, both ISC and CSC during lactation affected the composition of fecal microbes and changed the expression of intestinal mucosal genes related to nutrient metabolism and absorption of piglets.

This study was conducted to evaluate the effects of feeding fermented wheat bran (FWB) and yeast culture (YC) on growth performance, immune levels, and intestinal microflora in growing-finishing pigs. In total, 96 crossbred pigs were randomly distributed into four treatments with four replicates pens and six pigs per pen. This study was performed using a 2 × 2 factor design: 1) CON (basal diet), 2) FWB (basal diet + 5% FWB), 3) YC (basal diet + 2% YC), and 4) FWB + YC (basal diet + 5% FWB + 2% YC). Dietary FWB supplementation significantly increased the average daily gain and significantly decreased the feed gain ratio of growing-finishing pigs (P < 0.05). Supplementation of FWB and YC improved the immune capacity and reduced the inflammation level of growing-finishing pigs (P < 0.05). In addition, pigs fed FWB, YC, and FWB + YC diets showed better intestinal development and morphology compared with those CON pigs. The relative abundance of Streptococcus in the FWB group was significantly lower than that in the CON group (P < 0.05), and the relative abundance of probiotics (unclassified_f_Lachnospiraceae, Turicibacter) increased significantly (P < 0.05). Furthermore, the relative abundance of probiotics (Lactobacillus, norank_f_Muribaculaceae) in the YC group was significantly increased compared with the CON group (P < 0.05). The results of this study observed positive effects of FWB and YC on growing-finishing pigs, which provides insights into the application of biological feed in swine industry.

In order to explore the differentially expressed genes (DEGs) related to the high incidence of sham-chewing behavior in pregnant sows, 50 sows (Large White × Landrace) were used to observe sham-chewing behavior in early, middle and late gestation. And then according to the number of sham-chewing observed, sows were divided into high incidence of sham-chewing (group H) and low incidence of sham-chewing (group L). After weaning, 4 sows were randomly selected from group H and group L, samples of brain were collected after slaughter, and hypothalamus was sequenced with transcriptome. After bioinformatics analysis, the DEGs were analyzed and enriched in GO and KEGG, afterwards the candidate genes were screened. qRT-PCR was used to verify the accuracy of the sequencing data. Hypothalamic transcriptome results showed that compared with group L, 1286 genes were significantly differentially expressed in group H, among which 934 genes were up-regulated and 352 genes were down-regulated. The results of GO enrichment analysis showed that in biological process, cell component and molecular function, the most significant terms of enrichment are trans-synaptic signaling, neuron part and gated channel activity. These terms are strongly related to synaptic plasticity, synaptic transmission, neurodevelopment and neuropsychiatric diseases, FGF12, SNAP25, CAMK1D, SYNDIG1, GABRD, RBFOX1, CRH, HCN1, HCN4, GLRA3, GRIN2A, ANXA2, MBP and NTF4 genes with high fold change and more in-depth functional research were selected for sham-chewing and depressive emotion-related genes. KEGG enrichment analysis results indicated that differential expressed genes were significantly enriched in 29 pathways, such as neuroactive ligand-receptor interaction, cAMP and Ras-MAPK signaling pathway. In conclusion, the results of hypothalamic transcriptome indicate that there is a molecular genetic basis for depression-like symptoms in group H, the 14 DEGs screened above might be candidate genes involved in high incidence of sham-chewing and depression-like emotion.

Ammonia (NH3) emission is a common threat to farm animals. Selenium (Se) is known for its antioxidant property and can resist several stressors affecting farm animals. The aims of this study were (Ⅰ) to determine how excess NH3 exert nephrotoxic effects in pigs and (Ⅱ) to investigate whether selenomethionine has an alleviative effect on NH3 toxicity. Two diets supplemented with different doses of Se (0.22 mg/kg or 0.50 mg/kg) and two concentrations of NH3 (< 5 mg/m3 or 89.8 mg/m3) were used in a 2 × 2 factorial design trial for a period of 30 days. The results showed that NH3 exposure caused apoptosis and increased the number of apoptotic cells in pig kidneys. Further, the activities of antioxidant enzymes were decreased, and the transcriptional and translational levels of endoplasmic reticulum stress-related genes, Bcl-2 and Caspase family members were increased under NH3 exposure. In addition, Wnt/β-catenin signaling pathway was suppressed after NH3 treatment. Dietary supplement with selenomethionine appears to offer protection against NH3-induced kidney injury in pigs and the pathologic changes above were alleviated. Our findings provide additional insight into the mechanism of NH3 toxicity in pigs while elucidating the role of Se as a potential antidote against NH3 poisoning.

Ammonia is a major harmful gas in the environment of livestock and poultry. Studies have shown that excessive ammonia inhalation has adverse effects in pig heart. However, the mechanism of ammonia-induced cardiac toxicity in pigs has not been reported. L-selenomethionine is a kind of organic selenium (Se) which is easily absorbed by the body. Therefore, in this study, twenty-four 125-day-old pigs were randomly divided into 4 groups: C (control) group, A (ammonia) group, Se group (Se content: 0.5 mg kg−1), and A (ammonia) + Se group. The mechanism of ammonia-induced cardiotoxicity and the alleviating effect of L-selenomethionine were examined. The results in the A group showed as follows: a large number of myocardial fiber edema and cytoplasmic bleakness were observed in the heart; a large number of mitochondrial autophagy were observed; ATP content, ATPase activities and hematological parameters decreased significantly; Endoplasmic reticulum stress (ERS) markers (GRP78, IRE1α, ATF4, ATF6, and CHOP) were significantly induced in the mRNA and protein levels; PI3K/AKT/mTOR signaling pathway was activated; and autophagy key genes and proteins (Beclin-1, LC3, ATG3, and ATG5) were significantly up-regulated. The results of comparison between the A + Se group and the A group were as follows: the degree of edema of cardiac muscle fiber in the A + Se group was somewhat relieved; the level of mitochondrial autophagy decreased; ATP content and ATPase activities increased significantly; the mRNA and protein levels of ERS markers were significantly down-regulated; the expression level of PI3K/AKT/mTOR signaling pathway was decreased; and the mRNA and protein levels of key autophagy genes were decreased. However, the changes of these indexes in the A + Se group were still significantly different from those in the C group. Our results indicated that L-selenomethionine supplementation inhibited ammonia-induced cardiac autophagy by activating the PI3K/AKT/mTOR signaling pathway, which confirmed that L-selenomethionine could alleviate the cardiac injury caused by excessive ammonia inhalation to a certain extent. This study aims to enrich the toxicological mechanism of ammonia and provide valuable reference for future intervention of ammonia toxicity.

Selenium (Se) is an essential trace element to maintain homeostasis in humans and animals. The aim of the present study was to clarify the mechanism of Se deficiency-induced inflammation in the pig's brain. Twenty-four healthy pigs were randomly divided into two groups (n = 12/group): control group (group C) was fed diet with 0.3 mg/kg inorganic Se, and Se-deficient group (group L) was fed diet with 0.007 mg/kg inorganic Se. At the 90th day of the experiment, the histology in the pig's brain was observed by the microscope, the NO levels and iNOS activity were assayed, and the mRNA and protein expression levels of inflammatory cytokines (iNOS, COX-2, NF-κB, and PTGEs) and HSPs (HSP27, HSP40, HSP60, HSP70, and HSP90) were detected by real-time quantitative PCR and Western blot. Compared with group C, both of NO levels and iNOS activity were increased in group L, and the mRNA and protein expression levels of inflammatory cytokines (iNOS, COX-2, NF-κB, and PTGEs) and HSPs (HSP27, HSP40, HSP60, HSP70, and HSP90) were also upregulated; histological observation displayed inflammatory response in the brain of pig. In summary, diet with Se deficiency can activate the iNOS/NF-κB pathway to upregulate the expression of inflammatory cytokines, thereby leading to inflammatory lesions in the pig's brain, and HSPs are involved in the compensatory regulation of inflammation. This study provides a reference for the prevention of pig brain inflammation from the perspective of nutrition.

Cadmium (Cd) is a widely distributed environmental pollutant with immunotoxicity and endocrine toxicity. M1/M2 macrophages participate in the immune response and exert an essential influence on fibrosis. Nevertheless, whether Cd can induce porcineadrenal fibrosis by affecting the polarization of M1/M2 macrophages and its potential regulatory mechanism have not been explored. We added 20 mg/kg CdCl2 to the pig diet for 40 days to investigate the fibrogenic effect of subacute Cd exposure on the adrenal gland. The results indicated that the ACTH and CORT in serum were decreased by 15.26 % and 21.99 %, respectively. The contents of adrenal mineral elements Cd, Cr, Mn were increased up to 34, 1.93, 1.42 folds and Co, Zn, Sn were reduced by 21.57 %, 20.52 %, 15.75 %. Concurrently, the pro-oxidative indicators (LPO, MDA and H2O2) were increased by 1.85, 2.20, 2.77 folds and 3.60, 11.15, 4.11 folds upregulated mRNA levels of TLR4, NF-κB, NLRP3 were observed. Subsequently, the expression of M1 macrophages polarization markers (IL-6, iNOS, TNF-α, CCL2 and CXCL9) were raised by 2.03, 2.30, 2.35, 1.58, 1.56 folds, while M2 macrophages (IL-4, CCL24, Arg1, IL-10, MRC1) showed a 62.34 %, 31.88 %, 50.26 %, 74.00 %, 69.34 % downregulation. The expression levels of AMPK subunits and genes related to glycolysis, oxidative phosphorylation (OXPHOS) and fatty acid oxidation (FAO) were also markedly increased. Additionally, the expression level of TGF-β1, Smad2/3 and downstream pro-fibrotic markers was obviously upregulated. Taken together, we conclude that Cd activates the oxidative stress-mediated TLR4/NF-κB/NLRP3 inflammatory signal transduction, leading to porcine adrenal fibrosis by promoting macrophage polarization toward M1.

Cadmium (Cd) is a heavy metal harmful to animals and humans. Cd exposure causes inflammation or necroptosis in many tissues, including adrenal tissue. However, the current researches on the effects of Cd2+ in adrenal tissues are not enough. Therefore, in our experiment Cd chloride (CdCl2) was added to the piglet's diet at a concentration of 20 mg/kg to study the effects of Cd2+ exposure on the porcine adrenal tissue. Our results showed that Cd2+ exposure could cause inflammation by activating the nuclear factor kappa-B (NF-κB) pathway, which in turn induced necroptosis in adrenal tissue with the activated mitogen-activated protein kinase (MAPK) pathway. The expression increase of inflammatory factors and necroptosis downstream genes, and the downregualtion of cysteinyl aspartate specific proteinase 8 (Caspase 8) proved that Cd2+ exposure caused inflammation and necroptosis in adrenal tissue. We conclude that this report provides more basic theoretical data for exploring the mechanism of adrenal injury.

An enriched environment can promote adaptability of animals to cope with complex environments. A total of 18-week-old 216 laying hens were randomly divided into 2 groups; of which, one group was housed in conventional battery cages (CC, n = 36), and the others were housed in furnished cages (FC, n = 180). At the end of 64 wk of age, 24 chickens of each group were selected for 4-hour transport treatment. The spleen tissues of laying hens were collected before transportation (BT), immediately after transportation, and at 48 h after transportation to detect the expression of the heat shock protective response signaling pathway and inflammatory factors. Serum samples were collected to detect the content of immune cytokines. Transport stress decreased heat shock proteins (HSP; including Small HSP, HSP27, HSP40, HSP60, HS70, HSP90, HSP110) in the CC group (P 0.05) immediately after transportation. At 48 h after transportation, mRNA levels of HSP (except for Small HSP and HSP40) in the FC group were upregulated, which were higher than those at BT (P

Cadmium (Cd) is one common environmental pollutant with systemic toxicity. Lipoxin A4 (LXA4) can regulate transforming growth factor-β (TGF-β) pathway and alleviate tissue injury via binding to formyl peptide receptor 2 (FPR2). The activation of nuclear factor-κB (NF-κB) pathway can promote the occurence of necroptosis. However, whether Cd exposure induces necroptosis in swine myocardium and the role of FPR2/TGF-β/NF-κB pathway in this process are unclear. Hence, we established Cd-exposed swine myocardial injury model by feeding a CdCl2 added diet (20 mg Cd/kg diet). Hematoxylin-eosin (H&E) staining was used to observe the morphological changes, and inductively coupled plasma mass spectrometry (ICP-MS) was performed to detect the levels of ion elements in myocardium. We further detected LXA4 and its receptor FPR2, TGF-β, Nrf2, NF-κB pathway and necroptosis related-genes expressions by RT-PCR and western blot. The results showed that Cd exposure induced necrotic cell death and ion homeostasis imbalance in swine myocardium. Moreover, Cd exposure increased the LXA4 content, inhibited the FPR2 expression, activated TGF-β pathway and suppressed Nrf2 pathway, activating the NF-κB pathway. In addition, Cd exposure increased the expressions of necroptosis related-genes TNF-α, TNFR1, RIP1, RIP3 and MLKL. It indicated Cd exposure induced necroptosis via FPR2/TGF-β/NF-κB pathway, revealing the potential mechanism of Cd-induced cardiotoxicity in swine myocardium.

Pregnant sows in the confined environment have poor welfare and frequently perform stereotypic behaviors. In order to clarify whether highly stereotypic behavior is a sign of increased stress and successfully contributes to coping with or adaptation to adverse environment, fifty pregnant sows (Large White × Landrace) housed in stalls were selected to observe behaviors and analyze physiological parameters [cortisol, major acute phase protein (Pig-MAP) and C-reactive protein (CRP)], and immunological parameters [immunoglobin A (IgA), immunoglobin G (IgG), interleukin 6 (IL-6), interleukin 10 (IL-10), tumor necrosis factor alpha (TNF-α) and interferon gamma (IFN-γ)] in early, middle and late gestation (27th, 62nd and 91st day). A repeated-measures analysis and Friedman test was performed to analyze the differences of behaviors and physiological and immunological parameters. The results showed that lateral lying behavior increased significantly with the progress of sows' gestation, while standing and ventral lying behaviors significantly decreased (p0.05). Sham-chewing, bar-biting, trough-biting and rooting behaviors significantly reduced (p0.05). Furthermore, there was no significant difference in physiological and immune levels in different gestational periods (p0.05). The results also indicated that sham-chewing behavior was positively correlated with serum cortisol, IL-6, IL-10, and negatively correlated with serum IFN-γ in each gestational period (p0.05). Trough-biting behavior was positively correlated with serum TNF-α in middle and late gestation (p0.05). Rooting behavior was positively correlated with serum IgG in each gestational period, and positively correlated with serum Pig-MAP, IL-6, and IL-10 in middle and late gestation (p0.05). In conclusion, the sows with a high incidence of stereotypic behaviors tried to improve stress and humoral immunity to cope with the confined environment, and long-term confined sows might be in a chronic stress state.

The sham-chewing behavior indicates the poor welfare of sows and quite frequently occurs in confined environment. To evaluate the behavior, physiology and immunity of pregnant sows with different frequency of sham-chewing, 63 sows (Large White × Landrace) housed in individual gestation crate were divided into the high (H), middle (M) and low (L) frequency groups based on the sham-chewing frequency analyzed by Hierarchical cluster using SPSS 20.0 systems. The behavioral data, including sham-chewing, standing, sitting, ventral lying, lateral lying, bar-biting and rooting, was observed in early, middle and late pregnancy (days 26–27, 61–62 and 91–92) of the sows. Serum CRP, IL-10 and IFN-γ contents were detected by enzyme-linked immunosorbent assay (ELISA). The results showed that the frequency of sham-chewing significantly reduced with the increase of the pregnancy period (P < 0.001) and the difference among three groups was significant in early and middle pregnancy (P < 0.001). Standing behavior in Group H and M decreased significantly (P = 0.004; P = 0.020). Sitting, ventral lying and lateral lying behaviors were significantly lower in early pregnancy than those in late pregnancy (P < 0.05). In late pregnancy, bar-biting of Group H was significantly increased (P = 0.018), while rooting was decreased (P = 0.033). The CRP level was significantly higher in Group H than L in middle pregnancy (P = 0.031). The IFN-γ level was significantly lower in Group H (P < 0.05). In late pregnancy, IL-10 level was significantly higher in group H and M than L (P < 0.05). Therefore, the pregnant sows with different frequency of sham-chewing showed differences in standing and lateral lying behaviors. The expression of sham-chewing reflects the higher level of stress and lower level of immunity and may indicate the maladaptation of pregnancy sows to the confined environment.

Cadmium (Cd) pollution has become an important public and environmental health issue. Xenobiotic receptors (XRs, aryl hydrocarbon receptor, AHR; constitutive androstane receptor, CAR; pregnane X receptor, PXR) modulate downstream cytochrome P450 enzymes (CYP450s) expression to metabolize xenobiotics and environmental contaminants. However, the underlying mechanisms of cardiotoxicity induced by Cd(II) in swine and the roles of XRs and CYP450s remain poorly understood. In this study, the cardiotoxicity of Cd(II) was investigated by establishing a Cd(II)-exposed swine model (CdCl2, 20 mg Cd/Kg diet). Terminal-deoxynucleotidyl transferase mediated nick end labeling (TUNEL) assay and transmission electron microscope were used to observe the apoptosis. Antioxidant capacity was evaluated by free radicals contents and antioxidant enzymes activities. RT-PCR and western blot were used to measure the expression of XRs, CYP450s and apoptosis-related genes. Our results revealed that Cd(II) exposure activated the XRs and increased the CYP450s expression, contributing to the production of reactive oxygen species (ROS). Cd(II) exposure restrained the antioxidant capacity, causing oxidative stress. Moreover, mitogen-activated protein kinase (MAPK) pathway including c-Jun N-terminal kinase (JNK), extracellular signal-regulated kinase (ERK) and P38 mitogen-activated protein kinase (P38) was activated, triggering the mitochondrial apoptotic pathway. In brief, we concluded that Cd(II) caused mitochondrial pathway apoptosis in swine myocardium via the oxidative stress-MAPK pathway, and XRs-mediated CYP450s expression might participate in this process through promoting the ROS.

Background: Social contact in the early suckling period as an enriched social environment can reduce the aggressive behaviors of piglets at weaning regrouping, and improve their production performance and welfare. We speculated that the social environment could modulate the maturation of piglet intestinal microbiota at later growth stages. Therefore, we performed microbial 16S rRNA gene sequencing from fecal samples, collected at 16, 35, 42, and 63 days of age, to investigate the structure and function of intestinal microbiota in piglets that experienced early social contact. Results: The results showed that the litter weight of the piglets was not significantly different between the control (CON) and social contact (SC) groups at 35 days of age (P > 0.05), but the weight of piglets in the SC group significantly decreased at 63 days compared to the CON group (P < 0.05). While the proportion of Firmicutes were the most abundant bacterial phylum detected in both groups at all times, the proportion of Firmicutes and the Firmicutes-to-Bacteroides ratio were significantly higher in the SC group compared to the CON group and the proportion of Bacteroides was lower in the SC group compared to the CON group at 35, 42, and 63 days of age (P < 0.05). At the genus level, early social contact had a significant positive effect on the level of Lactobacillus at 35 and 42 days of age (P < 0.05), but a negative effect on Prevotella at 35, 42, and 63 days of age (P < 0.05). Furthermore, functional analysis of the microbial composition showed that the changes induced by early social contact mainly altered the relative abundance of metabolic and related pathways. From 35 days of age, the social contact notably had a negative effect on the abundance microbial pathways for protein digestion and absorption and lipid metabolism (P < 0.05). Conclusions: Early social contact truly changed the taxonomy of fecal microbiota in piglets, which in turn, impacted the potential for microbial function within the piglet intestine. At present, we speculate that providing continuous social contact negatively influences the nutrient metabolism for the growing piglets.

An enriched environment is widely used to improve domestic animals’ welfare and promote their natural behaviors. Music can reduce abnormal behavior in humans, nonhuman primates, and rodents. However, little is known about the effects of music on pigs. This study aims to explore the effects of repeated music stimulation on the behavior, physiology, and immunity of growing pigs. A total of 72 hybrid piglets (Large White × Duroc × Minpig) were randomly divided into three groups, including music (Mozart K.448, 60 to 70 dB), noise (recorded mechanical noise, 80 to 85 dB), and control (natural background sound, 0.05). In conclusion, short-term music stimulus (8 d) reduced the stress response, whereas long-term music stimulus (60 d) enhanced the immune responses. In addition, the noise increased the aggressive behavior, and long-term noise reduced the immunity of the growing pigs.

Selenium (Se) deficiency is one of the crucial factors related to nervous system disease and necroptosis. MicroRNAs (miRNAs) play vital roles in regulating necroptosis. However, the mechanism of Se deficiency-induced necroptosis in the pig brain tissue and the role that miRNAs play in this process are unclear. Therefore, in this study, in vitro and pig models of Se deficiency were replicated, and electron microscopy, quantitative real-time polymerase chain reaction (qRT-PCR) and western blot assays were performed. The results showed that brain cells typically undergo necrotic changes, and that Se deficiency suppresses mir-29a-3p, which increases the levels of TNFRSF1A (TNFR1). Subsequently, a distinct increase in the necroptosis markers (RIPK1, RIPK3, and MLKL) and an evident decrease in caspase 8 was observed. And the expression of 10 selenoproteins was decreased. Moreover, the in vitro experiments showed that the expression of mir-29a-3p decreased as the Se content in the medium decreased and the application of an mir-29a-3p inhibitor increased the number of necrotic cells and the accumulation of ROS, and these effects were inhibited by necrostatin-1 (Nec-1) and N-acetyl-cysteine (NAC), respectively. Taken together, we proved that Se deficiency induced necroptosis both in vitro and in vivo through the targeted regulation of TNFR1 by mir-29a-3p in the pig brain.

In order to investigate the effect of gentle handling on the behavior, performance, and meat quality of pigs from weaning to slaughter, 144 6-week-old weaned hybrid Min piglets (a native breed) were selected and divided into a handling group (HG: 9 pens × 8 pigs) and a control group (CG: 9 pens × 8 pigs). After 6 weeks of handling, we observed and then evaluated the pigs’ responses to a handler with behavioral scores. Moreover, we measured heart rate and production performance. Three pigs were randomly selected from each of the 18 pens and divided into a handling group (HG: n = 27) and a control group (CG: n = 27), and the HG pigs were given gentle handling until slaughter. Subsequently, we evaluated meat quality and the production performance of six pigs from each group. The results show that AA test (approaching–avoidance test) scores in HG pigs, the number of contacts with the handler and absence of contact with the handler, were significantly higher than in the CG pigs (p &lt, 0.01). The occurrences of avoidance and looking at the handler were lower in the HG than in the CG group (p &lt, 0.01), however, heart rate was not found to be significantly different between the two groups (p = 0.63). No significant difference was found in the average daily gain, average daily feed intake, and feed conversion ratio between the two groups during the two periods (p &gt, 0.05). The b* value was determined 45 min after slaughter, and it was significantly lower in the HG than that in the CG group (p = 0.002). Furthermore, 2 h after slaughter, the L value of the HG group was significantly higher than that of the CG group (p = 0.047), but no difference was observed in carcass quality or other meat quality indicators between the two groups (p &gt, 0.05). The results indicate that gentle handling could reduce pigs’ anxiety and increase their willingness to approach the handler, increasing the intimacy of the pigs and handlers. However, long-term gentle handling had little effect on pig performance, carcass quality, and meat quality.

This study was performed to compare changes in posture, nursing behavior, and production performance of two breeds of sows. The posture, postural changes, nursing behavior, and production performance of hybrid Damin (Large White × Min pig, n = 32) and Landrace × Large White (n = 32) sows were observed by video recording for 72 h after farrowing and from 07:00 to 09:00 h and 13:00 to 15:00 h within any successive two-day period of each week from the two to five weeks postpartum. The production performances were compared between the two breeds. Except standing at days 1 to 3 postpartum and sitting at day 2 postpartum, there were significant differences in postures between the two breeds of sows. The frequency of ventral-to-lateral recumbency at day 1 and weeks 4 and 5 postpartum and sitting-to-lying at days 1 and 3 and weeks 4 and 5 postpartum was significantly lower for the Damin than for Landrace × Large White sows. The frequency of standing-to-lying in the first 72 h postpartum was significantly higher for the Damin than for Landrace × Large White sows. At days 2 and 3 postpartum, piglet loss was significantly lower for the Damin than for Landrace × Large White sows. The duration of parturition and farrowing interval were significantly longer for the Landrace × Large White than for Damin sows. The number of live piglets was higher for the Damin than for Landrace × Large White sows. The birth weight of litters and weaning weight of piglets were lower for Damin than for Landrace × Large White piglets. These data suggest that the Damin sows showed stronger maternal instincts through their behaviors and postural changes compared with the Landrace × Large White sows.

This study investigated the effects of two factors, enriched environment (EE) and different crossbreeds, on the maternal behavior and physiology of Min-pig hybrid sows. The analysis was performed on a total of 72 multiparous sows, including Duroc &times, Min pig (DM), Landrace &times, Min pig (LM), and Landrace &times, Yorkshire (LY) sows, using a total of 24 sows per cross. The sows were housed in two different farrowing pens, one with straw (EE) and one without straw (barren environment (BE)). The results showed that nest-building behavior, including the frequency, total duration, and bout duration, was significantly higher in EE sows than in BE sows (p &lt, 0.01). The frequency and duration of prepartum nest-building behavior were higher in DM and LM sows than in LY sows (p &lt, 0.0001). During the first three days postpartum, EE sows spent a shorter time in ventral recumbency compared with BE sows (p &lt, 0.05). The oxytocin (p &lt, 0.05) and prolactin (p &lt, 0.01) concentrations of EE sows were significantly higher than in BE sows, however, the concentration of cortisol followed the opposite (p &lt, 0.01). The concentration of oxytocin was significantly higher in DM and LM sows than in LY sows (p &lt, 0.01). In conclusion, both EE increased the expression of hormones related to parental behaviors and prenatal nesting and nursing behavior of sows. Furthermore, an EE can also reduce stress in sows. Min-pig hybrids may inherit highly advantageous characteristics of maternal behavior of Min-pig sows.

Cadmium (Cd) is a toxic substance that pollutes the environment with multiple organs. Long-term exposure to Cd can cause fibrosis in the lungs and other organs of animal body. This article explored the effects of subacute Cd exposure on pig liver fibrosis, as well as the polarization of microRNA (miRNA) and M1/M2 macrophages during this process. Based on the establishment of the pig subacute CdCl2 exposure model, we used immunofluorescence staining, Masson staining, qRT-PCR and western blotting to conduct further research. The results showed that Cd exposure can increase the expression of miRNA-21, decrease the expression of TGF-β and SMAD7, increase the expression of PI3K/AKT signaling pathway, cause the M1/M2 imbalance and the increase of M1 polarization. Meantime, it causes the secretion of inflammatory cytokines (TNF-α, IL-1β, and IL-6), and causes an imbalance in the expression of TIMP1, MMP2, and MMP9, which are related to the degree of fibrosis. And the expression of α-SMA, COL1 and COL3 were up-regulated. In the pig, these results indicate that liver fibrosis caused by subacute CdCl2 exposure is induced by the M1 polarization of macrophages through the PI3K/AKT signaling pathway activated by miRNA-21 signaling pathway. These research results not only enrich the theoretical basis and reference value of Cd toxicology research, but also provide new references and new research ideas for comparative medicine.

Chlorpyrifos (CPF) is an organophosphate insecticide and can cause cell death of animals. In the study, the common carp were exposed to CPF at 0 μg/L (the control group), 1.16 μg/L (the low dose group), 11.6 μg/L (the medium dose group), and 116 μg/L (the high dose group), respectively. The carp were euthanized at the 30th day and gills were collected immediately. The ultrastructural and histopathological observations showed obvious necrosis characteristics and inflammatory injury in the CPF-treated groups. CPF exposure activated the MAPK pathway, in which the mRNA and protein expressions of extracellular signal-regulated (ERK), p38 MAP kinase (p38), and c-Jun N-terminal kinase (JNK) were increased; the mRNAs and proteins of NF-κB and TNF-α were activated; and the mRNAs and proteins of necroptosis related genes were changed (the mRNA and protein expression of RIPK1, RIPK3, MLKL, and FADD were increased and caspase-8 was decreased) with concentration dependency. Taken together, we concluded that CPF exposure activated the MAPK/NF-κB/TNF-α pathway, promoted inflammatory injure and evoked necroptosis in common carp gills. In addition, CPF-induced inflammation and necroptosis was concentration dependency. The toxic effects of CPF on gills provided data for both aquaculture and toxicological studies.
